A large-scale study was conducted to explore AFB1 contamination in feed samples using Thin Layer Chromatography, following an AOAC protocol. Samples were identified which were not compliant with the maximum limits for AFB1 as regulated in the United States and Pakistan. Of a total of 923 samples, 51 samples (5.5%) were not compliant according to Pakistan and 267 (28.9%) were not compliant with US-FDA standards. The overall prevalence of non-compliant samples of compound feed was 26.2% (n = 117) according to US-FDA standards, while none of the samples were non-compliant according to Pakistan standards. Among feed ingredients, the overall prevalence of non-compliant samples was 10.7% (n = 51) and 31.4% (n = 150) according to Pakistan and USFDA standards, respectively. The aim of this study was to evaluate the effects of different anti-mycotoxin feed additives on the concentration of mycotoxins in milk, urine, and blood plasma of dairy cows fed diets artificially contaminated with mycotoxins. Secondarily, performance, total-tract apparent digestibility of nutrients, and blood parameters were evaluated. Twelve multiparous cows (165 ± 45 DIM, 557 ± 49 kg BW, and 32.1 ± 4.57 kg/d milk yield at the start of the experiment) were blocked according to parity, milk yield, and DIM and used in a 4 × 4 Latin square design experiment with 21-d periods, where the last 7 d were used for sampling and data analysis. Treatments were (1) mycotoxin group (MTX), basal diet (BD) without anti-mycotoxin feed additives; (2) hydrated sodium calcium aluminosilicate (HSCA), HSCA added to the BD at 25 g/cow per day; (3) mycotoxin deactivator (MD; Mycofix Plus, dsm-firmenich) added to the BD at 15 g/cow per day (MD15); and (4) MD added to the BD at 30 g/cow per day (MD30). Cows from all treatments were challenged with a blend of mycotoxins containing 404 μg of aflatoxin B 1 , 5,025 μg of deoxynivalenol (DON), 8,046 μg of fumonisins (FUM), 195 μg of T2 toxin (T2), and 2,034 μg of zearalenone (ZEN) added daily to the BD during the last 7 d of each period. Neither performance (milk yield and composition) nor nutrient digestibility was affected by treatments. All additives reduced aflatoxin M 1 (AFM1) concentration in milk, whereas MD15 and MD30 group had lower excretion of AFM1 in milk than HSCA. Deoxynivalenol, FUM, T2, or ZEN were not detected in milk of MD15 and MD30. Concentrations in milk of DON, FUM, T2, and ZEN were similar between MTX and HSCA. Except for AFM1, none of the analyzed mycotoxins were detected in urine of MD30 group. Comparing HSCA to MD treatments, the concentration of AFM1 was greater for HSCA, whereas MD30 was more efficient at reducing AFM1 in urine than MD15. Aflatoxin M1, DON, FUM, and ZEN were not detected in the plasma of cows fed MD30, and DON was also not detected in MD15 group. Plasma concentration of FUM was lower for MD15, similar plasma FUM concentration was reported for HSCA and MTX. Plasma concentration of ZEN was lower for MD15 than MTX and HSCA. Serum concentrations of haptoglobin and hepatic enzymes were not affected by treatments. Blood concentration of sodium was lower in HSCA compared with MD15 and MD30 groups. In conclusion, the mycotoxin deactivator proved to be effective in reducing the secretion of mycotoxins in milk, urine, and blood plasma, regardless of the dosage. This reduction was achieved without adverse effects on milk production or total-tract digestibility in cows fed multi-mycotoxin-contaminated diets over a short-term period. Ochratoxin A (OTA), a secondary fungal metabolite known for its nephrotoxic effects, is prevalent in various feeds and food items. Our recent study suggests that OTA-induced nephrotoxicity is linked to the Sigma-1 receptor (Sig-1R)-mediated mitochondrial pathway apoptosis in human proximal tubule epithelial-originated kidney-2 (HK-2) cells. However, the contribution of Sig-1R to OTA-induced nephrotoxicity involving other forms of regulated cell death, such as ferroptosis, remains unexplored. In this investigation, cell viability, malondialdehyde (MDA) levels, glutathione (GSH) levels, and protein expressions in HK-2 cells treated with OTA and/or Ferrostatin-1/blarcamesine hydrochloride/BD1063 dihydrochloride were assessed. The results indicate that a 24 h-treatment with 1 μM OTA significantly induces ferroptosis by inhibiting Sig-1R, subsequently promoting nuclear receptor coactivator 4 (NCOA4), long-chain fatty acid-CoA ligase 4 (ACSL4), arachidonate 5-lipoxygenase (ALOX5), autophagy protein 5 (ATG5), and ATG7, inhibiting ferritin heavy chain (FTH1), solute carrier family 7 member 11 (SLC7A11/xCT), glutathione peroxidase 4 (GPX4), peroxiredoxin 6 (PRDX6), and ferroptosis suppressor protein 1 (FSP1), reducing GSH levels, and increasing MDA levels (P < 0.05). In conclusion, OTA induces ferroptosis by inhibiting Sig-1R, subsequently promoting ferritinophagy, inhibiting GPX4/FSP1 antioxidant systems, reducing GSH levels, and ultimately increasing lipid peroxidation levels in vitro. The conducive conditions of warm and humid climates can facilitate mold proliferation and subsequent mycotoxin production during food processing and distribution, thereby posing a potential risk to consumer health. However, there exists a significant lack of research regarding the diversity of molds and the presence of ochratoxin A (OTA) and patulin (PAT) in food products available in the Yangzhou market. This study was conducted to assess OTA contamination levels and fungal presence in 57 cereal-based food samples, as well as PAT contamination levels and fungal presence in 50 types of foods, including apples, hawthorn berries, pears, and their derivatives. Ochratoxin A (OTA) was detected in 17 out of 57 cereal-based food samples, with concentrations ranging from 0.93 to 32.69 μg/kg. The contamination rate was determined to be 31.48%, and no samples exceeded the established regulatory limits. Furthermore, seven apple products were identified as contaminated with patulin (PAT), exhibiting concentrations between 26.85 and 192.78 μg/kg. Additionally, three food samples derived from hawthorn showed PAT contamination levels ranging from 29.83 to 88.56 μg/kg. Through purification on potato dextrose agar (PDA) medium, observation of colony morphology, and analysis of internal transcribed spacer (ITS) sequences, a total of 35 fungal strains belonging to 13 genera were identified in cereal-based foods. The predominant genera in cereals included Talaromyces, Fusarium, Aspergillus, and Penicillium. Additionally, twelve fungal strains from five genera (Penicillium, Cladosporium, Aureobasidium, Curvularia, and Alternaria) were isolated and identified in fruits and their derivatives. The findings indicate that OTA and PAT toxins are one of the important risk factors that threaten consumer health. Oat products have gained widespread recognition as a health food due to their rich and balanced nutritional profile and convenience. However, the unique matrix composition of oats, which differs significantly from other cereals, presents specific challenges for mycotoxin analysis. This study presents an ultrahigh-performance liquid chromatography–tandem mass spectrometry (UHPLC-MS/MS) method enhanced with an innovative egg white gel pretreatment for the simultaneous analysis of 13 regulated and unregulated trichothecenes in oats. The method demonstrated excellent performance with high accuracy (> 87.5%), repeatability (< 5.7%), and reproducibility (< 8.1%). Analysis of 100 commercial oat products revealed a concerning detection rate (78%) for at least one of the 11 trichothecenes investigated. Notably, deoxynivalenol, exceeding the standard limit in 2% of samples, exhibited the highest detection rate (62%). Additionally, concerning co-occurrence patterns and positive correlations were observed, highlighting potential synergistic effects. The first-time detection of unregulated mycotoxins (T-2 triol, 4,15-diacetoxyscirpenol, 15-acetoxyscirpenol, and neosolaniol) underscores the need for comprehensive monitoring. This method, while developed for oats, shows potential for broader application to other cereals, though further investigation and confirmation are necessary. In the current study, Ochratoxin A (OTA) levels and prevalence in beer were meta‐analyzed, and non‐carcinogenic risk was calculated using the target hazard quotient. Thirty papers with 70 data reports were included in our study. The pooled prevalence of OTA was 50.60%, 95% CI (confidence interval) (36.97–64.19). Five countries with the highest levels of OTA in beer were South Africa (1170.750 μg/L), Slovakia (31.300 μg/L), Portugal (3.140 μg/L), Tunisia (1.052 μg/L), and Greece (0.707 μg/L). The pooled levels of OTA were 0.089 μg/L, 95% CI (0.077–0.101 μg/L). Meta‐regression shows that OTA prevalence in beer decreased over time insignificantly (p value: .084). Except in South Africa and Slovakia, consumers in other countries are at an acceptable non‐carcinogenic risk due to OTA in beer. Maize forms a key component in poultry feed formulation in Ghana. Nonetheless, there is an existing challenge of low-quality maize due to insect infestation and mycotoxin contamination resulting in poor chicken growth and performance including diminished egg laying. This study compared the effectiveness of ZeroFly® Hermetic (ZFH) storage bags to polypropylene (PP) bags to mitigate insect pest and mycotoxin levels over a 6-month storage period. The study was conducted in threelayer poultry farms in Dormaa Ahenkro, Bono Region, Ghana. One storehouse was used in each poultry farm. In each storehouse, there were thirty-two 100-kg-capacity ZFH and same number of 100-kg PP bags, each containing 100 kg of maize, that is, altogether sixty-four 100-kg-capacity bags in each storehouse. The experiment was set up in a Randomized Complete Block Design with two treatments (ZFH and PP bags), three replications (storehouses), and two sub-replications. In each storehouse, two bags in each treatment were destructively sampled monthly. After 6 months of storage, the predominant insects found were Sitophilus spp. and Sitotroga cerealella, and a larger mean number of insects (live and dead) per 500-g sample was found in the PP bags (149.67 ± 8.67), compared to ZFH bags; 4.67 ± 0.73. The percentage of insect damaged kernels after 6 months was significantly higher in PP bags (67.67 ± 2.62) than ZFH bags (5.00 ± 0.50). Weight loss in the PP bags was > 30%, compared to < 0.3% in ZFH bags. Aflatoxin and fumonisin levels of maize in both types of bags were below the recommended safe threshold of 15 ppb and 4 ppm, respectively. The moisture content of maize in ZFH bags was maintained over the 6-month period whereas that of maize in PP bags increased. Also, crude protein, crude fat, and carbohydrate levels in maize were higher in ZFH bags than PP bags. Data showed ZFH bags conserved maize quality better than the PP bags thereby making maize wholesome for food and feed. Further research to compare the growth and performance of layer poultry in terms of egg laying related to diets of feed from maize stored in ZHF and PP bags is recommended. [ABSTRACT FROM AUTHOR]

Maize (Zea mays L.) is a vital crop susceptible to Gibberella ear rot (GER), a disease caused by Fusarium graminearum, resulting in significant yield losses and mycotoxin production. This study aimed to investigate the correlation between ear characteristics and GER resistance in 74 maize inbred lines (42 with non-stiff stalks and 32 stiff stalks) adapted to the northern Italian environment. Mycotoxin analysis was performed to assess the presence of deoxynivalenol (DON) and zearalenone (ZEA). The results showed a positive correlation between the husk traits, like the husk number and husk cover, and GER resistance in both heterotic groups. A positive correlation was also found between the DON and ZEA concentrations. In addition, we conducted a genome-wide association study (GWAS) which identified novel quantitative trait loci (QTLs) associated with the husk number, husk cover, ear attitude, and infection score. These QTLs can be utilized in marker-assisted selection for breeding new GER-resistant maize varieties. Our study provides valuable insights into the genetic basis of ear traits and their relationship with GER resistance, which can contribute to an improvement in the environmental and economical sustainability of the corn growing system. Fusarium graminearum (Fg) is the primary causal agent of Fusarium head blight (FHB) in wheat, barley, and other small grains in North America and worldwide. FHB results in yield reduction and contaminates grain with mycotoxins that pose threats to human and livestock health. Three genetically distinct North American (NA) populations of Fg have been characterized, which are generally associated with differences in their predominant trichothecene chemotype: NA1/15-acetyl-deoxynivalenol (15-ADON), NA2/3-acetyl-deoxynivalenol (3-ADON), and NA3/3α-acetoxy, 7,15-dihydroxy-12,13-epoxytrichothec-9-ene (NX-2). Recent studies found that the NA3 population had significantly less spread on point-inoculated wheat spikes than the NA1 and NA2 populations, and NX toxins are important for Fg spread and initial infection in wheat. In this follow-up study, to compare the effect of the three populations on initial infection and mycotoxin production on different hosts, we dip-inoculated spikes of the moderately resistant wheat cultivar Alsen and the susceptible barley cultivar Voyager using five strains from each population to evaluate disease, trichothecene mycotoxin accumulation, and trichothecene production per unit of fungal biomass. In dip-inoculated wheat spikes, the NA3 population produced significantly more trichothecene per unit of fungal biomass and accumulated higher levels of trichothecene per plant biomass than the NA1 and NA2 populations, regardless of the disease levels caused by the three populations. In contrast to its critical role during wheat infection, NX toxins had no effect on barley infection. In dip-inoculated barley, the NA1 population was more infectious and caused more severe FHB symptoms than the NA2 and NA3 populations; however, the NA3 population produced significantly higher toxin per unit of fungal biomass in infected barley tissues than the NA1 population. This study delineates the mycotoxin-producing capabilities of Rhizopus microsporus strains isolated from stored grains and nuts. Aflatoxins, particularly aflatoxin B1, are significant due to their deleterious effects on human and animal health and their economic impact on vital agricultural commodities. Employing polymerase chain reaction (PCR) for identification, one isolate was confirmed to produce aflatoxin B1 and was associated with R. microsporus. Thin-layer chromatography (TLC) and ammonia gas tests substantiated the isolate's toxin-producing ability. High-performance liquid chromatography (HPLC) quantified aflatoxin B1 concentration, revealing a yield of 395.19 PPb. This study's findings are unprecedented, marking the first known instance of such isolates producing toxins globally. DNA extraction from fungal hyphae, PCR analysis, nucleotide sequencing, and submission to the National Center for Biotechnology Information (NCBI) database further corroborated the isolate's classification as R. microsporus. The quality of food is one of the emergent points worldwide. Many microorganisms produce toxins that are harmful for human and animal health. In particular, mycotoxins from Fusarium fungi are strictly controlled in cereals. Simple and robust biosensors are necessary for 'in field' control of the crops and processed products. Nucleic acid-based sensors (aptasensors) offer a new era of point-of-care devices with excellent stability and limits of detection for a variety of analytes. Here we report the development of a surface-enhanced Raman spectroscopy (SERS)-based aptasensor for the detection of T-2 and deoxynivalenol in wheat grains. The aptasensor was able to detect as low as 0.17% of pathogen fungi in the wheat grains. The portable devices, inexpensive SERS substrate, and short analysis time encourage further implementation of the aptasensors outside of highly equipped laboratories. [ABSTRACT FROM AUTHOR]

Context: The contamination of feedstuffs with multi-mycotoxins and their transfer to milk is an important concern because it can affect the health of both animals and humans. Aims: This study determined the nutrient composition and quantified the mycotoxins present in dairy feedstuffs and in cow's milk. Methods: Samples of feedstuffs and milk were collected from 11 dairy farms in northern Tunisia for physicochemical composition and mycotoxin analyses. The extraction of mycotoxins was performed by the QuEChERS procedure and the presence of 26 mycotoxins was checked using liquid or gas chromatography with tandem mass spectrometry (LC–MS/MS and GC–MS/MS). Key results: Nutrient contents differ among feedstuffs (P < 0.05). The pH values of oat, ryegrass and pulp silages varied from 4.6 to 5.3. Dry-matter (DM) content was highest for concentrate feeds and hay (90% and 86% DM respectively). Crude protein (CP) content was highest in concentrate feeds (15.8% DM). The neutral detergent fibre, acid detergent fibre and acid detergent lignin contents were highest in green grass, hay, and silage. Eight Fusarium mycotoxins, enniatins (ENA1, ENB, ENB1), beauvericine (BEA), HT-2 toxin (HT-2), deoxynivalenol (DON), 15-acetyldeoxynivalenol (15-ADON), Fumonisin B2 (FB2) and one Alternaria mycotoxin tentoxin (TENT) were detected at low concentrations. There was a significant (P < 0.05) difference in the levels of contamination by ENB between the samples of forages and concentrate feeds. The ENs (ENA1, ENB, ENB1) were the most frequently detected combination of mycotoxins. Milk analysis for fat, CP, lactose and dehydrated dry extract averaged 3.49%, 3.10%, 4.71%, and 8.77% respectively; urea concentration was of 34.01 mg/dL, milk pH was 6.5, and a somatic cell count of 1168.63 103 cells/mL was found. Most milk samples (82%) were contaminated with ENB. Conclusions: The levels of mycotoxin contamination in feedstuffs and milk were low and do not present a risk for dairy cattle and consumer health. Implications: Mycotoxins in feedstuffs and milk should be regularly analysed to make sure that quality and safety standards are met. The safety and nutritional quality of milk for human consumption depends on the quality of the feed sources consumed by dairy cattle. Our study has shown that mycotoxins, harmful toxins, detected in some feed sources (e.g. forage silages, hay and concentrates) could be transferred to milk. Strategies for efficient management of feed sources should be developed and the transferability of mycotoxins to milk should be controlled to ensure that the food supply chain is safe and free from harmful toxins. MXene is a rapidly emerging group of two-dimensional (2D) multifunctional nanomaterials, drawing huge attention from researchers of a broad scientific field. Reporting the synthesis of MXene was the following breakthrough in 2D materials following the discovery of graphene. MXene is considered the most recent developments of materials, including transition metal carbonitrides, nitrides, and carbides synthesized by etching or mechanical-based exfoliation of selective MAX phases. MXene has a plethora of prodigious properties such as unique interlayer spacing, high ion and electron transport, large surface area, excellent thermal and electrical conductivity, exceptional volumetric capacitance, thermal shock, and oxidation resistance, easily machinable and inherently hydrophilic, and biocompatibility. Owing to the abundance of tailorable surface function groups, these properties can be further enhanced by surface functionalization with covalent and non-covalent modifications via numerous surface functionalization methods. Therefore, MXene finds their way to a plethora of applications in numerous fields including catalysis, membrane separation, energy storage, sensing, and biomedicine. Here, the focus is on reviewing the structure, synthesis techniques, and functionalization methods of MXene. Furthermore, MXene-based detection platforms in different sensing applications are survived. Great attention is given to reviewing the applications of MXene in the detection of biomolecules, pathogenic bacteria and viruses, cancer biomarkers food contaminants and mycotoxins, and hazardous pollutants. Lastly, the future perspective of MXene-based biosensors as a next-generation diagnostics tool is discussed. Crucial visions are introduced for materials science and sensing communities to better route while investigating the potential of MXene for creating innovative detection mechanisms. This study reviews global levels of ochratoxin A (OTA) in infant formula and cereal-based foods, using Monte Carlo simulation to assess risks. The review found 24 studies on global OTA levels in infant food and cereal-based products, using databases including PubMed, Scopus, Web of Science and Embase until March 2024. We estimated OTA exposure in infant food based on concentration, intake and body weight. The exposure and hazard quotient margin were calculated using BMDL10 and TDI values. Monte Carlo simulation evaluated human health risks from OTA in infant formula and cereal-based foods. A global study from 14 countries shows varying levels, surpassing EU limits in Tunisia, Ecuador, the USA, and generally in Africa, notably in infant cereals, which had higher levels than formula. Globally, OTA was present in 29.3% of the 3348 samples analyzed, with Lebanon at 95.2% and Brazil at 0%. Analysis indicates only non-carcinogenic risk for infants. While health risks for infants are mostly low, ongoing research and monitoring are vital to minimize OTA exposure in infant food. [ABSTRACT FROM AUTHOR]

This study explores the implementation of the One Sample Strategy (OSS), a co-regulation program aimed at managing mycotoxin risk in Texas maize. Fumonisin-contaminated cereals and oilseeds that contain greater than 5 mg kg−1 of the toxin (B1, B2, and B3) are a risk for equids and rabbits, and levels greater than 60 mg kg−1 are a risk to ruminants. The OSS, previously successful in managing aflatoxin risk in Texas maize, was evaluated for its effectiveness in handling fumonisin risk in maize, specifically as it relates to ruminants. In 2017, 25 analysts across seven firms qualified to participate in the program. To ensure greater accuracy in testing, working control samples were provided to the participating OSS firms with the requirement that their results fall within +/− 20% of the target concentration. Ninety-four percent of the working controls met this specification. The capability to grind maize to the OSS prescribed particle size was met by 100% of participants. To verify testing accuracy, file samples collected from each OSS firm were analysed by UPLC–MS/MS. The 177 fumonisin verification samples analysed by Office of the Texas State Chemist (OTSC) were correlated (r = 0.93) with co-regulation laboratories. Results were plotted in an operating curve to depict type I and type II errors. Error analysis revealed a type I error rate of 13% and type II error rate of 2% for the 5 mg kg−1 guidance level, and 6% and 8%, respectively, for the 60 mg kg−1 guidance level. For 2017, 994 official reports of analysis for fumonisin in whole maize in the Texas High Plains were issued by the seven laboratories that employed 25 OTSC-credentialed analysts. The OSS co-regulation program, supported by a quality systems approach and government regulations, has proven effective in managing fumonisin risk in Texas maize, enhancing both market confidence and livestock safety. The resistance gene Lr34 conditions durable disease resistance to many biotrophic wheat pathogens and has been incorporated into many wheat cultivars throughout the world. Lr67 is a similar adult plant resistance gene that also conditions resistance to multiple wheat diseases. Both genes significantly reduce disease severity and work in an additive manner with other rust resistance genes. To determine the effect of Lr34 and Lr67 on Fusarium head blight (FHB), two doubled haploid populations, segregating for each of these genes, were developed in the Thatcher wheat background by crossing near‐isogenic lines. Progeny from these populations were tested in five Fusarium graminearum‐inoculated disease nurseries and assessed for FHB symptoms, using a visual rating index (VRI), and deoxynivalenol (DON) accumulation. Both Lr34 and Lr67 significantly reduced visual symptoms of FHB and DON overall, though those effects were not significant in all environments. The overall reduction in the group of progeny with the resistant allele compared to the group with the susceptible allele for VRI was 9.4% for Lr34 and 6.8% for Lr67, while for DON it was 16.4% for Lr34 and 14.9% for Lr67. Maize gluten meal (MGM) is a by-product of maize starch and ethanol, produced by the wet milling process. Its high protein content makes it a preferred ingredient in feed. Given the high prevalence of mycotoxins in maize globally, they pose a significant challenge to use of MGM for feed: wet milling could concentrate certain mycotoxins in gluten components, and mycotoxin consumption affects animal health and can contaminate animal-source foods. To help confront this issue, this paper summarizes mycotoxin occurrence in maize, distribution during MGM production and mycotoxin risk management strategies for MGM through a comprehensive literature review. Available data emphasize the importance of mycotoxin control in MGM and the necessity of a systematic control approach, which includes: good agriculture practices (GAP) in the context of climate change, degradation of mycotoxin during MGM processing with SO2 and lactic acid bacteria (LAB) and the prospect of removing or detoxifying mycotoxins using emerging technologies. In the absence of mycotoxin contamination, MGM represents a safe and economically critical component of global animal feed. With a holistic risk assessment-based, seed-to-MGM-feed systematic approach to reducing and decontaminating mycotoxins in maize, costs and negative health impacts associated with MGM use in feed can be effectively reduced. Ochratoxin A (OTA) is a type of mycotoxin commonly found in raw and processed foods. It is essential to be aware of this toxin, as it can harm your health if consumed in high quantities. OTA can induce toxic effects in various cell models. However, a more comprehensive understanding of the harmful effects of OTA on human astrocytes is required. This study evaluated OTA's neurotoxic effects on the Gibco® Human Astrocyte (GHA) cell line, its underlying mechanisms, and the antioxidant N‐acetylcysteine (NAC) ability to prevent them. OTA exposure within 5–30 μM has induced concentration‐dependent cytotoxicity. In the OTA‐treated cells, the levels of reactive oxygen species (ROS) were found to be significantly increased, while the glutathione (GSH) contents were found to decrease considerably. The western blotting of OTA‐treated cells has revealed increased Bax, cleaved caspase‐9/caspase‐3 protein levels, and increased Bax/Bcl‐2 ratio. In addition, exposure to OTA has resulted in the induction of antioxidant responses associated with the protein expressions of Nrf2, HO‐1, and NQO1. On the other hand, the pretreatment with NAC has partially alleviated the significant toxic effects of OTA. In conclusion, our findings suggest that oxidative stress and apoptosis are involved in the OTA‐induced cytotoxicity in GHA cells. NAC could act as a protective agent against OTA‐induced oxidative damage. Our findings suggest that oxidative stress and apoptosis are involved in the ochratoxin A (OTA)‐induced cytotoxicity in Gibco® Human Astrocyte (GHA) cells. N‐acetylcysteine (NAC) could act as a protective agent against OTA‐induced oxidative damage. This study has reported that NAC could alleviate the OTA‐induced cytotoxicity in GHA cells for the first time. These days, a growing consumer demand and scientific interest can be observed for nutraceuticals of natural origin, including apiculture products. Due to the growing emphasis on environmental protection, extensive research has been conducted on the pesticide and heavy metal contamination of bee products; however, less attention is devoted on other food safety aspects. In our review, scientific information on the less‐researched food safety hazards of honey, bee bread, royal jelly, propolis, and beeswax are summarized. Bee products originating from certain plants may inherently contain phytotoxins, like pyrrolizidine alkaloids, tropane alkaloids, matrine alkaloids, grayanotoxins, gelsemium alkaloids, or tutin. Several case studies evidence that bee products can induce allergic responses to sensitive individuals, varying from mild to severe symptoms, including the potentially lethal anaphylaxis. Exposure to high temperature or long storage may lead to the formation of the potentially toxic 5‐hydroxymethylfurfural. Persistent organic pollutants, radionuclides, and microplastics can potentially be transferred to bee products from contaminated environmental sources. And lastly, inappropriate beekeeping practices can lead to the contamination of beekeeping products with harmful microorganisms and mycotoxins. Our review demonstrates the necessity of applying good beekeeping practices in order to protect honeybees and consumers of their products. The article provides a detailed review of recent studies examining the role of probiotics in the degradation of aflatoxins, highly toxic compounds in food that pose significant health risks. Regulatory standards for aflatoxin levels in foodstuffs have been established to protect public health. Various methods--chemical, physical, and biological--have been developed for decontaminating and detoxifying aflatoxins. Among biological methods, probiotics have shown substantial potential in decontaminating aflatoxins. Studies indicate that probiotics can reduce the bioavailability of aflatoxins through adsorption or binding to their cell walls, facilitating their excretion from the body. The extent of aflatoxin absorption by probiotics is influenced by several factor s, including the complexity of the food matrix. Emerging food processing technologies also offer promising results in aflatoxin degradation. Combining these emerging technologies with probiotics can enhance the overall effectiveness of decontamination efforts. In addition, environmental factors influenced by climate change could affect aflatoxin production, necessitating advanced decontamination techniques. The reviewed studies underscore the importance of optimizing probiotic strains and their conditions of use to maximize their effectiveness in aflatoxin degradation, as well as exploring the potential of combining these biological methods with novel technological approaches. AbstractAnimal waste is a potential pollution hazard as it can harbour contaminants, such as antimicrobial residues, mycotoxins, and pesticides, becoming a risk to the public, animal, and environmental health. To assess this risk, 15 experimental broiler chickens orally received contaminants to evaluate excretion levels. An analytical method was previously developed to detect 18 substances in poultry droppings using high-performance liquid chromatography coupled to a tandem mass spectrometer (HPLC-MS/MS). Contaminants including tetracycline, 4-epi-tetracycline, oxytetracycline, 4-epi-oxytetracycline, chlortetracycline, 4-epi-chlortetracycline, tylosin, erythromycin, enrofloxacin, ciprofloxacin, flumequine, florfenicol, sulfachloropyridazine, sulfadiazine, 2,4-dichlorophenoxyacetic acid, zearalenone, alpha- and beta-zearalenol, were extracted with EDTA-McIlvain and acetonitrile. This method showed a p-value < 0.05, RSD < 25%, and R2 > 0.95 in the calibration curves linearity for all analytes. The limit of quantification, selectivity, decision limit for confirmation, matrix effect, precision, and recovery parameters were validated according to European Union document 2021/808/EC, technical report CEN/TR 16059, SANTE/11813/2017 and according to the Veterinary International Conference on Harmonization: VICH GL2 and GL49. This method confirmed the detection of most analytes 12–36 h post-administration and simultaneously detected and quantified mixed contaminants. Thereby, poultry droppings are a potential matrix for spreading contaminants in animal production before slaughter and their control will minimize environmental impacts and mitigate antimicrobial resistance. Aflatoxin B1 (AFB1), the most potent toxic and carcinogenic secondary fungal metabolite, has frequently been reported in food/feed. Nowadays, herbal extracts are considered safe dietary additives to reduce the toxicity of such compounds. The protective capability of various combinations of hydro‐alcoholic extracts (HAEs) of ginger, turmeric, and Shirazi thyme, against the toxicity of AFB1 on the RAW264.7 cell line was investigated. The RAW264.7 cells were exposed to six different concentrations of AFB1 (0.09, 0.18, 0.37, 0.75, 1.5, and 3 μg mL−1) for 48 h to determine the IC50 of AFB1. AFB1 was estimated to have an IC50 of 1.5 μg mL−1 for RAW264.7 cells. Then, the cells were simultaneously incubated with 1.5 μg mL−1 AFB1 and the HAEs for 24 h. The HAEs significantly reduced the toxicity of AFB1 in RAW264.7 cells. HAE of Shirazi thyme showed the highest amount of total phenol content (TPC) and the highest DPPH• activity. In addition, a combination of ginger, turmeric, and Shirazi thyme extract showed the highest antioxidant activity. Rutin, quercetin, and apigenin were the main phenolic components of ginger HAE. A significantly positive correlation was observed between TPC of hydro‐alcoholic extract with ferric reducing antioxidant power (FRAP) and 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide (MTT) values. The experiment aimed to explore the effects of different water contents and storage time on the content of mycotoxins in the mushroom bran of Flammulina velutifolia. This study set up 13%, 30%, 48%, 62%, and 76% moisture gradients of Flammulina velutifolia bran, and measured the content of aflatoxin B1 (AFB1), zearalenone (ZEN), ochratoxin A (OTA), and T-2 toxin (T-2) in Flammulina velutifolia bran, respectively. The dynamic changes in the content of Flammulina velutifolia bran naturally stored for 1~7 days were recorded, and the correlation between the content of each Flammulina velutifolia toxin and storage time was analyzed. Combined with the requirements of the Feed Hygiene Standard for the content of Flammulina velutifolia bran in feed raw materials, it was determined whether the Flammulina velutifolia bran exceeded the limit of mycotoxins. The results showed that at a moisture content of 13%, the contents of AFB1, ZEN, OTA, and T-2 in the mushroom bran of Flammulina velutifolia showed no significant differences within 1~7 days after storage (P>0.05), and none of them exceeded the standard threshold. At 30%, 48%, 62%, and 76% moisture content, the content of AFB1, ZEN, OTA, and T-2 in Flammulina velutifolia bran showed an increasing trend with prolonged storage time. At a moisture content of 30%, the content of AFB1, ZEN, OTA, and T-2 in the Flammulina velutifolia bran did not exceed the standard threshold. At 48%, 62%, and 76% moisture content, the content of AFB1, ZEN, OTA, and T-2 in the Flammulina velutifolia bran did not exceed the standard within 1~2 days after storage. At a moisture content of 62%, the content of AFB1 in the Flammulina velutifolia bran exceeded the standard threshold after the 3rd day, and all three contents exceeded the standard threshold on the 5th day. At 62% and 76% moisture content, both ZEN and OTA in the Flammulina velutifolia bran exceeded the standard threshold on the 7th, with T-2 content not exceeding the standard threshold. At low moisture content (13%), there was no significant correlation between fungal toxin content and storage time (P>0.05). Under medium to high moisture content (48% and 76%), there was a significant positive correlation between fungal toxin content and storage time (P<0.05), and there was a significant positive correlation between different fungal toxins (P<0.05). The study indicates that when safely storing Flammulina velutifolia bran, it is necessary to control the moisture content and storage time. When the moisture content of Flammulina velutifolia bran is below 13% or high moisture Flammulina velutifolia bran is used within two days of leaving the factory, it can ensure that the content of mycotoxins does not exceed the standard. Mycotoxins are secondary fungal metabolites harmful to humans and animals. Patulin (PAT) is a toxin found in different food products but especially in apples and their derivative products. The most common fungi producers of this compound are Aspergillus clavatus and Penicillium expansum. The production of patulin, as other mycotoxins, can be impacted by diverse phenomena such as water and nutrient availability, UV exposure, and the presence of antagonistic organisms. Consequently, gaining a comprehensive understanding of climate and environmental conditions is a crucial step in combating patulin contamination. In this study, moulds were isolated from 40 apple samples collected from seven locations across Hungary: Csenger, Damak, Pallag, Lövőpetri, Nagykálló, and Újfehértó. A total of 183 moulds were morphologically identified, with 67 isolates belonging to the Alternaria, 45 to the Aspergillus, and 13 to the Penicillium groups. The location possessed a higher influence than farming method on the distribution of mould genera. Despite the requirement of higher temperature, Aspergillus species dominated only for the region of Újfehértó with approximately 50% of the isolates belonging to the genus. Four of the seven locations assessed: Csenger, Debrecen-Pallag, Nyírtass and Nagykálló, were dominated by Alternaria species. All isolates belonging to the genera Aspergillus and Penicillium were tested for the presence of the isoepoxidone dehydrogenase (idh) gene, a key player in the patulin metabolic pathway. To guarantee patulin production, this ability was confirmed with TLC assays. The only Aspergillus strain that presented a positive result was the strain Aspergillus clavatus B9/6, originated from the apple cultivar Golden Reinders grown in Debrecen-Pallag by integrated farming. Of the Penicillium isolates only one strain, B10/6, presented a band of the right size (500-600 bp) for the idh gene. Further sequencing of the ITS gene showed that this strain should be classified as Talaromyces pinophilus. Fusarium head blight (FHB) is a plant disease caused by various species of the Fusarium fungus. One of the major concerns associated with Fusarium spp. is their ability to produce mycotoxins. Mycotoxin contamination in small grain cereals is a risk to human and animal health and leads to major economic losses. A reliable site-specific precise Fusarium spp. infection early warning model is, therefore, needed to ensure food and feed safety by the early detection of contamination hotspots, enabling effective and efficient fungicide applications, and providing FHB prevention management advice. Such precision farming techniques contribute to environmentally friendly production and sustainable agriculture. This study developed a predictive model, Sága, for on-site FHB detection in wheat using imaging spectroscopy and deep learning. Data were collected from an experimental field in 2021 including (1) an experimental field inoculated with Fusarium spp. (52.5 m × 3 m) and (2) a control field (52.5 m × 3 m) not inoculated with Fusarium spp. and sprayed with fungicides. Imaging spectroscopy data (hyperspectral images) were collected from both the experimental and control fields with the ground truth of Fusarium-infected ear and healthy ear, respectively. Deep learning approaches (pretrained YOLOv5 and DeepMAC on Global Wheat Head Detection (GWHD) dataset) were used to segment wheat ears and XGBoost was used to analyze the hyperspectral information related to the wheat ears and make predictions of Fusarium-infected wheat ear and healthy wheat ear. The results showed that deep learning methods can automatically detect and segment the ears of wheat by applying pretrained models. The predictive model can accurately detect infected areas in a wheat field, achieving mean accuracy and F1 scores exceeding 89%. Maize, one of the most important cereal crops in Bangladesh, is severely contaminated by fumonisin, a carcinogenic secondary metabolite produced by Fusarium including Fusarium proliferatum. Biocontrol with Bacillus strains is an effective approach to controlling this F. proliferatum as Bacillus has proven antagonistic properties against this fungus. Therefore, the present study aimed to determine how native Bacillus strains can reduce fumonisin in maize cultivated in Bangladesh, where BDISO76MR (Bacillus subtilis) strains showed the highest efficacy both in vitro in detached cob and in planta under field conditions. The BDISO76MR strain could reduce the fumonisin concentration in detached cob at 98.52% over untreated control, by inhibiting the conidia germination and spore formation of F. proliferatum at 61.56% and 77.01%, respectively in vitro. On the other hand, seed treatment with formulated BDISO76MR showed higher efficacy with a reduction of 97.27% fumonisin contamination compared to the in planta cob inoculation (95.45%) over untreated control. The highly cytotoxic macrocyclic trichothecene Isororidin A (C29H40O9) was isolated from the fungus Myrothesium verrucaria endophytic on the wild medicinal plant 'Datura' (Datura stramonium L.) and was characterized by one‐ (1D) and two‐dimensional (2D) NMR spectroscopy. The three‐dimensional structure of Isororidin A has been confirmed by X‐ray crystallography at 0.81 Å resolution from crystals grown in the orthorhombic space group P212121, with one molecule per asymmetric unit. Isororidin A is the epimer of previously described (by X‐ray crystallography) Roridin A at position C‐13′ of the macrocyclic ring. [ABSTRACT FROM AUTHOR]

Alternaria alternata is a prevalent postharvest pathogen that generates diverse mycotoxins, notably alternariol (AOH) and alternariol monomethyl ether (AME), which are recurrent severe contaminants. Nitrogen sources modulate fungal growth, development, and secondary metabolism, including mycotoxin production. The GATA transcription factor AreA regulates nitrogen source utilization. However, little is known about its involvement in the regulation of nitrogen utilization in A. alternata. To examine the regulatory mechanism of AaAreA on AOH and AME biosynthesis in A. alternata, we analyzed the impact of diverse nitrogen sources on the fungal growth, conidiation and mycotoxin production. The use of a secondary nitrogen source (NaNO3) enhanced mycelial elongation and sporulation more than the use of a primary source (NH4Cl). NaNO3 favored greater mycotoxin accumulation than did NH4Cl. The regulatory roles of AaAreA were further clarified through gene knockout. The absence of AaAreA led to an overall reduction in growth in minimal media containing any nitrogen source except NH4Cl. AaAreA positively regulates mycotoxin biosynthesis when both NH4Cl and NaNO3 are used as nitrogen sources. Subcellular localization analysis revealed abundant nuclear transport when NaNO3 was the sole nitrogen source. The regulatory pathway of AaAreA was systematically revealed through comprehensive transcriptomic analyses. The deletion of AaAreA significantly impedes the transcription of mycotoxin biosynthetic genes, including aohR, pksI and omtI. The interaction between AaAreA and aohR, a pathway-specific transcription factor gene, demonstrated that AaAreA binds to the aohR promoter sequence (5'-GGCTATGGAAA-3'), activating its transcription. The expressed AohR regulates the expression of downstream synthase genes in the cluster, ultimately impacting mycotoxin production. This study provides valuable information to further understand how AreA regulates AOH and AME biosynthesis in A. alternata, thereby enabling the effective design of control measures for mycotoxin contamination. DON and ZEN residues in the blood and urine of dairy cows can be used to predict the outer exposure to DON and ZEN expressed per kilogram diet for a risk evaluation based on comparisons to critical dietary concentrations. This method was used to evaluate the exposure of dairy cows from 12 farms located in Brandenburg, Germany, fed rations with unknown DON and ZEN concentrations (N = 244). The corresponding diet concentrations predicted by different methods from analyzed blood and urine samples varied significantly amongst farms from 0 to 1.6 mg/kg for DON and 0 to 3.0 mg/kg for ZEN at a reference dry matter content of 88% but independently of lactational state (post-partum vs. early lactation). This significant variation was noticed below the critical dietary DON concentration of 5 mg/kg, while the ZEN concentration in one farm exceeded the critical ZEN level of 0.5 mg/kg markedly. Predicted DON concentrations of rations increased with the proportion of maize silage, while the high ZEN concentration found in one farm was most likely related to a higher proportion of sugar beet pulp supposedly highly contaminated by ZEN. Exceeding the critical dietary ZEN concentration and significant variations in DON contents below the critical level was not related to performance, reproductive performance, and health-related traits of cows. For a more consistent evaluation of possible associations between the inner exposure of cows to DON and ZEN, more frequent longitudinal observations of both mycotoxin residue levels and performance and health traits are required. ABSTRACT: The aim of this study was to evaluate the effects of different anti-mycotoxin feed additives on the concentration of mycotoxins in milk, urine, and blood plasma of dairy cows fed diets artificially contaminated with mycotoxins. Secondarily, performance, total-tract apparent digestibility of nutrients, and blood parameters were evaluated. Twelve multiparous cows (165 ± 45 DIM, 557 ± 49 kg BW, and 32.1 ± 4.57 kg/d milk yield at the start of the experiment) were blocked according to parity, milk yield, and DIM and used in a 4 × 4 Latin square design experiment with 21-d periods, where the last 7 d were used for sampling and data analysis. Treatments were (1) mycotoxin group (MTX), basal diet (BD) without anti-mycotoxin feed additives; (2) hydrated sodium calcium aluminosilicate (HSCA), HSCA added to the BD at 25 g/cow per day; (3) mycotoxin deactivator (MD; Mycofix Plus, dsm-firmenich) added to the BD at 15 g/cow per day (MD15); and (4) MD added to the BD at 30 g/cow per day (MD30). Cows from all treatments were challenged with a blend of mycotoxins containing 404 μg of aflatoxin B1, 5,025 μg of deoxynivalenol (DON), 8,046 μg of fumonisins (FUM), 195 μg of T2 toxin (T2), and 2,034 μg of zearalenone (ZEN) added daily to the BD during the last 7 d of each period. Neither performance (milk yield and composition) nor nutrient digestibility was affected by treatments. All additives reduced aflatoxin M1 (AFM1) concentration in milk, whereas MD15 and MD30 group had lower excretion of AFM1 in milk than HSCA. Deoxynivalenol, FUM, T2, or ZEN were not detected in milk of MD15 and MD30. Concentrations in milk of DON, FUM, T2, and ZEN were similar between MTX and HSCA. Except for AFM1, none of the analyzed mycotoxins were detected in urine of MD30 group. Comparing HSCA to MD treatments, the concentration of AFM1 was greater for HSCA, whereas MD30 was more efficient at reducing AFM1 in urine than MD15. Aflatoxin M1, DON, FUM, and ZEN were not detected in the plasma of cows fed MD30, and DON was also not detected in MD15 group. Plasma concentration of FUM was lower for MD15, similar plasma FUM concentration was reported for HSCA and MTX. Plasma concentration of ZEN was lower for MD15 than MTX and HSCA. Serum concentrations of haptoglobin and hepatic enzymes were not affected by treatments. Blood concentration of sodium was lower in HSCA compared with MD15 and MD30 groups. In conclusion, the mycotoxin deactivator proved to be effective in reducing the secretion of mycotoxins in milk, urine, and blood plasma, regardless of the dosage. This reduction was achieved without adverse effects on milk production or total-tract digestibility in cows fed multi-mycotoxin-contaminated diets over a short-term period. Greater reductions in mycotoxin secretion were observed with full dose of MD.

Abstract Like many traditional Chinese herbal medicines, preparations from Radix Dipsaci are at risk of contamination by harmful mycotoxins; however, there have been no reports of actual contamination. In this study, we developed an analytical method to simultaneously detect eight mycotoxins in Radix Dipsaci and estimate the exposure risk for consumers. We have developed an analytical method utilizing ultra-high performance liquid chromatography and tandem mass spectrometry to accurately determine the levels of AFB1, AFB2, AFG1, AFG2, OTA, ZEN, T-2 and ST mycotoxins in 45 batches of Radix Dipsaci sourced from major medicinal herb markets across five regions in China. We also analyzed migration of mycotoxins from the raw herbs into water decoction. Based on these results and data on human consumption of the herbal medicine, we estimated risk of exposure and acceptable exposure limits to mycotoxins in the Radix Dipsaci using the “margin of exposure (MOE)” method. Of the 45 batches of Radix Dipsaci, 48.89% contained at least one of the eight mycotoxins, 24.44% contained one, 17.78% contained two and 6.67% contained three. The most frequent mycotoxins were aflatoxin B1, present in 35.56% of batches (at 0.25–34.84 μg/kg); aflatoxin G1, 15.56% (1.99–44.05 μg/kg); and ochratoxin A, 22.22% (16.11–143.38 μg/kg). These three mycotoxins transferred from the raw herb into water decoction at respective rates of 20.20%, 29.14%, and 24.80%. The 95th percentile values of the MOE risk factors for health effects of AFB1 were below 10,000 at high doses but above 10,000 at low doses of Radix Dipsaci long-term treatment. With the reduction in duration of exposure years, the MOE values of AFB1 and AFG1 gradually reverted to within the acceptable range. The mean, 50th, and 95th percentile values of the MOE risk factors for health effects of OTA exceeded 10,000 regardless of whether consumers received a low or high dose of Radix Dipsaci treatment for durations ranging from 1 to lifetime. Based on this exposure and a typical human diet, we have estimated the respective 20-year exposure limits for Radix Dipsaci to be 5.821 μg/kg, 4.035 μg/kg, and 56.073 μg/kg for the three mycotoxins under consideration. Contamination with multiple mycotoxins is frequently observed in Radix Dipsaci, and the three most prevalent contaminants have been found to leach into water decoctions, thereby posing a potential health hazard for individuals consuming this herbal preparation. This work highlights the need to monitor herbal medicines for mycotoxin contamination in order to protect consumers.

Fusarium graminearum is the main pathogenic fungus causing wheat head blight. It not only causes severe crop yield reduction, but also produces fungal toxins such as deoxynivalenol (DON) and 15-acetyl deoxynivalenol (15-ADON), which are dangerous food pollutants to people and livestock. For the genetic functional study of Fusarium graminearum, it is often necessary to culture mycelium on LTB medium in the laboratory to investigate its regulatory effect on DON and 15-ADON synthesis. However, the detection of DON and 15-ADON in the mycelium is focused on extracellular, with less focus on the detection of intracellular. Simultaneous detection of intracellular and extracellular DON and 15-ADON can provide more in-depth information of fungal toxin production and secretion functions. In the study, a method based on gas chromatography-mass spectrometry (GC-MS) for simultaneous detection of DON and 15-ADON was developed. The solvents for extracting DON and 15-ADON can be completely separated and the calibration curves of each extract show good linear relationship within a certain range. The mass spectrometric characteristic scan shows high ionization efficiency for both DON and 15-ADON. The retention time of DON and 15-ADON in chromatogram are 6.98 and 7.68 min, respectively. Qualitative analysis of DON was performed using m/z 295, 235, and 193 under single ion monitor (SIM) mode, while m/z 392, 235, and 193 were used for 15-ADON. The highest intensity ion m/z 235 was selected for the quantification of DON, and m/z 193 for the quantification of 15-ADON. This method was used to detect the intracellular and extracellular levels of DON and 15-ADON in wild-type PH-1 and tri5 mutant with DON biosynthesis deficiency. The intracellular DON and 15-ADON levels in PH-1 are determined to be (149.13±9.15) μg/g and (1833.31±185.33) μg/g, extracellular levels are (5910.35±468.23) μg/g and (45222.12±2726.81) μg/g, respectively. The study identified a mutant strain with the deletion of the first enzyme involved in the synthesis pathway of DON and 15-ADON，both DON and 15-ADON are not detected in the extracellular and intracellular of the tri5 mutant. Additionally, this research confirms that after 7 days of cultivation in LTB medium, the wild-type strain exhibits higher levels of 15-ADON in both intracellular and extracellular compared to DON, with extracellular levels of DON and 15-ADON being higher than intracellular levels. This method can provide a methodological reference for the functional study of toxin synthesis in Fusarium graminearum.

Fungal toxins (generally recognised as mycotoxins) contamination in foods with an agricultural base represents an important food safety issue. In this regard, food processing may play a key role in mycotoxin mitigation through different strategies. The present study focused on investigating the mycotoxin content in grinding before wet milling combined with steeping on a lab scale. Experimental groups included corn kernels giving steeping treatments spiked in five concentrations for each mycotoxin, including aflatoxins (B1, B2, G1, and G2), zearalenone, ochratoxin A, and deoxynivalenol, and mycotoxin levels were measured using HPLC. According to validation results, the detection and quantification limit was 0.033–20 and 0.1–100 µg.kg−1, respectively. All measured mycotoxins significantly decreased after dry-grinding and steeping procedures (p < 0.05), and this reduction was significantly (p < 0.05) higher in comparison to other studies that investigated the effect of steeping of whole corn kernels, which represents a vital role of dry grinding before wet milling. Dry grinding before corn wet milling help to distribute and availability of mycotoxins, which could enhance their water solubility and consequently reduce the mycotoxin content of corn during the steeping procedure. The global yield of wheat is limited by Fusarium head blight (FHB), the most damaging disease of wheat accompanied by mycotoxin contamination. Use of resistant cultivars, from an economical point of view, is the most effective control method for plant diseases. Many naturally occurring secondary metabolites in plants are involved in resistance mechanisms against FHB, especially phenolic compounds with antioxidant properties which caused various colouration of the grain in wheat. The objective of this paper was to evaluate the resistance of wheat with different grain colour on the base of accumulation of deoxynivalenol (DON) in grain and other important FHB traits after inoculation with Fusarium culmorum. Visual symptom score (VSS), Fusarium damaged kernels (FDK) and reduction of grain weight per spike (GWS-R) were determined. This study compared current conventional red wheat cultivars and coloured-grain wheat cultivars or lines with blue aleurone, purple pericarp, red grain and white grain. It was found that the cultivars with a purple pericarp (e.g. Rufia) had the lowest DON content and FDK. Statistically significant interactions between grain colour and year were found for all the variables: DON, VSS, FDK, GWS-R. Red grain materials had the lowest DON levels of all the groups studied in 2016 and 2017, but not in 2018. Koji mold (Aspergillus oryzae) is a key microorganism in brewing and fermentation in Japan. We isolated koji molds from the environment in Niigata Prefecture. Eighty-one environmental samples were placed on isolation medium made from steamed rice with wood ash and 36 Aspergillus section Flavi -like strains were obtained. Of those, 26 strains did not produce aflatoxin. We studied their morphology, sequence of ITS region, calmodulin gene, aflatoxin biosynthetic homologous gene cluster and α-amylase gene and fermentation-related enzyme activities. Furthermore, DNA-seq analysis of 14 strains from 26 non-aflatoxin producing strains were conducted and compared the three mycotoxin biosynthetic gene clusters (aflatoxin, cyclopiazonic acid, and aflatrem) and fermentation-related genes against those of reference strain A. oryzae RIB40. In some strains, gene sequences confirmed the absence of mycotoxin production, but differences in fermentation-related enzyme activities could not be explained well by amino acid substitutions. We classified the 26 isolates into 6 morphology types based on the appearance of colonies and mating types, and it was found that strains of the same morphology type had similar enzymatic profiles and gene sequences. Mycotoxins, secondary metabolites produced by various fungi, pose a significant threat to food and feed safety worldwide due to their toxic effects on human and animal health. Traditional methods of mycotoxin management often involve chemical treatments, which may raise concerns about residual toxicity and environmental impact. In recent years, there has been growing interest in exploring natural alternatives for preventing mycotoxin contamination and detoxification. This review provides an overview of the current research on the use of natural products for mitigating mycotoxin risks in food and feed. It encompasses a wide range of natural sources, including plant-derived compounds, microbial agents, and enzymatic control. The mechanisms underlying the efficacy of these natural products in inhibiting mycotoxin synthesis, adsorbing mycotoxins, or enhancing detoxification processes are discussed. Challenges and future directions in the development and application of natural products for mycotoxin management are also addressed. Overall, this review highlights the promising role of natural products as sustainable and eco-friendly alternatives for combating mycotoxin contamination in the food and feed supply chain. Citrinin (CIT) is a nephrotoxic mycotoxin, produced by several species of Penicillium, Aspergillus, and Monascus. The foodstuffs most frequently contaminated with CIT include cereals, cereal products, and red yeast rice. Studies on the occurrence of CIT in food have shown that the CIT concentrations in processed cereal-based products are generally lower than in unprocessed industry cereal samples. One possible explanation is the reaction of CIT with major food components such as carbohydrates or proteins to form modified CIT. Such modified forms of CIT are then hidden from conventional analyses, but it is possible that they are converted back into the parent mycotoxin during digestion. The aim of this study is therefore to investigate reactions of CIT with food matrix during thermal processes and to gain a deeper understanding of the degradation of CIT during food processing. Yogurt, a milk-derived product, is susceptible to mycotoxin contamination. While various methods have been developed for the analysis of dairy products, only a few have been specifically validated for yogurt. In addition, these methods are primarily focus on detecting aflatoxins and zearalenone. This study aimed to conduct a preliminary investigation into the presence of regulated, emerging, and modified mycotoxins in natural and oat yogurts available in the Spanish market. For this, a QuEChERS-based extraction method was optimized and then validated to detect and quantify 32 mycotoxins using ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). The method was in-house validated for the analysis of natural and oat yogurt in terms of linearity, matrix effect, sensitivity, accuracy, and precision. Satisfactory performance characteristics were achieved; for most of the analytes, LOQs were lower than 2 ng/g, and recoveries ranged from 60 to 110% with a precision, expressed as the relative standard deviation of the recovery, lower than 15%. Subsequently, the validated method was applied to analyze commercial yogurt samples, revealing a notable incidence of beauvericin and enniatins, with some analogues found in up to 100% of the samples. Alternariol methyl ether was also frequently found, appearing in 50% of the samples. Additionally, the study identified regulated toxins such as fumonisins, ochratoxin A , and HT-2 toxin. Mycotoxins, such as aflatoxin B1 (AFB1), deoxynivalenol (DON), fumonisins (FBs), ochratoxin A (OTA), T-2 toxin (T-2), and zearalenone (ZEN), can contaminate animal feeds and pose risks to animal health and production performance. These mycotoxins are commonly found in cereals and grains, with the increased use of cereals in pet food, there is a rising concern about mycotoxin contamination among pet owners. To address this, we analyzed imported brands of feline and canine food from the Chinese market produced in 2021–2022. Ninety-three samples were analyzed, comprising 45 feline food and 48 canine food samples. Among them, 14 were canned food and 79 were dry food. The results indicate that AFB1, DON, FBs, OTA, T-2, and ZEN occurred in 32.26%, 98.92%, 22.58%, 73.12%, 55.91%, and 7.53% of the samples, respectively. The most prevalent mycotoxin was DON, followed by OTA, T-2, AFB1, and FBs, whereas ZEN was less frequently detected. The mean concentrations of the six mycotoxins in pet feed samples were 3.17 μg/kg for AFB1, 0.65 mg/kg for DON, 2.15 mg/kg for FBs, 6.27 μg/kg for OTA, 20.00 μg/kg for T-2, and 30.00 μg/kg for ZEN. The levels of mycotoxins were generally below the limits of the Pet Feed Hygiene Regulations of China and the EU. Notably, a substantial majority of the pet food samples (88 out of 93) were contaminated by two or more mycotoxins. AFB1, FBs, OTA, and ZEN occurred slightly more often in feline food than in canine food. Except for OTA, the contamination rates for the other five mycotoxins in canned food were lower than those in dry food. Moreover, except for AFB1, the levels of the other five mycotoxins in canned foods were lower than those in dry foods. ABSTRACT: Notoginseng Radix et Rhizoma (Sanqi in Chinese) is a precious traditional Chinese herbal medicine. It has the effect of dispersing blood stasis and stopping bleeding, reducing swelling and fixing pain. However, it tends to contaminate with harmful fungi during storage, which may make it much less effective. In order to understand the fungal contamination of Notoginseng Radix et Rhizoma and master its composition of the exogenous fungi. The surface fungi of Notoginseng Radix et Rhizoma samples collected from six Chinese provinces and districts were investigated by using dilution plate method. Detection of aflatoxins by UPLC-MS/MS. The results showed that Penicillium citrinum was dominantly isolated from Notoginseng Radix et Rhizoma samples from No.1 to No.4. Aspergillus flavus, which produces aflatoxin, was dominantly isolated from Notoginseng Radix et Rhizoma samples from No.5 and No.6. In addition, kinds of mycotoxin were assayed which were produced by three of those identified A. flavus. All three fungi strains produced aflatoxin B1 (AFB1) and one strain HBSQ1-5 additionally produced other three kinds of mycotoxin, AFB2, AFG1 and AFG2. It is the results implied that it will be very important to take serious cautions when using Notoginseng Radix et Rhizoma. As well as, understanding the composition of the exogenous fungi of Notoginseng Radix et Rhizoma and the strains of toxin-producing fungi, which can play an important role in guiding the storage of Notoginseng Radix et Rhizoma..

The productivity of domestic animals and the safety of food products derived from them are jeopardised by mycotoxins in animal feed. To control them, feed additives are used, which limit the absorption of mycotoxins in the gastrointestinal tract of animals by binding to them. The study aimed to evaluate the effectiveness of a new in vitro model in experiments on the binding of mycotoxins from buffers and contaminated feed and to confirm the effect of a single sorbent or mixture in binding them.

Background and Aim: Mycotoxins such as aflatoxin B1 and ochratoxin A (OTA) are secondary metabolites in molds that grow in raw materials or commercial feed. This interaction has a synergistic effect on mortality, body weight, feed intake, embryo abnormalities, egg production, and lymphoid organ atrophy. This study was conducted to determine the effect of a mycotoxin detoxifier on the blood profile of broilers that were given feed contaminated with mycotoxin, such as the number of heterophils, lymphocytes, monocytes, mean corpuscular hemoglobin (MCH), and MCH concentration (MCHC). Materials and Methods: A total of 20 day-old chicks (DOC) of Cobb broilers were given four treatments with five replicates. The number of chickens used in this research was determined using statistical calculations, and the data obtained was homogeneous so that the population was represented. Treatments included negative control with basal feed (C-), positive control with mycotoxins contamination (C+), treatment 1: Mycotoxins contamination and mycotoxin detoxification 1.1 g/kg (T1), and treatment 2: Mycotoxins contamination and mycotoxin detoxification 1.6 g/kg (T2). Mycotoxin contamination comprised 0.1 mg/kg aflatoxin B1 and 0.1 mg/kg OTA. The treatment period for chickens was 28 days, from 8 to 35 days. A battery cage was used in this study. Chickens were kept in a closed, ventilated room and the room temperature (27°C) was monitored during the treatment period. Results: Based on the results of statistical data processing, a significant difference (p < 0.05) was observed between chickens fed mycotoxin-contaminated feed (C+) and chickens not fed mycotoxin-contaminated feed (C-) and chickens given 1.6 g/kg mycotoxin detoxification (T2). Mycotoxin detoxification at a dose of 1.6 g/kg had a significant (p < 0.05) effect on the heterophil, lymphocyte, and heterophil lymphocyte ratio, leukocyte, erythrocyte, and hemoglobin levels of the blood broiler in this experiment. On other parameters such as monocytes, MCH, and MCHC, treatment 2 at dose 1.6 g/kg was the best treatment, although there was no significant effect with C- and T1. Conclusion: The administration of mycotoxin detoxifiers at a dose of 1.6 g/kg increased the number of heterophils and the ratio of heterophil lymphocytes, leukocytes, erythrocytes, and hemoglobin in broilers fed mycotoxin-contaminated feed.

Ochratoxin A (OTA), which is one of the most important mycotoxins in terms of human health, can be found in cereal products such as bread, “bazlama” (traditional flatbread), and pita bread, as well as cereals such as wheat, barley, and corn. This study aimed to determine the effect of different fermentation times, baking, and storage for various periods on the presence of OTA in sourdough bazlama. Bazlama flour was contaminated with OTA concentrations of 5 and 10 μg/kg. After two different fermentation times (1.5 and 3 h), baking at 300 ± 5°C, and storage at room temperature (25 ± 2°C) for 0, 5, and 10 days, the change in OTA levels of bazlama samples was determined by the high‐performance liquid chromatography with fluorescence detector (HPLC‐FLD) method. The effect of different storage periods on the presence of OTA is insignificant. Although a general decrease in OTA level has been determined, it has been found that long‐term fermentation (at least 3 h) was more effective, especially in flours with a high concentration (10 μg/kg) of OTA contamination. It has been determined that bazlama made from contaminated flours with OTA levels of 5 and 10 μg/kg contained OTA levels exceeding 3 μg/kg when long‐term fermentation was not used. This is the maximum permitted limit set by the Turkish Food Codex and the European Commission, indicating that it is not suitable for consumption in this position. [ABSTRACT FROM AUTHOR]

Agricultural food commodities are highly susceptible to contamination by fungi and mycotoxins, which cause great economic losses and threaten public health. New technologies such as gamma ray irradiation, ultraviolet radiation, electron beam irradiation, microwave irradiation, pulsed light, pulsed electric fields, plasma, ozone, etc. can solve the problem of fungal and mycotoxin contamination which cannot be effectively solved by traditional food processing methods. This paper summarizes recent advancements in emerging food decontamination technologies used to control various fungi and their associated toxin contamination in food. It discusses the problems and challenges faced by the various methods currently used to control mycotoxins, looks forward to the new trends in the development of mycotoxin degradation methods in the future food industry, and proposes new research directions. Introduction: The gastrointestinal tract is integral to defending against external contaminants, featuring a complex array of immunological, physical, chemical, and microbial barriers. Mycotoxins, which are toxic metabolites from fungi, are pervasive in both animal feed and human food, presenting substantial health risks. Methods: This review examines the pharmacological, toxicological, and microbiological impacts of natural products on mycotoxicosis, with a particular focus on the gut-x axis. The analysis synthesizes current understanding and explores the role of natural products rich in polysaccharides, polyphenols, flavonoids, and saponins. Results: The review highlights that mycotoxins can disrupt intestinal integrity, alter inflammatory responses, damage the mucus layer, and disturb the bacterial balance. The toxins' effects are extensive, potentially harming the immune system, liver, kidneys, and skin, and are associated with serious conditions such as cancer, hormonal changes, genetic mutations, bleeding, birth defects, and neurological issues. Natural products have shown potential anticancer, antitumor, antioxidant, immunomodulatory, and antitoxic properties. Discussion: The review underscores the emerging therapeutic strategy of targeting gut microbial modulation. It identifies knowledge gaps and suggests future research directions to deepen our understanding of natural products' role in gut-x axis health and to mitigate the global health impact of mycotoxininduced diseases. Mycotoxins, especially aflatoxin B1 (AFB1) and fumonisin B1 (FMB1), are common contaminants in cereal-based foods. Instances of contamination are predicted to increase due to the current challenges induced by climate change. Despite the health benefits of whole grains, the presence of mycotoxins in bran remains a concern. Nonetheless, previous research indicates that wheat bran can adsorb mutagens. Therefore, this study investigated the capacity of maize, wheat, and oat brans to adsorb AFB1 and FMB1 under varying in vitro conditions, including pH, binding time, temperature, particle size, and the amount of bran utilized. Maize bran demonstrated a high AFB1 adsorption capacity (>78%) compared to wheat and oat brans. However, FMB1 was not adsorbed by the brans, possibly due to its hydrophilic nature. Lower temperature (≤25 °C) enhanced AFB1 adsorption efficacy in wheat and oat bran, while for maize bran, the highest adsorption occurred at 37 °C. A linear model following Henry's law best explained AFB1 adsorption by the brans. Further studies identified the pericarp layer of bran as the primary site of AFB1 adsorption, with the initial liquid volume being a critical factor. The study concludes that bran could potentially act as an effective bioadsorbent. Further research is essential to confirm the adsorption efficacy and the bioavailability of AFB1 through in vivo experiments. [ABSTRACT FROM AUTHOR]

BACKGROUND: Maize is frequently contaminated with deoxynivalenol (DON) and fumonisins B1 (FB1) and B2 (FB2). In the European Union, these mycotoxins are regulated in maize and maize‐derived products. To comply with these regulations, industries require a fast, economic, safe, non‐destructive and environmentally friendly analysis method. RESULTS: In the present study, near‐infrared hyperspectral imaging (NIR‐HSI) was used to develop regression and classification models for DON, FB1 and FB2 in maize kernels. The best regression models presented the following root mean square error of cross validation and ratio of performance to deviation values: 0.848 mg kg−1 and 2.344 (DON), 3.714 mg kg−1 and 2.018 (FB1) and 2.104 mg kg−1 and 2.301 (FB2). Regarding classification, European Union legal limits for DON and FB1 + FB2 were selected as thresholds to classify maize kernels as acceptable or not. The sensitivity and specificity were 0.778 and 1 for the best DON classification model and 0.607 and 0.938 for the best FB1 + FB2 classification model. Spices are food flavouring agents that are highly used in Iraq. However, they may be contaminated by toxicogenic fungi and subsequent production of mycotoxins. The aim of this study was to investigate the contamination of commonly used spices with fungi using polymerase chain reaction (PCR) assay and to detect fungal mycotoxin using high‐performance liquid chromatography. Thirty‐five spices (seven samples from each black pepper, red pepper, turmeric, cumin and ginger) were cultured on an appropriate medium to identify various fungi species. Later on, the toxigenicity of Aspergillus flavus and Aspergillus niger was determined using a PCR assay. The fungal mycotoxins, including aflatoxins and ochratoxins, were then determined through a high‐performance liquid chromatography using the validated Quick, Easy, Cheap, Effective, Rugged, and Safe (QuECHERS) method. Aspergillus species were the predominantly isolated fungi, followed by Penicillium and Fusarium. The PCR results indicate the high toxigenicity of A. flavus as 85.7% of the strains had aflQ/aflR genes and 79% had PKS15KS/PKS15C‐MeT genes. Regarding mycotoxin contamination in spices, the highest rates of aflatoxins and ochratoxins were found in black pepper (5.913 μg/kg) and red chilli (6.9055 μg/kg), respectively. Deoxynivalenol (DON) is a common mycotoxin that contaminates cereals. Therefore, the development of sensitive and efficient detection methods for DON is essential to guarantee food safety and human health. In this study, an enzyme cascade amplification-based immunoassay (ECAIA) using a dual-functional alkaline phosphatase-linked single-chain fragment variable fusion tracer (scFv-ALP) and MnO2 nanosheets was established for DON detection. The scFv-ALP effectively catalyzes the hydrolysis of ascorbyl-2-phosphate (AAP) to produce ascorbic acid (AA). This AA subsequently interacts with MnO2 nanosheets to initiate a redox reaction that results in the loss of oxidizing properties of MnO2. In the absence of ALP, MnO2 nanosheets can oxidize 3,3′,5,5′-tetramethylbenzidine (TMB) to produce the blue oxidized product of TMB, which exhibits a signal at a wavelength of 650 nm for quantitative analysis. After optimization, the ECAIA had a limit of detection of 0.45 ng/mL and a linear range of 1.2–35.41 ng/mL. The ECAIA exhibited good accuracy in recovery experiments and high selectivity for DON. Moreover, the detection results of the actual corn samples correlated well with those from high-performance liquid chromatography. Overall, the proposed ECAIA based on the scFv-ALP and MnO2 nanosheets was demonstrated as a reliable tool for the detection of DON in corn samples. [ABSTRACT FROM AUTHOR]
