Increasing evidence shows the oncogenic function of FAM83D in human cancer, but how FAM83D exerts its oncogenic function remains largely unclear. Here, we investigated the importance of FAM83D/FBXW7 interaction in breast cancer (BC). We systematically mapped the FBXW7-binding sites on FAM83D through a comprehensive mutational analysis together with co-immunoprecipitation assay. Mutations at the FBXW7-binding sites on FAM83D led to that FAM83D lost its capability to promote the ubiquitination and proteasomal degradation of FBXW7; cell proliferation, migration, and invasion in vitro; and tumor growth and metastasis in vivo, indicating that the FBXW7-binding sites on FAM83D are essential for its oncogenic functions. A meta-evaluation of FAM83D revealed that the prognostic impact of FAM83D was independent on molecular subtypes. The higher expression of FAM83D has poorer prognosis. Moreover, high expression of FAM83D confers resistance to chemotherapy in BCs, which is experimentally validated in vitro. We conclude that identification of FBXW7-binding sites on FAM83D not only reveals the importance for FAM83D oncogenic function, but also provides valuable insights for drug target.

Objective To analyze the effects of pain management nursing combined with collaborative nursing on pain, psychological state and quality of life in patients with advanced tumors. Methods A total of 114 patients with advanced tumors admitted to Ankang Central Hospital from January 2021 to January 2023 were selected. According to the different research methods, the patients were divided into two groups: the control group (50 cases) received collaborative nursing, and the observation group (50 cases) received pain management nursing combined with collaborative nursing. The two groups of patients were matched by PSM, and the pain outcome [American Pain Society Patient Outcome Questionnaire-Revised (APS-POQ-R)], mental state [Self-Rating Depression Scale (SDS), Self-Rating Anxiety Scale (SAS)] and quality of life [World Health Organization Quality of Life-BREF (WHOQOL-BREF)] of the two groups of patients were evaluated before and after the intervention. Results After intervention, the scores of pain degree, pain belief and pain impact on life in observation group were lower than those in the control group, and the scores of pain control satisfaction were higher than those in the control group (P＜0.05). The scores of SDS and SAS in the observation group were lower than those in the control group (P＜0.05). The score of physiology, psychology, environment and social relation in the observation group were higher than those in the control group (P＜0.05). Conclusion The combination of pain nursing and collaborative nursing can effectively relieve the pain degree of patients with advanced tumors, and significantly improve their mental state and quality of life, which is worthy of clinical promotion and application.

The aim of this study was to analyze the role of Ezrin in esophageal squamous cell carcinoma (ESCC) and investigate potential therapeutic targets for ESCC by interfering with Ezrin expression. Bioinformatics analysis revealed that Ezrin expression differed significantly among patients with different clinical stage ESCC. Moreover, there was a significant correlation between Ezrin and yes-associated protein/connective tissue growth factor (YAP1/CTGF) levels in esophageal cancer. Sixty paraffin-embedded ESCC tissue samples were examined and Ezrin and YAP1/CTGF levels were determined using immunohistochemistry. The positive expression rates of Ezrin and YAP1/CTGF were significantly lower in adjacent tissues than in ESCC tissues. Furthermore, knockdown of Ezrin expression inhibited colony formation and reduced cell migration and invasion. Compared with control ESCC cells, protein expression levels of YAP1 and CTGF were significantly downregulated in cells with Ezrin knocked down. We conclude that Ezrin may be involved in ESCC progression through the Hippo signaling pathway.

In order to study the enzymatic properties of laccase derived from Lactobacillus and its potential application in biogenic amine degradation, the gene encoding laccase from Lactobacillus plantarum was heterologously expressed and the recombinant laccase was enzymatically characterized. The optimum reaction temperature and pH, the effects of metal ions and chelators on laccase activity, the tolerance of the recombinant laccase to different concentrations of ethanol and the degradation of biogenic amines by the recombinant laccase were determined. The results showed that the optimum reaction temperature and pH of the purified recombinant laccase was 40 ℃ and 4.0, respectively. Low concentrations of Mg2+, Zn2+ and Ca2+ could promote the activity of the recombinant laccase in different degrees, Zn2+ and Ca2+ being most effective. On the contrary, high concentrations of Mn2+, Fe2+ and ethylenediaminetetraacetic acid could inhibit the activity of recombinant laccase by 85% or more. Toward the substrate 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), the Michaelis constant and maximum reaction rate of the recombinant laccase were 2.49 mmol/L and 0.22 mmol/(L·min), respectively. In addition, the recombinant laccase had a good tolerance to ethanol at low concentration, and the residual activity was more than 75% after incubation in 20% (V/V) ethanol for 1 h. The degradation rates of tyramine and tryptamine by the recombinant laccase were both 100%, while the degradation rates of spermidine, phenethylamine and histamine were 49.60%, 25.27% and 21.80%, respectively. Based on the above results, the recombinant laccase from L. plantarum has great application potential in the degradation of biogenic amines in low-alcohol fermented beverages and fermented foods.

This paper reviewed the clinical data of 5 newborn infants with acute peripheral vascular lesion, and summarized individualized surgical treatment and nursing management. The types of peripheral vascular lesion included mural thrombus at initiation site of left external iliac vein in 2 cases, false aortic aneurysm of posterior tibial of left leg in 1 case, thrombophlebitis of left leg in 1 case, false aortic aneurysm of at left elbow in 1 case. Based on the analysis of cause analysis, implementation of individualized surgical treatment and nursing strategy may achieve good clinical outcomes in the treatment of acute peripheral vascular lesion in neonates.
