Few studies have reported the associations of granulocyte colony-stimulating factor (G-CSF) with cytokine release syndrome (CRS), neurotoxic events (NEs) and efficacy after chimeric antigen receptor (CAR) T-cell therapy for relapsed or refractory (R/R) B-cell acute lymphoblastic leukemia (B-ALL). We present a retrospective study of 67 patients with R/R B-ALL who received anti-CD19 CAR T-cell therapy, 41 (61.2%) patients received G-CSF (G-CSF group), while 26 (38.8%) did not (non-G-CSF group). Patients had similar duration of grade 3–4 neutropenia between the two groups. The incidences of CRS and NEs were higher in G-CSF group, while no differences in severity were found. Further stratified analysis showed that the incidence and severity of CRS were not associated with G-CSF administration in patients with low bone marrow (BM) tumor burden. None of the patients with low BM tumor burden developed NEs. However, there was a significant increase in the incidence of CRS after G-CSF administration in patients with high BM tumor burden. The duration of CRS in patients who used G-CSF was longer. There were no significant differences in response rates at 1 and 3 months after CAR T-cell infusion, as well as overall survival (OS) between the two groups. In conclusion, our results showed that G-CSF administration was not associated with the incidence or severity of CRS in patients with low BM tumor burden, but the incidence of CRS was higher after G-CSF administration in patients with high BM tumor burden. Public key encryption with equality test (PKEET) plays an important role in the cloud storage. It allows a third party to test whether two ciphertexts contain the same message without decryption. Since the third party is not fully trusted, it is sometimes necessary to prevent the third party from testing the ciphertexts all the time. To this end, we propose the notion of revocable public key encryption with equality test (R-PKEET). We give the construction of R-PKEET in the random oracle model, which employs time key to update trapdoor and partial ciphertexts. In details, our scheme enables a user to revoke the third party's test right by sending a time key to the cloud server, which is responsible for updating partial ciphertexts using the time key. Compared with related works, our scheme achieves both lightweight revocation and lower computational complexity by using Shamir's secret sharing and Lagrange interpolating polynomial. Systemic lupus erythematosus (SLE) is an autoimmune disease associated with an imbalance of T helper 17 (Th17) to regulatory T cells (Tregs). However, the underlying mechanism remains unclear. Increasing evidence suggests that circular RNAs play a crucial role in SLE. Although circETS1 was discovered 30 years ago, detailed exploration of its functions remains limited. In this study, we measured the expression levels of circETS1 in peripheral blood mononuclear cells (PBMCs) and CD4+ T cells of patients with SLE by quantitative polymerase chain reaction. The impact of circETS1 expression on the Th17/Treg balance and underlying mechanism were evaluated using double-luciferase reporter, gain-/loss-of-function, and rescue assays. Receiver operating characteristic (ROC) curve analysis was conducted to assess the diagnostic value of circETS1. Both circETS1 and FOXP3 expression were downregulated in the PBMCs and CD4+ T cells of patients with SLE (n = 28) compared with those in the cells of healthy controls (n = 20). Mechanistically, we found that circETS1 can bind directly to the microRNA miR-1205, acting as a sponge to upregulate the transcription of FOXP3, thereby maintaining the Th17/Treg balance. Notably, ROC analysis showed that the expression level of circETS1 in PBMCs had an area under the curve of 0.873 (95% confidence interval: 0.771–0.976; p <.001) for diagnosing SLE, with a sensitivity of 80.00% and a specificity of 89.29%. Finally, we found negative correlations between the level of circETS1 in PBMCs and disease severity (according to the Systemic Lupus Erythematosus Disease Activity Index) in patients with SLE (r = –0.7712, 95% CI: –0.8910 to –0.5509; p <.001). The imbalance in Th17/Treg cells in SLE may be attributed, in part, to the circETS1/miR-1205/ FOXP3 pathway. We present a retrospective study performed on 113 patients with R/R MM who received single anti-BCMA CAR T-cell, combined with anti-CD19 CAR T-cell or anti-CD138 CAR T-cell therapy. Eight patients were given G-CSF after successful management of CRS, and no CRS re-occurred thereafter. Of the remaining 105 patients that were finally analyzed, 72 (68.6%) received G-CSF (G-CSF group), and 33 (31.4%) did not (non G-CSF group). We mainly analyzed the incidence and severity of CRS or NEs in two groups of patients, as well as the associations of G-CSF timing, cumulative dose and cumulative time with CRS, NEs and efficacy of CAR T-cell therapy. Both groups of patients had similar duration of grade 3–4 neutropenia, and the incidence and severity of CRS or NEs.There were also no differences in the incidence and severity of CRS or NEs between patients with the timing of G-CSF administration ≤3 days and those >3 days after CAR T-cell infusion. The incidence of CRS was greater in patients receiving cumulative doses of G-CSF >1500 μg or cumulative time of G-CSF administration >5 days. Among patients with CRS, there was no difference in the severity of CRS between patients who used G-CSF and those who did not. The duration of CRS in anti-BCMA and anti-CD19 CAR T-cell–treated patients was prolonged after G-CSF administration. There were no significant differences in the overall response rate at 1 and 3 months between the G-CSF group and the non–G-CSF group. Our results showed that low-dose or short-time use of G-CSF was not associated with the incidence or severity of CRS or NEs, and G-CSF administration did not influence the antitumor activity of CAR T-cell therapy. MicroRNAs play a critical regulatory role in different cancers, but their functions in renal cell carcinoma (RCC) have not been elucidated. Reportedly, miR-142-3p is involved in the tumorigenesis and the development of RCC in vitro and is clinically correlated with the poor prognosis of RCC patients. However, the molecular target of miR-142-3p and the underlying mechanism are unclear. In this study, we found that miR-142-3p was upregulated in RCC tumor tissues and downregulated in exosomes compared to normal tissues. The expression of miR-142-3p was inversely associated with the survival of patients with kidney renal clear cell carcinoma (KIRC). RhoBTB3 was reduced in RCC, and miR-142-3p plays an inverse function with RhoBTB3 in KIRC. The direct interaction between RhoBTB3 and miR-142-3p was demonstrated by a dual luciferase reporter assay. miR-142-3p promoted metastasis in the xenograft model, and the suppression of miR-142-3p upregulated RhoBTB3 protein expression and inhibited the mRNAs and proteins of HIF1A, VEGFA, and GGT1. Also, the miR-142-3p overexpression upregulated the mRNA of HIF1A, VEGFA, and GGT1. In conclusion, miR-142-3p functions as an oncogene in RCC, especially in KIRC, by targeting RhoBTB3 to regulate HIF-1 signaling and GGT/GSH pathways, which needs further exploration. Background: CD5-positive diffuse large B-cell lymphoma (DLBCL) is a clinically rare subtype of DLBCL with aggressive clinical manifestations and a poor prognosis. It has been demonstrated that the prognostic nutritional index (PNI), an indicator of nutritional status and systemic inflammation, is a significant prognostic factor for several types of lymphoma. The objective of this multicenter retrospective study was to explore the prognostic value of the PNI in patients with CD5-positive DLBCL.Methods: In total, 207 patients with CD5-positive DLBCL were recruited from 11 centers of the Huaihai Lymphoma Working Group. Maximally selected rank statistics analysis was used to identify optimal cutoff points for the PNI. A Cox proportional hazards model was used for univariable and multivariable analyses. Kaplan-Meier curves were used to calculate survival rates and draw survival curves, and the log-rank test was used to compare differences between groups.Results: The median age at diagnosis was 61 years, and the 5-year overall survival rate was 47.5%. According to the maximally selected rank statistics analysis, a score of 49.7 was the optimal cutoff point for the PNI. Subgroup analysis showed that the PNI could re-stratify patients in BCL-2-negative, MYC-negative, high-intermediate-risk and high-risk International Prognostic Index, BCL-6-positive and BCL-6-negative, high Ki-67 score (≥0.9), Ann Arbor stage III/IV, Eastern Cooperative Oncology Group performance status ≥2, and germinal center B subgroups. Multivariable analysis revealed that PNI, age, Eastern Cooperative Oncology Group performance status, albumin level, and red blood cell count were independent prognostic factors for CD5-positive DLBCL.Conclusions: The PNI was a significant prognostic indicator for CD5-positive DLBCL and was able to re-stratify the prognosis for clinicopathologic subgroups of patients with CD5-positive DLBCL. Increased proliferation and impaired death of fibroblast-like synovial cells play an important role in the development of rheumatoid arthritis (RA). Survivin plays an important role in the prodromal stage and prognosis of RA and has been introduced as a biomarker of joint injury in RA patients. The purpose of this study was to explore whether propionate alleviates RA through miR-140-5p/survivin pathway. The synovial tissues of RA patients were collected to detect the expression levels of miR-140-5p and survivin; normal human fibroblast-like synovial cells (HLSs) and RA fibroblast-like synovial cells (RA-FLSs) were cultured and treated with 10 mM of sodium propionate (SP), then the expressions of miR-140-5p and survivin, cell viability and apoptosis were detected; collagen induced arthritis (CIA) rat model was constructed and treated with SP, then the tissue inflammation level and the expression levels of miR-140-5p and Survivin were detected. The expression of miR-140-5p decreased in synovial tissues of RA patients and RA-FLSs cells, while the expression of survivin increased significantly in RA patients. SP promoted miR-140-5p expression and apoptosis in RA-FLSs cells and inhibited survivin expression and cell viability of RA-FLSs cells. In addition, miR-140-5p plays a protective role by targeting survivin. Importantly, in the CIA rat model, SP reduced joint inflammatory response, and the miR-140-5p inhibitor weakened the protective effect of SP. SP can alleviate RA by promoting the expression of miR-140-5p and inhibiting the excessive proliferation and death impairment of RA-FLSs cells induced by survivin. [ABSTRACT FROM AUTHOR]

Objectives. The study evaluated the association between lipoprotein-associated phospholipase A2 (Lp-PLA2) gene polymorphisms and coronary heart disease (CHD), in order to explore the molecular genetics of CHD. Methods. Groups of CHD patients (n = 283) and healthy controls (n = 261) were involved in this study. R92H, V279F, and A379V polymorphisms of LP-PLA2 gene were confirmed using polymerase chain reaction (PCR) and direct DNA sequencing. These polymorphisms and their interaction were also analyzed as potential risk factors of CHD. Results. In this study population, the genotypes of R92H (GG, GA, and AA), V279F (CC, AC, and AA) and A379V (GG, GA, and AA) were studied. There was a significantly difference in frequencies of R92H between CHD patients and controls (P < 0.05). In contrast, no significant difference in frequencies of V279F and A379V existed between CHD patients and controls. Furthermore, R92H and A379V were in strong linkage disequilibrium. Conclusions. With the expansion of cloud computing, an increasing amount of sensitive data is being encrypted and stored in public clouds to alleviate storage and management burdens. Secure equivalence-based retrieval of ciphertexts for multiple users is crucial in a cloud environment where diverse user data resides for processing purposes. Public key encryption with equality test (PKEET) has been introduced as a cryptographic tool to verify if two ciphertexts under different public keys contain the same message. However, existing PKEET schemes often face misuse of trapdoors due to their unlimited lifespan, potentially leading to unauthorized disclosure of user privacy. In this paper, we propose a novel approach called fast and forward-secure equivalence-based comparability (FFEC) for multiple users by employing a forward-secure PKEET (FS-PKEET). This restricts the retrieval process only to ciphertexts generated prior to the most recent trapdoor update. We present a concrete FS-PKEET scheme based on bilinear pairing and demonstrate its security using Bilinear Diffie–Hellman (BDH) assumption in the random oracle model. Comprehensive performance evaluation shows that our work has much efficiency of decryption, trapdoor generation and test execution thanks to greatly reducing the cost of trapdoor generation and thus is practical for the application of secure ciphertext information retrieval in cloud environment. • Propose a stronger public key encryption with equality test. • Accurately control the granularity of time-based authorization. • Formally prove its security against two adversaries in random oracle model. • Show its practical application in cloud environment thanks to its significant high efficiency. This study aimed to explore the mechanism by which systemic lupus erythematosus (SLE) activity is promoted through Treg inhibition from the perspective of ceRNA. qRT-PCR was used to detect the expressions of circETS1, miR-1205, and FoxP3 in clinical SLE patient samples. Overexpression of circETS1and miR-1205, along with knockdown of miR-1205 and FoxP3 were conducted in CD4+ T cells, while the proliferation of helper T cell 17 (Th17) and regulatory T cell (Treg) was detected. Arescue assay was performed to verify the molecular mechanism of circETS1/miR-1205/Foxp3 mRNA axis in regulating CD4+ T cell differentiation. In the in vivo experiment, the expression of miR-1205 in SLE mice was intervened, and renal function, inflammatory factors, and serum complement were measured. Additionally, Treg/Th17 cell ratio was detected by flow cytometry. In SLE patients, Treg cells were found to decrease, while Th17 cells increased. Transfection with circETS1 overexpression led to CD4+ T cells differentiating into Treg cells, causing an imbalance in the Th17/Treg ratio. Transfection of miR-1205 mimic and si-FoxP3 could reverse the effect of circETS1 overexpression. Moreover, inhibiting the expression of miR-1205 showed therapeutic effects on SLE mice. circETS1 inhibits Treg via the miR-1205/FoxP3 axis, thereby promoting SLE activity, which may become a new target for SLE treatment. Group signature (GS) allows each member in a group to do signatures anonymously on behalf of the group under management of a group authority. Membership revocation has been a central issue in GS and widely studied so far. The mechanism of verifier local revocation for GS, first formalized by Boneh and Shacham, has an advantage of making the signers free from fetching the up-to-date information of the revoked users and practicality in the scenario of periodically update in the large population of group users. Most of work related to group signature with verifier-local revocation either can only achieve selfless anonymity or have inefficient constructions due to complicate primitives. Aiming to a recent chosen ciphertext attack-almost-full anonymous notion for GS, this paper presents an efficient GS with verifier local revocation in the standard model by adding a new primitive plaintext searchable encryption into Groth's GS under Canard et al.'s framework. Background: Survivin have been shown to play a crucial role in rheumatoid arthritis (RA); however, the regulatory mechanism of survivin in RA has not been fully elucidated. This study aims to investigate the effect of survivin on the proliferation and apoptosis of human RA fibroblast‐like synoviocyte (RA‐HFLS) cells in RA and its underlying mechanism through the NOTCH pathway. Methods: The RA synovial tissues of 65 RA patients with partial resection of synovium of knee joint by arthroscopy were collected. The expression of survivin in synovial tissue was detected by immunohistochemistry, and the correlation of survivin expression and clinical‐pathological parameters of patients was analyzed. In vitro, the proliferation of HFLS and RA‐HFLS were detected by MTT; the apoptosis of HFLS and RA‐HFLS were detected by flow cytometry; the expression of survivin proteins, key protein factors (Notch1, Jagged1, Hes1) in the Notch pathway, and angiogenesis‐related proteins (vascular endothelial growth factor receptor 1 [VEGFR1], Ang1, Ang2) were determined by western blot. Results: We found that survivin was highly expressed in RA synovial tissues and RA‐HFLS cells, and was positively correlated with erythrocyte sedimentation rate, cyclic citrullinated peptide, C‐reactive protein, Disease Activity Score of 28 joints and other pathological indexes. Knockdown survivin induces RA‐HFLS cell apoptosis, suppresses proliferation and the expression of VEGFR1, Ang1, Ang2. In addition, blocking Notch pathway using FLI‐06 significantly down‐regulated survivin expression. When survivin is up‐regulated, it promotes RA‐HFLS cell proliferation, the expression of VEGFR1, Ang1, Ang2 and suppresses apoptosis by activating the NOTCH. Conclusion: This study confirmed that survivin promotes RA‐HFLS cell proliferation, the expression of angiogenesis‐related proteins and suppresses apoptosis by activating the NOTCH pathway. ID-based encryption with equality test (IBEET) allows a tester to compare ciphertexts encrypted under different public keys for checking whether they contain the same message. In this paper, we first introduce group mechanism into IBEET and propose a new primitive, namely group ID-based encryption with equality test (G-IBEET). With the group mechanism: (1) group administrator can authorize a tester to make comparison between ciphertexts of group users, but it cannot compare their ciphertexts with any ciphertext of any user who is not in the group. Such group granularity authorization can make IBEET that adapts to group scenario; (2) for the group granularity authorization, only one trapdoor, named group trapdoor, should be issued to the tester, which can greatly reduce the cost of computation, transmission and storage of trapdoors in traditional IBEET schemes; (3) G-IBEET can resist the insider attack launched by the authorized tester, which is an open problem in IBEET. In the era of cloud computing, public key encryption with keyword search (PEKS) is an extremely useful cryptographic tool for searching on encryption data, whose strongest security notion is indistinguishability encryption against chosen ciphertext attack (ind-cca). Adballa et al. presented a transformation from identity based encryption (IBE) to PEKS in the Theory of Cryptography Conference 2010. This paper proposes a new framework of ind-cca secure PEKS in the standard model. Our main technical tool is a newly introduced notion of smooth projective hash function with key mapping, in which the hash key hk is mapped into another mapping projection key mhp besides the classical projection key hp. Background: Many studies focused on health inequality between migrant older adults and local older adults, while few study concerned the health inequalities between urban-to-urban and rural-to-urban migrant older adults. This study aimed to compare physical health and mental health between these two groups in Hangzhou, Zhejiang Province, China, and to explore the relationship between cognitive social capital, social integration and health among migrant older adults.Methods: A two-stage stratified sampling method was employed to recruit participants from May to August 2013 in Hangzhou. Measurement data were compared with student's t-tests and multivariate analysis of variance (MANOVA). Multiple linear regression was adopted in this study.Results: A total of 1000 of participants who met the inclusion criteria were analyzed, consisting of 527 (52.7%) urban-to-urban and 473 (47.3%) rural-to-urban migrant older adults. There were no statistically significant difference in physical health and mental health between urban-to-urban and rural-to-urban groups on the whole. However, urban-to-urban migrant older adults had a higher reciprocity and social integration than did in rural-to-urban group (13.36 vs. 12.50, p < 0.01; 40.07 vs. 38.50, p < 0.01). And both of cognitive social capital and social integration were positively related to physical health (social reciprocity: t = 6.69, p < 0.01; social trust: t = 3.27, p < 0.01; social integration: t = 5.66, p < 0.01) and mental health (social reciprocity: t = 4.49, p < 0.01; social trust: t = 5.15, p < 0.01; social integration: t = 10.02, p < 0.01). Overall, the female, widowed, and the oldest among migrant older adults had a worse health.Conclusions: Social capital and social integration were played important roles in health of migrant older adults. The incident direction of earthquake motion is an important factor affecting the seismic response of underground structures. In this study, a three-dimensional (3D) oblique incidence method of SV waves is proposed and the effects of incident angles of SV waves on the seismic response of a lined arched tunnel are evaluated. Based on wave field decomposition principle and equivalent node force method and together with viscous-spring artificial boundary, the oblique incidence method of SV waves is implemented by transforming seismic wave field into the equivalent nodal forces acting on the artificial boundaries. By deriving the distance of the incident waves and the reflected wave on free surface to artificial boundaries, this method can comprehensively consider the phase difference of the seismic wave propagation and the influence of the damping effect of the rock medium on the seismic wave propagation. The method is programed into a dynamic finite element program and its effectiveness is examined by a numerical example. Consequently, the oblique incidence method is applied to evaluate the seismic behaviors of the tunnel. The numerical results reveal that (1) the oblique incidence of the seismic wave results in a larger seismic response; (2) the response amplitudes of the stress and displacement increase with the increase of incident angles and reaching the maximum in the case of 30° incident angle; (3) the damage extent increases with an increase in the incident angles, and the oblique incidence of the seismic wave is believed to increase the spatial difference of damage distribution. Eukaryotic initiation factor 3 (EIF3) is one of the largest and most complex translation initiation factors, which consists of 13 subunits named eukaryotic translation initiation factor 3 subunit A (EIF3a) to EIF3m. EIF3a is the largest subunit of EIF3. Previous studies suggested that EIF3a is a housekeeping gene, recent results have found that EIF3a is closely related to the tumorigenesis and drug resistance. Circular RNAs (circRNAs) derived from biologically important gene can play an important role in gene regulation. However, the mechanism underlying circRNAs' biological functions is not well understood yet. In this work, we screened 31 EIF3a‐derived circRNAs, in which two circEIF3as were identified to be correlated with cisplatin drug sensitivity in lung cancer. Two circEIF3as were found involved in RNA‐binding proteins‐mediated biological processes and may be related to translational regulation according to bioinformatics analyses. CircEIF3as, the transcriptional initiation factor EIF3a transcribed circRNAs, are associated with both drug sensitivity and translation regulation. These findings mean that they may have a functional synergy effect with EIF3a or be valuable therapeutic targets for treatment like EIF3a. This is the first study that exploits circRNAs screening from EIF3a in lung cancer, our findings provide a novel perspective on the function of EIF3a and circEIF3as in lung cancer. Accumulating evidence has shown that lncRNAs and circRNAs are novel regulators of gene expression. The discovery of numerous lncRNAs and circRNAs, and investigation into their structures and functions will contribute to our understanding of the pathogenesis of diseases as well as better prevention, diagnosis, and treatment of diseases. There is a close relationship between circRNAs and lncRNAs regarding their origins and functions. Recent studies have shown that non-coding linear and circular transcripts can be transcribed from the same gene and are potential super-enhancers modulating gene transcription. In this review, we summarize the categories, characteristics, biological functions and databases of both lncRNAs and circRNAs, focusing on their transcriptions derived from the same gene, which might give us a deeper understanding of and enable us to better recognize and distinguish their physiological roles. A new metal–organic polyhedron Cu-MOP with octahedral geometry was constructed by 2,8-dibenzothiophenedicarboxylic acid and dicopper paddlewheel. Cu-MOP exhibited H 2 and CO 2 sorption behavior. Stem cell-based regenerative medicine has great potential to advance the therapeutic treatment of human diseases. Among the various stem cell platforms, mesenchymal stem cells (MSCs) represent one of the most promising options. Currently, there are over 500 clinical trials based on MSCs registered at the NIH's ClinicalTrials.gov website. Although successful expansion of MSCs in vitro has been well-established, higher-yield, billion-cell expansion of MSCs remains a bottleneck. In this study, we successfully demonstrated large-scale culture of human adipose-derived mesenchymal stem cells (AdMSCs) in an industrial, single-use vessel at 3.75 L scale. The vessel offers a precision-controlled environment for the ideal growth of stem cells under simulated hypoxic physiological conditions. Stem cells and their culture media were monitored, analyzed, and controlled, allowing production of AdMSCs in substantial quantities. It is widely cultured under aerobic conditions. High cell density cultivation of E. coli is a powerful technique for the production of recombinant proteins. Indeed, 30% of the FDAapproved biopharmaceuticals on the market are produced in E. coli. In this study, bioceramics of alumina, zirconia, and silica were synthesized through Nd:YAG laser sintering process aiming at studying the microstructure and phase composition based on the various laser processing parameters used. Optimal parameters of scanning speed and average laser power for selective laser sintering were determined. Effect of shifting of consecutive single vector on the process of forming the monolayer was also studied. Following the synthesis, the structure and composition were characterized by scanning electron microscopy and analyzing X-ray diffraction. Synovial tissues from mouse OA model and control groups were collected and miR-9-5p expression levels and macrophage markers were measured with qPCR. The function of miR-9-5p in macrophage polarization was analyzed by flow cytometry and qPCR. Various databases were employed to screen the target genes one of which was validated with dual-luciferase analysis. Following the validation, rescue research was applied, and the signaling pathways were analyzed with Western blotting. Finally, the role of miR-9-5p in the progression of OA was validated in the mouse model. MiR-9-5p was highly expressed in the synovial tissues of the OA model and was positively associated with M1 markers. Function analysis demonstrated that miR-9-5p could promote the progression of OA by promoting M1 polarization and inhibiting M2 polarization in vivo and in vitro. The mechanism analysis demonstrated that miR-9-5p could regulate macrophage polarization via NF-κB and AMPK signaling pathways by inhibiting SIRT1 expression. Nowadays, therapeutic monoclonal antibodies (mAbs) are predominantly produced with mammalian cell culture systems such as those using Chinese hamster ovary (CHO) cells. Efforts are underway to reduce the costs of this process to meet the increasing global demand in biopharmaceuticals; meanwhile, cheaper and faster expression systems are being investigated as alternatives. The yeast, Pichia pastoris, has become a substantial workhorse for recombinant protein production. However, the N-linked glycosylation in P. pastoris, namely high mannose glycosylation, is significantly different from that in CHO or other mammalian cells, including human cells. In this study, a SuperMan5 strain of P. pastoris was constructed using Pichia GlycoSwitch® technology to successfully produce a more mammalian-like immunoglobulin G (IgG) fragment crystallizable (Fc), which showcases the potential of P. pastoris as a next-generation mAb production platform. Importantly, in this study, a strong methanol-independent promoter, PUPP, was applied, which only requires glycerol feeding for protein production. Most P. pastoris promoters used for protein expression are derived from genes in the methanol metabolism pathway, creating safety concerns due to the flammable nature of methanol, especially at large scale. Here, a fed-batch SuperMan5 P. pastoris fermentation was carried out in which methanol induction, as well as its affiliated safety risks, were eliminated. Background: Owing to the increase in life expectancy and sickliness, caring for older adults has become a major challenge in China, where the traditional care system is disintegrating and community- and home-based care have been introduced to respond to this 'silver wave'. However, there is limited knowledge of the dilemmas associated with caring for older adults and the acceptability of community- and home-based care for this population.Methods: Participants were recruited from Xihu District, Hangzhou, from June to July 2017. In-depth interviews were conducted using semi-structured questionnaires. Audio recording, verbatim transcription, and thematic analysis were conducted.Results: A total of 64 older adults from four communities were interviewed. Half of the participants had multiple chronic diseases. The very old individuals and those with severe diseases and in poor financial conditions were observed to be struggling the most. Health status, financial capability, and personality were the main factors affecting the care process. Participants cited the following reasons for staying away from nursing homes: misunderstanding, negative environment, a sense of shame, loneliness, and financial limitations. Community- and home-based cares are popular forms of old-age care; however, some participants exhibited a lack of knowledge about such services.Conclusion: A multi-layered old-age care system is urgently needed for older adults in Zhejiang Province. Further, it is important that such a system integrates the care provided through community- and home-based services with that offered by nursing homes. Accumulating evidence has shown that lncRNAs and circRNAs are novel regulators of gene expression. The discovery of numerous lncRNAs and circRNAs, and investigation into their structures and functions will contribute to our understanding of the pathogenesis of diseases as well as better prevention, diagnosis, and treatment of diseases. There is a close relationship between circRNAs and lncRNAs regarding their origins and functions. Recent studies have shown that non-coding linear and circular transcripts can be transcribed from the same gene and are potential super-enhancers modulating gene transcription. In this review, we summarize the categories, characteristics, biological functions and databases of both lncRNAs and circRNAs, focusing on their transcriptions derived from the same gene, which might give us a deeper understanding of and enable us to better recognize and distinguish their physiological roles. Carotenoids are the most important pigments determining the color of C. × morifolium; however, it is still unknown whether the changes of plastid ultrastructure affect carotenoids accumulation. In this study, we compared the change of carotenoid composition, content, and the plastid ultrastructures in the different developmental stages of capitulum among fourteen C. × morifolium cultivars from seven color groups. We found that the carotenoids and plastids detected at the early stage of capitulum development were similar in all cultivars, including violaxanthin, lutein, and β-carotene, which were present in proplastids and immature chloroplasts. Immature chloroplasts were degraded completely, forming loosely broken plastids during the development of the capitulum in white and pink cultivars. Meanwhile, a number of lipid vesicles appeared at proplastids, which resulted in only trace amounts of carotenoid accumulation in these cultivars. For yellow, orange, red, and brown cultivars, a great number of chromoplasts were found, which contained diverse ultrastructures, such as plastoglobules, tubules, and lipid droplets; these chromoplasts were derived from proplastids or chloroplasts. Compared with the early stage of capitulum development, these cultivars accumulated large amounts of carotenoids, primarily including lutein, lutein derivatives, and their isomers. In green cultivars, proplastids and immature chloroplasts were completely transformed into mature chloroplasts. These chloroplasts mainly contained violaxanthin, lutein, β-carotene, and two new components, (9Z)-lutein and (9'Z)-lutein, compared with carotenoid components presented in proplastids and immature chloroplasts. This research will be helpful for understanding the mechanisms of carotenoid metabolism of C. × morifolium. Furthermore, we found that two different chromoplast transformation patterns could be present in the same tissue cell, which contributed to the research on plastid differentiation and development in higher plants. Background: Histone H2AX phosphorylation at the site of Tyr-142 can participates in multiple biological progressions, which is including DNA repair. Ras pathway is closely involved in human cancers. Our study investigated the effects of Ras pathway via regulating H2AX.Y142ph.Methods: Gastric cancer cell line SNU-16 and MKN1 cells were transfected with Ras for G12D and T35S site mutation. The phosphorylation of H2A.XY142 and ERK1/2, WSTF and MDM2 was detected by western blot. Cell viability, cell colonies and migration was analyzed by MTT assay, soft-agar colony formation assay, and Transwell assay, respectively. The expression of Ras pathway related downstream factors, EYA3 and WSTF was detected by qRT-PCR. The relationship between Ras and downstream factors were detected by ChIP. The cell cycle progression was measured by flow cytometry.Results: RasG12D/T35V transection decreased the phosphorylation of H2A.XY142 and activated phosphorylation of ERK-1/2. H2A.XY142 inhibited cell viability, colonies and migration. H2A.XY142ph altered the expression of Ras downstream factors. CHIP assay revealed that RasG12D/T35V could bind to the promoters of these Ras pathway downstream factors. Silence of EYA3 increased H2A.XY142ph and inhibited cell viability, migration and percent cells in S stage. Furthermore, silence of EYA3 also changed the downstream factors expression. WSTF and H2A.XY142ph revealed the similar trend and MDM2 on the opposite.Conclusion: Ras/ERK signal pathway decreased H2A.XY142ph and promoted cell growth and metastasis. The seismic response of hydraulic tunnels is a complex nonlinear process. What makes the case even more interesting is that the large amount of water in hydraulic tunnels which is likely to induce considerable hydrodynamic pressure acted on tunnel structures during earthquakes. In this work, a full three-dimensional (3D) dynamic finite element model is adopted to conduct a comprehensive assessment of the seismic behaviors of a hydraulic tunnel system. In this analysis, the plastic-damage model is employed to reflect the nonlinear mechanical behaviors of the concrete lining, and a fluid-solid coupling method based on an explicit weighted residual approach is proposed to consider the effects of the hydrodynamic pressures on the seismic response of the hydraulic tunnel. The numerical results indicate that the hydrodynamic pressure contributes to a greater seismic response of the tunnel structure. When the hydrodynamic pressure is considered, the magnitudes of the maximum principal stresses are likely to increase by 50% and the displacement amplitudes are approximately 2 cm more than that of without hydrodynamic pressure. The hydrodynamic pressure exacerbates the damage degree of the tunnel structure, and the waist suffers the most severe damage. [ABSTRACT FROM AUTHOR]

Purpose: An estimated 9 million elderly people accompanied their adult children to urban areas in China, raising concerns about their social capital and mental health following re-location. The aim of this study was to examine the effect of migration on social capital and depression among this population.Methods: Multistage stratified cluster sampling was applied to recruit the migrant and urban elderly in Hangzhou from May to August, 2013. Data were collected from face-to-face interviews by trained college students using a standardized questionnaire. Social capital measurements included cognitive (generalized trust and reciprocity) and structure (support from individual and social contact) aspects. Depression was measured by Geriatric Depression Scale-30 (GDS-30). Chi-square tests and binary logistic regression models were used for analysis.Results: A total of 1248 migrant elderly and 1322 urban elderly were eligible for analysis. After adjusting for a range of confounder factors, binary logistic regression models revealed that migrant elderly reported significantly lower levels of generalized trust [OR = 1.34, 95% CI (1.10-1.64)], reciprocity [OR = 1.55, 95% CI (1.29-1.87)], support from individual [OR = 1.96, 95% CI (1.61-2.38)] and social contact [OR = 3.27, 95% CI (2.70-3.97)]. In the full adjusted model, migrant elderly were more likely to be mentally unhealthy [OR = 1.85, 95% CI (1.44-2.36)] compared with urban elderly.Conclusions: Migrant elderly suffered from a lower mental health status and social capital than their urban counterparts in the emigrating city. Transforming data into ciphertexts and storing them in the cloud database is a secure way to simplify data management. Public key encryption with keyword search (PEKS) is an important cryptographic primitive as it provides the ability to search for the desired files among ciphertexts. As a variant of PEKS, certificateless public key authenticated encryption with keyword search (CLPAEKS) not only simplifies certificate management but also could resist keyword guessing attacks (KGA). In this paper, we analyze the security models of two recent CLPAEKS schemes and find that they ignore the threat that, upon capturing two trapdoors, the adversary could directly compare them and distinguish whether they are generated using the same keyword. To cope with this threat, we propose an improved security model and define the notion of strong trapdoor indistinguishability. We then propose a new CLPAEKS scheme and prove it to be secure under the improved security model based on the intractability of the DBDH problem and the DDH problem in the targeted bilinear group. Chimeric antigen receptor (CAR) T-cell therapy is a new and effective method in relapsed or refractory (R/R) multiple myeloma (MM). This study was aimed to explore the risk factors of infection events. We retrospectively analyzed 68 patients with R/R MM who received CAR T-cell therapy at the Affiliated Hospital of Xuzhou Medical University from June 2017 to June 2021.35 patients received anti-CD19 combined with anti-BCMA CAR T-cell therapy and 33 patients received anti-BCMA CAR T-cell therapy alone. Infection events in patients who received ≥4 prior lines of treatment or with grade 3–5 cytokines released syndrome (CRS) mainly occurred within 4 months after CAR T-cell infusion(CTI). The duration of infection-free survival was positively correlated with progression-free survival of patients with R/R MM (R2 = 0.962, p < 0.001) and the first infection event was closely accompanied by the disease relapse or progression. Treatment lines (p = 0.05), duration of ANC<500 cells/mm3 after CTI (p = 0.036), CRS grade (p = 0.007) and treatment response (p < 0.001) were the independent risk factors associated with infection for a multivariable model. The infection incidence was higher in patients with dual CAR T-cell therapy than with mono CAR T-cell therapy18 months after CTI although no statistic differences were observed within 18 months. Infections after CTI were closely associated with more lines of prior treatment, longer duration of ANC<500 cells/mm3, higher grade CRS and poor treatment response. It can be used as a highly efficient and recyclable chiral organocatalyst for Michael addition of cyclohexanone with ( E)-β-nitroalkenes in methanol at room temperature. The Michael addition affords the corresponding products in satisfactory yields of isolated products (78-98%) with high diastereoselectivities and excellent enantioselectivities (up to 98% dr and up to 99% ee). Single-use, stirred-tank bioreactor systems have been used in large-scale production for a number of years. Bench-scale, stirred-tank bioreactors have not been commercially-available for single-use until recently. The New Brunswick™ CelliGen® BLU pitched-blade bioreactor was introduced in 2009, and the CelliGen BLU packed-bed bioreactor, in 2012. Little information is currently available on the utility of these bioreactors for bench-scale production of recombinant products. Thus, we designed this study to perform multiple comparisons with these single-use bioreactors and their tradi-tional glass vessel counterparts. The data comparisons included: (1) CelliGen BLU pitched-blade vs. glass pitched-blade; and (2) CelliGen BLU pitched blade in batch mode vs. CelliGen BLU packed-bed in perfusion mode. Chinese hamster ovary (CHO) cells were used to measure alkaline phosphatase (ALKP) pro-duction in each bioreactor. The final measured concentration of ALKP, after eight days of batch-mode culture in the single-use, pitched-blade bioreactor, was 1.6 U/mL compared to 2.1 U/mL in the reusable bioreactor. After six perfusion harvests in the single-use, packed-bed bioreactor, the combined ALKP production was 16.2 U/mL compared to 17.4 U/mL in the reus-able bioreactor in batch mode. Multiple batch culture runs in the pitched-blade bioreactor would be required to match the output of a single run in the packed-bed bioreactor in perfusion mode. Results demonstrate that there are no significant differences between the reusable and single-use systems for bench-scale production of recombinant proteins. Our results also suggest that the CelliGen BLU packed-bed bioreactor, when operated in perfusion mode, is superior to the CelliGen BLU pitched-blade bioreactor when operated in batch mode, confirming our studies from 2012. It is tricky to determine whether two ciphertexts contain the same message when the messages are encrypted with different public keys. public key encryption with equality test (PKEET) addresses this problem without decryption. By integrating PKEET with identity-based encryption, identity-based encryption with equality test (IBEET) simplifies the certificate management in PKEET. In this paper, we first propose an IBEET scheme that can resist offline message recovery attacks (OMRA) and requires neither the dual-tester setting nor the group mechanism. With the help of some mathematical assumptions, we demonstrate the security of our scheme. Experiment results reveal that our scheme is efficient. From the perspective of usability, we explain why our scheme is more appropriate to be applied in healthcare social Apps than other OMRA-resistant schemes. • This is the first OMRA-resistant IBEET scheme that is free of the dual-tester setting and the group mechanism. • We prevent the adversaries from launching OMRA in two ways: Prevent the adversary from generating new ciphertexts. Our scheme combines IBEET with authenticated encryption (Wang et al., 2019). The sender encrypts the message using its private key as an additional input; Prevent the adversary from abusing the trapdoors. We introduce Type-6 authorization (Zhang et al., 2019) into IBEET. That is, a receiver (Alice) specifies a target receiver (Bob) and generates the trapdoor. Through the trapdoor, the tester could only compare Alice's specific ciphertext with Bob's all ciphertexts, but could not compare Alice's specific ciphertext with any ciphertext of any other receiver than Bob. • We demonstrate that, in the random oracle model, our OMRA-resistant IBEET scheme satisfies OW-ID-CCA security and IND-ID-CCA security. Objective To explore the molecular mechanism of Res in regulation of pulmonary fibrosis (PF). Methods Rats were injected with bleomycin (BLM) to establish a PF model and treated with resveratrol (Res) and/or miR-21 agomir. After 14 days, lung tissues were collected for Hematoxylin-eosin and Masson’s staining, and real-time quantitative polymerase chain reaction and Western blot were performed to detect fibrosis-related protein expression and the activation of the TGF-β1/Smad pathway. In vitro , MRC-5 cells were pretreated with TGF-β1, Res, and/or miR-21 agomir. After 48 h, total soluble collagen was detected with a Sircol Soluble Collagen Assay. Subsequently, a miR-21 mimic was transfected into MRC-5 cells, and a luciferase reporter assay was employed to verify whether miR-21 targeted Smad7. Results Res reversed the increased levels of miR-21 induced by BLM and alleviated serious PF symptoms, but agomiR-21 treatment effectively impaired the above manifestations. In vivo , miR-21 inhibited the decreases of TGF-β1 and p-Smad2/3 that were induced by Res. In vitro , miR-21 significantly disrupted the positive effect of Res on TGF-β-induced collagen deposition, as well as the levels of Fn, α-SMA, p-Smad2, and Smad7. In addition, Smad7 was found to be a direct target of miR-21-5p. TGF-β stimulation led to an enormous increase in p-c-Jun, c-Jun, and c-Fos, which were significantly reduced by Res. Finally, miR-21 sharply reduced the increased phosphorylation levels of ERK, JNK and p38 that were induced by Res. In the identity-based broadcast encryption (IBBE), only these users whose identities are chosen in the ciphertext computing can decrypt the encrypted message. In this paper, we introduce an extension of IBBE, namely Identity-Based Broadcast Encryption for Inner Product (IBBE-IP), where message encryption is replaced by inner product encryption (IPE) introduced by Abdalla et al. (PKC 2015). Precisely, in the IBBE-IP, the private key is associated with a pair of an identity and a vector (I D , y →). The user with private key of (I D , y →) can decrypt the encrypted vector x → for an identity set S selected by the encryptor and learn the inner product 〈 x → , y → 〉 if and only if I D ∈ S. Differing from the IBBE, the decryption in the IBBE-IP yields the inner product associated with the encrypted vector without leaking any information of the vector. The encrypted vector is protected as long as the number of selected identities is less than their length. We present a construction of IBBE-IP with constant-size private keys and it supports unbounded private key queries, which was unachieved in the previous works of IPE in the public-key setting. As a relatively successful agroforestry system, whether the addition of more intercropped species in the rubber–tea agroforestry system extends the benefits is still unknown but worth exploring because diverse cash crop production helps farmers resist market risks and may bring the ecological functions of rubber agroforestry systems closer to those of natural forests. Therefore, understanding the effects of plant interspecific competition on both plants and soil with the increase in intercropped species number in such agroforestry systems is vital for the improvement of rubber–tea agroforestry systems. For these reasons, we selected a monocultural rubber plantation, rubber–tea, rubber–orange–tea and jungle-like rubber–tea agroforestry systems for studying plant water absorbing patterns, plant water use efficiency, and the nutrient status of plants and soil through stable isotope (13C, 2H and 18O) techniques and nutrient diagnosis (C, N, P, K, Ca and Mg) of plant organs (leaves, stems and roots), soil and litter. We found that rubber trees primarily absorbed water from deeper soil layers with increases in their intercropped species number and the formation of increasingly obvious hydrologic niche differentiation between rubber trees and the intercropped plants. In addition, soil nutrient status first improved but then declined with an increase in the number of intercropped species. However, only a slight impact was observed on the nutrient status and water use efficiency of rubber trees, even in the complex rubber–tea agroforestry system. However, the intercropped species experienced resource shortages, especially P shortages, since their water-absorbing zones almost overlapped. Therefore, we suggest that more diverse intercropped species compositions in rubber–tea agroforestry systems would offset the intercropping benefits for both plants and soil. Effect of cryogenic burnishing on the corrosion product film formation mechanism of Ti-6Al-4V titanium alloy has been investigated in this work. The corrosion product films of both burnished and unburnished specimens exhibited three distinct oxide layers. Compared with un-burnished specimens, burnished specimens possessed a thicker subsurface layer with high-valent oxides and additional Al 2 O 3 in the inner layer. The chemical activity, diffusion, and distribution of alloy elements were changed by high-density dislocations, nano-grains, twins and compressive residual stress induced by cryogenic burnishing, resulting in the enhancement of the compactness, thickness, growth rate and protection of corrosion product film on the Ti-6Al-4V alloy. In this paper, we propose a two-round k -out-of- n oblivious transfer scheme with the minimum communication cost. In our proposed scheme, the messages sent by the receiver R to the sender S consist of only three elements, which is independent of n and k , while the messages from S to R are ( n + 1 ) elements when the sender holds n secrets. Our scheme features a nice property of universal parameter, where the system parameter can be used by all senders and receivers. The proposed k -out-of- n oblivious transfer scheme is the most efficient two-round scheme in terms of the number of messages transferred between two communicating parties in known constructions. Nucleolar and spindle‐associated protein 1 (NUSAP1) is an important functional protein during mitosis, and its abnormal expression is implicated in progression of different types of tumors. However, the role of NUSAP1 in gliomas remains unclear. Methods: NUSAP1 expression in gliomas with different grades was investigated based on GEO glioma datasets. Kaplan‐Meier survival analysis was used to evaluate its prognostic significance. In vitro assays were also performed to evaluate effects of NUSAP1 on malignant phenotypes of glioma cells by silencing NUSAP1. Results: NUSAP1 expression was correlated not only with glioma grade but also with prognosis of glioma patients. NUSAP1 depletion suppressed proliferation of U251 cells by inducing cell cycle arrest at G2/M phase and apoptosis. NUSAP1 depletion rendered U251 cells impaired migratory ability as well. Conclusion: NUSAP1 is a potential prognosis marker for glioma patients and therapeutic strategies targeting NUSAP1 might hold promise in improving glioma treatment. [ABSTRACT FROM AUTHOR]

Lung cancer is the leading cause of cancer death worldwide due to its high incidence and mortality. As the most common lung cancer, non-small cell lung cancer (NSCLC) is a terrible threat to human health. Despite improvements in diagnosis and combined treatments including surgical resection, radiotherapy and chemotherapy, the overall survival for NSCLC patients still remains poor. DNA damage is considered to be the primary cause of lung cancer development and is normally recognized and repaired by the intrinsic DNA damage response machinery. The role of DNA repair pathways in radio(chemo)therapy-resistant cancers has become an area of significant interest in the clinical setting. Meanwhile, some studies have proved that genetic and epigenetic factors can alter the DNA damage response and repair, which results in changes of the radiation and chemotherapy curative effect in NSCLC. In this review, we focus on the effect of genetic polymorphisms and epigenetic factors such as miRNA regulation and lncRNA regulation participating in DNA damage repair in response to radio(chemo)therapy in NSCLC. These may provide novel information on the radio(chemo)therapy of NSCLC based on the individual DNA damage response. [ABSTRACT FROM AUTHOR]
