Rodents play an important role in the transmission of zoonotic diseases. This study investigated the prevalence of Leptospira spp. and Bartonella spp. in Mastomys natalensis and its ectoparasites (fleas and mites) in selected villages of Morogoro, Tanzania. Mastomys natalensis were captured live in fallow habitats using Sherman® traps and anesthetized using Halothane. Blood samples were obtained from the retroorbital sinus Ectoparasites were removed from the fur using a hard brush and preserved in 70 % ethanol. Real time–qPCR was used to detect Leptospira spp. and Bartonella spp. from Mastomys natalensis blood and ectoparasites respectively. The study revealed a relatively larger number of males than females captures. Leptospira spp. was demonstrated in one out of 100 Mastomys natalensis. For Bartonella spp., prevalence of (14 %) was recorded in mites with a higher proportion in mites from adult male Mastomys natalensis than females. Upon Sanger sequencing, four positive samples showed a complete sequence of the ITS gene. Indicating that all samples belonged to Uncultured Bartonella. Low prevalence of Leptospira spp. and a high prevalence of Bartonella spp. was observed in Mastomys natalensis. Further exploration of rodent pathogens is recommended to raise awareness of the role of commensal rodents in disease transmission via their ectoparasites. [ABSTRACT FROM AUTHOR]

Abstract Background Leptospira are the parasitic bacterial organisms associated with a broad range of mammalian hosts and are responsible for severe cases of human Leptospirosis. The epidemiology of leptospirosis is complex and dynamic. Multiple serovars have been identified, each adapted to one or more animal hosts. Adaptation is a dynamic process that changes the spatial and temporal distribution of serovars and clinical manifestations in different hosts. Serotyping based on repertoire of surface antigens is an ambiguous and artificial system of classification of leptospiral agents. Molecular typing methods for the identification of pathogenic leptospires up to individual genome species level have been highly sought after since the decipherment of whole genome sequences. Only a few resources exist for microbial genotypic data based on individual techniques such as Multiple Locus Sequence Typing (MLST), but unfortunately no such databases are existent for leptospires. Results We for the first time report development of a robust MLST method for genotyping of Leptospira. Genotyping based on DNA sequence identity of 4 housekeeping genes and 2 candidate genes was analyzed in a set of 120 strains including 41 reference strains representing different geographical areas and from different sources. Of the six selected genes, adk, icdA and secY were significantly more variable whereas the LipL32 and LipL41 coding genes and the rrs2 gene were moderately variable. The phylogenetic tree clustered the isolates according to the genome-based species. Conclusion The main advantages of MLST over other typing methods for leptospires include reproducibility, robustness, consistency and portability. The genetic relatedness of the leptospires can be better studied by the MLST approach and can be used for molecular epidemiological and evolutionary studies and population genetics.

Investigation of home ranges, sex ratio and recruitment of the multimammate rat (Mastomys natalensis) in semi-arid areas of Tanzania was conducted in maize and fallow fields using the capture-mark-release (CMR) technique. The aim of this study was to generate useful data for the management of M. natalensis. The relative home range size of M. natalensis was significantly higher during the wet [544 m2 ± 25 standard error (SE)] than during the dry (447 m2 ± 18 SE) season, in males (521 m2 ± 23 SE) than in females (450 m2 ± 17 SE) and in adults (576 m2 ± 34 SE) than in juveniles (459 m2 ± 16 SE). However, there were no significant differences between habitats. Sex ratio was not significantly different (p = 0.44) between habitats. Recruitment was significantly higher (p = 0.000) in maize fields (mean = 0.43) than in fallow land (mean = 0.32) and differed significantly over time (p < 0.0001) with the highest recruitment recorded from April to July and the lowest from October to December. Management strategies should focus on managing rodents inhabiting maize fields using methods that affect their recruitment in order to reduce the population increase of M. natalensis. The prevalence of mycobacteria in rodents and insectivores are studied in Morogoro, Tanzania, by using direct smear examination (DSE) after acid-fast staining technique. The findings have shown that potentially pathogenic mycobacteria have posed a risk for other animals and humans, especially HIV/AIDS patients, having a weakened immune system.
