1,6-Enynes have recently stimulated enormous attention toward paving the way to unique cascade cyclizations offering complex cyclic motifs from linear substrates. We describe herein a general approach to napthyridinones via the Pd-catalyzed annulation of 1,6-enynes with 2-iodoanilines. This protocol represents a rare carbo-aminative annulative cyclization via the 6-endo-trig mode, subduing the well-documented exo-trig/dig cyclizations. The regioselective aryl palladation of alkyne followed by Heck-type intramolecular coupling before isomerization were key in realizing this cascade.

Surface microtexturing accomplishes a substantial role in enhancing the interfacial functional performance of machine-driven components. For producing good-quality square micropatterns, maskless electrochemical micromachining (EMM) is a viable option. Maskless EMM is employed for producing micropatterns, wherein the workpiece remains close by interaction with the mask tightly enclosed to the coated tool surface, and the electrolysis of job involves in the enclosed cell. The special micropatterning setup is developed to produce the good quality square micropatterns including micropatterning cell, pulsed electrical unit and perpendicular cross flow electrolyte system. The developed flow method on the job surface in this unit produces better good quality micropatterns. A reused textured tool and developed flow method is applied in maskless EMM in an enclosed unit, which eradicates the sludges and fabricate micropatterns with higher depth. This unique method is an efficient approach for improving the dimensional accuracy of micropatterns. The influences of input criteria, viz. voltage, duty ratio and frequency are studied on surface roughness, current efficiency, length overcut, and textured depth using this technique. The experimental outcomes show that the best parametric combination, viz. 30% duty ratio, 5 kHz frequency and 8 V voltage, enhances the micropatterned quality. A study has been approached based on micrographs for attaining the best input criteria combination.

We demonstrated a dual-cyclizative coupling of ynone oxime ethers with acrylamides for the synthesis of isoxazolyl 2-oxindoles. The cascade is triggered by a palladium(ii)-catalyzed ynone oxime ether cyclization, and is completed by a Heck-type coupling.

Quantum leap in the field of C-H functionalization has expediently provided comprehensive solutions to access valuable molecules that previously required convoluted synthetic attempts. Dual C-H functionalization, unsymmetrically, with a single bifunctional reagent is an incisive tactic. Propargyl alcohols (PAs), despite containing reactive C≡C bond, were not however explored as building blocks via oxidative cleavage. Annulations via C-H activation orchestrate versatile and synthetically attractive strategies. In this unprecedented work, we disclose PA as a new bifunctional reagent for unsymmetrical dual C-H functionalization of biphenylamine for regioselectively annulated outcomes. On tuning conditions, the annulation bifurcated towards an unusual dual oxidative cyclization. The methodology accommodates a wide range of PAs and showcases late-stage diversification of some natural products.

We present here a rhodium-catalyzed oxidative three-point double annulation of enaminones with propargylic alcohols via a C-H and a C-N bond activation to access arylnaphthalene-based lignan derivatives. The key step in the reaction is the regioselective insertion of propargylic alcohol into the rhoda-cycle, a result of hydroxyl rhodium coordination. Necessary control experiments and KIE studies were conducted to determine the mechanism.

Given their omnipresence in natural products and pharmaceuticals, isochromenone congeners are one of the most privileged scaffolds to synthetic chemists. Disclosed herein is a dual (ortho/meta) C-H and C-C activation of phenacyl ammonium salts (acylammonium as traceless directing group) toward annulation with propargylic alcohols to accomplish rapid access for novel isochromenones by means of rhodium catalysis from readily available starting materials. This operationally simple protocol features broad substrate scope and wide functional group tolerance. Importantly, the protocol circumvents the need of any stoichiometric metal oxidants and proceeds under aerobic conditions.

A cascade reaction of enynones with enaminones via cyclative coupling to engineer diverse multisubstituted furans and furano-pyrrole bisheterocycles is described and is proposed to involve carbene insertion and C–C migrations to deliver a totally rearranged product.

In the realm of transition-metal catalyzed arene functionalization, rhodium(III) catalysis is considered as exemplary due to its propensity to activate C-H bonds to obtain comprehensive molecular assembly. Herein, we demonstrate a new rhodium(III) catalyzed assembly of polyheterocyclic scaffolds via C-H activation and regioselective annulation of 4-arylbut-3-yn-1-amines with 4-hydroxy-2-alkynoates. Heterocyclization and trans-metalation prior to annulation is the key for initiation of this relay redox-neutral catalytic cascade.

A regio- and stereoselective synthesis of unsymmetrically substituted pentafulvenes is reported via the condensation of readily available ynones and amino crotonates under very mild conditions. The mechanism of this 3 + 2 annulation involved a vinylogous Michael addition followed by an intramolecular enamine aldol condensation. Substrates with o-bromo tether further cyclized to pentannulated hydroquinolines through an isomerization/SNAr in the same reaction pot at the elevated temperature.

2-Aminoindole-3-carboxylates undergo a Lewis-acid-catalyzed decarboxylative annulation with ynals to afford dihydrochromeno-fused δ-carbolines through a 2,3-aza migration, via a spirocyclic intermediate generated from an initial [3 + 2] spirocycloaddition. Brønsted acid interference changes the path from a [3 + 2] to a [4 + 2] addition. 2-Aminoindoles without an ester functional group at C3 underwent a different condensation, followed by hetero-Diels-Alder reaction to generate chromeno-fused α-carbolines.

A regioselective carbonickelation followed by cyclization, an arylative cyclization, of ortho functional diaryl acetylenes is achieved apparently through an electrophile driven alkyne polarization. A series of selectively substituted di aryl isoquinoline, pyridine and indene derivatives are thus accessed from diarylacetylenes with azide, carbonyl and cyanide tethers.

An unprecedented [2+2+1] hydrative annulation of 1,6-enynes with terminal alkynes is achieved using catalytic cationic Rh(I). Thus, a modular assembly of cyclopenta[b]benzofuranols with two consecutive quarternary stereocenters is achieved from readily available alkynes. The reaction is proposed to go through a sequence of 5-membered rhoda-cycle formation, regioselective acetylene insertion, 1,5 H-shift, substrate controlled stereoselective addition of water molecule followed by 1,2-rhodium migration gave contracted rhoda-cycle D and reductive elimination. Necessary control/labelling experiments were conducted to gain insight in to the mechanism.

A general method for highly functionalized 2-pyrones via a base-mediated sequential vinylogous addition and cyclization of γ-phosphonyl/sulfonyl crotonates and ynones are described. An exclusive E-geometry with respect to the newly generated olefin substituent at C3 of pyrone was observed. Imino glyoxalates and glycine imines similarly reacted with ynones to deliver 3-imino pyrones.

The efficiency and selectivity of annulation reactions are often difficult to control in the presence of multiple potential reactive centers, like in the case of allylic sulfur ylides (ASY). Here, we describe a novel base mediated [3+3] benzannulation of ASY and readily available alkynones, which accomplishes the regioselective formation of multisubstituted thioanisoles, highly sought after chemical scaffolds. A new reactivity pattern of ASY has been unearthed, where it acted as both a 3C component and sulfur source in benzannulation. Use of a widely available base, operational simplicity and broad substrate scope are the additional salient features of the conversion.

[4 + 2] oxidative Diels-Alder reaction of readily available alkynols with maleimide is achieved for the rapid access of pthalimide-fused multicyclic compounds. The reaction is proposed to go through a sequence of Sc(OTf)

Stereoselective E-iodosulfonylation of internal alkynes for synthesizing highly defined tetrasubstituted olefins using sodium phenyl sulfinate and iodine is achieved. Most of the alkynes explored are non-symmetrical and the regioselection is in full control. Aryl alkynes with various with-drawing groups like keto, aldehyde, ester, amide, cyano, sulfone, bromo, chloro and iodo on the other end are identified as successful precursors to yield the highly functionalized olefins. Alkynes directly tethered with heteroatoms are also found as suitable substrates.

A formal intramolecular vicinal 1,2-diamination of alkynes is achieved for the synthesis of indole-cyclic urea fused derivatives through a double cyclization process from readily available aminophenyl propargyl alcohols. This sequential triple C-N bond construction event was possible using isocyanate as urea precursor and Ag(I) catalyst as alkyne activating agent. Control experiments reveal that the cyclization, followed by 1,3-allylic amino dehydroxylation, is preceded by urea formation.

Access to furanyl- and pyrrolyl-3-carboxamides from readily available 3-alkyne-1,2-diols and 1-amino-3-alkyn-2-ols using isocyanate as amido surrogate is demonstrated. The approach constitutes a successful unprecedented combination of heteropalladation and isocyanate insertion, a new avenue for novel amide bond constructions. The mechanism likely involves a 6-membered oxaaminopalladacycle as the key intermediate.

Convenient and general Hg(ii)/Pt(iv) catalyzed syntheses of γ-butyrolactams and α,β-unsaturated γ-butyrolactones/lactams are described via intramolecular electrophilic cyclizations of bromoalkynes with tosylamino and hydroxyl tethers. The reaction features the use of wet solvents, the exclusion of any base and additive, mild conditions and practical yields. We also synthesised few chiral lactams through this pathway. Additionally, it is shown that the NHTs group distanced further from the homopropargylic position assists regioselective bromoalkyne hydration to yield useful α-bromoketones. Furthermore, Boc protected bromo homo propargyl amines undergo 6-endo-dig cyclization through Boc oxygen to give bromomethylene substituted oxazinones.

Palladium catalyzed conversion of 1-(2-aminophenyl)-propargyl alcohols to 3-alkynyl quinolines is realized via a cascade that involves aminopalladation, oxidative coupling with alkynes and dehydration. The method is shown to have a broad substrate scope with respect to propargyl alcohols as well as alkynes. Vinyl ketones as coupling partners in the same reaction afforded 3-alkenyl quinolines with equal ease.

A syn-arylative nickelation followed by nucleophilic syn-selective cyclization of o-propargyloxy benzaldehydes is achieved toward the synthesis of chromanol skeletons with alkenyl substitution at C3. The capture of the intermediate vinyl nickel in its cis geometry is done also with a Michael acceptor to synthesize 4-alkylated derivatives. This protocol is equally applicable to o-propargylamino benzaldehydes to access 3,4-disubstituted tetrahydro-hydroquinolines.

An anti-carbonickelative cyclization via reversible alkenylnickel E/Z isomerization of 2-azido phenyl propargyl alcohols with aryl boronic acids is achieved using Ni(acac)2 as the catalyst to access 2,3-diaryl quinolines. It represents a rare example of trapping the vinyl metal intermediate with a nitrogen center, a non-carbon center electrophile.

Phosphorylation and dephosphorylation are the key mechanisms for mycobacterial physiology and play critical roles in mycobacterial survival and in its pathogenesis. Mycobacteria evade host immune mechanism by inhibiting phagosome – lysosome fusion in which mycobacterial protein tyrosine phosphatase A (PtpA;TP) plays an indispensable role. Tyrosine kinase (PtkA;TK) activated by autophosphorylation; phosphorylates TP, which subsequently leads to increase in its phosphatase activity. The phosphorylated TP is secreted in phagosome of macrophage. In the present study, we have shown that the phosphorylation at two sites of TP; Y128 and Y129 are critical for TK-mediated phosphatase activity. The disruption of this interaction between TK and TP inhibits activation of later which further leads to the decrease in intracellular survival of mycobacteria. Furthermore, the proof of concept has been established using benzylbenzofurans and benzofuranamides, which inhibit the growth and intracellular survival of mycobacteria, associate with the functional sites of TP and contend with the TK. This binding was further restated by looking at the anchorage of protein–protein and the protein–inhibitor complexes in the homology-based structure models and by surface plasmon resonance analysis.

Conjugated enynes, enol ethers, and enynones are versatile building blocks that can be elaborated by a wide variety of synthetic transformations. The selective synthesis of such units is a prerequisite for their effective utilization. The synthesis of conjugated 2-phenoxyenynes through a palladium-catalyzed cross-addition of terminal alkynes to phenylethynyl ethers (hydroalkynylation) is now presented. The reaction is highly regio-, stereo-, and chemoselective, and shows excellent tolerance toward functional groups. The addition further features very mild reaction conditions (room temperature) and an inexpensive catalytic system (without a ligand and with a cheaply available Pd catalyst). The thus synthesized enynyl ethers with allylic hydroxy tethers, which survived the reaction, were shown to be ready precursors for valuable 1-en-4-yn-3-ones.

A regio- and stereoselective thiocyanate addition to ynones is achieved using KSCN in AcOH at 70 °C. The reaction is extendable to ynals, ynesulfones, ynoic acids and ynoates. Adducts from ynones were readily transformed into thiazine-2-thione derivatives under slightly modified reaction conditions. In contrast, thiocyanated adducts from ynamides underwent an in situ decyanative amido cyclization towards isothiazolones. None of these events needed any transition metal or catalyst, attaining a high synthetic value.

Addition of arylboronic acids to directing group tethered acetylenes in a regio and stereoselective manner using an inexpensive catalytic system is achieved for the first time to access highly sought after allyl/homoallyl alcohol/amine units. The apparent vinylnickel intermediate was successfully trapped by the Michael electrophiles to get defined tri- and tetra-substituted olefins. An interesting selectivity switch was observed with internal alkynes.

The copper-catalyzed ketenimine formation reaction of 1-(o-acetamidophenyl)propargyl alcohols with various sulfonyl azides is found to undergo a concomitant intramolecular nucleophile attack to generate 1,2-dihydro-2-iminoquinolines after aromatization (via elimination of acetyl and hydroxy groups) and tautomerization. The reaction produces 4-substituted and 3,4-unsubstituted title compounds in moderate to good yields under mild reaction conditions.

A method to prepare 3-substituted-2-formyl indole derivatives from N-Boc oaminoarylketones and ethoxyacetylene through a cascade of reactions in a single operation that included a metal/catalyst free nucleophile triggered 5-exo-dig cyclization and acid mediated 1,3-allyl alcohol isomerization (1,3-AAI) is described. A variety of aryl, vinyl and alkynyl groups are introduced at C3 of indole-2-carboxaldehyde while having a high functional group compatibility. The 3-alkynyl adducts, which are highly valuable substrates for diversity oriented synthesis, are further transformed to useful carboline and carbazole derivatives through novel pathways.

The reaction between readily accessible propargyl acetates and sulfonyl azides in the presence of CuI catalyst yields trans-α,β-unsaturated N-tosylamides via N-sulfonyl ketenimine formation followed by a probable 1,3-OAc migration ([3,3]-sigmatropic rearrangement). The reaction is very general, allowing all kinds of substitution, including alkyl, aryl (electron-donating, -withdrawing, and -neutral), heteroaryl, and vinyl groups, on the C-terminal of acrylamide. Also, the method affords the products at ambient temperature with excellent diastereoselectivity in moderate to good yields.

A remarkably simple method for the synthesis of di- and trisubstituted functionalised oxazoles under metal- and catalyst-free conditions is described. An iterative bromination and debromination of N-acylated amino acid derivatives with NBS as the sole reagent cleanly led to various substituted oxazoles.

A general method for synthesizing 4-imino tetrahydropyridine derivatives is achieved, from readily available β-enaminones and sulfonyl azides, which comprises a sequential copper catalyzed ketenimine formation and its hitherto inaccessible intramolecular hydrovinylation. The products are shown as ready precursors for highly valuable 4-sulfonamidopyridine derivatives via DDQ mediated oxidation.

A general method for isoxazoles from readily available ynones using trimethylsilyl azide as an amino surrogate, likely via an unprecedented hydroazidation of the alkyne and denitrogenative cyclization, is demonstrated. The method neither required any catalyst nor demanded unusual conditions to afford the products with outstanding functional group compatibility.

An efficient route for the synthesis of the title 2-azidomethyl heterocycles (IV) and (II), resp., based on Ag(I)-catalyzed intramolecular cyclization and γ-allylic azidation of readily available propargyl alcohols is developed.

Synthesis of Z-β-aryl-α,β-unsaturated esters from readily available 1-aryl-3-phenoxy propargyl alcohols is achieved via a BF3-mediated syn-selective Meyer–Schuster rearrangement under ambient conditions. The reaction mechanism is postulated to involve an electrophilic borylation of an allene intermediate as the key step to kinetically control the stereoselectivity.

The title reaction proceeds under mild conditions and tolerates a wide range of functional groups in both partners.

Depending on the tether between the substrates′ bromoalkyne and the hydroxy or tosylamino group, respectively, the selective formation of saturated or unsaturated lactams, unsaturated lactones, oxazinones or α-bromoketones is observed.
