Type 2 diabetes is a polygenic disease and characterized by hyperglycemia and insulin resistance, and it is strongly associated with obesity. However, the mechanism by which obesity contributes to onset of type 2 diabetes is not well understood. We generated rat strains with a hyperglycemic quantitative trait locus (QTL) derived from the Otsuka Long-Evans Tokushima Fatty rat and a fa/fa (Lepr–/–) locus derived from the Zucker Fatty rat. Phenotypes for plasma glucose, and insulin levels were measured, and RNA and protein levels were determined using reverse transcription quantitative PCR and Western blot analyses, respectively. Compared with the obese control strain F344-fa (Lepr–/–), plasma glucose levels of the obese F344-fa-nidd6 (Lepr–/– and Nidd6/of) significantly increased, and plasma insulin levels significantly decreased. These phenotypes were not observed in the lean strains, suggesting that the Nidd6/of locus harbors a diabetogenic gene associated with obesity. We measured the expression of 41 genes in the Nidd6/of QTL region of each strain and found that the mRNA expression levels of the two genes significantly differed between the obese strains. The two genes, pleckstrin homology domain-containing, family S member 1 (Plechs1) and peroxiredoxin III (Prdx3), were differentially expressed only in the obese rats, suggesting that these two genes are involved in the mild elevation of blood glucose levels and insulin resistance in obesity.

Ticks transmit viruses responsible for severe emerging and re-emerging infectious diseases, some of which have a significant impact on public health. In Japan, little is known about the distribution of tick-borne viruses. In this study, we collected and tested ticks to investigate the distribution of tick-borne arboviruses in Kyoto, Japan, and isolated the first Thogoto virus (THOV) to our knowledge from Haemaphysalis longicomis in far-eastern Asia. The Japanese isolate was genetically distinct from a cluster of other isolates from Africa, Europe and the Middle East. Various cell lines derived from mammals and ticks were susceptible to the isolate, but it was not pathogenic in mice. These results advance understanding of the distribution and ecology of THOV.

京都市は，毎年，国内外から多くの観光客が訪れる国際観光都市の一つである。活発な人の移動や物流により，国内外で発生している感染症が京都市に持ち込まれると，その感染が京都市内や国内外に急速に拡大する危険性がある。特に，蚊等の節足動物媒介性感染症については，近年の世界的な温暖化に伴う流行域の拡大が懸念されており，日本においても注意を要する感染症の一つとなっている。媒介動物である蚊等の環境中での生息種や，その数，保有する病原微生物等を常に把握しておくことは，公衆衛生上重要である。そこで，本研究では，2013年5月から2014年3月まで，京都市市街地にあるK寺の境内において，夜間活動性の吸血蚊をCDCトラップで，昼間活動性の吸血蚊を人囮法で捕獲し，その季節消長を観察した。Aedes（Ae.）albopictus（Skuse）（53%），Ae. japonicas（Theobald）（0.2%）， Culex（Cx.）pipiens complex（46.1%），Cx. tritaeniorhynchus（Giles）（0.4%），および Armigeres subalbatus（Coquillett）（0.3%）の3属5種，総計1,407個体の蚊が捕獲された。Cx. pipiens complexの鑑別をPCR法で行ったところ，鑑別した蚊の約95％がCx. pipience pallens（Coquillett）であり，約3％がCx. pipiens form molestus（Forskal）であった。捕獲された夜間活動性の吸血蚊のうち，約98％がAe. albopictus（Skuse）であり，捕獲数のピークは7月であった。一方，昼間活動性の吸血蚊は，主としてCx. pipiens complexとAe. albopictu（s Skuse）であった。それぞれの捕獲数のピークは異なり，各々，6月と7月であった。, Kyoto is one of the historically important and famous international cities for site-seeing. Every year, many visitors come to Kyoto City from all over the world. Once a certain infectious disease is imported into Kyoto City from visitor’s home town in which the outbreak of the disease occur, the disease might be spread rapidly to whole the city and even to other countries. Especially, arthropodborne diseases have been a world public health problem because of on going global warming. In this study, we conducted vector surveillance for mosquito-borne diseases at a temple “K” located at urban area in Kyoto City, from May, 2013 to March, 2014. Mosquitoes were captured using CDC-dry ice trap for night-active mosquitoes, and using human-trap for day-time active ones. We captured 1,407 mosquitoes belonging to three genera including 5 species, Aedes（Ae.）albopictus（Skuse）（53%）, Ae.japonicas（Theobald）（0.2%）, Culex（Cx.）pipiens complex（46.1%）, Cx. tritaeniorhynchus（Giles）（0.4%），and Armigeres subalbatus（Coquillett）（0.3%）. We genetically classified Cx. pipiens complex further into Cx. pipience pallens（Coquillett）（around 95%）and Cx. pipiens form molestus（Forskal）（around 3%）. Ae. albopictus（Skuse）was predominant among mosquitoes captured as day-time active ones. The population peak of Ae. albopictus（Skuse）during research period, was June, 2013. マダニは多くの感染症をヒトや動物に伝播することが知られている。環境中には多くのマダニ種が生息しており，野生動物や家畜，ヒトを吸血し，その時に病原体を伝播する。京都市内でもダニの刺咬被害が報告されていることから，市内でダニが媒介する感染症が発生し流行することが危惧されている。そこで本研究では，京都市山科区で害獣駆除されたイノシシから捕集した68匹のマダニについて，その種の割合を検討した。その結果，Haemaphysalis（H.）longicornis（30.9％），H. flava（27.9％）およびAmblyomma testudinarium（41.2％）の3種類のマダニ種が捕集された。また，イノシシには幼ダニや若ダニに比べ，成ダニが多く寄生していた。この結果は，発育段階が進んだマダニがイノシシに寄生することを示している。, It is well known that ticks transmit many kinds of zoonotic diseases to human. The patient was 85-year-old woman. Five years and 8 months previously, bisphosphonate was prescribed for osteoporosis at a local clinic. Deformity of the thigh occurred when she tried to stand after sitting. X-ray findings revealed a transverse fracture of the right femoral diaphysis. Intramedullary nailing of the right femur was performed. Bone union was slightly delayed, but it was eventually achieved. Bisphosphonates have been reported to severely suppress bone turnover, resulting in the occurrence of fractures at the diaphysis or metaphysis of the femur. This type of fracture is characteristically induced by minor trauma and usually shows a simple transverse fracture. Bone union is delayed and these patients complain of prodromal symptoms. Because our case met all of the above criteria, it was concluded that the cause of her fracture was severely suppressed bone turnover (SSBT). We treated a patient who had a pathologic fracture associated with bisphosphonate therapy. Careful follow-up will be required, because it has been reported that such fractures can also occur on the contralateral side.
