Amphiphilic polycations with quaternary ammonium groups show a strong tendency to porate lipid membranes. The polycations with a high capacity to open pores in biomembranes are the most cytotoxic to cells.

Although the key factor affecting the biocompatibility of IONPs is the core size, there is a lack of regular investigation concerning the impact of the parameter on the toxicity of these nanomaterials. Therefore, such studies were carried out in this paper. Their purpose was to compare the influence of PEG-coated-magnetite NPs with the core of 5, 10 and 30 nm on six carefully selected cell lines. The proliferation rate, viability, metabolic activity, migration activity, ROS levels and cytoskeleton architecture of cells have been evaluated for specified incubation periods. These were 24 and 72-h long incubations with IONPs administered in two doses: 5 and 25 µg Fe/ml. A decrease in viability was observed after exposure to the tested NPs for all the analyzed cell lines. This effect was not connected with core diameter but depended on the exposure time to the nanomaterials. IONPs increased not only the proliferation rate of macrophages—being phagocytic cells—but also, under certain conditions stimulated tumor cell divisions. Most likely, the increase in proliferation rate of macrophages contributed to the changes in the architecture of their cytoskeleton. The growth in the level of ROS in cells had been induced mainly by the smallest NPs. This effect was observed for HEK293T cells and two cancerous lines: U87MG (at both doses tested) and T98G (only for the higher dose). This requires further study concerning both potential toxicity of such IONPs to the kidneys and assessing their therapeutic potential in the treatment of glioblastoma multiforme.

Recombinant human bone morphogenetic protein-2 (rhBMP-2) plays a key role in the stem cell response, not only via its influence on osteogenesis, but also on cellular adhesion, migration, and proliferation. However, when applied clinically, its supra-physiological levels cause many adverse effects. Therefore, there is a need to concomitantly retain the biological activity of BMP-2 and reduce its doses. Currently, the most promising strategies involve site-specific and site-directed immobilization of rhBMP-2. This work investigated the covalent and electrostatic binding of rhBMP-2 to ultrathin-multilayers with chondroitin sulfate (CS) or diazoresin (DR) as the topmost layer. Angle-resolved X-ray photoelectron spectroscopy was used to study the exposed chemical groups. The rhBMP-2 binding efficiency and protein state were studied with time-of-flight secondary ion mass spectrometry. Quartz crystal microbalance, atomic force microscopy, and enzyme-linked immunosorbent assay were used to analyze protein–substrate interactions. The effect of the topmost layer was tested on initial cell adhesion and short-term osteogenesis marker expression. The results show the highest expression of selected osteomarkers in cells cultured on the DR-ended layer, while the cellular flattening was rather poor compared to the CS-ended system. rhBMP-2 adhesion was observed only on negatively charged layers. Cell flattening became more prominent in the presence of the protein, even though the osteogenic gene expression decreased.

(Ce,Gd,Nd,Sm,Pr)O2-δ and (Ce,Gd,La,Nd,Pr)O2-δ rare-earth high-entropy oxides (HEOx) with the oxygen vacancy-ordered Ia-3 structure are doped with a small amount of molybdenum, to prepare two series of materials, (Ce,Gd,Nd,Sm,Pr,Mox)O2-δ and (Ce,Gd,Nd,Sm,Pr,Mox)O2-δ with x = (0, 0.1, 0.2, 0.3 and 0.5). The first-ever stabilization of the fluorite-type Fm-3m structure (i.e. with disordered vacancies) in the ceria-based HEOx is achieved. Structural, microstructural and transport properties of the compounds are systematically studied. The substitution level of x ≥ 0.3 allows to fully stabilize Fm-3m symmetry, with samples exhibiting good homogeneity. Influence of Mo on the oxygen vacancy concentration and charge state of the elements is studied with the use of Raman spectroscopy and XPS methods, showing decrease of the vacancy content and dominating presence of Mo6+, Ce4+ and mixed Pr3+/4+ states. Measured for the first time in this group of materials, the electrical conductivity of the Mo-doped oxides indicates the mixed ionic-electronic behavior.

Hydrophobically modified chondroitin sulfate (CS) is widely used in the preparation of nano-sized drug delivery systems. Herein, the behavior of amphiphilic CSs in aqueous media and the drug accumulation inside the formed micelle-like structures were studied using experimental methods and molecular dynamics simulations. In particular, we focused on the impact of the degree of substitution (DS) with hydrophobic groups and the presence of drug on the morphology of the nanostructures and their molecular organization. Our results show that with increasing DS, the morphology of the amphiphilic CS nanostructures changes from irregular, loosely packed nanogels to cylindrical micelles with a core-shell architecture. These structures can efficiently accumulate hydrophobic drugs. However, the drug molecules preferentially locate at the interface between the hydrophobic part and the hydrophilic corona formed by the CS chains. Our work provides detailed information that may be relevant to the development of amphiphilic polysaccharide-based drug delivery systems.

The formation of poly[(3-allylamino-2-hydroxypropyl)trimethylammonium chloride] (PAH-GTMAC, a strong polycation) monolayers on various substrates was thoroughly studied. Molecular dynamics simulations and experimental techniques (dynamic light scattering, electrophoresis, streaming potential measurements, and quartz crystal microbalance (QCM-D)) were applied to characterize the system. The density, cross-sectional area, end-to-end length, diffusion coefficient, number of uncompensated charges of the PAH-GTMAC molecules were theoretically evaluated. The QCM-D measurements allowed to determine the PAH-GTMAC adsorption kinetics on these substrates interpreted in terms of a hybrid random sequential adsorption model. The dynamic hydration of the PAH-GTMAC monolayer on these substrates was evaluated for the first time. The intermetallic was melted in an electric arc furnace. Its phase composition was confirmed using X-ray diffraction, light microscopy, and differential scanning calorimetry. Then Al2Cu corrosion kinetics was studied. Chemical composition of the solution after corrosion was determined using inductively coupled plasma-optical emission spectroscopy. The surface of corroded specimens was analyzed using scanning electron microscopy and X-ray photoelectron spectroscopy. Subsequent electrochemical studies involved determination of open-circuit potential, electrochemical impedance spectra, and polarization curves. It was found that the Al2Cu phase corrodes selectively and vanadium isopolyoxoanions increase this process both at pH = 1.3 and 2.5 with two exceptions. Corrosion inhibition was observed for 100 and 200 mM of Na3VO4 at pH 1.3, with inhibition efficiency 78% and 62% respectively, due to precipitation of V2O5.

Adsorption kinetics of human serum albumin (HSA) at a gold substrate was studied using the quartz microbalance (QCM), atomic force microscopy (AFM) and X-ray photoelectron spectroscopy (XPS). Measurements were performed at pH 3.5 for various bulk suspension concentrations and ionic strengths. The QCM experimental data were compared with the dry coverage of HSA derived from AFM and from the solution of the mass transfer equation acquired using a hybrid random sequential adsorption model. In this way, the water factor and the dynamic hydration function for HSA monolayers were quantitatively evaluated as a function of dry coverage for various ionic strengths. A comparison of these results with previously known for a silica sensor confirmed that the QCM kinetic measurements are sensitive to the roughness of the sensor characterized in terms of the rms parameter. For the more rough gold sensor (rms = 2.5 nm, average surface feature size 6 nm) the QCM mass transfer rate constant was 2.6 times lower than for the silica sensor characterized by rms = 0.86 nm. This gives for the gold sensor the apparent water factor equal to one and zero hydration function. Moreover, the hydration function increased for larger HSA coverage and was dependent on ionic strength in contrast to the silica substrate. This unexpected behavior was interpreted in terms of the buoyancy effect where the HSA molecules adsorbing in cavities existing at rough surfaces replace the stagnant (hydrodynamically bound) water. Hence, these results confirm that the sensor roughness of the size comparable with protein molecule dimensions exerts a decisive influence on their adsorption kinetic derived from QCM measurements.

Interaction of polycations with lipid membranes is a very important issue in many biological and medical applications such as gene delivery or antibacterial usage. In this work, we address the influence of hydrophobic substitution of strong polycations containing quaternary ammonium groups on the polymer-zwitterionic membrane interactions. In particular, we focus on the polymer tendency to adsorb on or/and incorporate into the membrane. We used complementary experimental and computational methods to enhance our understanding of the mechanism of the polycation-membrane interactions. Polycation adsorption on liposomes was assessed using dynamic light scattering (DLS) and zeta potential measurements. The ability of the polymers to form hydrophilic pores in the membrane was evaluated using a calcein-release method. The polymer-membrane interaction at the molecular scale was explored by performing atomistic molecular dynamics (MD) simulations. Our results show that the length of the alkyl side groups plays an essential role in the polycation adhesion on the zwitterionic surface, while the degree of substitution affects the polycation ability to incorporate into the membrane. Both the experimental and computational results show that the membrane permeability can be dramatically affected by the amount of alkyl side groups attached to the polycation main chain.

International audience; We show that the properties of superparamagnetic iron oxide nanoparticles suspended in liquids can be effectively studied using Magnetic Circular Dichroism in Resonant Inelastic X-ray Scattering. Analysis of the spectral shape and magnetic contrast produced by this experiment enables an assessment of the site distribution and magnetic state of metal ions in the spinel phase. The selective magnetization profile of particles as derived from the field dependence of dichroism empowers an estimation of particle size distribution. Furthermore, the new proposed methodology discriminates sizes that are below the detection limits of X-ray and light scattering probes and that are difficult to spot in TEM.

There is a wide spectrum of malignant diseases that are connected with the clonal proliferation of plasma cells, which cause the production of complete immunoglobulins or their fragments (heavy or light immunoglobulin chains). These proteins may accumulate in tissues, leading to end organ damage. The quantitative determination of immunoglobulin free light chains (FLCs) is considered to be the gold standard in the detection and treatment of multiple myeloma (MM) and amyloid light-chain (AL) amyloidosis. In this study, a silver nanoparticle-based diagnostic tool for the quantitation of FLCs is presented. The optimal test conditions were achieved when a metal nanoparticle (MNP) was covered with 10 particles of an antibody and conjugated by 5&ndash, 50 protein antigen particles (FLCs). The formation of the second antigen protein corona was accompanied by noticeable changes in the surface plasmon resonance spectra of the silver nanoparticles (AgNPs), which coincided with an increase of the hydrodynamic diameter and increase in the zeta potential, as demonstrated by dynamic light scattering (DLS). A decrease of repulsion forces and the formation of antigen&ndash, antibody bridges resulted in the agglutination of AgNPs, as demonstrated by transmission electron microscopy and the direct formation of AgNP aggregates. Antigen-conjugated AgNPs clusters were also found by direct observation using green laser light scattering. The parameters of the specific immunochemical aggregation process consistent with the sizes of AgNPs and the protein particles that coat them were confirmed by four physical methods, yielding complementary data concerning a clinically useful AgNPs aggregation test.

Surface potential of biomaterials is a key factor regulating cell responses, driving their adhesion and signaling in tissue regeneration. In this study we compared the surface and zeta potential of smooth and porous electrospun polycaprolactone (PCL) fibers, as well as PCL films, to evaluate their significance in bone regeneration. The ’ surface potential of the fibers was controlled by applying positive and negative voltage polarities during the electrospinning. The surface properties of the different PCL fibers and films were measured using X-ray photoelectron spectroscopy (XPS) and Kelvin probe force microscopy (KPFM), and the zeta potential was measured using the electrokinetic technique. The effect of surface potential on the morphology of bone cells was examined using advanced microcopy, including 3D reconstruction based on a scanning electron microscope with a focused ion beam (FIB-SEM). Initial cell adhesion and collagen formation were studied using fluorescence microscopy and Sirius Red assay respectively, while calcium mineralization was confirmed with energy-dispersive x-ray (EDX) and Alzarin Red staining. These studies revealed that cell adhesion is driven by both the surface potential and morphology of PCL fibers. Furthermore, the ability to tune the surface potential of electrospun PCL scaffolds provides an essential electrostatic handle to enhance cell-material interaction and cellular activity, leading to controllable morphological changes.

Surface-initiated atom transfer radical polymerization (SI-ATRP) was used for grafting of model brushes (PNIPAM and PMMA) from various planar surfaces: indium tin oxide (ITO), glass and silicon oxide. All polymerizations for a given monomer were conducted at the same conditions in the presence of a sacrificial initiator. Both PNIPAM and PMMA brushes were found to be thicker when grafted from ITO with respect to the other surfaces demonstrating a significant influence of the ITO substrate on SI-ATRP kinetics. It was shown that ITO significantly increases the polymerization rate of the grafted chains due to not negligible desorption of metal ions which may undergo redox reactions also with the copper catalyst. Moreover, the molar masses of the grafted PNIPAM and PMMA chains were found generally larger (3–6 times for PNIPAM) than the ones of free polymers generated in solution, regardless of the substrate type, indicating significant differences in the rates of SI-ATRP and ATRP in the solution.

Poly(N-isopropylacrylamide) (PNIPAM) brushes were grafted from ZnO nanostructured layers with nanorod and hexagonal grains morphologies via Atom Transfer Radical Polymerization in various methanol/water mixtures. The brushes’ growth was followed using Quartz Crystal Microbalance and the obtained hybrid systems were characterized microscopically and using XPS profiling. The studied nanostructured ZnO has high potential for e.g., photovoltaic, sensor applications that may be enhanced in nanocomposites with spatial and compositional control on the organic/inorganic interface. Such systems were realized by grafting polymer brushes from both the convex tops of nanorods and concave areas between them. It was revealed that the growth of PNIPAM brushes on such nanostructured substrates may be controlled by varying the solvent composition. The polymerization performed at higher methanol content (xMe ≈ 0.31) led to decoration of the nanorods only with thin polymer layers around them while for the lower content the polymerization proceeded much faster leading to formation of the thick coating covering their tops. Importantly, such a layer did not block the brushes growth in the confined intercolumnar space. Thus, fine tuning of the solvent mixture should lead to desired relations between the mass/thickness of polymer brushes grafted from convex and concave areas that are crucial for applications of such nanocomposites.

Scattering-mode Scanning Near-Field Optical Microscopy (sSNOM) allows one to obtain absorption spectra in the mid-IR region for samples as small as 20 nm in size. This configuration has made it possible to measure FTIR spectra of the protein complement of membranes. (Amenabar 2013) We now show that mid-IR sSNOM has the sensitivity required to measure spectra of phospholipids in individual bilayers in the spectral range 800 cm-1–1400 cm-1. We have observed the main absorption bands of the dipalmitoylphosphatidylcholine headgroups in this spectral region above noise level. We have also mapped the phosphate absorption band at 1070 cm-1 simultaneously with the AFM topography. We have shown that we could achieve sufficient contrast to discriminate between single and multiple phospholipid bilayers and other structures, such as liposomes. This work opens the way to further research that uses nano-IR spectroscopy to describe the biochemistry of cell membranes and model systems.

Oral contraceptive (OC) use is associated with increased intrarenal renin-angiotensin-aldosterone system (RAA System) activity and risk of nephropathy, though the contribution of progestins contained in the OC in the regulation of angiotensin-dependent control of the renal circulation has not been elucidated. A total of 18 OC users (8 non-diabetic, 10 Type 1 diabetic) were studied in high salt balance, a state of maximal RAA System suppression. Progestational and androgenic activity of the progestin in each OC was standardized to that of the reference progestin norethindrone. Renal plasma flow (RPF) was measured by para-aminohippurate clearance at baseline and in response to angiotensin-converting enzyme (ACE) inhibition. There was a positive correlation between OC progestational activity and the RPF response to ACE inhibition (r=0.52, P=0.03). Similar results were noted with OC androgenic activity (r=0.54, P=0.02). On subgroup analysis, only non-diabetic subjects showed an association between progestational activity and angiotensin-dependent control of the renal circulation (r=0.71, P=0.05 non-diabetic; r=0.14, P=0.7 diabetic; P=0.07 between groups). Similar results were noted with respect to androgenic activity (r=0.88, P=0.005 non-diabetic; r=-0.33, P=0.3 diabetic; P=0.002 between groups). Our results suggest that the OC progestin component is a significant influence on the degree of angiotensin-dependent control of the renal circulation, though these findings may not apply to women with diabetes.

Acyl-CoA:lysophosphatidylcholine acyltransferase (LPCAT; EC 2.3.1.23) catalyzes the acyl-CoA-dependent acylation of lysophosphatidylcholine (LPC) to produce PC and CoA. LPCAT activity may affect the incorporation of fatty acyl moieties at the sn-2 position of PC where PUFA are formed and may indirectly influence seed TAG composition. LPCAT activity in microsomes prepared from microspore-derived cell suspension cultures of oilseed rape (Brassica napus L. cv Jet Neuf) was assayed using [1-14C]acyl-CoA as the fatty acyl donor. LPCAT activity was optimal at neutral pH and 35 degrees C, and was inhibited by 50% at a BSA concentration of 3 mg mL(-1). At acyl-CoA concentrations above 20 microM, LPCAT activity was more specific for oleoyl (18:1)-CoA than stearoyl (18:0)- and palmitoyl (16:0)-CoA. Lauroyl (12:0)-CoA, however, was not an effective acyl donor. LPC species containing 12:0, 16:0, 18:0, or 18:1 as the fatty acyl moiety all served as effective acyl acceptors for LPCAT, although 12:0-LPC was somewhat less effective as a substrate at lower concentrations. The failure of LPCAT to catalyze the incorporation of a 12:0 moiety from acyl-CoA into PC is consistent with the tendency of acyltransferases to discriminate against incorporation of this fatty acyl moiety at the sn-2 position of TAG from the seed oil of transgenic B. napus expressing a medium-chain thioesterase.

cDNAs encoding acyl-CoA:diacylglycerol acyltransferase (DGAT, EC 2.3.1.20), designated BnDGAT1 and BnDGAT2, were obtained from a microspore-derived cell suspension culture of oilseed rape (Brassica napus L. cv Jet Neuf). BnDGAT2 shares a very high level of identity with BnDGAT1, but is a smaller protein lacking the relatively hydrophilic N-terminal segment found in BnDGAT1. Both transcripts were produced in the cell suspension cultures and the cDNAs were functionally expressed in transformed yeast (Pichia pastoris) cells. Sucrose-mediated changes in triacylglycerol (TAG) metabolism and expression of BnDGAT1 were examined in the cell suspension cultures following transfer of cells from media containing 6% (w/v) sucrose to media containing 14% sucrose. TAG content and DGAT activity of the cells increased transiently within the first 12 h after transfer (HAT). The rapid decline in TAG content observed at 12 HAT was inversely related to an increase in TAG lipase (EC 3.1.1.3) activity. The transient increases in TAG content and DGAT activity correlated with the elevated amounts of BnDGAT1 polypeptide. Transcript levels were also induced, but levels of mRNA encoding BnDGAT1 were not tightly correlated with DGAT activity and amount of polypeptide suggesting some control of expression at the post-transcriptional level. In general, the rapid changes in TAG content were closely associated with the changes in the activity of TAG-metabolizing enzymes and expression of BnDGAT1.

Uric acid is associated with hypertension and increased renin–angiotensin system activity, although this relationship diminishes after chronic exposure to high levels. Uric acid is more strongly associated with poor outcomes in women compared to men, although whether this is due to a sex‐specific uric acid‐mediated pathophysiology or reflects sex differences in baseline uric acid levels remains unknown. We examined the association between uric acid and vascular measures at baseline and in response to angiotensin‐II challenge in young healthy humans. Fifty‐two subjects (17 men, 35 premenopausal women) were studied in high‐salt balance. Serum uric acid levels were significantly higher in men compared to women (328 ± 14 μmol/L vs. 248 ± 10 μmol/L, P < 0.001), although all values were within normal sex‐specific range. Men demonstrated no association between uric acid and blood pressure, either at baseline or in response to angiotensin‐II. In stark contrast, a significant association was observed between uric acid and blood pressure at baseline (systolic blood pressure, P = 0.005; diastolic blood pressure, P = 0.02) and in response to angiotensin‐II (systolic blood pressure, P = 0.035; diastolic blood pressure, P = 0.056) in women. However, this sex difference lost significance after adjustment for baseline uric acid. When all subjects were stratified according to high (>300 μmol/L) or low (≤300 μmol/L) uric acid levels, only the low uric acid group showed a positive association between uric acid and measures of vascular tone at baseline and in response to angiotensin‐II. Differences in uric acid‐mediated outcomes between men and women likely reflect differences in exposure to increased uric acid levels, rather than a sex‐specific uric acid‐mediated pathophysiology., The lower baseline uric acid levels in women compared to men may account for differences in uric acid‐associated vascular outcomes rather than sex‐mediated differences in pathophysiology.
