Abstract

The Marine and Aquatic Research Experience (MARE) is a student-generated, faculty-guided not mandated, model for hands-on, team-based, environmental science research for undergraduates, of any class rank and any academic major. MARE is unique in many ways compared to traditional undergraduate research models. MARE was envisioned and established in the fall of 1998 as part of two Research-Based Learning (RBL) Critical Connection Courses (CCC) entitled Design of Inquiry in Science and Implementation of Inquiry in Science. The initial goal was to study the dynamic processes occurring in Winyah Bay, South Carolina. As we began to establish research directions, MARE members organized themselves into crews to allow collaboration between members with similar research interests. Currently, there are 28 members in MARE, composing five crews. As students involved in MARE since the beginning, we have witnessed the evolution of the team as a whole. We felt it was time for us to take a step back and critically examine ourselves. We have formulated a list of the benefits and challenges of working in a team doing scientific research as undergraduates. Following our presentation on student-driven team-based research, there was a roundtable discussion focusing on three main issues: how MARE is funded, what the personal benefits of team-based research are, and what are the roles of the individual members. Overall, we came to the conclusion that team-based research is possible and rewarding due to the unique research and learning opportunities it provides students. The rewards of this experience directly reflect the ideals of Critical Connection Courses and Research-Based Learning, illustrating the possibilities of these educational innovations. 5. Use of Progesterone Concentrations in Plasma and Milk in the Diagnosis of Pregnancy in Domestic Cattle

Abstract

The concentrations of progesterone in plasma, whole milk or fat-free milk taken 21 days after AI and at other times have been used to diagnose pregnancy in about 500 Friesian heifers and cows. Those cattle diagnosed not pregnant (15–35%) by this method were in 95–100% of cases so confirmed by subsequent evidence. About 20% of those diagnosed pregnant by the progesterone method later showed oestrus or had no palpable foetus 70 days after AI and it is suggested that much of this difference represents embryo mortality. Diagnosis of pregnancy from a single sample (fat-free milk) by the progesterone method was probably most accurate when the sample was taken either 21 ± 1 days or 24 ± 1 days after AI. Substantially better accuracy was probably obtained when information from samples taken at both these times was combined.

Abstract

Prostate-specific antigen (PSA) is a routinely used marker of prostate cancer; however, the cut-off values for unambiguous positive/negative prostate cancer diagnoses are not defined. Therefore, despite the best effort, certain percentage of misdiagnosed cases is being recorded every year. For this reason, search for more specific diagnostic markers is of great interest. In this study, systematic comparison of PSA and metallothionein (MT) levels in blood serum of 46 prostate cancer-diagnosed patients is presented. It is clearly demonstrated that PSA levels vary significantly and despite normal total PSA values in the range of 0 - 4 ng/mL were obtained in over 36.9% of cases, positive prostate cancer was diagnosed by biopsy. In contrary, MT levels were considerably elevated in all tested samples and no significant variations were observed. These results are indicating the potential of MT as an additional prostate cancer marker reducing, in combination with PSA, the probability of false positive/negative diagnosis. To increase the throughput of the screening, chip-based capillary electrophoresis was suggested as a rapid and effective method for the fingerprinting analysis of prostate cancer from diseased blood sera., (Copyright © 2011 WILEY-VCH Verlag GmbH & Co. Abstract

Tumour markers are substances produced by malignant cells or by the organism as a response to cancer development. Determination of their levels can, therefore, be used to monitor the risk, presence and prognosis of a cancer disease or to monitor the therapeutic response or early detection of residual disease. Time-consuming imaging methods, examination of cerebrospinal fluid or tumour tissue and assays for hormones and tumour markers have been used for cancer diagnosis. However, no specific marker for diagnosis of childhood solid tumours has been discovered yet. In this study, metallothionein (MT) was evaluated as a prospective marker for such diseases. Serum metallothionein levels of patients with childhood solid tumours were determined using differential pulse voltammetry - Brdicka reaction. A more than 5-fold increase in the amount of metallothionein was found in sera of patients suffering from cancer disease, compared with those in sera of healthy donors. The average metallothionein level in the sera of healthy volunteers was 0.5 ± 0.2 μmol · dm⁻³ and was significantly different (p<0.05, determined using the Schefe test) from the average MT level found in serum samples of patients suffering from childhood solid tumours (3.4 ± 0.8 μmol · dm⁻³). Results found in this work indicate that the MT level in blood serum can be considered as a promising marker for diagnostics, prognosis and estimation of therapy efficiency of childhood tumours.
