30. Enhanced Non-Associative Long-Term Potentiation in Immature Granule Cells in the Dentate Gyrus of Adult Rats

Abstract

The dentate gyrus is one of the few sites of neurogenesis in the adult brain. Integration of new-generated granule cells into the hippocampal circuitry provides a substrate for structural plasticity, fundamental for normal function of adult hippocampus. However, mechanisms of synaptic plasticity that mediate integration of new-generated granule cells into the existing circuitry remain poorly understood. Especially mechanisms of plasticity at GABA-ergic synapses remain elusive. Here, we show that postsynaptic spiking without presynaptic activation can induce heterosynaptic, non-associative plasticity at GABA-ergic inputs to both immature and mature granule cells. In both immature and mature neurons, plastic changes were bidirectional and individual inputs could express long-term potentiation (LTP) or long-term depression (LTD), or do not change. However, properties of non-associative plasticity dramatically change with maturation of newly generated granule cells: while in immature cells there was a clear predominance of non-associative LTP and net potentiation across the inputs, in mature neurons, potentiation and depression were balanced with no net change on average. We conclude that GABA-ergic inputs to granule cells are plastic, and that the rules for induction of non-associative plasticity change with maturation. We propose that potentiation-biased non-associative plasticity of GABA-ergic transmission might help to counter-balance an increase of excitatory drive that is facilitated by enhanced LTP at glutamatergic synapses in maturating granule cells. Such mechanism might help to build a strong GABA-ergic input to surviving active new cells, necessary for normal function of mature granule cells, which operate under a tight inhibitory control and generate sparse spiking activity.

Abstract

Background The recently developed genetically encoded calcium indicator (GECI), called NTnC, has a novel design with reduced size due to utilization of the troponin C (TnC) as a Ca²⁺-binding moiety inserted into the mNeonGreen fluorescent protein. NTnC binds two times less Ca²⁺ ions while maintaining a higher fluorescence brightness at the basal level of Ca²⁺ in neurons as compared with the calmodulin-based GECIs, such as GCaMPs. In spite of NTnC’s high brightness, pH-stability, and high sensitivity to single action potentials, it has a limited fluorescence contrast (F-Ca²⁺/F⁺Ca²⁺) and slow Ca²⁺ dissociation kinetics. Results Herein, we developed a new NTnC-like GECI with enhanced fluorescence contrast and kinetics by replacing the mNeonGreen fluorescent subunit of the NTnC indicator with EYFP. Similar to NTnC, the developed indicator, named iYTnC2, has an inverted fluorescence response to Ca²⁺ (i.e. becoming dimmer with an increase of Ca²⁺ concentration). In the presence of Mg²⁺ ions, iYTnC2 demonstrated a 2.8-fold improved fluorescence contrast in vitro as compared with NTnC. The iYTnC2 indicator has lower brightness and pH-stability, but similar photostability as compared with NTnC in vitro. Stopped-flow fluorimetry studies revealed that iYTnC2 has 5-fold faster Ca²⁺ dissociation kinetics than NTnC. When compared with GCaMP6f GECI, iYTnC2 has up to 5.6-fold faster Ca²⁺ association kinetics and 1.7-fold slower dissociation kinetics. During calcium transients in cultured mammalian cells, iYTnC2 demonstrated a 2.7-fold higher fluorescence contrast as compared with that for the NTnC. iYTnC2 demonstrated a 4-fold larger response to Ca²⁺ transients in neuronal cultures than responses of NTnC. iYTnC2 response in neurons was additionally characterized using whole-cell patch clamp. Abstract

In terrestrial gastropod snail Helix lucorum L. we studied the changes after a 16-day exposure to microgravity in: behavior, neural responses to adequate motion stimulation, intersensory interactions between the photosensory pathways and the statocyst receptors, and in expression of the HPeP gene in the statocyst receptors. In behavioral experiments it was found that the latency of body position change to sudden orientation change (flip from horizontal to downwards position) was significantly reduced in the postflight snails. Extracellularly recorded neural responses of the statocyst nerve to adequate motion stimulation in the postflight snails were independent of the motion direction while in the control animals an orientation selectivity was observed. Significant differences in the HPeP gene mRNA expression pattern in the statocyst receptor neurons were observed in postflight (30h) and control snails. Obtained results confirm the possibility to elucidate the influence of microgravity exposure on mechanisms and function of gravireceptors using this simple model animal.
