The genetic diversity assessment of agricultural crops is crucial for breeding programs aimed at enhancing crop yield, resistance to diseases, and adaptation to changing environmental conditions. In the present investigation, a comparative genetic relationship in sixteen onion genotypes was assessed utilizing dominant (RAPD) and co-dominant (SSR) marker systems. Ten RAPD and nine SSR markers showed genetic diversity remarkably and produced 503 and 107 amplicons respectively. Spearman rank correlation was used to compare the different efficiency parameters in two marker systems with respect to sixteen onion genotypes. The genetic relationship based on similarity matrix values between a pair of cultivars was higher for SSR markers than for the RAPD marker system. OPC-04 (RAPD primer) and ACM-004 (SSR primer) witnessed the highest poly-morphic bands along with other polymorphic markers that proved to be useful in grouping onion genotypes. Finally, dendrograms were constructed and compared following the mantel test to find out the genetic diversity among the germplasms. An Elite Spring Wheat Yield Trial (EWYT) was conducted with 50 diverse genotypes of Wheat (Triticum aestivum L.) and analyzed by multivariate analysis during 2018-19 rabi season. Correlation analysis revealed positive association of plant height, grains per spike, spike length, test weight and biomass with grain yield. Path analysis revealed the highest direct effect on grain yield/metre by biomass/metre (1.508) which was also positively associated with grain yield/metre (0.833). PCA analysis showed 11 principal components out of which five principal components (PC1 to PC5) exhibited eigen value greater than one. The correlation between AUDPC and physiological characters indicated that chlorophyll loss was associated with high spot blotch disease incidence at later stages of crop growth. This study investigates the response of lentil genotypes to Stemphylium blight disease over two cropping years, 2021 and 2022. In addition, this research aims to assess the marker-trait association between Stemphylium disease progress and the selected SSRs. Visual symptoms were assessed using Hashemi's disease scoring system, leading to the calculation of disease parameters such as Percent Disease Index (PDI) and Area Under Disease Progress Curve (AUDPC). The results revealed variations in disease progress among genotypes, with some exhibiting low disease indices, particularly genotype IC241072. Genetic diversity analysis using SSR markers identified 30 alleles and grouped genotypes into four clusters. Principal Coordinate Analysis (PCoA) confirmed the genetic distances among genotypes. Marker-trait association analysis linked the microsatellite locus LCSSR04_130362_118bp to Stemphylium blight resistance, supported by the identification of a TCP family transcription factor in close proximity. The validation of the association indicated a significant difference between resistant and susceptible alleles. This comprehensive approach combining field screening, genetic diversity analysis, and marker-trait associations provides valuable insights into the genetic basis of Stemphylium blight resistance in lentil genotypes, offering potential candidates for future breeding programs. Grass pea has the potential to become a miracle crop if the stigma attached to it as a toxic plant is ignored. In light of the following, we conducted transcriptome analyses on the high and low ODAP-containing cultivars i.e., Nirmal and Bidhan respectively in both normal and salt stress conditions. In this study, genes that work upstream and downstream to β-ODAP have been found. Among these genes, AAO3 and ACL5 were related to ABA and polyamine biosynthesis, showing the relevance of ABA and polyamines in boosting the β-ODAP content in Nirmal. Elevated β-ODAP levels in salt stress-treated Bidhan may have evolved tolerance by positively regulating the expression of genes involved in phenylpropanoid and jasmonic acid biosynthesis. Although the concentration of β-ODAP in Bidhan increased under salt stress, it was lower than in stress-treated Nirmal. Despite this, the expression of stress-related genes that work downstream to β-ODAP was found higher in stress-treated Bidhan. This could be because stress-treated Nirmal has lower GSH, proline, and higher H 2 O 2 , resulting in the development of severe oxidative stress. Overall, our research not only identified new genes linked with β-ODAP, but also revealed the molecular mechanism by which a low β-ODAP-containing cultivar developed tolerance against salinity stress. • Bidhan is a low-β-ODAP cultivar having salinity stress tolerance. • β-ODAP production is largely regulated by genes involved in ABA and polyamine biosynthesis. • Elevated β-ODAP in stress-treated Bidhan develops tolerance by positive regulation of phenylpropanoid and jasmonic acid biosynthesis related genes. • Elevated level of GSH and proline provides salinity stress tolerance to Bidhan.
