Hipofīzes neiroendokrīnais audzējs (PitNET) ir labdabīgs smadzeņu audzējs, kas ir viens no visbiežāk sastopamajiem audzējiem smadzeņu dobumā. Molekulārie un šūnu mehānismi, kas veicina PitNET patoģenēzi, vēl nav labi izprasti. Somatostatīna receptoru analogi (SSA) tiek izmantoti audzēju ārstēšanai, tomēr daļai pacientu ir novērota rezistence pret SSA, kuru būtiski ietekmē dažādu gēnu ekspresija. Šajā darbā PitNET šūnu līnijās tika validēti un funkcionāli noraksturoti PitNET transkriptomu sekvencēšanā atklātie G-proteīna signalizācijas regulatora 4 (RGS4) un miR-625-5p kandidātfaktori un novērtēta SSA transkriptomiskā un funkcionālā ietekme uz somatotrofo GH3 šūnu līniju. Iegūtie rezultāti parādīja, ka RGS4 kā GTPāzi aktivizējošā proteīna inhibēšana pastiprinā SSA ietekmi uz PitNET šūnu proliferāciju un apoptozi modulējot cAMF un MAPK signālpārneses ceļus. Savukārt miR-625-5p izslēgšana GH3 šūnās palielina SSA antiproliferatīvo darbību. GH3 šūnu līnijas transkiptomā netika novērotas nozīmīgas izmaiņas pēc SSA stimulācijas, kas būtu pielīdzināmas no PitNET audiem iegūto sfēru transkriptoma izmaiņām, liecinot par to, ka GH3 šūnās pilnībā neatspoguļo PitNET sfērās notiekošos procesus., Pituitary neuroendocrine tumor (PitNET) noncancerous brain tumor, that is one of the most common tumors in the brain cavity. The molecular and cellular mechanisms that contribute to the pathogenesis of PitNET are not yet well understood. Somatostatin receptor analogues (SSAs) are used to treat tumors, but resistance to SSA has been reported in some patients and is significantly affected by the expression of various genes. In this study, candidate proteins of the G-protein signaling regulator 4 (RGS4) and miR-625-5p detected by PitNET transcriptome sequencing were validated and functionally characterized in PitNET cell lines and the transcriptomic and functional effects of SSA on the somatotrophic GH3 cell line were evaluated. The results showed that inhibition of RGS4 as a GTPase activating protein potentiates the effect of SSA on PitNET cell proliferation and apoptosis by modulating cAMP and MAPK signaling pathways and knockdown of miR-625-5p in GH3 cells increases the antiproliferative activity of SSA. No significant changes were observed in the transcriptome of the GH3 cell line after SSA stimulation comparable to changes in the transcriptome of the PitNET-derived spheres, suggesting that GH3 cells do not fully reflect the processes in the PitNET spheres.

2. tipa cukura diabēts ir metabolo traucējumu grupa, kam raksturīga hiperglikēmija un insulīna rezistence. Maģistra darba mērķis bija izveidot diabēta pacientiem specifiskas cilmes šūnu līnijas no taukaudu paraugiem. No taukaudiem iegūtās cilmes šūnas tika diferencētas par adipocītiem, kuriem tika ierosināta insulīna rezistence. Tika noteikta to gēnu ekspresija, kas iesaistīti glikozes homeostāzes, transporta un iekaisumu procesos - normālos un insulīna rezistences apstākļos un novērtēta arī pioglitazona iedarbība uz in vitro izraisīto insulīna rezistenci pacientiem specifiskajās šūnu līnijās. Darba rezultāta iegūtas pacientiem specifiskās ADSC šūnu kultūras, kas, diferencējot, uzrāda adipocītiem raksturīgo gēnu ekspresijas izmaiņas insulīna rezistences un pioglitazona darbības rezultātā., Type 2 diabetes is a group of metabolic disorders that are characterized by hyperglycemia and insulin resistance. Adipose tissue derived stem cells were differentiated into adipocytes and then insulin resistance was induced. Gene expression was determined for the genes play role in glucose homeostasis and transport, and inflammatory processes both in normal and insulin resistance conditions. Effect of pioglitazone was assessed in in vitro induced insulin resistant patient specific cell lines. As a result patient specific ADSC cell lines were obtained showed gene expression characteristic for adipocytes under insulin resistance conditions and pioglitazone.

Aptaukošanās un ar to saistītās slimības pēdējā laikā ir pieaugoša problēma Rietumvalstīs, tādēļ plaši tiek pētīti procesi taukaudos. Darba mērķis bija noskaidrot hidroksikarbonskābes (HCA) receptoru ligandu ietekmi uz adipoģenēzi un iekaisuma procesiem, kā arī to ietekmi uz HCA receptoru gēnu ekspresiju cilvēka adipocītos un makrofāgos. Rezultāti liecina, ka neviens no HCA1-3 receptoru ligandiem neietekmē ne adipoģenēzei raksturīgo transkripcijas faktoru, ne pašu HCA receptoru gēnu ekspresiju SGBS adipocītos un diferencētās adipozo audu cilmes šūnās. Savukārt HCA2 un HCA3 receptoru ligandi samazina LPS paaugstināto iekaisuma citokīnu gēnu ekspresiju SGBS adipocītos un THP-1 makrofāgos., Obesity is an increasing problem in Western world and it is associated with many health problems, therefore processes in the adipose tissue are widely investigated. The aim of the present work was to determine the effect of hydroxycarboxylic acid receptor (HCA) ligands on adipogenesis and inflammatory responses as well as their effect on HCA receptor gene expression in human adipocytes and macrophages. The results show that none of the HCA1-3 receptor ligands has an effect on the gene expression of adipogenic transcription factors or the gene expression of HCA receptors in SGBS adipocytes and differentiated adipose tissue stem cells. HCA2 and HCA3 receptor ligands decrease LPS stimulated proinflammatory cytokines gene expression in SGBS adipocytes and THP-1 macrophages.
