Abstract

Background and aimType I interferons (IFNs) are highly expressed in the gut mucosa of celiac disease (CD) gut mucosa and stimulates immune response prompted by gluten ingestion, but the processes that maintain the production of these inflammatory molecules are not well understood. Adenosine deaminase acting on RNA 1 (ADAR1), an RNA-editing enzyme, plays a crucial role in inhibiting self or viral RNAs from activating auto-immune mediated responses, most notably within the type-I IFN production pathway. The aim of this study was to assess whether ADAR1 could contribute to the induction and/or progression of gut inflammation in patients with celiac disease.Material and methodsADAR1 expression was assessed by Real time PCR and Western blotting in duodenal biopsy taken from inactive and active celiac disease (CD) patients and normal controls (CTR). To analyze the role of ADAR1 in inflamed CD mucosa, lamina propria mononuclear cells (LPMC) were isolated from inactive CD and ADAR1 was silenced in with a specific antisense oligonucleotide (AS) and then incubated with a synthetic analogue of viral dsRNA (poly I:C). IFN-inducing pathways (IRF3, IRF7) in these cells were evaluated with Western blotting and inflammatory cytokines were evaluated with flow cytometry. Lastly, the role of ADAR1 was investigated in a mouse model of poly I:C-driven small intestine atrophy.ResultsReduced ADAR1 expression was seen in duodenal biopsies compared to inactive CD and normal controls. Ex vivo organ cultures of duodenal mucosal biopsies, taken from inactive CD patients, stimulated with a peptic-tryptic digest of gliadin displayed a decreased expression of ADAR1. ADAR1 silencing in LPMC stimulated with a synthetic analogue of viral dsRNA strongly boosted the activation of IRF3 and IRF7 and the production of type-I IFN, TNF-α and IFN-γ. Administration of ADAR1 antisense but not sense oligonucleotide to mice with poly I:C-induced intestinal atrophy, significantly increased gut damage and inflammatory cytokines production.ConclusionsThese data show that ADAR1 is an important regulator of intestinal immune homeostasis and demonstrate that defective ADAR1 expression could provide to amplifying pathogenic responses in CD intestinal mucosa.

Abstract

A fundamental study is made of a rotating boundary layer (BL) in the inboard region of a tilt-rotor blade, using an embedded laser Doppler velocimetry probe inside a rotating blade. Velocity measurements have been performed to get insight into the BL behavior around a blade section close to the rotor hub. BL transition and separation phenomena are investigated on the basis of both chordwise and spanwise velocity profiles measured for different collective pitch angles on a two-bladed hovering rotor. The velocity experimental data bank appears to be in good agreement with theoretical velocity profiles in attached flow conditions (laminar, transitional, or turbulent). Moreover, validation of a transitional criterion based on the second thickness of energy [[delta].sub.3] of the BL is extended to the rotational flow configuration. Usually such a configuration classically exhibits a separation of the BL for high collective pitch angles. However, in the particular case of a high twisted tilt-rotor blade operating in hover, centrifugal effects are shown to be responsible for an important delay in the BL separation process.

Abstract

An analysis of propeller/wing aerodynamic interaction at low subsonic speed indicates that tilt angle parameter and flow configuration have a significant influence on wake distortions due to half-wing. The propeller/nacelle/half-wing configuration under distinct wind-tunnel flow configuration activates different conditions of flight. The existence of half-wing alters propeller wake geometry. The close vortex wing surface interaction generates a low-pressure peak on the wing's upper side. Experimental procedure for the analysis is discussed.

Abstract

The embedded laser velocimetry technique gives two-dimensional velocity measurements for boundary-layer characterization around curved walls and airfoils oscillating in pitching motion. A decrease in the frequency parameter, k strongly influences the cyclic separation or reattachment process. An increase in the k value produces a time delay on the transition, separation and reattachment processes.

Abstract

Carcinomas of the left colon represent a neoplasm of older patients (late onset), but epidemiologic evidence has been showing an increasing incidence in patients 50 years or younger (early onset). In this study, we investigate pathologic and molecular features of early- and late-onset carcinoma of the left colon.We selected 22 patients 50 years or younger and 21 patients 70 years or older with left-sided colorectal carcinoma (CRC). All samples were evaluated for pathologic features, microsatellite instability, and KRAS and BRAF mutations. Moreover, both groups were analyzed to identify CpG island methylator phenotype features and assessed with restriction landmark genome scanning (RLGS) to unveil differential DNA methylation patterns.Early-onset patients had advanced pathologic stages compared with late-onset patients (P = .0482). All cases showed a microsatellite stable profile and BRAF wild-type sequence. Early-onset patients (43%) more frequently had mutations at KRAS codon 12 compared with late-onset patients (14%) (P =.0413). RLGS showed that patients younger than 50 years who had CRC had a significantly lower percentage of methylated loci than did patients 70 years or older (P = .04124), and differential methylation of several genomic loci was observed in the two groups.Our results suggest that left-sided CRCs may present differential patterns of aberrant DNA methylation when they are separated by age.

Abstract

The commercial interest in functional foods that contain live microorganisms, also named probiotics, is paralleled by the increasing scientific attention to their functionality in the digestive tract. This is especially true of yogurts that contain strains of lactic-acid bacteria of intestinal origin, among these, Lactobacillus delbrueckii ssp. bulgaricus is extensively used in the dairy industry and it has been demonstrated to be a probiotic strain. In this work we describe high cell density cultivations of this microorganism also focusing on the stereospecific production of lactic acid. Key parameters such as medium composition (bactocasitone concentration) and diverse aeration conditions were explored. The results showed that the final concentration of biomass in anaerobic fermentation was lower than the one obtained in microaerophilic conditions, while it gave a very high productivity of lactic acid which was present as a racemic mixture in the permeate. Fermentation experiments carried out with air sparging, even at very low flow-rate, led to the production of the sole L(+) lactic acid giving sevenfold increase in biomass yield in respect to the batch cultivation. Finally, a mathematical model was developed to describe the microfiltration bioprocess applied in this research considering an inhibition kinetic and enucleating a suitable mathematical description for the decrease of the transmembrane flux.
