Sedimentary ancient DNA can indicate ecosystem-wide changes. Here, the authors show association between warm phases and high diatom abundance in the Antarctic Scotia Sea, in addition to presenting ancient eukaryote sedimentary DNA spanning the last approximately 1 million years.

A new high-resolution record for the Antarctic Zone shows persistent anti-phasing of high interglacial ocean productivity and high glacial dust deposition, suggesting a close inter-hemispheric coupling of cryosphere, ocean and atmosphere.

Human and bovine respiratory syncytial virus (HRSV and BRSV) are closely genetically related and cause respiratory disease in their respective host. Whereas HRSV vaccines are still under development, a multitude of BRSV vaccines are used to reduce clinical signs. To enable the design of vaccination protocols to entirely stop virus circulation, we aimed to investigate the duration, character and efficacy of the immune responses induced by natural infections. The systemic humoral immunity was monitored every two months during two years in 33 dairy cattle in different age cohorts following a natural BRSV outbreak, and again in selected individuals before and after a second outbreak, four years later. Local humoral and systemic cellular responses were also monitored, although less extensively. Based on clinical observations and economic losses linked to decreased milk production, the outbreaks were classified as moderate. Following the first outbreak, most but not all animals developed neutralising antibody responses, BRSV-specific IgG1, IgG2 and HRSV F- and HRSV N-reactive responses that lasted at least two years, and in some cases at least four years. In contrast, no systemic T cell responses were detected and only weak IgA responses were detected in some animals. Seronegative sentinels remained negative, inferring that no new infections occurred between the outbreaks. During the second outbreak, reinfections with clinical signs and virus shedding occurred, but the signs were milder, and the virus shedding was significantly lower than in naïve animals. Whereas the primary infection induced similar antibody titres against the prefusion and the post fusion form of the BRSV F protein, memory responses were significantly stronger against prefusion F. In conclusion, even if natural infections induce a long-lasting immunity, it would probably be necessary to boost memory responses between outbreaks, to stop the circulation of the virus and limit the potential role of previously infected adult cattle in the chain of BRSV transmission.

The induction of long-lasting clinical and virological protection is needed for a successful vaccination program against the bovine respiratory syncytial virus (BRSV). In this study, calves with BRSV-specific maternally derived antibodies were vaccinated once, either with (i) a BRSV pre-fusion protein (PreF) and MontanideTM ISA61 VG (ISA61, n = 6), (ii) BRSV lacking the SH gene (ΔSHrBRSV, n = 6), (iii) a commercial vaccine (CV, n = 6), or were injected with ISA61 alone (n = 6). All calves were challenged with BRSV 92 days later and were euthanized 13 days post-infection. Based on clinical, pathological, and proteomic data, all vaccines appeared safe. Compared to the controls, PreF induced the most significant clinical and virological protection post-challenge, followed by ΔSHrBRSV and CV, whereas the protection of PreF-vaccinated calves was correlated with BRSV-specific serum immunoglobulin (Ig)G antibody responses 84 days post-vaccination, and the IgG antibody titers of ΔSHrBRSV- and CV-vaccinated calves did not differ from the controls on this day. Nevertheless, strong anamnestic BRSV- and PreF-specific IgG responses occurred in calves vaccinated with either of the vaccines, following a BRSV challenge. In conclusion, PreF and ΔSHrBRSV are two efficient one-shot candidate vaccines. By inducing a protection for at least three months, they could potentially improve the control of BRSV in calves.

Achieving safe and protective vaccination against respiratory syncytial virus (RSV) in infants and in calves has proven a challenging task. The design of recombinant antigens with a conformation close to their native form in virus particles is a major breakthrough. We compared two subunit vaccines, the bovine RSV (BRSV) pre-fusion F (preF) alone or with nanorings formed by the RSV nucleoprotein (preF+N). PreF and N proteins are potent antigenic targets for neutralizing antibodies and T cell responses, respectively. To tackle the challenges of neonatal immunization, three groups of six one-month-old calves with maternally derived serum antibodies (MDA) to BRSV received a single intramuscular injection of PreF, preF+N with MontanideTM ISA61 VG (ISA61) as adjuvant or only ISA61 (control). One month later, all calves were challenged with BRSV and monitored for virus replication in the upper respiratory tract and for clinical signs of disease over one week, and then post-mortem examinations of their lungs were performed. Both preF and preF+N vaccines afforded safe, clinical, and virological protection against BRSV, with little difference between the two subunit vaccines. Analysis of immune parameters pointed to neutralizing antibodies and antibodies to preF as being significant correlates of protection. Thus, a single shot vaccination with preF appears sufficient to reduce the burden of BRSV disease in calves with MDA.

Diacylglycerol acyltransferase-1 (DGAT1) is a potential therapeutic target for treatment of obesity and related metabolic diseases. However, the degree of DGAT1 inhibition required for metabolic benefits is unclear. Here we show that partial DGAT1 deficiency in mice suppressed postprandial triglyceridemia, led to elevations in glucagon-like peptide-1 (GLP-1) and peptide YY (PYY) only following meals with very high lipid content, and did not protect from diet-induced obesity. Maximal DGAT1 inhibition led to enhanced GLP-1 and PYY secretion following meals with physiologically relevant lipid content. Finally, combination of DGAT1 inhibition with dipeptidyl-peptidase-4 (DPP-4) inhibition led to further enhancements in active GLP-1 in mice and dogs. The current study suggests that targeting DGAT1 to enhance postprandial gut hormone secretion requires maximal inhibition, and suggests combination with DPP-4i as a potential strategy to develop DGAT1 inhibitors for treatment of metabolic diseases.

African swine fever (ASF) is a contagious disease of wild boar and domestic pigs notifiable to the World Organisation for Animal Health due to its high socio-economic impact. ASF is caused by the complex ASF virus (ASFV), and it can present different clinical manifestations that can be confused with other diseases; for this reason, laboratory testing is necessary for the proper diagnosis of clinically suspected animals. Despite the efforts put into it over decades, no treatment or safe vaccine is globally available, and disease control is based on early diagnosis and the implementation of strict biosecurity measures. In this context, rapid tests have the potential to accelerate and facilitate the identification of infected animals by giving fast on-site results. In this work, we improved the available point-of-care assays for the diagnosis of the disease by the development of a more specific antigen test and a more sensitive antibody test. This antibody detection test allowed for the earlier detection of infected animals than two commercial indirect ELISAs (statistically significant). Moreover, we developed a combined dual rapid test, unifying, in the same cassette, an antigen detection strip and an antibody detection strip. In this study, we confirmed that this combo approach is a useful tool for implementing rapid tests in the field since it increases the percentage of positive samples detected, even when PCR turns negative, while maintaining a good specificity.

The arrival of a new invasive alga, Rugulopteryx okamurae, in the Strait of Gibraltar (SoG) in 2015 marked an unprecedented milestone in the North African and, later, in the European marine ecosystems. Nowadays, it is colonising vast infralittoral areas and significantly modifying some habitats and associated communities of the southern Iberian Peninsula. In recent expeditions, a high amount of free drifted thalli of this alga has been detected in different circalittoral and bathyal habitats of the northern SoG and the Alboran Sea. The present study combines quantitative data of this alga obtained with the use of a remotely operated vehicle (ROV) and a bottom otter trawl. The coverage–entanglement level of the drifted thalli on circalittoral and bathyal benthic invertebrates (e.g., not covering, covering only the basal part, covering one-third of the invertebrate, etc.) was also annotated from picture frames taken in locations with abundant drifted thalli. In underwater images, drifted thalli were mainly detected in circalittoral and bathyal bottoms of the northern SoG and the north-western Alboran Sea, between 50 to ca. 450 m depth. Nevertheless, abundant drifted thalli were also detected in bottom otter trawl samples from circalittoral bottoms of the north-central and north-eastern Alboran Sea. Small benthic organisms (e.g., encrusting sponges, hydrozoans, etc.) generally displayed low coverage–entanglement levels of drifted thalli. Nevertheless, large sessile and colonial benthic organisms with a complex three-dimensional morphology (e.g., gorgonians, colonial scleractinians) reached high levels of R. okamurae thalli entangled in different parts of their colonies. The drifted R. okamurae thalli entangled in these colonial suspension feeding organisms may hinder their feeding capability in the long term, resulting in habitat deterioration in the near future.

African swine fever (ASF) is a viral disease of swine with a huge impact due to its high mortality. Lately, the disease has actively spread around the world, affecting new areas from which it had been eradicated long ago. To date, ASF control is carried out by the implementation of strict biosecurity measures such as the early identification of infected animals. In this work, two fluorescent rapid tests were developed to improve the sensitivity of point-of-care diagnosis of ASF. For antigen (Ag) detection in blood, a double-antibody sandwich fluorescent lateral flow assay (LFA) was developed, employing a newly developed recombinant antibody to the VP72 of the virus. To complement the diagnosis, a double-recognition fluorescent LFA was developed using the VP72 for the detection of specific antibodies (Ab) in sera or blood. Both assays statistically improved the detection of the disease when compared to the commercial colorimetric assays INgezim® ASFV CROM Ag and INgezim® PPA CROM Anticuerpo, respectively, with higher statistical significance between 11 and 39 days post-infection. From the observation of results, it can be concluded that the combination of both Ag-LFA and Ab-LFA assays would facilitate the identification of infected animals, regardless of post-infection time.

Clinoform depositional features along the Iberian Mediterranean margin are investigated in this study, with the aim of establishing the causes of their varied shapes and other characteristics. We have analyzed the broad-scale margin physiography and seismic stratigraphic patterns based on high-resolution bathymetric data and previously interpreted seismic data. In addition, we have evaluated regional supply conditions and the uplift-subsidence regime of the different shelf sectors. The upper Quaternary record is strongly dominated by shelf-margin regressive wedges affected by the prevailing 100 ka cyclicity. However, the margins exhibit considerable lateral variability, as the result of the balance between the amount of sediment supply and the uplift-subsidence relationship. Three major shelf sectors with distinct morpho-sedimentary features have been defined. The relatively narrow northern shelves (Roses, La Planassa and Barcelona) are supplied by discrete river outlets that collectively constitute a linear source and are mainly affected by tectonic tilting. The wide middle shelves (Ebro Shelf, the Gulf of Valencia, and the Northern Arc) receive the sediment supply from the large Ebro River and other medium rivers. Although the tectonic regime changes laterally (strong subsidence in the north and uplift in the south), shelf growth is maintained by lateral advection of sediments. The southern shelves (the Southern Arc and the northern Alboran Shelf) are very abrupt and narrow because of the uplifting Betic Cordillera, and the torrential fluvial regimes that determine a very efficient sediment by-pass toward the deep basin. Submarine canyons deeply incised in the continental margin constitute a key physiographic feature that may enhance the transport of sediment to the deep sea or individualize shelf sectors with specific sedimentation patterns, as occurs in the Catalan margin.

This research looked at the liaison between the library information professionals and academic staff of academic institutions stressing the aspect of information literacy. The general aims of this research were to emphasize the importance of training in information literacy in higher education, for students, academic staff and library staff and to stress that essential collaboration between librarians and faculty staff is basic for successful results. The research method used to collect data was the interview. Interviews were conducted with both librarians and lecturers at Thames Valley University and at some colleges of the University of London
