Objectives To describe a patient with BRCA1 mutation, mucoepidermoid parotid, multiple breasts, and thyroid cancers. Case Report A women was diagnosed at 33-years-age with a triple-negative breast cancer (right breast), at 43-years-age with a triple-negative breast cancer in left breast and at 53-years-age with a primary papillary-thyroid carcinoma. At 55-years-age, she was diagnosed with a primary mucoepidermoid carcinoma in right parotid, and concomitantly, her right nipple was affected by Paget's disease and a recurrent carcinoma in right breast (HR + /HER2 = 3 +). At 57-years-age, after the recurrence of a triple-negative breast cancer (left breast), a geneticist evaluated the patient's family history, including one stomach, one non-smoking-related lung, and two smoking-related laryngeal cancers. Genetic testing revealed a BRCA1 mutation (Chr17:41:251.867). The patient's daughter (a non-cancer patient) tested negative for the mutation. Both remain under medical supervision. Conclusions We suggest that BRCA1 mutations are associated with non-breast and non-ovarian cancers such as salivary gland cancer.

INTRODUCTION: PMS1 is part of the cluster of genes related to Mismatch repair genes (MMR) and has been used for investigation in oncogenetic panels. The MMR genes play an important role in tumor control and progression, however, the role of PMS1 in this process still poorly understood and information about its role in increasing the risk of developing a hereditary cancer predisposition syndrome (SHPC) is not completely understood.AIM: To characterize clinically and genetically cancer patients with NCCN criteria for SHPC and carriers of variants in the PMS1 gene.METHODOLOGY: A total of 368 patients suspected of having SHPC, according to the National Comprehensive Cancer Network (NCCN) criteria, were investigated using a Next-Generation Sequencing (NGS) in a panel containing 31 genes. Those that showed variation in the PMS1 gene were grouped, and the tumors were characterized in clinical and molecular aspects.RESULTS: From the 368 patients analyzed, 6.8% (25/368) patients presented Variants of Uncertain Significance (VUS) in PMS1. There was a case of Breast Cancer (BC) with a variant in PMS1, not described in ClinVar, presented in heterozygosis, probably pathogenic [Chr2:190.738,325 NM_000534: c.2578delA: p.(Arg860.Glufs*14)]. Besides, this case was associated with a family history (FH) of cancer breast cancer and melanoma. It is worth mentioning that only one patient was a man, with colorectal cancer (CRC), and his FH also was CRC in addition to stomach cancer. The only change founded was the presence of VUS NM_000534:c.1615A>G:p.(Met539Val) which was identified in 32% (8/25). This VUS was the only genetic alteration observed in 20% (5/25) patients with tumors in the breast (3), thyroid (1), CRC (1), and ovary (1). One of these patients had bilateral breast cancer and thyroid tumor. The patients had their age at diagnosis ranged from 25 to 65 years, with an average of 41 years. The carriers of variants exclusively in PMS1 [40% (10/25)] had different tumors: breast (6); CRC (3); ovary (2); lymphoma (1); thyroid (1); others 32% (n = 8/25) that presented additional VUS in different genes: [ATM (2); BARD (1); CDH1 (1); CHECK2 (1); NBN (1); APC (1); POLE (1)]. Different pathogenic variants (PV) [28% (n = 7/25)] also were identified, besides the previous PMS1 VUS described. The PV was founded in ATM (1); BRCA1 (3); NF1 (1); TP53 (1); MUTYH-heterozygosis (1), and PMS1 (1) gene. A double pathogenic variation was identified in a patient (MUTYH in heterozygosis and BRCA1). The findings in PMS1, even currently classified as VUS, should highlight observation for the clinical and familiar history. Genetic panels should be increasingly used in the investigation of SPHC.CONCLUSION: The roles of PMS1 in cancer progression need further investigations. A few numbers of reports have been identified on germline PMS1 mutations, considering defined disease phenotypes. Therefore, the performance of genetic panels, including the PMS1 gene, in SHPC further investigations will expand our knowledge and permit precise genetic counseling. INTRODUCTION: HBOC syndrome is directly associated with a variant in BRCA1/2 genes and the diagnosis may increase anxiety and depression levels because of the need for preventive measures. Psychic impacts on women with mutations have been further explored in the literature, although the emotional aspects linked to the diagnosis in male breast cancer is poorly known. AIM: This work aimed to compare the anxiety, depression, and quality of life levels in male and female cancer patients HBOC-suspected, immediately after the result of the genetic test. METHODOLOGY: A retrospective case-control study was carried out with five men matched for age with 39 women, both with National Comprehensive Cancer Network (NCCN) criteria for HBOC. The Hospital Anxiety and Depression (HAD) scale and the WHOQOL-bref quality of life inventory were used. Sociodemographic, clinical, molecular, and psychological data were analyzed using χ2/Fisher's exact and Mann-Whitney, Friedman/Dunn, and Wilcoxon tests (SPSS, 20.0; p 45 years old, all recruited women had breast cancer as the primary tumor and eight of them (20.5%) had ovary cancer. We had two male breast cancer and three prostate cancer Gleason>7. Sociodemographic characteristics did not differ between the genders (p> 0.05). Men had a higher prevalence of pathogenic mutation (p=0,031). Women had a higher prevalence of BRCA1 mutations (p=0,014) while men had of BRCA2 mutation (p=0,001). There was no difference in the anxiety and depression levels immediately after receiving the result, but it was observed that only women had lower quality of life in the physical domain (p=0,048). INTRODUCTION: The male breast cancer (MBC) is a rare disease, responsible for about 0.2% of all cancers and 0.1% of deaths for male cancer. Despite the rarity of the disease, statistics indicate that the incidence of MBC has been increasing significantly. The main risk factors for the development of MBC include old age, hormonal imbalance, radiation exposure, and family history of breast cancer. Hereditary breast and ovarian cancer (HBOC) is a syndrome associated with mutations in the BRCA1 and BRCA2 genes that have been related to MBC that as a rare disease, still poorly understood. METHODOLOGY: Family history analyzes, type and tumor staging, molecular markers expression, hormone receptors (RE, RP, and HER2), and cell proliferation marker (KI67) analysis were performed by immunohistochemistry. The presence of genetic mutations and the frequency of these mutations were evaluated in 31 genes of HBOC suspected patients treated at the Haroldo Juaçaba Hospital between the years 2009 to 2020. RESULTS: A total of 236 patients were diagnosed with breast cancer and submitted to a genetic test. Six of them were male patients aged between 54 and 73 years. His family history indicated that 83.3% had first-degree relatives affected by breast cancer. Different pathological staging was founded after tumors evaluation with the presence of micrometastases (P3) and higher levels of invasion in the lymph nodes. The expression of the cell proliferation marker (KI67) indicated low levels of expression except for patient P5. All patients presented expression of two hormone receptors (ER and RP) and no expression for HER2, except for one patient (P1), who had bilateral breast cancer, with HER2 expression (+2) on the right breast (first tumor) and no RP and HER2 expression in the left breast (second tumor), besides a moderate expression of KI67 (40%) (Table 1). The oncogenetic evaluation indicated that 83.33% of the patients had mutations described in the clinical database (ClinVar) 60% of them were pathogenic. The variants founded were in the PALB2 (16.66%), BRCA1 (33.33%), and BRCA2 (33.33%) genes (Table 1). The pathogenic mutations founded were located in the BRCA 1 and BRCA2 genes. One patient (P5) did not present mutations or VUS. The patient without the mutation (P5) presented a tumor with T3N1 pathological staging, low expression of estrogen (10%) and progesterone (10%) receptors, absence of HER2 expression, and high capacity of proliferation, indicated by the expression of KI67 (80%). The results indicate heterogeneity in the histological and molecular patterns of the patients evaluated, besides the oncogenetic patterns that may be associated with HBOC, especially those whose variants founded were pathogenic.CONCLUSION: Due to the low frequency of male breast cancer, the oncological data of these patients are relevant for epidemiological, pathological, and oncogenetic characterization, improving the characterization and identification of patterns for this pathology.Research Sponsor: Ministerio da Saude, Brasil (PRONON) Table 1. Emotional distress associated with genetic testing for hereditary breast and ovarian cancer syndrome (HBOC) is reported to interfere with adherence to treatment and prophylactic measures and compromise quality of life. To determine levels of anxiety, depression, and quality of life in patients tested for pathogenic BRCA1/2 mutations and identify risk factors for the development of adverse psycho-emotional effects. Cross-sectional observational trial involving 178 breast or ovarian cancer patients from a referral cancer hospital in Northeastern Brazil. Information was collected with the Hospital Anxiety and Depression Scale (HADS) and the World Health Organization (WHO) Quality of Life (QoL) questionnaire (WHOQOL-BREF). Patients suspected of HBOC had higher levels of anxiety than depression. The presence of (probably) pathogenic BRCA1/2 mutations did not affect levels of anxiety and depression. High schooling, history of psychiatric disease, and use of psychotropic drugs were directly associated with high anxiety. High schooling was too inversely associated with QoL as such a breast tumor. Anxiety and depression were directly correlated and both reduced significantly QoL. Our results highlight the importance of psychological support and screening of risk factors for anxiety and depression and low QoL in HBOC patients at the time of testing.

Background Penile cancer is one of the most aggressive male tumors. Although it is preventable, the main etiologic causes are lifestyle behaviors and viral infection, such as human papillomavirus (HPV). Long-term epigenetic changes due to environmental factors change cell fate and promote carcinogenesis, being an important marker of prognosis. We evaluated epidemiological aspects of penile squamous cell carcinoma (SCC) and the prevalence of HPV infection using high-risk HPV (hrHPV) and p16INK4A expression of 224 participants. Global DNA methylation was evaluated through 5-methylcytosine (5mC) and 5-hydroxymethylcytosine (5hmC). Results The incidence of HPV was 53.2% for hrHPV and 22.32% for p16INK4a. hrHPV was not related to systemic or lymph node metastasis and locoregional recurrence, nor influenced the survival rate. P16INK4a seems to be a protective factor for death, which does not affect metastasis or tumor recurrence. Lymph node and systemic metastases and locoregional recurrence increase the risk of death. An increased 5mC mark was observed in penile SCC regardless of HPV infection. However, there is a reduction of the 5hmC mark for p16INK4a + (P = 0.024). Increased 5mC/5hmC ratio (> 1) was observed in 94.2% of penile SCC, irrespective of HPV infection. Despite the increase in 5mC, it seems not to affect the survival rate (HR = 1.06; 95% CI 0.33–3.38). Conclusions P16INK4a seems to be a good prognosis marker for penile SCC and the increase in 5mC, an epigenetic mark of genomic stability, may support tumor progression leading to poor prognosis.

Brazilian Somalis is a locally-adapted breed of rams raised in tropical climate and native pastures. The present study was conducted to evaluate gene expression and proteome of the reproductive tract of such rams. Samples were collected from testes, epididymides, seminal vesicles and bulbourethral glands of four rams. Expression of clusterin (CLU), osteopontin (OPN) and prostaglandin D2 synthase (PGDS) genes were evaluated in all samples by real-time PCR. Shotgun proteomic analysis was performed using samples from the head, corpus and cauda epididymides and from all other structures as well. Gene ontology terms and protein interactions were obtained from UniProtKB databases and MetaCore v.6.8 platform. CLU trasncripts were detected in the testes, epididymides, seminal vesicles and bulbourethral glands of the Somalis rams. The initial region and body of the epididymis had the greatest CLU expression. OPN mRNA was localized in all tissues of the ram reproductive tract. PGDS mRNA was detected in the testes and epididymides. Lable-free mass spectrometry allowed the identification of 137 proteins in all samples. Proteins of the epididymis head mainly participate in cellular processes and response to stimulus, participating in catalityc activity and binding. Proteins of epididymis body acted as regulatory proteins and in cellular processes, with binding and catalytic activity. Cauda epididymis molecules were associated with cellular processes and regulation, with binding function and catalytic activity as well. Testis proteins were mainly linked to cell processes and response to stimuli, and had catalytic function. Seminal vesicle proteins were involved in regulation and mainly with binding functions. Most bulbourethral gland proteins participated in cellular processes. The present study is the first to evaluate the proteome and gene expressions in the reproductive tract of Brazilian Somalis rams. Such pieces of information bring significant cointribution for the understanding of the reproductive physiology of locally-adapted livestock.

Five adult females received an ovarian stimulation treatment and the FF was collected by laparoscopy from small/medium (3.5 mm) follicles. Concentrations of soluble proteins in FF samples were measured and proteins were analyzed by 1-D SDS-PAGE followed by tryptic digestion and tandem mass spectrometry. Data from protein list defined after a Mascot database search were analyzed using the STRAP software tool. For the protein concentration, no significant difference (P> 0.05) was observed between small/medium and large follicles: 49.2 +/- 22.8 and 56.7 +/- 27.4 mu g/mu l, respectively. Mass spectrometry analysis identified 13 major proteins, but with no significant difference (P> 0.05) between follicle size class. This study provides insight into elucidating folliculogenesis in brown brocket deer. e20548 Background: Lung cancer has the highest incidence and mortality worldwide. In Brazil, it is the second most common, and individualized therapy, although more efficient than traditional chemotherapy, is still a challenge for health services, especially in developing countries, as socioeconomic disparities make access to genetic testing difficult or treatment. Thus, it is understood that greater knowledge about the genetic profile and therapeutic response of different populations can contribute to policy changes that guarantee expanded access to individualized therapy. Methods: the EGFR gene mutation status was correlated with clinical-epidemiological, socioeconomic, treatment, and survival parameters of patients with lung adenocarcinoma treated from 2014 to 2021 at an oncology referral center in northeastern Brazil. Data were obtained retrospectively from the patient's charts at the Hospital Haroldo Juaçaba/Instituto do Câncer do Ceará. The X²/Fisher and Log-Rank Mantel-Cox tests were used to associate clinical variables and overall survival curves. The Research Ethics Committee approved the research under protocol number 3.544.754. Results: 586 patients tested, 157 (26.8%) had an EGFR mutation, with exon 19 deletion (n = 83, 52.9%). the most frequent followed by L858R (n = 54, 34.4%) and others (n = 30, 12.8%). The EGFR mutation was inversely associated with smoking history (p < 0.001) and previous use of carboplatin plus paclitaxel (p < 0.001). The median overall survival was 33 (95%CI = 25-41) months, and the presence of a mutation was not associated with a worse outcome (p = 0.146). Interestingly, patients with sensitivity mutation who used gefitinib (64.1%) did not show a significant difference in overall survival (p = 0.218). However, when stratifying analyzes according to mutation type, it was observed that patients with exon 19 deletion treated with gefitinib had greater median overall survival (32, 95%CI = 27-37 months) than patients with this mutation not treated with gefitinib (median = 15, 95%CI = 2-25 months) (p < 0.001). Patients with the L858R type sensitivity mutation (p = 0.523) or other mutations (p = 0.439) did not experience significant benefits from gefitinib use. Conclusions: In the population studied, the exon 19 deletion was the most frequent and the only one associated with a substantial benefit from using gefitinib.

e22529 Background: Genetic tests have been increasingly requested to identify families at risk of hereditary cancer. In a family with a known mutation, the offspring of a carrier, whether male or female, have a 50% risk of inheriting the mutation. Most studies on the psychological implications associated with genetic testing have focused on women and few studies have focused on the male population. Methods: A total of 56 men with a personal history of cancer underwent screening for mutations in a panel of 31 genes associated with hereditary cancer. Assessment of quality of life (QoL) and levels of anxiety and depression was performed after patients received the genetic test result. For this, the HADS and WHOQOL-BREF scales were used. Wilcoxon, X², and Spearman correlation tests were used (SPSS v20.0 for Windows). Results: Of the 56 participants, 28.6% had a pathogenic mutation, 32.1% had a Variant of Undetermined Significance (VUS), and 39.3% had no mutation. The presence of the mutation was not associated with QoL (p = 0.967) or anxiety (p = 0.436) or depression (p = 0.945). Mean QoL was 77.95±7.38 (range = 50.00-95.20), mean anxiety was 5.14±2.62 (range = 1-14) and mean depression scores were 4.14±2.55 (range = 0-14). QoL was inversely correlated with levels of anxiety (p < 0.001, r = -0.496) and depression (p < 0.001, r = -0.494), parameters that were directly correlated (p = 0.037, r = 0.279). Most patients had QoL > 80 (n = 38, 67.9%), and only 16 (28.6%) and 13 (23.2%) men had HAD scores > 5. Conclusions: The result of carrying a mutation did not influence the quality of life and anxiety or depression levels in the male population evaluated. However, it was observed that levels of anxiety and depression can compromise the quality of life. The data from the present study reinforce the importance of considering the psychological aspects in the process of genetic investigation for hereditary cancer. In addition, it suggests that educational levels must be investigated to assess whether the low levels of anxiety and depression observed are associated with a low level of understanding of the clinical significance of carrying a pathogenic mutation.

e22528 Background: The familiar history is indispensable to clinical investigation during genetic counseling and the new molecular techniques allowed the identification of hereditary cancer predisposition syndromes (CPS). However, the classification systems of these syndromes are difficulty and very complex. In 2015, the American College of Medical Genetics and Genomics (ACMG) and the Association for Molecular Pathology (AMP) published guidelines for the assessment of variants in genes associated with Mendelian diseases to reduce the subjectivity bias of mutation variants classified as risk. Methods: To show the difficulty of classification of these variants we describe a case report of a family from countryside of Brazil in which there is a high rate of inbreeding. This family has three brothers with polyposis and colorectal cancer between 51 and 60 years-old. Results: The index-case had multiple polyps, developed colorectal cancer at 51 years-old and had a familiar history of two brother dead by colorectal cancer after polyposis and nine brother without familiar history of cancer, a father dead at 38 years-old by non-specific lung-disease and a mother alive at 83 years-old. There was no description of other cases of cancer among uncles and grandparents. The index-case performe a NSG genetic painel to CPS and showed a homozygosis variant in MUTYH gene: NM_001048174.1: c.253T > C:p.(Trp85Arg), that was classified as variant of uncertain significance (VUS). MUTYH is a base excision repair enzyme, it plays a crucial role in the correction of DNA errors and may be considered a cell protective factor and is associated with MUTYH-related polyposis (MAP). MAP is an autosomal recessive CPS and presents a phenotype that overlaps attenuated polyposis and familial adenomatous polyposis, increasing the risk of colorectal cancer in 43-63% up to 60 years or 80% if there is no surveillance/tracking. In view of the family history that suggests autosomal recessive inheritance, with a homozygous variant absent in the database of population controls (gnomAD) and in which in silico predictors indicate the effect of this variant as deleterious, the management of this family must be carried out considering it as a potential high-risk pathogenic variant. Conclusions: While the classification criteria for the variants are being refined, we reinforce the importance of a careful family history in the approach to CPS in order to offer an appropriate clinal conduct in the prevention of hereditary cancer.

Metal-based compounds have emerged as a novel strategy to improve our arsenal of medicines. Among these compounds, carbon monoxide donor agents have been developed mainly as metal complexes. These compounds have shown exciting biological activities, among them strong anti-inflammatory, antibiotic, and anticancer activity, although some of those properties might be associated with the metal moiety as well. Aiming to prepare a new ruthenium complex containing CO, we prepared a cis-[RuCl(CO)(dppb)(dppz)]PF6 complex (dppb = 1,4-bis(diphenylphosphino)butane and dppz = dipyrido[3,2-a:2′,3′-c]phenazine), which was fully characterized along with DFT studies. This compound exhibited great thermal stability in phosphate buffer solution, but showed fast and efficient release of CO upon light irradiation, including blue LED. Surprisingly, DNA binding was quite weak, despite the dppz ligand moiety, suggesting that steric effects prevented intercalation binding. Nonetheless, this compound exhibited promising antibiotic activity against S. aureus, S. epidermidis and C. albicans in a low micromolar range. However, there was no further improvement upon light irradiation suggesting that there is no role of CO. Additionally, cytotoxicity assay using cancer cells showed quite low IC50 (92 nM, HCT-116) along with moderate to low toxicity assayed in lymphocytes and Artemia. Altogether, these results supported that this compound has great potential as a biological agent, whereas neither CO release nor DNA binding seemed to have a direct role in its activity.

e13670 Background: Loss-of-function mutations in PALB2 gene are associated with increased risk for breast cancer and possibly pancreatic, ovarian, male breast, prostate, colorectal as others cancers. In Brazil it has been estimated that up to 1,516 new cases of hereditary breast cancer for 2020 in the North and Northeast regions. Analysis of susceptibility gene mutations helps identify precisely the high-risk patient and their families, whom need specific and personalized clinical management as high-risk individuals. Methods: Twenty-six patients with pathogenic mutations in PALB2 gene identify by next-generation sequencing from states of Bahia (11), Ceará (9), Pernambuco (5) and Rondônia (1) in the North and Northeast regions were analyzed. Results: Most of the patients analyzed had only breast cancer (80%), including two cases of male breast cancer (9,5%); the others were isolated cases of endometrial cancer (4%), breast and pancreas cancers (4%), breast and lung cancers (4%), only ovarian cancer (4%) and ovarian and breast cancers (4%). Most cancers were stage II or III (65%). Family history of cancer was observed in 22/26 (84%); the most common tumors were breast, prostate, pancreas and thyroid. The founder mutations were more frequent in exons: 4 (58%) and 12 (15%). Eleven variants were found as follow: c.1240C > T (19%); c.3256delC (15%); c.1671_1674delTATT (11.5%); c.355delC (11.5%); NC_000016.9:g.(?_23632673)_(23652488_?)del (11,5%). The greatest variety of mutations was found in the state of Bahia, probably due to the greater number of patients included (42%). Conclusions: These data suggest that changes in clinical management of PALB2 patients are needed since the phenotype observed exhibited pattern of hereditary tumors, including male breast cancer. Besides that, PALB2 gene should be included in painel gene analysis in patients from the North and Northeast of Brazil because its high frequency of pathogenic variants.

e13532 Background: The Lynch syndrome (LS) is the most common inherited syndrome associated with colorectal cancer (CRC). The hallmark of LS is DNA mismatch repair deficiency. The amount of variants of uncertain significance (VUS) in suspected LS confounds diagnosis, requiring surveillance of variant reclassifications. In this paper we describe a new and most likely pathogenic VUS in PMS2. Methods: Patient screened for personal and familial cancer and evaluated with NGS gene panels for 31 genes. The protein structure was analyzed with the software Pymol. Results: A woman diagnosed with breast cancer (BC) at 60 years and CRC at 65 years was investigated due to her extensive family history of cancer (21 maternal relatives), including 6/9 siblings [gastric n = 1, lung n = 1, BC n = 4 (one of whom with BC, endometrium and CRC)], 8/12 aunts and uncles, all of whom died of cancer (gastric n = 4, CRC n = 1, CNS n = 1, BC n = 1; uterus/gallbladder n = 1), and 7 cousins (CNS n = 3, lung n = 1, gastric n = 2, suspected ovary n = 1). The new PMS2 gene variant c.134A > G(p.Asn45Ser), which was also detected in two sisters with CRC submitted to segregation analysis, occupied the same locus as a known pathogenic variant, c.134A > C(p.Asn45Thr). The structural analysis showed that the mutated locus encodes a serine instead of an asparagine in position 45 at the enzyme’s active site. Asn 45 is a conserved amino acid that binds to ATP in protein structure (PDB = 1EA6). Interestingly, this is close to position 42-44, for which another potentially pathogenic VUS (p.Leu42_Glu44del) has been described. Conclusions: Based on our patient’s personal and family history and on the locus variant and protein structure analysis, the new PMS2 gene variant described in this paper is most likely pathogenic.

e13603 Background: Identifying carriers of hereditary cancer predisposition syndromes (HCPS) improves prevention and treatment. However, due to the elevated cost of genetic testing, information on the prevalence and incidence of HCPS is scarce, depriving patients of assertive treatment―a scenario observed worldwide, especially in regions like Northeastern Brazil. In this study, we determined the prevalence of hereditary breast and ovarian cancer (HBOC) in patients from Ceará (Brazil) based on NCCN criteria. Methods: The patients (n = 267) were evaluated using NGS gene panels, while retrospective data were analyzed statistically with the chi-squared test, Fisher’s test and multinomial logistic regressions ( p< 0.05; SPSS 20.0). Results: BRCA1/2 was detected in 21.3% (57/267) of the sample, but BRCA1 was more prevalent than BRCA2 (70%; 40/57 vs 30%; 17/57) ( p< 0.001). Having two or more primary tumors ( p= 0.031) or primary ovarian cancer ( p= 0.029) was associated with the pathogenic mutation BRCA1/2. The latter was detected in patients with bilateral tumors (40%) and male breast cancer (45%), and in patients under 45 years (67%), with familial breast cancer (27%) and familial ovarian cancer (41%). In the multivariate analysis, the triple-negative phenotype was strongly associated with mutated BRCA1/2 ( p= 0.003). Conclusions: The prevalence of HBOC in our sample is compatible with rates reported worldwide. The study variables (bilaterality, male sex, age < 45 years, family history of breast or ovarian cancer, and triple-negative phenotype) reinforce the diagnostic criteria published by the NCCN.

e13602 Background: Breast cancer (BC) is associated with several hereditary syndromes (HS). Patients with clinical criteria for hereditary breast and ovarian cancer (HBOC) should be evaluated for the presence of pathogenic BRCA1/2 variants, although the absence of BRCA1/2 variants does not rule out HS. The purpose of this study was to evaluate the prevalence of pathogenic non- BRCA1/2 variants in patients with suspected HBOC. Methods: Suspected HBOC (NCCN criteria) patients (n = 267) from a Northeastern Brazilian referral cancer hospital were submitted to NGS 31-gene painel. Individuals testing negative for BRCA1/2 mutation (n = 210) remained in the present presentation. Data were analized by the chi-squared test and Fisher’s test ( p< 0.05; SPSS 20.0). Results: Pathogenic/likely pathogenic (P/LP) non- BRCA1/2 variants were detected in 13.3% (n = 28/210) of the patients, all of whom had BC. The most prevalent variant was PALB2 (31%), followed by ATM and MUTYH (14.2%), and TP53 and PMS2 (10.7%). Among these patients 10.7% (n = 3/28) had triple negative tumors. Other gene variants ( BARD1, CHEK2, MLH1, MSH2, PMS1 and XRCC2) were observed at lower frequencies (3.5% each). DNA mismatch repair (MMR)/Lynch syndrome (LS) mutated genes were identified in 4/24 patients with BC. Rare microdeletions were identified in PALB2 (chr16:23652431-23652678 [exons 1-10] and TP53 (chr17:7.573.927-7.579.891 [exons 2-10]) , to our knowledge for the first time in the medical literature (ClinVar). In addition, one patient displayed a double pathogenic variant in ATM (c.67C > T: p.[Arg23*]) and PALB2 (chr16:23652431-23652678 [exons 1-10]). Patients with and without P/LP did not differ with regard to clinical-pathological parameters or therapeutic profile. Conclusions: Patients with suspected HBOC should be evaluated for non- BRCA1/2 variants, including MMR. We also suggest considering BC among the malignancies associated with LS.

The present study was conducted to describe the major seminal plasma proteome of rabbits and potential associations between seminal proteins and semen criteria. Semen samples were collected from 18 New Zealand adult rabbits, and seminal plasma proteins were analyzed by 2-D SDS-PAGE and tandem mass spectrometry. Sperm motility, vigor, concentration, morphology and membrane sperm viability were evaluated. Rabbits ejaculated 364 ± 70 million sperm/ml, with 81 ± 6.1% motile cells, 3.8 ± 0.2 vigor and 66.7 ± 2.5% sperm with normal morphology. Based on the viability and acrosome integrity assay, there were 65.8 ± 2.5% live sperm with intact acrosome and most spermatozoa had both intact acrosome and functional membrane. On average, 2-D gels of rabbit seminal plasma had 232 ± 69.5 spots, as determined by PDQuest software (Bio Rad, USA). Mass spectrometry allowed the identification of 137 different proteins. The most abundant proteins in rabbit seminal plasma were hemoglobin subunit zeta-like, annexins, lipocalin, FAM115 protein and albumin. The intensity of the spots associated with these five proteins represented 71.5% of the intensity of all spots detected in the master gel. Multiple regression models were estimated using sperm traits as dependent variables and seminal plasma proteins as independent ones. Also, sperm motility had positive association with beta-nerve growth factor and cysteine-rich secretory protein 1-like and a negative one with galectin-1. The percentage of rabbit sperm with intact membrane was related to seminal plasma protein FAM115 complex and tropomyosin. Then, the population of morphologically normal sperm in rabbit semen was positively linked to carcinoembryonic antigen-related cell adhesion molecule 6-like and down regulated by seminal plasma isocitrate dehydrogenase. Based on another regression model, the variation in the percentage of live sperm with intact acrosome was partially explained by the amount of leukocyte elastase inhibitor and the peptidyl-prolyl cis-trans isomerase A in the rabbit seminal fluid. The current study reports the identification of 137 proteins of rabbit seminal plasma. Major proteins of seminal secretion relate primarily to prevention of damages caused by lipid peroxide radicals and oxidative stress, membrane functionality, transport of lipids to the sperm membrane and temperature regulation. Moreover, finding seminal plasma proteins as indicators of semen parameters will improve assisted reproductive technologies.

Molecular components of sperm and in the media surrounding them influence male fertility. In this regard, seminal plasma proteins and metabolites modulate various reproductive events, including sperm motility and capacitation, cell protection, acrosome reaction, fertilization and embryonic development. Empirical associations between seminal proteins and metabolites and fertility indicate that these molecules are potential molecular markers of male reproductive status in cattle and other species.

Lectins are comprised of a large family of proteins capable of the specific and reversible recognition of carbohydrates. Legume lectins, the most studied plant lectins, show high structural similarity, but with modifications that imply a variation in the intensity of some biological activities. In this work, the primary and tertiary structures of Canavalia grandiflora (ConGF) were determined. ConGF, a lectin isolated from C. grandiflora seeds, is able to induce relaxant activity in rat aortic rings. The complete sequence of ConGF comprises 237 amino acids. This particular protein has primary sequence variations commonly found in lectins from Dioclea and Canavalia genera. The protein structure was solved at 2.3 Å resolution by X-ray crystallography. An X-Man molecule was modeled into the carbohydrate recognition domain. Still, ConGF (30 and 100 μg mL(-1)) elicited 25% of vasorelaxation (IC50=34.48 ± 5.07 μg mL(-1)) in endothelialized aortic rings. A nonselective inhibitor of nitric oxide blocked ConGF relaxant effect, showing mediation by nitric oxide. Key distances between ConGF carbohydrate recognition domain residues were determined in order to explain this effect, in turn revealing some structural aspects that could differentiate lectins from the Canavalia genera with respect to different efficacy in vasorelaxant effect.

Lectins from Diocleinae subtribe belong to the family of legume lectins and are characterized by high identity between their amino acids sequences. It has been shown that punctual differences in amino acid sequences, such as one single amino acid or an alternative conformation, represent changes in biological activities caused by these lectins. Therefore, a more detailed understanding of three-dimensional structures of these proteins is essential for accurate analyzing the relationship between structure and function. In this study lectins purified from the seeds of Dioclea violacea (DVL) and Dioclea rostrata (DRL) were compared with regard to crystal structure and vasorelaxant properties. Differences in structure of lectins were found to be reflected in differences in vasorelaxant effects based on their high specificity and selectivity for cell glycans. Binding activity was related to the position of specific residues in the carbohydrate recognition domain (CRD). DVL complexed structure was solved by X-ray crystallography and was compared to native DVL and DRL. Therefore, DVL was co-crystallized with X-Man, and a molecular modeling with X-Man complexed with DVL was done to compare the complexed and native forms adjusted fit. The relatively narrow and deep CRD in DVL promotes little interaction with carbohydrates; in contrast, the wider and shallower CRD in DRL favors interaction. This seems to explain differences in the level of relaxation induced by DVL (43%) and DRL (96%) in rat aortic rings.

RATIONALE Lectins are a family of proteins capable of deciphering the glycan code. Several authors have published works about crystallization and mass spectrometry analyses of ConA-like lectins. However, mass spectrometry has never been used to characterize lectin crystal content. In this study, Canavalia grandiflora lectin (ConGF), a ConA-like lectin, was crystallized, part of its primary structure sequenced and the pro-inflammatory activity evaluated. In addition, the crystal content was analyzed by mass spectrometry. METHODS ConGF was crystallized in the presence of X-Man by hanging-drop vapor diffusion at 293 K and the protein crystal content was analyzed by electrospray ionization in a SYNAPT HDMS mass spectrometer. Partial sequence was obtained by protein digestion with several proteolytic enzymes and the peptides sequenced by liquid chromatography/tandem mass spectrometry (LC/MS/MS). The pro-inflammatory potential of ConGF was also evaluated in the model of rat paw edema. RESULTS The protein crystals consist of mature α chain and β and γ fragments measuring 25 612 ± 2 Da, 12 962 ± 2 Da and 12 667 ± 2 Da, respectively. The crystal belongs to the orthorhombic space group I222 (unit cell parameters: a = 67.70, b = 55.90, c = 107.46 A), assuming a monomer in the asymmetric unit. The solvent content was calculated as 43.50% and the protein content as 2.5 µg. Furthermore, a significant part of the primary structure (65.8%) was determined by mass spectrometry. CONCLUSIONS As far as we know this is the first report of lectin crystal content characterized by mass spectrometry. Like other ConA-like lectins, GonGF induced paw edema however differing in potency and duration. The observed pro-inflammatory activity suggests that ConGF might be a useful tool in the study of inflammation processes and structure/function relationships. The crystal structure and pro-inflammatory property of a lectin from the seeds of Dioclea wilsonii (DwL) were analyzed to gain a better understanding of structure/function relationships of Diocleinae lectins. Following crystallization and structural determination by standard molecular replacement techniques, DwL was found to be a tetramer based on PISA analysis, and composed by two metal-binding sites per monomer and loops which are involved in molecular oligomerization. DwL presents 96% and 99% identity with two other previously described lectins of Dioclea rostrata (DRL) and Dioclea grandiflora (DGL). DwL differs structurally from DVL and DRL with regard to the conformation of the carbohydrate recognition domain and related biological activities. The structural analysis of DwL in comparison to other Diocleinae lectins can be related to the differences in the dose-dependent pro-inflammatory effect elicited in Wistar rats, probably via specific interactions with mast cells complex carbohydrate, resulting in significant paw edema. DwL appears to be involved in positive modulation of mast cell degranulation via recognition of surface carbohydrates. Since this recognition is dependent on site volume and CRD configuration, edematogenesis mediated by resident cells varies in potency and efficacy among different Diocleinae lectins.

DwL, a lectin extracted from the seeds of Dioclea wilsonii, is a metalloprotein with strong agglutinating activity against rabbit and ABO erythrocytes, inhibited by glucose and mannose. DwL was purified by affinity chromatography on a Sephadex G-50 column and ion exchange chromatography on a HiTrap SP XL column. SDS-PAGE revealed three electrophoretic bands corresponding to the α (25,634 ± 2 Da), β (12,873 ± 2 Da) and γ (12,779 ± 2 Da) chains. Protein sequencing was done by Tandem Mass Spectrometry. The primary sequence featured 237 amino acids and was highly homologous to other reported Diocleinae lectins. A complete X-ray dataset was collected at 2.0 Å for X-Man-complexed DWL crystals produced by the vapor diffusion method. The crystals were orthorhombic and belonged to the space group I222, with the unit-cell parameters a = 59.6, b = 67.9 and c = 109.0 Å. DWL differed in potency from other ConA-like lectins and was found to induce neutrophil migration in rats, making it particularly useful in structural/functional studies of this class of proteins.

Diocleinae lectins are highly homologous in their primary structure which features metal binding sites and a carbohydrate recognition domain (CRD). Differences in the biological activity of legume lectins have been widely investigated using hemagglutination inhibition assays, isothermal titration microcalorimetry and co-crystallization with mono- and oligosaccharides. Here we report a new lectin crystal structure (ConBr) extracted from seeds of Canavalia brasiliensis, predict dimannoside binding by docking, identify the α-aminobutyric acid (Abu) binding pocket and compare the CRD of ConBr to that of homologous lectins. Based on the hypothesis that the carbohydrate affinity of lectins depends on CRD configuration, the relationship between tridimensional structure and endothelial NO synthase activation was used to clarify differences in biological activity. Our study established a correlation between the position of CRD amino acid side chains and the stimulation of NO release from endothelium.

The lectin from seeds of Dioclea virgata (DvirL) was purified in a single step affinity chromatography, sequenced by tandem mass spectrometry and submitted to crystallization and biological experiments. DvirL has a molecular mass of 25,412 ± 2 Da and the chains β and γ has 12,817 Da ± 2 and 12,612 Da ± 2, respectively. Primary sequence determination was assigned by tandem mass spectrometry and revealed a protein with 237 amino acids and 87% of identify with ConA. The protein crystals were obtained native and complexed with X-Man using vapor-diffusion method at a constant temperature of 293 K. A complete X-ray dataset was collected at 1.8 Å resolution. DvirL crystals were found to be orthorhombic, belonging to the space group I222, with a unit cell parameters a = 647.5 Å, b = 86.6 Å, c = 90.2 Å. Molecular replacement search found a solution with a correlation coefficient of 77.1% and an R(factor) of 44.6%. The present study also demonstrated that D. virgata lectin presents edematogenic and antinociceptive activities in rodents electing this protein as a candidate to structure/function analysis.

Light absorption of $\ensuremath{\alpha}$-glycine crystals grown by slow evaporation at room temperature was measured, indicating a $5.11\ifmmode\pm\else\textpm\fi{}0.02\phantom{\rule{0.3em}{0ex}}\mathrm{eV}$ energy band gap. Structural, electronic, and optical absorption properties of $\ensuremath{\alpha}$-glycine crystals were obtained by first-principles quantum mechanical calculations using density functional theory within the generalized gradient approximation in order to understand this result. To take into account the contribution of core electrons, ultrasoft and norm-conserving pseudopotentials, as well as an all electron approach were considered to compute the electronic density of states and band structure of $\ensuremath{\alpha}$-glycine crystals. They exhibit three indirect energy band gaps and one direct $\ensuremath{\Gamma}\text{\ensuremath{-}}\ensuremath{\Gamma}$ energy gap around $4.95\phantom{\rule{0.3em}{0ex}}\mathrm{eV}$. The optical absorption related to transitions between the top of the valence band and the bottom of the conduction band involves $\mathrm{O}\phantom{\rule{0.2em}{0ex}}2p$ valence states and $\mathrm{C},\mathrm{O}\phantom{\rule{0.2em}{0ex}}2p$ conduction states, with the carboxyl group contributing significantly to the origin of the energy band gap. The calculated optical absorption is highly dependent on the polarization of the incident radiation due to the spatial arrangement of the dipolar glycine molecules; in the case of a polycrystalline sample, the first-principles calculated optical absorption is in good agreement with the measurement when a rigid energy shift is applied.

Single crystals of monohydrated $L$-asparagine have been grown from aqueous solutions using $\mathrm{Mn}{\mathrm{Cl}}_{2}$ as doping material. Electron paramagnetic resonance (EPR) was used to determine the incorporation sites of ${\mathrm{Mn}}^{2+}$ ions in the crystal structure. Depending on small $p\mathrm{H}$ changes and crystal growth kinetics in the aqueous solutions, ${\mathrm{Mn}}^{2+}$ ions are incorporated in two chemically distinct sites in asparagine crystals. The first shows isotropic six-line hyperfine-split EPR spectra, whereas the second shows anisotropic multiple line splitting due to ${\mathrm{Mn}}^{2+}$ fine structure $(S=5∕2)$ and hyperfine interaction $(I=5∕2)$. Angular dependencies of the ${\mathrm{Mn}}^{2+}$ EPR spectra in three mutually perpendicular crystal planes were measured and analyzed. The results are discussed in terms of the metal incorporation site symmetry in the crystal structure of monohydrated $L$-asparagine.

Lectins from the Diocleinae subtribe (Leguminosae) are highly similar proteins that promote various biological activities with distinctly differing potencies. The structural basis for this experimental data is not yet fully understood. Dioclea rostrata lectin was purified and crystallized by hanging-drop vapour diffusion at 293 K. The crystal belongs to the orthorhombic space group I222, with unit-cell parameters a = 61.51, b = 88.22, c = 87.76 A. Assuming the presence of one monomer per asymmetric unit, the solvent content was estimated to be about 47.9%. A complete data set was collected at 1.87 A resolution.
