Kysyn, millaisia olivat muistitiedon kertomukset pakkosiirrosta. Aineistonani käytän kahdenkymmenen vuosina 1941–1942 karkotetun inkerinsuomalaisen henkilöhistoriallista haastattelua. Analysoin kertomuksia vastakertomuksen käsitteen avulla. Esitän ensiksi, että pakkosiirrettyjen inkerinsuomalaisten haastateltujen kertomukset pakkosiirrosta olivat usein vastakertomuksina neuvostovallan toimia vastaan, jotka leimasivat suomalaiset vihollisiksi ja toteuttivat epäinhimillisellä tavalla suomalaisten pakkosiirrot kaukaisiin paikkoihin alkeellisiin oloihin. Karkotetut kärsivät syrjinnästä, nälästä, sairauksista, pakkotyöstä ja surkeista elinoloista kuolleisuuden ollessa korkea. Toiseksi esitän, että inkerinsuomalaisten muistitieto sisälsi myös myönteisiä kertomuksia Siperiassa eletystä ajasta vastoin inkerinsuomalaisten hallitsevaa kärsimyskertomusta tai tutkijoiden luomaa kuvaa siitä. Paikallisten ihmisten apu ja paikoin parempi ruokatilanne pelastivat monen hengen. Muistitieto nosti esille neuvostoarjen kohenemisen monissa karkotuspaikoissa 1940-luvun kuluessa, mutta samalla haastateltavat kuvasivat totalitaristisen neuvostojärjestelmän edelleen ongelmalliseksi.

Sensorimotor adaptation, or the capacity to flexibly adapt movements to changes in the body or the environment, is crucial to our ability to move efficiently in a dynamic world. The field of sensorimotor adaptation is replete with rigorous behavioural and computational methods, which support strong conceptual frameworks. An increasing number of studies have combined these methods with electroencephalography (EEG) to unveil insights into the neural mechanisms of adaptation. We review these studies: discussing EEG markers of adaptation in the frequency and the temporal domain, EEG predictors for successful adaptation and how EEG can be used to unmask latent processes resulting from adaptation, such as the modulation of spatial attention. With its high temporal resolution, EEG can be further exploited to deepen our understanding of sensorimotor adaptation.

Sensorimotor adaptation to wedge prisms can alter the balance of attention between left and right space in healthy adults, and improve symptoms of spatial neglect after stroke. Here we asked whether the orienting of spatial attention to visual stimuli is affected by a different form of sensorimotor adaptation that involves physical perturbations of arm movement, rather than distortion of visual feedback. Healthy participants performed a cued discrimination task before and after they made reaching movements to a central target. A velocity-dependent force field pushed the hand aside during each reach, and required participants to apply compensatory forces toward the opposite side. We used event-related potentials (ERPs) to determine whether electroencephalography (EEG) responses reflecting orienting (cue-locked N1) and disengagement (target-locked P1) of spatial attention are affected by adaptation to force fields. After adaptation, the cue-locked N1 was relatively larger for stimuli presented in the hemispace corresponding to the direction of compensatory hand force. P1 amplitudes evoked by invalidly cued targets presented on the opposite side were reduced. This suggests that force field adaptation boosted attentional orienting responses toward the side of hand forces, and impeded attentional disengagement from that side, mimicking previously reported effects of prism adaptation. Thus, remapping between motor commands and intended movement direction is sufficient to bias ERPs, reflecting changes in the orienting of spatial attention in the absence of visuo-spatial distortion or visuo-proprioceptive mismatch. Findings are relevant to theories of how sensorimotor adaptation can modulate attention, and may open new avenues for treatment of spatial neglect.

Although age-related declines in cognitive, sensory and motor capacities are well documented, current evidence is mixed as to whether or not aging impairs sensorimotor adaptation to a novel dynamic environment. More importantly, the extent to which any deficits in sensorimotor adaptation are due to general impairments in neural plasticity, or impairments in the specific processes that drive adaptation is unclear. Here we investigated whether there are age-related differences in electrophysiological responses to reaching endpoint and trajectory errors caused by a novel force field, and whether markers of error processing relate to the ability of older adults to adapt their movements. Older and young adults (N = 24/group, both sexes) performed 600 reaches to visual targets, and received audio-visual feedback about task success or failure after each trial. A velocity-dependent curl field pushed the hand to one side during each reach. We extracted ERPs time-locked to movement onset [kinematic error-related negativity (kERN)], and the presentation of success/failure feedback [feedback error-related negativity (fERN)]. At a group level, older adults did not differ from young adults in the rate or extent of sensorimotor adaptation, but EEG responses to both trajectory errors and task errors were reduced in the older group. Most interestingly, the amplitude of the kERN correlated with the rate and extent of sensorimotor adaptation in older adults. Thus, older adults with an impaired capacity for encoding kinematic trajectory errors also have compromised abilities to adapt their movements in a novel dynamic environment.

Publisher´s version (útgefin grein)., Whereas aerobic training has found to be beneficial for inhibitory control, less is known on the efficiency of other exercise types in children. The present study compared the effects of aerobic and coordinative training on behavioral and neurophysiological measures of inhibitory control. Forty-five children were randomly assigned (1:1:1 ratio) to groups performing aerobic training, coordinative training or assisted homework sessions over 10 weeks. Before and after intervention, all participants completed a Flanker task. The P300 component of event-related potentials elicited from the task was recorded via electroencephalography. Additionally, aerobic fitness and gross-motor skills were assessed using 20 m Shuttle Run and Heidelberg Gross-Motor Test, respectively. Statistical analyses revealed no time by group interactions for the P300 component (amplitude, latency), p = 0.976, eta(2) = 0.007, and behavioral performance (reaction time, accuracy), p = 0.570, eta(2) = 0.045. In contrast, there was a significant group-difference in pre- to post-test changes in aerobic fitness, p = 0.008, eta(2) = 0.246, with greater improvements following aerobic and coordinative training compared to assisted homework sessions. In conclusion, no differences regarding the efficiency of aerobic and coordinative training for the enhancement of inhibitory control were found as both exercise programs failed to elicit changes in speed and accuracy of stimulus evaluation and the allocation of attentional resources., The German Research Foundation (BU 1837/5-2). It is well-established that expertise developed through continuous and deliberate practice has the potential to delay age-related decline in fine motor skills. However, less is known about the underlying mechanisms, that is, whether expertise leads to a higher performance level changing the initial status from which age-related decline starts or if expertise-related changes result in qualitatively different motor output and neural processing providing a resource of compensation for age-related changes. Thus, as a first step, this study aims at a better understanding of expertise-related changes in fine motor control with respect to force output and respective electrophysiological correlates. Here, using a multidimensional approach, we investigated fine motor control of experts and novices in precision mechanics during the execution of a dynamic force control task. On the level of force output, we analyzed precision, variability, and complexity. We further used dynamic mode decomposition (DMD) to analyze the electrophysiological correlates of force control to deduce brain network dynamics. Experts' force output was more precise, less variable, and more complex. Task-related DMD mean mode magnitudes within the α-band at electrodes over sensorimotor relevant areas were reduced in experts, and lower DMD mean mode magnitudes related to the force output in novices. Our results provide evidence for expertise dependent central adaptions with distinct and more complex organization and decentralization of sensorimotor subsystems. Results from our multidimensional approach can be seen as a step forward in understanding expertise-related changes and exploiting their potential as resources for healthy aging.

Visuomotor adaptation involves the learning of a new mapping between a spatial goal and well-learned movements. In order to learn a new visuomotor transformation, visual attention is needed to monitor movements and their visual consequences. Once a transformation is learnt, it can be executed automatically without attentional control. Using steady-state visual evoked potentials (SSVEPs) measured from EEG activity, we examined how visual attention changes during the early phase of visuomotor adaptation. SSVEPs were elicited by a green disc flickering at 15 Hz which was either the movement target or the cursor that participants controlled. Participants performed an adapted continuous visuomotor adaptation task with either 60° or 120° screen cursor rotation, and changes in 15-Hz SSVEP power were examined. Participants' performance improved over time in all conditions, with the rate of learning significantly influenced by the degree of rotation. SSVEPs at 15 Hz showed a significant change over time with adaptation for 60° rotations, but not for 120° rotations, such that SSVEPs elicited by the stimuli were significantly lower for 60° compared with 120° rotation conditions over the last adaptation blocks. This suggests that visual attention to the movement target and feedback reduces over time as performance improves during visuomotor adaptation for easier rotations, but must be maintained throughout the task for more difficult 120° rotations that might require more strategic control.

The pure exposure to extensive tactile stimulation, without the requirement of attention or active training, has been revealed to enhance sensorimotor functioning presumably due to an induction of plasticity in the somatosensory cortex. The induced effects, including increased tactile acuity and manual dexterity have repeatedly been observed in basic as well as clinical research. However, results vary greatly in respect to the strength and direction of the effects on the behavioral and on the brain level. Multiple evidences show that differences in the stimulation protocols (e.g., two vs. multiple stimulation sites) and parameters (e.g., duration, frequency, and amplitude) might contribute to this variability of effects. Nevertheless, stimulation protocols have not been comprehensively compared yet. Identifying favorable parameters for tactile stimulation interventions is especially important because of its possible application as a treatment option for patients suffering from sensory loss, maladaptive plasticity, or certain forms of motor impairment. This review aims to compare the effects of different tactile stimulation protocols and to assess possible implications for tactile interventions. Our goal is to identify ways of optimizing stimulation protocols to improve sensorimotor performance. To this end, we reviewed research on tactile stimulation in the healthy population, with a focus on the effectiveness of the applied parameters regarding psychophysiological measures. We discuss the association of stimulation-induced changes on the behavioral level with alterations in neural representations and response characteristics.

The authors examined age-related differences in fine motor control during a bimanual coordination task. The task required the modulation of fingertip forces in the precision grip according to a visually presented sinusoidal antiphase pattern (force range 2-12 N; frequency 0.2 Hz). Thirty-four right-handed participants of three age groups (young, early middle-aged, and late middle-aged) practiced 30 trials of the task. Accuracy and variability of relative timing and relative forces at minima and maxima of the sine wave were analyzed for hand-hand and hand-stimulus couplings and compared between age groups. Analysis showed for relative timing and force weaker hand-hand than hand-stimulus coupling as well as lower accuracy and higher variability for minima as compared to maxima. Further, we analyzed practice effects by comparing the first and last trials and characterized the course of practice by detecting the transition of a steeper to a shallower acquisition slope for the different age groups. Late middle-aged participants demonstrated poorer performance than both other groups for all parameters. All groups improved performance to a similar amount. However, an age-related difference in acquisition strategy is visible. Late middle-aged participants seemed to have focused on improvement of force amplitude, whereas young and early middle-aged focused on timing.

From the conceptual and methodological framework of the dynamical systems approach, force control results from complex interactions of various subsystems yielding observable behavioral fluctuations, which comprise both deterministic (predictable) and stochastic (noise-like) dynamical components. Here, we investigated these components contributing to the observed variability in force control in groups of participants differing in age and expertise level. To this aim, young (18–25 yr) as well as late middle-aged (55−65 yr) novices and experts (precision mechanics) performed a force maintenance and a force modulation task. Results showed that whereas the amplitude of force variability did not differ across groups in the maintenance tasks, in the modulation task it was higher for late middle-aged novices than for experts and higher for both these groups than for young participants. Within both tasks and for all groups, stochastic fluctuations were lowest where the deterministic influence was smallest. However, although all groups showed similar dynamics underlying force control in the maintenance task, a group effect was found for deterministic and stochastic fluctuations in the modulation task. The latter findings imply that both components were involved in the observed group differences in the variability of force fluctuations in the modulation task. These findings suggest that between groups the general characteristics of the dynamics do not differ in either task and that force control is more affected by age than by expertise. However, expertise seems to counteract some of the age effects.NEW & NOTEWORTHY Stochastic and deterministic dynamical components contribute to force production. Dynamical signatures differ between force maintenance and cyclic force modulation tasks but hardly between age and expertise groups. Differences in both stochastic and deterministic components are associated with group differences in behavioral variability, and observed behavioral variability is more strongly task dependent than person dependent.

Repetitive tactile stimulation is a well-established tool for inducing somatosensory cortical plasticity and changes in tactile perception. Previous studies have suggested that baseline performance determines the amount of stimulation-induced learning differently in specific populations. Older adults with lower baseline performance than young adults, but also experts, with higher baseline performance than non-experts of the same age, have been found to profit most from such interventions. This begs the question of how age-related and expertise-related differences in tactile learning are reflected in neurophysiological correlates. In two experiments, we investigated how tactile learning depends on age (experiment 1) and expertise (experiment 2). We assessed tactile spatial and temporal discrimination accuracy and event-related potentials (ERPs) in 57 persons of different age and expertise groups before and after a 30-min tactile stimulation intervention. The intervention increased accuracy in temporal (found in experiment 1) and spatial (found in experiment 2) discrimination. Experts improved more than non-experts in spatial discrimination. Lower baseline performance was associated with higher learning gain in experts and non-experts. After the intervention, P300 latencies were reduced in young adults and amplitudes were increased in late middle-aged adults in the temporal discrimination task. Experts showed a steeper P300 parietal-to-frontal gradient after the stimulation. We demonstrated that tactile stimulation partially reverses the age-related decline in late middle-aged adults and increases processing speed in young adults. We further showed that learning gain depends on baseline performance in both non-experts and experts. In experts, however, the upper limit for learning seems to be shifted to a higher level.

Salient and sudden sensory events generate a remarkably large response in the human brain, the vertex wave (VW). The VW is coupled with a modulation of a voluntarily-applied isometric force. In the present study, we tested whether the VW is also related to executing high-precision movements. The execution of a voluntary high-precision movement remains relatively independent of the brain activity reflected by the preceding VW. The apparent relationship between the positive VW and movement onset time is explained by goal-related but stimulus-independent neural activities. These results highlight the need to consider such goal-related but stimulus-independent neural activities when attempting to relate event-related potential amplitude with perceptual and behavioural performance.Salient and fast-rising sensory events generate a large biphasic vertex wave (VW) in the human electroencephalogram (EEG). We recently reported that the VW is coupled with a modulation of concomitantly-applied isometric force. In the present study, in five experiments, we tested whether the VW is also related to high-precision visuomotor control. We obtained three results. First, the saliency-induced increase in VW amplitude was paralleled by a modulation in two of the five extracted movement parameters: a reduction in the onset time of the voluntary movement (P 0.005) and an increase in movement accuracy (P 0.005). Second, spontaneous trial-by-trial variability in vertex wave amplitude, for a given level of stimulus saliency, was positively correlated with movement onset time (P 0.001 in four out of five experiments). Third, this latter trial-by-trial correlation was explained by a widespread EEG negativity independent of the occurrence of the positive VW, although overlapping in time with it. These results indicate that (i) the execution of a voluntary high-precision movement remains relatively independent of the neural processing reflected by the preceding VW, with (ii) the exception of movement onset time, for which saliency-based contextual effects are dissociated from trial-by-trial effects. These results also indicate that (iii) attentional effects can produce spurious correlations between event-related potentials (ERPs) and behavioural measures. Although sudden salient stimuli trigger characteristic EEG responses coupled with distinct reactive components within an ongoing isometric task, the results of the present study indicate that the execution of a subsequent voluntary movement appears largely protected from such saliency-based modulation, with the exception of movement onset time.

Laser-accelerated electron pulses have been used to irradiate human tumors grown on mice’s ears during radiobiological experiments. These experiments have been carried out with the JETI laser system at the Institute of Optics and Quantum Electronics in Jena, Germany. To treat a total of more than 50 mice, a stable and reliable operation of the laser-electron accelerator with a dose rate exceeding 1 Gy/min was necessary. To achieve this, a sufficient number of electrons at energies in excess of 5 MeV had to be generated. The irradiation time for a single mouse was a few minutes. Furthermore, the particle pulses’ parameters needed to remain achievable for a time period of several weeks. Due to the online detection of the radiation dose, the unavoidable shot-to-shot fluctuations, currently still typical for laser-based particle accelerators, could be compensated. The results demonstrate that particle pulses generated with laser-based accelerators have the potential to be a future alternative for conventional particle accelerators used for the irradiation of tumors.

Performance of a task carried out with two hands separately is better than the performance of the same task done with both hands at the same time. This so-called bilateral deficit may be reduced or counteracted by long-term practice. Little is known about age-related changes. We examined age- and expertise-related differences in the bilateral deficit in force initiation. Participants performed static and dynamic force modulation tasks either with the right and left hand separately or both hands simultaneously. In order to examine age-related differences, we compared novices of fine motor control (service employees) from three age groups, covering the working age (young n = 13, early middle-aged n = 10 and late middle-aged n = 12). To assess the influence of expertise, we considered precision mechanics as experts in fine motor control. To ensure the acquisition of expertise, only early middle-aged (n = 10) and late middle-aged (n = 14) experts were recruited. Regardless of the task, bimanual force initiation was slower than unimanual force initiation. This bilateral deficit was (1) more pronounced in the static than in the dynamic task, (2) higher in early and late middle-aged than in younger novices, and (3) lower in experts as compared to novices. Based on our results, we assume both interhemispheric inhibition and division of attention to contribute to the bilateral deficit and the expertise- and age-related differences, respectively. The results are promising for the possibility to overcome constraints of bilateral hand movements by long-term practice.

Tactile perception declines with age on both behavioral and neurophysiological levels. Less well understood is how neurophysiological changes relate to tactile discrimination performance in middle adulthood. A tactile discrimination task was conducted while ERPs were measured in three groups of healthy adults aged 20 to 66 years. Accuracy was lowest in late middle adulthood (56-66 years) while somatosensory ERP components (P50, N70, P100, N140) were comparable across age groups. The cognitive P300 revealed age-related differences in scalp distribution typical for older adults to already be present in late middle adulthood. Increased recruitment of frontal cognitive processes was positively related to performance in later middle adulthood. Our results further the understanding of age-related differences in tactile perception during middle adulthood and the importance of cognitive processes to compensate for age-related decline. We report on results from the fully diode-pumped chirped-pulse amplification laser system Polaris. Pulses were amplified to a maximum energy of 54.2 J before compression. These pulses have a full width at half-maximum spectral bandwidth of 18 nm centered at 1033 nm and are generated at a repetition rate of 0.02 Hz. To the best of our knowledge, these are the most energetic broadband laser pulses generated by a diode-pumped laser system so far. Due to the limited size of our vacuum compressor, only attenuated pulses could be compressed to a duration of 98 fs containing an energy of 16.7 J, which leads to a peak power of 170 TW. These pulses could be focused to a peak intensity of 1.3×1021 W/cm2. Having an ultra-high temporal contrast of 1012 with respect to amplified spontaneous emission these laser pulses are well suited for high-intensity laser–matter experiments.

There are well-documented differences in the way that people typically perform identical motor tasks with their dominant and the nondominant arms. According to Yadav and Sainburg's ( Neuroscience 196: 153–167, 2011) hybrid-control model, this is because the two arms rely to different degrees on impedance control versus predictive control processes. Here, we assessed whether differences in limb control mechanisms influence the rate of feedforward compensation to a novel dynamic environment. Seventy-five healthy, right-handed participants, divided into four subsamples depending on the arm (left, right) and direction of the force field (ipsilateral, contralateral), reached to central targets in velocity-dependent curl force fields. We assessed the rate at which participants developed predictive compensation for the force field using intermittent error-clamp trials and assessed both kinematic errors and initial aiming angles in the field trials. Participants who were exposed to fields that pushed the limb toward ipsilateral space reduced kinematic errors more slowly, built up less predictive field compensation, and relied more on strategic reaiming than those exposed to contralateral fields. However, there were no significant differences in predictive field compensation or kinematic errors between limbs, suggesting that participants using either the left or the right arm could adapt equally well to novel dynamics. It therefore appears that the distinct preferences in control mechanisms typically observed for the dominant and nondominant arms reflect a default mode that is based on habitual functional requirements rather than an absolute limit in capacity to access the controller specialized for the opposite limb.

It has been repeatedly shown that precise finger force control declines with age. The tasks and evaluation parameters used to reveal age-related differences vary between studies. In order to examine effects of task characteristics, young adults (18-25 years) and late middle-aged adults (55-65 years) performed precision grip tasks with varying speed and force requirements. Different outcome variables were used to evaluate age-related differences. Age-related differences were confirmed for performance accuracy (TWR) and variability (relative root mean square error, rRMSE). The task characteristics, however, influenced accuracy and variability in both age groups: Force modulation performance at higher speed was poorer than at lower speed and at fixed force levels than at force levels adjusted to the individual maximum forces. This effect tended to be stronger for older participants for the rRMSE. A curve fit confirmed the age-related differences for both spatial force tracking parameters (amplitude and intercept) and for one temporal parameter (phase shift), but not for the temporal parameter frequency. Additionally, matching the timing parameters of the sine wave seemed to be more important than matching the spatial parameters in both young adults and late middle-aged adults. However, the effect was stronger for the group of late middle-aged, even though maximum voluntary contraction was not significantly different between groups. Our data indicate that changes in the processing of fine motor control tasks with increasing age are caused by difficulties of late middle-aged adults to produce a predefined amount of force in a short time.

This study investigates the association between physical activity level and attentional control in a group of younger and older middle-aged adults (MA). Participants performed a Flanker task; two types of conflict were analyzed: response and perceptual conflict. For perceptual conflict, behavioral findings suggest that, irrespective of age, physical activity is positively associated with attentional control. For response conflicts, only highly active younger MA showed better attentional control, indicated by increased amplitudes of the event related potential N2 and reduced interference costs by distracting information. Physical activity did not modulate P3 amplitudes. The findings are discussed with respect to physical activity as functional approach to maintain cognitive functioning across the lifespan.

Age-related decline of fine motor control commences even in middle adulthood. Less is known, however, whether age-related changes can be postponed through continuous practice. In this study we tested how age and professional expertise influence fine motor control in middle-aged adults. Forty-eight right-handed novices and experts (35 to 65 years) performed submaximal precision grip force modulation tasks with index or middle finger opposing the thumb, either with the right hand or the left hand. Novices revealed expected age-related differences in all performance measures (force initialization, mean applied force, variability), whereas experts outperformed novices in all outcome measures. Expertise seems to contribute to maintaining manual skills into older age, as indicated by the age and expertise interaction for the force initialization.

We present few-femtosecond shadowgraphic snapshots taken during the non-linear evolution of the plasma wave in a laser wakefield accelerator with transverse synchronized few-cycle probe pulses. These snapshots can be directly associated with the electron density distribution within the plasma wave and give quantitative information about its size and shape. Our results show that self-injection of electrons into the first plasma wave period is induced by a lengthening of the first plasma period. The Nemertodermatida are a small group of microscopic marine worms. Recent molecular studies have demonstrated that they are likely to be the earliest extant bilaterian animals. What was the nervous system (NS) of a bilaterian ancestor like? In order to answer that question, the NS of Nemertoderma westbladi was investigated by means of indirect immunofluorescence technique and confocal scanning laser microscopy. The antibodies to a flatworm neuropeptide GYIRFamide were used in combination with anti-serotonin antibodies and phalloidin-TRITC staining. The immunostaining revealed an entirely basiepidermal NS. A ring lying outside the body wall musculature at the level of the statocyst forms the only centralisation, the “brain”. No stomatogastric NS has been observed. The GYIRFamide immunoreactive part of the “brain” is formed of loosely packed nerve fibres with multiple small neurones and a few large ones. The peptidergic and aminergic patterns of the NS do not correspond to each other: the former is more developed on the ventral side, the latter is more pronounced on the dorsal side. A pair of GYIRFamide immunoreactive nerve cords innervates the ventral side of the animal, the mouth and the male genital opening. The nemertodermatids studied to-date display no common NS pattern. Possible synapomorphies of the Acoelomorpha are discussed. The study demonstrates that the nemertodermatid NS possesses a number of plesiomorphic features and appears more primitive than the NS in other worms, except the Xenoturbellida. The bilaterian ancestor supposedly possessed only a basiepidermal nerve net and had no centralised brain-like structures and no stomatogastric NS.

The phenomenon of asexual multiplication is rare in the animal kingdom, but it occurs in all main flatworm taxa. In the present paper, we review data regarding the presence of different forms of asexual multiplication in flatworms and argue that the presence of a population of totipotent or pluripotent stem cells, "neoblasts", is a primitive feature of decisive importance for the developing potential of flatworms. Next we present information on the role of stem cells in fission, head regeneration, and pharynx regeneration of planarians. Furthermore, the tracing of neoblasts in lower flatworms and cestodes is presented, and the results indicating heterogeneity of the neoblast pool are discussed. Finally, the mode by which the neoblasts are stimulated to divide, migrate, and differentiate and the nature of the interactions are discussed. We focus on (i) biogenic amines and neuropeptides, (ii) the role of neuropeptides in the early stage of regeneration, (iii) the evidence for the influences of growth factors and nitric oxide, and (iv) the influence of weak electromagnetic fields. We discuss the pattern in which a gradient system of morphogens and (or) a hierarchical system of inductions is expressed in development.

Raikova, O. I., Reuter, M., Gustafsson, M. K. S., Maule, A. G., Halton, D. W. & Jondelius, U. (2004). Evolution of the nervous system in Paraphanostoma (Acoela). —Zoologica Scripta, 33, 71–88. According to recent molecular studies, the Acoela are the earliest extant bilaterian group. Their nervous system displays a striking variety of patterns. The aim of the present investigation was to study the variability of the nervous system in a monophyletic group of the Acoela. Six species of Paraphanostoma were chosen for the study. Using immunocytochemical methods and confocal scanning laser microscopy, the immunoreactive patterns of serotonin (5-HT) and the neuropeptide GYIRFamide were described in detail. The study has demonstrated that the brains in Paraphanostoma species, although diverse in detail, still follow the same general pattern. 18S rDNA sequences were used to generate a hypothesis of the phylogeny within the group. Characters of the nervous system revealed in this study were coded and analysed together with 18S rDNA data. Several synapomorphies in the nervous system characters were identified. However, numerous parallelisms in the nervous system evolution have occurred. Data obtained demonstrate that the genus Paraphanostoma is closely related to Childia and should belong to the same family, Childiidae.

Efficient motor control requires motor planning. Age-related changes in motor control are well described, e.g. increased movement variability and greater antagonistic muscle co-activation, as well as less functional and less regional specific brain activation. However, less is known about age-related changes in motor planning. By use of transcranial magnetic stimulation we investigated differences in corticomotor facilitation during motor planning in 17 young (25 ± 3 years) and 17 older healthy adults (70 ± 13 years) in a delayed movement paradigm for wrist movements. Motor evoked potentials (MEPs) were recorded for the flexor and extensor carpi radialis during movement preparation of wrist flexion and extension as well as during rest. We found that MEPs were less specifically facilitated during planning in older as compared to younger adults, as indicated by an Age × Condition × Muscle interaction. Young participants showed significantly facilitated MEPs in the respective muscle needed for wrist flexion or extension. By contrast MEPs in older participants were less specifically modulated. We conclude that age relates to dedifferentiated activation of the primary motor cortex already during preparation of distinct movements which might contribute to less efficient motor control in older adults.

The use of well-characterized antibodies raised to neuronal signal substances and their application through immunocytochemistry and confocal scanning laser microscopy has revolutionized studies of the flatworm nervous system (NS). Data about flatworm neuropeptides and the spatial relationship between neuropeptides and other neuronal signal substances and muscle fibers are presented. Neuropeptides form a large part of the flatworm NS. Neuropeptides are especially important as myoexcitatory transmitters or modulators, controlling the musculature of the attachment organs, the stomatogastric and the reproductive systems.

In order to broaden the information about the organisation of the nervous system in the Plathelminthes, an immunocytochemical (ICC) study of the free-living flatworm Castrella truncata (Dalyellioida) has been performed. This is the first time a representative of the taxon Rhabdocoela has been studied with the ICC technique. Antibodies to 5-HT and FMRF-amide and confocal scanning laser microscope were used. 5-HT and FMRF-amide immunoreactivity was observed in the neuropile of the brain, in the cerebral neurones, in the three pairs of longitudinal nerve cords and the adjoining neurones, in the pharyngeal nerve ring and in twelve neurones associated with the pharynx. 5-HT and FMRF-amide immunoreactivity occurs in separate sets of neurones. Only FMRF-amide immunoreactivity was observed in a peculiar ellipse-shaped structure in the brain, and in the genital plexus. The type of orthogon is discussed. Staining of the muscular system with TRITC conjugated phalloidin revealed muscle patterns that have not been described previously.

The long-term goal to integrate laser-based particle accelerators into radiotherapy clinics not only requires technological development of high-intensity lasers and new techniques for beam detection and dose delivery, but also characterization of the biological consequences of this new particle beam quality, i.e. ultra-short, ultra-intense pulses. In the present work, we describe successful in vivo experiments with laser-driven electron pulses by utilization of a small tumour model on the mouse ear for the human squamous cell carcinoma model FaDu. The already established in vitro irradiation technology at the laser system JETI was further enhanced for 3D tumour irradiation in vivo in terms of beam transport, beam monitoring, dose delivery and dosimetry in order to precisely apply a prescribed dose to each tumour in full-scale radiobiological experiments. Tumour growth delay was determined after irradiation with doses of 3 and 6 Gy by laser-accelerated electrons. Reference irradiation was performed with continuous electron beams at a clinical linear accelerator in order to both validate the dedicated dosimetry employed for laser-accelerated JETI electrons and above all review the biological results. No significant difference in radiation-induced tumour growth delay was revealed for the two investigated electron beams. These data provide evidence that the ultra-high dose rate generated by laser acceleration does not impact the biological effectiveness of the particles.

Tactile expertise, resulting from extensive use of hands, has previously been shown to improve tactile perception in blind people and musicians and to be associated with changes in the central processing of tactile information. This study investigated whether expertise, due to precise and deliberate use of the fingers at work, relates to improved tactile perception and whether this expertise interacts with age. A tactile pattern and a frequency discrimination task were conducted while ERPs were measured in experts and nonexperts of two age groups within middle adulthood. Independently of age, accuracy was better in experts than in nonexperts in both tasks. Somatosensory N70 amplitudes were larger with increasing age and for experts than for nonexperts. P100 amplitudes were smaller in experts than in nonexperts in the frequency discrimination task. In the pattern discrimination task, P300 difference wave amplitude was reduced in experts and late middle-aged adults. In the frequency discrimination task, P300 was more equally distributed in late middle-aged adults. We conclude that extensive, dexterous manual work leads to acquisition of tactile expertise and that this expertise might delay, but not counteract, age effects on tactile perception. Comparable neurophysiological changes induced by age and expertise presumably have different underlying mechanisms. Enlarged somatosensory N70 amplitudes might result from reduced inhibition in older adults but from enhanced, specific excitability of the somatosensory cortex in experts. Regarding P300, smaller amplitudes might indicate fewer available resources in older adults and, by contrast, a reduced need to engage as much cognitive effort to the task in experts.

By combining phalloidin-TRITC staining with confocal scanning laser microscopy (CSLM), the pattern of the musculature in two species of Rotifera, Euchlanis dilatata unisetata and Brachionus quadridentatus is revealed. The same general muscle pattern prevails in both species. The major components of the body wall musculature are: 1. retractor muscles (5 pairs in E. dilatata unisetata and 3 pairs in B. quadridentatus); 2. Two pairs of dorso-ventral muscles; 3. Two pairs of perpendicular muscles (in E. dilatata unisetata); 4. retractors of the corona (median, lateral and ventral); 5. Foot retractors. In addition, three pairs of cutaneo-visceral muscles and visceral muscles (including mastax muscles) are described. The sphincter of the corona was found only in B. quadridentatus. The high degree of muscle differentiation points to a high level of development of rotifer muscular system.

The phylogenetic position of the Xenoturbellida is highly disputed. Are they primitive flatworms? Are they related to Deuterostomia? Do they form a sister taxon to other Bilateria? Are they bivalve molluscs? In order to provide more data for this discussion, a study of the nervous system of Xenoturbella westbladi and its relation to the musculature was performed, using 5-HT and FMRFamide immunocytochemistry, TRITC-conjugated phalloidin fluorescence for staining of F-actin filaments, confocal scanning laser microscopy and transmission electron microscopy. The nervous system comprises solely an intraepidermal net of nerve cells and processes. No ganglia or any other internal nervous structures could be detected. No evidence of 5-HT- or FMRFamide-immunoreactive innervation below the subepidermal membrane complex was obtained. The 5-HT and FMRFamide immunoreactivity occurs in separate sets of neurones. On the ultrastructural level, three types of neurones were observed: (1) the predominating ”light” neurones, (2) the smaller ”dark” neurones and (3) the bipolar sensory neurones bearing a single cilium with a long bipartite rootlet. Non-synaptic, paracrine, release sites are common and synapses are inconspicuous. In the layer of epidermal cells, close to the lateral furrow, F-actin filaments were observed. They reach from the basal membrane to the surface. The organisation of the nervous system appears very simple. Our results are compatible with the hypothesis of Xenoturbellida forming a sister taxon to Bilateria. No evidence was obtained for inclusion of X. westbladi in either the Mollusca or Plathelminthes.

This is the first detailed description of the nitrergic nervous system in a fluke. In this study, the authors analysed the distribution of the nicotinamide adenine dinucleotide phosphatediaphorase (NADPH-d) reactivity in neuronal and nonneuronal tissues of the adult fluke Fasciola hepatica and compared this with the distribution of the musculature using tetramethylrhodamine isothiocyanate-phalloidin. To assess the correlation between the number of muscle cells in different parts of the fluke and the NADPH-d-stained cells, the nuclei were stained with Hoechst 333 42, which is specific for chromatin. The spatial relation between the NADPH-d-positive nerves and the 5-hydroxytryptamine (serotonin; 5-HT)-immunoreactive (-IR) and GYIRFamide-IR nervous elements was also examined. The methods complement each other. NADPH-d-positive staining occurs in both in neuronal tissue and nonneuronal tissue. Large, NADPH-d-stained neurones were localised in the nervous system. The oral and ventral suckers are innervated with many large NADPH-d-stained neurones. In addition, the NADPH-d staining reaction follows closely the muscle fibres in both the suckers, in the body, and in the ducts of the reproductive organs. The presence of NADPH-d activity along muscle fibres in F. hepatica and in other flatworms supports a possible myoinhibitory role for nitric oxide. In order to test the role of nitric oxide in flatworms, Mesocestoides vogae tetrathyridia were incubated together with l -arginine, which is the substrate for nitric oxide synthesis, or with NG-nitro- l -arginine, which is an irreversible inhibitor of nitric oxide synthase. Normally, tetrathyridia attach to each other with the aid of their suckers, forming clusters. The rate of cluster formation was followed during the incubations. l -Arginine stimulated, and NG-nitro- l -arginine clearly inhibited, the cluster formation. This is the first time that an effect of nitric oxide has been observed in a flatworm. In addition, the pattern of the NADPH-diaphorase histochemical reaction in the nervous system and the pattern of F-actin filaments in the musculature stained with TRITC-labelled phalloidin were studied. NADPH-d staining occurred in the brain and the main nerve cords but also followed the muscle fibres stained with phalloidin. The pattern of the NADPH-d reaction was compared with that of 5-HT immunoreactivity. The implications of the results are discussed in relation to the background of data on neuronal signal substances in M. vogae.

Summary The study shows that the regenerating pharynx of Girardia tigrina forms a simple and valuable model system for studies of pattern formation in the nervous system and its relationship to the developing muscles. Immunocytochemistry was used, with antisera raised against 5-HT and the native planarian neuropeptide GYIRFamide. We studied the sequential development of the innervation in the regenerating pharynx, and using TRITC-labelled phalloidin we followed the corresponding differentiation and maturation of pharynx musculature. The peptidergic and aminergic neuronal cell types develop according to different time schedules and different modes. Throughout the process, the GYIRFarnide-IR elements are in contact with the old parts of the nervous system, while the apical 5-HT-IR elements develop de novo. In the regenerating pharynx the GYIRFamide-IR nerves develop in a proximodistal direction. The first 5-HT-IR cell bodies appear in the tip of the pharynx and are symmetrically placed. They have no contact to the rest of the nervous system. From these cell bodies processes grow disto-proximally and fasciculate with tibres from the proximal part. A striking parallelism between the appearance of GYIRFamide-IR nerves and muscle fibres stained with TRITC-phalloidin was observed. The GYIRFamide-IR nerves cling to the muscle fibres. These results suggest that the contact between muscle Iibres and GYIRF-IR nerves is essential for the function of pharynx. The delayed appearance of 5-HT-IR nervous elements is discussed in terms of the influence of 5-HT on sprouting of nerve fibres and synapse formation. The development of the pharynx in tail parts after fission is compared with that after pharynx amputation. The faster rate observed for pharynx regenerating after amputation in comparison with that in regenerating tail parts indicates the importance of the remains of the old nervous system in the pharynx structure.

In order to solve the question whether Plathelminthes belonging to the taxon Acoela have a brain and an orthogon of the common flatworm type, an immunocytochemical study of the pattern of serotonin (5-HT) in four species was performed. In all species the 5-HT immunoreactivity revealed no ganglionic cell mass typical for other Plathelminthes, only a symmetrical brain-like structure composed of commissural fibres associated with a few cell bodies. 5-HT immunoreactivity was detected in three to five pairs of longitudinal nerve cords, connected by an irregular network of immunoreactive transverse fibres. No regular orthogon was visualised. All the surface sensilla were strongly immunoreactive. The unique commissural brain and cordal nervous system found in Acoela support the view of a deep gap lying between Acoela and other Plathelminthes.
