Nanoparticles (NPs) coated with hyaluronic acid (HA) seem to be increasingly promising for targeted therapy due to HA chemical versatility, which allows them to bind drugs of different natures, and their affinity with the transmembrane receptor CD-44, overexpressed in tumor cells. However, an essential aspect for clinical use of NPs is formulation stability over time. For these reasons, analytical techniques capable of characterizing their physico-chemical properties are needed. In this work, poly(lactide-co-glycolide) (PLGA) NPs with an average diameter of 100–150 nm, coated with a few 10 s of nm of HA, were synthesized. For stability characterization, two complementary investigative techniques were used: Dynamic Light Scattering (DLS) and Surface-Enhanced Raman Scattering (SERS) spectroscopy. The first technique provided information on size, polidispersity index, and zeta-potential, and the second provided a deeper insight on the NP surface chemicals, allowing distinguishing of HA-coated NPs from uncoated ones. Furthermore, in order to estimate formulation stability over time, NPs were measured and monitored for two weeks. SERS results showed a progressive decrease in the signal associated with HA, which, however, is not detectable by the DLS measurements.

For a healthy indoor environment, it is important to understand which materials and factors favor the generation of high levels of indoor radon. A preliminary multivariate statistical analysis was carried out on two datasets concerning indoor radon and building materials in the Campania Region using Principal Component Analysis (PCA) and the k-means partitional analysis technique. A total of 13 parameters related to building materials were used. The results show the greater contribution of building materials of volcanic origin to the concentration of indoor radon and thoron activity and the different influence of the parameters of the 31 materials analyzed. The same analyses applied to the indoor radon values of 694 rooms in the Campania Region were equally effective in assessing the structural characteristics of indoor environments that most influence indoor radon levels. The study provided an effective assessment of the influence on radon activity of several environmental parameters, which are often not adequately considered.

The effectiveness of the increase in passive ventilation was studied as remedial action for high concentrations of indoor radon gas activity. The rationale has been to apply the provisions of Directive 2013/59/EURATOM, partially implemented by a regional law of Puglia Region (Southern Italy). Residential buildings were selected based on characteristics common to those of commercial premises, which are the recipients of legislative obligations. The final goal of this study was to evaluate the feasibility of carrying out the same remediation activity for both types of buildings. From a previous survey in Puglia in 2013–2014, 18 houses were selected and passive ventilation strategy was applied in order to evaluate its effect on the radon activity concentration. A specially designed protocol defined frequency and time of windows operation. The average value of the annual measurement revealed a significant reduction of the internal radon activity concentration: mitigation ranged from 21 ± 5% to 58 ± 2% with an average value of 33 ± 3%. The results of the study showed that passive ventilation is an effective method of mitigation of the internal radon concentration if correctly designed according to the local characteristics of the buildings and habits of the occupants.

The success of future long-term manned missions in Space is strictly related to the development and setup of sustainable closed artificial ecosystems, defined as Bioregenerative Life Support Systems (BLSSs). In these BLSSs, the cultivation of higher plants represents the core element, given its role in primary resources (e.g., oxygen, water) regeneration and in fresh food production, also helping to counteract the emergence of Space-induced diseases in astronauts. However, Space farming may be constrained by Space ionizing radiation for its influence on plant growth and biomass production, depending on radiation properties and plant intrinsic factors (e.g., type of radiation, dose, plant species, cultivar, developmental stage at the time of irradiation). This study aimed to evaluate the effects of the different types and doses of low and high-LET (Linear Energy Transfer) radiation on morpho-anatomical traits and antioxidant content of Brassica rapa L. subsp. sylvestris var. esculenta microgreens. Dry seeds were exposed to different doses (0-control, 0.3, 1, 10, 20, and 30 Gy) of X-rays (the reference Low-LET radiation) at Istituto Nazionale Tumori - IRCCS - Fondazione G. Pascale, and to two types of Heavy Ions, namely 12C and 56Fe (to simulate the High-LET radiation) at different doses (0-control, 0.3, 1, 10, 20, and 25 Gy) at GSI Helmholtzzentrum für Schwerionenforschung GmbH. After the irradiation, seeds were sowed and microgreens were cultivated in a growth chamber under controlled environmental conditions, monitoring germination and survival, as well as growth performances. At the harvest, morpho-biometric traits, such as stem elongation, fresh and dry biomass, and total leaf area were measured. Leaf functional anatomical traits (e.g., lamina thickness, localization of phenolics, stomatal frequency) were quantified through light and epifluorescence microscopy and digital image analysis. The antioxidant charge of microgreens was evaluated considering antioxidant capacity, and polyphenols, chlorophylls, and carotenoid content. Results highlighted that the responses of microgreens, from irradiated seeds, were dose and radiation-type specific. This study furnished useful insights to evaluate plant radio-resistance at early stages of development, namely seedling establishment, that is a very critical phase in the plant life cycle. Gained information is useful to support the decision actions about the choice of suitable species to be cultivated in the BLSSs in Space and the definition of the shielding requirements for Space cultivation facilities.Part of the results presented here is based on the experiment Bio_08_DeMicco, which was performed at the SIS18 at the GSI Helmholtzzentrum fuer Schwerionenforschung, Darmstadt (Germany) in the frame of FAIR Phase-0.

Space exploration always has fascinated scientists, and in the last decades, the interest in this goal has increased exponentially. However, the realization of crewed space exploration missions or the permanence of human settlements on orbital stations or planetary habitats requires Life Support Systems (LSS) based on the interaction among abiotic and biotic elements, resembling terrestrial ecosystems. As on Earth, plants in Space would be involved in air regeneration through photosynthesis, water recovery, waste processing and food production, providing the astronauts with renewed resources and fresh food.The extraterrestrial environment may be considered one the most extreme for organisms’ survival because of several Space factors constraining biological life (e.g., altered gravity, fluid-dynamic and microgravity interaction, modified pressure, temperature). Among them, ionizing radiation influences severely plant growth and metabolic processes. Our study explores the response of tomato (Solanum lycopersicum L. ‘Microtom’), one of most widely cultivated crops, to different doses of sparsely ionizing radiation (IR), with two main goals: 1) to evaluate the morpho-functional mechanisms conferring radioresistance to guarantee their ecological role in closed, controlled environments; 2) to assess the possibility to use IR as a “biostimulant” to produce plant-derived functional food richer in functional compounds.Different doses of X-rays (0.3, 1, 10, 20, and 30 Gy, 6 MV energy), were delivered on dry seeds (DS) and germinated seeds (GS) and compared to not-irradiated controls, to define a dose-response curve and check possible negative/positive outcomes on seedlings. After the irradiation treatments, seeds/seedlings were transferred to a climatic chamber and cultivated under controlled environmental conditions of light, temperature, relative humidity and photoperiod. For our study, we adopted a multidisciplinary approach that merges anatomical analyses with measurements of photosynthetic efficiency and biochemical traits, including polyphenols and other antioxidant compounds linked with the nutritional value of derived food. The growth and photosynthetic performance of DS and GS plants were followed during the whole plant life cycle, while anatomical traits and fruit antioxidant properties were determined at full maturity of the specific organs. The results showed that the outcomes of radiation are dose-specific and dependent on the irradiated target stage, being SS plants more high-performing in photosynthetic activity and antioxidant content in fruits than SGs. Furthermore, some doses of X-rays act as a booster of bioactive compounds in fruits of both SS and SG plants. The outcomes of this research will be helpful to optimize crop production in Space and controlled environment agriculture systems. Moreover, the fine analysis of the relations between anatomical, eco-physiological and biochemical traits will furnish valuable insights to understand mechanisms of plant acclimation to stress, useful to manage cultivation factors to improve resource use optimization in controlled environments cultivations on Earth in line with sustainable development goals.

In this paper, an in-depth and systematic study of the radiological characterization of three types of Puglia region natural limestones (Pietra Leccese, Pietra Mazzara and Carparo) was carried out. The investigation was performed by XRF spectroscopy for a chemical analysis, and gamma spectroscopy of the specific activity concentration of natural radionuclides 226Ra, 232Th, and 40K. Although the limestone does not fall within the category included by Italian Legislative Decree 101/2020, the gamma index was calculated using the results of the gamma spectroscopy measurements. For Pietra Mazzara and Carparo stones, the gamma index was found to be less than the reference value, conversely Pietra Leccese was found to be higher. To obtain a more complete evaluation of the external exposure, radium equivalent activity and external radiation hazard were calculated for all analyzed stones. The results suggest the need to broadly consider the radiological risk for these stones, and for limestone more generally, when used as a building material.

Phaseolus vulgaris L. plants were irradiated with different doses (0.3, 10, 50 and 100 Gy) of X-rays in order to obtain a reference curve of response to ionizing radiations for this species. Growth analysis, gas exchange and chlorophyll a fluorescence measurements were performed to estimate the radio-resistance of bean plants. Specifically, there was a negative influence of X-rays on the net photosynthesis rate at 50 and 100 Gy, already on the day of irradiation. Experimental data showed a recovery over time in the gas exchange while the theoretical maximum photochemical efficiency of the photosystem II (Fv/Fm) was fairly constant throughout the period of measurements (20 days) and for all the experimental conditions. On the other hand, the quantum yield of PSII linear electron transport (ΦPSII) and non-photochemical quenching (NPQ) were deeply influenced over time by X-ray dose, suggesting a decrease in the functionality of the photosynthetic apparatus at the highest radiation doses. The growth was affected only at the highest doses of radiation with a significant and severe reduction of leaf expansion and number of leaves per plant. Despite the arrest in growth, X-ray exposure seems to trigger an increased photochemical activity probably signifying that P. vulgaris plants have a fairly elevated resistance to this kind of ionizing radiation. Our current results will provide a complete analysis of the photosystem II (PSII) response of P. vulgaris to different doses (0.3, 10, 50 and 100 Gy) of X-rays, providing sound references for both space-oriented and radioecology questions.

Exploration of deep space can lead to several health problems due to exposure to cosmic radiation. To increase health and safety in interplanetary exploratory missions, passive shielding is considered the best solution, though not without faults. In fact, shielding can be complex for several reasons, such as the deficiency in biological and physical measurements. Another fundamental aspect related to the shielding problem is the space radiation environment. In fact, galactic cosmic rays, solar particle events, and trapped particle radiations are considered some of the major health risk to astronauts. To protect the space crew, both passive and active shielding is used. In particular, the passive method is considered more efficient to limit the absorbed dose. Fundamental is also the choice of the shielding material. In fact, some materials, like polyethylene, is deemed one of the most effective tools against space radiation.

The study concerns the analysis of 220Rn (thoron) recorded in the surface soil in two sites of the Campi Flegrei caldera (Naples, Southern Italy) characterized by phases of volcanic unrest in the seven-year period 1 July 2011–31 December 2017. Thoron comes only from the most surface layer, so the characteristics of its time series are strictly connected to the shallow phenomena, which can also act at a distance from the measuring point in these particular areas. Since we measured 220Rn in parallel with 222Rn (radon), we found that by using the same analysis applied to radon, we obtained interesting information. While knowing the limits of this radioisotope well, we highlight only the particular characteristics of the emissions of thoron in the surface soil. Here, we show that it also shows some clear features found in the radon signal, such as anomalies and signal trends. Consequently, we provide good evidence that, in spite of the very short life of 220Rn compared to 222Rn, both are related to the carrier effect of CO2, which has significantly increased in the last few years within the caldera. The hydrothermal alterations, induced by the increase in temperature and pressure of the caldera system, occur in the surface soils and significantly influence thoron’s power of exhalation from the surface layer. The effects on the surface thoron are reflected in both sites, but with less intensity, the same behavior of 222Rn following the increasing movements and fluctuations of the geophysical and geochemical parameters (CO2 flux, fumarolic tremor, background seismicity, soil deformation). An overall linear correlation was found between the 222−220Rn signals, indicating the effect of the CO2 vector. The overall results represent a significant step forward in the use and interpretation of the thoron signal.

222Rn gas represents the major contributor to human health risk from environmental radiological exposure. In confined spaces radon can accumulate to relatively high levels so that mitigation actions are necessary. The Italian legislation on radiation protection has set a reference value for the activity concentration of radon at 300 Bq/m3. In this study, measurements of the annual radon concentration of 62 bank buildings spread throughout the Campania region (Southern Italy) were carried out. Using devices based on CR-39 solid-state nuclear track detectors, the 222Rn level was assessed in 136 confined spaces (127 at underground floors and 9 at ground floors) frequented by workers and/or the public. The survey parameters considered in the analysis of the results were: floor types, wall cladding materials, number of openings, door/window opening duration for air exchange. Radon levels were found to be between 17 and 680 Bq/m3, with an average value of 130 Bq/m3 and a standard deviation of 120 Bq/m3. About 7% of the results gave a radon activity concentration above 300 Bq/m3. The analysis showed that the floor level and air exchange have the most significant influence. This study highlighted the importance of the assessment of indoor radon levels for work environments in particular, to protect the workers and public from radon-induced health effects.

Radiological characterization of tuff of Ciglio area in Ischia Island was performed to assess the potential radiological hazard associated with its use. For this purpose, high resolution gamma-ray spectroscopy was used. In five green tuff samples was measured the activity concentration of natural gamma-ray emitting radionuclides 226Ra, 232Th and 40K so, the mean gamma index was calculated. The mean value of the gamma index resulted lower than the reference level and it meets the dose criterion for the safety use of green tuff as building material.

The cytoskeleton is involved in several biological processes, including adhesion, motility, and intracellular transport. Alterations in the cytoskeletal components (actin filaments, intermediate filaments, and microtubules) are strictly correlated to several diseases, such as cancer. Furthermore, alterations in the cytoskeletal structure can lead to anomalies in cells’ properties and increase their invasiveness. This review aims to analyse several studies which have examined the alteration of the cell cytoskeleton induced by ionizing radiations. In particular, the radiation effects on the actin cytoskeleton, cell adhesion, and migration have been considered to gain a deeper knowledge of the biophysical properties of the cell. In fact, the results found in the analysed works can not only aid in developing new diagnostic tools but also improve the current cancer treatments.

Tuscany, with its mixed geology, offers a great variety of stones that are used to build entire villages. In this paper, two of these building materials are considered: Rosso Ammonitco and Pietra Serena. In particular, the activity concentration of 226Ra, 232Th and 40K have been analysed in order to estimate the value of the gamma index. In addition to this, the X-ray fluorescence spectroscopy has been carried out to obtain a chemical characterization of the stones. Although these kind of stones are not subjected to radiological characterization, the approach used was the one reported in Radiation Protection 112, also implemented in the Italian Legislative Decree 101/2020, because of their lithographical peculiarity and their employment in construction.

Radiological risk affects the quality of the environment in buildings since population and workers can be potentially exposed to high levels of radiation. Radon gas emanating from both subsoil and building materials represents the most important source of radiation exposure for people. This study investigates the sustainability concept of a small rural village of Ischia Island, named Ciglio, in relation to radiation protection legislation concerning the radiological risk for workers. Radon activity concentration was measured in typical green-tuff dwellings and in water samples collected from a local waterfall E-Perm devices. Moreover, for green tuff as building material, the radon emanation coefficient was calculated by gamma spectroscopy. The results highlight the importance of performing environmental radon monitoring and investigating the radon content of building materials, especially in geographical areas characterized by traditional use of typical stones for constructions. In conclusion, the sustainable development of rural buildings is possible if the radiological risk for inhabitants and workers is assessed in line with the national radiation protection legislation.

A large body of literature has demonstrated that the mechanical properties of microenvironment have a key role in regulating cancer cell adhesion, motility, and invasion. In this work, we have introduced two additional parameters, named cell trajectory extension and area traveled by cell, to describe the tendency of normal tissue and metastatic cancer cells to move in a directional way when they interact with physio-pathological substrates, characterized by stiffnesses of 1–13 kPa, before and after treatment with 2 doses of X-rays (2 and 10 Gy). We interpreted these data by evaluating also the impact of substrate stiffness on 2 morphological parameters which indicate not only the state of cell adhesion, but also cell polarization, prerequisite to directional movement, and the formation of protrusions over cell perimeters. We believe that a so wide analysis can give an efficient and easily readable overview of effects of radiation therapy on cell-ECM crosstalk when used as therapeutic agent.

The structural and mechanical properties of the microenvironmental context have a profound impact on cancer cell motility, tumor invasion, and metastasis formation. In fact, cells react to their mechanical environment modulating their adhesion, cytoskeleton organization, changes of shape, and, consequently, the dynamics of their motility. In order to elucidate the role of extracellular matrix stiffness as a driving force in cancer cell motility/invasion and the effects of ionizing radiations on these processes, we evaluated adhesion and migration as biophysical properties of two different mammary cell lines, over a range of pathophysiological stiffness (1&ndash, 13 kPa) in a control condition and after the exposure to two different X-ray doses (2 and 10 Gy, photon beams). We concluded that the microenvironment mimicking the normal mechanics of healthy tissue has a radioprotective role on both cell lines, preventing cell motility and invasion. Supraphysiological extracellular matrix stiffness promoted tumor cell motility instead, but also had a normalizing effect on the response to radiation of tumor cells, lowering their migratory capability. This work lays the foundation for exploiting the extracellular matrix-mediated mechanism underlying the response of healthy and tumor cells to radiation treatments and opens new frontiers in the diagnostic and therapeutic use of radiotherapy.

Purpose: The aim of the study was to exploit the feasibility of thermoluminescent dosimeters (TLDs) in radiation therapy techniques in which high dose per fraction is involved. Methods: Dose–response of TLD-100 (LiF: Mg, Ti) was investigated in both 6-MV photon and 6-MeV electron beams. The element correction factor (ECF) generation method was applied to check the variability of the TLDs response. Two batches of 50 TLDs were divided into groups and exposed in the dose range 0 to 30 Gy. Regression analysis was performed with both linear and quadratic models. For each irradiation beam, the calibration curves were obtained in 3 dose range 0 to 8 Gy, 0 to 10 Gy, and 0 to 30 Gy. The best-fitting model was assessed by the Akaike Information Criterion test. Results: The ECF process resulted a useful tool to reduce the coefficients of variation from original values higher than 5% to about 3.5%, for all the batches exposed. The results confirm the linearity of dose–response curve below the dose level of 10 Gy for photon and electron beam and the supralinear trend above. Conclusion: The TLDs are suitable dosimeters for dose monitoring and verification in radiation treatment involving dose up to 30 Gy in a single fraction.

This is a seven-year study (1/7/2011-31/12/2017) of radon monitoring at two sites of Campi Flegrei caldera (Neaples, Southern Italy) that in the last 70 years experienced repeated phases of volcanic unrest. The sites are equipped with devices for radon detection, based on the spectrometry analysis of the α-particles of radon daughters. A hybrid method, as combination of three known methods, is applied for the identification of residuals (anomalies) and trends of the time series of Radon. The results are compared with the following indicators of current caldera unrest: the tremor caused by the major fumarolic vent registered by a seismic station; the cumulative of background seismicity; the maximum vertical deformation acquired by GPS networks during the current phase of uplift; the temperature-pressure of the hydrothermal system estimated based on gas geo-indicators. The comparisons show strong correlation among independent signals and suggest that the extension of the area affected by current Campi Flegrei crisis is larger than the area of seismicity and of intense hydrothermal activity from which the radon stations are 1–4 km away. These results represent an absolute novelty in the study of a such calderic area and mark a significant step forward in the use and interpretation of the radon signal.

In this study, for the first time, was measured the concentration of the natural gamma-ray emitting radionuclides of 238U, 232Th series and 40K in building materials used in Puglia region. The selected samples consist on calcarenitic stones: Pietra Leccese, Pietra Mazzara and Carparo. The results show that the Pietra Leccese specific activity of 226Ra, 238U, 40K are higher than Pietra Mazzara and Carparo ones. 232Th radionuclide activity is in accordance for all samples. Consequently, the gamma index, useful for gamma-ray exposure estimation, was calculated. This value for each analysed sample is below the reference level reported in European Directive 59/2013.

Proton therapy represents a technologically advanced method for delivery of radiation treatments to tumors. The determination of the biological effectiveness is one of the objectives of the MoVe IT (Modeling and Verification for Ion Beam Treatment Planning) project of the National Institute for Nuclear Physics (INFN) CSN5. The aim of the present work, which is part of the project, was to evaluate the performance of the thermoluminescent dosimeters (TLDs-100) for dose verification in the proton beam line. Four irradiation experiments were performed in the experimental room at the Trento Proton Therapy Center, where a 150 MeV monoenergetic proton beam is available. A total of 80 TLDs were used. The TLDs were arranged in one or two rows and accommodated in a specially designed water-equivalent phantom. In the experimental setup, the beam enters orthogonally to the dosimeters and is distributed along the proton beam profile, while the irradiation delivers doses of 0.8 Gy or 1.5 Gy in the Bragg peak. For each irradiation stage, the depth–dose curve was determined by the TLD readings. The results showed the good performance of the TLDs-100, proving their reliability for dose recordings in future radiobiological experiments planned within the MoVe IT context.

The aim of this study was to assess a potential relationship between indoor radon exposure and the incidence of lung cancer worldwide. A systematic literature search was carried out in PubMed, Web of Science, and Google Scholar to identify relevant studies published in English conducted in the last 15 years until January 2016. Summary relative risks (RR) and the corresponding 95% confidence intervals (CIs) were calculated using a random-effects model and the influence of moderators using a mixed-effects model. Heterogeneity was assessed using the Q, I2 and H2 tests, and the source of heterogeneity was detected by meta-regression analysis. Publication bias was evaluated with Egger’s regression symmetry test and the contour-enhanced funnel plot. Leave-one-out sensitivity analysis was performed. Twenty-five lung cancer studies (case-control studies) with 13,569 cases and 22,701 controls were included. Indoor Radon exposure was significantly associated with increased risk for lung cancer (RR, 1.19; 95% CI, 1.02–1.39). Study location analysis showed that radon exposure was associated with increased risk for lung cancer from forty degrees absolute latitude (RR, 1.09; 95% CI, 0.92–1.31), to fifty degrees (RR 1.26; 95% CI, 1.08–1.48), to sixty degrees (RR, 1.46; 95% CI, 1.12–1.91). Indoor radon exposure may be associated with increased risk for lung cancer.

Astrobiology studies the origin and evolution of life on Earth and in the universe. According to the panspermia theory, life on Earth could have emerged from bacterial species transported by meteorites, that were able to adapt and proliferate on our planet. Therefore, the study of extremophiles, i.e. bacterial species able to live in extreme terrestrial environments, can be relevant to Astrobiology studies. In this work we described the ability of the thermophilic species Geobacillus thermantarcticus to survive after exposition to simulated spatial conditions including temperature's variation, desiccation, X-rays and UVC irradiation. The response to the exposition to the space conditions was assessed at a molecular level by studying the changes in the morphology, the lipid and protein patterns, the nucleic acids. G. thermantarcticus survived to the exposition to all the stressing conditions examined, since it was able to restart cellular growth in comparable levels to control experiments carried out in the optimal growth conditions. Survival was elicited by changing proteins and lipids distribution, and by protecting the DNA's integrity.

The International Commission on Radiation Protection (ICRP) in 2011 recommended the lowering of the annual eye lens dose limit from 150 mSv/year to 20 mSv/year in order to reduce the risk of X-ray-induced lens opacity in medical staff. The purpose of this study was to assess the status of knowledge of the new eye lens dose limit and of the radioprotection culture among operators. To this end, a questionnaire was administered to physicians, X-ray technicians, and nurses working in five hospitals of the Campania region, Italy. A total of 64 questionnaires were collected in the hospital departments in which procedures involving ionizing radiation were routinely performed. The data analyzed yielded the following results: 12 operators affirmed to know the new eye lens dose limit, 53 operators routinely wore lead aprons, and 23 operators used lead glasses. Four workers performed eye lens dosimetry through specific dosimeters. A significant lack of knowledge of the reduced eye lens dose limit suggests the need to implement radioprotection-training programs aimed at raising awareness about the importance of health care in the workplace and at reducing the risk of radio-induced effects to the eye lens.
