Taking into account the requirements to reduce the release of harmful emissions into the environment, the EU's environmental standards when transitioning to the Euro 7 standard in 2025 will actually lead vehicles having to operate without producing emissions in all driving situations. Carmakers believe that the new, much stricter regulations will mark the end of the internal combustion engine era. For example, in 2030, the manufacturer SEAT will cease its activities, leaving behind the Cupra brand, which will be exclusively electric in the future. This trend will apply not only to private vehicles (passenger cars), but also to utility vehicles, which is the subject of our research, namely the spatial tubular frame in the Bogdan ERCV27 garbage truck, presented in the form of a solid model. The peculiarity of the studied model is the installation of a battery block behind the driver's cabin, causing an additional load to be placed on the spatial frame of the garbage truck, which in terms of its architecture is more like the body of a bus. During the conditions involving various modes of operation of a full-scale Bogdan ERCV27 garbage truck sample, questions about the strength and uniformity of its load-bearing spatial frame inevitably arise, which are decisive, even at the stage of designing and preparing the technical documentation. The main static load mode, which, despite its name, also covers dynamic conditions, was modeled using the appropriate coefficient kd = 2.0. The maximum stresses on the model during the "bending" mode were 381.13 MPa before structure optimization and 270.5 MPa as a result of the improvement measures. The spatial frame mass was reduced by 4.13%. During the "torsion" mode, the maximum deformation values were 12.1–14.5 mm, which guarantees the normal operation of the aggregates and units of the truck. [ABSTRACT FROM AUTHOR]

Among the various types of polymeric materials, geosynthetics deserve special attention. A geosynthetic is a product made from synthetic polymers that is embedded in soils for various purposes. There are some basic functions of geosynthetics, namely, erosion control, filtration, drainage, separation, reinforcement, containment, barrier, and protection. Geosynthetics for erosion control are very effective in preventing or limiting soil loss by water erosion on slopes or river/channel banks. Where the current line runs through the undercut area of the slope, the curvature of the arch is increased. If this phenomenon is undesirable, the meander arch should be protected from erosion processes. The combination of geosynthetics provides the best resistance to erosion. In addition to external erosion, internal erosion of soils is also a negative phenomenon. Internal erosion refers to any process by which soil particles are eroded from within or beneath a water-retaining structure. Geosynthetics, particularly geotextiles, are used to prevent internal erosion of soils in contact with the filters. Therefore, the main objective of this review paper is to address the many ways in which geosynthetics are used for erosion control (internal and external). Uveal melanoma (UM) is the most common primary intraocular tumor in adults, with no standardized treatment for advanced disease. Based on preliminary bioinformatical analyses DTYMK and PARP1 were selected as potential therapeutic targets. High levels of both proteins were detected in uveal melanoma cells and correlated with increased tumor growth and poor prognosis. In vitro tests on MP41 (BAP1 positive) and MP46 (BAP1 negative) cancer cell lines using inhibitors pamiparib (PARP1) and Ymu1 (DTYMK) demonstrated significant cytotoxic effects. Combined treatment had synergistic effects in MP41 and additive in MP46 cell lines, reducing cell proliferation and inhibiting the mTOR signaling pathway. Furthermore, the applied inhibitors in combination decreased cell motility and migration speed, especially for BAP1-negative cell lines. Our hypothesis of the double hit into tumoral DNA metabolism as a possible therapeutic option in uveal melanoma was confirmed since combined targeting of DTYMK and PARP1 affected all tested cytophysiological parameters with the highest efficiency. Our in vitro findings provide insights into novel therapeutic avenues for managing uveal melanoma, warranting further exploration in preclinical and clinical settings. [ABSTRACT FROM AUTHOR]

Implementing rain garden (RG) designs is widespread worldwide to reduce peak flow rates, promote stormwater infiltration, and treat pollutants. However, inadequate RG design degrades its hydrological behaviour, requiring the development and validation of an appropriate hydrological model for the design and analysis of structures. This study aimed to improve a hydrological infiltration model based on Darcy's law by taking into account the height of the water column (HWC) at the surface of the RG and the filtration coefficients of soil materials. The model was tested by simulating the hydrological characteristics of a rain garden based on a single rain event of critical intensity (36 mm/h). Using the validated model, design curves were obtained that predict the performance of the RG as a function of the main design parameters of the structure: water column height, ratio of catchment area to structure area, layer thickness, and soil filtration coefficient. The hydrological efficiency of the RG was evaluated in terms of the time of complete saturation, filling of the structure with water, and determining the change in HWC caused by changes in the parameters. The filtration coefficient and thickness of the upper and intermediate infiltration layers of the RG are the main parameters that affect the depth of saturation of the layers of the structure and the HWC on the surface. The model is not very sensitive to the model parameters related to the lower gravel layer. If the top layer's thickness increases by 10 cm, it takes longer to fill the structure with water, and the HWC on the surface reaches 0.341 m. The rain garden's performance improves when the filtration coefficient of the top layer is 7.0 cm/h. Complete saturation and filling of the structure with rainwater do not occur within 7200 s, and the water column reaches a height of 0.342 m at this filtration coefficient. However, the rain garden's effectiveness decreases if the filtration coefficient of the upper and intermediate layers exceeds 15 cm/h and 25 cm/h, respectively, or if the catchment area to RG area ratio decreases to values below 15. The modelling results confirm that considering the HWC in RG hydrological models is essential for designing structures to minimise the risk of overflow during intense rainfall events. Background Accurate risk stratification of uveal melanoma (UM) patients is important for determining the interval and frequency of surveillance. Loss of BAP1 expression has been shown to be strongly associated with UM-related death and metastasis. Methods In this study of 164 enucleated UMs, we assessed the prognostic role of preferentially expressed antigen in melanoma (PRAME) expression and Ki67 proliferation index measured by digital quantitation using QuPath programme in patients with BAP1-positive and BAP1-loss UMs. Results In univariate analyses with log-rank tests and Kaplan-Meier curves, PRAME further stratified only overall survival (OS) in BAP1-positive and BAP1-loss tumour groups. However, Ki67 further stratified both OS and disease-free survival (DFS) in BAP1-positive and BAP1-loss tumour groups. In multivariate analyses, Ki67 percentage and BAP1 were independent survival predictors for both OS and DFS, whereas PRAME was not a significant covariate. In model comparisons, combined Ki67 and BAP1 performed better than combined PRAME and BAP1 in risk-stratifying patients for both OS and DFS. Ki67 was better than PRAME in risk stratification of BAP1-positive UMs. Low Ki67 index correlated with significantly prolonged DFS in BAP1-loss UMs. Conclusion A panel of Ki67 and BAP1 could be a helpful risk stratification strategy for UM. Changes in river channel morphological parameters are influenced by anthropogenic factors, such as climatic changes, river catchment management changes, and hydrotechnical development of rivers. To assess the intensity of individual pressures and the resulting changes in abiotic and biotic factors in the riverbed, water quality monitoring is conducted, including the assessment of the hydromorphological status. The assessment can be based on the River Habitat Survey (RHS) which is a synthetic method that includes the evaluation of habitat character and river quality based on their morphological structure. The input data, which characterise any river include physical features of hydrotechnical structures, bed granulation, occurrence of bedforms, visible morphodynamic phenomena, and a sediment transport pattern. The RHS method allows to determine two quantitative indices used to evaluate the hydromorphological status: Habitat Modification Score (HMS), which determines the extent of transformation in the morphology of a watercourse, and Habitat Quality Assessment (HQA), which is based on the presence and diversity of natural elements in a watercourse and river valley. The proposed method can be divided into three stages. The first assesses the river section hydromorphological indices, describing the degree of technical modification (HMS) and the ecological quality of the reach (HQA), using the RHS method. The second stage describes morphological changes resulting from the technical regulation and estimates indices for the regulated reach. Finally, we compare HQA and HMS indices before and after the regulation. This comparison is described by numerical indicators and related to reference values. [ABSTRACT FROM AUTHOR]
