We studied a Triops population from a Mediterranean temporary pond located in Guadalcázar, a township in the western part of Córdoba Province. There are four morphologically similar species of Triops occurring in the SW Iberian Peninsula. In particular, the two species found in southern Andalusia, T. baeticus and T. gadensis, cannot reliably be told apart based on known morphological characters. We thus used sequences of 12S rDNA as a molecular marker to soundly identify the population to species rank. Our results demonstrate that the Triops population studied belongs to T. gadensis. This result is rather unexpected given that the species was thought to be restricted to a small distribution range along the Atlantic coast in Cádiz Province. The population is located at approximately 170 km from previously known sites of T. gadensis and shows a previously unknown haplotype, the sixth 12S haplotype reported for the species. Based on a molecular clock designed for dating divergence events among conspecific samples of Triops, the haplotype is estimated to have diverged at 10.3–29.4 ka BP. We thus suggest that the population has high conservation value, and its habitat, including the surroundings, should be legally protected. We further propose that an open habitat type as pastureland should be retained for the surroundings of the site in order to support its ecological function in the frame of meta-populations dynamics, and as a possible source population from which new localities could be populated via passive dispersal.

The distribution, status, and phenology of the anostracan Phallocryptus spinosus in Sicily are described. The only population of the species to date known to occur with certainty on the island inhabits a few temporary water bodies located within the Natura2000 site “ITA010006 - Paludi di Capo Feto e Margi Spanò”. In these sites, the species co-occurs with an interesting halophilous flora and crustacean fauna. Based on available data, the concentration of dissolved salts in the water bodies seem to be the main driver of the life histories of the co-existing populations of the anostracans Artemia salina and Phallocryptus spinosus.

The large branchiopod fauna of Sardinia is reviewed based both on literature and newly collected data. Based on the available evidence, 13 taxa are present on the island (8 Anostraca, 2 Notostraca, and 3 Spinicaudata). Among them, the finding of the spinicaudatan Leptestheria dahalacensis is new for Sardinia, while the spinicaudatans Cyzicus bucheti and Eulimnadia sp. were overlooked in the most recent synopses on the fauna of the island due to misidentifications. Conversely, Cyzicus tetracerus and Limnadia lenticularis, previously erroneously reported based on misidentifications, must be excluded from the fauna of Sardinia. The finding of Eulimnadia sp. is the first record in Europe and the northernmost record of the genus. The occurrence of Leptestheria dahalacensis in Sardinia is rather unexpected and probably due to its accidental introduction linked with rice cultures. At least four of the 13 Sardinian large branchiopod species are absent from the Italian mainland and Sicily, stressing the uniqueness of its fauna and its significant contribution to the Mediterranean inland water crustacean diversity.

Ectosymbiotic temnocephalan flatworms belonging to the genus Temnosewellia Damborenea and Cannon, 2001 were collected on Cherax destructor Clark, 1936 in an aquaculture farm in Sicily, Italy. This represents the first record of a temnocephalan species for the fauna of the island. Morphological and molecular identification of the collected specimens proved that they belong to the allochthonous species Temnosewellia minor Haswell, 1888, which was introduced along with crayfishes bred in aquaculture farms. The phylogenetic analyses carried out for the molecular identification of the Sicilian population highlighted some inconsistencies in the grouping of the Temnosewellia sequences available online, thus stressing the opportunity of a careful re-examination of the voucher samples and their identifications. The risks of a its unwary introduction in the wild and the need of monitoring its possible impacts on native biota are briefly discussed.

Scarce data are currently available about the distribution of the Mediterranean medicinal leech Hirudo verbana in Italy, and most of the known occurrence localities are based on records collected in the nineteenth and the first half of the twentieth century, which were not confirmed in the last decades, mostly due to a lack of surveys. Accordingly, the available knowledge on the distribution of the species is far from being updated and representative, although a significant negative trend of H. verbana throughout the country is supposed. The lack of sound distribution data is a significant shortfall, which hinders the implementation of the monitoring of the species as required by the Article 17 of the “Habitats Directive” on the species of Union concern. To provide recent, validated distributional data for the Mediterranean medicinal leech in Italy to be used as baseline data for further surveys and monitoring, we present herein a set of unpublished observations collected in the last decades in peninsular Italy, Sicily, and Sardinia. Moreover, we report observation of H. verbana feeding on amphibians, a feeding habit to date poorly documented for the Mediterranean medicinal leech. Based on both published and novel data, H. verbana is characterised by a large but fragmented distribution in Italy. Therefore, dedicated monitoring programs and the collection of validated occasional observations are highly desirable to get a clearer picture of the real distribution of the species.

This study evaluates, for the first time, the reducing capacity, radical scavenger activity, and in vitro antitumor and anti-inflammatory effects of chitosan, astaxanthin, and bio-phenols extracted from the exoskeleton of Sicilian Procambarus clarkii, the most widespread species of invasive crayfish in the Mediterranean region. Among the extracted compounds, astaxanthin exhibited the highest antioxidant activity in all assays. Chitosan and polyphenols demonstrated reducing and radical scavenging activity; chitosan showed significant ferric ion reducing capacity in the FRAP test, while bio-phenolic compounds displayed notable radical scavenging activity in the DPPH and ABTS assays. Both astaxanthin and polyphenols showed dose-dependent cytotoxicity on two different cancer cell lines, with IC50 values of 1.45 µg/mL (phenolic extract) and 4.28 µg/mL (astaxanthin extract) for HepG2 cells and 2.45 µg/mL (phenolic extract) and 4.57 µg/mL (astaxanthin extract) for CaCo-2 cells. The bio-phenolic extract also showed potential anti-inflammatory effects in vitro by inhibiting nitric oxide production in inflamed RAW 264.7 macrophages, reducing the treated/control NO ratio to 77% and 74% at concentrations of 1.25 and 1.5 μg/mL, respectively. These results suggest that P. clarkii exoskeletons could be a valuable source of bioactive molecules for biomedical, pharmaceutical, and nutraceutical application while contributing to the sustainable management of this invasive species. The occurrence of the medicinal leech Hirudo verbana in the inland waters of Sicily has been lately overlooked. In the present note, the occurrence and distribution of this species is reviewed based both on the review of the available literature data and field collecting. Although a noteworthy reduction in the distribution range of the species seems to have taken place in Sicily in the course of the XX century, Hirudo verbana was confirmed to be still present in several sites located both within and out of Natura2000 sites. The Sicilian populations of the species should be included in the frame of the monitoring activities established by the Article 17 of the EU Council Directive 92/43/EEC (“Habitats Directive”).

In the frame of the activities of the LIFE11 NAT/GR/1014 ForOpenForests, some water bodies occurring in the "Ethnikos Drymos Oitis” (GR2440004) and “Oros Kallidromo” (GR2440006) (Sterea Ellada) were investigated with the aim of providing a first census of the composition and diversity of their crustacean fauna. Overall, the sampling of 15 water bodies (7 of them listed as “Mediterranean temporary ponds” sensu EU “Habitats Directive”) led to the finding of 13 branchiopod, 11 copepod, and 7 ostracod taxa, including 4 species new for mainland Greece, i.e. the copepods Arctodiaptomus alpinus (Imhoff, 1885) and Diaptomus cf. serbicus, and the branchiopods Leptestheria dahalacensis (Rüppel, 1837) and Wlassicsia pannonica Daday, 1904. The comparative analysis of the observed species assemblages and richness suggests that the protection of those ponds identified as “priority habitats” according to the “Habitats Directive” is effective for the specialized and peculiar crustacean biota of these ecosystems, but it is not sufficient in order to preserve efficiently the whole diversity of temporary pond-dwelling crustaceans occurring in the study area. Therefore, the implementation of synergistic conservation measures dedicated to both “priority” and “non-priority” habitats is desirable.

Properly managing aquatic organisms is crucial, including protecting endemic species and controlling invasive species. From a circular economy perspective, the sustainable use of aquatic species as a source of bioactive molecules is an area that is increasingly being explored. This includes the use of non-edible portions of seafood, which could pose considerable risks to the environment due to current methods of disposal. Therefore, it is of paramount importance to ensure that the exploitation of these resources does not result in the transfer of pollutants to the final product. This study analyzed two types of non-edible parts from the crayfish Procambarus clarkii: the abdominal portion of the exoskeleton (AbE) and the whole exoskeleton (WE), including the cephalothorax. These portions could potentially be utilized in the context of eradication activities regulated by local authorities. A screening analysis of four classes of pollutants, including pesticides, per- and polyfluoroalkyl substances (PFAS), phthalic acid esters (PAEs), and trace elements (TEs), was performed. The only analytes detected were TEs, and significant differences in the contamination profile were found between AbE and WE. Nevertheless, the levels recorded were comparable to or lower than those reported in the literature and below the maximum levels allowed in the current European legislation for food, suggesting that their potential use is legally permitted. The first record of the red swamp crayfish in Sicily dates back to 2003 and, since then, the species seemed to be confined to a few localities in western Sicily. A small “citizen science” project carried out from November 2016 onwards led to the creation of the “Sicilian Procambarus working group” (SPwg), which aims at monitoring the distribution and impact of the species in Sicily. To date, the SPwg found the red swamp crayfish in five new sites on the island, thus doubling the number of local sites of occurrence. The new Procambarus clarkii sites lie in different river basins, some of them located several hundred kilometres from the invaded areas known to date, suggesting the existence of multiple independent releases of the species in the wild. The need of better informing the local population on the risks exerted by invasive species on biological diversity, and of carefully monitoring the impact of P. clarkii on the Sicilian inland water biota is briefly stressed.

Temporary ponds are the most peculiar and representative water bodies in the arid and semi-arid regions of the world, where they often represent diversity hotspots that greatly contribute to the regional biodiversity. Being indissolubly linked to these ecosystems, the so-called “large branchiopods” are unanimously considered flagship taxa of these habitats. Nonetheless, updated and detailed information on large branchiopod faunas is still missing in many countries or regions. Based on an extensive bibliographical review and field samplings, we provide an updated and commented checklist of large branchiopods in Tunisia, one of the less investigated countries of the Maghreb as far as inland water crustaceans are concerned. We carried out a field survey from 2004 to 2012, thereby collecting 262 crustacean samples from a total of 177 temporary water bodies scattered throughout the country. Large branchiopod crustaceans were observed in 61% of the sampled sites, leading to the identification of fifteen species. Among these, the halophilic anostracan Branchinectella media is here reported for the first time for the country; conversely, four of the species reported in literature were not found during the present survey. Based on literature and novel data, the known large branchiopod fauna of Tunisia now includes 19 species, showing a noteworthy species richness when the limited extension of the country is considered. For each species, the regional distribution is described and an annotated list of references is provided. Under a conservation perspective, the particular importance of the temporary ponds occurring in the Medjerda river alluvial plain is further stressed. In this location, several large branchiopod taxa with different ecological requirements converge and form unique and species-rich assemblages that should be preserved.

The chemical valorization of widespread molecules in renewable sources is a field of research widely investigated in the last decades. In this context, we envisaged that indole-3-carbinol, present in different Cruciferae plants, could be a readily available building block for the synthesis of various classes of indoles through a palladium-catalyzed Tsuji–Trost-type reaction with O and S soft nucleophiles. The regiochemical outcome of this high-yielding functionalization shows that the nucleophilic substitution occurs only at the benzylic position. Interestingly, with this protocol, the sulfonyl unit could be appended to the indole nucleus, providing convenient access to new classes of molecules with potential bioactivity. Dopamine is a key neurotransmitter involved in a series of biologically relevant processes and its derivatives have sparked significant interest as intriguing synthetic targets. This class of compounds is indeed not only considerable for the potential biological activities but is also promising for diverse applications in material science. In light of this, our research was focused on the synthesis of 6-aryldopamine derivatives starting from 4-(2-aminoethyl)phenol through a sequential protocol, whose main steps are hydroxylation, halogenation, and Suzuki cross-coupling. Simple Summary: The genus Craspedacusta comprises invasive freshwater jellyfish species present in all continents except Antarctica. Due to the morphological plasticity of the medusa stage, the number of species in the genus Craspedacusta is still disputed. Here, we shed new light on the distribution of the genetic lineages of these non-native species across the Italian peninsula, Sicily, and Sardinia. Since the first Italian record in 1946 and up to the last available review in 2017, Craspedacusta medusae were reported in 40 Italian water bodies. In the present study, we report 21 new records of Craspedacusta medusae presence since its latest finding in 2017. Furthermore, we present results of the molecular analyses conducted on the collected medusae. Our findings show the presence of two distinctive genetic lineages of Craspedacusta in Italy: (i) a group whose distribution ranges from central to northern Italy; and (ii) a group that comprises three populations from northern Italy and the single Sicilian population known to date. Olindiid freshwater jellyfishes of the genus Craspedacusta Lankester, 1880 are native to eastern Asia; however, some species within the genus have been introduced worldwide and are nowadays present in all continents except Antarctica. To date, there is no consensus regarding the taxonomy within the genus Craspedacusta due to the morphological plasticity of the medusa stages. The species Craspedacusta sowerbii Lankester, 1880 was first recorded in Italy in 1946, and until 2017, sightings of the jellyfish Craspedacusta were reported for 40 water bodies. Here, we shed new light on the presence of the freshwater jellyfishes belonging to the genus Craspedacusta across the Italian peninsula, Sardinia, and Sicily. First, we report 21 new observations of this non-native taxon, of which eighteen refer to medusae sightings, two to environmental DNA sequencing, and one to the finding of polyps. Then, we investigate the molecular diversity of collected Craspedacusta specimens, using a Bayesian analysis of sequences of the mitochondrial gene encoding for Cytochrome c Oxidase Subunit I (mtDNA COI). Our molecular analysis shows the presence of two distinctive genetic lineages: (i) a group that comprises sequences obtained from populations ranging from central to northern Italy; (ii) a group that comprises three populations from northern Italy—i.e., those from the Lake Levico, the Lake Santo of Monte Terlago, and the Lake Endine—and the single known Sicilian population. Owing to the "Racovitzan impediment", the groundwater fauna of most biogeographical regions is currently inadequately known, thus hampering our understanding of subterranean biodiversity and its protection. Based on an extensive bibliographical review accompanied by fieldwork to localize occurrence sites, a checklist of crustacean taxa reported to date from Sicilian groundwater is provided, and their distribution is described. Among the 63 taxa recorded to date, 43 belong to the class Copepoda (orders Cyclopoida and Harpacticoida), 15 to the class Malacostraca (orders Amphipoda, Bathynellacea, Isopoda, and Thermosbaenacea), and 5 to the class Ostracoda (order Podocopida). Conversely, to date, no representatives of the copepod order Calanoida nor species of the class Branchiopoda have been recorded from groundwater habitats on the island. Several taxa require accurate taxonomic revision or are yet to be formally described and are thus at present left in open nomenclature. Finally, the date of publication of several copepod and amphipod taxa is amended. A high incidence of stygobites (i.e., obligate groundwater dwellers) has been observed in malacostracans, whereas nearly half of the recorded copepods were non-stygobites. This pattern is mirrored by the incidence of endemic species, which is higher in malacostracans than in copepods. The only non-stygobitic crustacean species endemic to Sicily observed in the frame of the present review is the asellid isopod Proasellus montalentii. The paucity of information currently available on the Sicilian groundwater ostracods prevents us from drawing conclusions regarding this crustacean group. The origin and composition of Sicilian groundwater crustacean fauna can be explained by considering three major faunal assemblages: the presence of ancient paleoendemic taxa, likely of Miocene origin, the colonization of the groundwater of the island during late Pliocene and Pleistocene land connections with peninsular Italy, and the direct colonization of these environments from the sea; no species of African origin have been discovered to date. Based on currently available data, the groundwater of southeastern Sicily hosts the highest species richness and some of the most biogeographically interesting taxa. Unfortunately, a progressive lowering and salinization of the local aquifers possibly due to climate change and its overexploitation threats this fauna, and several taxa are disappearing even before their discovery and description. A gold-catalyzed protocol to obtain functionalized 3H-pyrrolo [1,2,3-de] quinoxalines from suitable substituted N-alkynyl indoles has been proposed. The mild reaction conditions were revealed to be compatible with different functional groups, including halogen, alkoxyl, cyano, ketone, and ester, allowing the isolation of title compounds with yields from good to high. A reaction mechanism has been proposed, and theoretical calculations have been provided to rationalize the final step of the hypothesized reaction mechanism.
