Abstract

The advancements in human-centered artificial intelligence (HCAI) systems for Industry 5.0 is a new phase of industrialization that places the worker at the center of the production process and uses new technologies to increase prosperity beyond jobs and growth. HCAI presents new objectives that were unreachable by either humans or machines alone, but this also comes with a new set of challenges. Our proposed method accomplishes this through the knowlEdge architecture, which enables human operators to implement AI solutions using a zero-touch framework. It relies on containerized AI model training and execution, supported by a robust data pipeline and rounded off with human feedback and evaluation interfaces. The result is a platform built from a number of components, spanning all major areas of the AI lifecycle. We outline both the architectural concepts and implementation guidelines and explain how they advance HCAI systems and Industry 5.0. In this article, we address the problems we encountered while implementing the ideas within the edge-to-cloud continuum. Further improvements to our approach may enhance the use of AI in Industry 5.0 and strengthen trust in AI systems.

2. DNA Interactions and Biological Activity of 2,9-Disubstituted 1,10-Phenanthroline Thiosemicarbazone-Based Ligands and a 4-Phenylthiazole Derivative

Abstract

Four 1,10-phenanthroline derivatives (1–4) were synthesized as potential telomeric DNA binders, three substituted in their chains with thiosemicarbazones (TSCs) and one 4-phenylthiazole derivative. The compounds were characterized using NMR, HRMS, FTIR-spectroscopy and combustion elemental analysis. Quadruplex and dsDNA interactions were preliminarily studied, especially for neutral derivative 1, using FRET-based DNA melting assays, equilibrium dialysis (both competitive and non-competitive), circular dichroism and viscosity titrations. The TSC derivatives bind and stabilize the telomeric Tel22 quadruplex more efficiently than dsDNA, with an estimated 24-fold selectivity determined through equilibrium dialysis for compound 1. In addition, cytotoxic activity against various tumor cells (PC-3, DU145, HeLa, MCF-7 and HT29) and two normal cell lines (HFF-1 and RWPE-1) was evaluated. Except for the 4-phenylthiazole derivative, which was inactive, the compounds showed moderate cytotoxic properties, with the salts displaying lower IC50 values (30–80 μM), compared to the neutral TSC, except in PC-3 cells (IC50 (1) = 18 μM). However, the neutral derivative was the only compound that exhibited a modest selectivity in the case of prostate cells (tumor PC-3 versus healthy RWPE-1). Cell cycle analysis and Annexin V/PI assays revealed that the compounds can produce cell death by apoptosis, an effect that has proven to be similar to that demonstrated by other known 1,10-phenanthroline G4 ligands endowed with antitumor properties, such as PhenDC3 and PhenQE8.

Abstract

The complement system is considered the first line of defense against pathogens. Hijacking complement regulators from blood is a common evasion tactic of pathogens to inhibit complement activation on their surfaces. Here, we report hijacking of the complement C4b-binding protein (C4bp), the regulator of the classical and lectin pathways of complement activation, by the sporozoite (SPZ) stage of the Plasmodium falciparum parasite. This was shown by direct binding of radiolabeled purified C4bp to live SPZs as well as by binding of C4bp from human serum to SPZs in indirect immunofluorescence assays. Using a membrane-bound peptide array, peptides from the N-terminal domain (NTD) of P. falciparum circumsporozoite protein (CSP) were found to bind C4bp. Soluble biotinylated peptide covering the same region on the NTD and a recombinantly expressed NTD also bound C4bp in a dose-dependent manner. NTD-binding site on C4bp was mapped to the CCP1-2 of the C4bp α-chain, a common binding site for many pathogens. Native CSP was also co-immunoprecipitated with C4bp from human serum. Preventing C4bp binding to the SPZ surface negatively affected the SPZs gliding motility in the presence of functional complement and malaria hyperimmune IgG confirming the protective role of C4bp in controlling complement activation through the classical pathway on the SPZ surface. Incorporating the CSP-C4bp binding region into a CSP-based vaccine formulation could induce vaccine-mediated immunity that neutralizes this immune evasion region and increases the vaccine efficacy.

Abstract

This paper provides an analysis on the emergence of new technologies in the financial industry and their application to financial and investment activities, where organizations are highly equipped with the technology needed to overshadow traditional financial institutions. In addition, three of the most important and controversial areas of this sector -challenges, regulation and collaboration- are reviewed and analyzed in depth through a systematic literature review. VOS Viewer software has been used to classify the different keywords, according to their co-citation, following clustering techniques. Thanks to the studies carried out by other researchers, it has been possible to structure clearly the distribution and trends in the financial sector, mentioning also possible future trends and gaps to be researched in the sector.

5. Anopheles midgut epithelium evades human complement activity by capturing factor H from the blood meal.

Abstract

Hematophagous vectors strictly require ingesting blood from their hosts to complete their life cycles. Exposure of the alimentary canal of these vectors to the host immune effectors necessitates efficient counteractive measures by hematophagous vectors. The Anopheles mosquito transmitting the malaria parasite is an example of hematophagous vectors that within seconds can ingest human blood double its weight. The innate immune defense mechanisms, like the complement system, in the human blood should thereby immediately react against foreign cells in the mosquito midgut. A prerequisite for complement activation is that the target cells lack complement regulators on their surfaces. In this work, we analyzed whether human complement is active in the mosquito midgut, and how the mosquito midgut cells protect themselves against complement attack. We found that complement remained active for a considerable time and was able to kill microbes within the mosquito midgut. However, the Anopheles mosquito midgut cells were not injured. These cells were found to protect themselves by capturing factor H, the main soluble inhibitor of the alternative complement pathway. Factor H inhibited complement on the midgut cells by promoting inactivation of C3b to iC3b and preventing the activity of the alternative pathway amplification C3 convertase enzyme. An interference of the FH regulatory activity by monoclonal antibodies, carried to the midgut via blood, resulted in increased mosquito mortality and reduced fecundity. By using a ligand blotting assay, a putative mosquito midgut FH receptor could be detected. Thereby, we have identified a novel mechanism whereby mosquitoes can tolerate human blood.

Abstract

AI is one of the biggest megatrends towards the 4th industrial revolution. Although these technologies promise business sustainability as well as product and process quality, it seems that the ever-changing market demands, the complexity of technologies and fair concerns about privacy, impede broad application and reuse of Artificial Intelligence (AI) models across the industry. To break the entry barriers for these technologies and unleash its full potential, the knowlEdge project will develop a new generation of AI methods, systems, and data management infrastructure. Subsequently, as part of the knowlEdge project we propose several major innovations in the areas of data management, data analytics and knowledge management including (i) a set of AI services that allows the usage of edge deployments as computational and live data infrastructure as well as a continuous learning execution pipeline on the edge, (ii) a digital twin of the shop-floor able to test AI models, (iii) a data management framework deployed along the edge-to-cloud continuum ensuring data quality, privacy and confidentiality, (iv) Human-AI Collaboration and Domain Knowledge Fusion tools for domain experts to inject their experience into the system, (v) a set of standardisation mechanisms for the exchange of trained AI models from one context to another, and (vi) a knowledge marketplace platform to distribute and interchange trained AI models. In this paper, we present a short overview of the EU Project knowlEdge –Towards Artificial Intelligence powered manufacturing services, processes, and products in an edge-to-cloud-knowledge continuum for humans [in-the-loop], which is funded by the Horizon 2020 (H2020) Framework Programme of the European Commission under Grant Agreement 957331. Our overview includes a description of the project’s main concept and methodology as well as the envisioned innovations. Abstract

The COVID-19 pandemic has already caused more than 150,000,000 cases worldwide. In Spain this has lead to a massive and simultaneous saturation of all sanitary regions. Coherently, the quick and consistent understanding of the COVID-19 disease requires of the combined analysis of thousands of medical records generated by dozens of different institutions. In the context of the publicly funded CIBERES-UCI-COVID project, we have gathered, cleaned and preprocessed data from heterogeneous sources - more than 30 hospitals, with different data entry systems - in order to produce a unified database, of more than 6.000 patients, that is used in several clinical studies being carried by different multidisciplinary groups. In this paper, we identify the complexities we encountered, the solutions we applied, and we summarise the statistical and machine learning techniques we have applied for the studies. © 2021 The authors and IOS Press.

Abstract

This work aimed to develop a textural quality classification function for frozen hake fillets using instrumental measurements. Frozen hake fillets were stored to provide a wide range of textures. Raw and cooked fillets were analyzed for texture using Kramer and Warner-Bratzler shear cells and a puncture device. Apparent viscosity, dimethylamine and protein solubility were also measured. Principal Component Analysis indicated three factors accounting for 86.5% of the variance. Four clusters were found, ranging from excellent quality (low texture, high viscosity) to very poor (high texture, low viscosity). Parameters required for classification were viscosity, maximum force from Kramer and maximum force and energy from the puncture test., This research was funded by project COR002/94, Comunidad de Madrid.

Abstract

In this article, we present the designof an mHealth system to support senior citizens with chronic diseases in managing their adherenceto pharmacological treatment. The DIALCAT system consisting of a smart pill dispenser and a mobile application will be tested in a clinical study for 18 months by patients suffering diabetes with mild cognitive impairment. The clinical trial aims to study the effectof diabetes as an accelerator of cognitive impairment and Alzheimer’s disease and the impact of adherenceto treatment. DIALCAT is a RIS3CAT project co-fundedunder the European ERDF programme.
