4. From hidden hearing loss to supranormal auditory processing by neurotrophin 3-mediated modulation of inner hair cell synapse density.

Abstract

Loss of synapses between spiral ganglion neurons and inner hair cells (IHC synaptopathy) leads to an auditory neuropathy called hidden hearing loss (HHL) characterized by normal auditory thresholds but reduced amplitude of sound-evoked auditory potentials. It has been proposed that synaptopathy and HHL result in poor performance in challenging hearing tasks despite a normal audiogram. However, this has only been tested in animals after exposure to noise or ototoxic drugs, which can cause deficits beyond synaptopathy. Furthermore, the impact of supernumerary synapses on auditory processing has not been evaluated. Here, we studied mice in which IHC synapse counts were increased or decreased by altering neurotrophin 3 (Ntf3) expression in IHC supporting cells. As we previously showed, postnatal Ntf3 knockdown or overexpression reduces or increases, respectively, IHC synapse density and suprathreshold amplitude of sound-evoked auditory potentials without changing cochlear thresholds. We now show that IHC synapse density does not influence the magnitude of the acoustic startle reflex or its prepulse inhibition. In contrast, gap-prepulse inhibition, a behavioral test for auditory temporal processing, is reduced or enhanced according to Ntf3 expression levels. These results indicate that IHC synaptopathy causes temporal processing deficits predicted in HHL. Furthermore, the improvement in temporal acuity achieved by increasing Ntf3 expression and synapse density suggests a therapeutic strategy for improving hearing in noise for individuals with synaptopathy of various etiologies. The auditory neuropathy hidden hearing loss (HHL) is characterised by the loss of cochlear inner hair cell synapses, which reduce the amplitude of sound-evoked auditory potentials. This study shows that loss of these synapses degrades temporal auditory processing and that the generation of supernumerary synapsis results in above-normal temporal auditory processing in mice, potentially improving hearing in HHL patients. The use of electrochemical stimuli to manipulate intercomponent interactions is a powerful method for controlling and observing submolecular motion in hydrogen bonded molecular shuttles. In a benzylic amide macrocycle-based rotaxane system, the motion resulting from reduction and reoxidation of the naphthalimide station was unequivocally shown to be reversible shuttling of the macrocycle along the thread.

Abstract

A simple, inexpensive, conceptually uncomplicated student experiment involving adsorption of polyoxometalate on graphite electrodes has been developed for teaching surface electrochemistry. The experiment has been used for fourth-year students. Among things students will become familiar with are cyclic voltammetry, capacitive current, and spontaneous chemisorption. Much research is being done in the area of modified electrodes and students should be introduced to it.

23. Aspects of helium production and transport in the continents

Abstract

This work examines the isotopic composition and abundance of helium in a number of different crustal environments with a view to understanding its production and transport in the crust. The work was largely carried out using existing instrumentation, but a dedicated interface was also built to allow computer control of a quadrupole mass spectrometer for rare gas abundance pattern determination. Conventional calculations of the radiogenic 3He/4He production ratio based on the assumption of a chemically homogeneous composition cannot adequately account for the 3He/4He ratio in waters from the Carnmenellis Granite of SW England. Alpha tracking and back-scattered electron microscopy show that the majority of the U and Th are concentrated in volumetrically insignificant accessory minerals. A new model has been devised, taking this heterogeneity into account, to explain the Carnmenellis data. Helium isotopic analysis of the granite itself revealed isotopic disequilibrium with the circulating waters. This may be related to differential release of 3He and 4He associated with different formation sites. A survey was made of the helium abundance and isotopic composition of groundwaters from the Pannonian Basin of Hungary in order to study the behaviour of mantle-derived fluids in an area of major recent crustal extension and volcanism. More than 80 samples were analysed covering most of the basin, and almost all contained a component of mantle-derived helium. Although 3He/4He ratio is not clearly correlated with the surface expression of volcanism, it may act as an indicator of intrusion at depth. The 3He flux through the Hungarian crust is ≈4 atoms.cm-2.s-2. If the mechanism of extraction is partial melting, then by analogy with melt production at mid-ocean ridges, addition of a 20-40 metre layer of basalt (for 5-10% partial melting) must be added to the Hungarian crust in a million years to support the present day 3He flux.

Abstract

International audience; Commercial PC105 titanium dioxide nanoparticles were studied under mechanical milling process. The effect of milling time and speed on the structural and electronic properties of TiO 2 powder was then investigated using X-ray powder diffraction (XRD), dynamic light scattering (DLS), transmission electronic microscopy (TEM), electron param-agnetic resonance (EPR) and UV-visible spectroscopy. The related photo-catalytic properties of the milled nanoparticles were probed following the degradation rate of methylene orange (MO) under UV-light irradiation and through EPR spin-scavenging approach. Comparison with pristine powder shows that milled nanoparticles are significantly less reactive upon illumination, despite decreased radius and hence, higher specific area. Such low yield of reactive species is attributed to the apparition of the amorphous TiO 2 and brookite phase upon milling, as well as increased charge carrier recombination as pointed out by the presence of sacrificial electron donor.

Abstract

The synthesis, structures and magnetic properties of some new [Fe.sup.III] and [Fe.sup.II] complexes are described, which has shown the rich coordination chemistry of the multidentate hydrazido salicyl derivative ligands bearing sulfur groups. The studies have shown an unexpected spontaneous reduction of the new [Fe.sup.III][(HL).sub.2]Cl,S complex to the new [Fe.sup.II][([H.sub.2]L).sub.2][Cl.sub.2] high-spin mononuclear complex.

Abstract

Residual dipolar couplings (RDCs) of the wild-type nuclease are compared with those of a fragment that retains a folded OB-fold subdomain structure despite missing the last 47 o 149 residues. The results suggested that RDCs are more sensitive to structural changes in folded than unfolded proteins.

Abstract

Here, we investigate remodeling of hippocampal cholinergic inputs after noise exposure and determine the relevance of these changes to tinnitus. To assess the effects of noise exposure on the hippocampus, guinea pigs were exposed to unilateral noise for 2 hr and 2 weeks later, immunohistochemistry was performed on hippocampal sections to examine vesicular acetylcholine transporter (VAChT) expression. To evaluate whether the changes in VAChT were relevant to tinnitus, another group of animals was exposed to the same noise band twice to induce tinnitus, which was assessed using gap‐prepulse Inhibition of the acoustic startle (GPIAS) 12 weeks after the first noise exposure, followed by immunohistochemistry. Acoustic Brainstem Response (ABR) thresholds were elevated immediately after noise exposure for all experimental animals but returned to baseline levels several days after noise exposure. ABR wave I amplitude‐intensity functions did not show any changes after 2 or 12 weeks of recovery compared to baseline levels. In animals assessed 2‐weeks following noise‐exposure, hippocampal VAChT puncta density decreased on both sides of the brain by 20–60% in exposed animals. By 12 weeks following the initial noise exposure, changes in VAChT puncta density largely recovered to baseline levels in exposed animals that did not develop tinnitus, but remained diminished in animals that developed tinnitus. These tinnitus‐specific changes were particularly prominent in hippocampal synapse‐rich layers of the dentate gyrus and areas CA3 and CA1, and VAChT density in these regions negatively correlated with tinnitus severity. The robust changes in VAChT labeling in the hippocampus 2 weeks after noise exposure suggest involvement of this circuitry in auditory processing. After chronic tinnitus induction, tinnitus‐specific changes occurred in synapse‐rich layers of the hippocampus, suggesting that synaptic processing in the hippocampus may play an important role in the pathophysiology of tinnitus.
