Abstract

We present an updated ephemeris and physical parameters for the exoplanet WASP-77 A b. In this effort, we combine 64 ground- and space-based transit observations, 6 space-based eclipse observations, and 32 radial velocity observations to produce the most precise orbital solution to date for this target, aiding in the planning of James Webb Space Telescope (JWST) and Ariel observations and atmospheric studies. We report a new orbital period of 1.360029395 +- 5.7e-8 days, a new mid-transit time of 2459957.337860 +- 4.3e-5 BJDTDB (Barycentric Julian Date in the Barycentric Dynamical Time scale; arXiv:1005.4415) and a new mid-eclipse time of 2459956.658192 +- 6.7e-5 BJDTDB. Furthermore, the methods presented in this study reduce the uncertainties in the planet mass to 1.6654 +- 4.5e-3 Mjup and orbital period to 1.360029395 +- 5.7e-8 days by factors of 15.1 and 10.9, respectively. Through a joint fit analysis comparison of transit data taken by space-based and citizen science-led initiatives, our study demonstrates the power of including data collected by citizen scientists compared to a fit of the space-based data alone. Additionally, by including a vast array of citizen science data from ExoClock, Exoplanet Transit Database (ETD), and Exoplanet Watch, we can increase our observational baseline and thus acquire better constraints on the forward propagation of our ephemeris than what is achievable with TESS data alone., Comment: Updated a co-author name. Abstract

NASA's Transiting Exoplanet Survey Satellite (TESS) has identified over 7,000 candidate exoplanets via the transit method, with gas giants among the most readily detected due to their large radii. Even so, long intervals between TESS observations for much of the sky lead to candidates for which only a single transit is detected in one TESS sector, leaving those candidate exoplanets with unconstrained orbital periods. Here, we confirm the planetary nature of TIC 393818343 b, originally identified via a single TESS transit, using radial velocity data and ground-based photometric observations from citizen scientists with the Unistellar Network and Exoplanet Watch. We determine a period of $P$ = 16.24921 $\substack{+0.00010 \\ -0.00011}$ days, a mass $M_{P}$ = 4.34 $\pm$ 0.15 $M_{J}$, and semi-major axis $a$ = 0.1291 $\substack{+0.0021 \\ -0.0022}$ au, placing TIC 393818343 b in the "warm Jupiter" population of exoplanets. With an eccentricity $e$ = 0.6058 $\pm$ 0.0023, TIC 393818343 b is the most eccentric warm Jupiter to be discovered by TESS orbiting less than 0.15 au from its host star and therefore an excellent candidate for follow-up, as it may inform our future understanding of how hot and warm Jupiter populations are linked., Comment: 13 pages, 4 figures, 3 tables. Abstract

NASA Citizen Scientists have used Exoplanet Transit Interpretation Code (EXOTIC) to reduce 40 sets of time-series images of WASP-12 taken by privately owned telescopes and a 6-inch telescope operated by the Center for Astrophysics | Harvard & Smithsonian MicroObservatory (MOBs). Of these sets, 24 result in clean transit light curves of WASP-12 b which are included in the NASA Exoplanet Watch website. We use priors from the NASA Exoplanet Archive to calculate the ephemeris of the planet and combine it with ETD (Exoplanet Transit Database), ExoClock, and TESS (Transiting Exoplanet Survey Satellite) observations. Combining these datasets gives an updated ephemeris for the WASP-12 b system of 2454508.97923 +/- 0.000051 BJDTDB with an orbital period of 1.09141935 +/- 2.16e-08 days which can be used to inform the efficient scheduling of future space telescope observations. The orbital decay of the planet was found to be -6.89e-10 +/- 4.01e-11 days/epoch. Abstract

El presente artículo de revisión tuvo como objetivo describir al salario emocional desde la perspectiva organizacional en países de América Latina y establecer las principales características y medios para lograr su implementación efectiva, del proceso de revisión sistemática a 80 artículos científicos en las bases de búsqueda de Scopus y Web of Science, se seleccionaron para la elaboración del presente artículo de revisión 60 investigaciones por su pertinencia y relevancia, empleando los principios de ética investigativa, resultando que la implementación del salario emocional en países de América Latina, dependerá del sector, el país y la adaptabilidad organizacional. Sin embargo, del análisis realizado es indudable que el salario emocional es una estrategia de suma importancia que genera bienestar emocional y psicológico; permitiendo el incremento del desempeño laboral en los colaboradores. Finalmente, se concluye que esta se caracteriza por la flexibilidad y la generación de un ambiente laboral positivo, además de promover el desarrollo profesional y personal de los colaboradores, fomentando la adquisición de competencias organizacionales.

Abstract

The increasing availability of complex survey data, and the continued need for estimates of demographic and health indicators at a fine spatial and temporal scale, which leads to issues of data sparsity, has led to the need for spatio-temporal smoothing methods that acknowledge the manner in which the data were collected. The open source R package SUMMER implements a variety of methods for spatial or spatio-temporal smoothing of survey data. The emphasis is on small-area estimation. We focus primarily on indicators in a low and middle-income countries context. Our methods are particularly useful for data from Demographic Health Surveys and Multiple Indicator Cluster Surveys. We build upon functions within the survey package, and use INLA for fast Bayesian computation. This paper includes a brief overview of these methods and illustrates the workflow of accessing and processing surveys, estimating subnational child mortality rates, and visualizing results with both simulated data and DHS surveys.

13. Modeling microbial abundances and dysbiosis with beta-binomial regression

Abstract

Using a sample from a population to estimate the proportion of the population with a certain category label is a broadly important problem. In the context of microbiome studies, this problem arises when researchers wish to use a sample from a population of microbes to estimate the population proportion of a particular taxon, known as the taxon's relative abundance. In this paper, we propose a beta-binomial model for this task. Like existing models, our model allows for a taxon's relative abundance to be associated with covariates of interest. However, unlike existing models, our proposal also allows for the overdispersion in the taxon's counts to be associated with covariates of interest. We exploit this model in order to propose tests not only for differential relative abundance, but also for differential variability. The latter is particularly valuable in light of speculation that dysbiosis, the perturbation from a normal microbiome that can occur in certain disease conditions, may manifest as a loss of stability, or increase in variability, of the counts associated with each taxon. We demonstrate the performance of our proposed model using a simulation study and an application to soil microbial data.

Abstract

For more than a decade, the Joint Center for Structural Genomics (JCSG; www.jcsg.org) worked toward increased three-dimensional structure coverage of the protein universe. This coordinated quest was one of the main goals of the four high-throughput (HT) structure determination centers of the Protein Structure Initiative (PSI; www.nigms.nih.gov/Research/specificareas/PSI). To achieve the goals of the PSI, the JCSG made use of the complementarity of structure determination by X-ray crystallography and nuclear magnetic resonance (NMR) spectroscopy to increase and diversify the range of targets entering the HT structure determination pipeline. The overall strategy, for both techniques, was to determine atomic resolution structures for representatives of large protein families, as defined by the Pfam database, which had no structural coverage and could make significant contributions to biological and biomedical research. Furthermore, the experimental structures could be leveraged by homology modeling to further expand the structural coverage of the protein universe and increase biological insights. Here, we describe what could be achieved by this structural genomics approach, using as an illustration the contributions from 20 NMR structure determinations out of a total of 98 JCSG NMR structures, which were selected because they are the first three-dimensional structure representations of the respective Pfam protein families. The information from this small sample is representative for the overall results from crystal and NMR structure determination in the JCSG. There are five new folds, which were classified as domains of unknown functions (DUF), three of the proteins could be functionally annotated based on three-dimensional structure similarity with previously characterized proteins, and 12 proteins showed only limited similarity with previous deposits in the Protein Data Bank (PDB) and were classified as DUFs.

Abstract

Toca 511 (vocimagene amiretrorepvec), a nonlytic, amphotropic retroviral replicating vector (RRV), encodes and delivers a functionally optimized yeast cytosine deaminase (CD) gene to tumors. In orthotopic glioma models treated with Toca 511 and 5-fluorocytosine (5-FC) the CD enzyme within infected cells converts 5-FC to 5-fluorouracil (5-FU), resulting in tumor killing. Toca 511, delivered locally either by intratumoral injection or by injection into the resection bed, in combination with subsequent oral extended-release 5-FC (Toca FC), is under clinical investigation in patients with recurrent high-grade glioma (HGG). If feasible, intravenous administration of vectors is less invasive, can easily be repeated if desired, and may be applicable to other tumor types. Here, we present preclinical data that support the development of an intravenous administration protocol. First we show that intravenous administration of Toca 511 in a preclinical model did not lead to widespread or uncontrolled replication of the RVV. No, or low, viral DNA was found in the blood and most of the tissues examined 180 days after Toca 511 administration. We also show that RRV administered intravenously leads to efficient infection and spread of the vector carrying the green fluorescent protein (GFP)-encoding gene (Toca GFP) through tumors in both immune-competent and immune-compromised animal models. However, initial vector localization within the tumor appeared to depend on the mode of administration. Long-term survival was observed in immune-competent mice when Toca 511 was administered intravenously or intracranially in combination with 5-FC treatment, and this combination was well tolerated in the preclinical models. Enhanced survival could also be achieved in animals with preexisting immune response to vector, supporting the potential for repeated administration. On the basis of these and other supporting data, a clinical trial investigating intravenous administration of Toca 511 in patients with recurrent HGG is currently open and enrolling.

45. Measurement and classification of scapular kinematics

Abstract

Shoulder pain is associated with abnormal movement of the scapula. Quantitative measurement and classification of abnormal scapular kinematics, however, is difficult due to the gliding nature of the scapula beneath the skin surface, and large variation in data. The aims of this study were to determine the validity and reliability of the acromion marker cluster (AMC) in measuring scapular kinematics during the arm lowering phase, to measure poor control of the scapula in people with a history of shoulder pain and a group with shoulder impingement, and objectively classify poor control scapular control. The validity and reliability of the AMC was compared to the scapular locator. The AMC was valid during sagittal and scapular plane arm lowering movements (max root mean square error = 6.1°), but had poor to fair reliability (ICC = 0.12 – 0.76). The AMC was used to determine whether a group of people with a history of pain, and those with shoulder impingement, exhibited poor scapular control when performing a scapular repositioning clinical manoeuvre, compared to a healthy control group. The history of pain group exhibited an increased position of scapular downward rotation (-7.9° ± 6.3) at the end of the manoeuvre when compared to the healthy control group (0.3° ± 4.4). There were no statistical differences in people with shoulder impingement. A statistical classification technique based on the Dempster-Shafer Theory of Evidence (DST) was used to objectively classify participants as having poor scapular control. Six kinematic variables were was used as input in to the DST classifier which classified 5 out of the 6 history of pain participants as having poor scapular control, with an accuracy of 72%. The DST multivariate classifier was reasonably successful at classifying participants with poor scapular control suggesting its potential use for future analysis of abnormal scapular kinematics.

Abstract

Patients with the most common and aggressive form of high-grade glioma, glioblastoma multiforme, have poor prognosis and few treatment options. In 2 immunocompetent mouse brain tumor models (CT26-BALB/c and Tu-2449-B6C3F1), we showed that a nonlytic retroviral replicating vector (Toca 511) stably delivers an optimized cytosine deaminase prodrug activating gene to the tumor lesion and leads to long-term survival after treatment with 5-fluorocytosine (5-FC). Survival benefit is dose dependent for both vector and 5-FC, and as few as 4 cycles of 5-FC dosing after Toca 511 therapy provides significant survival advantage. In the virally permissive CT26-BALB/c model, spread of Toca 511 to other tissues, particularly lymphoid tissues, is detectable by polymerase chain reaction (PCR) over a wide range of levels. In the Tu-2449-B6C3F1 model, Toca 511 PCR signal in nontumor tissues is much lower, spread is not always observed, and when observed, is mainly detected in lymphoid tissues at low levels. The difference in vector genome spread correlates with a more effective antiviral restriction element, APOBEC3, present in the B6C3F1 mice. Despite these differences, neither strain showed signs of treatment-related toxicity. These data support the concept that, in immunocompetent animals, a replicating retroviral vector carrying a prodrug activating gene (Toca 511) can spread through a tumor mass, leading to selective elimination of the tumor after prodrug administration, without local or systemic pathology. This concept is under investigation in an ongoing phase I/II clinical trial of Toca 511 in combination with 5-FC in patients with recurrent high-grade glioma (www.clinicaltrials.gov NCT01156584).
