Background: Combining a mobile application-based vestibular diary called the DizzyQuest and an iPad-based hearing test enables evaluation of the relationship between experienced neuro-otological symptoms and hearing thresholds in daily life setting. The aim was to investigate the relationship between self-reported hearing symptoms and hearing thresholds in patients with Meniere's disease (MD), using the DizzyQuest and the iPad-based hearing test simultaneously. Methods: The DizzyQuest was administered for 3 weeks in 21 patients. Using the experience-sampling-method (ESM), it assessed hearing loss and tinnitus severity for both ears separately. Each day after the DizzyQuest, an iPad-based hearing test was used to measure hearing thresholds. A mixed model regression analysis was performed to investigate relationships between hearing thresholds and self-reported hearing loss and tinnitus severity. Results: Fifteen patients were included. Overall, pure-tone averages (PTAs) were not correlated with self-reported hearing loss severity and tinnitus. Individual differences in PTA results between both ears did not significantly influence the difference in self-reported hearing loss severity between both ears. Self-reported hearing loss and tinnitus scores were significantly higher in ears that corresponded with audiometric criteria of MD (p < 0.001). Self-reported tinnitus severity significantly increased with self-reported hearing loss severity in affected (p = 0.011) and unaffected ears (p < 0.001). Conclusion: Combining the DizzyQuest and iPad-based hearing test, facilitated assessment of self-reported hearing loss and tinnitus severity and their relationship with hearing thresholds, in a daily life setting. This study illustrated the importance of investigating neuro-otological symptoms at an individual level, using multiple measurements. Background: Most questionnaires currently used for assessing symptomatology of vestibular disorders are retrospective, inducing recall bias and lowering ecological validity. An app-based diary, administered multiple times in daily life, could increase the accuracy and ecological validity of symptom measurement. The objective of this study was to introduce a new experience sampling method (ESM) based vestibular diary app (DizzyQuest), evaluate response rates, and to provide examples of DizzyQuest outcome measures which can be used in future research. Methods: Sixty-three patients diagnosed with a vestibular disorder were included. The DizzyQuest consisted of four questionnaires. The morning- and evening-questionnaires were administered once each day, the within-day-questionnaire 10 times a day using a semi-random time schedule, and the attack questionnaire could be completed after the occurrence of a vertigo or dizziness attack. Data were collected for 4 weeks. Response rates and loss-to-follow-up were determined. Reported symptoms in the within-day-questionnaire were compared within and between patients and subgroups of patients with different vestibular disorders. Results: Fifty-one patients completed the study period. Average response rates were significantly higher than the desired response rate of > 50% (p < 0.001). The attack-questionnaire was used 159 times. A variety of neuro-otological symptoms and different disease profiles were demonstrated between patients and subgroups of patients with different vestibular disorders. Conclusion: The DizzyQuest is able to capture vestibular symptoms within their psychosocial context in daily life, with little recall bias and high ecological validity. The DizzyQuest reached the desired response rates and showed different disease profiles between subgroups of patients with different vestibular disorders. Background: The DizzyQuest, an app-based vestibular diary, provides the opportunity to capture the number and nature of vertigo attacks in daily life. To accomplish this, the DizzyQuest provides different strategies: event sampling using an attack questionnaire, and time sampling using an evening questionnaire. Objective of this study was to investigate whether the number and nature of reported vertigo attacks was comparable between the two questionnaires. Methods: Fifty-seven patients, who reported vertigo attacks, used the DizzyQuest for on average 24 days. The number and nature (including symptoms, triggers and duration) of vertigo attacks were compared between the attack and the evening questionnaire. Results: The attack questionnaire was used 192 times. In contrast, at least 749 new vertigo attacks were reported in 446 evening questionnaires. A vertigo attack was not always reported in both questionnaires during the same day. Vertigo attacks that were most likely captured by both questionnaires were not always reported the same in both questionnaires regarding triggers and duration. Conclusion: Event sampling using an attack questionnaire has low recall bias and, therefore, reliably captures the nature of the attack, but induces a risk of under-sampling. Time sampling using an evening questionnaire suffers from recall bias, but seems more likely to capture less discrete vertigo attacks and it facilitates registration of the absence of vertigo attacks. Background: Self‐injurious behaviour (SIB) can be classified as intentional, direct injuring of body tissue usually without suicidal intent. In its non‐suicidal form it is commonly seen as a clinical sign of borderline personality disorder, autism, PTSD, depression, and anxiety affecting a wide range of ages and conditions. In rhesus macaques SIB is most commonly manifested through hair plucking, self‐biting, self‐hitting, and head banging. SIB in the form of self‐biting is observed in approximately 5–15% of individually housed monkeys. Recently, glial cells are becoming recognised as key players in regulating behaviours. Method: The goal of this study was to determine the role of glial activation, including astrocytes, in macaques that had displayed SIB. To this end, we performed immunohistochemistry and next generation sequence of brain tissues from rhesus macaques with SIB. Results: Our studies showed increased vimentin, but not nestin, expression on astrocytes of macaques displaying SIB. Initial RNA Seq analyses indicate activation of pathways involved in tissue remodelling, neuroinflammation and cAMP signalling. Stromal/stem cell differentiation is controlled by a vast array of regulatory mechanisms. Included within these are methods of mRNA gene regulation that occur at the level of epigenetic, transcriptional, and/or posttranscriptional modifications. Current studies that evaluate the posttranscriptional regulation of mRNA demonstrate microRNAs (miRNAs) as key mediators of stem cell differentiation through the inhibition of mRNA translation. miRNA expression is enhanced during both adipogenic and osteogenic differentiation; however, the mechanism by which miRNA expression is altered during stem cell differentiation is less understood. Here we demonstrate for the first time that adipose-derived stromal/stem cells (ASCs) induced to an adipogenic or osteogenic lineage have differences in strand preference (-3p and -5p) for miRNAs originating from the same primary transcript. Furthermore, evaluation of miRNA expression in ASCs demonstrates alterations in both miRNA strand preference and 5'seed site heterogeneity. Additionally, we show that during stem cell differentiation there are alterations in expression of genes associated with the miRNA biogenesis pathway. Quantitative RT-PCR demonstrated changes in the Argonautes (AGO1-4), Drosha, and Dicer at intervals of ASC adipogenic and osteogenic differentiation compared to untreated ASCs. Specifically, we demonstrated altered expression of the AGOs occurring during both adipogenesis and osteogenesis, with osteogenesis increasing AGO1-4 expression and adipogenesis decreasing AGO1 gene and protein expression. These data demonstrate changes to components of the miRNA biogenesis pathway during stromal/stem cell differentiation. Identifying regulatory mechanisms for miRNA processing during ASC differentiation may lead to novel mechanisms for the manipulation of lineage differentiation of the ASC through the global regulation of miRNA as opposed to singular regulatory mechanisms. Describing fish habitat associations and their relevance to conservation remains a central challenge in stream fish ecology. Unfortunately, there are limited opportunities to investigate these associations in unaltered systems and identify critical habitats used by native fishes. Investigation of fish habitat associations in tallgrass prairie is especially vital, owing to their widespread destruction. Our study aim was to identify habitat factors associated with the distribution and density of fishes in two protected tallgrass prairie stream watersheds in eastern Kansas: Kings Creek on the Konza Prairie Biological Station (KPBS) and Fox Creek on the Tallgrass Prairie National Preserve (TPNP). We sampled fishes and measured eight habitat variables at three sites on KPBS (2006-2011) and four sites on TPNP (2008-2011). Multiple regression suggested that species richness was positively associated with pool area (partial r = 0.70) and discharge (partial r = 0.50) in Fox Creek (df = 15, Adj. R² = 0.60, P < 0.001). In Kings Creek, species richness was only associated with pool area (dr = 17, R² = 0.44, P < 0.001). Redundancy analyses showed common prairie fish species exhibit ontogenetic habitat associations, partitioning adults in deep and juveniles in shallow pools. Strong species area relationships in these minimally altered systems indicates large volume habitats have greater species richness, suggesting water diversions or extractions that reduce habitat are likely to cause declines in native biodiversity. A key component in the calculation of reference crop evapotranspiration (ETr) is the weather data. If the weather data have been collected from a station under nonreference conditions, the data itself may contain errors, which will in turn yield inaccurate ETr estimates. It was proposed by Allen in 1996 that data used for evapotranspiration be scrutinized by comparing daily minimum temperature (Tmin) and the daily average dew point temperature (Tdew). If the difference between Tmin and Tdew is greater than 3°C, then the site is considered to be arid (nonreference) and adjustments are recommended for temperature and dew point data. In Arizona, normal weather conditions often occur where Tmin and Tdew do not approach each other. This study examined the appropriateness of applying the conditions set forth by Allen to temperature data collected in central Arizona. Two weather stations were set up in a 35.5 ha alfalfa field in central Arizona to measure dry bulb and wet bulb temperatures. Additionally, plant temperature data were collected to verify field conditions. Daily data were taken for 1.5 years at the University of Arizona’s Maricopa Agricultural Center. Of the 611 days of data collected, the difference between Tmin and Tdew was greater than 3°C on 329 days, indicating that these data were not taken under reference conditions. Among these data, 178 days were verified as nonreference but 151 were verified as actually being under reference conditions. Making adjustments for these days (151 days) resulted in a 47 mm decrease in ETr estimation, which mostly occurred during the summer. [ABSTRACT FROM AUTHOR]

Introduction: For tests reporting continuous results, primary studies usually provide test performance at multiple but often different thresholds. This creates missing data when performing a meta‐analysis at each threshold. A standard meta‐analysis (no imputation [NI]) ignores such missing data. A single imputation (SI) approach was recently proposed to recover missing threshold results. Here, we propose a new method that performs multiple imputation of the missing threshold results using discrete combinations (MIDC). Methods: The new MIDC method imputes missing threshold results by randomly selecting from the set of all possible discrete combinations which lie between the results for 2 known bounding thresholds. Imputed and observed results are then synthesised at each threshold. This is repeated multiple times, and the multiple pooled results at each threshold are combined using Rubin's rules to give final estimates. We compared the NI, SI, and MIDC approaches via simulation. Results: Both imputation methods outperform the NI method in simulations. There was generally little difference in the SI and MIDC methods, but the latter was noticeably better in terms of estimating the between‐study variances and generally gave better coverage, due to slightly larger standard errors of pooled estimates. Given selective reporting of thresholds, the imputation methods also reduced bias in the summary receiver operating characteristic curve. Simulations demonstrate the imputation methods rely on an equal threshold spacing assumption. A real example is presented. Conclusions: The SI and, in particular, MIDC methods can be used to examine the impact of missing threshold results in meta‐analysis of test accuracy studies. [ABSTRACT FROM AUTHOR]

Weedy rice can severely impact rice production through yield reduction because of its competitive ability to reduce growth resources for cultivated rice. The characteristic weedy traits have made weedy rice very challenging to manage as they have the same agro-morphological characteristics with cultivated rice. The study examined the relationships between selected cultivated rice (Oryza sativa L.) cultivars, weedy rice biotypes, and wild rice collected from different locations in the Philippines, using simple sequence repeat markers. Cluster analysis, using UPGMA, enabled the genetic differentiation and relationships examination of the test materials. Subgroups of 13 with at least two biotypes formed 100% similarity based on post-harvest data, with the cultivated rice cultivars forming one subgroup. With polymorphic SSR markers, five major clusters range from three (group I) to 62 biotypes (group III). There was 100% similarity observed for 15 subgroups ranging from two to 10 biotypes. The wild rice cultivars formed species-specific groupings. Subgroups with 100% similarity came from the same province; likewise, one with 100% similarity came from both Iloilo and Batangas; and still another from cultivated rice cultivar and weedy rice biotype from Pangasinan. The possible relationships of weedy rice biotypes with wild rice relatives (>65% similarity) include two biotypes related to Oryza minuta, one for O. meyeriana, and 22 biotypes for O. rufipogon. Concerning cultivated rice cultivars, high similarity (>80%) was observed in 22 biotypes closely related to PSB Rc 82, 13 biotypes to NSIC Rc 222, six to NSIC Rc 160, three biotypes each to NSIC Rc 215, NSIC Rc 152, NSIC Rc 64, NSIC Rc 18, and NSIC Rc 10, and one biotype each to IR64 and NSIC Rc 14.
