Animal African trypanosomosis, caused by blood protozoan parasites transmitted mainly by tsetse flies, represents a major constraint for millions of cattle in sub-Saharan Africa. Exposed cattle include trypanosusceptible indicine breeds, severely affected by the disease, and West African taurine breeds called trypanotolerant owing to their ability to control parasite development, survive and grow in enzootic areas. Until now the genetic basis of trypanotolerance remains unclear. Here, to improve knowledge of the biological processes involved in trypanotolerance versus trypanosusceptibility, we identified bovine genes differentially expressed in five West African cattle breeds during an experimental infection by Trypanosoma congolense and their biological functions. To this end, whole blood genome-wide transcriptome of three trypanotolerant taurine breeds (N’Dama, Lagune and Baoulé), one susceptible zebu (Zebu Fulani) and one African taurine x zebu admixed breed (Borgou) were profiled by RNA sequencing at four time points, one before and three during infection. As expected, infection had a major impact on cattle blood transcriptome regardless of the breed. The functional analysis of differentially expressed genes over time in each breed confirmed an early activation of the innate immune response, followed by an activation of the humoral response and an inhibition of T cell functions at the chronic stage of infection. More importantly, we highlighted overlooked features, such as a strong disturbance in host metabolism and cellular energy production that differentiates trypanotolerant and trypanosusceptible breeds. N’Dama breed showed the earliest regulation of immune response, associated with a strong activation of cellular energy production, also observed in Lagune, and to a lesser extent in Baoulé. Susceptible Zebu Fulani breed differed from other breeds by the strongest modification in lipid metabolism regulation. Overall, this study provides a better understanding of the biological mechanisms at work during infection, especially concerning the interplay between immunity and metabolism that seems differentially regulated depending on the cattle breeds.

BackgroundBeyond the question of short-term survival, days spent at home could be considered a patient-centered outcome in critical care trials.Research questionWhat are the days spent at home and health care trajectories during the year after surviving critical illness?Study design and methodsData were extracted on adult survivors spending at least 2 nights in a French ICU during 2018 who were treated with invasive mechanical ventilation or vasopressors or inotropes. Trauma, burn, organ transplant, stroke, and neurosurgical patients were excluded. Stays at home, death, and hospitalizations were reported before and after ICU stay, using state sequence analysis. An unsupervised clustering method was performed to identify cohorts based on post-ICU trajectories.ResultsOf 77,132 ICU survivors, 89% returned home. In the year after discharge, these patients spent a median of 330 (interquartile range [IQR], 283-349) days at home. At 1 year, 77% of patients were still at home and 17% had died. Fifty-one percent had been re-hospitalized, and 10% required a further ICU admission. Forty-eight percent used rehabilitation facilities, and 5.7%, hospital at home. Three clusters of patients with distinct post-ICU trajectories were identified. Patients in cluster 1 (68% of total) survived and spent most of the year at home (338 [323-354] days). Patients in cluster 2 (18%) had more complex trajectories, but most could return home (91%), spending 242 (174-277) days at home. Patients in cluster 3 (14%) died, with only 37% returning home for 45 (15-90) days.InterpretationMany patients had complex health care trajectories after surviving critical illness. Wide variations in the ability to return home after ICU discharge were observed between clusters, which represents an important patient-centered outcome.

Despite a growing interest in emotions in sport psychology, little has been written about the contextual elements triggering athletes' emotional experience. This single case study aimed to analyze the inducers of an elite table tennis player's emotional experience throughout his career. He was ranked among the 15 best players in the world, and his career spanned more than 20 years. Inspired by the critical-incident method, we conducted a lengthy interview with the player, during which he related the most significant moments of his career. The categorization of the 96 critical incidents collected highlighted four inducers of positive emotions and five inducers of negative emotions, emerging over five periods (exponential progression, first difficulties, major difficulties, second career, and end of career). These findings contribute to the development of an innovative single-case-study design, allowing an understanding of the contextual origin of athletes' emotions over the long term. Finally, highlights are discussed and recommendations for the practice of sport psychology are formulated. [ABSTRACT FROM AUTHOR]

Introduction: In France, land use is dominated by a narrow range of crops. As crop diversification is increasingly promoted to transform the food system, outlets for diversification crops need to be identified. We assess the scope for introducing diversification crops from farmers' fields to canteen plates, and the agroecological, environmental, work-related and nutritional impacts of their introduction. Methods: We focus on three diversification crops: chickpea, squash, and millet, to partly replace soft wheat, potato, and durum wheat, respectively. For each crop, we define three scenarios of low (1), medium (2) and high (3) integration into institutional catering on a national scale. We assess the scenarios against the amount of diversification cropland area needed and their expected impacts. Results: Diversification crops can be developed through institutional catering. Taking the case of chickpea, in Scenario 1, chickpea flour is introduced in a cake served once every 20 days, replacing 30% of the soft wheat flour used. This amounts to 874 ha cropped. In Scenario 2, chickpea flour partly replaces wheat flour in all preparations, and 4,048 ha are required. In Scenario 3, raw chickpeas are also introduced into new recipes and 20,958 ha are needed. All three diversification crops support the establishment of agroecological cropping systems, with enhanced nutrient cycling in particular, and reduce their environmental footprint, notably by avoiding pesticide use. Diversification crops do not generate work constraints for farmers and cooks. However, they have a lower productivity per unit area than the reference crops, although they do improve several nutrition parameters such as fiber intake. Discussion: We show for the first time that institutional catering provides a tangible outlet for developing diversification crops and improving the sustainability of agriculture. Our scenarios can support concerted actions between farmers, supply-chain actors, cooks and policy makers, as well as communication to eaters surrounding their impacts on agriculture. Sugarcane (Saccharum spp.) is a major crop for sugar and bioenergy production. Its highly polyploid, aneuploid, heterozygous, and interspecific genome poses major challenges for producing a reference sequence. We exploited colinearity with sorghum to produce a BAC-based monoploid genome sequence of sugarcane. A minimum tiling path of 4660 sugarcane BAC that best covers the gene-rich part of the sorghum genome was selected based on whole-genome profiling, sequenced, and assembled in a 382-Mb single tiling path of a high-quality sequence. A total of 25,316 protein-coding gene models are predicted, 17% of which display no colinearity with their sorghum orthologs. We show that the two species, S. officinarum and S. spontaneum, involved in modern cultivars differ by their transposable elements and by a few large chromosomal rearrangements, explaining their distinct genome size and distinct basic chromosome numbers while also suggesting that polyploidization arose in both lineages after their divergence.
