The study introduces a preliminary approach to enhance the supervision of food processing operators. Specifically, it showcases a digital model of an online compliance validation system with environmental conditions designed for food processing and preservation items that are sensitive to climate factors. The technology demonstration involves four fundamental stages carried out in a complete experimental cycle, where intelligent sensors gather and transmit data through a low-power wireless wide-area network from a farm specializing in sunflower seed storage. Subsequently, six consecutive scripts are executed, five of which are coded in Python. The integration of distributed data technology brings objectivity to the process of securely storing environmental parameters and objectively verifying their compliance with recommended hygienic, bio certification standards. As a result, monitoring the food production process becomes more publicly accessible and with higher rates of objectivity in decisions, enabling prospective buyers and certification agencies to assess the quality of food products with great precision. Therefore, the suggested digital model has the potential to ensure the utmost transparency in the food processing sector.

The advancement of smart metering technology is progressing steadily and inevitably across various key economic sectors. The utilizatio.n of remote sensors in agriculture presents unique characteristics and specific challenges. In this study, an on-site experiment was carried out on a Slovakian production farm to analyze the transition from traditional measurement methods to smart meters, focusing on timing decisions related to soil preparation and sowing and their relation to scientifically justified dates. Consequently, a clear distinction was observed in terms of the timing decisions made regarding agricultural activities during traditional, combined, and scientifically based approaches in meteorological data readings. This study contrasts these three scenarios and deliberates on the factors that need to be carefully evaluated before incorporating remote sensors into agricultural processes. This study serves as a valuable resource for individuals involved in the adoption of smart metering practices in the Eastern European agricultural sector and promotes an improved understanding of the interactions within smart-sensing, scientific developments, and land management that contribute to the goal of land-system sustainability.

Ammonia (NH3) emissions have a negative impact on the welfare of breeding animals, human health, and the environment. These influences of modern intensive agriculture have led to numerous protocols, national regulations, and European Directives. Following previous regulatory measures, the Commission Implementing Decision European Union (EU) 2017/302 on 15 February 2017 has established best available technique (BAT) conclusions, under Directive 2010/75/EU of the European Parliament and the Council, for the intensive rearing of poultry and pigs. This applies to intensive poultry and pig producers with a capacity of over 40,000 poultry, 750 sows, or 2000 fattening pigs. Due to the application of this directive, air emissions have been reduced by between 40% and 75% over the last 15 years. The integrated permit monitors the entire environmental burden of the farm on its surroundings (air pollution, water, soil pollution, waste production, energy use). This review aims to provide a critical overview of how member states (including the United Kingdom) are approaching the implementation of IPPC (Integrated Pollution Prevention and Control) and the conclusions of BAT in their legislation and related documents, and how they monitor NH3 emissions from intensive livestock farming. The data for this review were obtained from 2019 to 2020.

The influence of atmospheric plasma discharge (APD) of the Gliding Arc type and low-pressure microwave plasma discharge (LPMD) on the class of various seeds was investigated. Pea, wheat, and sunflower seeds, representing legumes, cereals, and oilseeds, respectively, were selected for the treatment. Our study aimed to verify the effect of plasma treatment on the water contact angle and the wettability of the seeds. Treatment time, working gas flow and microwave power were varied to determine their impact. All treated and untreated variants were used to conduct the water contact angle measurements to compare their wettability. APD treatment seemed to be utterly ineffective for improving the wettability for most process parameters. On the contrary, LPMD manifested a much more efficient impact. The maximum effect was found for the 800 W microwave power applied to the pea seed for 30 s. The contact angle achieved decreased by approximately 50% compared to the untreated sample. These results indicate that LPMD may be an effective alternative to traditional pre-sowing seed treatments used in agriculture for water intake enhancement. Still, it is strongly dependent on the seed’s type and the used process parameters.

Rolling bearings play a pivotal role in rotating machinery. The remaining useful life prediction and fault diagnosis of bearings are crucial to condition-based maintenance. However, traditional data-driven methods usually require manual extraction of features, which needs rich signal processing theory as support and is difficult to control the efficiency. In this study, a bearing remaining life prediction and fault diagnosis method based on short-time Fourier transform (STFT) and convolutional neural network (CNN) has been proposed. First, the STFT was adopted to construct time-frequency maps of the unprocessed original vibration signals that can ensure the true and effective recovery of the fault characteristics in vibration signals. Then, the training time-frequency maps were used as an input of the CNN to train the network model. Finally, the time-frequency maps of testing signals were inputted into the network model to complete the life prediction or fault identification of rolling bearings. The rolling bearing life-cycle datasets from the Intelligent Management System were applied to verify the proposed life prediction method, showing that its accuracy reaches 99.45%, and the prediction effect is good. Multiple sets of validation experiments were conducted to verify the proposed fault diagnosis method with the open datasets from Case Western Reserve University. Results show that the proposed method can effectively identify the fault classification and the accuracy can reach 95.83%. The comparison with the fault diagnosis classification effects of backpropagation (BP) neural network, particle swarm optimization-BP, and genetic algorithm-BP further proves its superiority. The proposed method in this paper is proved to have strong ability of adaptive feature extraction, life prediction, and fault identification.

This paper considers the evolution of processes applied in agriculture for field operations developed from non-organized handmade activities into very specialized and organized production processes. A set of new approaches based on the application of metaheuristic optimization methods and smart automatization known as Agriculture 4.0 has enabled a rapid increase in in-field operations’ productivity and offered unprecedented economic benefits. The aim of this paper is to review modern approaches to agriculture machinery movement optimization with applications in sugarcane production. Approaches based on algorithms for the division of spatial configuration, route planning or path planning, as well as approaches using cost parameters, e.g., energy, fuel and time consumption, are presented. The combination of algorithmic and economic methodologies including evaluation of the savings and investments and their cost/benefit relation is discussed.

Abstract Background Monocytes comprise subsets of classical, intermediate and non-classical monocytes with distinct anti- or pro-tumor effects in breast cancer (BC). They are modulated by estrogen, and can contribute to BC control by endocrine therapy in preclinical models. Methods To elucidate whether changes in monocyte subsets are associated with treatment and response, we investigated peripheral blood samples of 73 postmenopausal women with estrogen receptor (ER) positive BC, who received aromatase inhibitor therapy with or without the mucin-1 vaccine tecemotide in the ABCSG34 trial. Blood was retrieved at baseline, midterm and end of therapy, and was analyzed for the distribution and ER expression of monocyte subsets by flow cytometry. Results When 40 healthy, age-matched women were compared with BC patients before treatment start, ER levels of monocytes did not differ, yet patients presented with a higher frequency of classical and fewer non-classical monocytes. Endocrine therapy triggered a significant increase in ER levels in all monocyte subsets, without affecting subset distribution. Vaccination had no overall impact on subset frequency and ER expression. Yet, a shift from intermediate to classical monocytes during therapy correlated with changes in plasma cytokines and chemokines and was significantly associated with low residual cancer burden in vaccinated patients. Without tecemotide, baseline ER levels in classical monocytes were significantly higher in women with good response to endocrine therapy. Conclusions This study identified classical monocytes to be associated with ER positive BC and with patient response to neoadjuvant endocrine treatment and cancer vaccination.

Abstract Background Epirubicin/cyclophosphamide (EC) and docetaxel (D) are commonly used in a sequential regimen in the neoadjuvant treatment of early, high-risk or locally advanced breast cancer (BC). Novel approaches to increase the response rate combine this treatment with immunotherapies such as PD-1 inhibition. However, the expected stimulatory effect on lymphocytes may depend on the chemotherapy backbone. Therefore, we separately compared the immunomodulatory effects of EC and D in the setting of a randomized clinical trial. Methods Tumor and blood samples of 154 patients from the ABCSG-34 trial were available (76 patients received four cycles of EC followed by four cycles of D; 78 patients get the reverse treatment sequence). Tumor-infiltrating lymphocytes, circulating lymphocytes and 14 soluble immune mediators were determined at baseline and at drug change. Furthermore, six BC cell lines were treated with E, C or D and co-cultured with immune cells. Results Initial treatment with four cycles of EC reduced circulating B and T cells by 94% and 45%, respectively. In contrast, no comparable effects on lymphocytes were observed in patients treated with initial four cycles of D. Most immune mediators decreased under EC whereas D-treatment resulted in elevated levels of CXCL10, urokinase-type plasminogen activator (uPA) and its soluble receptor (suPAR). Accordingly, only the exposure of BC cell lines to D induced similar increases as compared to E. While treatment of BC cells with E was associated with cell shrinkage and apoptosis, D induced cell swelling and accumulation of cells in G2 phase. Conclusion The deleterious effect of EC on lymphocytes indicates strong immunosuppressive properties of this combination therapy. D, in contrast, has no effect on lymphocytes, but triggers the secretion of stimulatory proteins in vivo and in vitro, indicating a supportive effect on the immune system. Underlying differences in the induced cell death might be causal. These divergent immunomodulatory effects of epirubicin/cyclophosphamide and docetaxel should be considered when planning future combinations with immunotherapies in breast cancer.

Background: Proof of the T790M resistance mutation is mandatory if patients with EGFR -mutated non-small cell lung cancer (NSCLC) progress under first- or second-generation tyrosine kinase inhibitor therapy. In addition to rebiopsy, analysis of plasma circulating tumor DNA is used to detect T790M resistance mutation. We studied whether sputum is another feasible specimen for detection of EGFR mutations. Methods: Twenty-eight patients with advanced EGFR -mutated NSCLC were included during stable and/or progressive disease. The initial activating EGFR mutations (exon 19 deletions or L858R mutations) at stable disease and at progressive disease (together with T790M) were assessed in simultaneously collected plasma and sputum samples and detected by droplet digital polymerase chain reaction (ddPCR). Results: Activating EGFR mutations were detected in 47% of the plasma samples and 41% of sputum samples during stable disease, and in 57% of plasma samples and 64% of sputum samples during progressive disease. T790M was detected in 44% of the plasma samples and 66% of the sputum samples at progressive disease. In ddPCR T790M-negative results for both specimens (plasma and sputum), negativity was confirmed by rebiopsy in 5 samples. Concordance rate of plasma and sputum for T790M was 0.86, with a positive percent agreement of 1.0 and a negative percent agreement of 0.80. Conclusions: We demonstrated that EGFR mutation analysis with ddPCR is feasible in sputum samples. Combination of plasma and sputum analyses for detection of T790M in NSCLC patients with progressive disease increases the diagnostic yield compared with molecular plasma analysis alone.

Background Patients with metastatic breast cancer (MBC) have a considerable symptom burden and may require extensive care for a long period of time. Palliative care (PC) has the potential to improve their quality of care and reduce their use of medical services. However, the role of specialised PC (SPC) in patients with MBC remains unclear.Patients and methods We performed a retrospective analysis of the medical records of patients diagnosed with breast cancer (BC) from 2008 to 2018 at an university-based referral centre to examine the extent of early and late integration of SPC services for patients with MBC. A descriptive analysis of the patients was also established.Results In all, 932 patients were diagnosed with BC from 2008 to 2018; 225 of these patients had or developed metastases related to their BC. In addition, 132 patients received SPC (58.7%) and 93 patients did not receive SPC (41.3%). The median probability of overall survival (OS) for patients who did not receive SPC services was 3.6 years (95% CI 2.0 to 5.1) and 1.8 years (95% CI 1.3 to 2.3) (p

Background: To assess the clinical relevance of genome-wide somatic copy-number alterations (SCNAs) in plasma circulating tumor DNA (ctDNA) from advanced epidermal growth factor receptor (EGFR)-mutated lung adenocarcinoma patients. Methods: We included 43 patients with advanced EGFR T790M-positive lung adenocarcinoma who were treated with osimertinib after progression under previous EGFR-TKI therapy. We performed genomic profiling of ctDNA in plasma samples from each patient obtained pre-osimertinib and after patients developed resistance to osimertinib. SCNAs were detected by shallow whole-genome plasma sequencing and EGFR mutations were assessed by droplet digital PCR. Results: SCNAs in resistance-related genes (rrSCNAs) were detected in 10 out of 31 (32%) evaluable patients before start of osimertinib. The presence of rrSCNAs in plasma before the initiation of osimertinib therapy was associated with a lower response rate to osimertinib (50% versus 81%, p = 0.08) and was an independent predictor for shorter progression-free survival (adjusted HR 3.33, 95% CI 1.37–8.10, p = 0.008) and overall survival (adjusted HR 2.54, 95% CI 1.09–5.92, p = 0.03). Conclusions: Genomic profiling of plasma ctDNA is clinically relevant and affects the efficacy and clinical outcome of osimertinib. Our approach enables the comprehensive assessment of SCNAs in plasma samples of lung adenocarcinoma patients and may help to guide genotype-specific therapeutic strategies in the future.

Posterior polymorphous corneal dystrophy (PPCD) is characterized by abnormal proliferation of corneal endothelial cells. It was shown that TGF-β2 present in aqueous humor (AH) could help maintaining the corneal endothelium in a G1-phase-arrest state. We wanted to determine whether the levels of this protein are changed in AH of PPCD patients.We determined the concentrations of active TGF-β2 in the AH of 29 PPCD patients (42 samples) and 40 cadaver controls (44 samples) by ELISA. For data analysis the PPCD patients were divided based on either the molecular genetic cause of their disease as PPCD1 (37 samples), PPCD3 (1 sample) and PPCDx (not linked to a known PPCD loci, 4 samples) or on the presence (17 samples) or absence (25 samples) of secondary glaucoma or on whether they had undergone penetrating keratoplasty (PK, 32 samples) or repeated PK (rePK, 7 samples).The level of active TGF-β2 in the AH of all PPCD patients (mean ± SD; 386.98 ± 114.88 pg/ml) in comparison to the control group (260.95 ± 112.43 pg/ml) was significantly higher (P = 0.0001). Compared to the control group, a significantly higher level of active TGF-β2 was found in the PPCD1 (P = 0.0005) and PPCDx (P = 0.0022) groups. Among patients the levels of active TGF-β2 were not significantly affected by gender, age, secondary glaucoma or by the progression of dystrophy when one or repeated PK were performed.The levels of active TGF-β2 in the AH of PPCD patients are significantly higher than control values, and thus the increased levels of TGF-β2 could be a consequence of the PPCD phenotype and can be considered as another feature characterizing this disease.

Overexpression of ecotropic viral integration site 1 (EVI1) is associated with aggressive disease in acute myeloid leukemia (AML). Despite of its clinical importance, little is known about the mechanism through which EVI1 confers resistance to antileukemic drugs. Here, we show that a human myeloid cell line constitutively overexpressing EVI1 after infection with a retroviral vector (U937_EVI1) was partially resistant to etoposide and daunorubicin as compared to empty vector infected control cells (U937_vec). Similarly, inducible expression of EVI1 in HL-60 cells decreased their sensitivity to daunorubicin. Gene expression microarray analyses of U937_EVI1 and U937_vec cells cultured in the absence or presence of etoposide showed that 77 and 419 genes were regulated by EVI1 and etoposide, respectively. Notably, mRNA levels of 26 of these genes were altered by both stimuli, indicating that EVI1 regulated genes were strongly enriched among etoposide regulated genes and vice versa. One of the genes that were induced by both EVI1 and etoposide was CDKN1A/p21/WAF, which in addition to its function as a cell cycle regulator plays an important role in conferring chemotherapy resistance in various tumor types. Indeed, overexpression of CDKN1A in U937 cells mimicked the phenotype of EVI1 overexpression, similarly conferring partial resistance to antileukemic drugs.

Posterior polymorphous corneal dystrophy (PPCD) is a rare autosomal dominant genetically heterogeneous disorder. Nineteen Czech PPCD pedigrees with 113 affected family members were identified, and 17 of these kindreds were genotyped for markers on chromosome 20p12.1- 20q12. Comparison of haplotypes in 81 affected members, 20 unaffected first degree relatives and 13 spouses, as well as 55 unrelated controls, supported the hypothesis of a shared ancestor in 12 families originating from one geographic location. In 38 affected individuals from nine of these pedigrees, a common haplotype was observed between D20S48 and D20S107 spanning approximately 23 Mb, demonstrating segregation of disease with the PPCD1 locus. This haplotype was not detected in 110 ethnically matched control chromosomes. Within the common founder haplotype, a core mini-haplotype was detected for D20S605, D20S182 and M189K2 in all 67 affected members from families 1-12, however alleles representing the core mini-haplotype were also detected in population matched controls. The most likely location of the responsible gene within the disease interval, and estimated mutational age, were inferred by linkage disequilibrium mapping (DMLE+2.3). The appearance of a disease-causing mutation was dated between 64-133 generations. The inferred ancestral locus carrying a PPCD1 disease-causing variant within the disease interval spans 60 Kb on 20p11.23, which contains a single known protein coding gene, ZNF133. However, direct sequence analysis of coding and untranslated exons did not reveal a potential pathogenic mutation. Microdeletion or duplication was also excluded by comparative genomic hybridization using a dense chromosome 20 specific array. Geographical origin, haplotype and statistical analysis suggest that in 14 unrelated families an as yet undiscovered mutation on 20p11.23 was inherited from a common ancestor. Prevalence of PPCD in the Czech Republic appears to be the highest worldwide and our data suggests that at least one other novel locus for PPCD also exists.
