1. Phonon collapse and anharmonic melting of the 3D charge-density wave in kagome metals

Abstract

Abstract The charge-density wave (CDW) mechanism and resulting structure of the AV3Sb5 family of kagome metals has posed a puzzling challenge since their discovery four years ago. In fact, the lack of consensus on the origin and structure of the CDW hinders the understanding of the emerging phenomena. Here, by employing a non-perturbative treatment of anharmonicity from first-principles calculations, we reveal that the charge-density transition in CsV3Sb5 is driven by the large electron-phonon coupling of the material and that the melting of the CDW state is attributed to ionic entropy and lattice anharmonicity. The calculated transition temperature is in very good agreement with experiments, implying that soft mode physics are at the core of the charge-density wave transition. Contrary to the standard assumption associated with a pure kagome lattice, the CDW is essentially three-dimensional as it is triggered by an unstable phonon at the L point. The absence of involvement of phonons at the M point enables us to constrain the resulting symmetries to six possible space groups. The unusually large electron-phonon linewidth of the soft mode explains why inelastic scattering experiments did not observe any softened phonon. We foresee that large anharmonic effects are ubiquitous and could be fundamental to understand the observed phenomena also in other kagome families.

Abstract The kagome metals AV3Sb5 (A = K, Rb, Cs) present an ideal sandbox to study the interrelation between multiple coexisting correlated phases such as charge order and superconductivity. So far, no consensus on the microscopic nature of these states has been reached as the proposals struggle to explain all their exotic physical properties. Among these, field-switchable electric magneto-chiral anisotropy (eMChA) in CsV3Sb5 provides intriguing evidence for a rewindable electronic chirality, yet the other family members have not been likewise investigated. Here, we present a comparative study of magneto-chiral transport between CsV3Sb5 and KV3Sb5. Despite their similar electronic structure, KV3Sb5 displays negligible eMChA, if any, and with no field switchability. This is in stark contrast to the non-saturating eMChA in CsV3Sb5 even in high fields up to 35 T. In light of their similar band structures, the stark difference in eMChA suggests its origin in the correlated states. Clearly, the V kagome nets alone are not sufficient to describe the physics and the interactions with their environment are crucial in determining the nature of their low-temperature state.

Abstract

Abstract Lung diseases, including lung cancer, are rising causes of global mortality. Despite novel imaging technologies and the development of biomarker assays, the detection of lung cancer remains a significant challenge. However, the lung communicates directly with the external environment and releases aerosolized droplets during normal tidal respiration, which can be collected, stored and analzsed as exhaled breath condensate (EBC). A few studies have suggested that EBC contains extracellular vesicles (EVs) whose microRNA (miRNA) cargos may be useful for evaluating different lung conditions, but the cellular origin of these EVs remains unknown. In this study, we used nanoparticle tracking, transmission electron microscopy, Western blot analyses and super resolution nanoimaging (ONi) to detect and validate the identity of exhaled EVs (exh‐EVs). Using our customizable antibody‐purification assay, EV‐CATCHER, we initially determined that exh‐EVs can be selectively enriched from EBC using antibodies against three tetraspanins (CD9, CD63 and CD81). Using ONi we also revealed that some exh‐EVs harbour lung‐specific proteins expressed in bronchiolar Clara cells (Clara Cell Secretory Protein [CCSP]) and Alveolar Type II cells (Surfactant protein C [SFTPC]). When conducting miRNA next generation sequencing (NGS) of airway samples collected at five different anatomic levels (i.e., mouth rinse, mouth wash, bronchial brush, bronchoalveolar lavage [BAL] and EBC) from 18 subjects, we determined that miRNA profiles of exh‐EVs clustered closely to those of BAL EVs but not to those of other airway samples. When comparing the miRNA profiles of EVs purified from matched BAL and EBC samples with our three tetraspanins EV‐CATCHER assay, we captured significant miRNA expression differences associated with smoking, asthma and lung tumor status of our subjects, which were also reproducibly detected in EVs selectively purified with our anti‐CCSP/SFTPC EV‐CATCHER assay from the same samples, but that confirmed their lung tissue origin. Our findings underscore that enriching exh‐EV subpopulations from EBC allows non‐invasive sampling of EVs produced by lung tissues.

Abstract

Background SEA-CD40 is an investigational, non-fucosylated, humanized monoclonal IgG1 antibody that activates CD40, an immune-activating tumor necrosis factor receptor superfamily member. SEA-CD40 exhibits enhanced binding to activating FcγRIIIa, possibly enabling greater immune stimulation than other CD40 agonists. A first-in-human phase 1 trial was conducted to examine safety, pharmacokinetics, and pharmacodynamics of SEA-CD40 monotherapy in patients with advanced solid tumors and lymphoma.Methods SEA-CD40 was administered intravenously to patients with solid tumors or lymphoma in 21-day cycles with standard 3+3 dose escalation at 0.6, 3, 10, 30, 45, and 60 µg/kg. An intensified dosing regimen was also studied. The primary objectives of the study were to evaluate the safety and tolerability and identify the maximum tolerated dose of SEA-CD40. Secondary objectives included evaluation of the pharmacokinetic parameters, antitherapeutic antibodies, pharmacodynamic effects and biomarker response, and antitumor activity.Results A total of 67 patients received SEA-CD40 including 56 patients with solid tumors and 11 patients with lymphoma. A manageable safety profile was observed, with predominant adverse events of infusion/hypersensitivity reactions (IHRs) reported in 73% of patients. IHRs were primarily ≤grade 2 with an incidence associated with infusion rate. To mitigate IHRs, a standardized infusion approach was implemented with routine premedication and a slowed infusion rate. SEA-CD40 infusion resulted in potent immune activation, illustrated by dose dependent cytokine induction with associated activation and trafficking of innate and adaptive immune cells. Results suggested that doses of 10–30 µg/kg may result in optimal immune activation. SEA-CD40 monotherapy exhibited evidence of antitumor activity, with a partial response in a patient with basal cell carcinoma and a complete response in a patient with follicular lymphoma.Conclusions SEA-CD40 was tolerable as monotherapy and induced potent dose dependent immune cell activation and trafficking consistent with immune activation. Evidence of monotherapy antitumor activity was observed in patients with solid tumors and lymphoma. Further evaluation of SEA-CD40 is warranted, potentially as a component of a combination regimen.Trial registration number NCT02376699.

Abstract

Current use of liquid biopsy is based on cell-free DNA (cfDNA) and the evaluation of mutations or methylation pattern. However, expressed RNA can capture mutations, changes in expression levels due to methylation, and provide information on cell of origin, growth, and proliferation status. We developed an approach to isolate cell-free total nucleic acid (cfDNA) and used targeted next generation sequencing to sequence cell-free RNA (cfRNA) and cfDNA as new approach in liquid biopsy. We demonstrate that cfRNA is overall more sensitive than cfDNA in detecting mutations. We show that cfRNA is reliable in detecting fusion genes and cfDNA is reliable in detecting chromosomal gains and losses. cfRNA levels of various solid tumor biomarkers were significantly higher (P 0.98) of solid tumors, B-cell lymphoid neoplasms, T-cell lymphoid neoplasms, and myeloid neoplasms. Abstract

Our multidimensional identities determine how we interact with each other, shaping social networks through group-based connection preferences. While interactions along single dimensions have been extensively studied, the dynamics driving multidimensional connection preferences remain largely unexplored. In this work, we develop a network model of multidimensional social interactions to tackle two crucial questions: What is the structure of our latent connection preferences, and how do we integrate information from our multidimensional identities to connect with others? To answer these questions, we systematically model different latent preference structures and preference aggregation mechanisms. Then, we compare them using Bayesian model selection by fitting empirical data from high school friendship networks. We find that a simple latent preference model consistently outperforms more complex alternatives. The calibrated model provides robust measures of latent connection preferences in real-world networks, bringing insights into how one- and multidimensional groups interact. Finally, we develop natural operationalizations of dimension salience, revealing which aspects of identity are most relevant for individuals when forming connections.

Abstract

Here, we report on a phase IIa study to determine the intubation rate, survival, viral clearance, and development of endogenous Abs in patients with COVID-19 pneumonia treated with convalescent plasma (CCP) containing high levels of neutralizing anti–SARS-CoV-2 Abs. Radiographic and laboratory evaluation confirmed all 51 treated patients had COVID-19 pneumonia. Fresh or frozen CCP from donors with high titers of neutralizing Abs was administered. The nonmechanically ventilated patients (n = 36) had an intubation rate of 13.9% and a 30-day survival rate of 88.9%, and the overall survival rate for a comparative group based on network data was 72.5% (1625/2241). Patients had negative nasopharyngeal swab rates of 43.8% and 73.0% on days 10 and 30, respectively. Patients mechanically ventilated had a day-30 mortality rate of 46.7%; the mortality rate for a comparative group based on network data was 71.0% (369/520). All evaluable patients were found to have neutralizing Abs on day 3 (n = 47), and all but 1 patient had Abs on days 30 and 60. The only adverse event was a mild rash. In this study on patients with COVID-19 disease, we show therapeutic use of CCP was safe and conferred transfer of Abs, while preserving endogenous immune response.

23. Unveiling homophily beyond the pool of opportunities

Abstract

Unveiling individuals' preferences for connecting with similar others (choice homophily) beyond the structural factors determining the pool of opportunities, is a challenging task. Here, we introduce a robust methodology for quantifying and inferring choice homophily in a variety of social networks. Our approach employs statistical network ensembles to estimate and standardize homophily measurements. We control for group size imbalances and activity disparities by counting the number of possible network configurations with a given number of inter-group links using combinatorics. This method provides a principled measure of connection preferences and their confidence intervals. Our framework is versatile, suitable for undirected and directed networks, and applicable in scenarios involving multiple groups. To validate our inference method, we test it on synthetic networks and show that it outperforms traditional metrics. Our approach accurately captures the generative homophily used to build the networks, even when we include additional tie-formation mechanisms, such as preferential attachment and triadic closure. Results show that while triadic closure has some influence on the inference, its impact is small in homophilic networks. On the other hand, preferential attachment does not perturb the results of the inference method. We apply our method to real-world networks, demonstrating its effectiveness in unveiling underlying gender homophily. Our method aligns with traditional metrics in networks with balanced populations, but we obtain different results when the group sizes or degrees are imbalanced. This finding highlights the importance of considering structural factors when measuring choice homophily in social networks.

Social polarization is a growing concern worldwide, as it strains social relations, erodes trust in institutions, and thus threatens democratic societies. Academic efforts to understand this phenomenon have traditionally approached it from a one-dimensional perspective, focusing on bipolar or dichotomous systems. However, political conflicts often involve not only two, but multiple potentially dissenting factions. The most representative examples are multi-party democracies, where the multilateral tensions among different parties often lead to gridlock and uncertainty. Despite the prevalence of these multipolar systems, there is still a lack of suitable analytical tools to study their intricate polarization patterns. In this work, we develop an analytical framework consisting of an inherently multipolar model for unbiased ideological spaces, a method to infer multidimensional opinions from interaction networks, and novel multidimensional polarization metrics that quantify several aspects of ideological polarization and bring new insights into the analysis of high-dimensional opinion distributions. Crucially, our multidimensional framework does not assume the underlying ideological structure, such as conservative vs progressive, liberal vs authoritarian, etc. Instead, it reveals the natural space that best describes the social landscape, which does not necessarily correspond to traditional categories. We illustrate the application of this framework in quadripolar and pentapolar real-world democratic processes, finding non-trivial ideological structures with clear connections to the underlying social context. Abstract

PurposeAutosomal dominant cone rod dystrophy 7 (CORD7) was initially linked to the gene RIMS1 and reported in a 4-generation British family in 1998. The purpose of this study was to investigate the legitimacy of this association, and to correctly characterize the genetic cause of this condition.MethodsThe allele frequency of RIMS1 c.2459G>A, p.Arg820His, was investigated in the Genomes Aggregation Dataset (gnomAD) datasets and whole genome sequencing (WGS) was performed for 4 members of the CORD7 family with filtering of rare pathogenic variants in a virtual gene panel comprising all genes known to be associated with inherited retinal dystrophy (IRD). Cytogenetic analysis was performed to rule out interchromosomal translocation.ResultsRIMS1 p.Arg820His has a maximal carrier frequency of >1:5000 in Europeans. A previously well-characterized PROM1 variant: c.1118C>T, p.Arg373Cys, was detected in 9 affected members of the CORD7 family who underwent WGS or direct sequencing. One affected family member is now known to have macular dystrophy in the absence of RIMS1 p.Arg820His. Clinical analysis of affected family members and 27 individuals with retinopathy associated with the same - PROM1 - variant showed consistent phenotypes.ConclusionsThe case for pathogenicity of RIMS1 p.Arg820His is not strong based on its presence on 10 alleles in the gnomAD dataset and absence from additional CORD affected individuals. The finding of a known pathogenic variant in PROM1 correlates well with the phenotypic characteristics of the affected individuals, and is likely to account for the condition. Clear evidence of association between RIMS1 and a retinal dystrophy is yet to be described.

34. First-mover advantage explains gender disparities in physics citations

Abstract

Mounting evidence suggests that publications and citations of scholars in the STEM fields (Science, Technology, Engineering and Mathematics) suffer from gender biases. In this paper, we study the physics community, a core STEM field in which women are still largely underrepresented and where these gender disparities persist. To reveal such inequalities, we compare the citations received by papers led by men and women that cover the same topics in a comparable way. To do that, we devise a robust statistical measure of similarity between publications that enables us to detect pairs of similar papers. Our findings indicate that although papers written by women tend to have lower visibility in the citation network, pairs of similar papers written by men and women receive comparable attention when corrected for the time of publication. These analyses suggest that gender disparity is closely related to the first-mover and cumulative advantage that men have in physics, and is not an intentional act of discrimination towards women., Comment: 21 pages, 8 tables, 10 figures

Abstract

We develop a theory to measure the variance and covariance of probability distributions defined on the nodes of a graph, which takes into account the distance between nodes. Our approach generalizes the usual (co)variance to the setting of weighted graphs and retains many of its intuitive and desired properties. Interestingly, we find that a number of famous concepts in graph theory and network science can be reinterpreted in this setting as variances and covariances of particular distributions. As a particular application, we define the maximum variance problem on graphs with respect to the effective resistance distance, and characterize the solutions to this problem both numerically and theoretically. We show how the maximum variance distribution is concentrated on the boundary of the graph, and illustrate this in the case of random geometric graphs. Our theoretical results are supported by a number of experiments on a network of mathematical concepts, where we use the variance and covariance as analytical tools to study the (co-)occurrence of concepts in scientific papers with respect to the (network) relations between these concepts., Comment: 38 pages, 9 figures

48. Racial/Ethnic Differences in the Relationship Between Stressful Life Events and Quality of Life in Adolescents.

Abstract

PurposeStressful life events (SLEs) increase allostatic load and require adaptation. Experiencing SLEs has been associated with decreased health-related quality of life (HRQOL) among adolescents. This study examined racial/ethnic and developmental differences in the relationship between SLEs and HRQOL from preadolescence to midadolescence.MethodsData were from 4,824 participants in the Healthy Passages project, a population-based prospective longitudinal survey of fifth, seventh, and 10th grade adolescents in the U.S. HRQOL was measured with Pediatric Quality of Life Inventory and SLEs with items addressing family-related SLEs (e.g., the parent's death, separation, and divorce; family member's injury/illness; residential change; new child in the household).ResultsAdolescents, regardless of race/ethnicity, reported the highest SLEs and the lowest HRQOL in early adolescence. Analysis of an autoregressive model with cross-lagged effects showed that the concurrent relationships between SLEs and HRQOL were significantly negative across preadolescence, early adolescence, and midadolescence in African-American, Latinx, and white groups. Furthermore, adolescents had a negative cross-lagged association from SLEs in early adolescence to HRQOL in pre adolescence, but this was not the case among the other racial/ethnic groups.ConclusionsBecause the negative relationship between family-related SLEs and HRQOL persisted throughout stages of adolescent development, health services targeting adolescents should provide comprehensive family-centered care to alleviate the impact of family-related life stress. Relationships between family life stress and HRQOL varied by racial/ethnic groups, which should be considered by health professionals, teachers, and parents, and in prevention efforts. Latinx adolescents may be particularly vulnerable to time-lagged effects of such family-related stress.

50. A LEGO® Serious Play Activity to Help Teamwork Skills Development amongst Business Students

Abstract

Methods for teaching higher education students how to learn teamwork skills are constantly evolving. Over the last few years, serious games have become popular in business schools for stimulating teamwork among students by making them focus on the game while pursuing a team objective. Among these games, LEGO® Serious Play has shown enormous potential. However, the number of results published on its application to the development of teamwork skills remains scarce. This paper seeks to explain the development of a teaching activity designed to make all first year undergraduate students in a business school work in teams. The way in which LEGO® Serious Play can position students within the research process is discussed, as is the potential for LEGO® Serious Play as a participatory method. Additionally, we summarize teacher and student metrics related to their experience with the activity in terms of teamwork performance and results, and we show the effect of contextual factors such as daily or weekly schedule or team size. This 'one shot' teamwork activity with first year students is expected to have an effect later when they are able to re-create the processes used to work as a team.
