Abstract We perform a detailed model-independent phenomenological analysis of B ¯ → D ¯ D $$ \overline{B}\to \overline{D}D $$ decays. Employing an SU(3) F analysis including symmetry-breaking contributions together with a conservative power counting for various suppression effects, we obtain updated Standard Model predictions for all branching fractions and CP asymmetries from a fit to the available experimental data, testable at Belle II and the LHC experiments. These results include all relevant suppressed contributions, thereby providing upper limits on subleading Standard Model (SM) effects like “penguin pollution”, enabling searches for physics beyond the SM. Importantly, allowing in the same fit for the production fractions of charged and neutral B mesons to be different, we find f d /f u = 0.86 ± 0.05, which is 2.5σ away from unity, which, if confirmed, would have important phenomenological consequences.

The article presents results of using remote sensing images and machine learning to map and assess land potential based on time-series of potential Fraction of Absorbed Photosynthetically Active Radiation (FAPAR) composites. Land potential here refers to the potential vegetation productivity in the hypothetical absence of short–term anthropogenic influence, such as intensive agriculture and urbanization. Knowledge on this ecological land potential could support the assessment of levels of land degradation as well as restoration potentials. Monthly aggregated FAPAR time-series of three percentiles (0.05, 0.50 and 0.95 probability) at 250 m spatial resolution were derived from the 8-day GLASS FAPAR V6 product for 2000–2021 and used to determine long-term trends in FAPAR, as well as to model potential FAPAR in the absence of human pressure. CCa 3 million training points sampled from 12,500 locations across the globe were overlaid with 68 bio-physical variables representing climate, terrain, landform, and vegetation cover, as well as several variables representing human pressure including: population count, cropland intensity, nightlights and a human footprint index. The training points were used in an ensemble machine learning model that stacks three base learners (extremely randomized trees, gradient descended trees and artificial neural network) using a linear regressor as meta-learner. The potential FAPAR was then projected by removing the impact of urbanization and intensive agriculture in the covariate layers. The results of strict cross-validation show that the global distribution of FAPAR can be explained with an R2 of 0.89, with the most important covariates being growing season length, forest cover indicator and annual precipitation. From this model, a global map of potential monthly FAPAR for the recent year (2021) was produced, and used to predict gaps in actual vs. potential FAPAR. The produced global maps of actual vs. potential FAPAR and long-term trends were each spatially matched with stable and transitional land cover classes. The assessment showed large negative FAPAR gaps (actual lower than potential) for classes: urban, needle-leave deciduous trees, and flooded shrub or herbaceous cover, while strong negative FAPAR trends were found for classes: urban, sparse vegetation and rainfed cropland. On the other hand, classes: irrigated or post-flooded cropland, tree cover mixed leaf type, and broad-leave deciduous showed largely positive trends. The framework allows land managers to assess potential land degradation from two aspects: as an actual declining trend in observed FAPAR and as a difference between actual and potential vegetation FAPAR.

Abstract Latent and sensible heat flux observations are essential for understanding land–atmosphere interactions. Measurements from the eddy covariance technique are widely used but suffer from systematic energy imbalance problems, partly due to missing large eddies from sub‐mesoscale transport. Because available energy drives the development of large eddies, we propose an available energy based correction method for turbulent flux measurements. We apply our method to 172 flux tower sites and show that we can reduce the energy imbalance from −14.99 to −0.65 W m−2 on average, together with improved consistency between turbulent fluxes and available energy and associated increases in r2 at individual sites and across networks. Our results suggest that our method is conceptually and empirically preferable over the method implemented in the ONEFlux processing. This can contribute to the efforts in understanding and addressing the energy imbalance issue, which is crucial for the evaluation and calibration of land surface models.

PEX19 binding sites are essential parts of the targeting signals of peroxisomal membrane proteins (mPTS). In this study, we characterized PEX19 binding sites of PEX11, the most abundant peroxisomal and glycosomal membrane protein from Trypanosoma brucei and Saccharomyces cerevisiae. TbPEX11 contains two PEX19 binding sites, one close to the N-terminus (BS1) and a second in proximity to the first transmembrane domain (BS2). The N-terminal BS1 is highly conserved across different organisms and is required for maintenance of the steady-state concentration and efficient targeting to peroxisomes and glycosomes in both baker’s yeast and Trypanosoma brucei. The second PEX19 binding site in TbPEX11 is essential for its glycosomal localization. Deletion or mutations of the PEX19 binding sites in TbPEX11 or ScPEX11 results in mislocalization of the proteins to mitochondria. Bioinformatic analysis indicates that the N-terminal region of TbPEX11 contains an amphiphilic helix and several putative TOM20 recognition motifs. We show that the extreme N-terminal region of TbPEX11 contains a cryptic N-terminal signal that directs PEX11 to the mitochondrion if its glycosomal transport is blocked.

Somatoform disorders (SD), commencing during adolescence, represent a major problem in health care systems. While literature underlines the high presence of mental health problems among children and adolescents afflicted by somatic symptoms in the general population, limited evidence is available on the prevalence of comorbid somatic symptoms in child and adolescent psychiatric populations. We assessed the prevalence of somatic symptoms, depression, and anxiety by validated questionnaires in an inpatient cohort. We further screened for the presence of SD. Out of 434 inpatients aged 11–17 years, 371 were included and a total of n = 288 (77.6%) children and adolescents participated in the study. A total of 93.8% of the inpatients reported somatic symptoms within the past six months and still almost half (45.7%) of the sample reported at least one somatic symptom within the last seven days prior to inquiry. Relating to the past six months, 59.5% were positively screened for SD, and 44.6% reported symptoms eligible for positive screening within the past seven days prior to the survey. Somatoform symptomatology was highly associated with anxiety and depression scores, but functional decline was amenable to the number of somatic symptoms only. We provide evidence that somatic symptoms are frequent in children and adolescents being treated in child and adolescent psychiatry and are relevant to everyday functioning. Screening for somatic symptoms should be introduced in the routine diagnostic procedures for early detection of SD in the commencing stages.

Mineral nitrogen (N) fertilizer use is essential to maintain high-yielding cropping systems that presently provide food for nearly half of humanity. Simultaneously, it causes a range of detrimental impacts such as greenhouse gas emissions, eutrophication, and contamination of drinking water. There is growing recognition of the need to balance crop production with the impacts of fertilizer use. Here we provide a global assessment of the potential to reduce mineral fertilizer use through four interventions: capping surpluses, enhancing manure cycling to cropland, cultivation of off-season green manures, and cycling of human excreted N to cropland. We find that the combined potential of these interventions is a reduction in global N fertilizer use by 21%–52%. The availability of interventions is spatially heterogeneous with most cropland having three to four interventions available with alternative N sources tending to be more abundant on cropland already receiving fertilizer. Our assessment highlights that these locally in part already practiced interventions bear great opportunities to mitigate synthetic N use and dependency globally. Yet, their limited adoption underpins the need for cross-sectoral policies to overcome barriers to their implementation and agronomic research on their robust scaling.

In the context of renal proteinuric diseases, TRPC6 has been shown to play an important role in ultrafiltration associated with the slit diaphragm through the control of the intracellular Ca2+ concentration in the podocytes of glomeruli. However, to date, the properties of TRPC6 have been studied mainly in cell lines or in animal models. Therefore, the aim of the study presented here was to investigate the presence and distribution of TRPC6 in human kidneys in order to possibly verify the applicability of the results previously obtained in nonhuman experiments. For this purpose, kidneys from nine cadavers were prepared for immunohistochemical staining and were supplemented with a fresh human kidney obtained by nephrectomy. TRPC6 was detected in glomeruli and in the parietal epithelial cells of Bowman’s capsule. Larger amounts were detected in the tubular system and collecting ducts. In contrast to the peritubular capillary bed, which showed no immune reaction, the cortical resistance vessels showed mild TRPC6 staining. In conclusion, our studies on the expression of TRPC6 in human kidney tissue support the translational concept of the involvement of TRPC6 in various renal diseases and reveal new aspects of the distribution of TRPC6 in the human kidney.

To elucidate the redundancy in the components for the targeting of membrane proteins to the endoplasmic reticulum (ER) and/or their insertion into the ER membrane under physiological conditions, we previously analyzed different human cells by label-free quantitative mass spectrometry. The HeLa and HEK293 cells had been depleted of a certain component by siRNA or CRISPR/Cas9 treatment or were deficient patient fibroblasts and compared to the respective control cells by differential protein abundance analysis. In addition to clients of the SRP and Sec61 complex, we identified membrane protein clients of components of the TRC/GET, SND, and PEX3 pathways for ER targeting, and Sec62, Sec63, TRAM1, and TRAP as putative auxiliary components of the Sec61 complex. Here, a comprehensive evaluation of these previously described differential protein abundance analyses, as well as similar analyses on the Sec61-co-operating EMC and the characteristics of the topogenic sequences of the various membrane protein clients, i.e., the client spectra of the components, are reported. As expected, the analysis characterized membrane protein precursors with cleavable amino-terminal signal peptides or amino-terminal transmembrane helices as predominant clients of SRP, as well as the Sec61 complex, while precursors with more central or even carboxy-terminal ones were found to dominate the client spectra of the SND and TRC/GET pathways for membrane targeting. For membrane protein insertion, the auxiliary Sec61 channel components indeed share the client spectra of the Sec61 complex to a large extent. However, we also detected some unexpected differences, particularly related to EMC, TRAP, and TRAM1. The possible mechanistic implications for membrane protein biogenesis at the human ER are discussed and can be expected to eventually advance our understanding of the mechanisms that are involved in the so-called Sec61-channelopathies, resulting from deficient ER protein import.

Abstract We analyze in detail the angular distributions in $${\bar{B}}\rightarrow D^*\ell {{\bar{\nu }}}$$ B ¯ → D ∗ ℓ ν ¯ decays, with a focus on lepton-flavour non-universality. We investigate the minimal number of angular observables that fully describes current and upcoming datasets, and explore their sensitivity to physics beyond the Standard Model (BSM) in the most general weak effective theory. We apply our findings to the current datasets, extract the non-redundant set of angular observables from the data, and compare to precise SM predictions that include lepton-flavour universality violating mass effects. Our analysis shows that the number of independent angular observables that can be inferred from current experimental data is limited to only four. These are insufficient to extract the full set of relevant BSM parameters. We uncover a $$\sim 4\sigma $$ ∼ 4 σ tension between data and predictions that is hidden in the redundant presentation of the Belle 2018 data on $${\bar{B}}\rightarrow D^*\ell {{\bar{\nu }}}$$ B ¯ → D ∗ ℓ ν ¯ decays. This tension specifically involves observables that probe $$e-\mu $$ e - μ lepton-flavour universality. However, we find inconsistencies in these data, which renders results based on it suspicious. Nevertheless, we discuss which generic BSM scenarios could explain the tension, in the case that the inconsistencies do not affect the data materially. Our findings highlight that $$e-\mu $$ e - μ non-universality in the SM, introduced by the finite muon mass, is already significant in a subset of angular observables with respect to the experimental precision.

Local studies and modeling experiments suggest that shallow groundwater and lateral redistribution of soil moisture, together with soil properties, can be highly important secondary water sources for vegetation in water-limited ecosystems. However, there is a lack of observation-based studies of these terrain-associated secondary water effects on vegetation over large spatial domains. Here, we quantify the role of terrain properties on the spatial variations of dry season vegetation decay rate across Africa obtained from geostationary satellite acquisitions to assess the large-scale relevance of secondary water effects. We use machine learning based attribution to identify where and under which conditions terrain properties related to topography, water table depth, and soil hydraulic properties influence the rate of vegetation decay. Over the study domain, the machine learning model attributes about one-third of the spatial variations of vegetation decay rates to terrain properties, which is roughly equally split between direct terrain effects and interaction effects with climate and vegetation variables. The importance of secondary water effects increases with increasing topographic variability, shallower groundwater levels, and the propensity to capillary rise given by soil properties. In regions with favorable terrain properties, more than 60% of the variations in the decay rate of vegetation are attributed to terrain properties, highlighting the importance of secondary water effects on vegetation in Africa. Our findings provide an empirical assessment of the importance of local-scale secondary water effects on vegetation over Africa and help to improve hydrological and vegetation models for the challenge of bridging processes across spatial scales.

Ribbon synapses reliably transmit synaptic signals over a broad signalling range. Rod photoreceptor ribbon synapses are capable of transmitting signals generated by the absorption of single photons. The high precision of ribbon synapses emphasizes the need for particularly efficient signalling mechanisms. Synaptic ribbons are presynaptic specializations of ribbon synapses and are anchored to the active zone. Synaptic ribbons bind many synaptic vesicles that are delivered to the active zone for continuous and faithful signalling. In the present study we demonstrate with independent antibodies at the light- and electron microscopic level that rabconnectin-3α (RC3α)—alternative name Dmx-like 2 (DMXL2)—is localized to the synaptic ribbons of rod photoreceptor synapses in the mouse retina. In the brain, RC3α-containing complexes are known to interact with important components of synaptic vesicles, including Rab3-activating/inactivating enzymes, priming proteins and the vesicular H+-ATPase that acidifies the synaptic vesicle lumen to promote full neurotransmitter loading. The association of RC3α/DMXL2 with rod synaptic ribbons of the mouse retina could enable these structures to deliver only fully signalling-competent synaptic vesicles to the active zone thus contributing to reliable synaptic communication.

Background Biodiversity varies in space and time, and often in response to environmental heterogeneity. Indicators in the form of local biodiversity measures–such as species richness or abundance–are common tools to capture this variation. The rise of readily available remote sensing data has enabled the characterization of environmental heterogeneity in a globally robust and replicable manner. Based on the assumption that differences in biodiversity measures are generally related to differences in environmental heterogeneity, these data have enabled projections and extrapolations of biodiversity in space and time. However so far little work has been done on quantitatively evaluating if and how accurately local biodiversity measures can be predicted. Methods Here I combine estimates of biodiversity measures from terrestrial local biodiversity surveys with remotely-sensed data on environmental heterogeneity globally. I then determine through a cross-validation framework how accurately local biodiversity measures can be predicted within (“predictability”) and across similar (“transferability”) biodiversity surveys. Results I found that prediction errors can be substantial, with error magnitudes varying between different biodiversity measures, taxonomic groups, sampling techniques and types of environmental heterogeneity characterizations. And although errors associated with model predictability were in many cases relatively low, these results question–particular for transferability–our capability to accurately predict and project local biodiversity measures based on environmental heterogeneity. I make the case that future predictions should be evaluated based on their accuracy and inherent uncertainty, and ecological theories be tested against whether we are able to make accurate predictions from local biodiversity data.

In human cells, approximately 30% of all polypeptides enter the secretory pathway at the level of the endoplasmic reticulum (ER). This process involves cleavable amino-terminal signal peptides (SPs) or more or less amino-terminal transmembrane helices (TMHs), which serve as targeting determinants, at the level of the precursor polypeptides and a multitude of cytosolic and ER proteins, which facilitate their ER import. Alone or in combination SPs and TMHs guarantee the initial ER targeting as well as the subsequent membrane integration or translocation. Cytosolic SRP and SR, its receptor in the ER membrane, mediate cotranslational targeting of most nascent precursor polypeptide chains to the polypeptide-conducting Sec61 complex in the ER membrane. Alternatively, fully-synthesized precursor polypeptides and certain nascent precursor polypeptides are targeted to the ER membrane by either the PEX-, SND-, or TRC-pathway. Although these targeting pathways may have overlapping functions, the question arises how relevant this is under cellular conditions and which features of SPs and precursor polypeptides determine preference for a certain pathway. Irrespective of their targeting pathway(s), most precursor polypeptides are integrated into or translocated across the ER membrane via the Sec61 channel. For some precursor polypeptides specific Sec61 interaction partners have to support the gating of the channel to the open state, again raising the question why and when this is the case. Recent progress shed light on the client spectrum and specificities of some auxiliary components, including Sec62/Sec63, TRAM1 protein, and TRAP. To address the question which precursors use a certain pathway or component in intact human cells, i.e., under conditions of fast translation rates and molecular crowding, in the presence of competing precursors, different targeting organelles, and relevant stoichiometries of the involved components, siRNA-mediated depletion of single targeting or transport components in HeLa cells was combined with label-free quantitative proteomics and differential protein abundance analysis. Here, we present a summary of the experimental approach as well as the resulting differential protein abundance analyses and discuss their mechanistic implications in light of the available structural data.

TRPC channels are critical players in cochlear hair cells and sensory neurons, as demonstrated in animal experiments. However, evidence for TRPC expression in the human cochlea is still lacking. This reflects the logistic and practical difficulties in obtaining human cochleae. The purpose of this study was to detect TRPC6, TRPC5 and TRPC3 in the human cochlea. Temporal bone pairs were excised from ten body donors, and the inner ear was first assessed based on computed tomography scans. Decalcification was then performed using 20% EDTA solutions. Immunohistochemistry with knockout-tested antibodies followed. The organ of Corti, the stria vascularis, the spiral lamina, spiral ganglion neurons and cochlear nerves were specifically stained. This unique report of TRPC channels in the human cochlea supports the hypothesis of the potentially critical role of TRPC channels in human cochlear health and disease which has been suggested in previous rodent experiments.

In the absence of targeted treatment options, neoadjuvant chemotherapy (NACT) is applied widely for triple-negative breast cancer (TNBC). Response to NACT is an important parameter predictive of oncological outcomes (progression-free and overall survival). An approach to the evaluation of predictive markers enabling therapy individualization is the identification of tumor driver genetic mutations. This study was conducted to investigate the role of SEC62, harbored at 3q26 and identified as a driver of breast cancer pathogenesis, in TNBC. We analyzed SEC62 expression in The Cancer Genome Atlas database, and immunohistologically investigated SEC62 expression in pre- and post-NACT tissue samples from 64 patients with TNBC treated at the Department of Gynecology and Obstetrics/Saarland University Hospital/Homburg between January 2010 and December 2018 and compared the effect of SEC62 on tumor cell migration and proliferation in functional assays. SEC62 expression dynamics correlated positively with the response to NACT (p ≤ 0.01) and oncological outcomes (p ≤ 0.01). SEC62 expression stimulated tumor cell migration (p ≤ 0.01). The study findings indicate that SEC62 is overexpressed in TNBC and serves as a predictive marker for the response to NACT, a prognostic marker for oncological outcomes, and a migration-stimulating oncogene in TNBC.

Abstract Hydrological interactions between vegetation, soil, and topography are complex, and heterogeneous in semi‐arid landscapes. This along with data scarcity poses challenges for large‐scale modeling of vegetation‐water interactions. Here, we exploit metrics derived from daily Meteosat data over Africa at ca. 5 km spatial resolution for ecohydrological analysis. Their spatial patterns are based on Fractional Vegetation Cover (FVC) time series and emphasize limiting conditions of the seasonal wet to dry transition: the minimum and maximum FVC of temporal record, the FVC decay rate and the FVC integral over the decay period. We investigate the relevance of these metrics for large scale ecohydrological studies by assessing their co‐variation with soil moisture, and with topographic, soil, and vegetation factors. Consistent with our initial hypothesis, FVC minimum and maximum increase with soil moisture, while the FVC integral and decay rate peak at intermediate soil moisture. We find evidence for the relevance of topographic moisture variations in arid regions, which, counter‐intuitively, is detectable in the maximum but not in the minimum FVC. We find no clear evidence for wide‐spread occurrence of the “inverse texture effect” on FVC. The FVC integral over the decay period correlates with independent data sets of plant water storage capacity or rooting depth while correlations increase with aridity. In arid regions, the FVC decay rate decreases with canopy height and tree cover fraction as expected for ecosystems with a more conservative water‐use strategy. Thus, our observation‐based products have large potential for better understanding complex vegetation‐water interactions from regional to continental scales.

Abstract We provide updated predictions for the hadronic decays $$\bar{B}_s^0\rightarrow D_s^{(*)+} \pi ^-$$ B ¯ s 0 → D s ( ∗ ) + π - and $$\bar{B}^0\rightarrow D^{(*)+} K^-$$ B ¯ 0 → D ( ∗ ) + K - . They are based on $${\mathcal {O}}(\alpha _s^2)$$ O ( α s 2 ) results for the QCD factorization amplitudes at leading power and on recent results for the $$\bar{B}_{(s)} \rightarrow D_{(s)}^{(*)}$$ B ¯ ( s ) → D ( s ) ( ∗ ) form factors up to order $$\mathcal{O}(\Lambda _\mathrm{QCD}^2/m_c^2)$$ O ( Λ QCD 2 / m c 2 ) in the heavy-quark expansion. We give quantitative estimates of the matrix elements entering the hadronic decay amplitudes at order $$\mathcal{O}(\Lambda _\mathrm{QCD}/m_b)$$ O ( Λ QCD / m b ) for the first time. Our results are very precise, and uncover a substantial discrepancy between the theory predictions and the experimental measurements. We explore two possibilities for this discrepancy: non-factorizable contributions larger than predicted by the QCD factorization power counting, and contributions beyond the Standard Model. We determine the $$f_s/f_d$$ f s / f d fragmentation fraction for the CDF, D0 and LHCb experiments for both scenarios.

Abstract We carry out a comprehensive analysis of the full set of $${\bar{B}}_q \rightarrow D_q^{(*)}$$ B¯q→Dq(∗) form factors for spectator quarks $$q=u,d,s$$ q=u,d,s within the framework of the Heavy-Quark expansion (HQE) to order $${\mathcal {O}}\left( \alpha _s, 1/m_b, 1/m_c^2\right) $$ Oαs,1/mb,1/mc2 . In addition to the available lattice QCD calculations we make use of two new sets of theoretical constraints: we produce for the first time numerical predictions for the full set of $${\bar{B}}_s \rightarrow D_s^{(*)}$$ B¯s→Ds(∗) form factors using Light-Cone sum rules with $$B_s$$ Bs -meson distribution amplitudes. Furthermore, we reassess the QCD three-point sum rule results for the Isgur-Wise functions entering all our form factors for both $$q=u,d$$ q=u,d and $$q=s$$ q=s spectator quarks. These additional constraints allow us to go beyond the commonly used assumption of $$SU(3)_F$$ SU(3)F symmetry for the $${{\bar{B}}}_s\rightarrow D_s^{(*)}$$ B¯s→Ds(∗) form factors, especially in the unitarity constraints which we impose throughout our analysis. We find the coefficients of the IW functions emerging at $${\mathcal {O}}\left( 1/m_c^2\right) $$ O1/mc2 to be consistent with the naive $${\mathcal {O}}\left( 1\right) $$ O1 expectation, indicating a good convergence of the HQE. While we do not find significant SU(3) breaking, the explicit treatment of $$q=s$$ q=s as compared to a simple symmetry assumption renders the unitarity constraints more effective. We find that the (pseudo)scalar bounds are saturated to a large degree, which affects our theory predictions. We analyze the phenomenological consequences of our improved form factors by extracting $$|V_{cb}|$$ |Vcb| from $${\bar{B}}\rightarrow D^{(*)}\ell \nu $$ B¯→D(∗)ℓν decays and producing theoretical predictions for the lepton-flavour universality ratios R(D), $$R(D^*)$$ R(D∗) , $$R(D_s)$$ R(Ds) and $$R(D_s^*)$$ R(Ds∗) , as well as the $$\tau $$ τ - and $$D_q^*$$ Dq∗ polarization fractions for the $${\bar{B}}_q\rightarrow D_q^{(*)}\tau \nu $$ B¯q→Dq(∗)τν modes.

Abstract We carry out an analysis of the full set of ten $$\bar{B}\rightarrow D^{(*)}$$ B¯→D(∗) form factors within the framework of the Heavy-Quark Expansion (HQE) to order $$\mathcal {O}\left( \alpha _s,\,1/m_b,\,1/m_c^2\right) $$ Oαs,1/mb,1/mc2 , both with and without the use of experimental data. This becomes possible due to a recent calculation of these form factors at and beyond the maximal physical recoil using QCD light-cone sum rules, in combination with constraints from lattice QCD, QCD three-point sum rules and unitarity. We find good agreement amongst the various theoretical results, as well as between the theoretical results and the kinematical distributions in $$\bar{B}\rightarrow D^{(*)}\lbrace e^-,\mu ^-\rbrace \bar{\nu }$$ B¯→D(∗){e-,μ-}ν¯ measurements. The coefficients entering at the $$1/m_c^2$$ 1/mc2 level are found to be of $$\mathcal {O}(1)$$ O(1) , indicating convergence of the HQE. The phenomenological implications of our study include an updated exclusive determination of $$|V_{cb}|$$ |Vcb| in the HQE, which is compatible with both the exclusive determination using the BGL parametrization and with the inclusive determination. We also revisit predictions for the lepton-flavour universality ratios $$R_{D^{(*)}}$$ RD(∗) , the $$\tau $$ τ polarization observables $$P_\tau ^{D^{(*)}}$$ PτD(∗) , and the longitudinal polarization fraction $$F_L$$ FL . Posterior samples for the HQE parameters are provided as ancillary files, allowing for their use in subsequent studies.

Deforestation contributes to global greenhouse gas emissions and must be reduced if the 1.5°C limit to global warming is to be realized. Protected areas represent one intervention for decreasing forest loss and aiding conservation efforts, yet there is intense human pressure on at least one-third of protected areas globally. There have been numerous studies addressing the extent and identifying drivers of deforestation at the local, regional, and global level. Yet few have focused on drivers of deforestation in protected areas in high thematic detail. Here we use a new crowdsourced data set on drivers of tropical forest loss for the period 2008–2019, which has been collected using the Geo-Wiki crowdsourcing application for visual interpretation of very high-resolution imagery by volunteers. Extending on the published data on tree cover and forest loss from the Global Forest Change initiative, we investigate the dominant drivers of deforestation in tropical protected areas situated within 30° north and south of the equator. We find the deforestation rate in protected areas to be lower than the continental average for the Latin Americas (3.4% in protected areas compared to 5.4%) and Africa (3.3% compared to 3.9%), but it exceeds that of unprotected land in Asia (8.5% compared to 8.1%). Consistent with findings from foregoing studies, we also find that pastures and other subsistence agriculture are the dominant deforestation driver in the Latin Americas, while forest management, oil palm, shifting cultivation and other subsistence agriculture dominate in Asia, and shifting cultivation and other subsistence agriculture is the main driver in Africa. However, we find contrasting results in relation to the degree of protection, which indicate that the rate of deforestation in Latin America and Africa in strictly protected areas might even exceed that of areas with no strict protection. This crucial finding highlights the need for further studies based on a bottom up crowdsourced, data collection approach, to investigate drivers of deforestation both inside and outside protected areas.

Tree-ring chronologies encode interannual variability in forest growth rates over long time periods from decades to centuries or even millennia. However, each chronology is a highly localized measurement describing conditions at specific sites where wood samples have been collected. The question whether these local growth variabilites are representative for large geographical regions remains an open issue. To overcome the limitations of interpreting a sparse network of sites, we propose an upscaling approach for annual tree-ring indices that approximate forest growth variability and compute gridded data products that generalize the available information for multiple tree genera. Using regression approaches from machine learning, we predict tree-ring indices in space and time based on climate variables, but considering also species range maps as constraints for the upscaling. We compare various prediction strategies in cross-validation experiments to identify the best performing setup. Our estimated maps of tree-ring indices are the first data products that provide a dense view on forest growth variability at the continental level with 0.5° and 0.0083° spatial resolution covering the years 1902–2013. Furthermore, we find that different genera show very variable spatial patterns of anomalies. We have selected Europe as study region and focused on the six most prominent tree genera, but our approach is very generic and can easily be applied elsewhere. Overall, the study shows perspectives but also limitations for reconstructing spatiotemporal dynamics of complex biological processes. Synaptic ribbons are presynaptic specializations that define eponymous ribbon synapses. Synaptic ribbons are largely composed of RIBEYE, a protein containing an N-terminal A-domain and a carboxyterminal B-domain that is identical with CtBP2, a NAD(H)-binding transcriptional co-repressor. Previously we showed that synaptic ribbons are completely absent in RIBEYE knockout mice in which the RIBEYE A-domain-encoding exon had been deleted, but CtBP2 is still made, demonstrating that the A-domain is required for synaptic ribbon assembly. In the present study, we asked whether the RIBEYE B-domain also has an essential role in the assembly of synaptic ribbons. For this purpose, we made use of RIBEYE knockin mice in which the RIBEYE B-domain was replaced by a fluorescent protein domain, whereas the RIBEYE A-domain was retained unchanged. We found that replacing the RIBEYE B-domain with a fluorescent protein module destabilizes the resulting hybrid protein and causes a complete loss of synaptic ribbons. Our results thus demonstrate an essential role of the RIBEYE B-domain in enabling RIBEYE assembly into synaptic ribbons, reinforcing the notion that RIBEYE is the central organizer of synaptic ribbons.

Robust carbon monitoring systems are needed for land managers to assess and mitigate the changing effects of ecosystem stress on western United States forests, where most aboveground carbon is stored in mountainous areas. Atmospheric carbon uptake via gross primary productivity (GPP) is an important indicator of ecosystem function and is particularly relevant to carbon monitoring systems. However, limited ground-based observations in remote areas with complex topography represent a significant challenge for tracking regional-scale GPP. Satellite observations can help bridge these monitoring gaps, but the accuracy of remote sensing methods for inferring GPP is still limited in montane evergreen needleleaf biomes, where (a) photosynthetic activity is largely decoupled from canopy structure and chlorophyll content, and (b) strong heterogeneity in phenology and atmospheric conditions is difficult to resolve in space and time. Using monthly solar-induced chlorophyll fluorescence (SIF) sampled at ∼4 km from the TROPOspheric Monitoring Instrument (TROPOMI), we show that high-resolution satellite-observed SIF followed ecological expectations of seasonal and elevational patterns of GPP across a 3000 m elevation gradient in the Sierra Nevada mountains of California. After accounting for the effects of high reflected radiance in TROPOMI SIF due to snow cover, the seasonal and elevational patterns of SIF were well correlated with GPP estimates from a machine-learning model (FLUXCOM) and a land surface model (CLM5.0-SP), outperforming other spectral vegetation indices. Differences in the seasonality of TROPOMI SIF and GPP estimates were likely attributed to misrepresentation of moisture limitation and winter photosynthetic activity in FLUXCOM and CLM5.0 respectively, as indicated by discrepancies with GPP derived from eddy covariance observations in the southern Sierra Nevada. These results suggest that satellite-observed SIF can serve as a useful diagnostic and constraint to improve upon estimates of GPP toward multiscale carbon monitoring systems in montane, evergreen conifer biomes at regional scales.

Although river water storage contributes to Total Terrestrial Water Storage (TWS) variations obtained from GRACE satellite gravimetry, it is unclear if computationally expensive river routing schemes are required when GRACE data is used for calibration and validation in global hydrological modeling studies. Here, we investigate the role of river water storage on calibration and validation of a parsimonious global hydrological model. In a multi-criteria calibration approach, the model is constrained against either GRACE TWS or TWS from which river water storage is removed. While we find that removing river water storage changes the TWS constraint regionally and globally, there are no significant implications for model calibration and the resulting simulations. However, adding modeled river water storage a-posteriori to calibrated TWS simulations improves model validation against seasonal GRACE TWS variations globally and regionally, especially in tropics and Northern low- and wetlands. While our findings justify the exclusion of explicit river routing for global model calibration, we find that the inclusion of river water storage is relevant for model evaluation.

Abrupt land changes may have long-lasting effects on local biodiversity. Here, Jung et al. show that past abrupt land change reduces species richness and abundance, and alters assemblage composition, with recovery often taking more than 10 years.

Importing proteins into the endoplasmic reticulum (ER) is essential for about 30% of the human proteome. It involves the targeting of precursor proteins to the ER and their insertion into or translocation across the ER membrane. Furthermore, it relies on signals in the precursor polypeptides and components, which read the signals and facilitate their targeting to a protein-conducting channel in the ER membrane, the Sec61 complex. Compared to the SRP- and TRC-dependent pathways, little is known about the SRP-independent/SND pathway. Our aim was to identify additional components and characterize the client spectrum of the human SND pathway. The established strategy of combining the depletion of the central hSnd2 component from HeLa cells with proteomic and differential protein abundance analysis was used. The SRP and TRC targeting pathways were analyzed in comparison. TMEM109 was characterized as hSnd3. Unlike SRP but similar to TRC, the SND clients are predominantly membrane proteins with N-terminal, central, or C-terminal targeting signals.

Abstract We perform a general model-independent analysis of b → cτ ν ¯ τ $$ b\to c\tau {\overline{\nu}}_{\tau } $$ transitions, including measurements of ℛ D , ℛ D∗, their q 2 differential distributions, the recently measured longitudinal D* polarization F L D ∗ $$ {F}_L^{D\ast } $$ , and constraints from the B c → τ ν ¯ τ $$ {B}_c\to \tau {\overline{\nu}}_{\tau } $$ lifetime, each of which has significant impact on the fit. A global fit to a general set of Wilson coefficients of an effective low-energy Hamiltonian is presented, the solutions of which are interpreted in terms of hypothetical new-physics mediators. From the obtained results we predict selected b → cτ ν ¯ τ $$ b\to c\tau {\overline{\nu}}_{\tau } $$ observables, such as the baryonic transition Λ b → Λ c τ ν ¯ τ $$ {\Lambda}_b\to {\Lambda}_c\tau {\overline{\nu}}_{\tau } $$ , the ratio ℛ J/ψ , the forward-backward asymmetries A FB D ∗ $$ {\mathcal{A}}_{\mathrm{FB}}^{D\left(\ast \right)} $$ , the τ polarization asymmetries P τ D ∗ $$ {\mathcal{P}}_{\tau}^{D\left(\ast \right)} $$ , and the longitudinal D* polarization fraction F L D ∗ $$ {F}_L^{D\ast } $$ . The latter shows presently a slight tension with any new-physics model, such that an improved measurement could have an important impact. We also discuss the potential change due the very recently announced preliminary ℛ D ∗ $$ {\mathrm{\mathcal{R}}}_{\mathcal{D}\left(\ast \right)} $$ measurement by the Belle collaboration.

We discuss the impact of the recent untagged analysis of B0→D⁎lν¯l decays by the Belle Collaboration on the extraction of the CKM element |Vcb| and provide updated SM predictions for the b→cτν observables R(D⁎), Pτ, and FLD⁎. The value of |Vcb| that we find is about 2σ from the one from inclusive semileptonic B decays, and is very sensitive to the slope of the form factor at zero recoil which should soon become available from lattice calculations.

Abstract B decays proceeding via b → cℓν transitions with ℓ = e or μ are tree-level processes in the Standard Model. They are used to measure the CKM element V cb , as such forming an important ingredient in the determination of e.g. the unitarity triangle; hence the question to which extent they can be affected by new physics contributions is important, specifically given the long-standing tension between V cb determinations from inclusive and exclusive decays and the significant hints for lepton flavour universality violation in b → cτ ν and b → sℓℓ decays. We perform a comprehensive model-independent analysis of new physics in b → cℓν, considering all combinations of scalar, vector and tensor interactions occuring in single-mediator scenarios. We include for the first time differential distributions of B → D ∗ ℓν angular observables for this purpose. We show that these are valuable in constraining non-standard interactions. Specifically, the zero-recoil endpoint of the B → Dℓν spectrum is extremely sensitive to scalar currents, while the maximum-recoil endpoint of the B → D ∗ ℓν spectrum with transversely polarized D ∗ is extremely sensitive to tensor currents. We also quantify the room for e-μ universality violation in b → cℓν transitions, predicted by some models suggested to solve the b → cτ ν anomalies, from a global fit to B → Dℓν and B → D ∗ ℓν for the first time. Specific new physics models, corresponding to all possible tree-level mediators, are also discussed. As a side effect, we present V cb determinations from exclusive B decays, both with frequentist and Bayesian statistics, leading to compatible results. The entire numerical analysis is based on open source code, allowing it to be easily adapted once new data or new form factors become available.

Summary: By vacuolar patch-clamp and Ca2+ imaging experiments, we show that the yeast vacuolar transient receptor potential (TRPY) channel 1 is activated by cytosolic Ca2+ and inhibited by Ca2+ from the vacuolar lumen. The channel is cooperatively affected by vacuolar Ca2+ (Hill coefficient, 1.5), suggesting that it may accommodate a Ca2+ receptor that can bind two calcium ions. Alanine scanning of six negatively charged amino acid residues in the transmembrane S5 and S6 linker, facing the vacuolar lumen, revealed that two aspartate residues, 401 and 405, are essential for current inhibition and direct binding of 45Ca2+. Expressed in HEK-293 cells, a significant fraction of TRPY1, present in the plasma membrane, retained its Ca2+ sensitivity. Based on these data and on homology with TRPV channels, we conclude that D401 and D405 are key residues within the vacuolar vestibule of the TRPY1 pore that decrease cation access or permeation after Ca2+ binding. : Structural Biology; Protein Structure Aspects; Biophysics Subject Areas: Structural Biology, Protein Structure Aspects, Biophysics

The Unc119 protein mediates transport of myristoylated proteins to the photoreceptor outer segment, a specialized primary cilium. This transport activity is regulated by the GTPase Arl3 as well as by Arl13b and Rp2 that control Arl3 activation/inactivation. Interestingly, Unc119 is also enriched in photoreceptor synapses and can bind to RIBEYE, the main component of synaptic ribbons. In the present study, we analyzed whether the known regulatory proteins, that control the Unc119-dependent myristoylated protein transport at the primary cilium, are also present at the photoreceptor synaptic ribbon complex by using high-resolution immunofluorescence and immunogold electron microscopy. We found Arl3 and Arl13b to be enriched at the synaptic ribbon whereas Rp2 was predominantly found on vesicles distributed within the entire terminal. These findings indicate that the synaptic ribbon could be involved in the discharge of Unc119-bound lipid-modified proteins. In agreement with this hypothesis, we found Nphp3 (Nephrocystin-3), a myristoylated, Unc119-dependent cargo protein enriched at the basal portion of the ribbon in close vicinity to the active zone. Mutations in Nphp3 are known to be associated with Senior–Løken Syndrome 3 (SLS3). Visual impairment and blindness in SLS3 might thus not only result from ciliary dysfunctions but also from malfunctions of the photoreceptor synapse.

Summary: Multiple sclerosis (MS) is a demyelinating disease caused by an auto-reactive immune system. Recent studies also demonstrated synapse dysfunctions in MS patients and MS mouse models. We previously observed decreased synaptic vesicle exocytosis in photoreceptor synapses in the EAE mouse model of MS at an early, preclinical stage. In the present study, we analyzed whether synaptic defects are associated with altered presynaptic Ca2+ signaling. Using high-resolution immunolabeling, we found a reduced signal intensity of Cav-channels and RIM2 at active zones in early, preclinical EAE. In line with these morphological alterations, depolarization-evoked increases of presynaptic Ca2+ were significantly smaller. In contrast, basal presynaptic Ca2+ was elevated. We observed a decreased expression of Na+/K+-ATPase and plasma membrane Ca2+ ATPase 2 (PMCA2), but not PMCA1, in photoreceptor terminals of EAE mice that could contribute to elevated basal Ca2+. Thus, complex Ca2+ signaling alterations contribute to synaptic dysfunctions in photoreceptors in early EAE.

Photosynthesis of terrestrial ecosystems in the Arctic-Boreal region is a critical part of the global carbon cycle. Solar-induced chlorophyll Fluorescence (SIF), a promising proxy for photosynthesis with physiological insight, has been used to track gross primary production (GPP) at regional scales. Recent studies have constructed empirical relationships between SIF and eddy covariance-derived GPP as a first step to predicting global GPP. However, high latitudes pose two specific challenges: (a) Unique plant species and land cover types in the Arctic–Boreal region are not included in the generalized SIF-GPP relationship from lower latitudes, and (b) the complex terrain and sub-pixel land cover further complicate the interpretation of the SIF-GPP relationship. In this study, we focused on the Arctic-Boreal vulnerability experiment (ABoVE) domain and evaluated the empirical relationships between SIF for high latitudes from the TROPOspheric Monitoring Instrument (TROPOMI) and a state-of-the-art machine learning GPP product (FluxCom). For the first time, we report the regression slope, linear correlation coefficient, and the goodness of the fit of SIF-GPP relationships for Arctic-Boreal land cover types with extensive spatial coverage. We found several potential issues specific to the Arctic-Boreal region that should be considered: (a) unrealistically high FluxCom GPP due to the presence of snow and water at the subpixel scale; (b) changing biomass distribution and SIF-GPP relationship along elevational gradients, and (c) limited perspective and misrepresentation of heterogeneous land cover across spatial resolutions. Taken together, our results will help improve the estimation of GPP using SIF in terrestrial biosphere models and cope with model-data uncertainties in the Arctic-Boreal region.

Endoplasmic reticulum (ER) functioning requires a constant supply of ATP, but the exchanger required for ATP uptake into the ER is unknown. Here, the authors report that SLC35B1, here named AXER, or ATP/ADP exchanger into the ER, can transport ATP into the ER.

Abstract Background CredelioTM (lotilaner) is an oral ectoparasiticide from the isoxazoline class developed for the treatment of flea and tick infestations in cats. It is formulated as a pure S-enantiomer in flavoured chewable tablets. The pharmacokinetics of lotilaner were investigated after intravenous or oral administration and under fed or fasted conditions in cats. Twenty-six adult cats were enrolled in a pharmacokinetic study evaluating either intravenous or oral administration of lotilaner. Following the oral administration at a dosage of 6 mg/kg, under fed or fasted conditions, or intravenous administration of 3 mg/kg, blood samples were collected up to 35 days after treatment. Lotilaner blood concentrations were measured using a validated liquid chromatography/tandem mass spectrometry method. Pharmacokinetic parameters were calculated by non-compartmental analysis. In addition, in vivo enantiomer stability of lotilaner was evaluated in a separate bioanalytical study. Results Following oral administration in fed cats, lotilaner was readily absorbed and peak blood concentrations reached within four hours. The terminal half-life was 33.6 days. Food enhanced the absorption, providing close to 100% oral bioavailability and reduced the inter-individual variability. Following intravenous administration, lotilaner had a low clearance of 0.13 l/kg/day, large volumes of distribution Vz and Vss of 5.34 and 5.37 l/kg, respectively and a terminal half-life of 28.7 days. In addition, there was no in vivo racemization of lotilaner. Conclusions The pharmacokinetic properties of lotilaner administered orally as a flavoured chewable tablet (CredelioTM) were studied in detail. With a Tmax of 4 h and a terminal half-life of 33.6 days under fed conditions, lotilaner provides a rapid onset of flea and tick killing activity with consistent and sustained efficacy for at least one month in cats.

Abstract We perform model-independent analyses extracting limits for the electric dipole moment of the electron and the P,T-odd scalar-pseudoscalar (S-PS) nucleon-electron coupling from the most recent measurements with atoms and molecules. The analysis using paramagnetic systems, only, is improved substantially by the inclusion of the recent measurement on HfF+ ions, but complicated by the fact that the corresponding constraints are largely aligned, owing to a general relation between the coefficients for the two contributions. Since this same relation does not hold in diamagnetic systems, it is possible to find atoms that provide essentially orthogonal constraints to those from paramagnetic ones. However, the coefficients are suppressed in closed-shell systems and enhancements of P,T-odd effects are only prevalent in the presence of hyperfine interactions. We formulate the hyperfine-induced time-reversal-symmetry breaking S-PS nucleon-electron interaction in general atoms in a mixed perturbative and variational approach, based on electronic Dirac-wavefunctions including the effects of electron correlations. The method is applied to the Hg atom, yielding the first direct calculation of the coefficient of the S-PS nucleon-electron coupling in a diamagnetic system. This results in additionally improved model-independent limits for both the electron EDM and the nucleon-electron coupling from the global fit. Finally we employ this fit to provide indirect limits for several paramagnetic systems under investigation.

Abstract The renormalization group equations (RGEs) in Standard Model effective theory are usually either solved analytically, neglecting the scale dependence of gauge and Yukawa couplings, or numerically without such approximations. We present analytic solutions of RGEs that take into account the dominant scale dependence of the anomalous-dimension matrix due to the running of the QCD coupling α s and of the top-Yukawa coupling. We consider first the case for which a given operator is generated directly through mixing with the parent operator whose Wilson coefficient is non-vanishing at the new physics scale. Subsequently we consider the case of two-step running, in which two operators do not mix directly, but only via a third mediator operator. We generalize these solutions to an arbitrary number of operators and show how even in this case analytic solutions can be obtained.

Various landmark studies have revealed structures and functions of the Sec61/SecY complex in all domains of live demonstrating the conserved nature of this ancestral protein translocase. While the bacterial homolog of the Sec61 complex resides in the plasma membrane, the eukaryotic counterpart manages the transfer of precursor proteins into or across the membrane of the endoplasmic reticulum (ER). Sec61 complexes are accompanied by a set of dynamically recruited auxiliary proteins assisting the transport of certain precursor polypeptides. TRAP and Sec62/Sec63 are two auxiliary protein complexes in mammalian cells that have been characterized by structural and biochemical methods. Using these ER membrane protein complexes for our proof-of-concept study, we aimed to detect interactions of membrane proteins in living mammalian cells under physiological conditions. Bimolecular luminescence complementation and competition was used to demonstrate multiple protein–protein interactions of different topological layouts. In addition to the interaction of the soluble catalytic and regulatory subunits of the cytosolic protein kinase A, we detected interactions of ER membrane proteins that either belong to the same multimeric protein complex (intra-complex interactions: Sec61α–Sec61β, TRAPα–TRAPβ) or protein complexes in juxtaposition (inter-complex interactions: Sec61α–TRAPα, Sec61α–Sec63, and Sec61β–Sec63). In the process, we established further control elements like synthetic peptide complementation for expression profiling of fusion constructs and protease-mediated reporter degradation demonstrating the cytosolic localization of a reporter complementation. Ease of use and flexibility of the approach presented here will spur further research regarding the dynamics of protein–protein interactions in response to changing cellular conditions in living cells.

Abstract Background Lotilaner is a new oral ectoparasiticide from the isoxazoline class developed for the treatment of flea and tick infestations in dogs. Lotilaner blood concentrations were measured using a validated liquid chromatography/tandem mass spectrometry (LC-MS/MS) method. Pharmacokinetic parameters were calculated by non-compartmental analysis. In addition, in vivo enantiomer stability was evaluated in an analytical study. Results Following oral administration in fed animals, lotilaner was readily absorbed and peak blood concentrations reached within 2 hours. The terminal half-life was 30.7 days. Food enhanced the absorption, providing an oral bioavailability above 80% and reduced the inter-individual variability. Moreover, the time of feeding with respect to dosing (fed 30 min prior, fed at dosing or fed 30 min post-dosing) or the reduction of the food ration to one-third of the normal daily ration did not impact bioavailability. Following intravenous administration, lotilaner had a low clearance of 0.18 l/kg/day, large volumes of distribution Vz and Vss of 6.35 and 6.45 l/kg, respectively and a terminal half-life of 24.6 days. In addition, there was no in vivo racemization of lotilaner. Conclusions The pharmacokinetic properties of lotilaner administered orally as a flavoured chewable tablet (Credelio™) were studied in detail. With a Tmax of 2 h and a terminal half-life of 30.7 days under fed conditions, lotilaner provides a rapid onset of flea and tick killing activity with consistent and sustained efficacy for at least 1 month.

We perform a comprehensive analysis of scalar contributions in b→cτν transitions including the latest measurements of R(D(⁎)), the q2 differential distributions in B→D(⁎)τν, the τ polarization asymmetry for B→D⁎τν, and the bound derived from the total width of the Bc meson. We find that scalar contributions with the simultaneous presence of both left- and right-handed couplings to quarks can explain the available data, specifically R(D(⁎)) together with the measured differential distributions. However, the constraints from the total Bc width present a slight tension with the current data on B→D⁎τν in this scenario, preferring smaller values for R(D⁎). We discuss possibilities to disentangle scalar new physics from other new-physics scenarios like the presence of only a left-handed vector current, via additional observables in B→D(⁎)τν decays or additional decay modes like the baryonic Λb→Λcτν and the inclusive B→Xcτν decays. We also analyze scalar contributions in b→uτν transitions, including the latest measurements of B→τν, providing predictions for Λb→pτν and B→πτν decays. The potential complementarity between the b→u and b→c sectors is finally investigated once assumptions about the flavour structure of the underlying theory are made.

Abstract We discuss Yukawa-enhanced contributions from Z-mediated new physics to down-type quark ΔF = 2 processes in the framework of the standard model gauge-invariant effective theory (SMEFT). Besides the renormalization group (RG) mixing of the Z-mediating ψ 2 H 2 D operators into ΔF = 2 operators, we include at the electroweak scale one-loop (NLO) matching corrections consistently, necessary for the removal of the matching scale dependence. We point out that the right-handed Z-mediated interactions generate through Yukawa RG mixing ΔF = 2 left-right operators, which are further enhanced through QCD RG effects and chirally enhanced hadronic matrix elements. We investigate the impact of these new effects on the known correlations between ΔF = 2 and ΔF = 1 transitions in the SMEFT framework and point out qualitative differences to previous parameterizations of Z-mediated new physics that arise for the left-handed case. We illustrate how specific models fit into our model-independent framework by using four models with vector-like quarks. We carry out model-independent analyses of scenarios with purely left-handed and purely right-handed new-physics Z couplings for each of the three sectors s → d, b → s and b → d. Specifically we discuss the correlations between ε′/ε, ε K , K L → μ + μ − K + → π + ν ν ¯ $$ {K}_L\to {\mu}^{+}{\mu}^{-}{K}^{+}\to {\pi}^{+}\nu \overline{\nu} $$ and K L → π 0 ν ν ¯ $$ {K}_L\to {\pi}^0\nu \overline{\nu} $$ in the Kaon sector, and ϕ s , B s → μ + μ − and B → K (*)(μ + μ − , ν ν ¯ $$ \nu \overline{\nu} $$ ) in the b → s sector and B d → μ + μ − in the b → d sector.

Abstract We study the patterns of flavour violation in renormalisable extensions of the Standard Model (SM) that contain vector-like quarks (VLQs) in a single complex representation of either the SM gauge group GSM or G SM ′ ≡ GSM ⊗ U(1)L μ − L τ . We first decouple VLQs in the M = (1 − 10) TeV range and then at the electroweak scale also Z, Z ′ gauge bosons and additional scalars to study the phenomenology. The results depend on the relative size of Z- and Z ′-induced flavour-changing neutral currents, as well as the size of |ΔF | = 2 contributions including the effects of renormalisation group Yukawa evolution from M to the electroweak scale that turn out to be very important for models with right-handed currents through the generation of left-right operators. In addition to rare decays like P → ℓ ℓ ¯ , P → P ′ ℓ ℓ ¯ , P → P ′ ν ν ¯ $$ P\to \ell \overline{\ell},P\to {P}^{\prime}\ell \overline{\ell},P\to {P}^{\prime}\nu \overline{\nu} $$ with P = K, B s , B d and |ΔF | = 2 observables we analyze the ratio ε ′ /ε which appears in the SM to be significantly below the data. We study patterns and correlations between these observables which taken together should in the future allow for differentiating between VLQ models. In particular the patterns in models with left-handed and right-handed currents are markedly different from each other. Among the highlights are large Z-mediated new physics effects in Kaon observables in some of the models and significant effects in B s,d -observables. ε ′ /ε can easily be made consistent with the data, implying then uniquely the suppression of K L → π 0 ν ν ¯ $$ {K}_L\to {\pi}^0\nu\ \overline{\nu} $$ . Significant enhancements of B r K + → π + ν ν ¯ $$ Br\left({K}^{+}\to {\pi}^{+}\nu \overline{\nu}\right) $$ are still possible. We point out that the combination of NP effects to |ΔF | = 2 and |ΔF | = 1 observables in a given meson system generally allows to determine the masses of VLQs in a given representation independently of the size of VLQ couplings.

Abstract The PREDICTS project—Projecting Responses of Ecological Diversity In Changing Terrestrial Systems (https://www.nhm.ac.uk/our-science/our-work/biodiversity/predicts.html)—has collated from published studies a large, reasonably representative database of comparable samples of biodiversity from multiple sites that differ in the nature or intensity of human impacts relating to land use. We have used this evidence base to develop global and regional statistical models of how local biodiversity responds to these measures. We describe and make freely available this 2016 release of the database, containing more than 3.2 million records sampled at over 26,000 locations and representing over 47,000 species. We outline how the database can help in answering a range of questions in ecology and conservation biology. To our knowledge, this is the largest and most geographically and taxonomically representative database of spatial comparisons of biodiversity that has been collated to date; it will be useful to researchers and international efforts wishing to model and understand the global status of biodiversity.

Forests play a crucial role in the global carbon (C) cycle by storing and sequestering a substantial amount of C in the terrestrial biosphere. Due to temporal dynamics in climate and vegetation activity, there are significant regional variations in carbon dioxide (CO2) fluxes between the biosphere and atmosphere in forests that are affecting the global C cycle. Current forest CO2 flux dynamics are controlled by instantaneous climate, soil, and vegetation conditions, which carry legacy effects from disturbances and extreme climate events. Our level of understanding from the legacies of these processes on net CO2 fluxes is still limited due to their complexities and their long-term effects. Here, we combined remote sensing, climate, and eddy-covariance flux data to study net ecosystem CO2 exchange (NEE) at 185 forest sites globally. Instead of commonly used non-dynamic statistical methods, we employed a type of recurrent neural network (RNN), called Long Short-Term Memory network (LSTM) that captures information from the vegetation and climate's temporal dynamics. The resulting data-driven model integrates interannual and seasonal variations of climate and vegetation by using Landsat and climate data at each site. The presented LSTM algorithm was able to effectively describe the overall seasonal variability (Nash-Sutcliffe efficiency, NSE = 0.66) and across-site (NSE = 0.42) variations in NEE, while it had less success in predicting specific seasonal and interannual anomalies (NSE = 0.07). This analysis demonstrated that an LSTM approach with embedded climate and vegetation memory effects outperformed a non-dynamic statistical model (i.e. Random Forest) for estimating NEE. Additionally, it is shown that the vegetation mean seasonal cycle embeds most of the information content to realistically explain the spatial and seasonal variations in NEE. These findings show the relevance of capturing memory effects from both climate and vegetation in quantifying spatio-temporal variations in forest NEE.

The approximate symmetry of the strong interactions under isospin transformations is among the most precise tools available to control hadronic matrix elements. It is crucial in extracting fundamental parameters, but also provides avenues for the search of phenomena beyond the Standard Model. The precision of the resulting predictions requires special care when determining the quantities they are to be tested with. Specifically, in the extraction of branching ratios often isospin symmetry is assumed at one point or another implicitly, implying a significant bias for precision analyses. We extract a bias-free value for the production asymmetry between charged and neutral B meson pairs at B factories and discuss its consequences for the determination of branching fractions generally, and isospin-violating observables like the rate asymmetries in B→J/ψK or B→K⁎γ decays specifically.

The plasma membrane transporter SOS1 (SALT-OVERLY SENSITIVE1) is vital for plant survival under salt stress. SOS1 activity is tightly regulated, but little is known about the underlying mechanism. SOS1 contains a cytosolic, autoinhibitory C-terminal tail (abbreviated as SOS1 C-term), which is targeted by the protein kinase SOS2 to trigger its transport activity. Here, to identify additional binding proteins that regulate SOS1 activity, we synthesized the SOS1 C-term domain and used it as bait to probe Arabidopsis thaliana cell extracts. Several 14-3-3 proteins, which function in plant salt tolerance, specifically bound to and interacted with the SOS1 C-term. Compared to wild-type plants, when exposed to salt stress, Arabidopsis plants overexpressing SOS1 C-term showed improved salt tolerance, significantly reduced Na+ accumulation in leaves, reduced induction of the salt-responsive gene WRKY25, decreased soluble sugar, starch, and proline levels, less impaired inflorescence formation and increased biomass. It appears that overexpressing SOS1 C-term leads to the sequestration of inhibitory 14-3-3 proteins, allowing SOS1 to be more readily activated and leading to increased salt tolerance. We propose that the SOS1 C-term binds to previously unknown proteins such as 14-3-3 isoforms, thereby regulating salt tolerance. This finding uncovers another regulatory layer of the plant salt tolerance program.
