Biotoxins produced by harmful algal blooms (HABs) are a substantial global threat to ocean and human health. Domoic acid (DA) is one such biotoxin whose negative impacts are forecasted to increase with climate change and coastal development. This manuscript serves as a review of DA toxicosis after environmental exposure in humans and wildlife, including an introduction to HAB toxins, the history of DA toxicosis, DA production, toxicokinetic properties of DA, susceptibility, clinical signs, DA detection methods and other diagnostic tests, time course of toxicosis, treatment, prognostics, and recommendations for future research. Additionally, we highlight the utility of California sea lions (CSLs; Zalophus californianus) as a model and sentinel of environmental DA exposure. [ABSTRACT FROM AUTHOR]

Klebsiella pneumoniae is a Gram-negative, facultative anaerobic, and non-motile bacillus frequently observed in mammals. A hypermucoviscous (HMV) phenotype of K. pneumoniae is reported in stranded California sea lions (CSL) causing pleuritis and bronchopneumonia as a persistent cause of strandings. We describe an increase in cases of HMV and non-HMV K. pneumoniae in stranded CSLs along the California coast between 2017 to 2022. 2,146 live-stranded CSLs were rescued by The Marine Mammal Center (Sausalito, California). 47 of these tested positive for K. pneumoniae , with a rise in annual incidence from 0.92% (3/331) in 2017 to 8.5% (14/165) and 7.2% (23/320) in 2021 and 2022, respectively. These strandings occurred across 965 kilometers along the North-Central California coast from Bodega Bay, Sonoma, to Avila Beach, San Luis Obispo. A similar pattern was observed in beach-cast CSL carcasses by the Long Marine Laboratory (LML) Stranding Network in Santa Cruz, California who had 8 CSLs test positive for K. pneumoniae with an annual incidence rate of 2.0% (1/50) and 10.1% (7/69) in 2021 and 2022, respectively, with no cases reported prior to 2021. We applied whole-genome sequencing (WGS) to 8 isolates obtained by LML in 2022. WGS analysis revealed high nucleotide similarity between any pair of isolates. Multilocus Sequence Typing showed that all isolates belonged to Sequence type 65 and contained the K2 polysaccharide capsule and siderophore aerobactin, which are all characteristics of hypervirulent K. pneumoniae. The C-terminal region of cardiac troponin I (cTnI) is known to be important in cardiac function, as removal of the last 17 C-terminal residues of human cTnI has been associated with myocardial stunning. To investigate the C-terminal region of cTnI, three C-terminal deletion mutations in human cTnI were generated: Δ1 (deletion of residue 210), Δ3 (deletion of residues 208-210), and Δ5 (deletion of residues 206-210). Mammalian two-hybrid studies showed that the interactions between cTnI mutants and cardiac troponin C (cTnC) or cardiac troponin T (cTnT) were impaired in Δ3 and Δ5 mutants when compared to wild-type cTnI. Two emaciated male northern elephant seal (NES) Mirounga angustirostris pups were admitted to The Marine Mammal Center (Sausalito, California, USA) and treated for malnutrition. Complete blood counts showed a progressive moderate to marked leukocytosis characterized by a predominance of large monomorphic mononuclear cells of probable lymphoid origin, frequently with flower-shaped nuclei. Both seals were euthanized due to suspected lymphoid neoplasia. At necropsy, most lymph nodes in both pups were markedly enlarged, some with distinct white nodules, the spleens were diffusely enlarged, and the intestinal mucosae were thickened. Histopathologic features consistent with disseminated large cell lymphoma were identified to varying degrees of severity in lymph nodes, bone marrow, liver, tonsils, spleen, liver, intestines, kidneys, lower urinary tract, and several other organs. Immunohistochemical staining of neoplastic cells was most consistent with B lymphocyte origin, with most cells staining positively for Pax 5 and CD20 with admixed small CD3-positive T lymphocytes and CD204-positive macrophages. PCR and sequencing identified a novel gammaherpesvirus, herein called miroungine gammaherpesvirus 3, from affected tissues. This virus is in a clade outside of named genera that utilize hosts in the suborder Caniformia. The present study is the first description of diffuse large B cell lymphoma with leukemic manifestation and concomitant detection of a novel gammaherpesvirus in free-living NESs. Further research regarding the prevalence of this new gammaherpesvirus and its associated pathogenesis in this species is indicated.
