Abstract

Euclid will cover over 14000 $deg^{2}$ with two optical and near-infrared spectro-photometric instruments, and is expected to detect around ten million active galactic nuclei (AGN). This unique data set will make a considerable impact on our understanding of galaxy evolution and AGN. In this work we identify the best colour selection criteria for AGN, based only on Euclid photometry or including ancillary photometric observations, such as the data that will be available with the Rubin legacy survey of space and time (LSST) and observations already available from Spitzer/IRAC. The analysis is performed for unobscured AGN, obscured AGN, and composite (AGN and star-forming) objects. We make use of the spectro-photometric realisations of infrared-selected targets at all-z (SPRITZ) to create mock catalogues mimicking both the Euclid Wide Survey (EWS) and the Euclid Deep Survey (EDS). Using these catalogues we estimate the best colour selection, maximising the harmonic mean (F1) of completeness and purity. The selection of unobscured AGN in both Euclid surveys is possible with Euclid photometry alone with F1=0.22-0.23, which can increase to F1=0.43-0.38 if we limit at z>0.7. Such selection is improved once the Rubin/LSST filters (a combination of the u, g, r, or z filters) are considered, reaching F1=0.84 and 0.86 for the EDS and EWS, respectively. The combination of a Euclid colour with the [3.6]-[4.5] colour, which is possible only in the EDS, results in an F1-score of 0.59, improving the results using only Euclid filters, but worse than the selection combining Euclid and LSST. The selection of composite ($f_{{\rm AGN}}$=0.05-0.65 at 8-40 $\mu m$) and obscured AGN is challenging, with F1<0.3 even when including ancillary data. Abstract

Background: Controversy exists regarding the optimal duration of the extended adjuvant endocrine treatment (ET) in patients with early-stage breast-cancer (eBC).We performed a systematic review and trial-level meta-analysis of all randomized clinical trials (RCTs) comparing a “limited-extended” adjuvant ET (defined as more than 5 but less than 7.5 years of treatment overall) versus a “full-extended” adjuvant ET (defined as more than 7.5 years of treatment overall) in eBC. Methods: To be eligible, RCTs had to i) compare a “limited-extended” adjuvant ET versus a “full-extended” adjuvant ET in patients with eBC; and ii) report disease-free survival (DFS) hazard ratio (HR) according to the disease nodal-status [i.e., nodal-negative (N-ve) versus nodal-positive (N + ve)].The primary endpoint was to assess the difference in efficacy of full-versus limited-extended ET, measured in terms of the difference in DFS log-HR, according to the disease nodal-status. Secondary endpoint was the difference in efficacy of full-versus limited-extended ET according to tumor size (i.e., pT1 vs pT2/3/4), histological grade (i.e., G1/G2 vs G3), patients’ age (i.e., ≤60 vs > 60 years) and previous type of ET (i.e., aromatase inhibitors vs tamoxifen vs switch strategy). Results: Three phase III RCTs fulfilled the inclusion criteria. A total of 6689 patients were included in the analysis, of which 3506 (53%) had N + ve disease.The full-extended ET provided no DFS-benefit as compared with the limited-extended ET in patients with N-ve disease (pooled DFS-HR = 1.04, 95%CI: 0.89 to 1.22; I2 = 18%).Conversely, in patients with N + ve disease the full-extended ET significantly improved DFS, with a pooled DFS-HR of 0.85 (95%CI: 0.74 to 0.97; I2 = 0%).There was a significant interaction between the disease nodal-status and the efficacy of the full-versus limited-extended ET (p-heterogeneity = 0.048).The full-extended ET provided no significant DFS-benefit as compared with the limited-extended ET in all the other subgroups analyzed. Conclusions: Patients with eBC and N + ve disease can obtain a significant DFS-benefit from the full-extended as compared with the limited-extended adjuvant ET.

Abstract

The Euclid space mission will cover over 14 000 deg2 with two optical and near-infrared spectro-photometric instruments, and is expected to detect around ten million active galactic nuclei (AGN). This unique data set will make a considerable impact on our understanding of galaxy evolution in general, and AGN in particular. For this work we identified the best colour selection criteria for AGN, based only on Euclid photometry or including ancillary photometric observations, such as the data that will be available with the Rubin Legacy Survey of Space and Time (LSST) and observations already available from Spitzer/IRAC. The analysis was performed for unobscured AGN, obscured AGN, and composite (AGN and star-forming) objects. We made use of the spectro-photometric realisations of infrared-selected targets at all-z (SPRITZ) to create mock catalogues mimicking both the Euclid Wide Survey (EWS) and the Euclid Deep Survey (EDS). Using these mock catalogues, we estimated the best colour selection, maximising the harmonic mean (F1) of: (a) completeness, that is, the fraction of AGN correctly selected with respect to the total AGN sample; and (b) purity, that is, the fraction of AGN inside the selection with respect to the selected sample. The selection of unobscured AGN in both Euclid surveys (Wide and Deep) is possible with Euclid photometry alone with F1 = 0.22–0.23 (Wide and Deep), which can increase to F1 = 0.43–0.38 (Wide and Deep) if we limit out study to objects at z > 0.7. Such a selection is improved once the Rubin/LSST filters, that is, a combination of the u, g, r, or z filters, are considered, reaching an F1 score of 0.84 and 0.86 for the EDS and EWS, respectively. The combination of a Euclid colour with the [3.6]−[4.5] colour, which is possible only in the EDS, results in an F1 score of 0.59, improving the results using only Euclid filters, but worse than the selection combining Euclid and LSST colours. The selection of composite (fAGN = 0.05–0.65 at 8–40 μm) and obscured AGN is challenging, with F1 ≤ 0.3 even when including Rubin/LSST or IRAC filters. This is unsurprising since it is driven by the similarities between the broad-band spectral energy distribution of these AGN and star-forming galaxies in the wavelength range 0.3–5 μm. Abstract

The Euclid space mission will cover over 14 000 deg2 with two optical and near-infrared spectro-photometric instruments, and is expected to detect around ten million active galactic nuclei (AGN). This unique data set will make a considerable impact on our understanding of galaxy evolution in general, and AGN in particular. For this work we identified the best colour selection criteria for AGN, based only on Euclid photometry or including ancillary photometric observations, such as the data that will be available with the Rubin Legacy Survey of Space and Time (LSST) and observations already available from Spitzer/IRAC. The analysis was performed for unobscured AGN, obscured AGN, and composite (AGN and star-forming) objects. We made use of the spectro-photometric realisations of infrared-selected targets at all-z (SPRITZ) to create mock catalogues mimicking both the Euclid Wide Survey (EWS) and the Euclid Deep Survey (EDS). Using these mock catalogues, we estimated the best colour selection, maximising the harmonic mean (F1) of: (a) completeness, that is, the fraction of AGN correctly selected with respect to the total AGN sample; and (b) purity, that is, the fraction of AGN inside the selection with respect to the selected sample. The selection of unobscured AGN in both Euclid surveys (Wide and Deep) is possible with Euclid photometry alone with F1 = 0.22–0.23 (Wide and Deep), which can increase to F1 = 0.43–0.38 (Wide and Deep) if we limit out study to objects at z > 0.7. Such a selection is improved once the Rubin/LSST filters, that is, a combination of the u, g, r, or z filters, are considered, reaching an F1 score of 0.84 and 0.86 for the EDS and EWS, respectively. Abstract

Background: Failure to rescue (FTR) is defined as the inability to prevent death after the development of a complication. FTR is a parameter in evaluating multidisciplinary postoperative complication management. The aim of this study was to evaluate FTR rates after major liver resection for perihilar cholangiocarcinoma (pCCA) and analyze factors associated with FTR. Patients and Method: Patients who underwent major liver resection for pCCA at 27 centers were included. FTR was defined as the presence of a Dindo grade III or higher complication followed by death within 90 days after surgery. Liver failure ISGLS grade B/C were scored. Multivariable logistic analysis was performed to identify predictors of FTR and reported using odds ratio and 95% confidence intervals. Results: In the 2186 included patients, major morbidity rate was 49%, 90-day mortality rate 13%, and FTR occurred in 24% of patients with a grade III or higher complication. Across centers, major complication rate varied from 19 to 87%, 90-day mortality rate from 5 to 33%, and FTR ranged from 11 to 50% across hospitals. Age [1.04 (1.02–1.05) years], ASA 3 or 4 [1.40 (1.01–1.95)], jaundice at presentation [1.79 (1.16–2.76)], right-sided resection [1.45 (1.06–1.98)], and annual hospital volume < 6 [1.44 (1.07–1.94)] were positively associated with FTR. When liver failure is included, the odds ratio for FTR is 9.58 (6.76–13.68). Conclusion: FTR occurred in 24% of patients after resection for pCCA. Liver failure was associated with a nine-fold increase of FTR and hospital volume below six was also associated with an increased risk of FTR.

Abstract

CoQ10 (Coenzyme Q10) is an essential fat-soluble metabolite that plays a key role in cellular metabolism. A less-known function of CoQ10 is whether it may act as a plasma membrane-stabilizing agent and whether this property can affect cancer development and progression. Here, we show that CoQ10 and its biosynthetic enzyme UBIAD1 play a critical role in plasmamembrane mechanical properties that are of interest for breast cancer (BC) progression and treatment. CoQ10 and UBIAD1 increase membrane fluidity leading to increased cell stiffness in BC. Furthermore, CoQ10 and UBIAD1 states impair ECM (extracellular matrix)-mediated oncogenic signaling and reduce ferroptosis resistance in BC settings. Analyses on human patients and mouse models reveal that UBIAD1 loss is associated with BC development and progression and UBIAD1 expression in BC limits CTCs (circulating tumor cells) survival and lung metastasis formation. Overall, this study reveals that CoQ10 and UBIAD1 can be further investigated to develop therapeutic interventions to treat BC patients with poor prognosis. The roles of CoQ10 and producing enzymes in cancer have not been well explored. Here, the authors identify that the CoQ10-biosynthetic enzyme UBIAD1 is a tumor suppressor in breast cancer. CoQ10 induced alterations in plasma membrane mechanical properties lead to increased cellular stiffness and impaired tumorigenic signaling, as well as enhanced sensitivity to ferroptosis. Growth cones at the tips of extending axons navigate through developing organisms by probing extracellular cues, which guide them through intermediate steps and onto final synaptic target sites. Widespread focus on a few guidance cue families has historically overshadowed potentially crucial roles of less well-studied growth factors in axon guidance. In fact, recent evidence suggests that a variety of growth factors have the ability to guide axons, affecting the targeting and morphogenesis of growth cones in vitro. This review summarizes in vitro experiments identifying responses and signaling mechanisms underlying axon morphogenesis caused by underappreciated growth factors.

Abstract

The purpose of the present programme was to develop and validate a theoretically-grounded instrument to measure the planned and unplanned dimensions of lifestyle PA (PPA and UPA; Dunn, Andersen, & Jakicic, 1998). In Study 1, two samples of British adults (Internet: N = 742; paper: N = 563) were used to establish the content validity of the Brunel Lifestyle Physical Activity Questionnaire (BLPAQ). Exploratory factor analysis yielded a two-factor model (UPA and PPA) that produced acceptable fit indices using confirmatory factors analyses with both samples. The purpose of Study 2 was to examine the test-retest reliability of the BLPAQ over 5 weeks using a sample of leisure centre users, university staff members, and university students (N = 337). High correlations were observed between the two administrations (range = .93-.98; p < .01). Thereafter, the data were subjected to proportion of agreement (PoA) analysis as advocated by Nevill, Lane, Kilgor, Bowes, and Whyte (2001). Both PPA and UPA demonstrated satisfactorily high internal agreement (PoA > 95%). In Study 3, the BLPAQ was cross-validated using two criterion measures: the Baecke Questionnaire of Habitual Physical Activity (Baecke, Burema, & Frijters, 1982) and the Godin’s Leisure-Time Exercise Questionnaire (Godin & Shephard, 1985). Multiple linear regressions were performed to predict PPA and UPA from the subscales of the two reference measures. The predictive models differed markedly in terms of gender. Subsequently, the sample of 338 British adults was divided into two subsamples, and these were subjected to a cross-validation using the Limits of Agreement (LoA) methodology advocated by Bland and Altman (1986). The agreement plots revealed that both BLPAQ subscales demonstrated acceptable inter-sample agreement when compared to the criterion measures. In Study 4, a series of structural equation models were tested with the aim of predicting PPA and UPA using the variables that constitute the Theory of Planned Behaviour (TPB). The TPB was able to predict PPA but not UPA. The addition of a direct path between past behaviour to UPA did not result in a significant prediction. Further work is required to examine the factorial structure of the PPA subscale and to increase the number of items in the UPA subscale. In sum, the programme has contributed a valid and reliable theory-based measure of PA as well as evidence to support the utility of the TPB in PA research. However, the TPB framework may require the addition of predictors such as past behaviour and actual behavioural control.

Abstract

In this article we used transgenic zebrafish lines that express compartment-specific isoforms of the roGFP2-Orp1 and Grx1-roGFP2 biosensors, described in Panieri et al (2017) [1], to test the contribute of the pentose phosphate pathway and of the glutathione biosynthesis in the antioxidant capacity of myocardial and endothelial cells in vivo. The transgenic zebrafish embryos were subdued to metabolic inhibition and subsequently challenged with H2O2 or the redox-cycling agent menadione to respectively mimic acute or chronic oxidative stress. Confocal time-lapse recordings were performed to follow the compartmentalized H2O2 and EGSH changes in the cardiovascular tissues of zebrafish embryos at 48 h post fertilization. After sequential excitation at 405 nm and 488 nm the emission was collected between 500–520 nm every 2 min for an overall duration of 60 min. The 405/488 nm ratio was normalized to the initial value obtained before oxidants addition and plotted over time. Abstract

Since its introduction, the zebrafish has provided an important reference system to model and study cardiovascular development as well as lymphangiogenesis in vertebrates. A scientific workshop, held at the 2018 European Zebrafish Principal Investigators Meeting in Trento (Italy) and chaired by Massimo Santoro, focused on the most recent methods and studies on cardiac, vascular and lymphatic development. Daniela Panáková and Natascia Tiso described new molecular mechanisms and signaling pathways involved in cardiac differentiation and disease. Arndt Siekmann and Wiebke Herzog discussed novel roles for Wnt and VEGF signaling in brain angiogenesis. In addition, Brant Weinstein’s lab presented data concerning the discovery of endothelium-derived macrophage-like perivascular cells in the zebrafish brain, while Monica Beltrame’s studies refined the role of Sox transcription factors in vascular and lymphatic development. In this article, we will summarize the details of these recent discoveries in support of the overall value of the zebrafish model system not only to study normal development, but also associated disease states.

Abstract

Vascular mural cells (vMCs) are essential components of the vertebrate vascular system, controlling blood vessel maturation and homeostasis. Discrete molecular mechanisms have been associated with vMC development and differentiation. The function of hemodynamic forces in controlling vMC recruitment is unclear. Using transgenic lines marking developing vMCs in zebrafish embryos, we find that vMCs are recruited by arterial-fated vessels and that the process is flow dependent. We take advantage of tissue-specific CRISPR gene targeting to demonstrate that hemodynamic-dependent Notch activation and the ensuing arterial genetic program is driven by endothelial primary cilia. We also identify zebrafish foxc1b as a cilia-dependent Notch-specific target that is required within endothelial cells to drive vMC recruitment. In summary, we have identified a hemodynamic-dependent mechanism in the developing vasculature that controls vMC recruitment.
