A multidisciplinary approach allowed to investigate the formation and evolution of gypsum blocks within the Riato Conglomerate outcropping in the Catanzaro Basin (Calabria, Southern Italy). Macroscopically, the gypsum blocks seem to belong to the branching facies (primary facies) known in literature, however microscopic observations reveal a detrital origin (secondary facies). Pain and nociception are different phenomena. Nociception is the result of complex activity in sensory pathways. On the other hand, pain is the effect of interactions between nociceptive processes, and cognition, emotions, as well as the social context of the individual. Alterations in the nociceptive route can have different genesis and affect the entire sensorial process. Genetic problems in nociception, clinically characterized by reduced or absent pain sensitivity, compose an important chapter within pain medicine. This chapter encompasses a wide range of very rare diseases. Several genes have been identified. These genes encode the Nav channels 1.7 and 1.9 (SCN9A, and SCN11A genes, respectively), NGFβ and its receptor tyrosine receptor kinase A, as well as the transcription factor PRDM12, and autophagy controllers (TECPR2). Monogenic disorders provoke hereditary sensory and autonomic neuropathies. Their clinical pictures are extremely variable, and a precise classification has yet to be established. Additionally, pain insensitivity is described in diverse numerical and structural chromosomal abnormalities, such as Angelman syndrome, Prader Willy syndrome, Chromosome 15q duplication syndrome, and Chromosome 4 interstitial deletion. Studying these conditions could be a practical strategy to better understand the mechanisms of nociception and investigate potential therapeutic targets against pain.

Purpose: Fulfilling educational needs in pain management should be a lifelong process, even involving physicians board certified in pain medicine such as the anesthesiologists/pain therapists. The aim of the study was to investigate Italian anesthesiologists' self-perceived competency, confidence, and interest to attend educational programs in relation to their seniority in pain management. Methods: SIAARTI members were sent an online questionnaire addressing the following items: education, skills (both soft and hard skills), technical expertise and engaged to participate between December 2020 and January 2021. Participants rated their competence based on the following range (no knowledge, knowledge, competence) while their agreement to attend educational courses was assessed with a 5-point Likert-type scale. Results: Less than one in four participants declare to be dedicated to pain medicine activity with greater proportion among older (over 61 years) compared to younger ones (31-40 years). Regarding cancer and chronic non-cancer pain a positive gradient of self-perceived competence has been observed in relation to seniority. In contrast, no gradient of self-perceived competence was reported about musculoskeletal and low back pain. Participants self-perceived competent in both opioid use and prevention of opioid-related adverse event while feeling less competent when managing drugs with abuse potential. The lowest competence has been observed in pediatric pain along with the lowest interest to attend educational courses. Participants were much and very much interested to education regarding cancer, non-cancer, musculoskeletal and low back pain, invasive analgesic procedures but less regarding items for which they declared less competence, such as use of pain scales, pain management in children and use of drugs with abuse potential. Conclusion: This work provides first evidence of a summative assessment of competency and related educational needs' profile of anesthesiologists/pain therapists thus paving the way for developing a nationwide educational program to improve chronic pain care in Italy.

In the last few years Sr isotope geochemistry has contributed substantially to environmental and food traceability research. This is achievable because soils, plants and waters all have a peculiar Sr isotopic signature (87Sr/86Sr) inherited from the local geological substratum and affected by geological processes as well as the age and initial rubidium concentration of the rocks. Strontium ions released from the bedrock by weathering processes deriving by the interaction of circulating fluids with rocks, enter the environment and accumulates in water and soils. This reservoir of bioavailable Sr may represent a reliable tracer useful to determine the geographical origin of wines as it is known that strontium is taken first by plant roots, then by grapes, and lastly by wine, with no isotope fractionation when compared to the original 87Sr/86Sr ratio in the soil and rocks. As a result, the study of the Sr isotope ratio in the final product (wine) links directly to its geological origin thus representing a specific geofingerprint for any selected wine. Based on these premises this work aims at confirming the strong link between the product (wine) and its territory, with the final purpose to make it recognizable and distinguishable from similar products and protecting it from possible fraud and adulteration. In this work the 87Sr/86Sr systematics has been used to analyze a total of 39 samples (37 soil samples and 2 wine samples) from Campania (Italy). For a better understanding, both total Sr fraction and bioavailable Sr fraction were analyzed in soil samples (rhizospheric soils, bulk soils and samples collected from different horizons) in order to better investigate the environmental processes involved during the wine production cycle.

Cenozoic tectono-magmatism in Iran is widely considered to be related to subduction of the Neo-Tethys Ocean. We employed whole-rock and mineral geochemistry and isotopic data of intrusive rocks from Tafresh, central Urumieh-Dokhtar magmatic arc, to evaluate the role of the mantle in magmatism, to assess the timing of emplacement, and to interpret the tectonic setting. Rock compositions range from gabbro or gabbro-diorite (plagioclase + pyroxene ± olivine), to diorite (plagioclase + amphibole ± pyroxene), to granodiorite (quartz + plagioclase + K-feldspar + amphibole + biotite), exhibiting high-alumina calc-alkaline affinity. Major oxide and trace element variations vary systematically from less to more evolved rocks suggesting a major role for fractional crystallization processes. Zircon LA-ICP-MS U–Pb ages of major rock types are in the range of 24–19 Ma, whereas those of gabbroic dikes are ~17.5 Ma. ԐNd values range between -1.8 and 3.7, and (87Sr/86Sr)i is narrowly restricted to 0.705–0.706, suggesting a common mantle source. The enrichment in light rare earth element (REE) enrichment and flat heavy REE patterns couple depletion of Nb–Ta–Ti indicate that subducting oceanic crust had interacted with the overlying mantle wedge. High-alumina, mid-Mg# Tafresh plutonic rocks formed from hydrous melts from which Ca-pyroxene and magnetite crystallized earlier than plagioclase, whereas late-crystallizing zircon nucleated while magma traversed through lithospheric mantle and Cadomian crust. Modelling of isotope and incompatible-element patterns suggests the contribution of no more than ~5% molten sediment or other crustal components in Tafresh magma, at a developmental stage before most plagioclase and amphibole had crystallized. The Miocene Tafresh plutons originated during the final stages of subduction, before the collision between the Arabian and Eurasian plates.

Pigmented cereal grains with high levels of flavonoid compounds have attracted the attention of nutritional science backing the development of functional foods with claimed health benefits. In this study, we report results on the genetic factors controlling grain pigmentation in durum wheat using a segregant population of recombinant inbred lines (RILs) derived from a cross between an Ethiopian purple grain accession and an Italian amber grain cultivar. The RIL population was genotyped by the wheat 25K SNP array and phenotyped for total anthocyanin content (TAC), grain color, and the L*, a*, and b* color index of wholemeal flour, based on four field trials. The mapping population showed a wide variation for the five traits in the different environments, a significant genotype x environment interaction, and high heritability. A total of 5942 SNP markers were used for constructing the genetic linkage map, with an SNP density ranging from 1.4 to 2.9 markers/cM. Two quantitative trait loci (QTL) were identified for TAC mapping on chromosome arms 2AL and 7BS in the same genomic regions of two detected QTL for purple grain. The interaction between the two QTL was indicative of an inheritance pattern of two loci having complementary effects. Moreover, two QTL for red grain color were detected on chromosome arms 3AL and 3BL. The projection of the four QTL genomic regions on the durum wheat Svevo reference genome disclosed the occurrence of the candidate genes Pp-A3, Pp-B1, R-A1, and R-B1 involved in flavonoid biosynthetic pathways and encoding of transcription factors bHLH (Myc-1) and MYB (Mpc1, Myb10), previously reported in common wheat. The present study provides a set of molecular markers associated with grain pigments useful for the selection of essential alleles for flavonoid synthesis in durum wheat breeding programs and enhancement of the health-promoting quality of derived foods.

The isotope composition of chemical elements is an invaluable investigation tool widely used in Earth Sciences. Rocks, waters and gases acquire specific radiogenic (e.g., 87Sr/86Sr) and stable (e.g., δ18O) isotope ratios due to geological processes such as magmas’ genesis and evolution, past climatic changes, mixing among distinct reservoirs. The power of these tracers bears in the particular isotope signature any geological material acquires due to both its history and specific processes undergone. Isotope approaches have recently been applied to track air, soil and water pollutants, to identify the provenance of archaeological artifacts, and to reconstruct diet and migration paths of past animals and humans. In this framework, isotope tracers can be efficiently coupled to more traditional investigation techniques to solve forensic issues, including linking a suspect to a crime scene, identifying crime victims and the guilty of an environmental crime, tracking the provenance of drugs, identifying explosives. In this chapter, laboratory techniques for the preparation of variable materials aimed at their isotope analysis are illustrated. Examples of application of isotope analysis techniques are described for a variety of forensic problems.

Powdery mildew (PM) is an economically important foliar disease of cultivated cereals worldwide. The cultivation of disease-resistant varieties is considered the most efficient, sustainable and economical strategy for disease management. The objectives of the current study were to fine map the chromosomal region harboring the wild emmer PM resistance locus Pm36 and to identify candidate genes by exploiting the improved tetraploid wheat genomic resources. A set of backcross inbred lines (BILs) of durum wheat were genotyped with the SNP 25K chip array and comparison of the PM-resistant and susceptible lines defined a 1.5 cM region (physical interval of 1.08 Mb) harboring Pm36. The genetic map constructed with F2:3 progenies derived by crossing the PM resistant line 5BIL-42 and the durum parent Latino, restricted to 0.3 cM the genetic distance between Pm36 and the SNP marker IWB22904 (physical distance 0.515 Mb). The distribution of the marker interval including Pm36 in a tetraploid wheat collection indicated that the positive allele was largely present in the domesticated and wild emmer Triticum turgidum spp. dicoccum and ssp. dicoccoides. Ten high-confidence protein coding genes were identified in the Pm36 region of the emmer, durum and bread wheat reference genomes, while three added genes showed no homologous in the emmer genome. The tightly linked markers can be used for marker-assisted selection in wheat breeding programs, and as starting point for the Pm36 map-based cloning.

Since the Late Pleistocene, Campi Flegrei have been inhabited by humans because of the region's soil fertility, mild climatic conditions, and both commercially and strategically advantageous position in the Mediterranean basin. After a period of frequent eruptions that affected the environment and forced indigenous farmers temporarily to abandon the area after each event, in 740 BC Campi Flegrei were chosen by Chalcidian Greeks from Ischia to establish the settlement of Cuma, which was the first Greek colony in what is today the Italian mainland. Since its earliest colonisation, the Phlegraean area, because of its location and environmental characteristics, has been a cause of conflicts over its control, and has also witnessed a significant increase in population and urban development. Even the threat of ongoing bradyseismic activity, which has caused seismicity and ground deformation, and the AD 1538 Monte Nuovo eruption did not discourage people from living in the area. Campi Flegrei have undergone socio-economic development which for most of its history has been sustainable. Only with industrialisation over recent decades, has it become unsustainable. This unsustainable development has also caused an increase in the vulnerability of the area in terms of its volcanic risk. Knowledge of the history of urbanisation in Campi Flegrei may, therefore, assist the civil authorities in effecting socio-economic policies aimed at disaster risk reduction.

A review of the recent geochemical investigations carried out on products of the Campi Flegrei volcanic field is presented, with particular attention on radiogenic (Sr, Nd, Pb) and stable (B, O) isotope data. The studied volcanic rocks span the entire period of best known Campi Flegrei volcanic field activity, since ~58 ka to the last event, the AD 1538 Monte Nuovo eruption. The isotope data, combined with mineralogical, geochemical, geochronological and volcanological data have allowed reconstructing a complex behaviour of the Campi Flegrei volcanic field magmatic feeding system. They suggest that several, isotopically-distinct magmatic components fed the activity over various time periods. In particular, over the past 15 kyrs at least three isotopically-distinct magmatic components, located either in a deep and large reservoir or in shallower and smaller chambers, have variably interacted, eventually feeding the volcanic activity. These magmatic components originated through a complex interplay of magma supply from a variably-enriched mantle source, and crustal contamination processes occurring at depth. Mingling/mixing among these isotopically-distinct magmatic components, as well as closed-system differentiation of variable hybrid magmas and interaction with cumulate mushes, were the dominant processes acting in the Campi Flegrei caldera feeding system over the past 15 kyrs. Three examples including the Agnano-Monte Spina, Astroni and Averno 2 eruptions of this period are described to illustrate the interplay among the above-mentioned different processes.

Considerable debate persists regarding the petrogenesis of high-alumina basalts (HAB) which are purported to occur exclusively in subduction zones. Major and trace element, mineral chemistry, whole-rock Sr-Nd-Hf-isotopes and zircon U-Pb age data are reported for the Cenozoic Eshtehard HABs, in order to constrain the nature of mantle beneath the central Urumieh-Dokhtar magmatic arc, and further investigate the mechanism of HABs generation. Eshtehard HABs, chemically akin to those from continental arcs, include basaltic, basaltic andesite, andesitic and dacitic rocks. U-Pb geochronology of zircon yield ages of 47.2 ± 0.6 Ma, 43.9 ± 0.3, and 40.9 ± 0.5 to 39.4 ± 0.9 for basaltic, andesitic and dacitic samples, respectively. Basaltic andesitic dikes intruded into dacitic hosts yield age of ca. 20–18 Ma. eHf(t) values for Eocene zircons display a range variable from −6.4 to +6.5. Miocene zircons have higher eHf(t), ranging between −1.8 and +10.7. The studied rocks are characterized by enrichment in incompatible trace elements and have relatively homogeneous Sr-Nd isotopes. Integrated studies indicate that Eshtehard HABs were derived from the hydrated, dominantly depleted shallow asthenospheric mantle wedge (and possibly also in the lower lithosphere) overlying subducted oceanic lithosphere. Traversing lithospheric mantle and Cadomian crust, and assimilating crustal material while fractionating plagioclase, high-Ca pyroxene, magnetite, and amphibole, the primary melt formed Eshtehard high-alumina, low-Mg# basalts. We hypothesize that ponding of hydrous magma at the base of the crust allowed for further crustal assimilation and fractionation of Ca-pyroxene and magnetite without plagioclase and zircon nucleation; then, ascending magmas through the crust led to crystallization of plagioclase and eventually zircon. We suggest comparatively high water content (rather than high crystallization pressure), up to 4% sediment melt and less than 10% of continental crust materials were involved, as subordinate components, in the petrogenesis of Eshtehard HABs.

Thrombosis of small and large vessels is reported as a key player in COVID-19 severity. However, host genetic determinants of this susceptibility are still unclear. Congenital Thrombotic Thrombocytopenic Purpura is a severe autosomal recessive disorder characterized by uncleaved ultra-large vWF and thrombotic microangiopathy, frequently triggered by infections. Carriers are reported to be asymptomatic. Exome analysis of about 3000 SARS-CoV-2 infected subjects of different severities, belonging to the GEN-COVID cohort, revealed the specific role of vWF cleaving enzyme ADAMTS13 (A disintegrin-like and metalloprotease with thrombospondin type 1 motif, 13). We report here that ultra-rare variants in a heterozygous state lead to a rare form of COVID-19 characterized by hyper-inflammation signs, which segregates in families as an autosomal dominant disorder conditioned by SARS-CoV-2 infection, sex, and age. This has clinical relevance due to the availability of drugs such as Caplacizumab, which inhibits vWF–platelet interaction, and Crizanlizumab, which, by inhibiting P-selectin binding to its ligands, prevents leukocyte recruitment and platelet aggregation at the site of vascular damage.

We employed a multifaceted computational strategy to identify the genetic factors contributing to increased risk of severe COVID-19 infection from a Whole Exome Sequencing (WES) dataset of a cohort of 2000 Italian patients. We coupled a stratified k-fold screening, to rank variants more associated with severity, with training of multiple supervised classifiers, to predict severity on the basis of screened features. Feature importance analysis from tree-based models allowed to identify a handful of 16 variants with highest support which, together with age and gender covariates, were found to be most predictive of COVID-19 severity. When tested on a follow-up cohort, our ensemble of models predicted severity with good accuracy (ACC=81.88%; ROC_AUC=96%; MCC=61.55%). Principal Component Analysis (PCA) and clustering of patients on important variants orthogonally identified two groups of individuals with a higher fraction of severe cases. Our model recapitulated a vast literature of emerging molecular mechanisms and genetic factors linked to COVID-19 response and extends previous landmark Genome Wide Association Studies (GWAS). It revealed a network of interplaying genetic signatures converging on established immune system and inflammatory processes linked to viral infection response, such as JAK-STAT, Cytokine, Interleukin, and C-type lectin receptor signaling. It also identified additional processes cross-talking with immune pathways, such as GPCR signalling, which might offer additional opportunities for therapeutic intervention and patient stratification. Publicly available PheWAS datasets revealed that several variants were significantly associated with phenotypic traits such as “Respiratory or thoracic disease”, confirming their link with COVID-19 severity outcome. Taken together, our analysis suggests that curated genetic information can be effectively integrated along with other patient clinical covariates to forecast COVID-19 disease severity and dissect the underlying host genetic mechanisms for personalized medicine treatments.

The polymorphism L412F in TLR3 has been associated with several infectious diseases. However, the mechanism underlying this association is still unexplored. Here, we show that the L412F polymorphism in TLR3 is a marker of severity in COVID-19. This association increases in the sub-cohort of males. Impaired macroautophagy/autophagy and reduced TNF/TNF�� production was demonstrated in HEK293 cells transfected with TLR3L412F-encoding plasmid and stimulated with specific agonist poly(I:C). A statistically significant reduced survival at 28 days was shown in L412F COVID-19 patients treated with the autophagy-inhibitor hydroxychloroquine (p = 0.038). An increased frequency of autoimmune disorders such as co-morbidity was found in L412F COVID-19 males with specific class II HLA haplotypes prone to autoantigen presentation. Our analyses indicate that L412F polymorphism makes males at risk of severe COVID-19 and provides a rationale for reinterpreting clinical trials considering autophagy pathways. The sands of the Ofanto River (Southern Italy) include a volcaniclastic component represented by clinopyroxene, melanite garnet, amphibole and subordinate volcanic lithics. In order to infer the provenance of this component, we have conducted a microanalytical and isotopic study on the volcanic minerals of the sands collected in three different sites along the Ofanto River: (1) upstream sector, where only sedimentary rocks are drained; (2) middle course, along a tributary draining only the Monte Vulture volcanic rocks, (3) the mouth of the Ofanto. Moreover, minerals of the beach sands of the Tyrrhenian Cilento coast were analyzed for comparison. The microanalytical study and the Sr-Nd isotopic composition reveal the existence of two populations of pyroxenes characterized by distinct isotopic signatures and indicating a provenance from two different volcanic sources. One is confidently identifiable with Monte Vulture, and is evident in the minerals of the middle course and mouth sands. The pyroxenes of the upstream sector, isotopically similar to those of the Cilento coast, have a Campanian signature. The simultaneous occurrence of amphibole and garnet point to the explosive eruptions of Mount Vesuvius as the most probable sources of these minerals. The absence of Campanian pyroxenes at the mouth of the Ofanto River could be related to the occurrence of dams limiting the transport of solids along the river.
