9. Mutational analysis of flagellum-independent surface spreading of Serratia marcescens 274 on a low-agar medium

Abstract

Examination of swarming-defective mutants of Serratia marcescens 274 reveals that these mutants are defective in the generation of serrawettin W1, an extracellular cyclic lipopeptide. The serrawettin mutants with Fla defects exhibit spreading deficiency on low-agar media. Extracellular supply of serrawettin helps overcome the spreading deficiency. Surface translocation is not affected by the introduction of Fla defects into chemotaxis mutants, revealing spreading as a passive form of translocation.

Abstract

A simple one-step technique facilitates the simultaneous identification of three virulent-related genes, plcB, hlyA and prfA by PCR in milk samples that are artificially contaminated. Using phosphate buffered saline to wash the contaminated milk and concentrating Listeria monocytogenes to 1/10 of the original volume can eliminate the inhibitory influence of milk in the PCR identification of the virulent bacteria.

Abstract

The mechanism behind fractal morphogenesis and the conditions necessary for its generation were examined in gram-negative rods cultured in solid agar plates. Results showthat colony size and nutrient diffusion greatly influenced bacterial growth. Biosurfactants and high glucose concentrations were also critical for fractal morphogenesis. These findings are common among gram-negative rods and are consistent with those predicted by the diffusion-limited aggregation model.

Abstract

The chemical structure and function of serrawettin W2 of Serratia marcescens NS 25 was elucidated by degradation and spectroscopic analyses. The results indicate that serrawettin W2 is a cyclodepsipeptide with a proposed structure of 3-hydroxyldecanoyl-D-leucyl-L-seryl-L-threonyl-D-phenylalanyl-L-isoleucyl lactone. Serrawettin W2 is a nonionic surfactant. Studies involving serrawettin-producing and -nonproducing bacteria showed that serrawettin is involved in enhancing bacterial spreading on a surface environment.

The smallest pore size through which Listeria monocytogenes could pass, the time required for the pass-through, and other influencing factors are examined. Flagellar motility and expansive pressure generated by the growing bacterial population are indicated as the driving forces of infiltration.
