Abstract

Goldfish or Kinguio is a widely marketed species worldwide due to the ornamental market. There is some lack of acknowledgment of the production of the species under specific climatic conditions. To evaluate the effect of temperature on embryonic development and the first exogenous feeding of goldfish, an experiment was proposed. Fifteen incubators, organized in five treatments (18, 22, 26, 30, and 34 °C) with three replications each, were used to keep the fertilized goldfish eggs until the first exogenous feeding of the larvae. The main development events were observed to understand the possible effects of these temperatures on embryos and larvae of the species. Temperature influences embryo development and the time of first exogenous feeding of goldfish. The temperature of 34 °C was lethal to the species causing 100% of anomalies in the embryos and larvae. The experiment data allow us to conclude that the species presents a maximum thermal limit during embryogenesis, and these data are important to the aquaculture industry and to understand the effect of climate changes on goldfish. The data obtained in this experiment will assist in the management of invasive species and production of the species (aquaculture). Resumo O peixe dourado ou Kinguio é uma espécie amplamente comercializada em todo o mundo devido ao mercado ornamental. Existe alguma falta de conhecimento da produção da espécie em condições climáticas específicas. Para avaliar o efeito da temperatura no desenvolvimento embrionário e na primeira alimentação exógena do Kinguio, um experimento foi proposto. Quinze incubadoras, organizadas em cinco tratamentos (18, 22, 26, 30 e 34 °C) com três repetições cada, foram utilizadas para manter os ovos fertilizados de Kinguio até a primeira alimentação exógena das larvas. Os principais eventos do desenvolvimento foram observados para entender os possíveis efeitos dessas temperaturas em embriões e larvas da espécie. A temperatura influencia o desenvolvimento do embrião e o tempo da primeira alimentação exógena do Kinguio. A temperatura de 34°C foi letal para a espécie causando 100% de anomalias nos embriões e larvas. Os dados do experimento permitem concluir que a espécie apresenta um limite térmico máximo durante a embriogênese, sendo esses dados importantes para a indústria da aquicultura e para entender o efeito das mudanças climáticas no Kinguio. Os dados obtidos neste experimento auxiliarão no manejo de espécies invasoras e na produção da espécie (aquicultura).

21. Influence of garlic extract on larval performance and survival of juvenile angelfish Pterophyllum scalare during transport

Abstract

The objective of this study was to evaluate the properties of garlic as a growth promoter in larvae of angelfish Pterophyllum scalare and its benefits during transport of juveniles of this species. The experiment was conducted in recirculation aquaculture system, consisting of 20 tanks of 40 L equipped with independent water input and output. We used 1,400 larvae distributed among five treatments, with four repetitions, which totaled 20 experimental units. The treatments were 0, 50, 100, 200, and 400 mg of garlic extract per kilo of feed. The results showed that the inclusion of levels of garlic extract in the feed did not significantly effect the fish’s development or their transport. Neither did the inclusion of levels of garlic extract affect the survival of the larvae during the trial period. New research with extracts of higher dosages should be performed to elucidate the effect of garlic extract as a growth promoter. Resumo O estudo teve como objetivo avaliar as propriedades do alho como promotor de crescimento em larvas de acará bandeira Pterophyllum scalare e seus benefícios no transporte de juvenis da mesma espécie. O experimento foi conduzido em sistema de recirculação composto por 20 aquários de 40 L de volume útil, dotado de entrada e saída de água independentes e teve a duração de 40 dias. O delineamento experimental adotado foi o inteiramente casualizado (DIC), com cinco tratamentos e quatro repetições, totalizando 20 unidades experimentais. Foram utilizadas 1.400 larvas de acará bandeira (Pterophyllum scalare) divididas entre os tratamentos. Os tratamentos consistiram em: 0, 50, 100, 200 e 400 mg de extrato de alho por quilo de ração. Os resultados mostraram que não houve efeito significativo dos níveis de inclusão do extrato do alho sobre os índices zootécnicos avaliados e posteriormente no transporte dos juvenis. Também não foi observado influência na sobrevivência das larvas durante o período experimental. Novas pesquisas com dosagens maiores de extratos devem ser realizados para melhor elucidação do efeito do extrato de alho como promotor de crescimento.

27. Variance and covariance components for weaning weight for Herefords in three countries

Abstract

Records from the Hereford Associations of the United States (USA), Canada, and Uruguay were used to estimate genetic and phenotypic variances and covariances for weaning weight. Estimation was done using a complete animal model, relatively large data sets, and the same methodology for the three countries in order to determine whether genetic parameters for weaning weight were homogeneous across environments. Data were composed of 2,322,722,487,661, and 102,986 edited weaning weight records for USA, Canada, and Uruguay, respectively. Ten samples were obtained from each country by eliminating data from small herds with fewer than 500 records, selecting herds at random from the entire data set after removing the small herds, and then retaining the direct-sire-connected contemporary groups within each sample. The final sample sizes ranged from 9,832 to 46,377 records. An accelerated EM-REML algorithm was used in estimating the (co)variance components in each sample. The estimates were pooled by calculating the arithmetic mean of the 10 samples from within each country. Direct and maternal (in parentheses) heritability estimates were .24 (.16), .20 (.16), and .23 (.18) for USA, Canada, and Uruguay, respectively. Maternal heritabilities reported here are nearly 50% smaller than the values currently used in national genetic evaluation for the breed, which were estimated using sire-maternal grandsire models. Covariance between direct and maternal was negative in all countries, accounting for 6, 8, and 10% of the total phenotypic variation, and the total dam effect was 32.5, 37.0, and 34.0% in USA, Canada, and Uruguay, respectively. Total heritabilities were similar among the countries, with values of .19, .19, and .17 for the three respective countries. The similarity of genetic and environmental parameters across the three countries suggests that joint genetic evaluation is feasible across environments provided that the genotype x environment interaction is negligible and can be ignored. 35. Investigation of genotype x environment interactions for weaning weight for Herefords in three countries

Abstract

The objective of this study was to investigate the possibility of genotype x environment interactions for weaning weight (WWT) between different regions of the United States (US) and between Canada (CA), Uruguay (UY), and US for populations of Hereford cattle. Original data were composed of 487,661, 102,986, and 2,322,722 edited weaning weight records from CA, UY, and US, respectively. A total of 359 sires were identified as having progeny across all three countries; 240 of them had at least one progeny with a record in each environment. The data sets within each country were reduced by retaining records from herds with more than 500 WWT records, with an average contemporary group size of greater than nine animals, and that contained WWT records from progeny or maternal grand-progeny of the across-country sires. Data sets within each country were further reduced by randomly selecting among remaining herds. Four regions within US were defined: Upper Plains (UP), Cornbelt (CB), South (S), and Gulf Coast (GC). Similar sampling criteria and common international sires were used to form the within-US regional data sets. A pairwise analysis was done between countries and regions within US (UP-CB vs S-GC, UP vs CB, and S vs GC) for the estimation of (co)variance components and genetic correlation between environments. An accelerated EM-REML algorithm and a multiple-trait animal model that considered WWT as a different trait in each environment were used to estimate parameters in each pairwise analysis. Direct and maternal (in parentheses) estimated genetic correlations for CA vs UY, CA vs US, US vs UY, UP-CB vs S-GC, UP vs CB, and S vs GC were .88 (.84), .86 (.82), .90 (.85), .88 (.87), .88 (.84), and .87 (.85), respectively. The general absence of genotype x country interactions observed in this study, together with a prior study that showed the similarity of genetic and environmental parameters across the three countries, strongly indicates that a joint WWT genetic evaluation for Hereford cattle could be conducted using a model that treated the information from CA, UY, and US as a single population using single population-wide genetic parameters. Abstract

El objetivo del Proyecto de Investigación y Desarrollo del Merino Fino para Uruguay (INIA, SUL, SCHU) es desarrollar una alternativa de producción ovina que por medio de su difusión y posterior adopción, permita mejorar la sustentabilidad socioeconómica de los productores de lana de las regiones de Basalto y Cristalino, considerando las demandas actuales y futuras de la Cadena Agroindustrial de lana del País y de los mercados consumidores. Es así que se ha establecido un Núcleo de Selección Merino Fino (a partir de hembras nacionales y la importación de animales del exterior), el cuál ha logrado producir animales con diámetros de fibra sustancialmente menores al de la población nacional, con un progreso genético anual para dicha característica de -0,21ì logrando mantener el Peso de Vellón Limpio. El material genético producido en dicho núcleo ha sido diseminado hacia la cabaña nacional y hacia los productores comerciales. Adicionalmente el proyecto ha permitido establecer una evaluación genética nacional para Merino Fino y ha logrado que las Diferencias Esperadas en la Progenies para las características de interés económico sean utilizadas activamente como criterio de selección de los animales reproductores. Se dispone de información sobre alimentación, manejo sanitario y reproductivo y sus efectos sobre la reproducción, producción y calidad de lana, que demuestra la factibilidad de incorporar la producción de lanas finas y superfinas en sistemas productivos extensivos del país. // SUMMARY: The aim of this research and development Project of fine merino for Uruguay (INIA, SUL, SCHU) is todevelop a wool production alternative that through its extension and subsequent adoption will increase thesocioeconomic sustainability of Basalto and Cristalino wool producers of Uruguay, considering present andfuture demands of the Uruguayan wool Agro-industrial chain and of the consumer markets. Thus, a FineMerino selection nucleus has been established (originated by national females plus imported animals) whichhas managed to produce animals with substantial less fiber diameter than the national population, with a annualgenetic progress of -0,21ì maintaining the clean fleece weight. The genetic material generated has beendisseminated at the level of breeders and of commercial producers. The project has also allowed for a nationalgenetic evaluation for Fine Merino to be established and that the EPDs for the economically relevant traits beused as a selection criteria. Information is now available regarding feeding, sanitary and reproductive managementand its effects on reproduction, production and wool quality that shows the feasibility of incorporating fineand superfine wool production to the extensive production systems of Uruguay

37. Impact of Trypanosoma cruzi on antimicrobial peptide gene expression and activity in the fat body and midgut of Rhodnius prolixus.

Abstract

Background: Rhodnius prolixus is a major vector of Trypanosoma cruzi, the causative agent of Chagas disease in Latin America. In natural habitats, these insects are in contact with a variety of bacteria, fungi, virus and parasites that they acquire from both their environments and the blood of their hosts. Microorganism ingestion may trigger the synthesis of humoral immune factors, including antimicrobial peptides (AMPs). The objective of this study was to compare the expression levels of AMPs (defensins and prolixicin) in the different midgut compartments and the fat body of R. prolixus infected with different T. cruzi strains. The T. cruzi Dm 28c clone (TcI) successfully develops whereas Y strain (TcII) does not complete its life- cycle in R. prolixus. The relative AMP gene expressions were evaluated in the insect midgut and fat body infected on different days with the T. cruzi Dm 28c clone and the Y strain. The influence of the antibacterial activity on the intestinal microbiota was taken into account. Methods: The presence of T. cruzi in the midgut of R. prolixus was analysed by optical microscope. The relative expression of the antimicrobial peptides encoding genes defensin (defA, defB, defC) and prolixicin (prol) was quantified by RT-qPCR. The antimicrobial activity of the AMPs against Staphylococcus aureus, Escherichia coli and Serratia marcescens were evaluated in vitro using turbidimetric tests with haemolymph, anterior and posterior midgut samples. Midgut bacteria were quantified using colony forming unit (CFU) assays and real time quantitative polymerase chain reaction (RT-qPCR). Results: Our results showed that the infection of R. prolixus by the two different T. cruzi strains exhibited different temporal AMP induction profiles in the anterior and posterior midgut. Insects infected with T. cruzi Dm 28c exhibited an increase in defC and prol transcripts and a simultaneous reduction in the midgut cultivable bacteria population, Serratia marcescens and Rhodococcus rhodnii. In contrast, the T. cruzi Y strain neither induced AMP gene expression in the gut nor reduced the number of colony formation units in the anterior midgut. Beside the induction of a local immune response in the midgut after feeding R. prolixus with T. cruzi, a simultaneous systemic response was also detected in the fat body. Conclusions: R. prolixus AMP gene expressions and the cultivable midgut bacterial microbiota were modulated in distinct patterns, which depend on the T. cruzi genotype used for infection.
