Key message A total of 45 aquaporins was identified in Fagus sylvatica, 35 of which were differentially expressed in spring and summer in the leaves, phloem and xylem of 11-year-old trees. Abstract European beech (Fagus sylvatica) has been widely studied in terms of its water relations and local adaptation. However, to date, the underlying basis conferring adaptation to differences in water availability are unknown. Therefore, we examined the expression of aquaporins in trees of four different beech provenances representing the southern and northern range margins, as well as core populations, grown in a common garden. We sampled their xylem, phloem and leaf tissue, when leaves had fully expanded, and in late summer. A total of 45 aquaporin isoforms were identified in the beech genome, of which 35 were detected across all sampled tissues. In our phylogenetic analysis, beech aquaporins clustered into the five subfamilies found in other woody species. Members of the plasma membrane intrinsic protein subfamily generally displayed the highest levels of expression, followed by tonoplast intrinsic proteins. Isoforms of the remaining subfamilies, Noduline-26-like intrinsic proteins, small basic intrinsic proteins and uncharacterised intrinsic proteins, were expressed at very low to moderate levels. The expression of most isoforms was stable or declined from spring to summer. Leaves followed a different expression profile from that of vascular tissues, whereas both phloem and xylem were found to express the same FsMIPs. Tissue-specific aquaporin expression was very similar amongst the four beech provenances, indicating that there is no inherent difference in the capability of these provenances to regulate aquaporin activity. The general decrease in FsMIP expression toward the end of the growing period indicates that aquaporins are involved in tree water relations and growth.

Sunflecks are transient patches of direct radiation that provide a substantial proportion of the daily irradiance to leaves in the lower canopy. In this position, faster photosynthetic induction would allow for higher sunfleck-use efficiency, as is commonly reported in the literature. Yet, when sunflecks are too few and far between, it may be more beneficial for shade leaves to prioritize efficient photosynthesis under shade. We investigated the temporal dynamics of photosynthetic induction, recovery under shade, and stomatal movement during a sunfleck, in sun and shade leaves of Fagus sylvatica from three provenances of contrasting origin. We found that shade leaves complete full induction in a shorter time than sun leaves, but that sun leaves respond faster than shade leaves due to their much larger amplitude of induction. The core-range provenance achieved faster stomatal opening in shade leaves, which may allow for better sunfleck-use efficiency in denser canopies and lower canopy positions. Our findings represent a paradigm shift for future research into light fluctuations in canopies, drawing attention to the ubiquitous importance of sunflecks for photosynthesis, not only in lower-canopy leaves where shade is prevalent, but particularly in the upper canopy where longer sunflecks are more common due to canopy openness.

Leaf fall in the autumn opens the forest canopy, allowing more solar radiation to be transmitted to the forest floor. Those understorey species that remain physiologically active may benefit from the sunlight received by assimilating additional carbon while conditions remain favourable. We monitored water and leaf pigment content, as well as photosynthetic capacity in six understorey species growing in three adjacent stands differing in their canopy tree species. Leaf fall, transmitted light and microclimate were followed in each stand. We found that leaf fall started earlier in the Betula pendula, than in the Quercus robur stand, and light transmission changed accordingly. Concurrently, understorey leaf senescence was generally earlier in the birch than in the oak stand, itself earlier than in the evergreen Picea abies stand. Neither atmospheric CO , humidity, nor temperature differed between stands. A change in light quality and/or increase in quantity following leaf fall drove the difference in the timing of senescence in the understorey. Species with later senescence were more able to use the increased light after leaf fall. Together these findings help provides a mechanistic foundation to predict how ecosystem functioning and carbon balance will be impacted by phenological shifts in response to global changes.

The main tumor mass and its infiltrative area were analyzed. This work focused on the invading cancer cells after irradiation and their viability, aggressiveness, and recurrence potential were assessed using mitotic catastrophe quantification, MMP secretion analysis and neurosphere assays, respectively.Results: In vitro clonogenic assays showed that Au@DTDTPA(Gd) nanoparticles exerted a radiosensitizing effect on U87 cells, and in vivo experiments suggested a benefit of the combined treatment “RT 2× 5 Gy + Au@DTDTPA(Gd)” compared to RT alone. Invasion assays revealed that invasion distance tended to increase after irradiation alone, while the combined treatments were able to significantly reduce tumor invasion. Monitoring of U87-GFP tumor progression using organotypic cultures or intracerebral grafts confirmed the anti-invasive effect of Au@DTDTPA(Gd) on irradiated spheroids. Most importantly, the combination of Au@DTDTPA(Gd) with irradiation drastically reduced the number, the viability and the aggressiveness of tumor cells able to escape from U87 spheroids. Notably, the combined treatments significantly reduced the proportion of escaped cells with stem-like features that could cause recurrence.Conclusion: Combining Au@DTDTPA(Gd) nanoparticles and X-ray radiotherapy appears as an attractive therapeutic strategy to decrease number, viability and aggressiveness of tumor cells that escape and can invade the surrounding brain parenchyma. The sunlight received by plants is affected by cloudiness and pollution. Future changes in cloud cover will differ among regions, while aerosol concentrations are expected to continue increasing globally as a result of wildfires, fossil fuel combustion, and industrial pollution. Clouds and aerosols increase the diffuse fraction and modify the spectral composition of incident solar radiation, and both will affect photosynthesis and terrestrial ecosystem productivity. Thus, an assessment of how canopy and leaf-level processes respond to these changes is needed as part of accurately forecasting future global carbon assimilation. To review these processes and their implications: first, we discuss the physical basis of the effect of clouds and aerosols on solar radiation as it penetrates the atmosphere; second, we consider how direct and diffuse radiation are absorbed and transmitted by plant canopies and their leaves; and finally, we assess the consequences for photosynthesis at the canopy and ecosystem levels. Photobiology will be affected at the atmospheric level by a shift in spectral composition toward shorter or longer wavelengths under clouds or aerosols, respectively, due to different scattering. Changes in the microclimate and spectral composition of radiation due to an enhanced diffuse fraction also depend on the acclimation of canopy architectural and physiological traits, such as leaf area index, orientation, and clumping. Together with an enhancement of light-use efficiency, this makes the effect of diffuse solar radiation on canopy photosynthesis a multilayered phenomenon, requiring experimental testing to capture those complex interactions that will determine whether it produces the persistent enhancement in carbon assimilation that land-surface models currently predict.

Under field conditions, plants are subject to wind-induced movement which creates fluctuations of light intensity and spectral quality reaching the leaves, defined here as windflecks. Within this study, irradiance within two contrasting wheat (Triticum aestivum) canopies during full sun conditions was measured using a spectroradiometer to determine the frequency, duration and magnitude of low- to high-light events plus the spectral composition during wind-induced movement. Similarly, a static canopy was modelled using three-dimensional reconstruction and ray tracing to determine fleck characteristics without the presence of wind. Corresponding architectural traits were measured manually and in silico including plant height, leaf area and angle plus biomechanical properties. Light intensity can differ up to 40% during a windfleck, with changes occurring on a sub-second scale compared to similar to 5 min in canopies not subject to wind. Features such as a shorter height, more erect leaf stature and having an open structure led to an increased frequency and reduced time interval of light flecks in the CMH79A canopy compared to Paragon. This finding illustrates the potential for architectural traits to be selected to improve the canopy light environment and provides the foundation to further explore the links between plant form and function in crop canopies.

The plant area index (PAI) is a structural trait that succinctly parametrizes the foliage distribution of a canopy and is usually estimated using indirect optical techniques such as digital hemispherical photography. Critically, on-the-ground photographic measurements forgo the vertical variation of canopy structure which regulates the local light environment. Hence new approaches are sought for vertical sampling of traits. We present an uncrewed aircraft system (UAS) spherical photographic method to obtain structural traits throughout the depth of tree canopies. Our method explained 89% of the variation in PAI when compared with ground-based hemispherical photography. When comparing UAS vertical trait profiles with airborne laser scanning data, we found highest agreement in an open birch (Betula pendula/pubescens) canopy. Minor disagreement was found in dense spruce (Picea abies) stands, especially in the lower canopy. Our new method enables easy estimation of the vertical dimension of canopy structural traits in previously inaccessible spaces. The method is affordable and safe and therefore readily usable by plant scientists.

International audience; Through the variations in their dimension, density, anatomy or isotopes composition, tree rings have provided invaluable proxies to evaluate past changes in the environment. Whereas long-term records of changes in soil fertility are particularly desired for forest ecosystem studies, the use of the chemical composition of tree rings as potential marker is still controversial. Dendrochemistry has sometimes been considered as a promising approach to study past changes in soil chemistry, whereas some authors stated that element translocations in the wood preclude any possibility of reliable retrospective monitoring. Here, we aimed at testing whether the wood elemental content of fertilized oaks (Quercus petraea) differed from control trees >30 years after a NPKCaMg fertilization and, if so, if the date of fertilization could be retrieved from the ring analysis. The contents in N, Mg, P, K, Ca and Mn were measured for each of the 43 sampled trees and in every ring of the 58-year long chronology with a non-destructive method coupling a Wavelength Dispersive Spectroscope (WDS) with a Scanning Electron Microscope (SEM).The results showed significantly higher contents in Ca and lower contents in Mn in fertilized compared to control trees. However, there was no difference in elemental content between the rings of the fertilized trees built in the 20 years before and those built after fertilization. Thus, whereas the effect of fertilization on increasing ring width was dramatic, immediate and relatively short-lasting, the elemental composition of the entire ring sequence was impacted, precluding the dating of the event. These results question the possibility to reconstruct long-term changes in soil fertility based on dendrochemistry.

Light in canopies is highly dynamic since the strength and composition of incoming radiation is determined by the wind and the Sun's trajectory and by canopy structure. For this highly dynamic environment, we mathematically defined sunflecks as periods of high irradiance relative to the background light environment. They can account for a large proportion of the light available for photosynthesis. Based on high-frequency irradiance measurements with a CCD array spectroradiometer, we investigated how the frequency of measurement affects what we define as sunflecks. Do different plant canopies produce sunflecks with different properties? How does the spectral composition and strength of irradiance in the shade vary during a sunfleck? Our results suggest that high-frequency measurements improved our description of light fluctuations and led to the detection of shorter, more frequent and intense sunflecks. We found that shorter wind-induced sunflecks contribute most of the irradiance attributable to sunflecks, contrary to previous reports from both forests and crops. Large variations in sunfleck properties related to canopy depth and species, including distinct spectral composition under shade and sunflecks, suggest that mapping canopy structural traits may help us model photosynthesis dynamically.

• Water use efficiency (WUE), oftentimes estimated as transpiration efficiency (TE): the amount of biomass produced with regard to the water used, has not yet been used as a breeding trait to select poplar genotypes with simultaneously high productivity and conservation of water. Before its application as a selection target, evidence must be presented showing that WUE or its estimators remain constant with age and across environmental conditions. • We conducted a rainfall exclusion experiment in the field on two Populus euramericana (Moench.) and two Populus nigra (L.) genotypes, and assessed leaf-level (A/g s) and whole-plant WUE (DMT/WU as well as their components and related traits. Then, we aimed to compare these results with the same poplar genotypes grown in a glasshouse under contrasting water availability. • Despite a reduction of soil water content and whole-plant transpiration, growth was stimulated in the rainfall exclusion plot, likely as a result of an increased nitrogen assimilation. However, TE values between the glasshouse and the field were similar, and genotype ranking remained fairly constant for transpiration, carbon isotopic discrimination (∆, as a proxy for Wi) and TE. Moreover, even though the drivers of WUE in both experiments were different, increases of WUE measured as ∆ or TE was not associated with lower biomass production. Relatively good agreement was found between ∆ and TE in the field, absence of a similar correlation in the glasshouse is discussed. • These results suggest that ∆ may be a good proxy for TE, and could be used, both as a breeding target for genotype selection in glasshouses without impacting biomass production when planted in the field. However, reduced water availability modified the genotype ranking more significantly than between the field/glasshouse experiments, suggesting a diversity of poplar response to drought that should be considered in breeding strategies. Recent research has shown that plant acclimation to diverse patterns of light intensity modifies the dynamics of their stomatal response. Therefore, whether plants are grown in controlled conditions or in the field may impact their stomatal dynamics. We analysed the stomatal dynamics of two Populus euramericana and two Populus nigra genotypes grown in the field under contrasting water availability. By comparing their stomatal dynamics with that of the same genotypes grown in a glasshouse, we were able to test whether differences between these growing conditions interacted with genotypic differences in affecting stomatal dynamics and responses to soil water deficit. We found that, despite higher stomatal density and smaller size, in the field stomatal dynamics were much slower than in the glasshouse. Overall, differences among genotypes and their response to soil water deficit were much less pronounced in the field compared with the glasshouse. These results indicate that stomatal dynamics are regulated by both genotype-specific and environmental factors. Moreover, having slower stomata may be advantageous under some conditions. While stomatal dynamics were linked with whole-plant transpiration per leaf area in both experiments, the contribution of stomatal morphology varies dependent on the environmental conditions.

Poplar plantations, widely used for the production of woody biomass, might be at high risk from the climate change-induced increase in the frequency of drought periods. Therefore, selecting improved genotypes, which are highly productive but with a high water use efficiency (WUE), is becoming a major target. The use of automated weighing systems in controlled environments facilitates the estimation of cumulated water loss and whole plant transpiration efficiency (TE). Differences in TE and leaf level intrinsic WUE as well as the contribution of underlying ecophysiological traits were determined in three contrasting P. nigra genotypes. Strong differences in TE among the selected genotypes were congruent with differences in leaf level intrinsic WUE. Our data show that a high total leaf area was overcompensated by a low per leaf area transpiration rate, leading to higher TE in highly productive genotypes originating from cool locations. Nocturnal water loss was relatively low but contributed to variations in TE among genotypes. In response to drought, leaf level WUE increased but not TE, suggesting that carbon losses due to whole plant respiration could offset the drought-induced increase in intrinsic WUE.

Element content and expression of genes of interest on single cell types, such as stomata, provide valuable insights into their specific physiology, improving our understanding of leaf gas exchange regulation. We investigated how far differences in stomatal conductance (g(s)) can be ascribed to changes in guard cells functioning in amphistomateous leaves. g(s) was measured during the day on both leaf sides, on well-watered and drought-stressed trees (two Populus euramericana Moench and two Populus nigra L. genotypes). In parallel, guard cells were dissected for element content and gene expressions analyses. Both were strongly arranged according to genotype, and drought had the lowest impact overall. Normalizing the data by genotype highlighted a structure on the basis of leaf sides and time of day both for element content and gene expression. Guard cells magnesium, phosphorus, and chlorine were the most abundant on the abaxial side in the morning, where g(s) was at the highest. In contrast, genes encoding H+-ATPase and aquaporins were usually more abundant in the afternoon, whereas genes encoding Ca2+-vacuolar antiporters, K+ channels, and ABA-related genes were in general more abundant on the adaxial side. Our work highlights the unique physiology of each leaf side and their analogous rhythmicity through the day.

International audience; Recent findings were able to show significant variability of stomatal dynamics between species, but not much is known about factors influencing stomatal dynamics and its consequences on biomass production, transpiration and water-use efficiency (WUE). We assessed the dynamics of stomatal conductance (g(s)) to a change of irradiance or vapour-pressure deficit (VPD) in two Populus euramericana and two Populus nigra genotypes grown under control and drought conditions. Our objectives were to determine the diversity of stomatal dynamics among poplar genotypes, and if soil water deficit can alter it. Physiological and morphological factors were investigated to find their potential links with stomatal morphology, WUE and its components at the whole-plant level. We found significant genotypic variability of g(s) dynamics to both irradiance and VPD. Genotypes with faster stomatal dynamics were correlated with higher stomatal density and smaller stomata, and the implications of these correlations are discussed. Drought slowed g(s) dynamics, depending on genotype and especially during stomatal closing. This finding is contrary to previous research on more drought-tolerant species. Independently of the treatment, faster stomatal dynamics were negatively correlated with daily whole-plant transpiration, presenting new evidence of a previously hypothesized contribution of stomatal dynamics to whole-plant water use.

Pour apporter davantage de contexte et d'interprétabilitéà l'analyste SOC, cet article introduit une nouvelle approche d'apprentissage continu de la normalité et de détection de comportement anormal (nouveau ou déviant) avec la production d'une synthèse facilement interprétable par un analyste SOC. Cette approche s'appuie sur des stratégies IAévolutionnistes, des fonctions d'attentions auto paramétrables et des motivations intrinsèques de développement de connaissances comportementales. Dans un esprit d'accroissement de la confiance des analystes envers les algorithmes IA, ce modèle démontre de bonnes capacités de découverte de règles comportementales interprétables et de détection de comportements anormaux dans des logs applicatifs générés par un nouveau simulateur de Système d'Information (SI).
