Aims Numerous pre-clinical and clinical studies have recently demonstrated the significant role of phage therapy in treating multidrug-resistant bacterial infections. However, only a few researchers have focused on monitoring the phage-mediated adverse reactions during phage therapy. The present study aimed to demonstrated the oral acute and sub-acute toxicity of bacteriophages (Klebsiella pneumoniae XDR strain) in Charles Foster rats with special reference to immunological response and adverse effects. Methods and Results Bacteriophages were orally administered in dosages of 1010 PFU/ml and a 1015 PFU/ml to Charles Foster rats as a single dose (in acute toxicity study) and daily dosage for 28 days (in sub-acute toxicity study). One millilitre suspension of bacteriophages was administered through the oral gavage feeding tube. No adverse effect was observed in any of the experimental as well as in the control animals. Furthermore, an insignificant change in food and water intake and body weight was observed throughout the study period compared with the control group rats. On the 28th day of phage administration, blood was collected to estimate haematological, biochemical and cytokines parameters. The data suggested no difference in the haematological, biochemical and cytokine profiles compared to the control group. No significant change in any of the treatment groups could be observed on the gross and histopathological examinations. The cytokines estimated, interleukin-1 beta (IL-1β), IL-4, IL-6 and IFN-gamma, were found within the normal range during the experiment. Conclusions The results concluded that no adverse effect, including the severe detrimental impact on oral administration of high (1010 PFU/ml) and very high dose (1015 PFU/ml) of the bacteriophages cocktail. Significance and Impact of Study The high and long-term oral administration of bacteriophages did not induce noticeable immunological response as well.

Multidrug-resistant (MDR) Acinetobacter baumannii (A. baumannii) is one of the major pathogens present in burn wound infections. Biofilm formation makes it further challenging to treat with clinically available antibiotics. In the current work, we isolated the A. baumannii-specific bacteriophages (BPABΦ1), loaded into the chitosan microparticles followed by dispersion in gel, and evaluated therapeutic efficacy against MDR A. baumannii clinical strains. Isolated BPABΦ1 were found to belong to the Corticoviridae family, with burst size 102.12 ± 2.65 PFUs per infected host cell. The BPABΦ1 loaded chitosan microparticles were evaluated for quality attributes viz. size, PDI, surface morphology, in vitro release, etc. The developed formulation exhibited excellent antibiofilm eradication potential in vitro and effective wound healing after topical application.

The genus Vibrio includes pathogenic species that are naturally occurring in marine and estuarine environments globally. Routine monitoring for Vibrio species and environmental parameters influencing their incidence is critical to provide a warning system for the public when the risk of infection is high.

Vibrio spp. is typically found in salty waters and is indigenous to coastal environments. V. vulnificus and V. parahaemolyticus frequently causes food-borne and non-food-borne infections in the United States. Vibrio spp. is sensitive to changes in environmental conditions and various studies have explored their relationship with the environment and have identified water temperature as the strongest environmental predictor with salinity also affecting the abundance in some cases. It is unclear how additional environmental factors will affect intra-seasonal variance as well as the seasonal cycle. This study investigated the intra-seasonal variations in total and pathogenic V. parahaemolyticus and V. vulnificus organisms in oysters and surrounding waters from 2009 to 2012 at a few locations in the Chesapeake Bay. V. Vulnificus is always pathogenic, but it has been observed that there was greater sample-to-sample variability in pathogenic V. parahaemolyticus than in total V. parahaemolyticus. To determine the increase in the likelihood of vibrio presence when the value of a certain environmental parameter has changed, the odds ratio is examined for various values of environmental factors. The odds ratio that we employed measures the likelihood that the desired outcome would occur in samples with the vibrio in comparison to the likelihood that the desired outcome will occur in samples without the vibrio. This technique will give us the threshold value of the environmental variable above which the likelihood of vibrio spp. presence has increased drastically. With changing climate and environmental conditions, vibrio is posing increasing risks to human health. The findings of this study will demonstrate the effectiveness of the odds ratio technique in estimating the likelihood that vibrio abundance would increase when environmental conditions change, which can then be incorporated into prediction models to reduce the danger to the public's health.

Vibrio spp. are pathogenic bacteria native to warm and brackish water. Vibriosis- the disease caused by these pathogens in humans accounts for around 80000 illnesses and 100 deaths annually in the United States. Of all the species, V. vulnificus has the highest mortality rate of all seafood-borne pathogens in the United States. In this context, understanding the environmental conditions that lead to increased V. vulnificus growth and spread can aid in the development of early warning systems and targeted prevention strategies. Besides sea surface temperature (SST), biotic parameters like coastal chlorophyll are also determined to affect V. vulnificus incidence in humans locally. However, the precise role of coastal chlorophyll as a potential confounding variable is understudied. Moreover, the spatial scale to which the data for environmental variables could be obtained also poses characterization constraints for researchers since the commonly employed in-situ sampling-based methods usually work with discrete locations covering a small area. The present study uses the odds ratio analysis to determine SST and chlorophyll-a threshold values critical to V. vulnificus incidence. The analysis reveals a definite positive relationship between remotely derived environmental variables and the odds of V. vulnificus incidence, where a specific statistical value of SST and chlorophyll-a marks a clear distinction between low and high odds of V. vulnificus incidence. This finding translates into a consistent pattern when checked for counties of coastal Florida. We anticipate our methodology to help distinguish between high and low-risk conditions, enabling public health workers to take proactive measures to protect the health and well-being of the public.

The study objective was to evaluate the therapeutic effect of cannabidiol (CBD) on a combination of caecal slurry, lipopolysaccharide (LPS), and Escherichia coli (E. coli)-induced systemic inflammatory response syndrome (SIRS) in male Sprague Dawley rats.The therapeutic activity was monitored in behavioral tests and inflammatory biomarkers by the enzyme-linked immune sorbent assay (ELISA) method.Behavioral tasks were significantly increased like a tail flick response by 73.84% (p ≤ 0.001), grip strength by 33.56% (p ≤ 0.028), locomotor activity by 20.71% (p = 0.034) in the CBD (60 mg/kg) group compared to disease control (DC) group. Levels of inflammatory serum biomarkers like interleukin-1β (IL-1β), matrix metallopeptidase-9 (MMP-9), IL-6, and tumor necrosis factor-alpha (TNF-α) were significantly decreased by 29.56 (p = 0.041), 71.20 (p ≤ 0.001), 35.05 (p ≤ 0.001), and 75.56% (p = 0.002), respectively, in the CBD-60 compared with DC. Inflammatory cytokines levels, viz. IL-1β, MMP-9, IL-6, and TNF-α, in the liver were significantly (p ≤ 0.001) decreased by 81.01, 40.41, 22.84, and 69.46%, respectively, in CBD-60 to DC. Similarly, levels of inflammatory cytokines such as IL-1β and MMP-9 in the kidney were significantly (p ≤ 0.001) decreased by 80.90 and 43.93%, respectively, in CBD-60 compared to DC.Taken together, results suggest that CBD treatment significantly improved behavioral tasks and decreased the level of inflammatory cytokines under SIRS conditions that might provide an opportunity to manage acute and chronic inflammatory disorders.

The aim of the study was to develop, synthesis, and characterise a novel 1,3,5-trisubstituted-2-pyrazolines derivative, as well as to evaluate its antifungal activity. The reaction of chalcone derivatives with succinic hydrazide in the presence of pyridine yielded the 1,3,5-tri-substituted-2-pyrazolines derivatives. Total 20 compounds has been synthesized and characterized by the IR, 1HNMR and mass spectral analysis. Antifungal activity of the compounds carried out onfour fungal strains i.e. Saccharomyces cerevisiae, A. Niger, C. Albicans and R. Oryzae in two different concentration i.e. 50 and 100 µg/ml by Agar-diffusion method using Cup-plate method. The usual antifungal medicine was ketoconazole. All of the synthesized 1,3,5-trisubstituted pyrazole compounds (ME1-ME8, CL1-CL8, BR1-BR4) showed medium to best action against examined organisms, according to antifungal activity data. The antifungal activity of compounds against fungal strains (Saccharomyces cerevisiae, A. Niger, Candida albicans, and R. Oryzae) revealed the following order of action: CL-4 > BR-4 > CL-3 > CL-2 > ME-3> ME-2> CL-5 > CL-6 > ME-4 > ME-5 > ME-6 > ME-7 > CL-7 > CL-8>ME-8 >CL-1 >ME-1 > CL-5 > CL-6 > ME-4 > ME-5 > ME-6 > ME-7 > CL-7 > CL-8>ME-8 >CL-1 >ME-1. Electronegative groups (Br, Cl, F, and NO2) must be present at the third and fifth positions of the 1,3,5-pyrazoline ring for significant antifungal action. The presence of an electronegative group at the third and fifth positions may be required for the best action against bacterial and fungal strains, however the addition of F, NO2 has demonstrated moderate activity, while the substitution of methyl and methoxy groups may reduce the activity. The synthesized compounds in the BR-1 through BR-4 class are the most active.

Background: Scrub typhus (ST) is a rickettsial infection caused by Orientia tsutsugamushi, which presents with flu like symptoms. This disease has been reported from all over India but with slight variations in its pattern. For decreasing the prevalence, preventing new incidences, and predicting the course of the ST, therefore, it is crucial to gain knowledge and perception of local risk components associated with the disease. The present study aimed to investigate the epidemiological distribution and potential risk factors of O. tsutsugamushi Infection in Eastern Uttar Pradesh (EUP), India. Methods: The serums of 211 samples were collected from the suspected cases along with the detailed information about the participants such as age, location, and place recorded in case history form (CRF). IgM estimation was performed using enzyme-linked immunosorbent assay (ELISA) assay. Results: A total of 58 samples (27.4%) out of 211 ones were found to be positive for IgM antibodies against O. tsutsugamushi bacterium. Furthermore, the results were correlated with epidemiological data such as gender, rural or urban background, pets, and occupation. The results showed that 76.7% of the study participants were from rural areas or had bushes around their houses, 88.3% of them had pets/cattle or frequent encounter with rodents at their houses, and 30.3% of them had no toilet facilities at home. Conclusions: It was concluded that the proximity to pets/cattle, having rodents in closer vicinity, residing in places surrounded by vegetation/farm/bushy areas, and following occupations involving field work increased the chances of getting bitten by mites/chiggers. Overall, Orientia tsutsugamushi prevalence increased in EUP, with respect to clinical features, disease presentation, and laboratory diagnosis can help our community to reduce the mortality caused by this infectious disease.

Japanese encephalitis (JE) is a mosquito-borne flavivirus infection named Japanese Encephalitis Virus (JEV), prevalent in Asia-pacific countries, requires an accurate and rapid diagnosis to contain the outbreak of the disease. In cases of low viral load in early-stage infections, this task becomes difficult. Therefore, we have developed a surface-enhanced Raman spectroscopy (SERS) based biosensor for rapid, sensitive, and early-stage detection of JE antigen. In this work, silver nanoparticles were deposited over a glass coverslip and used as a substrate for designing the sensing platform. Silver Nanoparticles have good metallic properties and plasmon activity. Therefore, it amplifies the Raman signals and provides a suitable surface for the SERS substrate. The developed platform has been used for the detection of the Japanese encephalitis virus (JEV). The fabricated sensor shows a linear response from 5 ng/mL to 80 ng/mL with a limit of detection (LoD) of ∼7.6 ng/mL. Therefore, this method could be a significant addition to the diagnostic modalities for early, sensitive, and specific diagnoses of JE antigen even at the nanogram level.

Tetrahydrocurcumin (THC) is one of the major metabolites of curcumin (CUR), an ancient bioactive natural polyphenolic compound. This research article describes both the solid and liquid state characterization of THC using advanced spectroscopic and thermo-analytical techniques. Anti-inflammatory, anti-oxidant, and neuroprotective activities of THC were investigated using in vitro cell lines. Liquid chromatography-mass spectrometry analysis revealed that our sample comprised 95.15% THC, 0.51% tetrahydrodemethoxycurcumin (THDC), 3.40% hexahydrocurcumin, and 0.94% octahydrocurcumin. Gas chromatography-mass spectrometry analysis indicated the presence of 96.68% THC and 3.32% THDC. THC in solution existed as keto-enol tautomers in three different forms at different retention time, but the enol form was found to be dominant, which was also supported by nuclear magnetic resonance analysis. THC was thermally stable up to 335.55 °C. THC exhibited more suppression of cytokines (TNF-α, IL-1β, and MIP-1α) than CUR in a concentration-dependent manner in mouse splenocytes, while NK-cell and phagocytosis activity was increased in macrophages. THC showed a significant reduction of free radicals (LPO) along with improved antioxidant enzymes (SOD and catalase) and increased free radical scavenging activity against ABTS+ radicals in HepG2 cells. THC displayed higher protection capability than CUR from oxidative stress and neuronal damage by improving cell viability against H2O2 induced HepG2 cells and MPP+ induced SH-SY5Y cells, respectively, in a concentration-dependent manner. Thus, a variation of the biological activities of THC might rely on its keto-enol form and the presence of other THC analogs as impurities. During October 2020, Delta variant was detected for the first time in India and rampantly spread across the globe. It also led to second wave of pandemic in India which affected millions of people. However, there is limited information pertaining to the SARS-CoV-2 strain infecting the children in India.Here, we assessed the SARS-CoV-2 lineages circulating in the pediatric population of India during the second wave of the pandemic. Clinical and demographic details linked with the nasopharyngeal/oropharyngeal swabs (NPS/OPS) collected from SARS-CoV-2 cases (n = 583) aged 0-18 year and tested positive by real-time RT-PCR were retrieved from March to June 2021.Symptoms were reported among 37.2% of patients and 14.8% reported to be hospitalized. The E gene CT value had significant statistical difference at the point of sample collection when compared to that observed in the sequencing laboratory. Out of these 512 sequences 372 were VOCs, 51 were VOIs. Most common lineages observed were Delta, followed by Kappa, Alpha and B.1.36, seen in 65.82%, 9.96%, 6.83% and 4.68%, respectively in the study population.Overall, it was observed that Delta strain was the leading cause of SARS-CoV-2 infection in Indian children during the second wave of the pandemic. We emphasize on the need of continuous genomic surveillance in SARS-CoV-2 infection even amongst children.

This study aimed to detect the SARS-COV2 viral component directly from inoculated VTM without RNA extraction. Inoculated VTMs of already tested 50 positive and 50 negative samples were divided into three groups. Group I was treated with Proteinase K (PK) followed by 3-step-heat treatment at different temperatures (25°C, 60°C, and 98°C) and stored at 4°C. Group II was directly subjected to 3-step-heat treatment without PK exposure and stored at 4°C. And group III was set-up as standard group; it was processed using Qiagen’s column based QIAamp Nucleic Acid kit and the obtained nucleic acids were stored at 4°C. These stored samples were used as a template to execute real-time polymerase chain reaction, and results were noted. Group I demonstrated 96% and 88% sensitivity for N and ORF1ab genes respectively, whereas group II demonstrated 78% and 60% when compared to the results of standard group III. Overall group I showed better results than group II when compared to group III. Thus, in situations where gold-standard reagents are not available, PK exposure and heat treatment can be employed to carry out molecular detection of SARS-CoV2 viral component.

Vibrio spp. thrive in warm water and moderate salinity, and they are associated with aquatic invertebrates, notably crustaceans and zooplankton. At least twelve Vibrio spp. are known to cause infection in humans, and Vibrio cholerae is well documented as the etiological agent of pandemic cholera. Pathogenic non-cholera Vibrio spp., e.g., Vibrio parahaemolyticus and Vibrio vulnificus, cause gastroenteritis, septicemia, and other extra-intestinal infections. Incidence of vibriosis is rising globally, with evidence that anthropogenic factors, primarily emissions of carbon dioxide associated with atmospheric warming and more frequent and intense heatwaves, significantly influence environmental parameters, e.g., temperature, salinity, and nutrients, all of which can enhance growth of Vibrio spp. in aquatic ecosystems. It is not possible to eliminate Vibrio spp., as they are autochthonous to the aquatic environment and many play a critical role in carbon and nitrogen cycling. Risk prediction models provide an early warning that is essential for safeguarding public health. This is especially important for regions of the world vulnerable to infrastructure instability, including lack of 'water, sanitation, and hygiene' (WASH), and a less resilient infrastructure that is vulnerable to natural calamity, e.g., hurricanes, floods, and earthquakes, and/or social disruption and civil unrest, arising from war, coups, political crisis, and economic recession. Incorporating environmental, social, and behavioral parameters into such models allows improved prediction, particularly of cholera epidemics. We have reported that damage to WASH infrastructure, coupled with elevated air temperatures and followed by above average rainfall, promotes exposure of a population to contaminated water and increases the risk of an outbreak of cholera. Interestingly, global predictive risk models successful for cholera have the potential, with modification, to predict diseases caused by other clinically relevant Vibrio spp. In the research reported here, the focus was on environmental parameters associated with incidence and distribution of clinically relevant Vibrio spp. and their role in disease transmission. In addition, molecular methods designed for detection and enumeration proved useful for predictive modeling and are described, namely in the context of prediction of environmental conditions favorable to Vibrio spp., hence human health risk. Multiple organ dysfunction syndrome or failure is one of the major concerns against healthcare services in order to maintain the normal function. The present study aimed to explore the impact of the Biofield Energy Treated test formulation on the function of vital organs such as bones, heart, liver, lungs, and brain using standard activity parameters in specific cell-based assays. The test formulation and cells medium was divided into two parts, one untreated (UT) and other part received the Biofield Energy Treatment remotely by a renowned Biofield Energy Healer, Ariadne Esmene Afaganis, Canada and was labeled as the Biofield Treated (BT) test formulation/media. The test formulation was tested for cell viability, and the data suggested that the test formulation was found safe and non-toxic against all the cell lines. Cytoprotective activity among the experimental groups showed a significant improved activity by 94.4% at 1 µg/mL in untreated medium (UT-Med) + Biofield Treated Test Item (BT-TI) group in human cardiac fibroblasts cells (HCF) cells, while 84.4% at 10 µg/mL in BT-Med + BT-TI groups in human hepatoma cells (HepG2), and 124% increased cytoprotective action at 1 µg/mL in UT-Med + BT-TI group in adenocarcinomic human alveolar basal epithelial cells (A549) cells as compared with the untreated test group. ALP activity in MG-63 cells was significantly increased by 85.9% at 10 µg/mL in the UT-Med + BT-TI group, while in Ishikawa cells showed maximum increased ALP activity by 59.2% at 0.1 µg/mL in BT-Med + BT-TI groups as compared to the untreated group. The percent protection of HCF (heart) cells (decreased of LDH activity) was significantly increased by 53% and 40.5% at 1 and 10 µg/mL concentrations respectively, in UT-Med + BT-TI group, while BT-Med + UT-TI group showed increased protection by 68.5%, 70.7%, and 16.8% at 0.1, 1, and 10 µg/mL respectively, and 86.5%, 62.5%, and 34.2% improved cellular protection at 0.1, 1, and 10 µg/mL respectively, in BT-Med + BT-TI group as compared to the untreated test group. The percent protection of HepG2 (liver) cells (decreased of ALT activity) was reported by 33.5%, 63.2%, and 99.2% at 10 µg/mL in the UT-Med + BT-TI, BT-Med + UT-TI, and BT-Med + BT-TI groups, respectively compared to the untreated group. Cellular protection of A549 (lungs) cells (increased of SOD activity) in terms of percentage was increased by increased by 39.8% (at 10 µg/mL), 44% (at 25.5 µg/mL), and 59.7% (at 25.5 µg/mL) in the UT-Med + BT-TI, BT-Med + UT-TI, and BT-Med + BT-TI groups, respectively compared to untreated group. Serotonin level was significantly increased by 59.2% (at 0.1 µg/mL), 190.3% (at 0.1 µg/mL), and 201% (at 1 µg/mL) in the UT-Med + BT-TI, BT-Med + UT-TI, and BT-Med + BT-TI groups, respectively compared to untreated in human neuroblastoma cells (SH-SY5Y). However, the relative quantification (RQ) of vitamin D receptor (VDR) was significantly increased by 159.1% (at 50 µg/mL), 212.7% (at 1 µg/mL), and 278.3% (at 10 µg/mL) in the UT-Med + BT-TI, BT-Med + UT-TI, and BT-Med + BT-TI groups, respectively as compared to the untreated in MG-63 cells. Thus, the present data concluded that the overall multiple organ health using various standard biomarkers in specific cell lines were significantly improved with respect to health of bones, heart, liver, lungs, and brain after treatment with the Biofield Energy treated test formulation (The Trivedi Effect®). The aim of the present study was to determine the impact of Biofield Energy Treated test formulation using six different cell-lines. The test formulation/item (TI) and cell media (Med) was divided into two parts; one part was untreated (UT) and other part received Biofield Energy Treatment remotely by a renowned Biofield Energy Healer, Janice Patricia Kinney, USA and labeled as Biofield Energy Treated (BT) test item (TI)/ media. Based on cell viability assay, test formulation was found as safe at tested concentrations. Cytoprotective activity of test formulation showed a significant restoration of cell viability by 60.6% (10 µg/mL), 67.5% (63.75 µg/mL), and 117.5% (63.75 µg/mL) in UT-Med + BT-TI, BT-Med + UT-TI, BT-Med + BT-TI, respectively compared to untreated in human cardiac fibroblasts cells (HCF) cells. Moreover, restoration of cell viability was improved by 64% and 127.3% in UT-Med + BT-TI and BT-Med + UT-TI, respectively at 1 µg/mL compared to untreated in human liver cancer (HepG2) cells. Cellular restoration in A549 cells was improved by 314% and 112.3% at 1 µg/mL in BT-Med + UT-TI and BT-Med + BT-TI, respectively than untreated. ALP activity in Ishikawa cells was significantly increased by 175.5%, 547.2%, and 220.8% in UT-Med + BT-TI, BT-Med + UT-TI, and BT-Med + BT-TI, respectively at 0.1 µg/mL as compared to untreated. Additionally, in MG-63 cells showed increased ALP activity by 76.9%, 78.4%, and 79% in UT-Med + BT-TI, BT-Med + UT-TI, and BT-Med + BT-TI, respectively at 50 µg/mL compared to untreated. The percent cellular protection of HCF (heart) cells (decreased of LDH activity) was significantly increased by 60.6% (10 µg/mL), 67.5% (63.75 µg/mL), and 117.5% (63.75 µg/mL) in UT-Med + BT-TI, BT-Med + UT-TI, and BT-Med + BT-TI, respectively as compared to untreated. An improved HepG2 cells protection (represents decreased ALT activity) by 115.1% (1 µg/mL), 42.5% (25.5 µg/mL), and 60.8% (10 µg/mL) in UT-Med + BT-TI, BT-Med + UT-TI, BT-Med + BT-TI, respectively as compared to untreated. Percentage cellular protection of A549 (lungs) cells (represents increased of SOD activity) was significantly increased by 191.1% and 81.4% at 0.1 µg/mL in UT-Med + BT-TI and BT-Med + BT-TI, respectively as compared to untreated. Serotonin level was significantly increased by 31.8% (10 µg/mL) and 56.9% (25.5 µg/mL) in UT-Med + BT-TI and BT-Med + BT-TI, respectively compared to untreated in human neuroblastoma cells (SH-SY5Y). Relative quantification (RQ) of vitamin D receptor (VDR) was significantly increased by 304.3% (0.01 µg/mL), 128.4% (0.1 µg/mL), and 240% (0.1 µg/mL) in UT-Med + BT-TI, BT-Med + UT-TI, and BT-Med + BT-TI, respectively compared to untreated in MG-63 cells. The present study was planned to evaluate the efficacy of Pseudomonas aeruginosa specific phages in immunocompromised septicemia animal model as an alternative to antibiotics. Five different sets of experiments were performed: prophylactic administration of phage cocktail (3 lytic and unique) before and simultaneous with bacterial challenge; and therapeutic, that is, administration of phage cocktail 6, 12, and 24 hours after the bacterial challenge. No mortality was observed when simultaneous and late administration of phages was done with respect to the bacterial challenge. Contrary to this, administration of phage cocktail 100 µL (1012 PFU/mL) of volume after 6 hours of the infection resulted in a mortality rate of 60%. However, no mortality could be observed with reduced dose of cocktail, that is, 108, 109, and 1010 PFU administered 6 hours after bacterial challenge. Phage therapy in acute infections initiated with very small dosage under strict supervision may give better results. However, further studies to determine the quantity and frequency of dosage of phage cocktail for septicemia of various durations is strongly indicated.

The complexity of transmission of COVID-19 in the human population cannot be overstated. Although major transmission routes of COVID-19 remain as human-to-human interactions, understanding the possible role of climatic and weather processes in accelerating such interactions is still a challenge. The majority of studies on the transmission of this disease have suggested a positive association between a decrease in ambient air temperature and an increase in human cases. Using data from 19 early epicenters, we show that the relationship between the incidence of COVID-19 and temperature is a complex function of prevailing climatic conditions influencing human behavior that govern virus transmission dynamics. We note that under a dry (low-moisture) environment, notably at dew point temperatures below 0°C, the incidence of the disease was highest. Prevalence of the virus in the human population, when ambient air temperatures were higher than 24°C or lower than 17°C, was hypothesized to be a function of the interaction between humans and the built or ambient environment. An ambient air temperature range of 17 to 24°C was identified, within which virus transmission appears to decrease, leading to a reduction in COVID-19 human cases.

The study is aimed at establishing the optimal parameters for RNA purification of pooled specimens, in SARS-CoV-2 assay. This research work evaluates the difference of extracted RNA purity of pooled samples with and without treatment with isopropyl alcohol and its effect on real-time RT-PCR. As per the protocol of the Indian Council of Medical Research (ICMR), 5 sample pools were analysed using qRT-PCR. A total of 100 pooled samples were selected for the study by mixing 50 μL of one COVID-19 positive nasopharyngeal/oropharyngeal (NP/OP) specimen and 50 μL each of 4 known negative specimens. Pool RNA was extracted using the column-based method, and 1 set of pooled extracted RNA was tested as such, while RNA of the second set was treated additionally with chilled isopropyl alcohol (modified protocol). Further, the purity of extracted RNA in both the groups was checked using Microvolume Spectrophotometers (Nanodrop) followed by RT-PCR targeting E-gene and RNaseP target. The results showed that the purity index of extracted RNA of untreated pooled specimens was inferior to isopropyl alcohol-treated templates, which was observed to be 85% sensitivity and 100% specificity. The average Cq (E gene) in the unpurified and purified pool RNA group was 34.66 and 31.48, respectively. The nanodrop data suggested that purified RNA concentration was significantly increased with an average value of 24.73 ± 1.49 ng / uL , which might be the reason for high sensitivity and specificity. Thus, this group testing of SARS-CoV-2 cases using pools of 5 individual samples would be the best alternative for saving molecular reagents, personnel time, and can increase the overall testing capacity. However, purity of RNA is one of the important determinants to procure unfailing results, thus, this additional purification step must be included in the protocol after RNA has been extracted using commercially available kit before performing qRT-PCR.

Methods Air samples collected in the hospital room of two COVID-19 patients, one ready for discharge, the other newly admitted, were subjected to RT-qPCR and virus culture. The genomes of the SARS-CoV-2 collected from the air and isolated in cell culture were sequenced. Results Viable SARS-CoV-2 was isolated from air samples collected 2 to 4.8 m away from the patients. The genome sequence of the SARS-CoV-2 strain isolated from the material collected by the air samplers was identical to that isolated from the newly admitted patient. Estimates of viable viral concentrations ranged from 6 to 74 TCID50 units/L of air. Conclusions Patients with respiratory manifestations of COVID-19 produce aerosols in the absence of aerosol-generating procedures that contain viable SARS-CoV-2, and these aerosols may serve as a source of transmission of the virus.

The wound healing activity using scratch assay is considered as a convenient in vitro tool for the assessment of wound healing. The present study deals with the optimization of Biofield Energy Treatment (Consciousness Energy Healing Treatment-The Trivedi Effect®) in the HFF-1 cell line (Human Foreskin Fibroblast) and DMEM (Dulbecco’s Modified Eagle Medium) using scratch assay against positive control, recombinant Human Epidermal Growth Factor (Hu EGF, 30 ng/mL). This methodwas used for the determination of cell proliferation and migration of fibroblast quantitatively in the scratched wounded area. The scratched area was monitored after 24 hours of wound closure in the Biofield Energy Treated HFF-1 cells and the Biofield Energy Treated DMEM groups, and the representative photomicrographs were taken in each wells using WimScratch Image analysis software. The results showed that the Biofield Energy Treated DMEMwassignificantly higher percentage of fibroblast migration i.e. 51.8%, while the migration was altered in the Biofield Energy Treated HFF-1 cell line compared to the baseline control group. Vitamins, minerals, and nanocurcumin play a substantial role in various nutraceutical/pharmaceutical formulations that are widely used in therapeutics, cosmetics, and dietary supplements. The current study aimed to investigate the comparative in vitro immunomodulatory effect of a novel nanocurcumin-based formulation with curcumin in LPS-induced cytokine expression, NK cells’ activity, and phagocytosis. The proinflammatory cytokines (TNF-α, IL-1β, and MIP-1α) and NK cells’ activity were measured in cell supernatants using ELISA assay; however, phagocytosis activity was performed using colorimetric analysis. The chemical characterization of novel nanocurcumin-based formulation using LC–MS (R t 19.02 min) and mass spectra analysis (m/z 369.04) confirmed the presence of the curcumin in highest peak concentration. MTT assay in three tested cell-lines showed that the formulation was found non-toxic at all the tested concentrations. The expression of TNF-α, IL-1β, and MIP-1α in splenocytes was significantly (p ≤ 0.001) inhibited. Besides, the NK cells’ activity and phagocytosis (macrophage) were increased significantly (p ≤ 0.001). Overall, the promising results of this study indicated the significant immunomodulatory effect of nanocurcumin-based formulation compared to the curcumin, which could be used against various inflammatory disorders such as allergy, asthma, autoimmune diseases, coeliac disease, inflammatory bowel disease, etc.

There are many software easily available that can modify an image. With such a wide usage of images in communication networks it becomes difficult to conclude if an image is an original one or a forged one. This paper proposes the usage of image metadata, which is much smaller in size as compared to contents of an image, to authenticate an image. Further, the architecture of a system using a neural network is proposed which extracts attributes from the metadata of an image to predict if an image is an original one or a forged one. The system is also capable of predicting the operation applied and the software/tool used to modify the original image in case of a forged image. Experiments have been conducted on JPEG images which are modified with operations like Crop, Rotate, Resize, Compress, Compress_Rotate and Crop_Rotate to validate our system.

The major cause of high rate of mortality is the multiple organ dysfunction among critical care healthcare services. The aim of the current study was to evaluate the impact of the Biofield Energy Treated test formulation using standard and specific cell lines related with vital organs functioning. The test formulation and the specific cell media was divided into two parts; one part was untreated (UT) and other part received the Biofield Energy Treatment remotely by a renowned Biofield Energy Healer, Sakina Aleemah Ansari, USA and labeled as the Biofield Energy Treated (BT) test formulation/media. Cytoprotective activity showed improved cellular restoration by 105.4% (at 25 µg/mL), 32.8% (at 25 µg/mL), and 151.8% (at 10 µg/mL) in the UT-Med + BT-TI, BT-Med + UT-TI, and BT-Med + BT-TI groups respectively, as compared to the untreated test group in the human cardiac fibroblasts cells (HCF) cells, while improved restoration of cell viability by 22.4% (at 25.5 µg/mL), 67.1% (at 10 µg/mL), and 72.9% (at 10 µg/mL) in the UT-Med + BT-TI, BT-Med + UT-TI, and BT-Med + BT-TI groups respectively, as compared to the untreated test group in HepG2 cells. Cellular restoration in A549 cells was improved by 9.3%, 70.4%, and 14.1% at 0.1, 1, and 25.5 µg/mL respectively, in the BT-Med + UT-TI group, while 3% and 4.6% improved cellular restoration was reported at 10 and 25.5 µg/mL respectively, at BT-Med + BT-TI groups as compared to the untreated test group. ALP activity in Ishikawa cells was significantly increased by 68.4%, 41.1%, and 18.8% at 0.1, 10, and 50 µg/mL respectively, in the UT-Med + BT-TI group, while in MG-63 cells showed maximum increased ALP activity by 92.7%, 84.5%, and 93.2% respectively in the UT-Med + BT-TI, BT-Med + UT-TI, and BT-Med + BT-TI group respectively, at 50 µg/mL as compared to the untreated group. The maximum percent cellular protection of HCF (heart) cells (decreased of LDH activity) was significantly increased by 51.6%, 88.7%, and 53.7% in the UT-Med + BT-TI, BT-Med + UT-TI, and BT-Med + BT-TI groups respectively, at 10 µg/mL as compared to the untreated group. Alanine amino transferase (ALT) activity was reported in terms of percent cellular protection of HepG2 (liver) cells. The test data showed improved HepG2 cells protection (represents decreased ALT activity) by 33%, 90.2%, and 72.1% in the UT-Med + BT-TI, BT-Med + UT-TI, and BT-Med + BT-TI groups respectively, at 25 µg/mL as compared to the untreated test group. Percentage cellular protection of A549 (lungs) cells (represents increased of SOD activity) was increased by 21.5% at 25.5 µg/mL in the UT-Med + BT-TI, while 33.4%, 25.8%, and 21.5% at 1, 10, and 25.5 µg/mL respectively, in the BT-Med + UT-TI group, and 12.8% increased SOD activity at 25.5 µg/mL in the BT-Med + BT-TI groups as compared to the untreated group. Serotonin level was significantly increased 137.4% (at 0.1 µg/mL), 65.4% (at 0.1 µg/mL), and 77.3% (at 10 µg/mL) in the UT-Med + BT-TI, BT-Med + UT-TI, and BT-Med + BT-TI groups, respectively compared to untreated test group in human neuroblastoma cells (SH-SY5Y). Bone is a dynamic tissue, which continually adapting its structure during the development process. The aim of the study was to investigate the effect of Consciousness Energy Healing based vitamin D3 and DMEM medium on bone health parameters such as alkaline phosphatase enzyme (ALP) activity, collagen levels and bone mineralization using MG-63 cells. The test items (TI) i.e. vitamin D3 and DMEM medium were divided into two parts. The test samples received Consciousness Energy Healing Treatment by Lorraine Marie Hachfeld and samples were defined as the Biofield Energy Treated (BT) samples, while the other parts of each sample were denoted as the untreated test items (UT). Cell viability using MTT assay showed that cell viability was more than 79% with a safe and nontoxic profile on MG-63 cell line. The level of ALP was increased by 400%, 84%, and 179.4% at 0.1, 1, and 10µg/mL, respectively in the UT-DMEM+BT-TI, while 314.3% at 10µg/mL in the BT-DMEM+UT-TI group as compared with the untreated test item and DMEM group. BT-DMEM+BT-TI group showed an increased ALP level by 128% and 60.3% at 1 and 10µg/mL, respectively. Collagen was significantly increased by 180% and 128.1% at 0.1 and 1µg/mL, respectively in the UT-DMEM+BT-TI group, while 84.4% and 69.7% at 1 and 10µg/mL, respectively in BT-DMEM+UT-TI group as compared with the untreated group. Furthermore, BT-DMEM+BT-TI group showed a significant increased collagen level by 123.4% and 184.8% at 1 and 10 µg/mL, respectively as compared with the untreated test item and DMEM group. The percent of bone mineralization was significantly increased by 385.1% at 0.1µg/mL in the UT-DMEM+BT-TI group, while 53.2%, 30.2%, and 163.9% at 0.1, 1, and 10µg/mL, respectively in BT-DMEM+UT-TI group as compared with the untreated group. In addition, BT-DMEM+BT-TI group showed a significant increased bone mineralization by 283%, 65.1%, and 197.3% at 0.1, 1, and 10µg/mL, respectively as compared with the untreated group. A novel series of substituted benzothiazoles, bearing semicarbazone and thiosemicarbazone moieties, was designed, synthesized and evaluated for their antimicrobial activity and possible mode of action. Structures of the synthesized compounds were elucidated by 1H NMR, 13C NMR, IR and Mass spectral data. The results revealed that compounds SC06, SC09, TS05 and TS07 have potent antibacterial activity against both Gram-positive and Gram-negative strains. Compound TS05 displayed most potent activity with MIC values of 3.91, 7.81 and 1.56 µg/ml against S. aureus, E. coli and P. aeruginosa, respectively. The results from cytoplasmic membrane permeabilization assay, FACS study as well as DNA-binding assays, evaluated against clinically relevant pathogens S. aureus and E. coli, suggest membrane perturbing as well as intracellular mode of action of this class of compounds. In addition, hemolytic activity of the compounds was measured which indicated their low cytotoxicity.

A new proprietary herbomineral formulation was formulated, consisting of essential ingredients viz. herbal root extract ashwagandha and minerals (zinc, magnesium, and selenium). The aim of the study was to evaluate the immunomodulatory potential of Biofield Energy Healing (The Trivedi Effect®) Treatment on the herbomineral formulation in male Sprague Dawley rats. The test formulation was divided into two parts. One part was denoted as the control without any Biofield Energy Treatment, while the other part was defined as the Biofield Energy Treated sample, which received the Biofield Energy Healing Treatment remotely from twenty renowned Biofield Energy Healers. The experimental parameters studies were humoral immune response (primary and secondary titre), delayed type hypersensitivity reaction, animal weight parameters, feed and water intake, histopathology, hematological and serum biochemistry. Humoral immune response showed the primary and secondary antibody titre values were significantly (p≤0.05) increased by 5.33% and 112.5%, respectively in the Biofield Energy Treated test formulation (G4) group with respect to the disease control group (G2). Delayed type hypersensitivity results showed significant increase in paw edema by 88.24% (p≤0.05) and 25% at 24 and 48 hours of duration, respectively in the Biofield Energy Treated test formulation (G4) group compared with the G2 group. Hematological studies showed a significant increase in the RBC count, hemoglobin, and packed cell volume (PCV), while the platelet count was increased by 8.40% in the Biofield Energy Treated test formulation group (G4) compared with the disease control group (G2). Biochemical analysis exhibited an increased level of calcium and phosphorus by 2.34% and 7.38%, respectively while serum uric acid was significantly decreased by 18.34% along with a slight increase in the magnesium, potassium, and sodium ions in the Biofield Energy Treated test formulation (G4) compared with the G2 group. In conclusion, the Biofield Energy Treated Test Formulation would be the powerful immunomodulatory product, which was found to be safe at the tested doses. The aim of this study was to investigate the role of tetrahydrocurcumin (THC) against various skin health parameters using in vitro human foreskin fibroblast and melanoma cell lines (i.e. HFF-1 and B16-F10). The study was assessed using cell viability by MTT assay, identification of extracellular matrix component in HFF-1 cell line (i.e. collagen, elastin and hyaluronic acid), melanin synthesis in B16-F10 cells, cell viability against UVB-induced stress in HFF-1 cells, and in vitro wound healing by the scratch assay. THC was found to be safe and nontoxic up to the concentration of 10 µg/mL with improved level of collagen (37.90%), elastin (90.1%), and hyaluronic acid (74.19%) at 1 µg/mL. Besides, melanin was significantly inhibition by 78.5% at the lowest THC concentration of 0.1 µg/mL. UVB-protection rate was significantly improved by 61.2% and improved cell viability by THC in HFF-1 cells, which indicated protection from photoaging. In addition, THC showed significant wound healing activity (78.51%) and greater migration of fibroblast in HFF-1 cells at different time period. It can be concluded from the study that THC can protect the skin matrix with improved extracellular component synthesis and would healing via collagen synthesis in the skin, which improved the skin elasticity and tightness. Overall, it might be suggested that THC can be used as a safe skin whitening agent, wounds management, cosmetic applications, and treating various skin-related disorders.
