Microbial lipids are a valuable source of potential biofuels and essential polyunsaturated fatty acids. The optimization of the fermentation conditions is a strategy that affects the total lipid concentration. The genus Nigrospora sp. has been the target of investigations based on its potential bioherbicidal action. Therefore, this study developed a strategy to maximize the biomass concentration and lipid accumulation by Nigrospora sp. in submerged fermentation. Different media compositions and process variables were investigated in shaken flasks and bioreactor in batch and fed-batch modes. Maximum biomass concentration and lipid accumulations were 40.17 g/L and 21.32 wt% in the bioreactor, which was 2.1 and 5.4 times higher than the same condition in shaken flasks, respectively. This study presents relevant information to the production of fungal lipids since few investigations are exploring the fed-batch strategy to increase the yield of fungi lipids, as well as few studies investigating Nigrospora sp. to produce lipids.

Cutinases are serine esterases that belong to the α/β hydrolases superfamily. The natural substrates for these enzymes are cutin and suberin, components of the plant cuticle, the first barrier in the defense system against pathogen invasion. It is well-reported that plant pathogens produce cutinases to facilitate infection. Fusarium verticillioides, one important corn pathogens, is an ascomycete upon which its cutinases are poorly explored. Consequently, the objective of this study was to perform the biochemical characterization of three precursor cutinases (FvCut1, FvCut2, and FvCut3) from F. verticillioides and to obtain structural insights about them. The cutinases were produced in Escherichia coli and purified. FvCut1, FvCut2, and FvCut3 presented optimal temperatures of 20, 40, and 35 °C, and optimal pH of 9, 7, and 8, respectively. Some chemicals stimulated the enzymatic activity. The kinetic parameters revealed that FvCut1 has higher catalytic efficiency (Kcat/Km) in the p-nitrophenyl-butyrate (p-NPB) substrate. Nevertheless, the enzymes were not able to hydrolyze polyethylene terephthalate (PET). Furthermore, the three-dimensional models of these enzymes showed structural differences among them, mainly FvCut1, which presented a narrower opening cleft to access the catalytic site. The fungi-based technology provided encouraging scenarios in the transition from a conventionally based economic system to the potential security of sources closely associated with the agricultural sphere such as the agriculture. In recent years, the intensification of fungi-based processes has generated significant gains, additionally to the production of materials with significant benefits and strong environmental importance. Furthermore, the growing concern for human health, especially in the agriculture scenario, has fostered the investigation of organisms with high biological and beneficial potential for use in agricultural systems. Accordingly, this study offered a comprehensive review of the diversity of the soil fungal microbiome and its main applications in a biotechnological approach aimed at agriculture and food chain-related areas. Moreover, the spectrum of opportunities and the extensive optimization platform for obtaining fungi compounds and metabolites are discussed. Finally, future perspectives regarding the insurgency of innovations and challenges on the broad rise of visionary solutions applied to the biotechnology context are provided., (© 2023. This review presents scientific findings which indicate biomolecules are excellent candidates for the development of biopesticides. Efforts are being done to find routes to increase their concentrations in the cultivation media because this concentration facilitates applications, storage, and transportation. Some of these routes are co-fermentation and ultrasound-assisted fermentation. Ultrasonication increases metabolite production and growth rates by improvement of cell permeability and nutrient uptake rates through cell membranes. For example, 24% increase in the enzymatic activity of cellulases produced by Trichoderma reesei in solid-state fermentation was achieved with ultrasonication. Also, chitinase and β-1,3-glucanase productions were stimulated by ultrasound in Beauveria bassiana cultivation, presenting positive results. The common parameters evaluated in the production of biomolecules by ultrasound-assisted fermentation are the duty cycle, time of application, power, energetic density, and how long the sonication is maintained in the fermentation media. Many successful cases are reported and discussed, which include the final formulation of bioproducts for agricultural applications. In this trend, nanotechnology is a promising tool for the development of nanoformulations. Consumers increasingly prefer and seek functional beverages, which, given their characteristics, provide important bioactive compounds that help prevent and treat chronic diseases. Mead is a traditional fermented alcoholic beverage made from honey solution. The aging process of mead with oak chips is innovative and bestows functional characteristics to this beverage. Thus, in this study, we sought to develop and characterize a novel functional beverage by combining the health benefits of honey with the traditional aging process of alcoholic beverages in wood. Phenolic compounds, flavonoids, and antioxidant capacity were analyzed in mead using oak chips at different toasting levels and aged for 360 days. LC-ESI-QTOF-MS/MS was used to analyze the chemical profile of different meads. Over time, the aging process with oak chips showed a higher total phenolic and flavonoid content and antioxidant capacity. Eighteen compounds belonging to the classes of organic acids, phenolic acids, flavonoids, and tannins were identified in meads after 360 days. Our findings revealed that the addition of oak chips during aging contributed to p -coumaric, ellagic, abscisic, and chlorogenic acids, and naringenin, vanillin, and tiliroside significantly impacted the functional quality of mead.

This study describes the use of spray drying technology to obtain a powder containing biomolecules with herbicidal activity produced by submerged fermentation using Diaporthe schini . The efficiency of the bioherbicide was tested for the post-emergence control of Bidens pilosa L., Amaranthus viridis L., Echinochloa crusgalli (L.) Beauv., and Lolium multiflorum Lam. In the first step, different additives were used and lactose was the most suitable one because it resulted in high herbicidal activity and weed suppression. In the second step, process variables were investigated, including inlet air temperature, drying air flow rate, and feed flow rate. The highest herbicidal activity was obtained with an inlet air temperature of 100°C, and air and feed flow rates of 1.4 m 3 /min and 0.22 L/h, respectively. Maximum herbicidal activities were 38, 45, 21 and 18%, while weed heights reduction were 69.0, 74.3, 20.4 and 24.8% for B. pilosa , A. viridis, E. crusgalli and L. multiflorum , respectively. The bioherbicide was effective to suppress weed growth and spray drying is a promising technology for the production of solid formulations of bioherbicides.

Cell wall degrading enzymes (chitinase and β-1,3-glucanase) were produced by solid-state fermentation (SSF) using the fungus Trichoderma harzianum and different agro-industrial products, mainly residues. The influence of temperature (25-35 °C), initial moisture content (50-90% w/w), nutrient solution (1-2% v/w), and yeast extract (1-2% w/w) on enzyme activity was evaluated. The application of ultrasound during fermentation for different times (0-6 h/day) was also studied. White rice was the substrate that showed the highest chitinase and β-1,3-glucanase activities, which were 31.31 U/g for chitinase and 23.83 U/g for β-1,3-glucanase after 10 days of fermentation. Application of ultrasound waves during fermentation positively affected ( p  < 0.05) the enzyme activities. The best results for chitinase (51.88 U/g) and β-1,3-glucanase (39.22 U/g) were obtained with a 50% (w/w) moisture content and 4 h/day ultrasound application for 10 days of fermentation. Increases of 3.6-fold (from 14.37 to 51.88 U/g) and 3.8-fold (from 10.22 to 39.22 U/g) in activities for chitinase and β-1,3-glucanase, respectively, compared to non-sonicated fermentation, were obtained. Oleaginous microorganisms, including the fungus Umbelopsis isabellina, have emerged as a biotechnological alternative to obtain polyunsaturated fatty acid-rich oils, which are strongly linked to energy purposes (biofuel) than the food industry. Considering the composition of microbial oil and its use by the food industry, it is necessary to investigate strategies that increase its lipid stability. Ergo, this pioneering study aimed to microencapsulate the oil produced by Umbelopsis isabellina and evaluate its oxidative stability throughout the storage period against factors such as temperature and luminosity. The microbial oil was microencapsulated through the external ionic gelation technique, producing an encapsulation efficiency of 80% and proving to be a suitable method because it maintained oil composition. Combining microencapsulation and refrigerated storage led to the best effects on storage time, increasing the evaluated lipid stability through the peroxide values and conjugated diene formation. Moreover, saturated and monounsaturated fatty acid content increased, and polyunsaturated fatty acid content decreased during storage for both the free and microencapsulated oil, regardless of storage temperature, although microencapsulation reduced the changes. The results primarily demonstrate how microencapsulation prolongs the oxidative stability and unsaturated fatty acid content of the microbial oil by reducing its reactions to external environmental factors, thus facilitating its use in the food industry., (Copyright © 2021 Elsevier Ltd. Weed control is a critical factor to ensure productivity and quality for food production. Chemical control is the main method used worldwide, but the demand for healthier food and the farmers' health and environment concerns have led to an increase in the search for alternative control methods. In this way, the use of biomolecules produced by microorganisms that present phytotoxic activity against weeds, such as exopolysaccharides, is attracting attention. For this purpose, this work compared two techniques (solid-state and submerged fermentation) for bioherbicide production by Diaporthe schini . Physicochemical characterization of both fermented broth and evaluation of bioherbicidal effect in post-emergence of Amaranthus viridis , Bidens pilosa , Echinocloa crusgalli , and Lollium multiflorum were performed. Fungal broth obtained by submerged fermentation presented better physicochemical characteristics in terms of viscosity, density, and surface tension. Overall, it was more effective than the broth obtained by solid-state fermentation for weed control because it presented an average inhibition of 40% of weed growth and 45% lower surface tension if compared to the control test. Also, reductions of 1.4-4.2 times of root dry mass, 2.9-5.8 times of shoot dry mass and 1.2-3.9 times of weeds heights, if compared to the control test, were achieved.

Ruellia angustiflora (Acanthaceae) is known as flower-of-fire, and its leaves are traditionally employed to promote wound healing. This study was aimed at extracting and characterizing the chemical constituents of the extracts of R. angustiflora obtained by ultrasound-assisted and supercritical fluid extractions, and subsequently investigate their antioxidant potential and cyto-genotoxicity. The extract obtained by ultrasound (UAE-EtOH) was characterized by ultraperformance liquid chromatography-mass spectrometer (UPLC-MS), and the extract obtained via supercritical fluid (SFE-CO 2 ) by gas chromatograph-mass spectrometer (GC-MS). The antioxidant potential was verified by the antiradical activity against the 1,1-diphenyl-2-picrylhydrazyl radical (DPPH), and the cyto-genotoxicity evaluation to test cell viability and DNA damage was performed in peripheral blood mononuclear cells (PBMC) cultures. The SFE-CO 2 extract presented some fatty acids, triterpenes, tetraterpenes, tocopherols and phytosterols. The UAE-EtOH extract contained phenolic acids and flavonoids, and showed the highest antioxidant capacity. Neither extract was genotoxic or cytotoxic at the tested concentrations.

The residue from chicken mechanically separated meat (MSM) is a potential source for the extraction of collagen. However, this process requires the removal of many covalent crosslinks, which makes it quite complex. Ultrasound has been successfully used to extract collagen; it reduces the process time and increases the yield. However, information regarding the effects of this treatment on the structural and functional properties of proteins is still very limited. Therefore, the aims of the present study were to obtain collagen from chicken MSM residue and to test the effects of pre-treatment with ultrasonic probe and enzymatic extraction with pepsin in its yield, as well as to evaluate the properties of extracted collagen using gel electrophoresis, Fourier-transform infrared spectroscopy, solubility, and differential scanning calorimetry. Both the ultrasound and the enzymatic extraction had a positive effect on the extraction yield of collagen from chicken MSM residue without affecting its integrity. Using ultrasound led to an increase of up to 40% in yield when compared to treatments without ultrasound application. Five extraction treatments were considered. The extracted collagen exhibited high thermal stability (43.9-47.0 °C) and mainly type I structure. The use of ultrasound as pre-treatment, together with enzymatic extraction with pepsin, were effective in increasing the extraction yield of collagen from chicken MSM residue, as well as preserving the triple helical structure of the native collagen., (© Association of Food Scientists & Technologists (India) 2020.)

Several plant species synthesize biologically active secondary metabolites. Pyrrolizidine alkaloids are a large group of biotoxins produced by thousands of plant species to protect against the attack of insects and herbivores, but they are highly toxic for humans and animals. In this study, extracts from the aerial part of Senecio brasiliensis were obtained using different technologies: ultrasound-assisted extraction (UAE), pressurized liquid extraction (PLE), and microwave hydrodiffusion and gravity (MHG). The study aimed to evaluate the effectiveness of these technologies for the extraction of chemical compounds found in this plant, focusing on two pyrrolizidine alkaloids: integerrimine and senecionine. Influential parameters on yield and chemical composition were also evaluated: for UAE and MHG, temperature and pressure; for PLE, temperature, and percentage of ethanol. All the extraction techniques were efficient for the extraction of integerrimine and senecionine. The UAE and PLE stood out for the higher yields and number of compounds. The PLE presented a maximum yield of 18.63% for the matrix leaf and the UAE a maximum yield of 11.82% for the same matrix. The objective of this study was to evaluate the ethanol production by Wickerhamomyces sp. using soybean straw and hull hydrolysates obtained by subcritical water hydrolysis and, afterward, the biogas production using the fermented hydrolysates. Ethanol was produced using the straw and hull hydrolysates diluted and supplement with glucose, reaching 5.57 ± 0.01 g/L and 6.11 ± 0.11 g/L, respectively. The fermentation in a bioreactor with changing the pH to 7.0 allowed achieving maximum ethanol production of 4.03 and 3.60 g/L for straw and hull hydrolysates at 24 h, respectively. The biogas productions obtained for the fermented hydrolysates of straw with and without changing the pH were 739 ± 37 and 652 ± 34 NmL/g VSad , respectively. The fermented hydrolysate of hull without changing the pH presented 620 ± 26 NmL/g VSad . Ultrasound-assisted extraction (UAE) and pressurized hot water extraction (PHWE) were tested as advanced clean methods to obtain polysaccharides from Phoma dimorpha mycelial biomass. These methods were compared to conventional extraction (hot water extraction, HWE) in terms of polysaccharides-enriched fractions (PEF) yield. A central composite rotational design was performed for each extraction method to investigate the influence of independent variables on the yield and to help the selection of the condition with the highest yield using water as an extraction solvent. The best extraction condition of PEF yielded 12.02 wt% and was achieved when using UAE with direct sonication for 30 min under the intensity of 75.11 W/cm 2 and pulse factor of 0.57. In the kinetic profiles, the highest yield (15.28 wt%) was obtained at 50 °C under an ultrasound intensity of 75.11 W/cm 2 and a pulse factor of 0.93. Structural analysis of extracted polysaccharide was performed using Fourier-transform infrared spectroscopy, X-ray diffraction, scanning electron microscopy, and thermal property. The water solubility index, water holding capacity, and emulsification index of PEF were 31.3 ± 1.5%, 138.1 ± 3.2%, and 62.9 ± 2.3%, respectively. The submerged fermentation demonstrates the huge potential of Phoma dimorpha to produce polysaccharides with bioemulsifying properties as a biotechnologically cleaner alternative if compared to commercial petroleum-derived compounds. Furthermore, UAE and PHWE are green technologies, which can be operated at an industrial scale for PEF extraction.

The natural ability of microorganisms to secrete high levels of bioactive compounds make them attractive hosts for producing novel compounds. Microbial biopolymers have potential applications in most of the sectors of the world economy. According to the physicochemical properties, they present some advantages, such as biodegradability, reproducibility, and stability. Based on this context, the objective of this work was to evaluate different methods for concentration and characterisation of extracellular biopolymers produced by Phoma sp. Extracellular biopolymers were produced by submerged fermentation and were concentrated by hollow fibre membranes and by adsorption. The structural characterisation of purified biopolymers was determined by Fourier Transform Infrared spectroscopy. Phytotoxic effects were assessed through absorption assays in detached leaves of Cucumis sativus and evaluated on the seventh day after application. The surface tension was evaluated for each sample. Hollow-fibre microfiltration membrane presented a higher purification factor than hollow-fibre ultrafiltration membrane. Extracellular biopolymers were identified in the permeate and retentate fractions, but in higher concentration in the retentate fractions. The adsorption process was efficient for recovering more than 88% of extracellular biopolymers from cell-free fermented broth. The best performance was obtained by using silica and activated carbon as adsorbent, with a recovery higher than 93%. The herbicidal activity was proportional to the concentration of biopolymers and the results are very promising for future applications because a concentrated solution of biopolymers can increase weed control. Membrane processes can be used to develop a liquid formulation of bioherbicide, whereas adsorption can be used to develop a solid formula.

Fungal biopolymers have gained considerable attention from the scientific community for various applications due to their biological and physicochemical properties. The wide applications in several areas, especially in the food industry as a bioemulsifier and in the agricultural area as a biosurfactant, have expanded the knowledge on the production of fungal biopolymers to keep up with developments on this subject area. Recent scientific studies have disclosed novel routes, optimized parameters, increased yields, and other related approaches in order to produce and apply fungal bioemulsifiers and biosurfactants. However, there is a need to gather important information in order to provide a way forward. Therefore, this review presents an overview of properties, applications, and perspectives for encouraging further projects and investments in the near future by most categories of investors. The selection of culture media, the definition of cultivation parameters, extraction, recovery, and purification are the initial steps to indicate the conditions for scale-up. Indeed, scale-up is still one of the challenges in this biotechnological field, which could be solved by expanding the tests and operational productions in both pilot and industrial plants.

The chitinases have extensive biotechnological potential but have been little exploited commercially due to the low number of good chitinolytic microorganisms. The purpose of this study was to identify a chitinolytic fungal and optimize its production using solid state fermentation (SSF) and agroindustry substrate, to evaluate different chitin sources for chitinase production, to evaluate different solvents for the extraction of enzymes produced during fermentation process, and to determine the nematicide effect of enzymatic extract and biological control of Meloidogyne javanica and Meloidogyne incognita nematodes. The fungus was previously isolated from bedbugs of Tibraca limbativentris Stal (Hemiptera: Pentatomidae) and selected among 51 isolated fungal as the largest producer of chitinolytic enzymes in SSF. The isolate UFSMQ40 has been identified as Trichoderma koningiopsis by the amplification of tef1 gene fragments. The greatest chitinase production (10.76 U gds -1 ) occurred with wheat bran substrate at 55% moisture, 15% colloidal chitin, 100% of corn steep liquor, and two discs of inoculum at 30 °C for 72 h. Considering the enzymatic inducers, the best chitinase production by the isolated fungus was achieved using chitin in colloidal, powder, and flakes. The usage of 1:15 g/mL of sodium citrate-phosphate buffer was the best ratio for chitinase extraction of SSF. The Trichoderma koningiopsis UFSMQ40 showed high mortality of M. javanica and M. incognita when applied to treatments with enzymatic filtrated and the suspension of conidia.

The objective of this work was to concentrate, through a membrane separation process, the fatty acids from oil/solvent mixture. The oil was obtained by ultrasound-assisted extraction from freeze-dried cells of Mortierella isabellina. The concentration of the fatty acids was investigated using flat-sheet polymer membranes of ultrafiltration and nanofiltration. The effects of temperature and pressure were evaluated by the retention of the fatty acids. Oil retentions between 45.23 and 58.20% to ultrafiltration membrane and 43.50 and 56.00% to nanofiltration membrane were observed. The best condition for the ultrafiltration membrane was 4 bar and 40 °C and for nanofiltration membrane was 12 bar and 50 °C. The oil contains a high concentration of oleic acid and palmitic acid that is a desirable property for the biodiesel production. The results showed the applicability of this technology in the solvent recovery step whereas the oil recovered contains a high concentration of fatty acids.

Exopolysaccharides are secondary metabolites produced by microorganisms and are a subject of research in many fields of science and industry due to some of their confirmed properties, especially in the pharmaceutical and agrochemical areas. In this context, the objectives of this work were to evaluate the potential of Fusarium fujikuroi for producing exopolysaccharides and to concentrate such compounds in order to increase the herbicidal activity. Exopolysaccharides were produced by submerged fermentation and different concentration methods (membranes, lyophilization, and evaporation) were evaluated. The phytotoxic effects were assessed through absorption assays in detached leaves of Cucumis sativus and evaluated on the seventh day after application. The surface tension was evaluated for each concentration method. The production of exopolysaccharides in the crude broth without concentration was 5.94 g/L. When using the lyophilization method, a maximum yield of exopolysaccharides of 10.64 g/L was obtained. The membranes also presented satisfactory results of exopolysaccharides: 9.60 g/L. The increase of bioherbicidal activity and the lower surface tension are proportionally related to the increase of the concentration of exopolysaccharides.

Pecan cultivation has increased in recent years. Consequently, the amount of lignocellulosic residuals from its production has expanded. Thus, there is a necessity to explore and add value to their coproducts. The objective of this work was to obtain reducing sugars from pecan biomasses by the optimization of the subcritical water hydrolysis technology in a semi-continuous mode and the physicochemical and morphological characterization of these materials, such as SEM, TGA and FT-IR analysis. Temperatures of 180, 220 and 260 °C, water/solids mass ratio of 15 and 30 g water/g biomass and total reaction time of 15 min were used. The highest reducing sugar yield was 27.1 g/100 g of biomass, obtained at 220 °C and R-15 for pecan shells. Sugarcane bagasse, a largely available waste worldwide, was submitted to solid-state fermentation (SSF) using the fungus Metarhizium anisopliae, aiming to produce enzymes. The solid waste generated from SSF was tested as an alternative biosorbent to treat colored effluents containing crystal violet (CV) dye. The biosorbent, here named BW (bagasse waste), was characterized, and experimental tests were performed to verify the influence of pH and dosage on the CV biosorption. Isotherms and biosorption kinetics were performed, and the biosorption thermodynamic parameters were determined. The potential of BW was also evaluated for the treatment of a simulated textile effluent. The maximum biosorption capacity was 131.2 mg g -1 at 328 K, and the Liu was the most appropriate model to represent equilibrium data. The biosorption was spontaneous and endothermic. The use of BW in the simulated effluent showed that it is an efficient material, reaching color removal values of 85%. Therefore, the sugarcane bagasse generated from SSF can be considered a potential biosorbent to remove CV from textile effluents. This finding is relevant from the total environment viewpoint, since, at the same time, SSF generates enzymes and a solid waste, which in turn can be used as biosorbent to treat colored effluents.

Antioxidants are able of inhibiting free radicals and play an important role in human diet, protection of foods, anti-aging cosmetics, among others. Fungi have been highlighted in production of metabolites with biological activity, such as antioxidant. The main objective of this study was to select a fungus from resources of Brazilians Pampa and Atlantic Forest biomes with hability for production of antioxidants by submerged fermentation. The scavenging activity of the compounds was determined against radical 2.2-diphenyl-1-picrylhydrazyl (DPPH), superoxide and hydroxyl. Botryosphaeria dothidea exhibited higher potential for the production of antioxidants, and its supernatant showed percentage inhibition values of 94.47, 94.87 and 89.78% against DPPH, superoxide and hydroxyl radicals and IC 50 of 0.206 mg/mL. The identification of the volatile compounds present in the broth verified the presence of the hexahydropyrrolizin-3-one; 1.2-benzenedicarboxylic acid bis(2-methylpropyl) ester and 3.6-bis(2-methylpropyl)-2.5-piperazinedione. According to this study, Botryosphaeria dothidea presents great potential to produce compounds with antioxidant activity.

The adsorption of acid red 97 dye (RED 97) by the waste of the filamentous fungus Beauveria bassiana was analyzed. The adsorbent was obtained as a waste of a fermentative process, and characterized by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FT-IR), X-ray powder diffractometry (XRD), and specific surface area (BET). After the characterization, adsorption tests were carried out to determine the ideal conditions of pH, adsorbent mass, and contact time for the process. Adsorption isotherms, thermodynamic studies, and the treatment of textile effluent were also investigated. The adsorbent characterization allowed the visualization of its amorphous structure, with irregular and heterogeneous particles. The pore diameter was 51.9 nm and the surface area was 0.247 m 2 g -1 . 1.2 g L -1 of the adsorbent and pH of 2.0 were the ideal conditions for RED 97 adsorption. The pseudo-second-order kinetic model was the most appropriate to represent the experimental data, being the equilibrium reached in about 110 min. The Langmuir model was the most suitable to represent the equilibrium data, with maximum adsorption capacity of 194.1 mg g -1 at 45 °C. The adsorption processes was thermodynamically spontaneous, favorable, and exothermic. In the treatment of a real textile effluent, 5 g L -1 of the spores was capable to decolorize 70% of the solution. Therefore, spore wastes of Beauveria bassiana were promising for RED 97 adsorption.

The aim of this study was to develop and evaluate the physicochemical and antioxidant stability of nanoemulsions containing a Physalis peruviana calyx extract (CPp-NE) and free extracts under different storage conditions (7 and 25 °C) and with absence or incidence of light for 120 days. The calyx extracts were prepared with ethanol 60% and characterized for later preparation of the nanoemulsions by spontaneous emulsification. The formulations presented nanometric sizes, low polydispersity index, negative zeta potential, acid pH, rutin content (11 μg·mL -1 ), and encapsulation efficiency of 85%. Regarding the stability, the droplet size and PdI of the CPp-NE stored at refrigeration temperature in the dark, room temperature in the dark, and refrigeration temperature with light incidence were stable for 120 days and with no visible changes in the formulations. The antioxidant capacity was related to the reducing capacity, and the best results were found for nanoemulsions stored at room temperature and in absence of light. The potentiality of Phoma sp. inactive fungal biomass, waste from the bioherbicide production, was evaluated for the treatment of colored effluents containing Acid Red 18 (AR 18) dye. The batch experiments were performed to evaluate the following parameters: pH of the solution (2-10), dye concentration (50-200 mg L -1 ), adsorbent dose (0.5-2.5 g L -1 ), contact time (0-180 min) and temperature (298-328 K). The batch experiments using a synthetic dye solution revealed that Phoma sp. was efficient at pH of 2.0, 298 K and using a dosage of 1.25 g L -1 . The process was fast, being the equilibrium reached within 180 min. The maximum value of biosorption capacity was 63.58 mg g -1 , being the process favorable and exothermic. From the fixed bed assays, breakthrough curves were obtained, presenting a mass transfer zone of 7.08 cm and breakthrough time of 443 min. Phoma sp. was efficient to decolorize a simulated effluent, removing more than 90% of the color. From the obtained results, it can be concluded that Phoma sp. inactive biomass is a low-cost option to treat colored effluents in continuous and discontinuous biosorption modes. The application of lipases in liquid formulation instead of immobilized forms in the enzymatic biodiesel synthesis can make the process cost-efficient, more competitive, and sustainable. However, despite the benefits, the long reaction times required to achieve satisfactory yields is still a drawback of this biotechnological process. In this sense, employing the novel low-cost soluble NS40116 lipase, this paper proposes an innovative two-step hydroesterification reaction (TSHR) system as a technique of improving the reaction rate of an enzymatic biodiesel production. With the employment of two central composite statistical design to optimize the parameters of each of the reactions involved, the influence of the parameters "water concentration added to the reaction," "methanol-to-oil molar ratio," and "lipase load" on the process yield, besides the acid value of the samples, was investigated. After only 8 h of reaction, the highest fatty acid methyl ester yield reached was 97.1% with an acid value of 4.62 mg KOH g -1 utilizing a total of 8 wt% water, methanol-to-oil molar ratio of 6.3:1, and 0.70 wt% of lipase. Furthermore, the statistical models for both reactions indicated to be significant with 95% of reliability. Considering that the papers published using soluble lipases in a one-step batch process normally reach similar yields to those obtained in this research after 16 h to 24 h of reaction, the proposed system demonstrated to be a promising option of process configuration for the enzymatic production of biodiesel.

The potential of rice husks and straw as adsorbents after being processed by subcritical water hydrolysis (SWH) was investigated. The influences of temperature (453, 493 and 533 K) and liquid/solid ratio (7.5 and 15 g water/g biomass) on the rice straw and rice husks characteristics and on the adsorption capacity of 2-nitrophenol were evaluated at pH 4 and 7. Adsorption kinetics, equilibrium and thermodynamic parameters were also studied. The adsorption capacity was favored at pH 7. Pseudo-first-order model was suitable to predict the kinetic curves for 2-nitrophenol concentrations of 25, 50, 75 and 100 mg/L and the isotherm data obeyed the Freundlich model. Overall, the thermodynamic results revealed a spontaneous and exothermic process. The maximum adsorption capacity (92.97 ± 1.31 mg/g) was obtained for rice straw that has undergone an SWH at 453 K and 7.5 g water/g straw. This study is focused on the concentration of fermented broth from Phoma sp. to increase its herbicidal activity. For this purpose, biomolecules produced by submerged fermentation using Phoma sp. were concentrated by hollow fiber microfiltration membranes. The membrane feed was separated into two streams (retentate and permeate) and the crude broth was concentrated to 10, 30, 50, 70 and 90% (relative to the initial volume). The retentate samples were submitted to bioassays (triplicate) for evaluating their phytotoxic effects on five young leaves of species of Cucumis sativus and also on pre-emergence of weeds as Bidens pilosa and Amaranthus retroflexus . The highest herbicidal activity was 80.7% obtained for a concentration of 30% in the retentate fraction. At this condition, the bioherbicide presented severe damage symptoms on the detached leaves of Cucumis sativus if compared to the crude fermented broth. In the pre-emergence of B. pilosa and A. retroflexus , 100% control was obtained for assays performed in a germination chamber. For greenhouse assays using the substrate, the control rate of A. retroflexus was dependent of concentration of bioherbicide. The promising results achieved in the research with membrane separation process allow us to propose and develop further studies for evaluating this technology in the concentration of other metabolites produced by fermentation which also have bioherbicidal activity.

The prospection of bioherbicides has been an alternative to weed control, aiming at mitigating chemical risks to human, animal and environmental health due to extreme use of synthetic herbicides. In the present study, various fungi were isolated from plants with symptoms of fungal diseases for bioherbicide purposes against weeds ( Urochloa plantaginea , Euphorbia heterophylla and Bidens pilosa ). Fungi isolated were identified by molecular methods and enzymatic products obtained by fungi fermentation (cellulase, lipase, peroxidase, and amylase) were quantified. Bioherbicide selectivity study was performed on crops (soybean and corn), as well as on resistant weeds. Among the isolated fungi, Fusarium oxysporum , Fusarium ploriferatum, and Trichoderma koningiopsis presented bioherbicide potential. T. koningiopsis , in particular, presented the highest effect on Euphorbia heterophylla (popular name - Mexican fire plant), causing up to 60% of foliar damage, without presenting phytotoxicity against corn crop. New perspectives for weeds control and their use in corn crops were prospected, considering the bioherbicide selectivity described in this study.

Fungi are considered the most damaging microorganisms in agriculture. The indiscriminate use of chemical treatments in agricultural products causes the development of pest resistance and affects human health. An alternative to synthetic fungicides is the use of natural products such as plant extracts for the management of fungal diseases in plants. Extracts from different parts of Lupinus albescens (roots, stalks, leaves, and flowers) were obtained by extraction using supercritical carbon dioxide (CO 2 ) or compressed liquefied petroleum gas (LPG). Thereafter, the antioxidant activity of each extract was measured, and the antifungal activity in vitro of extracts was evaluated against Fusarium oxysporum and Fusarium verticillioides. For a concentration of 5000 mg/L, the half maximal inhibitory concentration (IC 50 ) ranged from 29.25 μg/mL to 192.96 μg/mL. Antifungal tests showed that all matrices presented inhibitory effect against both fungi tested. The extracts obtained from roots by CO 2 and LPG presented 70.1% and 65.1% inhibition against F. oxysporum, and 67.8% and 61.2% inhibition against F. verticillioides, respectively. These results suggest that the extracts obtained from L. albescens by extractions using supercritical CO 2 and compressed LPG might be a potential source of antioxidants and natural fungicides.

In the present study, it was presented a strategy to maximize the cutinase production by solid-state fermentation from different microorganisms and substrates. The best results were observed using Fusarium verticillioides, rice bran being the main substrate. Maximum yield of cutinase obtained by the strain was 16.22 U/g. For concentration, ethanol precipitation was used, and the purification factor was 2.4. The optimum temperature and pH for enzyme activity were 35 °C and 6.5, respectively. The enzyme was stable at a wide range of temperature and at all pH values tested. The concentrated cutinase was used as an adjuvant in a formulation containing cutinase + bioherbicide. The use of enzyme increased the efficiency of bioherbicide, since cutinase was responsible to remove/degrade the cutin that recovery the weed leaves and difficult the bioherbicide absorption. Cutinase showed to be a promising product to be used in formulation of bioherbicides.

This study evaluated the production of cellulolytic enzymes from different agricultural residues. The crude enzyme extract produced was characterized and applied for saccharification of some agricultural residues. Maximum cellulolytic activities were obtained using soybean hulls. All enzymatic activities were highly stable at 40 °C at a pH range of 4.5-5.5. For stability at low temperatures, the enzyme extract was stored at freezing temperature and cooling for about 290 days without major loss of activity. The K m values found for total cellulase (FPase), endoglucanase (CMCase), and xylanase were 19.73 mg ml -1 , 0.65 mg ml -1 , and 22.64 mg ml -1 , respectively, and V max values were 0.82 mol min -1  mg -1 , 0.62 mol min -1  mg -1 , and 104.17 mol min -1  mg -1 to cellulose, carboxymethyl cellulose, and xylan, respectively. In the saccharification tests, the total amount of total reducing sugars (TRS) released from 1 g of soybean hulls catalyzed by the enzymes present in the crude enzyme extract was 0.16 g g -1 dry substrate.

This work aimed at producing fermentable sugars and bioproducts from rice husks by subcritical water hydrolysis at 25 MPa in a semi-continuous mode. The influences of temperature (180 °C; 220 °C; 260 °C) and liquid/solid ratio (7.5 g water/g husks; 15 g water/g husks) on reducing sugar yield (Y RS ), efficiency (E), kinetic profiles (0-15 min), composition of sugars, inhibitors and organic acids, and physicochemical characteristics of the remaining solid material were evaluated and discussed in the work. The highest Y RS (18.0 ± 2.9 g/100 g husks) and E (39.5 ± 1.7 g sugars/100 g carbohydrates) were obtained at 220 °C and 7.5 gwater/g husks. In such condition, the hydrolyzed solutions presented cellobiose (18.0 g/L), xylose 17.7 g/L), arabinose (3.6 g/L), glucose (1.5 g/L), and levulinic acid (0.7 g/L). Biosurfactants have many advantages over synthetic surfactants but have higher production costs. Identifying microorganisms with high production capacities for these molecules and optimizing their growth conditions can reduce cost. The present work aimed to isolate and identify a fungus with high biosurfactant production capacity, optimize its growth conditions in a low cost culture medium, and characterize the chemical structure of the biosurfactant molecule. The fungal strain UFSM-BAS-01 was isolated from soil contaminated with hydrocarbons and identified as Fusarium fujikuroi. To optimize biosurfactant production, a Plackett-Burman design and a central composite rotational design were used. The variables evaluated were pH, incubation period, temperature, agitation and amount of inoculum in a liquid medium containing glucose. The partial structure of the biosurfactant molecule was identified by nuclear magnetic resonance spectrometry. F. fujikuroi reduced surface tension from 72 to 20mNm -1 under the optimized conditions of pH 5.0, 37°C and 7 days of incubation with 190rpm agitation. The partial identification of the structure of the biosurfactant demonstrated the presence of an α,β-trehalose. The present study is the first report of the biosynthesis of this compound by F. fujikuroi, suggesting that the biosurfactant produced belongs to the class of trehalolipids., (Copyright © 2018 Sociedade Brasileira de Microbiologia. The purpose of this review is to assist readers in understanding the importance of Lupinus albescens to nature, farmers, and scientists. L. albescens is mostly found in Argentina, Uruguay, Paraguay, and in "Campanha, Litoral and Missões" regions of State of Rio Grande do Sul (Brazil). Therefore, this review presents information and discussion on this plant that can encourage novel studies in a near future for exploring evermore the biological and physicochemical properties of L. albescens . The plant presents adaptive characteristics of soils with low content of nutrients, being an important plant for the recovering of degraded areas. In the last few years, there was an increase in scientific interest for exploring its chemical composition and biological activities. All plant matrices (i.e., roots, leaves, seeds, and stalks) are rich in antioxidant and antifungal compounds, especially stigmasterol. For example, the extracts obtained from the roots are reported with more than 50 wt% stigmasterol and 25 wt% ergosterol. This work investigates the use of blends of edible and nonedible raw materials as an alternative feedstock to fatty acid methyl esters (FAME) production through enzymatic catalysis. As biocatalyst, liquid lipase from Thermomyces lanuginosus (Callera™ Trans L), was used. Under reaction conditions of 35 °C, methanol to feedstock molar ratio of 4.5:1 and 1.45% of catalyst load, the best process performance was reached using 9% of water concentration in the medium-yield of 79.9% after 480 min of reaction. In terms of use of tallow mixed with soybean oil, the best yield was obtained when 100% of tallow was used in the process-84.6% after 480 min of reaction-behavior that was associated with the degree of unsaturation of the feedstock, something by that time, not addressed in papers of the area. The results show that tallow can be used as an alternative to FAME production, catalyzed by soluble lipase.

The separation of Jatropha curcas oil from azeotropes of ethyl alcohol-n-hexane and isopropyl alcohol-n-hexane using ceramic membranes with different cutoffs (5, 10 and 20 kDa) is presented. The mass ratios of oil:azeotropes (O:S) studied were 1:3 for feeding pressures of 0.1, 0.2 and 0.3 MPa, and 1:1 for the feeding pressure of 0.1 MPa. Isopropyl alcohol was the best solvent for the membranes conditioning to permeate n-hexane (240 kg/m 2  h). In the separation of J. curcas oil and azeotropes of solvents, both membranes showed oil retention and total flux decreases with time. Overall, the lowest decrease in the retentions was reached in the 5 kDa membrane, while the lowest decrease in the total flux was reached in the 20 kDa. In the separation of oil and ethyl alcohol-n-hexane azeotrope, the best retention at 60 min of the process was equal to 17.3 wt% in the 20 kDa membrane at 0.3 MPa and O:S ratio equalled to 1:3. In this condition, the total permeate flux was 17.5 kg/m 2  h. Different retentions and permeabilities are provided when changing the O:S ratio, the feeding pressure and the molecular weight cutoff of membranes.

In this study, a bioherbicide was produced by solid-state fermentation (SSF) using Diaporthe sp. Adjuvants were employed in a formulation to enhance the herbicidal activity towards the target (Cucumis sativus). The study was divided into two steps: (1) the fermentation condition for bioherbicide production was assessed; (2) evaluation of different formulations containing palm oil, Tween ® 80 and Span ® 80, in order to increase phytotoxicity. In step 1, the maximum herbicidal activity (1.23% of the leaves had lesions) was obtained at 25 °C, moisture content of 50 wt%, supplemented with 10 wt% of corn steep liquor and soybean bran and inoculum density of 15 wt%. In step 2, the formulation containing 8.2 wt% of palm oil, 8.2 wt% of Tween ® 80 and Span ® 80, resulting in an HLB of 12.8 showed the highest phytotoxicity on the leaves. At this condition, dry matter and height of target were reduced about 36% in comparison with control. Diaporthe sp. has the potential to produce molecules with herbicidal activity and the use of adjuvants enhanced three times its efficiency.

This work is focused on the optimization of the ultrasound-assisted extraction of antioxidant compounds with photoprotective effect from palm pressed fiber. The influence of ultrasound intensity and pulse cycle was investigated by means of a central composite rotational design. The optimized condition was ultrasound intensity of 120W.cm -2 and pulse factor of 0.4, yielding 3.24wt%. Compounds such as fatty acids, β-sitosterol, α-tocopherol, squalene, total phenolics and carotene were identified. The extract presented antioxidant activity towards synthetic (2,2-diphenyl-1-picrylhydrazyl - DPPH, 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) - ABTS) and biological radicals (Hydroxyl - OH), besides a sun protection factor of 15.01. Polar extracts from palm pressed fiber are promising candidates for use in cosmetic and pharmaceutical formulation since they present high antioxidant activity towards different radicals, combined with the high sun protection factor., (Copyright © 2016 Elsevier B.V. Production of a bioherbicide for biological control of weeds requires a series of steps, from selection of a suitable microbial strain to final formulation. Thus, this study aimed to select fungi for production of secondary metabolites with herbicidal activity using biological resources of the Brazilian Pampa biome. Phytopathogenic fungi were isolated from infected tissues of weeds in the Pampa biome. A liquid synthetic culture medium was used for production of metabolites. The phytotoxicity of fungal metabolites was assessed via biological tests using the plant Cucumis sativus L., and the most promising strain was identified by molecular analysis. Thirty-nine fungi were isolated, and 28 presented some phytotoxic symptoms against the target plant. Fungus VP51 belonging to the genus Diaporthe showed the most pronounced herbicidal activity. The objective of this work was to produce an herbicide by submerged fermentation in a stirred-tank bioreactor and to assess the potential herbicidal in pre-emergence, post-emergence, and in a detached leaves of Cucumis sativus var species. wisconsin (cucumber) and Sorghum bicolor (sorghum) species. Fermentations were carried out in a stirred-tank bioreactor with useful volume of 3L. Stirring rate (40, 50, and 60 rpm) and aeration (1, 2 and 3 vvm) were the variables studied for bioherbicide production. Fermented broth was fractioned with different solvents to identify the molecules produced by the fungus in a multi-dimensional gas chromatograph system. Bioherbicide showed 100% inhibition of germination of both species in the pre-emergence tests. From detached leaves tests were verified yellowish lesions in Cucumis sativus and necrotic lesions on leaves of Sorghum bicolor. Post-emergence test presented variation of the phytotoxicity from 25 to 66% for the species C. sativus and from 32 to 58% by S. bicolor. The metabolites produced by submerged fermentation of Phoma sp. presented activity in pre-emergence, post-emergence, and detached leaves of C. sativus and S. bicolor and it could be an alternative in the future for weed control.

Process modeling can lead to of advantages such as helping in process control, reducing process costs and product quality improvement. This work proposes a solid-state fermentation distributed parameter model composed by seven differential equations with seventeen parameters to represent the process. Also, parameters estimation with a parameters identifyability analysis (PIA) is performed to build an accurate model with optimum parameters. Statistical tests were made to verify the model accuracy with the estimated parameters considering different assumptions. The results have shown that the model assuming substrate inhibition better represents the process. It was also shown that eight from the seventeen original model parameters were nonidentifiable and better results were obtained with the removal of these parameters from the estimation procedure. Therefore, PIA can be useful to estimation procedure, since it may reduce the number of parameters that can be evaluated. FeWO4 particles were synthesized by a simple, rapid and facile microwave technique and their catalytic properties in heterogeneous photo-Fenton reaction were evaluated. This material was employed in the degradation of Amaranth azo dye. Individual and interactive effects of operational parameters such as pH, dye concentration and H2O2 dosage on the decolorization efficiency of Amaranth dye were evaluated by 2(3) central composite design. According to characterization techniques, a porous material and a well-crystallized phase of FeWO4 oxide were obtained. Regarding the photo-Fenton reaction assays, up to 97% color and 58% organic carbon removal were achieved in the best experimental conditions. In addition, the photo-Fenton process maintained treatment efficiency over five catalyst reuse cycles to indicate the durability of the FeWO4 catalyst. In summary, the results reveal that the synthesized FeWO4 material is a promising catalyst for wastewater treatment by heterogeneous photo-Fenton process.

In this study, the optimal conditions for production of cellulolytic enzymes by Trichoderma reesei NRRL-6156 using the solid-state fermentation were assessed in conical flasks and validated in a packed-bed bioreactor. Afterwards, the crude enzymatic extract obtained in the optimized condition was used for hydrolysis of sugarcane bagasse in water and ultrasound baths. The enzyme activities determined in this work were filter paper, exocellulase, endocellulase, and xylanase. The optimized condition for production was moisture content 68.6 wt% and soybean bran concentration 0.9 wt%. The crude enzymatic extract was applied for hydrolysis of sugarcane bagasse, being obtained 224.0 and 229 g kg(-1) at temperature of 43.4 °C and concentration of enzymatic extract of 18.6 % in water and ultrasound baths, respectively. The yields obtained are comparable to commercial enzymes.

Aiming to scale up and apply control and optimization strategies, currently is required the development of accurate plant models to forecast the process nonlinear dynamics. In this work, a mathematical model to predict the growth of the Kluyveromyces marxianus and temperature profile in a fixed-bed bioreactor for solid-state fermentation using sugarcane bagasse as substrate was built up. A parameter estimation technique was performed to fit the mathematical model to the experimental data. The estimated parameters and the model fitness were evaluated with statistical analyses. The results have shown the estimated parameters significance, with 95 % confidence intervals, and the good quality of process model to reproduce the experimental data.

The commercial inulinase obtained from Aspergillus niger was non-covalently immobilized on multiwalled carbon nanotubes (MWNT-COOH). The immobilization conditions for the carbon nanotubes were defined by the central composite rotational design (CCRD). The effects of enzyme concentration (0.8%-1.7% v/v) and adsorbent:adsorbate ratio (1:460-1:175) on the enzyme immobilization were studied. The adsorbent:adsorbate ratio variable has positive effect and the enzyme concentration has a negative effect on the inulinase immobilization (U/g) response at the 90% significance level. These results show that the lower the enzyme concentration and the higher the adsorbent:adsorbate ratio, better is the immobilization. According to the results, it is possible to observe that the carbon nanotubes present an effective inulinase adsorption. Fast adsorption in about six minutes and a loading capacity of 51,047 U/g support using a 1.3% (v/v) inulinase concentration and a 1:460 adsorbent:adsorbate ratio was observed. The effects of temperature on the immobilized enzyme activity were evaluated, showing better activity at 50 °C. The immobilized enzyme maintained 100% of its activity during five weeks at room temperature. The immobilization strategy with MWNT-COOH was defined by the experimental design, showing that inulinase immobilization is a promising biotechnological application of carbon nanotubes.

A solvothermal method was used to prepare zinc ferrite spinel oxide (ZnFe₂O₄) using ethylene glycol and 1,4 butanediol as solvent diols, and the influence of diols on the physical properties of ZnFe₂O₄ particles was investigated. The produced particles were characterized by X-ray powder diffraction (XRD), atomic force microscopy (AFM), Fourier transform infrared spectroscopy (FTIR) and nitrogen adsorption isotherms, and the catalytic activity for the organic pollutant decomposition by heterogeneous photo-Fenton reaction was investigated. Both solvents produced particles with cubic spinel structure. Microporous and mesoporous structures were obtained when ethylene glycol and 1,4 butanediol were used as diols, respectively. A higher pore volume and surface area, as well as a higher catalytic activity for the pollutant degradation were found when 1,4 butanediol was used as solvent.

Recent studies have shown that the chromatographic separation of mixtures of saccharides may be improved by making use of activated charcoal, a promising low cost material for the separation of sugars, including fructooligosaccharides. In this work, the development of a methodology to separate fructooligosaccharides from glucose, fructose and sucrose, using a fixed bed column packed with activated charcoal is proposed. The influence of temperature, eluant concentration and step gradients were evaluated to increase the separation efficiency and fructooligosaccharide purity. The final degree of fructooligosaccharide purification and separation efficiency were about 94% and 3.03 respectively, using ethanol gradient concentration ranging from 3.5% to 15% (v/v) at 40°C. The fixed bed column packed with the activated charcoal was shown to be a promising alternative for sugar separation, mainly those rich in fructooligosaccharides, leading to solutions of acceptable degrees of purification., (Copyright © 2014 Elsevier B.V. Fructooligosaccharides (FOS), a well-known prebiotic product, are obtained by enzymatic synthesis and consist of a mixture of mono- and disaccharides. In this work, a methodology for their separation and purification was developed using a zeolite fixed-bed column. The effects of column temperature (40-60°C), eluent flow rate (0.10-0.14 mL/min), injected to bed volume percent ratio (2.6-5.1%), and ethanol concentration in the eluent (40-60%, v/v) were investigated using a fractionary factorial design (2(4-1)), having the separation efficiency and purity as target responses. Additional experiments were performed as well, where the temperature and ethanol concentration were studied in a central composite design (2(2)). In this work, the zeolite fixed-bed column was shown to be a good alternative for FOS purification, allowing a FOS purity of 90% and separation efficiency of 6.86 between FOS and glucose, using an eluent at 45°C with 60% ethanol concentration., (© 2014 WILEY-VCH Verlag GmbH & Co. Papaya (Carica papaya L.) seeds were used as adsorbent to remove toxic pharmaceutical dyes (tartrazine and amaranth) from aqueous solutions, in order to extend application range. The effects of pH, initial dye concentration, contact time and temperature were investigated. The kinetic data were evaluated by the pseudo first-order, pseudo second-order and Elovich models. The equilibrium was evaluated by the Langmuir, Freundlich and Temkin isotherm models. It was found that adsorption favored a pH of 2.5, temperature of 298 K and equilibrium was attained at 180-200 min. The adsorption kinetics followed the pseudo second-order model, and the equilibrium was well represented by the Langmuir model. The maximum adsorption capacities were 51.0 and 37.4 mg g(-1) for tartrazine and amaranth, respectively. These results revealed that papaya seeds can be used as an alternative adsorbent to remove pharmaceutical dyes from aqueous solutions.
