1. Oxytocin promotes epicardial cell activation and heart regeneration after cardiac injury

Abstract

Cardiovascular disease (CVD) is one of the leading causes of mortality worldwide, and frequently leads to massive heart injury and the loss of billions of cardiac muscle cells and associated vasculature. Critical work in the last 2 decades demonstrated that these lost cells can be partially regenerated by the epicardium, the outermost mesothelial layer of the heart, in a process that highly recapitulates its role in heart development. Upon cardiac injury, mature epicardial cells activate and undergo an epithelial-mesenchymal transition (EMT) to form epicardium-derived progenitor cells (EpiPCs), multipotent progenitors that can differentiate into several important cardiac lineages, including cardiomyocytes and vascular cells. In mammals, this process alone is insufficient for significant regeneration, but it might be possible to prime it by administering specific reprogramming factors, leading to enhanced EpiPC function. Here, we show that oxytocin (OXT), a hypothalamic neuroendocrine peptide, induces epicardial cell proliferation, EMT, and transcriptional activity in a model of human induced pluripotent stem cell (hiPSC)-derived epicardial cells. In addition, we demonstrate that OXT is produced after cardiac cryoinjury in zebrafish, and that it elicits significant epicardial activation promoting heart regeneration. Oxytocin signaling is also critical for proper epicardium development in zebrafish embryos. The above processes are significantly impaired when OXT signaling is inhibited chemically or genetically through RNA interference. RNA sequencing data suggests that the transforming growth factor beta (TGF-β) pathway is the primary mediator of OXT-induced epicardial activation. Our research reveals for the first time an evolutionary conserved brain-controlled mechanism inducing cellular reprogramming and regeneration of the injured mammalian and zebrafish heart, a finding that could contribute to translational advances for the treatment of cardiac injuries.

Abstract

Rotating radio transients (RRATs) are neutron stars that emit detectable radio bursts sporadically. They are statistically distinct in the neutron star population, in many observable properties, but by their nature are practically difficult to study in depth. In this paper, we present the results from 1408 h of observations of RRAT candidates using the Irish station of the Low Frequency Array (LOFAR) at 150 MHz. As of October 2022, this census involved observing 113 sources, leading to 29 detections which were then followed up systematically. Single-pulse emission was detected from 25 sources, and periodic emission from 14 sources. 18 sources were found to have emission behaviour that is not discussed in prior works using LOFAR instruments. Four novel or modified source periods have been determined, ranging from 1.5-3.9 s, and 8 new or updated phase-coherent pulsar timing ephemerides have been produced using detected bursts. One unexpected single-pulse with a clearly-Galactic dispersion measure was detected as a part of this work, but has not been re-detected in follow-up observations. Observations are ongoing to expand the number of observed sources and further characterise and improve ephemerides for the detected sources. Abstract

Modern radio interferometers such as the LOw Frequency ARray (LOFAR) are capable of producing data at hundreds of gigabits to terabits per second. This high data rate makes the analysis of radio data cumbersome and computationally expensive. While high performance computing facilities exist for large national and international facilities, that may not be the case for instruments operated by a single institution or a small consortium. Data rates for next generation radio telescopes are set to eclipse those currently in operation, hence local processing of data will become all the more important. Here, we introduce the REAL-time Transient Acquisition backend (REALTA), a computing backend at the Irish LOFAR station (I-LOFAR) which facilitates the recording of data in near real-time and post-processing. Abstract

Background: No effective pharmacotherapy for acute respiratory distress syndrome (ARDS) exists, and mortality remains high. Preclinical studies support the efficacy of mesenchymal stem (stromal) cells (MSCs) in the treatment of lung injury. We aimed to test the safety of a single dose of allogeneic bone marrow-derived MSCs in patients with moderate-to-severe ARDS. Methods: The STem cells for ARDS Treatment (START) trial was a multicentre, open-label, dose-escalation, phase 1 clinical trial. Patients were enrolled in the intensive care units at University of California, San Francisco, CA, USA, Stanford University, Stanford, CA, USA, and Massachusetts General Hospital, Boston, MA, USA, between July 8, 2013, and Jan 13, 2014. Patients were included if they had moderate-to-severe ARDS as defined by the acute onset of the need for positive pressure ventilation by an endotracheal or tracheal tube, a PaO2:FiO2 less than 200 mm Hg with at least 8 cm H2O positive end-expiratory airway pressure (PEEP), and bilateral infiltrates consistent with pulmonary oedema on frontal chest radiograph. The first three patients were treated with low dose MSCs (1 million cells/kg predicted bodyweight [PBW]), the next three patients received intermediate dose MSCs (5 million cells/kg PBW), and the final three patients received high dose MSCs (10 million cells/kg PBW). Primary outcomes included the incidence of prespecified infusion-associated events and serious adverse events. The trial is registered with ClinicalTrials.gov, number NCT01775774. Findings: No prespecified infusion-associated events or treatment-related adverse events were reported in any of the nine patients. Serious adverse events were subsequently noted in three patients during the weeks after the infusion: one patient died on study day 9, one patient died on study day 31, and one patient was discovered to have multiple embolic infarcts of the spleen, kidneys, and brain that were age-indeterminate, but thought to have occurred before the MSC infusion based on MRI results. None of these severe adverse events were thought to be MSC-related. Interpretation: A single intravenous infusion of allogeneic, bone marrow-derived human MSCs was well tolerated in nine patients with moderate to severe ARDS. Based on this phase 1 experience, we have proceeded to phase 2 testing of MSCs for moderate to severe ARDS with a primary focus on safety and secondary outcomes including respiratory, systemic, and biological endpoints. Abstract

We describe and report first results from PALM-3000, the second-generation astronomical adaptive optics facility for the 5.1-m Hale telescope at Palomar Observatory. PALM-3000 has been engineered for high-contrast imaging and emission spectroscopy of brown dwarfs and large planetary mass bodies at near-infrared wavelengths around bright stars, but also supports general natural guide star use to V ~ 17. Using its unique 66 x 66 actuator deformable mirror, PALM-3000 has thus far demonstrated residual wavefront errors of 141 nm RMS under 1 arcsecond seeing conditions. PALM-3000 can provide phase conjugation correction over a 6.4 x 6.4 arcsecond working region at an observing wavelength of 2.2 microns, or full electric field (amplitude and phase) correction over approximately one half of this field. With optimized back-end instrumentation, PALM-3000 is designed to enable as high as 10e-7 contrast at ~1 arc second angular separation, after including post-observation speckle suppression processing. Abstract

Type-IIn supernovae (SNe), which are characterized by strong interaction of their ejecta with the surrounding circumstellar matter (CSM), provide a unique opportunity to study the mass-loss history of massive stars shortly before their explosive death. We present the discovery and follow-up observations of a Type IIn SN, PTF 09uj, detected by the Palomar Transient Factory (PTF). Serendipitous observations by GALEX at ultraviolet (UV) wavelengths detected the rise of the SN light curve prior to the PTF discovery. The UV light curve of the SN rose fast, with a time scale of a few days, to a UV absolute AB magnitude of about -19.5. Modeling our observations, we suggest that the fast rise of the UV light curve is due to the breakout of the SN shock through the dense CSM (n~10^10 cm^-3). Furthermore, we find that prior to the explosion the progenitor went through a phase of high mass-loss rate (~0.1 solar mass per year) that lasted for a few years. Abstract

Supernovae (SNe) are stellar explosions driven by gravitational or thermonuclear energy, observed as electromagnetic radiation emitted over weeks or more. In all known SNe, this radiation comes from internal energy deposited in the outflowing ejecta by either radioactive decay of freshly-synthesized elements (typically 56Ni), stored heat deposited by the explosion shock in the envelope of a supergiant star, or interaction between the SN debris and slowly-moving, hydrogen-rich circumstellar material. Here we report on a new class of luminous SNe whose observed properties cannot be explained by any of these known processes. These include four new SNe we have discovered, and two previously unexplained events (SN 2005ap; SCP 06F6) that we can now identify as members. These SNe are all ~10 times brighter than SNe Ia, do not show any trace of hydrogen, emit significant ultra-violet (UV) flux for extended periods of time, and have late-time decay rates which are inconsistent with radioactivity. Our data require that the observed radiation is emitted by hydrogen-free material distributed over a large radius (~10^15 cm) and expanding at high velocities (>10^4 km s^-1). These long-lived, UV-luminous events can be observed out to redshifts z>4 and offer an excellent opportunity to study star formation in, and the interstellar medium of, primitive distant galaxies., Comment: Accepted to Nature. Abstract

The Palomar Transient Factory (PTF) is a fully-automated, wide-field survey aimed at a systematic exploration of the optical transient sky. The transient survey is performed using a new 8.1 square degree camera installed on the 48-inch Samuel Oschin telescope at Palomar Observatory; colors and light curves for detected transients are obtained with the automated Palomar 60-inch telescope. PTF uses eighty percent of the 1.2-m and fifty percent of the 1.5-m telescope time. With an exposure of 60-s the survey reaches a depth of approximately 21.3 in g' and 20.6 in R (5 sigma, median seeing). Four major experiments are planned for the five-year project: 1) a 5-day cadence supernova search; 2) a rapid transient search with cadences between 90 seconds and 1 day; 3) a search for eclipsing binaries and transiting planets in Orion; and 4) a 3-pi sr deep H-alpha survey. PTF provides automatic, realtime transient classification and follow up, as well as a database including every source detected in each frame. This paper summarizes the PTF project, including several months of on-sky performance tests of the new survey camera, the observing plans and the data reduction strategy. Abstract

n this study, we present the results of nitrogen deposition on land from a set of 29 simulations from six different tropospheric chemistry models pertaining to present-day and 2100 conditions. Nitrogen deposition refers here to the deposition (wet and dry) of all nitrogen-containing gas phase chemical species resulting from NOx (NO + NO2) emissions. We show that under the assumed IPCC SRES A2 scenario the global annual average nitrogen deposition over land is expected to increase by a factor of ∼2.5, mostly because of the increase in nitrogen emissions. This will significantly expand the areas with annual average deposition exceeding 1 gN/m2/year. Using the results from all models, we have documented the strong linear relationship between models on the fraction of the nitrogen emissions that is deposited, regardless of the emissions (present day or 2100). On average, approximately 70% of the emitted nitrogen is deposited over the landmasses. For present-day conditions the results from this study suggest that the deposition over land ranges between 25 and 40 Tg(N)/year. By 2100, under the A2 scenario, the deposition over the continents is expected to range between 60 and 100 Tg(N)/year. Over forests the deposition is expected to increase from 10 Tg(N)/year to 20 Tg(N)/year. In 2100 the nitrogen deposition changes from changes in the climate account for much less than the changes from increased nitrogen emissions.

Abstract

Transfusion of red blood cells (RBCs) is one of the most valuable and widespread treatments in modern medicine. Lifesaving RBC transfusions are facilitated by the cold storage of RBC units in blood banks worldwide. Currently, RBC storage and subsequent transfusion practices are performed using simplistic workflows. More specifically, most blood banks follow the “first-in-first-out” principle to avoid wastage, whereas most healthcare providers prefer the “last-in-first-out” approach simply favoring chronologically younger RBCs. Neither approach addresses recent advances through -omics showing that stored RBC quality is highly variable depending on donor-, time-, and processing-specific factors. Thus, it is time to rethink our workflows in transfusion medicine taking advantage of novel technologies to perform RBC quality assessment. We imagine a future where lab-on-a-chip technologies utilize novel predictive markers of RBC quality identified by -omics and machine learning to usher in a new era of safer and precise transfusion medicine.

Abstract

Ambition to leverage the power of nature-based solutions (NbS) to address global challenges such as the interlinked climate change and biodiversity crises has reached unprecedented levels. Despite a growing consensus around the potential benefits of NbS, this ambition has not translated into sufficient actionable progress in practice. Cities, increasingly recognized as centres of transformative capacity, have become a focal point for achieving NbS ambitions. Nevertheless, the transition from theoretical discourse to practical implementation has been slow, with NbS frequently being overlooked until recently in political agendas. A key obstacle to progress is the lack of action-oriented research grounding theoretical discussions in empirical evidence on NbS policy design and application. This paper seeks to address these knowledge gaps by: (i) defining criteria for good practices in urban NbS policy design and implementation; (ii) assessing policy instruments from the Urban Governance Atlas (UGA) against these criteria using mixed statistical and qualitative methods; and (iii) identifying potential pathways and actionable measures to promote the adoption of good practices in practice. Drawing on the diverse UGA examples, with a particular focus on Europe and the CELAC region, the paper identifies seven good practice criteria and outlines four potential pathways and related actions to guide the development of NbS policy instruments across varied urban contexts. Concrete recommendations around actionable knowledge aim to tap the transformative potential of NbS in addressing pressing global issues.

Abstract

This technical report describes the third phase of a three-phase project to develop standards for training corrections personnel. (The first two phases involved a statewide job analysis of corrections officer, juvenile counselor, and probation officer jobs in California corrections agencies and development and validation of procedures for candidate selection.) The objectives of the third phase were to identify the knowledge and skills necessary for effective performance in the three occupations and to prepare models of basic training courses. This report is organized in five sections. Section 1 presents an overview of the systems approach used in the study. Section 2 describes the training needs analyses that were conducted to identify the knowledge and skills needed to perform the core tasks in the three occupations. Section 3 describes how the results of the needs analyses were used to develop performance objectives for the three basic training courses. Section 4 presents the purpose and content of the basic training course manuals, and section 5 provides recommendations for the preemployment training and continuing education of corrections officers, juvenile counselors, and probation officers. Abstract

The present study is the first to examine whether ongoing parental romantic relationship quality, as well as the adult children's relationship quality with their parents, are uniquely associated with adult children's romantic relationship dynamics. We use dyadic data from 3,241 families to assess associations between expressions of emotional intimacy and admiration within parents' romantic relationships, adult children's romantic relationships, and adult children's relationships with their parents. Structural equation modeling showed that ongoing parental romantic responsible actions and parent–adult child relationships are both uniquely associated with responsible actions within adult children's romantic relationships. Abstract

While epigenetic mechanisms such as DNA methylation and histone modification are known to be important for gene suppression, relatively little is still understood about the interplay between these systems. The UHRF1 protein can interact with both DNA methylation and repressive chromatin marks, but its primary function in humans has been unclear. To determine what that was, we first established stable UHRF1 knockdowns (KD) in normal, immortalized human fibroblasts using targeting shRNA, since CRISPR knockouts (KO) were lethal. Although these showed a loss of DNA methylation across the whole genome, transcriptional changes were dominated by the activation of genes involved in innate immune signalling, consistent with the presence of viral RNA from retrotransposable elements (REs). We confirmed using mechanistic approaches that 1) REs were demethylated and transcriptionally activated; 2) this was accompanied by activation of interferons and interferon-stimulated genes and 3) the pathway was conserved across other adult cell types. Restoring UHRF1 in either transient or stable KD systems could abrogate RE reactivation and the interferon response. Notably, UHRF1 itself could also re-impose RE suppression independent of DNA methylation, but not if the protein contained point mutations affecting histone 3 with trimethylated lysine 9 (H3K9me3) binding. Our results therefore show for the first time that UHRF1 can act as a key regulator of retrotransposon silencing independent of DNA methylation. [ABSTRACT FROM AUTHOR]

Abstract

It is well known that student-athletes are more diverse and higher academic achievers, on average, than their collegiate peers. However, little research has explored the mentoring relationships of student-athletes who conduct undergraduate research. Unique challenges and opportunities exist for student-athletes and faculty when it comes to mentoring undergraduate research in fields like science, technology, engineering and math (STEM). Using a collaborative autoethnographic methodological approach and apprenticeship theory, our personal narratives detail how an engineering professor mentored a former engineering student-athlete for undergraduate engineering education research. We offer several findings and recommendations.
