Abstract

Deciphering the complex cellular behaviours and advancing the biotechnology applications of filamentous fungi increase the requirement for genetically manipulating a large number of target genes. The current strategies cannot cyclically coedit multiple genes simultaneously. In this study, we firstly revealed the existence of diverse homologous recombination (HR) types in marker-free editing of filamentous fungi, and then, demonstrated that sgRNA efficiency-mediated competitive inhibition resulted in the low integration of multiple genetic sites during coediting, which are the two major obstacles to limit the efficiency of cyclically coediting of multiple genes. To overcome these obstacles, we developed a biased cutting strategy by Cas9 to greatly enhance the desired HR type and applied a new selection marker labelling strategy for multiple donor DNAs, in which only the donor DNA with the lowest sgRNA efficiency was labelled. Combined with these strategies, we successfully developed a convenient method for cyclically coediting multiple genes in different filamentous fungi. In addition, diverse HRs resulted in a useful and convenient one-step approach for gene functional study combining both gene disruption and complementation. This research provided both a useful one-step approach for gene functional study and an efficient strategy for cyclically coediting multiple genes in filamentous fungi.

17. Root Growth and Organic Acid Secretion of Tea Plants Affected by Phosphorus and Aluminum Interaction.

Abstract

To investigate the effects of phosphorus-aluminum interaction on the growth of tea plants, phosphorus and aluminum interaction treatments with three aluminum concentrations and five phosphorus concentrations were set to analyze the changes in root growth, secretion of organic acids and uptake of phosphorus and aluminum in tea plants. The results show that both low phosphorus (0.01 mmol·L-1) and high aluminum (1 mmol·L-1) significantly promoted the growth of new roots in tea plants, and the number of root tips, root length, average diameter and increase of dry matter in new roots reached the maximum under the combined treatment of low phosphorus and high aluminum. High aluminum was able to restore the growth of new roots impeded by high phosphorus (0.5 mmol·L-1) conditions. In addition to the high phosphorus treatment (0.5 mmol·L-1), increasing the concentration of phosphorus or aluminum in the environment could significantly promote their accumulation in the roots of tea plants. Within a certain range, phosphorus significantly promoted the accumulation of aluminum in shoots. However, aluminum inhibited the accumulation of phosphorus in shoots when phosphorus was sufficient (>0.05 mmol·L-1). When phosphorus was sufficient, high aluminum promoted the secretion of oxalic acid, malic acid and citric acid. Phosphorus application promoted the secretion of citric acid, while low phosphorus promoted the secretion of oxalic acid and malic acid. Low phosphorus and high aluminum synergistically promoted the secretion of malic acid. The results of two-way ANOVA show that phosphorus and aluminum concentrations and their interaction had highly significant effects on root length, root tip number, phosphorus and aluminum uptake and organic acid secretion in tea plants (p<0.01), which showed that phosphorus and aluminum interaction can affect root growth and organic acid secretion of tea plants significantly. 19. Production of biogenic copper oxide nanoparticles using paddy straw and an evaluation of its effects onCamellia sinensis

Abstract

The present investigation focused to produce the biogenic copper oxide nanoparticles and an assessment of its impact on Camellia sinensis. The biogenic copper oxide nanoparticles were synthesized using the extract of paddy straw and characterized by various standard techniques. Synthesized particles are spherical with a size of 24 ± 5 nm. Different concentrations of copper oxide nanoparticles were supplied to the tea plants and control tea plants were maintained. Copper oxide nanoparticles affect the level of photosynthetic pigments (chlorophylls and carotenoids), antioxidant enzymes, total protein, total flavonoids, and total amino acids. The assays determined that the high concentrations of biogenic copper oxide nanoparticles (400 mg/L) induce the oxidative stress in tea plants. However, the lower concentration of copper oxide nanoparticles (100 to 300 mg/L) did not alter the level of biochemical content of tea plants and it did not induce any changes in oxidative status of tea plants. The study concludes that higher concentration of copper oxide nanoparticles increases the ROS generation which decreases the level of biochemical components and meanwhile, the lower concentration of copper oxide nanoparticles did not show any negative impacts on the tea plants. Our results revealed the optimum concentrations of the nanoparticles are very essential to improve the plant growth.

25. Mycorrhizal Colonization Enhanced Sorghum bicolorTolerance under Soil Water Deficit Conditions by Coordination of Proline and Reduced Glutathione (GSH)

Abstract

Drought stress is an important limiting factor in crop production. Arbuscular mycorrhizal fungi (AMF) enhance plant drought tolerance through antioxidant activities. However, the coordination of nonenzymatic antioxidants against drought remains unclear. Here, we investigated the AMF symbiosis in drought tolerance of Sorghum bicolorby increasing proline and reducing glutathione (GSH). Glomus mosseaeinoculation increased grain yield, biochemical content, and bioactivities of millets. Under drought conditions, seedlings inoculated with G. mosseaehad higher SOD, POD, CAT, PPO, proline, and GSH activities compared to noninoculated controls. Meanwhile, a lower accumulation of MDA and H2O2was observed in the G. mosseaeseedlings. Furthermore, genes attributed to nonenzymatic antioxidants, such as GST29, P5CS2, FD3, GST, and GAD, were significantly up-regulated by G. mosseaeunder drought conditions. In conclusion, G. mosseaeinoculation enhanced the drought tolerance of S. bicolorby improving reactive oxygen species (ROS) scavengers, including proline and GSH, that regulate ROS production and prevent oxidative damage.
