Conservation management can be aided by knowledge of genetic diversity and evolutionary history, so that ecological and evolutionary processes can be preserved. The Button Wrinklewort daisy (Rutidosis leptorrhynchoides) was a common component of grassy ecosystems in south-eastern Australia. It is now endangered due to extensive habitat loss and the impacts of livestock grazing, and is currently restricted to a few small populations in two regions >500 km apart, one in Victoria, the other in the Australian Capital Territory and nearby New South Wales (ACT/NSW). Using a genome-wide SNP dataset, we assessed patterns of genetic structure and genetic differentiation of 12 natural diploid populations. We estimated intrapopulation genetic diversity to scope sources for genetic management. Bayesian clustering and principal coordinate analyses showed strong population genetic differentiation between the two regions, and substantial substructure within ACT/NSW. A coalescent tree-building approach implemented in SNAPP indicated evolutionary divergence between the two distant regions. Among the populations screened, the last two known remaining Victorian populations had the highest genetic diversity, despite having among the lowest recent census sizes. A maximum likelihood population tree method implemented in TreeMix suggested little or no recent gene flow except potentially between very close neighbours. Populations that were more genetically distinctive had lower genetic diversity, suggesting that drift in isolation is likely driving population differentiation though loss of diversity, hence re-establishing gene flow among them is desirable. These results provide background knowledge for evidence-based conservation and support genetic rescue within and between regions to elevate genetic diversity and alleviate inbreeding.

Many studies have quantified the distribution of heterozygosity and relatedness in natural populations, but few have examined the demographic processes driving these patterns. In this study, we take a novel approach by studying how population structure affects both pairwise identity and the distribution of heterozygosity in a natural population of the self-incompatible plant Antirrhinum majus. Excess variance in heterozygosity between individuals is due to identity disequilibrium, which reflects the variance in inbreeding between individuals; it is measured by the statistic g2. We calculated g2 together with FST and pairwise relatedness (Fij) using 91 SNPs in 22,353 individuals collected over 11 years. We find that pairwise Fij declines rapidly over short spatial scales, and the excess variance in heterozygosity between individuals reflects significant variation in inbreeding. Additionally, we detect an excess of individuals with around half the average heterozygosity, indicating either selfing or matings between close relatives. We use 2 types of simulation to ask whether variation in heterozygosity is consistent with fine-scale spatial population structure. First, by simulating offspring using parents drawn from a range of spatial scales, we show that the known pollen dispersal kernel explains g2. Second, we simulate a 1,000-generation pedigree using the known dispersal and spatial distribution and find that the resulting g2 is consistent with that observed from the field data. In contrast, a simulated population with uniform density underestimates g2, indicating that heterogeneous density promotes identity disequilibrium. Our study shows that heterogeneous density and leptokurtic dispersal can together explain the distribution of heterozygosity.

Summary Understanding the mechanisms causing phenotypic differences between females and males has long fascinated evolutionary biologists. An extensive literature exists on animal sexual dimorphism but less information is known about sex differences in plants, particularly the extent of geographical variation in sexual dimorphism and its life‐cycle dynamics.Here, we investigated patterns of genetically based sexual dimorphism in vegetative and reproductive traits of a wind‐pollinated dioecious plant, Rumex hastatulus, across three life‐cycle stages using open‐pollinated families from 30 populations spanning the geographic range and chromosomal variation (XY and XY1Y2) of the species.The direction and degree of sexual dimorphism was highly variable among populations and life‐cycle stages. Sex‐specific differences in reproductive function explained a significant amount of temporal change in sexual dimorphism. For several traits, geographical variation in sexual dimorphism was associated with bioclimatic parameters, likely due to the differential responses of the sexes to climate. We found no systematic differences in sexual dimorphism between chromosome races.Sex‐specific trait differences in dioecious plants largely result from a balance between sexual and natural selection on resource allocation. Our results indicate that abiotic factors associated with geographical context also play a role in modifying sexual dimorphism during the plant life‐cycle.

Many recent studies have addressed the mechanisms operating during the early stages of speciation, but surprisingly few studies have tested theoretical predictions on the evolution of strong reproductive isolation (RI). To help address this gap, we first undertook a quantitative review of the hybrid zone literature for flowering plants in relation to reproductive barriers. Then, usingPopulusas an exemplary model group, we analysed genome-wide variation for phylogenetic tree topologies in both early- and late-stage speciation taxa to determine how these patterns may be related to the genomic architecture of RI. Our plant literature survey revealed variation in barrier complexity and an association between barrier number and introgressive gene flow. Focusing onPopulus,our genome-wide analysis of tree topologies in speciating poplar taxa points to unusually complex genomic architectures of RI, consistent with earlier genome-wide association studies. These architectures appear to facilitate the ‘escape’ of introgressed genome segments from polygenic barriers even with strong RI, thus affecting their relationships with recombination rates. Placed within the context of the broader literature, our data illustrate how phylogenomic approaches hold great promise for addressing the evolution and temporary breakdown of RI during late stages of speciation.This article is part of the theme issue ‘Towards the completion of speciation: the evolution of reproductive isolation beyond the first barriers'.

Sexual dimorphism in resource allocation is expected to change during the life cycle of dioecious plants because of temporal differences between the sexes in reproductive investment. Given the potential for sex-specific differences in reproductive costs, resource availability may contribute to variation in reproductive allocation in females and males. Here, we used Rumex hastatulus, a dioecious, wind-pollinated annual plant, to investigate whether sexual dimorphism varies with life-history stage and nutrient availability, and determine whether allocation patterns differ depending on reproductive commitment. To examine if the costs of reproduction varied between the sexes, reproduction was either allowed or prevented through bud removal, and biomass allocation was measured at maturity. In a second experiment to assess variation in sexual dimorphism across the life cycle, and whether this varied with resource availability, plants were grown in high and low nutrients and allocation to roots, aboveground vegetative growth and reproduction were measured at three developmental stages. Males prevented from reproducing compensated with increased above- and belowground allocation to a much larger degree than females, suggesting that male reproductive costs reduce vegetative growth. The proportional allocation to roots, reproductive structures and aboveground vegetative growth varied between the sexes and among life-cycle stages, but not with nutrient treatment. Females allocated proportionally more resources to roots than males at peak flowering, but this pattern was reversed at reproductive maturity under low-nutrient conditions. Our study illustrates the importance of temporal dynamics in sex-specific resource allocation and provides support for high male reproductive costs in wind-pollinated plants.

†Background and Aims Populations of dioecious flowering plants commonly exhibit heterogeneity in sex ratios and deviations from the equilibrium expectation of equal numbers of females and males. Yet the role of ecological and demographic factors in contributing towards biased sex ratios is currently not well understood. †Methods Species-level studies from the literature were analysed to investigate ecological correlates of amongpopulation sex-ratio variation and metapopulation models and empirical data were used to explore the influence of demography and non-equilibrium conditions on flowering sex ratios. †Key Results The survey revealed significant among-population heterogeneity in sex ratios and this was related to the degree of sampling effort. For some species, sex-ratio bias was associated with the proportion of non-reproductive individuals, with greater male bias in populations with a lower proportion of individuals that were flowering. Male-biased ratios were also found at higher altitudes and latitudes, and in more xeric sites. Simulations and empirical data indicated that clonal species exhibited greater heterogeneity in sex ratios than non-clonal species as a result of their slower approach to equilibrium. The simulations also indicated the importance of interactions between reproductive mode and founder effects, with greater departures from equilibrium in clonal populations with fewer founding individuals. †Conclusions The results indicate that sex-based differences in costs of reproduction and non-equilibrium conditions can each play important roles in affecting flowering sex ratios in populations of dioecious plants.

Dioecious plant species commonly exhibit deviations from the equilibrium expectation of 1:1 sex ratio, but the mechanisms governing this variation are poorly understood. Here, we use comparative analyses of 243 species, representing 123 genera and 61 families to investigate ecological and genetic correlates of variation in the operational (flowering) sex ratio. After controlling for phylogenetic nonindependence, we examined the influence of growth form, clonality, fleshy fruits, pollen and seed dispersal vector, and the possession of sex chromosomes on sex-ratio variation. Male-biased flowering sex ratios were twice as common as female-biased ratios. Male bias was associated with long-lived growth forms (e.g., trees) and biotic seed dispersal and fleshy fruits, whereas female bias was associated with clonality, especially for herbaceous species, and abiotic pollen dispersal. Female bias occurred in species with sex chromosomes and there was some evidence for a greater degree of bias in those with heteromorphic sex chromosomes. Although the role of interactions among these correlates require further study, our results indicate that sex-based differences in costs of reproduction, pollen and seed dispersal mechanisms and sex chromosomes can each play important roles in affecting flowering sex ratios in dioecious plants.

The primary goal of restoration is to create self-sustaining ecological communities that are resilient to periodic disturbance. Currently, little is known about how restored communities respond to disturbance events such as fire and how this response compares to remnant vegetation. Following the 2003 fires in south-eastern Australia we examined the post-fire response of revegetation plantings and compared this to remnant vegetation. Ten burnt and 10 unburnt (control) sites were assessed for each of three types of vegetation (direct seeding revegetation, revegetation using nursery seedlings (tubestock) and remnant woodland). Sixty sampling sites were surveyed 6 months after fire to quantify the initial survival of mid- and overstorey plant species in each type of vegetation. Three and 5 years after fire all sites were resurveyed to assess vegetation structure, species diversity and vigour, as well as indicators of soil function. Overall, revegetation showed high (>60%) post-fire survival, but this varied among species depending on regeneration strategy (obligate seeder or resprouter). The native ground cover, mid- and overstorey in both types of plantings showed rapid recovery of vegetation structure and cover within 3 years of fire. This recovery was similar to the burnt remnant woodlands. Non-native (exotic) ground cover initially increased after fire, but was no different in burnt and unburnt sites 5 years after fire. Fire had no effect on species richness, but burnt direct seeding sites had reduced species diversity (Simpson's Diversity Index) while diversity was higher in burnt remnant woodlands. Variation in pollination intensity can influence the degree of gametophytic competition with diverse consequences for plant fitness. Such effects should be particularly important in wind-pollinated plants because they commonly possess uniovulate flowers. In several dioecious species, competition between female- and male-determining microgametophytes has been proposed as a cause of female-biased sex ratios. Here, we examine functional relations among pollination intensity, seed set, sex ratios, and sporophyte vigor in Rumex hastatulus, a wind-pollinated, dioecious plant. Using controlled pollinations, we applied a range of pollen grain numbers (1–71) to female flowers. Pollination intensity significantly predicted percent seed set, which was >80% when more than 15 pollen grains were added to stigmas. We estimated that the proportion of pollen grains participating in fertilization was 0.28 and the proportion of fertilized ovules developing into seed was 0.89. Variation in the timing of reproductive functions in dioecious organisms may result in adaptive changes in the direction of sexual dimorphism during the breeding season. For plants in which both pollen and seeds are wind-dispersed, it may be advantageous for male plants to be taller when pollen is dispersed and female plants to be taller when seeds are dispersed. We examined the dynamics of height dimorphism in Rumex hastatulus , an annual, wind-pollinated, dioecious plant from the southern USA. A field survey of seven populations indicated that females were significantly taller than males at seed maturity. However, a glasshouse experiment revealed a more complex pattern of height growth during the life cycle. No dimorphism was evident prior to reproduction for six of seven populations, but at flowering, males were significantly taller than females in all populations. This pattern was reversed at reproductive maturity, consistent with field observations. Males flowered later than females and the degree of height dimorphism was greater in populations with a later onset of male flowering. We discuss the potential adaptive significance of temporal changes in height dimorphism for pollen and seed dispersal, and how this may be optimized for the contrasting reproductive functions of the sexes.

In many angiosperm species, populations are reproductively subdivided into distinct sexual morphs including females, males and hermaphrodites. Sexual polymorphism is maintained by frequency-dependent selection, leading to predictable sex ratios at equilibrium. Charles Darwin devoted much of his book ‘The Different Forms of Flowers on Plants of the Same Species’ (1877) to investigating plant sexual polymorphisms and laid the foundation for many problems addressed today by integrating theory with empirical studies of the demography and genetics of populations. Here, we summarize our recent work on the ecological and genetic mechanisms influencing variation in sex ratios and their implications for evolutionary transitions among sexual systems. We present the results of a survey of sex ratios from 126 species from 47 angiosperm families and then address two general problems using examples from diverse angiosperm taxa: (i) the mechanisms governing biased sex ratios in dioecious species; (ii) the origins and maintenance of populations composed of females, males and hermaphrodites. Several themes are emphasized, including the importance of non-equilibrium conditions, the role of life history and demography in affecting sex ratios, the value of theory for modelling the dynamics of sex ratio variation, and the utility of genetic markers for investigating evolutionary processes in sexually polymorphic plant populations.

Summary 1. The role of genetic factors in species decline and extinction is the subject of a long-running controversy, with demographical factors often seen as more important for the immediate persistence of populations and species. One gene system that directly links genetic diversity with reproduction, through its influence on fertilization success, is the self-incompatibility (SI) locus in angiosperm plants. Despite the potential importance of SI allele diversity for demographical function, there are few direct measures of S-allele numbers in multiple populations, and no studies have simultaneously measured reproductive output, mate availability, S-allele diversity and pollinator service. 2. We used diallel crosses to estimate S-allele number and mate availability in seven populations of the forb Rutidosis leptorrhynchoides ranging in size from 5 to 100 000 plants. Seed set and correlated paternity were assessed from open-pollinated inflorescences to examine reproductive output and variance in female and male fitness. To assess whether populations were receiving adequate pollinator service, the amount of pollen deposited on open-pollinated stigmas was quantified. 3. We found that small populations ( 1000 plants) with higher numbers of S-alleles, despite the maintenance of pollinator service. Greater variance in seed set among plants and higher correlated paternity in small populations suggest that low S-allele numbers are directly limiting mate availability and causing the observed reproductive failure. 4. Synthesis and applications. Our work demonstrates that the loss of genetic diversity at the SI locus can have immediate detrimental effects on reproductive performance, despite the maintenance of pollinator service. These results highlight the importance of genetic factors in determining demographical outcomes when clear links between genotype and individual fitness exist, even when populations are not ecologically constrained. This result is important for the conservation and management of many plant species as approximately half of all angiosperm families contain species with genetically controlled SI and the loss of genetic diversity through habitat loss and fragmentation is an ever-increasing reality for plant species worldwide.

Through identifying and understanding ecologically important dimensions of plant trait variation we gain insight into why particular trait combinations are favoured and into the implications of trait differences among species. Here, we describe relationships among several poorly understood leaf and stem traits across species from several Australian vegetation types. Species with lower wood density (WD) consistently deployed more leaf area per unit shoot mass (LA/SM), as did the larger-leaved species within forested sites. Higher LA/SM is likely to lead to faster growth rates, implying a previously unrecognized implication to interspecific variation in leaf size and WD. Leaf : sapwood area ratio is one of several important traits contributing to a plant's water-use strategy, yet, we still only poorly understand how plants vary in the extent to which hydraulic properties and traits such as leaf size, WD and LM/SM are coordinated, and what the implications of this variation may be.

Summary 1The leaf size–twig size spectrum is an important spectrum of variation between species, although the costs and benefits of larger vs smaller leaf size are poorly understood. This study quantified the dry mass costs of deploying leaf area in relation to leaf size, across 70 species from four sites contrasted on the basis of rainfall and soil nutrients in east-temperate Australia. 2Leaf mass fraction beyond 10 mm2 of conductive cross-section (LMF10) varied threefold and was strongly positively correlated with leaf size, both across all species and within each habitat. This and other key correlations were significant both across species and as evolutionary divergences. 3An LMF10 advantage for larger-leaved species should translate into a proportional advantage in dry mass acquisition, yet species with small leaves persist and sustain populations. This raises the question as to what factors might counterbalance an LMF advantage associated with larger leaf size. 4Within some sites, specific leaf area (SLA) decreased with leaf size, which counterbalanced the LMF10 advantage. Within other sites, and across all sites, SLA did not decline with leaf size. In these comparisons other factors, such as avoidance of overheating, must counterbalance the LMF10 advantage to larger-leaved species. 5Wood density was negatively correlated with leaf size. However, the relationship was weaker within sites in the lower rainfall zone, and species in these sites had generally higher wood density. Sapwood cross-sectional area per unit leaf area (Huber value) varied approximately ninefold across all species, and was correlated with both leaf size and SLA across all species, but not within sites.

Seed germination is dependent on the interaction between the dormancy state of a seed and the presence of favourable environmental conditions. Thus, the spectacular pulse of seedling recruitment in many Australian vegetation communities following disturbances such as fire can be attributed to changes in microsite conditions and/ or the dormancy-breaking effect of the disturbance on accumulated seed banks. Grevillea rivularis is a threatened species endemic to the area immediately above Carrington Falls in the NSW Southern Highlands. Most of the population is confined to the riparian vegetation zone in woodland and heath, and is therefore subject to periodic disturbance from fire and flood. For this species, a pulse of seedling recruitment has been recorded after fire, flood and mechanical soil disturbance. The aims of this study were to examine the density and vertical distribution of the soil-stored seed bank and to investigate the role of heat and scarification as cues for germination of fresh and soil-stored seed. There was a large seed bank under the canopies of established individuals (194 ± 73 seeds m -2 ) and most seeds were found in the 0-2 cm and leaf-litter layers of the soil profile. The germination response of soil-stored and fresh seed was examined using a hierarchical series of laboratory experiments. Seeds of G. rivularis showed marked dormancy polymorphism. Thirty-six percent of soil-stored seed germinated without treatment, whereas no untreated fresh seeds germinated. Scarification or heating caused significant germination of dormant soil-stored seed, but only scarification resulted in germination of dormant fresh seeds. These results highlight important differences in the dormancy state of soil-stored and fresh seed. Thus, being a riparian species in a fire-prone environment, the dormancy mechanisms in seeds of G. rivularis suit this species to disturbance by both fire and flood.

Self-incompatibility systems function to prevent inbreeding, and work effectively in large, genetically diverse populations. However, a decrease in population size can reduce genetic diversity at the self-incompatibility locus, which leads to a reduction in mate availability and has important demographic implications for small populations. Currently, little is known about the response of self-incompatible polyploid species to a reduction in population size. In Rutidosis leptorrhynchoides there was a significant decrease in the within-population probability of fertilization with a decline in population size for diploid populations and a marginally significant relationship for tetraploid populations, suggesting that in small populations of both chromosome races fertilization success is reduced due to a decrease in self-incompatibility allele (S-allele) diversity. There was no significant difference between the slopes of the fertility-population size relationship for diploid and tetraploid populations which indicates a similar rate of decline in fertilization success with population size for both chromosome races. Fertilization success increased when crosses were undertaken between populations and this was significantly related to population size for diploid and tetraploid populations, indicating that small populations gain the greatest benefit to fertilization success from crossing between populations. For tetraploid populations the benefits of crossing between populations tended to decline more rapidly with increasing population size. These results suggest that for small populations that have reduced fertilization success, genetic rescue by introducing new genetic material from other populations is an important means of ameliorating mate limitation issues associated with reduced S-allele diversity in both diploid and tetraploid races.

Vegetation clearing and land-use change have depleted many natural plant communities to the point where restoration is required. A major impediment to the success of rebuilding complex vegetation communities is having regular access to sufficient quantities of high-quality seed. Seed-production areas (SPAs) can help generate this seed, but these must be underpinned by a broad genetic base to maximise the evolutionary potential of restored populations. However, genetic bottlenecks can occur at the collection, establishment and production stages in SPAs, requiring genetic evaluation. This is especially relevant for species that may take many years before a return on SPA investment is realised. Two recently established yellow box (Eucalyptus melliodora A.Cunn. ex Schauer, Myrtaceae) SPAs were evaluated to determine whether genetic bottlenecks had occurred between seed collection and SPA establishment. No evidence was found to suggest that a significant loss of genetic diversity had occurred at this stage, although there was a significant difference in diversity between the two SPAs. Complex population genetic structure was also observed in the seed used to source the SPAs, with up to eight groups identified. Plant survival in the SPAs was influenced by seed collection location but not by SPA location and was not associated with genetic diversity. There were also no associations between genetic diversity and plant growth. These data highlighted the importance of chance events when establishing SPAs and indicated that the two yellow box SPAs are likely to provide genetically diverse seed sources for future restoration projects, especially by pooling seed from both SPAs.

Understanding the relative importance of heterosis and outbreeding depression over multiple generations is a key question in evolutionary biology and is essential for identifying appropriate genetic sources for population and ecosystem restoration. Here we use 2455 experimental crosses between 12 population pairs of the rare perennial plant Rutidosis leptorrhynchoides (Asteraceae) to investigate the multi-generational (F 1 , F 2 , F 3 ) fitness outcomes of inter-population hybridization. We detected no evidence of outbreeding depression, with inter-population hybrids and backcrosses showing either similar fitness or significant heterosis for fitness components across the three generations. Variation in heterosis among population pairs was best explained by characteristics of the foreign source or home population, and was greatest when the source population was large, with high genetic diversity and low inbreeding, and the home population was small and inbred. Our results indicate that the primary consideration for maximizing progeny fitness following population augmentation or restoration is the use of seed from large, genetically diverse populations.
