Abstract

Apoptosis is a form of regulated cell death (RCD) that involves proteases of the caspase family. Pharmacological and genetic strategies that experimentally inhibit or delay apoptosis in mammalian systems have elucidated the key contribution of this process not only to (post-)embryonic development and adult tissue homeostasis, but also to the etiology of multiple human disorders. Consistent with this notion, while defects in the molecular machinery for apoptotic cell death impair organismal development and promote oncogenesis, the unwarranted activation of apoptosis promotes cell loss and tissue damage in the context of various neurological, cardiovascular, renal, hepatic, infectious, neoplastic and inflammatory conditions. Here, the Nomenclature Committee on Cell Death (NCCD) gathered to critically summarize an abundant pre-clinical literature mechanistically linking the core apoptotic apparatus to organismal homeostasis in the context of disease.

A study of the mechanism of action of the antimicrobial salivary peptide histatin-5 shows that it can aggregate negatively charged vesicles and is capable of fusing these vesicles, but only in the presence of Zn2+ ions. The Zn2+ ion induces a conformational change in the peptide in the presence of negatively charged vesicles as shown by circular dichroism spectroscopy. Two dimensional proton Nuclear magnetic resonance spectroscopy of histatin-5 in a trifluoroethanol/water mixture reveals that the peptide chain is helical and that zinc binding motif is most probably histidine mediated.

Abstract

The ability to precisely control cell‐loaded material systems is essential for in vitro testing of novel therapeutics poised to advance to clinic. In this report, unique patterns of cell migration are devised into an in vitro gel‐in‐gel model for the purpose of obtaining cell response data to potentially therapeutic chemical agonists. The model consists of co‐cultures in a cell‐loaded microgel invading an acellular "sorting" gel. Material properties including biophysical and chemical compositions of the sorting gel are carefully controlled to guide a desired cell‐specific behavior, leading to massive tumor cell invasion by amoeboid migration mechanisms. Optical transparency enables straightforward and high‐throughput measurements of outgrowth response in the presence of either chemical and photoradiation therapy. Important dosing and drug sensitivity information are obtained with the gel‐in‐gel model using no more than a light microscope, without further need for arduous genomic or proteomic screening of the tissue samples. Abstract

The properties of the synthetic RTN1-[C.sub.CT] peptide corresponding to the region from residues 186 to 208 were examined with relation to its ability to bind the metal ions in its N-terminal region. The results revealed that acetylated form of the metal-peptide complex have the ability to inhibit the histone deacetylase (HDAC) enzyme activity at micromolar concentrations and allowed to propose the Cu/Ni-RTN1-[C.sub.CT] complexes as models for the design of antitumor agents.

Abstract

A combination of electrophoretic and spectroscopic techniques is used to study the strong ability of reticulon paralogue (RTN1)-[C.sub.CT] peptide localized in the ER and expressed in the nervous system, to bind and condense the nucleic acid. The results revealed that the nucleic acid binding property of RTN1-C might have an important role in biological function of zinc-dependent histone deacetylase (HDAC) enzyme, HDAC8, which is regulated by an acetylation-deacetylation mechanism.

Abstract

The synthesis and characterization of a new peptide, an analogue of the histatin 5 named amino terminal C[u.sup.2+] and N[i.sup.2+] (ATCUN)-C16, which contains both metal-binding centers is reported. The results have shown that the salivary peptides preserve anticandidal activity and display a higher propensity to assume a stable conformation in a hydrophobic environment than do histatin 5 and the C16 peptide that contains the 16 residues of the C-terminal part of histatin 5.
