2. Eigenvalue problems and free boundary minimal surfaces in spherical caps

Abstract

Given a compact surface with boundary, we introduce a family of functionals on the space of its Riemannian metrics, defined via eigenvalues of a Steklov-type problem. We prove that each such functional is uniformly bounded from above, and we characterize maximizing metrics as induced by free boundary minimal immersions in some geodesic ball of a round sphere. Also, we determine that the maximizer in the case of a disk is a spherical cap of dimension two, and we prove rotational symmetry of free boundary minimal annuli in geodesic balls of round spheres which are immersed by first eigenfunctions.

Abstract

Manure N from cattle contributes to nitrate leaching, nitrous oxide, and ammonia emissions. Measurement of manure N outputs on commercial beef cattle operations is laborious, expensive, and impractical; therefore, models are needed to predict N excreted in urine and feces. Building robust prediction models requires extensive data from animals under different management systems worldwide. Thus, the study objectives were to 1) collate an international dataset of N excretion in feces and urine based on individual observations from beef cattle; 2) determine the suitability of key variables for predicting fecal, urinary, and total manure N excretion; and 3) develop robust and reliable N excretion prediction models based on individual observation from beef cattle consuming various diets. A meta-analysis based on individual beef data from different experiments was carried out from a raw dataset including 1,004 observations from 33 experiments collected from 5 research institutes in Europe (n = 3), North America (n = 1), and South America (n = 1). A sequential approach was taken in developing models of increasing complexity by incrementally adding significant variables that affected fecal, urinary, or total manure N excretion. Nitrogen excretion was predicted by fitting linear mixed models with experiment as a random effect. Simple models including dry matter intake (DMI) were better at predicting fecal N excretion than those using only dietary nutrient composition or body weight (BW). Simple models based on N intake performed better for urinary and total manure N excretion than those based on DMI. A model including DMI and dietary component concentrations led to the most robust prediction of fecal and urinary N excretion, generating root mean square prediction errors as a percentage of the observed mean values of 25.0% for feces and 25.6% for urine. Complex total manure N excretion models based on BW and dietary component concentrations led to the lowest prediction errors of about 14.6%. In conclusion, several models to predict N excretion already exist, but the ones developed in this study are based on individual observations encompassing larger variability than the previous developed models. In addition, models that include information on DMI or N intake are required for accurate prediction of fecal, urinary, and total manure N excretion. In the absence of intake data, equations have poor performance as compared with equations based on intake and dietary component concentrations.

11. A two-piece property for free boundary minimal hypersurfaces in the $(n+1)$-dimensional ball

Abstract

We prove that every hyperplane passing through the origin in $\rr^{n+1}$ divides an embedded compact free boundary minimal hypersurface of the euclidean $(n+1)$-ball in exactly two connected hypersurfaces. We also show that if a region in the $(n+1)$-ball has mean convex boundary and contains a nullhomologous $(n-1)$-dimensional equatorial disk, then this region is a closed halfball. Abstract

The ATLAS collaboration defines methods, establishes procedures, and organises advisory groups to manage the publication processes of scientific papers, conference papers, and public notes. All stages are managed through web systems, computing programs, and tools that are designed and developed by the collaboration. A framework called FENCE is integrated into the CERN GitLab software repository, to automatically configure workspaces where each analysis can be documented by the analysis team and managed by the relevant coordinators. Continuous integration is used to guide the writers in applying consistent and correct formatting when preparing papers to be submitted to scientific journals. Additional software assures the correctness of other aspects of each paper, such as the lists of collaboration authors, funding agencies, and foundations. The framework and the workflow therein provide automatic and easy support to the researchers and facilitates each phase of the publication process, allowing authors to focus on the article contents. The framework and its integration with the most up to date and efficient tools has consequently provided a more professional and efficient automatized work environment to the whole collaboration., Comment: 22 pages in total,11 figures, submitted to JINST. 32. The role of RUNX3 in acute myeloid leukaemia and normal human haematopoietic development

Abstract

The t(8;21)(q22;q22) is a common abnormality in acute myeloid leukaemia (AML). This translocation encodes the RUNX1-ETO fusion protein which blocks the differentiation of human CD34+ haematopoietic stem and progenitor cells (HSPC) and promotes their self-renewal, hallmarks of AML. Transcriptional repression of RUNX3 by RUNX1-ETO has been identified in t(8;21) AML but is strongly expressed in AML without core binding factor abnormalities. This study examines RUNX3 expression during normal and malignant human haematopoiesis and the consequences of increased and reduced RUNX3 expression for erythroid and myeloid cell development. Analysis of transcriptomic data showed that RUNX3 mRNA expression is downregulated during normal haematopoietic development. In AML, variable RUNX3 expression across different subtypes was observed, with core binding factor AML being associated with RUNX3 downregulation whilst AML patients with complex cytogenetics overexpressed RUNX3 compared to normal karyotype. The impact of altering RUNX3 expression on haematopoiesis was determined by transducing human HSPC with RUNX3 overexpression or shRNA knockdown vectors. RUNX3 overexpression inhibited both erythroid and myeloid growth and differentiation. Colony forming ability was also reduced but accompanied by increased self-renewal of progenitors. Reduced RUNX3 expression negatively impacted the survival of erythroid cells but did not cause significant effects in erythropoiesis and myelopoiesis. Attempts were also made to show whether RUNX3 could rescue the suppression of haematopoietic development seen in RUNX1-ETO-expresing cells. Further RNA-sequencing analysis showed that RUNX3 overexpression dysregulates the HSPC transcriptome, characterised by the repression of important erythroid-related genes, including KIT and LMO2. Differentiation markers GYPA, CD36 and ITGAM were also downregulated by RUNX3, validating the RUNX3-induced phenotypic changes during erythroid and myeloid development. In addition, cross-regulation among RUNX proteins was observed, as RUNX3 overexpression downregulated RUNX1 and RUNX2 levels in HSPC. In conclusion, this study shows that downregulation of RUNX3 expression is important for normal human erythroid and myeloid development. The ability of RUNX3 overexpression to inhibit differentiation suggest that RUNX3 could be a driver of leukaemogenesis.

Abstract

BackgroundAlcohol is a well-established risk factor for head and neck cancer (HNC). This study aims to explore the effect of alcohol intensity and duration, as joint continuous exposures, on HNC risk.MethodsData from 26 case-control studies in the INHANCE Consortium were used, including never and current drinkers who drunk ≤10 drinks/day for ≤54 years (24234 controls, 4085 oral cavity, 3359 oropharyngeal, 983 hypopharyngeal and 3340 laryngeal cancers). The dose-response relationship between the risk and the joint exposure to drinking intensity and duration was investigated through bivariate regression spline models, adjusting for potential confounders, including tobacco smoking.ResultsFor all subsites, cancer risk steeply increased with increasing drinks/day, with no appreciable threshold effect at lower intensities. For each intensity level, the risk of oral cavity, hypopharyngeal and laryngeal cancers did not vary according to years of drinking, suggesting no effect of duration. For oropharyngeal cancer, the risk increased with durations up to 28 years, flattening thereafter. The risk peaked at the higher levels of intensity and duration for all subsites (odds ratio = 7.95 for oral cavity, 12.86 for oropharynx, 24.96 for hypopharynx and 6.60 for larynx).ConclusionsPresent results further encourage the reduction of alcohol intensity to mitigate HNC risk.

Abstract

We prove that every plane passing through the origin divides an embedded compact free boundary minimal surface of the euclidean $3$-ball in exactly two connected surfaces. We also show that if a region in the ball has mean convex boundary and contains a nullhomologous diameter, then this region is a closed halfball. Moreover, we prove the regularity at the corners of currents minimizing a partially free boundary problem by following ideas by Gr\"uter and Simon. Here, we detected and isolated ZIKV in samples from Aedes aegypti, Aedes taeniorhynchus and Culex quinquefasciatus, collected during the Zika epidemic in Vitória, southeast Brazil. Using quantitative real-time polymerase chain reaction, ZIKV detection was performed in mosquito samples collected from February to April 2016.ResultsOverall, six pools of mosquitoes were positive for ZIKV: four of Cx. quinquefasciatus, one of Ae. aegypti and one of Ae. taeniorhynchus. Their genomes were sequenced.ConclusionsThese results support and strengthen the hypothesis that other mosquito species can also be involved in ZIKV transmission.

Abstract

IntroductionEarly childhood development can be described by an underlying latent construct. Global comparisons of children's development are hindered by the lack of a validated metric that is comparable across cultures and contexts, especially for children under age 3 years. We constructed and validated a new metric, the Developmental Score (D-score), using existing data from 16 longitudinal studies.MethodsStudies had item-level developmental assessment data for children 0-48 months and longitudinal outcomes at ages >4-18 years, including measures of IQ and receptive vocabulary. Existing data from 11 low-income, middle-income and high-income countries were merged for >36 000 children. Item mapping produced 95 'equate groups' of same-skill items across 12 different assessment instruments. A statistical model was built using the Rasch model with item difficulties constrained to be equal in a subset of equate groups, linking instruments to a common scale, the D-score, a continuous metric with interval-scale properties. D-score-for-age z-scores (DAZ) were evaluated for discriminant, concurrent and predictive validity to outcomes in middle childhood to adolescence.ResultsConcurrent validity of DAZ with original instruments was strong (average r=0.71), with few exceptions. Abstract

Background: Low prevalence of physical activity (PA) and a high prevalence of mental health problems are common among youth. The aim of this study was to evaluate the association between PA during adolescence and depression and anxiety disorders in young adulthood. Methods: Data from the 1993 Pelotas (Brazil) Birth Cohort Study were analyzed. Leisure-time PA and total PA (leisure plus commuting) were evaluated at 11, 15, and 18 years using self-reported data. PA was evaluated at each age separately and during adolescence, considering the number of times participants reached PA recommendations. Depression and anxiety were assessed through Mini International Neuropsychiatric Interview at 22 years. Crude and adjusted association analyses were performed using Poisson regression with robust error variance, providing prevalence ratios and 95% confidence intervals. Results: Three thousand two hundred and forty-seven participants were included in the study. In the adjusted analyses, active participants in leisure-time PA (≥300 min/wk) and total PA at 11 years were less likely to be depressed at age 22 (prevalence ratios: 0.54; 95% confidence interval, 0.33–0.89; prevalence ratios: 0.63; 95% confidence interval, 0.41–0.97). For leisure-time PA, the more PA recommendations were met during adolescence, the lower the prevalence of depression. There was no association when PA variables were evaluated separately at 15 and 18 years and between PA and anxiety after controlling for potential confounders. Conclusions: Early adolescence appears to be a sensitive period for PA benefits on depression in early adulthood. The more timepoints reaching PA recommendations during adolescence, the lower the risk of depression. On the other hand, PA during adolescence was not associated with anxiety in young adults.
