Simple Summary: Immune checkpoint inhibitors (ICIs), which can improve the immune system's ability to recognize and eliminate cancer cells, have significantly improved outcomes for patients with advanced kidney and bladder cancer. This success has prompted further research into the use of ICIs prior to definitive treatment in patients with localized genitourinary tumors at high risk for reoccurrence. Preliminary results from kidney and bladder cancer studies appear promising; however, further research is needed to evaluate a larger number of patients to demonstrate a survival benefit (compared to standard treatments) prior to the adoption of these treatment strategies. Conversely, the use of ICIs prior to definitive treatment thus far has demonstrated limited effectiveness in prostate cancer, likely due to dampening of the immune system response by the tumor and its surrounding cells. Overcoming these immune-suppressive mechanisms will require the development of biological tools that can better predict treatment response. This remains an area of active investigation. Genitourinary (GU) malignancies are common and associated with significant morbidity and mortality. In patients with localized GU cancers, surgical resection or definitive radiation remain the mainstays of treatment. Despite definitive treatment, many patients with high-risk localized disease experience recurrence. There is growing interest in using neoadjuvant immunotherapy to improve outcomes. This narrative review summarizes the current evidence for neoadjuvant immunotherapy in patients with localized high-risk GU cancers including renal cell carcinoma, urothelial carcinoma, prostate cancer, penile squamous cell carcinoma, and testicular germ cell tumors. We also discuss ongoing clinical trials and candidate biomarkers to optimize patient selection and improve treatment outcomes. [ABSTRACT FROM AUTHOR]

The quality of microseismic data greatly affects the accuracy of the source location results, especially in surface microseismic data where the first arrivals of P waves and S waves are not obvious or the low signal-to-noise ratio signals are difficult to identify.Secondly, due to the different focal mechanisms that induce microseismic, the signal characteristics received by different geophones are not the same.All of these factors have a certain impact on the accurate location of microseismic events.This paper proposes a method for selecting microseismic data based on cluster analysis to improve the imaging quality of microseismic event amplitude superposition locating.First, the Euclidean distance between the trace channels is calculated by cluster analysis, and the trace channels with larger and smaller distances are removed through the clustering graph, that is, trace channels with insignificant signal characteristics or severe noise interference are removed.Second, the cross-correlation calculation is performed on the signals of each channel, the trace channels with strong signal characteristic correlation being extracted, and those with unobvious P wave characteristics removed.Finally, the screened trace channels are superimposed and located with amplitude superposition.The comparison of the locating results with the original trace channels shows that the locating effect of the microseismic cluster analysis is more focused, the location of the focal point is clearer, and the locating results are significantly improved.

Biochar prepared from crop straw is an economical method for adsorbing bromocresol green (BCG) from textile industrial wastewater. However, there is limited research on the adsorption mechanism of biochar for the removal of BCG. This study utilized cucumber straw as raw material to prepare biochar with good adsorption potential and characterized its physicochemical properties. Through adsorption experiments, the effects of solution pH, biochar dosage, and initial dye concentration on adsorption performance were examined. The adsorption mechanism of cucumber straw biochar (CBC) for BCG was elucidated at the molecular level using adsorption kinetics, adsorption isotherm models, and density functional theory (DFT) calculations. Results show that the specific surface area of the CBC is 101.58 m2/g, and it has a high degree of carbonization, similar to the structure of graphite crystals. The presence of aromatic rings, –OH groups, and –COOH groups in CBC provides abundant adsorption sites for BCG. The adsorption process of CBC for BCG is influenced by both physical and chemical adsorption, and can be described by the Langmuir isotherm model, indicating a monolayer adsorption process. The theoretical maximum monolayer adsorption capacity (qm) of BCG at 298 K was calculated to be 99.18 mg/g. DFT calculations reveal interactions between BCG and CBC involving electrostatic interactions, van der Waals forces, halogen–π interactions, π–π interactions, and hydrogen bonds. Additionally, the interaction of hydrogen bonds between BCG and the –COOH group of biochar is stronger than that between BCG and the –OH group. Background and Purpose: Amyloid‐β (Aβ) peptide is one of the more important pathological markers in Alzheimer's disease (AD). The development of AD impairs autophagy, which results in an imbalanced clearance of Aβ. Our previous research demonstrated that AdipoRon, an agonist of adiponectin receptors, decreased the deposition of Aβ and enhanced cognitive function in AD. However, the exact mechanisms by which AdipoRon affects Aβ clearance remain unclear. Experimental Approach: We studied how AdipoRon affects autophagy in HT22 cells and APP/PS1 transgenic mice. We also investigated the signalling pathway involved and used pharmacological inhibitors to examine the role of autophagy in this process. Key Results: AdipoRon promotes Aβ clearance by activating neuronal autophagy in the APP/PS1 transgenic mice. Interestingly, we found that AdipoRon induces the nuclear translocation of GAPDH, where it interacts with the SIRT1/DBC1 complex. This interaction then leads to the release of DBC1 and the activation of SIRT1, which in turn activates autophagy. Importantly, we found that inhibiting either GAPDH or SIRT1 to suppress the activity of SIRT1 counteracts the elevated autophagy and decreased Aβ deposition caused by AdipoRon. This suggests that SIRT1 plays a critical role in the effect of AdipoRon on autophagic induction in AD. Conclusion and Implications: AdipoRon promotes the clearance of Aβ by enhancing autophagy through the AdipoR1/AMPK‐dependent nuclear translocation of GAPDH and subsequent activation of SIRT1. Granule size distribution of wheat starch is an important characteristic that could affect the functionality of wheat (Triticum aestivum L.) products. Lodging is a major limiting factor for wheat production. Few studies have been conducted to clarify how lodging influences the granule size distribution and viscosity parameters of starch in wheat grains. Two growing seasons, two high-yield winter wheat cultivars, and five artificial lodging treatments were imposed. The results indicated that lodging significantly reduced the content of starch and increased that of protein. Additionally, lodging caused a marked drop in both starch and protein yields. The relative loss of grain yield, starch yield, harvest index, and protein yield all differed remarkably among lodging treatments with a ranking of L2 > L1 > L4 > L3. Lodging also led to a reduction in the proportion (both by volume and by surface area) of B-type granules and a corresponding increase in that of A-type granules, and the more serious the lodging degree, the greater effect on the changes in these proportions. The smaller starch granules predominated in number, even though their collective contribution to the overall volume is was relatively minor. Meanwhile, it was found that the peak viscosity, hold viscosity, final viscosity, breakdown viscosity, and rebound value of wheat starch were significantly decreased by lodging. Correlation analysis showed that the peak and final viscosities were negatively correlated with volume percentages of A-type starch granules, but were positively correlated with B-type granules. This indicates that B-type granules have higher peak and final viscosities compared with A-type granules in wheat kernels. Lodging can reduce the proportion of B-type starch granules, and thus reduce the peak and the final viscosity in wheat grain. [ABSTRACT FROM AUTHOR]

The most common subtype of renal cell carcinoma is clear cell renal cell carcinoma (ccRCC). While localized ccRCC can be cured with surgery, metastatic disease has a poor prognosis. Recently, immunotherapy has emerged as a promising approach for advanced ccRCC. This review provides a comprehensive overview of the evolving immunotherapeutic landscape for metastatic ccRCC. Immune checkpoint inhibitors (ICIs) like PD-1/PD-L1 and CTLA-4 inhibitors have demonstrated clinical efficacy as monotherapies and in combination regimens. Combination immunotherapies pairing ICIs with antiangiogenic agents, other immunomodulators, or novel therapeutic platforms such as bispecific antibodies and chimeric antigen receptor (CAR) T-cell therapy are areas of active research. Beyond the checkpoint blockade, additional modalities including therapeutic vaccines, cytokines, and oncolytic viruses are also being explored for ccRCC. This review discusses the mechanisms, major clinical trials, challenges, and future directions for these emerging immunotherapies. While current strategies have shown promise in improving patient outcomes, continued research is critical for expanding and optimizing immunotherapy approaches for advanced ccRCC. Realizing the full potential of immunotherapy will require elucidating mechanisms of response and resistance, developing predictive biomarkers, and rationally designing combination therapeutic regimens tailored to individual patients.
