1. Structure design and breakaway failure analysis of fuse-pin for civil main landing gear

Abstract

The landing gear should be able to achieve safe and rapid separation from the aircraft body under overload conditions. To clarify the effect of breakaway groove on the breakaway load of emergency fuse-pin for civil main landing gear, a series of FEM simulations are conducted to study the influence of structural changes of breakaway groove on the breakaway load for outer-groove, long outer-groove, inner-groove and short inner-groove emergency fuse-pin. The breakaway load and failure mode are comparatively analyzed between inner-groove and outer-groove emergency fuse-pin by experimental tests. The results show that the breakaway failure of fuse-pin is extended from inside to outside. The breakaway loads increase with the increasing of the sectional area and exhibit a linear relationship. The breakaway load variation with the sectional area（shear strength coefficient）of through-hole pin is the maximum, followed by outer-groove pin, and the inner-groove pin is the minimum. Compared with other pins, the breakaway load of inner-groove pin is easy to control and less affected by sectional area and overall size. The fracture morphology is changed from ductile fracture of outer-groove pin to brittle fracture of inner-groove pin. Accordingly, the design of inner-groove pin meets the aviation regulations requirement for civil main landing gear.

Abstract

Interleukin‐4 (IL‐4)/IL‐4 receptor alpha (IL‐4Rα) signalling pathways play important roles in the complex process of bone formation and bone remodelling. However, whether IL‐4/IL‐4Rα participates in skeletogenesis during embryonic development is not completely understood. We used the anti‐IL‐4Rα monoclonal antibody (anti‐IL‐4Rα mAb) as a powerful investigational tool to evaluate the potential roles of IL‐4/IL‐4Rα in the chondrogenic differentiation of rat bone marrow mesenchymal stem cells (BMSCs) in vitro. Simultaneously, we explored the effect of IL‐4/IL‐4Rα on bone ossification during rat embryo‐fetal development. In this study, we found that, compared to the control group, IL‐4 can significantly promote the chondrogenic differentiation of BMSCs. Furthermore, following exposure to anti‐IL‐4Rα mAb in pregnant rats, unexpected phenomena were observed in fetal bone development, including non‐ossification of the fetal sternum, an incomplete ossification centre in long bones and a reduced number of ossification points in digit (toe) bones. To further investigate the underlying mechanism of the phenotype, we studied the rat sternum as the target organ, starting from different time points of sternum development in the embryonic stage. The results indicated that the retardation mainly occurred in the middle and late stages of embryonic development. This retardation was characterized by the inhibition of the differentiation process of mesenchymal stem cells into chondrocytes, resulting in reduced angiogenesis near the ossification centre, failure of osteoblasts to invade the centre of the cartilage body with the blood vessels and delayed formation of the primary ossification centre (POC). Overall, our study demonstrated the significant function of IL‐4/IL‐4Rα in chondrogenic differentiation of BMSCs and bone ossification during embryo‐fetal development. [ABSTRACT FROM AUTHOR]

Abstract

Over the past decades, mesenchymal stromal cells (MSCs) have been extensively investigated as a potential therapeutic cell source for the treatment of various disorders. Differentiation of MSCs from human induced pluripotent stem cells (iMSCs) has provided a scalable approach for the biomanufacturing of MSCs and related biological products. Although iMSCs shared typical MSC markers and functions as primary MSCs (pMSCs), there is a lack of lineage specificity in many iMSC differentiation protocols. Here, a stepwise hiPSC‐to‐iMSC differentiation method is employed via intermediate cell stages of neural crest and cytotrophoblast to generate lineage‐specific MSCs with varying differentiation efficiencies and gene expression. Through a comprehensive comparison between early developmental cell types (hiPSCs, neural crest, and cytotrophoblast), two lineage‐specific iMSCs, and six source‐specific pMSCs, are able to not only distinguish the transcriptomic differences between MSCs and early developmental cells, but also determine the transcriptomic similarities of iMSC subtypes to postnatal or perinatal pMSCs. Additionally, it is demonstrated that different iMSC subtypes and priming conditions affected EV production, exosomal protein expression, and cytokine cargo. [ABSTRACT FROM AUTHOR]
