Objectives Hepatocellular carcinoma (HCC) is difficult to diagnose early and progresses rapidly, making it one of the most deadly malignancies worldwide. This study aimed to evaluate whether salivary glycopattern changes combined with machine learning algorithms could help in the accurate diagnosis of HCC. Methods Firstly, we detected the alteration of salivary glycopatterns by lectin microarrays in 118 saliva samples. Subsequently, we constructed diagnostic models for hepatic cirrhosis (HC) and HCC using three machine learning algorithms: Least Absolute Shrinkage and Selector Operation, Support Vector Machine (SVM), and Random Forest (RF). Finally, the performance of the diagnostic models was assessed in an independent validation cohort of 85 saliva samples by a series of evaluation metrics, including area under the receiver operator curve (AUC), accuracy, specificity, and sensitivity. Results We identified alterations in the expression levels of salivary glycopatterns in patients with HC and HCC. The results revealed that the glycopatterns recognized by 22 lectins showed significant differences in the saliva of HC and HCC patients and healthy volunteers. In addition, after Boruta feature selection, the best predictive performance was obtained with the RF algorithm for the construction of models for HC and HCC. The AUCs of the RF-HC model and RF-HCC model in the validation cohort were 0.857 (95% confidence interval [CI]: 0.780–0.935) and 0.886 (95% CI: 0.814–0.957), respectively. Conclusions Detecting alterations in salivary protein glycopatterns with lectin microarrays combined with machine learning algorithms could be an effective strategy for diagnosing HCC in the future.

Glycosphingolipids (GSLs) are widely distributed in the phospholipid bilayer of various cell membranes, which play an important role in maintaining cell membrane stability, and regulate various cellular processes including adhesion, proliferation, apoptosis and recognition, as well as participate in various cellular activities. In addition, GSLs are not only involved in the process of apoptosis, but also regulate multiple signals in tumorigenesis and tumor development. The tumor-associated GSLs are expected to be used as diagnostic markers and immunotherapeutic targets for malignant tumors. These findings have important implications for the study of apoptosis and provide the new direction of tumor therapy. This review summarized the latest research progress of GSLs-mediated apoptosis and its effect on the genesis, development and metastasis of tumor cells. Moreover, we discussed the metabolic pathway of GSLs-mediated apoptosis and its application in tumor therapy, as well as the development prospect of targeted therapy strategies based on GSLs.

Aiming at the safety accident caused by the close distance between the crane arm and the high-voltage line when the crane is working near the high-voltage transmission line, this paper proposes a simulation model of the electric field intensity distribution of the high-voltage transmission line. First, through the simplification and equivalent treatment of the high-voltage line, the simulation modeling, and analysis of the electric field around the high-voltage line are carried out. Then, the theory of solving the electric field intensity of any point around the high-voltage line based on the charge simulation method is deduced, and the conclusion that the electric field intensity at the equal distance of the high-voltage line is approximately equal is obtained. Furthermore, the influence of the crane on the electric field strength of a high-voltage line is analyzed, and the linear correction function of electric field strength is obtained. Based on the research in this paper, the field strength sensor can be used to obtain the value of the electric field strength, calculate the distance between the crane and the high-voltage line, and make a safety warning response. The simulation results verify the feasibility and effectiveness of the proposed model and method.

Breast cancer is the most frequently diagnosed cancer in most countries. Early diagnosis of breast disease is necessary for its prognosis and treatment. Altered protein glycosylation has been shown to be expressed in precursor lesions of breast cancer, making them powerful early diagnostic biomarkers. The present study validated alterations of the N-glycan profiles of their salivary glycoproteins isolated by the Phaseolus vulgaris leucoagglutinin (PHA-E+L)-magnetic particle conjugates from 141 female subjects (66 healthy volunteers (HV), and 75 patients with breast disease including breast benign cyst (BB) or breast cancer in stage I/II (BC-I/II)) were analyzed and annotated by MALDI-TOF/TOF-MS. The results showed that there were 11, 20, 16, and 17 N-glycans recognized by PHA-E+L identified and annotated from the pooled salivary samples of HV, BB, BC-I, and BC-II, respectively. There were 3 N-glycans peaks (m/z 2459.8799, 2507.9139, and 2954.0547), 2 N-glycans peaks (m/z 1957.7265 and 2794.0427), and 2 N-glycans peaks (m/z 1866.6608 and 2240.8056) recognized by PHA-E+L that existed only in BB, BC-I, and BC-II, respectively. The present study compared the alternations of N-glycans from the salivary proteins isolated by PHA-E+L-magnetic particle conjugates among HV, BB, BC-I, and BC-II, which could provide information on N-glycans during the development of breast cancer in saliva to promote the study of its biomarkers.

Hepatocellular carcinoma (HCC) is a common malignancy with a poor prognosis. Dimethylaminomicheliolide (DMAMCL) is a novel antitumor agent that has been tested in phase I clinical trials; however, little is known regarding its effects in HCC. In this study, we found that DMAMCL reduces the viability of HCC cells in a dose- and time-dependent manner. In addition, DMAMCL causes cell cycle arrest at the G2/M phase and inhibits cell invasion and epithelial-mesenchymal transition (EMT). DMAMCL treatment also induces apoptosis via the intrinsic apoptotic pathway in HCC cells, which could be blocked by the pan-caspase inhibitor zVAD-fmk and silencing of Bax/Bak or overexpression of Bcl-2. Furthermore, DMAMCL treatment inactivates the PI3K/Akt pathway and leads to the generation of reactive oxygen species (ROS), which regulate apoptosis and inhibition of PI3K/Akt induced by DMAMCL. In vivo, DMAMCL inhibits tumor growth in mice bearing xenograft HCC tumors without noticeable toxicity. In summary, DMAMCL exerts antitumor effects both in vitro and in vivo and therefore may be applied as a potential therapeutic agent for HCC.

Colorectal cancer (CRC) is the most common carcinoma worldwide, but a lack of effective prognostic markers limits the clinical diagnosis and treatment. Yes-associated protein 1 (YAP1) is an effector of the HIPPO-pathway, which plays a critical role in cancer development and prognosis, including CRC. However, previous reports have suggested that it plays a dual role in CRC.A meta-analysis using RevMan software 5.4 and Stata 14.0 was performed to evaluate the relationship between YAP1 and clinical outcomes of CRC, after searching for eligible studies from the PubMed, Web of Science and Embase databases. Online datasets GEPIA and LOGpc were also used to calculate survival results and compare with the meta-analysis results. Besides, "DESeq" packages was used for expression analysis of YAP1 from TCGA dataset.YAP1 was over expression in the tissue of cancers comparing to normal tissues in patients with CRC from TCGA database (p=0.000164) and GEPIA database. A total of 10 studies involving 2305 patients from literatures were selected. Pooled HR indicated that over-expression of YAP1 was associated with poor clinical outcomes (HR=1.70, 95% CI: 1.28-2.26, p=0.0003). Subgroup analysis showed a clear correlation between over-expression of YAP1 and worse survival rate in Chinese patients (HR=1.94, 95% CI: 1.40-2.69, p=0.0001), nuclear YAP1 over-expression (HR=2.07, 95% CI: 1.29-3.31, p=0.003), 60 months follow-up duration (HR=1.89, 95% CI: 1.30-2.73, p=0.0008), IHC test (HR=1.65, 95% CI: 1.17-2.33, p=0.005), IHC combined with other tests (HR=1.77, 95% CI: 1.13-2.77, p=0.01) and multivariate analysis (HR=1.70, 95% CI: 1.24-2.31, p=0.0009). Nevertheless, disease-free survival (DFS) did not show significant result in the patients with CRC in our meta-analysis (HR=1.38, 95% CI: 0.51-3.75, p=0.52) as well as in the GEPIA and LOGpc databases. Meanwhile, YAP1 over-expression was also significantly associated with worse overall survival (OS) in GSE17536, GSE40967, GSE29623 and GSE71187.YAP1 over-expression is common in CRC tissues. Over-expression of YAP1 in CRC patients, particularly in the nucleus, might be related to shorter OS, maybe in the early stages. YAP1 could serve as a potential predictor of poor prognosis in CRC.

We derived a steady-state model of whole root pressure generation through the combined action of all parallel segments of fine roots. This may be the first complete analytical solution for root pressure, which can be applied to complex roots/shoots.The osmotic volume of a single root is equal to that of the vessel lumen in fine roots and adjacent apoplastic spaces. Water uptake occurs via passive osmosis and active solute uptake (, osmol s−1), resulting in the osmolal concentration Cr (mol·kg−1 of water) at a fixed osmotic volume. Solute loss occurs via two passive processes: radial diffusion of solute Km(Cr-Csoil), where Km is the diffusional constant and Csoil is the soil-solute concentration) from fine roots to soil and mass flow of solute and water into the whole plant from the end of the fine roots.The proposed model predicts the quadratic function of root pressure , where b and c are the functions of plant hydraulic resistance, soil water potential, solute flux, and gravitational potential.The present study investigates the theoretical dependencies of Pr on the factors detailed above and demonstrates the root pressure-mediated distribution of water through the hydraulic architecture of a 6.8-m-tall bamboo shoot.One sentence SummaryAn analytical solution for root pressure is derived and illustrated by applying it to the measured hydraulic architecture of a 7-meter bamboo shoot.

Zeranol (α-zearalanol) has been used as a growth promoter in livestock since 1969 in some non-EU countries; the residues of zeranol and its five analogues in animal origin foods may endanger human health due to their strong estrogenic and anabolic activities. Therefore, it is urgent to establish simple, rapid, real-time, broad-spectrum and high-sensitivity detection methods for the residues of zeranol and its analogues. In this study, an ultrasensitive indirect-competition enzyme-linked immunosorbent assay (ic-ELISA) was established for the rapid multi-residue detection of zeranol and its five analogues in cattle origin samples, which was based on a broad-spectrum monoclonal antibody (mAb) that specifically bound to zeranol and its analogues with high sensitivity. The half maximal inhibitory concentration (IC50) values for zeranol, β-zearalanol, zearalanone, α-zearalenol, β-zearalenol, and zearalenone were 0.103, 0.080, 0.161, 0.177, 0.254, and 0.194 ng mL−1, respectively, the recovery rates of cattle origin samples spiked with zeranol ranged from 79.2–104.2%, and the coefficient of variation (CV) values were less than 11.4%. Excellent correlation (R2 = 0.9845) was obtained between the results of HPLC-MS/MS and ic-ELISA. In conclusion, the developed ic-ELISA could be employed as an ultrasensitive and broad-spectrum detection method for monitoring trace ZEN residues in cattle origin foods.

Glycosylation is involved in several biological processes, and its alterations can reflect the process of certain diseases. Type 2 diabetes mellitus (T2DM) has attained the status of a global pandemic; however, the difference in salivary protein glycosylation between healthy subjects and patients with T2DM has not been fully understood. In the present study, salivary specimens from patients with T2DM (n = 72) and healthy volunteers (HVs, n = 80) were enrolled and divided into discovery and validation cohorts. A method combining the lectin microarray and lectin blotting was employed to investigate and confirm the altered glycopatterns in salivary glycoproteins. Then, lectin-mediated affinity capture of glycoproteins and MALDI-TOF/TOF-MS were performed to obtain the precise structural information of the altered glycans. As a result, the glycopatterns recognized by 5 lectins (LEL, VVA, Jacalin, RCA120 and DSA) showed significant alteration in the saliva of T2DM patients. Notably, the glycopattern of Galβ-1,4GlcNAc (LacNAc) recognized by LEL exhibited a significant increase in T2DM patients compared to HVs in both discovery and validation cohorts. The MALDI-TOF/TOF-MS results indicated that there were 10 and 7 LacNAc-containing N/O-glycans (e.g. m/z 1647.586, 11 688.613 and 1562.470) that were identified only in T2DM patients. Besides, the relative abundance of 3 LacNAc-containing N-glycans and 10 LacNAc-containing O-glycans showed an increase in the glycopattern in T2DM patients. These results indicated that the glycopattern of LacNAc is increased in salivary glycoproteins from T2DM patients, and an increase in LacNAc-containing N/O-glycans may contribute to this alteration. Our findings provide useful information to understand the complex physiological changes in the T2DM patients.

The present study was designed to highlight the effects of cold plasma (10 kV) treatment and priming with 2 mM salicylic acid (SA) and their combination (10 kV of plasma + 2 mM SA) on the physiological parameters and metabolism of two cultivars of Oryza sativa, i.e., Zhu Liang You 06 (ZY) and Qian You No. 1 (QY), under salinity stress (150 mM NaCl) and normal growth condition (0 mM NaCl). Seed germination and seedling growth were enhanced by SA priming and cold plasma treatment either alone or in combination under salinity stress. Photosynthetic pigments, photosynthetic gas exchange, and chlorophyll fluorescence were improved by cold plasma treatment and SA priming under salinity stress as compared to the untreated seeds. The activities of antioxidant enzymes were significantly improved by the combination of SA priming and cold plasma treatment in both cultivars under salinity stress. There were rapid changes in the cellular content of sodium (Na+) and calcium (Ca+), where the plants grown under saline conditions accumulate more Na+ and less Ca+ contents resulting in ionic imbalances. Interestingly, cold plasma and SA treatments diminished this action by reducing Na+ accumulation and increasing K+ and Ca+ contents in the plant cell under salinity stress. The activities of enzymes involved in secondary metabolism assimilation were up-regulated with cold plasma and SA priming either alone or combination under salinity stress. An increase in reactive oxygen species (ROS) accumulation and malondialdehyde (MDA) content was also observed under salinity stress condition. On contrast, seed treated with SA and plasma alone or combined resulted in a significant decrease in ROS and MDA contents under salinity stress. Our results indicated that SA priming and cold plasma treatment either alone or combined improved plant uptake of nutrients in both cultivars under stress conditions. The ultrastructural changes were observed to be more prominent in ZY than QY cultivar. Plants without SA priming or cold plasma treatments have a big vacuole due to the movement of ions into the vacuole directly from the apoplast into the vacuole through membrane vesiculation leading to membrane destabilization. However, SA priming and cold plasma treatment alone or combined helped the plants to recover their cell turgidity under salinity stress.

Low humidity monitoring has been attached to adequate attention in a wide range of industrial fields, such as medical device hermetic packaging, semiconductor manufacturing and transformer manufacturing. However, it remains challenging to detect slight moisture change in low-humidity environment for conventional ionic-type humidity sensors. Herein, Nickel (II) bromide (NiBr2) is explored to be of a reversible moisture-induced discoloration behavior and highly sensitive to moisture in low humidity environment. NiBr2 humidity sensor delivers a tremendous impedance change nearly to 5 orders of magnitude in the low relative humidity (RH) ranging from 7 % to 24 % and the response time is only about 2 s when RH increases from 11 % to 33 %. The excellent performance for low-humidity detection can be mainly attributed to the reversible bulk absorption behavior to water molecules that lead to a reversible material transition between NiBr2 and NiBr2⋅6H2O. Meanwhile, the sensitivity of the sensor toward the range of 7 %–24 % RH exhibits nearly 5 orders of magnitude enhancement than that in medium-high humidity range of 26 %–95 % RH, which indicates that the sensitivity of the sensor to water molecules caused by the bulk absorption behavior is better than that dominated by the surface adsorption behavior. The results demonstrate that the bulk absorption behavior-mediated reversible material transition provide efficient guidance to realize low-humidity detection.

Background: Aflatoxin B1 (AFB1 ) is a kind of toxic and carcinogenic mycotoxin. A time-resolved fluoroimmunoassay (TRFIA) was established for quantitative detection of AFB1 in feed using Eu3+ -labeled IgG as tracer.; Results: Monoclonal antibody (McAb) against AFB1 (9B11-D7) was prepared through immunization and cell fusion and was identified as high affinity, specificity and sensibility by enzyme-linked immunosorbent assay (ELISA). The 50% inhibition value (IC50 ) was 0.81 ng mL-1 , the limit of detection (LOD) was 0.10 ng mL-1 and detection range was 0.10-3.94 ng mL-1 . Goat anti-mouse immunoglobulin G (IgG) was modified by Eu3+ -DATT, generating Eu3+ -labeled IgG. Under optimal assay conditions, TRFIA was shown to be highly sensitive and specific in detection of AFB1 . The IC50 and LOD were 94.73 pg mL-1 and 3.55 pg mL-1 , respectively, and detection range was 3.55-1.11 × 103 pg mL-1 . Cross-reactivity with AFM1 , AFB2 , AFG1 and AFG2 was 31.26%, 37.6%, 127.46% and 35.74%, respectively, but zero with other analogues. In determination of AFB1 spiked in feed sample, TRFIA showed high accuracy and precision. The average recoveries ranged from 93.71% to 97.80%, and coefficient of variation was 1.25-3.73%. Good correlation between TRFIA and HPLC was demonstrated for determination of AFB1 in feeds, confirming the reliability of the developed method.; Conclusion: The developed TRFIA exhibited good potential for employment in the ultrasensitive detection of AFB1 in feed and could be used to determine total aflatoxins. © 2017 Society of Chemical Industry.; © 2017 Society of Chemical Industry.

Transition metal dichalcogenides (TMDs) have blazed a novel trail in the field of NO2 gas detection owing to the unique photoelectric properties and prospective gas sensing performance especially working at low operating temperature. However, the incomplete recoverability and susceptibility to humidity at room temperature restrict the practical applications of TMDs-based gas sensors. Herein, we demonstrate a fully recoverable WS2 based gas sensor with enhanced anti-humidity ability by combining the decoration of zero-dimensional PbS nanoparticles and the assistance of UV light. Through the control over the molar ratio of WS2/PbS, the optimized WS2/PbS (the molar is 10:1) gas sensor exhibits a remarkable elevated recovery rate from 33.4% to 99.8% with almost no baseline drift as compared to that in the pure WS2 gas sensor. More importantly, attributing to the irradiation of UV can accelerate the desorption of water molecules, WS2/PbS heterostructure based gas sensor manifests more stabilized anti-humidity performance with stable initial resistance and negligible response value drift (8.8%) in the range of 10%–90% relative humidity. Furthermore, UV-assisted WS2/PbS gas sensor also possesses prominent sensing performances including high sensitivity, low detection limit (∼20 ppb), favorable selectivity and outstanding long-term stability. This research will pave the way on designing high-performance TMDs-based gas sensors operating at room temperature.

Based on colloidal gold and broad-spectrum monoclonal antibody that binds to zeranol and its five analogues with high sensitivity, a lateral flow immunochromatographic assay (LFIA) in a competitive format was developed to specifically determine residues of zeranol, an illegal growth promoter in livestock. In this study, the assay had high sensitivity and was broad-spectrum only for zeranol and its five analogues, and the results were obtained within 10 min without needing sophisticated procedures. The cutoff values for zeranol and its five analogues were 10 ng/mL, and the IC50 values for zeranol, β-zearalanol, zearalanone, α-zearalenol, β-zearalenol and zearalenone were 1.250, 1.800, 1.775, 1.225, 1.709 and 1.319 ng/mL, respectively. The recovery rates were ranged from 85.6 to 93.9%, with the coefficient of variations less than 12.4%. The results demonstrated that the LFIA could be used for rapid, simultaneous, semi-quantitative and quantitative detection of residues of zeranol and its five analogous in milk.

Ultra-low humidity detection is significant in some important industrial fields, such as integrated circuit manufacturing, chip packaging. However, the traditional ionic-type humidity sensor is not competent for the detection of ultra-low humidity, but can only realize the detection of medium humidity to high humidity. Here, moisture-induced discoloration material Nickel(II) iodide (NiI2) is found to be highly sensitive to moisture, and both its electrical properties and color will change with environmental humidity variation. Especially, NiI2 exhibits the largest impedance change to date in the ultra-low humidity range compared with state of the art. The impedance variation is up to 2 orders of magnitude in the range of 0.4 %–11 % RH, which means that NiI2 can realize the detection of ultra-low humidity. The excellent performance of NiI2 in ultra-low humidity is attributed to the material transition characteristics susceptible to humidity rather than the traditional surface mechanism of water adsorption. Moreover, combining the reversible black-to-transparency moisture-induced discoloration behavior with humidity sensing properties, a flexible wristband based on NiI2 is developed to realize the dual-mode detection of wide range environmental humidity. These results demonstrate that NiI2 has great application prospects in professional ultra-low humidity detection and wearable humidity detection equipment.

A competitive ELISA (cELISA) for the quantitative detection of ochratoxin A (OTA) was developed that uses a clone selected from a phage random peptide display library. An anti-OTA monoclonal antibody (mAb) was employed as the target for clone selection from a phage random heptapepide library. After four rounds of panning, 22 positive phage clones, in which binding to anti-OTA mAb could be blocked by free OTA, were obtained. The peptide sequences were deduced from DNA sequencing, and the consensual amino acid sequence of the selected peptides is MPLWXDL (X is any amino acid residue). A cELISA for detecting OTA was established using the selected phage clones. In the most sensitive assay, the linear range of the inhibition curve was 125–8000 pg mL−1. The half-inhibitory concentration (IC50) was 481.8 pg mL−1 and the detection limit was 103.2 pg mL−1. The analysis of 15 domestic cereal samples in parallel using this cELISA, a commercial ELISA kit and HPLC demonstrated the effectiveness of the method for detection of OTA in cereal samples.

Luffa cylindrica (L.) Roem. is an economically important vegetable crop in China. However, the genomic information on this species is currently unknown. In this study, for the first time, a genome survey of L. cylindrica was carried out using next-generation sequencing (NGS) technology. In total, 43.40 Gb sequence data of L. cylindrica, about 54.94× coverage of the estimated genome size of 789.97 Mb, were obtained from HiSeq 2500 sequencing, in which the guanine plus cytosine (GC) content was calculated to be 37.90%. The heterozygosity of genome sequences was only 0.24%. In total, 1,913,731 contigs (>200 bp) with 525 bp N50 length and 1,410,117 scaffolds (>200 bp) with 885.01 Mb total length were obtained. From the initial assembled L. cylindrica genome, 431,234 microsatellites (SSRs) (≥5 repeats) were identified. The motif types of SSR repeats included 62.88% di-nucleotide, 31.03% tri-nucleotide, 4.59% tetra-nucleotide, 0.96% penta-nucleotide and 0.54% hexa-nucleotide. Eighty genomic SSR markers were developed, and 51/80 primers could be used in both “Zheda 23” and “Zheda 83”. Nineteen SSRs were used to investigate the genetic diversity among 32 accessions through SSR-HRM analysis. The unweighted pair group method analysis (UPGMA) dendrogram tree was built by calculating the SSR-HRM raw data. SSR-HRM could be effectively used for genotype relationship analysis of Luffa species.

An ultrasensitive method for the detection of antibiotics in milk is developed based on inexpensive, simple, rapid and portable lateral flow immunoassay (LFI) strip, in combination with high sensitivity surface-enhanced Raman spectroscopy (SERS). In our strategy, an immunoprobe was prepared from colloidal gold (AuNPs) conjugated with both a monoclonal antibody against neomycin (NEO-mAb) and a Raman probe molecule 4-aminothiophenol (PATP). The competitive interaction with immunoprobe between free NEO and the coated antigen (NEO-OVA) resulted in the change of the amount of the immobilized immunoprobe on the paper substrate. The LFI procedure was completed within 15min. The Raman intensity of PATP on the test line of the LFI strip was measured for the quantitative determination of NEO. The IC50 and the limit of detection (LOD) of this assay are 0.04ng/mL and 0.216pg/mL of NEO, respectively. There is no cross-reactivity (CR) of the assay with other compounds, showing high specificity of the assay. The recoveries for milk samples with added NEO are in the range of 89.7%-105.6% with the relative standard deviations (RSD) of 2.4%-5.3% (n=3). The result reveals that this method possesses high specificity, sensitivity, reproducibility and stability, and can be used to detect a variety of antibiotic residues in milk samples.
