Objective: To indicate the effect of Epigallocatechin gallate (EGCG) and Cisplatin (DDP) on proliferation of gastric cancer BGC-823 cells and the relative underlying mechanism. Methods: Cultured BGC-823 cells were treated by 5 μg/mL DDP, 25 μg/mL EGCG and combined 5 μg/mL DDP with 25 μg/mL EGCG, a blank group was used as control. Cell morphology was observed by 4’,6-diamidino-2-phenylindole (DAPI) staining. The ability of cell proliferation was detected by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT)assay. The cell cloning rate was determined by colony formation assay. The ability of cell migration was detected by cell scratch test. The cell cycle distributions and apoptosis were analyzed by flow cytometry, The expression of p19Arf, p53, p21Cip1 mRNA was determined by RT-qPCR. The protein levels of p19Arf, p53, p21Cip1 were measured by Western blot. Results: Compared with DDP or EGCG treatment alone, EGCG combined with DDP treatment significantly caused nuclear shrinkage, reduced the proliferation rate, the ability of cell clone and migration. Background: One of the most common treatment for gastric cancer is chemotherapy, however, multiple drug resistance (MDR) induce the therapeutic effect which result in the failure of anticancer therapy. Dihydromyricetin (DMY) was reported to have antitumor activities on various human cancer cells in vitro, our previous studies demonstrated that DMY combined with mitomycin has inhibitory effect on proliferation of gastric carcinoma cells. However, the underlying role of DMY reversing the MDR of gastric carcinoma is poor understood. The aim of this study was to evaluate the reversal effect of DMY on MDR and investigate the molecular mechanisms in vitro. Methods: Using MTT assay, we identified the toxicity of DMY on SGC7901 and SGC7901/5-FU cells. The effect of DMY on 5-FU induced apoptosis was evaluated by flow cytometry analysis. Using RT-PCR and Western blot, we determined the MDR1 mRNA and protein expression. Results: DMY induced growth inhibition in both SGC7901 and SGC7901/5-FU cells, the IC50 value was 13.64±1.15 µg/mL, 20.69±1.82 µg/mL respectively. DMY treatment sensitized SGC7901/5-FU cells to cytotoxicity of 5-FU. The combination of DMY with 5-FU increased the apoptosis rate (9.91%, 16.67%) comparing with 5-FU alone (5.25%). Comparing with the control group, the MDR1 mRNA and protein expression in SGC7901/5-FU cells after treatment of DMY decreased significantly (P< 0.05). Conclusion: In brief, our study demonstrated that DMY effectively reversed multi-drug resistance occurring in SGC7901/5-FU cells cultured in vitro, and the potential mechanism was involved in the downregulation of the MDR1 expression.

Liquid nitrogen (LN2), which can greatly improve the efficiency of hot dry rock (HDR) mining, is commonly used as a cooling material in the enhanced geothermal system (EGS). Physical property, triaxial compression, and permeability tests were undertaken on treated granite samples, for a better scientific understanding of the effect of the LN2 cooling method on the mechanical and permeability properties of the rocks after heat treatment. The experimental results indicated that the physical properties of the treated granite change significantly, such as the density and wave velocity are substantially reduced. Meanwhile, with the increase of treatment temperature, the macroscopic cracks on its surface are gradually generated and the volume is expanded clearly. In addition, the surface wettability of granite gradually increases with increasing temperature. Compared with the air/water cooling methods, under LN2 cooling condition, the mechanical properties decrease markedly. When the temperature exceeds 600°C, the granite strength decreases significantly to only 56.16% of the reference value. The deformation properties also change significantly, with a final strain of about 3% at failure for a sample at 800°C, showing an obvious ductile deformation characteristic. Further, an appreciable correlation also exists between the initial permeability of granite and temperature. Once the temperature exceeds 200°C, the increase in temperature contributes to the increase in initial permeability. In addition to the effect of temperature, the increase in load also leads to a change in the permeability coefficient. When the temperature reaches 600°C, the permeability of granite first decreases and then increases with the increases in axial stress. The results of this paper are valuable in understanding the effect of thermal shock by LN2 on the fracturing efficiency and permeability characteristics of dry hot rocks.

To explore the development mechanism of cracks in the process of rock failure, triaxial compression tests with simultaneous acoustic emission monitoring were performed on granite specimens using the MTS rock mechanics test system. The frequency-domain information of the acoustic emission signal was obtained by the fast Fourier transform. The Gutenberg–Richter law was used to calculate the acoustic emission signals and obtain the b-value dynamic curve in the loading process. Combined with the stiffness curve of granite specimens and acoustic emission signal in the time domain and frequency domain, the crack development characteristics in different stages were analyzed. The results showed that the acoustic emission signals of granite samples under triaxial compression can be divided into four stages: quiet period 1, active stage 1, quiet period 2, and active stage 2. b-value attained its maximum value in the active phase 2 when it is close to the sample loss, and then drops rapidly, which means the propagation of cracks and the formation of large cracks. The acoustic emission signal’s dominant frequency was not more than 500 kHz, mostly concentrated in the medium-frequency band (100–200 kHz), which accounted for more than 80%. The proportion of signals in each frequency band can reflect the distribution of the three kinds of cracks, while the change in low-frequency signals can reflect the breakthrough of microcracks and the formation time of macrocracks in granite samples. By fully analyzing the characteristics of acoustic emission signals in the time domain and frequency domain, the time and conditions of producing large cracks can be determined accurately and efficiently.

OBJECTIVE: To investigate the mechanism by which epigallocatechin gallate (EGCG) induces CHD5 gene demethylation and promotes the apoptosis of acute myeloid leukemia KG-1 and THP-1 cell lines. METHODS: KG-1 and THP-1 cells treated with 25, 50, 75, 100 or 150 μg/mL EGCG for 48 h were examined for CHD5 gene methylation using MSP and for cell proliferation using MTT assay. The changes in cell cycle and apoptosis of the two cell lines after treatment with EGCG for 48 h were detected using flow cytometry. The mRNA and protein expressions of DNMT1, CHD5, p19(Arf), p53 and p21(Cip1) in the cells were detected using RT-quantitative PCR and Western blot. RESULTS: EGCG dose-dependently reversed hypermethylation of CHD5 gene and reduced the cell viability in both KG-1 and THP-1 cells (P < 0.05). EGCG treatment caused obvious cell cycle arrest in G1 phase, significantly increased cell apoptosis, downregulated the expression of DNMT1 and upregulated the expressions of CHD5, p19(Arf), p53 and p21(Cip1) in KG-1 and THP-1 cells (P < 0.05). CONCLUSION: EGCG reduces hypermethylation of CHD5 gene in KG-1 and THP-1 cells by downregulating DNMT1 to restore its expression, which results in upregulated expressions of p19(Arf), p53 and p21(Cip1) and induces cell apoptosis.

The variability and intermittency of renewable energy and power load bring great pressure to the dispatch of Micro-Grid, especially intra-day dispatch. In order to reduce the intra-day dispatch pressure, this paper proposes a data-driven two-stage day-ahead dispatch model for islanded Micro-Grid. The first stage dispatch model considers multiple demand responses, and applies phase space reconstruction and machine learning to predict renewable energy output and power load. Multi-objective particle swarm optimization algorithm is used to solve the first stage dispatch model. For the obtained Pareto Front, weight multiple objectives by entropy weight method to get the optimal solution. Since the deviation between the predicted value and the actual value can lead to renewable energy curtailment or load loss, the role of the second stage is to predict the renewable energy curtailment and load loss after the first stage dispatch. Firstly, extreme gradient boosting is used to predict when renewable energy curtailment and load loss occur. Secondly, extreme learning machine is used to predict the amount of renewable energy curtailment and load loss at the corresponding time points. Finally, linear programming and mixed integer linear programming are used to solve the second stage dispatch model. By comparative cases analysis, simulation results show that the enhancement of demand response to system efficiency is at the cost of increasing dispatch cost and reducing reliability. In contrast, the proposed two-stage dispatch method using regulation reserve capacity not only reduces dispatch cost, but also improves system efficiency and reliability.

This work introduces the dynamic economic dispatch of the micro-grid considering the uncertainty of the renewable energy. Firstly, this paper applies scenario analysis method to deal with the uncertainty of the renewable energy. After scenario generation and scenario reduction, the most representative scenario is selected. Secondly, the integrated optimal dispatching model of the micro-grid is established on the basis of considering the economy and the environmental friendliness. Finally, a case study is carried out in selected scenarios, using differential evolution algorithm to solve the integrated optimal dispatching model of micro-grid.

In order to investigate the impact of electric vehicles’ charging-discharging behaviour and demand side response resources on the economic operation of photovoltaic grid-connected microgrid system, a multi-objective model of microgrid economic dispatching with electric vehicles, transferable load and other distributed generations (diesel engines and energy storage unit) is proposed in this paper. The model takes the comprehensive operating cost of microgrid, the utilization rate of photovoltaic energy and the power fluctuation between the microgrid and main grid as objectives. Moreover, four different cases of microgrid economic dispatch considering electric vehicles and transferable load are put forward, which are electric vehicles’ orderly charging and discharging and transferable load participating in demand response in Case 1, electric vehicles’ charging randomly and the transferable load participating in demand response in Case 2, electric vehicles orderly charging and discharging and transferable load not participating in demand response in Case 3, electric vehicles’ charging randomly and the transferable load not participating in the demand response in Case 4. Multi-objective Seeker Optimization Algorithm and the method of fuzzy membership function are applied in this study to obtain the optimal results. The simulation analysis shows that the orderly charging-discharging behaviour of electric vehicles and the participation of transferable load can effectively improve the economic costs, efficiency and security of microgrid economic operation.
