Lightweight soil is a novel technological material that boasts characteristics such as low density, high strength, thermal insulation, vibration isolation, environmental friendliness, and cost-effectiveness. These features make it highly suitable for a wide range of applications in geotechnical engineering, including roadbed backfill, soft foundation treatment, and tunnel load reduction. The influence of vibration loads resulting from transportation, earthquakes, waves, and other factors on the mechanical properties of lightweight soil has garnered considerable attention in recent research. This paper expounds on the influence of factors on the dynamic deformation characteristics and dynamic strength properties of lightweight soil. These factors include the mix ratio (such as the content of lightweight materials, dosage of curing agent, and moisture content), stress state, vibration frequency, dry–wet alternation effect, and freeze–thaw cycle. Additionally, we summarise the calculation model for the dynamic shear modulus and damping ratio of lightweight soil. The findings reveal that the incorporation of curing agents, such as cement and fly ash, substantially improves the resistance of lightweight soil to dynamic loads. Additionally, the distinctive pore structure of lightweight soil markedly enhances its vibration isolation effect. As dynamic strain increases, there is a nonlinear decrease in the dynamic modulus of lightweight soil while the damping ratio increases nonlinearly. Adjusting the content of lightweight materials and the dosage of curing agents can markedly improve the seismic reduction effect of lightweight soil, thus granting it greater dynamic stability. The coupling effects of dry–wet cycles, freeze–thaw cycles, and dynamic loads may lead to a degradation in the dynamic performance of lightweight soil. To extend its service life in practical engineering applications, the implementation of a waterproof layer is recommended. Model tests and numerical simulations substantiated the commendable dynamic stability and durability of lightweight soil in real-world engineering scenarios. Finally, following a comprehensive literature review, this paper identifies potential research directions. The study of dynamic characteristics in lightweight soil is still in its infancy, with the dynamic properties of novel solid waste lightweight soil remaining largely unexplored. Further exploration is required to fully understand the response mechanisms, mechanical properties, and constitutive models of lightweight soil under the combined effects of complex environmental factors and dynamic loads. Additionally, there is a need for continued research into the design and construction methods of lightweight soil across various engineering settings. In conclusion, this paper serves as a valuable reference for investigating the dynamics of lightweight soil and its extensive application in geotechnical engineering.

Objective To clarify the oil and gas accumulation patterns and main controlling factors of the thin weathered crust volcanic rock reservoir at the top of Member 2 of the Fengcheng Formation in the southern Mahu Sag(Manan Sag). Methods This paper provides a comprehensively analyses of the volcanic rock characteristics, reservoir properties, and oil and gas geological features. Furthmore, an oil and gas accumulation model for volcanic rocks in the slope area is established. Results Research has shown that Member 2 of the Fengcheng Formation in the southern Mahu Sag is mainly composed of basalt and tuff. The basalt with the volcanic overflow phase developed high-permeability reservoirs abundant in oil and gas. Overall, the crude oil is medium-high viscosity and light-medium waxy. Its distribution pattern of C20, C21, and C23 tricyclic terpanes resembles a mountain peak, showing a significantly higher relative content of tricyclic terpanes. In contrast to crude oil, the distribution of tricyclic terpanes in in-situ source rocks demonstrates an upward pattern, with maturity parameters of biomarkers also displaying lower maturity characteristics. Based on the consistent research results of the carbon isotope characteristics of crude oil in the study area and in the Fengcheng Formation source rocks, it is infered that the oil reservoir in the study area has undergone multistage oil and gas charging. Conclusion The volcanic rocks in the study area exhibit strong vertical and horizontal heterogeneities, with crude oil density decreasing with depth. It is believed that oil and gas mainly migrate from low to high parts along permeable strata below volcanic rocks and enter volcanic rock reservoirs under buoyancy. They accumulate reservoirs in structures or lithological traps with relatively good reservoir properties and preservation conditions. This is a typical slope area-structure lithology-multistage volcanic rock oil and gas reservoir formation model.

Abstract In the study of obesity and diabetes, mice are widely used for experimental research, and fasting is a common procedure used to reset metabolism in mouse models. The fasting duration for experimental mice varies greatly in nutritional and metabolic studies, ranging from 2 to 48 h. This study aims to assess the optimal fasting duration for mice fed low- and high-fat diets over a short period of time. C57BL/6J mice were fed a low-fat diet (LFD) or high-fat diet (HFD) and fasted for 4, 6, 8, 10, 12, or 24 h. The effects of different conditions after fasting on the metabolic level of mice were explored, and the data were collected for analysis. Our data indicate that fasting has inconsistent effects on mice fed a low-fat or high-fat diet. To compare the metabolic differences between mice in different dietary levels and thereby secure better scientific data, mice should fast for 6 h in animal experiments. Fasting for 6 h is also recommended when comparing glucose tolerance with insulin tolerance.

Walnut kernels, selected as the raw material, underwent hydrolysis with protein-glutaminase (PG) following pulping. This process laid the foundation for producing set-type walnut yogurt, involving steps such as blending, homogenization, sterilization, and fermentation. The study investigated the impact of protein-glutaminase and five distinct starter cultures labeled I~V on the quality of walnut yogurt. This was evaluated by examining the yogurt's fermentation characteristics, physicochemical indicators, and microstructure. Furthermore, walnut yogurt developed under these optimal conditions was compared to traditional milk yogurt to assess its quality and characteristics. The findings revealed a significant improvement in the fermentation characteristics and textural parameters of yogurt fermented by walnut milk following enzymatic hydrolysis compared to non-enzymatic walnut milk. The optimal conditions were identified as 0.2% enzyme concentration and a 40-minute reaction time, with the pH, acidity, hardness, and consistency of walnut yogurt being 4.3, 55°T, 85.30 g, and 583.96 g·s, respectively. Walnut yogurt developed using starter culture I exhibited the highest acidity level, maintaining minimal fluctuation over a 21-day storage period. In contrast, yogurts prepared with alternative starter cultures demonstrated varying degrees of stratification and sedimentation. The yogurt fermented with culture I showed superior viscoelastic properties, presenting a denser gel network. Conversely, yogurts fermented with cultures IV and V displayed reduced apparent viscosity, lower values of G' (storage modulus), and G'' (loss modulus), along with a looser gel structure. Additionally, walnut yogurt produced with starter culture I maintained bacterial counts exceeding 106 CFU/mL throughout the storage duration. Despite walnut yogurt requiring further refinement in aspects such as consistency, water-holding capacity, and whey release rate compared to milk yogurt, the overall impact of PG was positive, enhancing the gelation quality of walnut yogurt. The yogurt prepared with starter culture I (Lactobacillus bulgaricus and Streptococcus thermophilus) was notably improved, receiving favorable overall assessments.

Neuroinflammation is considered the principal pathogenic mechanism underlying neurodegenerative diseases, and the incidence of brain disorders is closely linked to dietary fat consumption and intestinal health. To investigate this relationship, 60 8-week-old C57BL/6J mice were subjected to a 20-week dietary intervention, wherein they were fed lard and soybean oil, each at 15% and 35% fat energy. At a dietary fat energy level of 35%, inflammation was observed in both the soybean oil and lard groups. Nevertheless, inflammation was more pronounced in the mice that were administered soybean oil. The process by which nerve cell structure is compromised, inflammatory factors are upregulated, brain antioxidant capacity is diminished, and the TLR4/MyD88/NF-κB p65 inflammatory pathway is activated resulting in damage to the brain-gut barrier. This, in turn, leads to a reduction in the abundance of Akkermansia and unclassified_f_Lachnospiraceae, as well as an increase in Dubosiella abundance, ultimately resulting in brain inflammation and damage. These results suggested that soybean oil induces more severe neuroinflammation compared to lard. Our study demonstrated that, at a dietary fat energy level of 35%, compared to soybean oil, lard could be the healthier option, the outcomes would help provide a reference basis for the selection of residents’ daily dietary oil.

Carbapenem-resistant Acinetobacter baumannii (CRAB) is a Priority 1 (Critical) pathogen urgently requiring new antibiotics. Polymyxins are a last-line option against CRAB-associated infections. This transcriptomic study utilized a CRAB strain to investigate mechanisms of bacterial killing with polymyxin B, colistin, colistin B, and colistin/sulbactam combination therapy. After 4 h of 2 mg/L polymyxin monotherapy, all polymyxins exhibited common transcriptomic responses which primarily involved disruption to amino acid and fatty acid metabolism. Of the three monotherapies, polymyxin B induced the greatest number of differentially expressed genes (DEGs), including for genes involved with fatty acid metabolism. Gene disturbances with colistin and colistin B were highly similar (89 % common genes for colistin B), though effects on gene expression were generally lower (0–1.5-fold in most cases) with colistin B. Colistin alone (2 mg/L) or combined with sulbactam (64 mg/L) resulted in rapid membrane disruption as early as 1 h. Transcriptomic analysis of this combination revealed that the effects were driven by colistin, which included disturbances in fatty acid synthesis and catabolism, and inhibition of nutrient uptake. Combination therapy produced substantially higher fold changes in 72 % of DEGs shared with monotherapy, leading to substantially greater reductions in fatty acid biosynthesis and increases in biofilm, cell wall, and phospholipid synthesis. This indicates synergistic bacterial killing with the colistin/sulbactam combination results from a systematic increase in perturbation of many genes associated with bacterial metabolism. These mechanistic insights enhance our understanding of bacterial responses to polymyxin mono- and combination therapy and will assist to optimize polymyxin use in patients.

Abstract Background Chinese cherry [Cerasus pseudocerasus (Lindl.) G.Don] (syn. Prunus pseudocerasus Lindl.) is an economically important fruiting cherry species with a diverse range of attractive colors, spanning from the lightest yellow to the darkest black purple. However, the MYB transcription factors involved in anthocyanin biosynthesis underlying fruit color variation in Chinese cherry remain unknown. Results In this study, we characterized the R2R3-MYB gene family of Chinese cherry by genome-wide identification and compared it with those of 10 Rosaceae relatives and Arabidopsis thaliana. A total of 1490 R2R3-MYBs were classified into 43 subfamilies, which included 29 subfamilies containing both Rosaceae MYBs and AtMYBs. One subfamily (S45) contained only Rosaceae MYBs, while three subfamilies (S12, S75, and S77) contained only AtMYBs. The variation in gene numbers within identical subfamilies among different species and the absence of certain subfamilies in some species indicated the species-specific expansion within MYB gene family in Chinese cherry and its relatives. Segmental and tandem duplication events primarily contributed to the expansion of Chinese cherry R2R3-CpMYBs. The duplicated gene pairs underwent purifying selection during evolution after duplication events. Phylogenetic relationships and transcript profiling revealed that CpMYB10 and CpMYB4 are involved in the regulation of anthocyanin biosynthesis in Chinese cherry fruits. Expression patterns, transient overexpression and VIGS results confirmed that CpMYB10 promotes anthocyanin accumulation in the fruit skin, while CpMYB4 acts as a repressor, inhibiting anthocyanin biosynthesis of Chinese cherry. Conclusions This study provides a comprehensive and systematic analysis of R2R3-MYB gene family in Chinese cherry and Rosaceae relatives, and identifies two regulators, CpMYB10 and CpMYB4, involved in anthocyanin biosynthesis in Chinese cherry. These results help to develop and utilize the potential functions of anthocyanins in Chinese cherry.

Abstract Background Fatigue affects nurses negatively and may influence life satisfaction. According to the stress process model, active coping might influence the impact of adverse conditions such as fatigue on well-being measures such as life satisfaction. However, no research examined the associations among nurses’ fatigue, coping styles, and life satisfaction. Methods The cross-sectional study was conducted in Liaoning Province, China. 482 effective questionnaires were collected (effective response rate of 80.3%). The questionnaire included Fatigue Scale-14, Simplified Coping Style Questionnaire and Satisfaction with Life Scale. The association among fatigue, coping styles and fatigue × coping styles interaction with life satisfaction was examined by hierarchical multiple regression analysis. The interaction was visualized by simple slope analysis. Results Mean score of life satisfaction was 22.74 ± 6.11. Active coping moderated the relationship between mental fatigue and life satisfaction. Abstract Asparagus is a nutritionally dense stem vegetable whose growth and development are correlated with its quality and yield. To investigate the dynamic changes and underlying mechanisms during the elongation and growth process of asparagus stems, we documented the growth pattern of asparagus and selected stem segments from four consecutive elongation stages using physiological and transcriptome analyses. Notably, the growth rate of asparagus accelerated at a length of 25 cm. A significant decrease in the concentration of sucrose, fructose, glucose, and additional sugars was observed in the elongation region of tender stems. Conversely, the levels of auxin and gibberellins(GAs) were elevated along with increased activity of enzymes involved in sucrose degradation. A significant positive correlation existed between auxin, GAs, and enzymes involved in sucrose degradation. The ABA content gradually increased with stem elongation. The tissue section showed that cell elongation is an inherent manifestation of stem elongation. The differential genes screened by transcriptome analysis were enriched in pathways such as starch and sucrose metabolism, phytohormone synthesis metabolism, and signal transduction. The expression levels of genes such as ARF, GA20ox, NCED, PIF4, and otherswere upregulated during stem elongation, while DAO, GA2ox, and other genes were downregulated. The gene expression level was consistent with changes in hormone content and influenced the cell length elongation. Additionally, the expression results of RT-qPCR were consistent with RNA-seq. The observed variations in gene expression levels, endogenous hormones and sugar changes during the elongation and growth of asparagus tender stems offer valuable insights for future investigations into the molecular mechanisms of asparagus stem growth and development and provide a theoretical foundation for cultivation and production practices.

Background Numerous investigations have examined the potential link between allergic rhinitis (AR) and attention deficit hyperactivity disorder (ADHD). However, some studies show no association between the two diseases. The connection between these two conditions remains inconclusive. This study aimed to conduct a meta-analysis exploring the correlation between AR and ADHD. Methods We conducted systematic searches of the MEDLINE, EMBASE, Cochrane Library, ERIC, PubMed, Web of Science, and CINAHL databases, up to the year 2023. Subsequently, we conducted a meta-analysis using R 4.2.2, where we computed the pooled odds ratio with a 95% confidence interval to assess the relationship between AR and ADHD within studies exhibiting similar characteristics. Statistical heterogeneity was evaluated by computing the value using the Cochrane Intervention Manual’s guidelines. Additionally, subgroup analyses were conducted by stratifying the study population according to gender, age, etc. Sensitivity analysis was performed by systematically removing individual studies. Results In this systematic review, we incorporated 12 eligible studies, collectively encompassing a sample size of 530,360 participants. Within the included studies, heterogeneity was observed, and the utilization of a random-effects model demonstrated a noteworthy correlation between children with ADHD and the presence of AR. Similarly, children with AR exhibited a significant correlation with the occurrence of ADHD. We also found some relationships in subgroup analyses. Conclusion A substantial correlation is evident between AR and ADHD in children and adolescents. AR may potentially contribute as a risk factor for the onset of ADHD, and conversely, ADHD may heighten the likelihood of developing AR.

Objective: Behcet's uveitis (BU) is a type of uveitis with a high rate of blindness, characterized by anterior segment inflammation, vitreous opacity, and retinal vasculitis. Its pathogenesis is still unclear. Fuchs syndrome (Fuchs) is another common type of uveitis, which clinically presents with anterior segment inflammation and vitreous opacity, but rarely causes blindness. This study aims to compare the gut microbiota and metabolites of two different types of uveitis to clarify whether the differences in clinical manifestations are relevant to the alterations in gut microbiota. Methods: Faecal samples were collected from new-onset BU (n = 11) patients without systemic treatment and other diseases. 16S rRNA and liquid chromatography-tandem mass spectrometry (LC-MS/MS) were performed to analyze gut microbes and metabolites. Fuchs (n = 15) was used as the disease control, and healthy controls (n = 18) without autoimmune diseases and systemic medication were included. Results: Microbial composition and metabolite profiles differed significantly among the three groups. Compared to controls, Fusicatenibacter and eight metabolites were specifically altered in BU patients, and Pantoea and five metabolites in Fuchs. Pathways involving delta-tocopherol, palmitic acid, and serotonin are significantly disrupted in BU patients. Pathways involving linoleic acid are dysregulated considerably in Fuchs. Microbial markers consisting of 4 genera and 7 metabolites can respectively distinguish BU patients from controls. AUC values of metabolite markers were greater than those of microbial markers. Furthermore, serum zonulin levels were significantly elevated in both types of uveitis, with no difference between them. Correlation analysis revealed correlations between zonulin levels and multiple microbes. Conclusions: Patients with BU and Fuchs syndrome showed significant differences in gut microbiota and metabolites. Disruption of the intestinal mucosal barrier was observed in both types of uveitis. However, the mechanism of different intestinal microbiota causing different clinical manifestations needs to be studied in the future.

IntroductionThe objective of this study is to develop a model based on indicators in the routine examination of neonates to effectively predict neonatal apnea.MethodsWe retrospectively analysed 8024 newborns from the MIMIC IV database, building logistic regression models and decision tree models. The performance of the model is examined by decision curves, calibration curves and ROC curves. Variables were screened by stepwise logistic regression analysis and LASSO regression.ResultsA total of 7 indicators were ultimately included in the model: gestational age, birth weight, ethnicity, gender, monocytes, lymphocytes and acetaminophen. The mean AUC (the area under the ROC curve) of the 5-fold cross-validation of the logistic regression model in the training set and the AUC in the validation set are 0.879 and 0.865, respectively. The mean AUC (the area under the ROC curve) of the 5-fold cross-validation of the decision tree model in the training set and the AUC in the validation set are 0.861 and 0.850, respectively. The calibration and decision curves in the two cohorts also demonstrated satisfactory predictive performance of the model. However, the logistic regression model performs relatively well.DiscussionOur results proved that blood indicators were valuable and effective predictors of neonatal apnea, which could provide effective predictive information for medical staff.

This study investigates the evolution process of hot corrosion in bi-phase Ti-6.5Al-1.5Zr-3.5Mo-0.3Si (TC11) titanium alloy induced by sodium chloride (NaCl). Through morphological progression and elemental distribution analyses, the integrated processes involved in the onset of internal corrosion during hot corrosion are elucidated, revealing intriguing phenomena that deviate from conventional mechanisms. The development of corrosion can be divided into three stages: First there is the rapid growth of the external corrosion layer induced by Cl; then a Cl and O induced internal corrosion is initiated when the external corrosion layer reaches a critical thickness, at the same time the external corrosion decreases; finally, the chlorine pressure is too low to act as the corrosion depth increases, O becomes a major contributor to deep internal corrosion, leading to O induced internal corrosion. Significant interphase selective corrosion was observed in this internal corrosion zone: the β phase was corroded while the α phase remained uncorroded, challenging the prevailing notion that the β phase is more corrosion-resistant in titanium alloys. This distinctive phenomenon results from the differences in oxygen solubility, element diffusion rates and the reactivity between the α and β phases.

Bread wheat (Triticum aestivum) is a staple food crop worldwide. The genetic dissection of important nutrient traits is essential for the biofortification of wheat to meet the nutritional needs of the world’s growing population. Here, 45,298 single-nucleotide polymorphisms (SNPs) from 55K chip arrays were used to genotype a panel of 768 wheat cultivars, and a total of 154 quantitative trait loci (QTLs) were detected for eight traits under three environments by genome-wide association study (GWAS). Three QTLs (qMn-3B.1, qFe-3B.4, and qSe-3B.1/qFe-3B.6) detected repetedly under different environments or traits were subjected to subsequent analyses based on linkage disequilibrium decay and the P-values of significant SNPs. Significant SNPs in the three QTL regions formed six haplotypes for qMn-3B.1, three haplotypes for qFe-3B.4, and three haplotypes for qSe-3B.1/qFe-3B.6. Phenotypic analysis revealed significant differences among haplotypes. These results indicated that the concentrations of several nutrient elements have been modified during the domestication of landraces to modern wheat. Based on the QTL regions, we identified 15 high-confidence genes, eight of which were stably expressed in different tissues and/or developmental stages. TraesCS3B02G046100 in qMn-3B.1 and TraesCS3B02G199500 in qSe-3B.1/qFe-3B.6 were both inferred to interact with metal ions according to the Gene Ontology (GO) analysis. TraesCS3B02G199000, which belongs to qSe-3B.1/qFe-3B.6, was determined to be a member of the WRKY gene family. Overall, this study provides several reliable QTLs that may significantly affect the concentrations of nutrient elements in wheat grain, and this information will facilitate the breeding of wheat cultivars with improved grain properties.

Abstract Background Monkeypox is an emerging infectious disease with confirmed cases and deaths in several parts of the world. In light of this crisis, this study aims to analyze the global knowledge pattern of monkeypox-related patents and explore current trends and future technical directions in the medical development of monkeypox to inform research and policy. Methods A comprehensive study of 1,791 monkeypox-related patents worldwide was conducted using the Derwent patent database by descriptive statistics, social network method and linear regression analysis. Results Since the 21st century, the number of monkeypox-related patents has increased rapidly, accompanied by increases in collaboration between commercial and academic patentees. Enterprises contributed the most in patent quantity, whereas the initial milestone patent was filed by academia. The core developments of technology related to the monkeypox include biological and chemical medicine. The innovations of vaccines and virus testing lack sufficient patent support in portfolios. Conclusions Monkeypox-related therapeutic innovation is geographically limited with strong international intellectual property right barriers though it has increased rapidly in recent years. The transparent licensing of patent knowledge is driven by the merger and acquisition model, and the venture capital, intellectual property and contract research organization model. Currently, the patent thicket phenomenon in the monkeypox field may slow the progress of efforts to combat monkeypox. Enterprises should pay more attention to the sharing of technical knowledge, make full use of drug repurposing strategies, and promote innovation of monkeypox-related technology in hotspots of antivirals (such as tecovirimat, cidofovir, brincidofovir), vaccines (JYNNEOS, ACAM2000), herbal medicine and gene therapy.

Abstract The neural circuits underlying sleep-wakefulness and general anesthesia have not been fully investigated. The GABAergic neurons in the bed nucleus of the stria terminalis (BNST) play a critical role in stress and fear that relied on heightened arousal. Nevertheless, it remains unclear whether BNST GABAergic neurons are involved in the regulation of sleep-wakefulness and anesthesia. Here, using in vivo fiber photometry combined with electroencephalography, electromyography, and video recordings, we found that BNST GABAergic neurons exhibited arousal-state-dependent alterations, with high activities in both wakefulness and rapid-eye movement sleep, but suppressed during anesthesia. Optogenetic activation of these neurons could initiate and maintain wakefulness, and even induce arousal from anesthesia. However, chronic lesion of BNST GABAergic neurons altered spontaneous sleep-wakefulness architecture during the dark phase, but not induction and emergence from anesthesia. Furthermore, we also discovered that the BNST-ventral tegmental area pathway might participate in promoting wakefulness and reanimation from steady-state anesthesia. Collectively, our study explores new elements in neural circuit mechanisms underlying sleep-wakefulness and anesthesia, which may contribute to a more comprehensive understanding of consciousness and the development of innovative anesthetics.

Abstract Kuding Tea (Ilex latifolia) is a bitter‐tasting herbal tea that was used for the treatment of symptoms related with diabetes mellitus, hypertension, and hyperlipidemia. However, Kuding Tea is also difficultly accepted by people in daily life because of its poor palatability. In this study, Kuding Tea, green tea (GT) (Camellia sinensis L.), and Luohan (Siraitia grosvenorii) fruits were formulated into a compound Kuding Tea (CKT) to improve the taste and health benefits of this beverage. High‐fat diet‐fed male C57BL/6J mice were used as animal models to explore the effects of CKT (6 or 12 mg/mL, water ad libitum) on body weight, food intake, liver function, blood glucose and lipids, and gene expression. L02 and 3T3‐L1 cells were used to further demonstrate the effects of CKT on fat accumulation and hepatic lipid deposition. Our results suggest that CKT can regulate glucose and lipid metabolism by decreasing body weight, reducing white adipose deposition, improving glucose tolerance, increasing the expression of brown adipose genes, and reducing fat accumulation in the liver, and CKT inhibited fat accumulation better than GT. In addition, a low dose (6 mg/mL) of CKT reduced the abundance of Desulfovibrio bacteria, positively associated with obesity, and increased that of norank_f__Muribaculaceae, Lachnospiraceae_NK4A136_group, and Alloprevotella, which are beneficial to glucose and lipid metabolism. This study suggests that CKT not only has a better palatability but also has potential preventive effects on high‐fat diet‐induced glucose–lipid metabolic diseases.

Abstract Single-cell whole-genome sequencing methods have undergone great improvements over the past decade. However, allele dropout, which means the inability to detect both alleles simultaneously in an individual diploid cell, largely restricts the application of these methods particularly for medical applications. Here, we develop a new single-cell whole-genome sequencing method based on third-generation sequencing (TGS) platform named Refresh-seq (restriction fragment ligation-based genome amplification and TGS). It is based on restriction endonuclease cutting and ligation strategy in which two alleles in an individual cell can be cut into equal fragments and tend to be amplified simultaneously. As a new single-cell long-read genome sequencing method, Refresh-seq features much lower allele dropout rate compared with SMOOTH-seq. Furthermore, we apply Refresh-seq to 688 sperm cells and 272 female haploid cells (secondary polar bodies and parthenogenetic oocytes) from F1 hybrid mice. We acquire high-resolution genetic map of mouse meiosis recombination at low sequencing depth and reveal the sexual dimorphism in meiotic crossovers. We also phase the structure variations (deletions and insertions) in sperm cells and female haploid cells with high precision. Refresh-seq shows great performance in screening aneuploid sperm cells and oocytes due to the low allele dropout rate and has great potential for medical applications such as preimplantation genetic diagnosis.

Abstract Background Late embryogenesis abundant (LEA) proteins play important roles in plant growth and development, as well as stresses responsiveness. Nowadays, it has been found that LEAs also have function in fruit ripening. However, the comprehensive analysis on a genome-wide basis of LEA family remains limited, and the role of LEA in fruit ripening has not been fully explored yet, especially in strawberry, an economic important plant and ideal material for studying fruit ripening. Results In this study, a total of 266 putative LEA proteins were identified and characterized in strawberry genome. Subcellular localization prediction indicated that they were mostly localized in chloroplast, cytoplasm and nucleus. Duplication events detection revealed that whole genome duplication or segmental was the main driver for the expansion of LEA family in strawberry. The phylogenetic analysis suggested that FaLEAs were classified into eight groups, among which, LEA2 was the largest subgroup with 179 members, followed by LEA3, dehydrin (DHN), LEA4 and SMP (seed maturation protein). The LEA1 and DHN groups were speculated to play dominant roles in strawberry fruit development and ripening, according to their larger proportion of members detected as differentially expressed genes during such process. Notably, the expression of FaLEA167 belonging to LEA1 group was altered by strawberry maturation, and inhibited by overexpression of negative regulators of ripening (a cytosolic/plastid glyceraldehyde-3-phosphate dehydrogenase, FaGAPC2 and a cytosolic pyruvate kinase, FaPKc2.2). Subsequently, overexpression of FaLEA167 significantly increased the percentage of fruit at green stage, while reduced the full red fruit proportion. In consistent, the anthocyanins content and the fruit skin color variable reflecting a range from greenness to redness (a* value) were significantly reduced. Whereas, FaLEA167 overexpression apparently up-regulated citric acid, soluble protein and malondialdehyde content, but had no obvious effects on total soluble solids, sugar, flavonoids, phenolics content and antioxidant capacity. Conclusions These findings not only provided basic information of FaLEA family for further functional research, but also revealed the involvement of FaLEA167 in negatively regulating strawberry fruit ripening, giving new insights into understanding of FaLEA functions.

The red color of red pears is primarily attributed to the accumulation of anthocyanins, which are biosynthesized via the phenylalanine pathway. However, research into the regulation of these processes is currently limited. The study found that increasing the expression of the transcription factor PbMYB5-like resulted in increased expression of genes involved in anthocyanin, proanthocyanidin (PAs), and lignin biosynthesis, as well as anthocyanin accumulation in pear epidermis. The experiments demonstrated that PbMYB5-like interacts with bHLH3, resulting in the formation of the PbMYB5 like-bHLH3-WD40 complex, which regulates the expression of structural genes such as CCR, F5H, LAR, ANS, UFGT, and others. Furthermore, PbMYB5-like has been demonstrated to directly bind to the promoters of CHI, F3H, ANS, and UFGT genes, thereby regulating the synthesis of related metabolites. The findings of this study provide a foundation for understanding the mechanisms by which PbMYB5-like regulates pear skin color, which is a pivotal aspect in pear breeding and germplasm innovation.

Background Primary health care plays an important role in providing populations with access to health care. However, it is currently facing unprecedented workforce shortages and high turnover worldwide. Objective This study examined the relationship between organizational culture and turnover intention among primary care providers in China. Methods A cross-sectional survey was administered in four large cities in China, Tianjin, Jinan, Shanghai, and Shenzhen, comprising 38 community health centers and 399 primary care providers. Organizational culture was measured using the Competing Value Framework model, which is divided into four culture types: group, development, hierarchy, and rational culture. Turnover intention was measured using one item assessing participants’ intention to leave their current position in the following year. We compared the turnover intention among different organizational culture types using a Chi-square test, while the hierarchical logistic regression was used to examine the relationship between organizational culture and turnover intention. Results The study found that 32% of primary care providers indicated an intention to leave. ABSTRACTDeeper integration of cross-organizational business process sharing and process mining has advanced the Industrial Internet. Privacy breaches and data security risks limit its use. Scrambling or anonymizing event data frequently preserves privacy in established studies. The scrambling mechanism or random noise injection corrupts event log process information and lowers process mining outcomes. This research presents a blockchain-based privacy-aware reversible shared image approach using chaotic image and privacy-aware theory for privacy-preserving process mining. Avoiding data loss, disclosure concerns, correlation attacks, and encrypted sharing is possible with the method. First, process data is turned into color images with chaotic image encryption to safeguard privacy and allow reversible reproduction. Second, the on-chain-off-chain paradigm helps handle information lightly; finally, attribute encryption of multi-view event data for correlation resistance and on-demand data encryption sharing. Simulations on common datasets reveal that: 1. The system performance of the proposed method outperforms the baseline method by 57%. 2. The strategy greatly enhances categorical and numerical data privacy. 3. It performs better in event data privacy protection and process mining fitness and precision. The proposed method ensures the secure flow of cross-organizational information in the Industrial Internet and provides a novel privacy-secure computational approach for the growing Artificial Intelligence.

Background Health information technology is one of the building blocks of a high-performing health system. However, the evidence regarding the influence of an electronic health record (EHR) on the quality of care remains mixed, especially in low- and middle-income countries. Objective This study examines the association between greater EHR functionality and primary care physician self-reported quality of care. Methods A total of 224 primary care physicians from 38 community health centres (CHCs) in four large Chinese cities participated in a cross-sectional survey to assess CHC care quality. Each CHC director scored their CHC’s EHR functionality on the availability of ten typical features covering health information, data, results management, patient access, and clinical decision support. Data analysis utilised hierarchical linear modelling. Results The availability of five EHR features was positively associated with physician self-reported clinical quality: share records online with providers outside the practice (β = 0.276, p = 0.04), access records online by the patient (β = 0.325, p = 0.04), alert provider of potential prescription problems (β = 0.353, p = 0.04), send the patient reminders for care (β = 0.419, p = 0.003), and list patients by diagnosis or health risk (β = 0.282, p = 0.04). However, no association was found between specific features availability or total features score and physician self-reported preventive quality. Conclusions This study provides evidence that the availability of EHR systems, and specific features of these systems, was positively associated with physician self-reported quality of care in these 38 CHCs. Future longitudinal studies focused on standardised quality metrics, and designed to control known confounding variables, will further inform quality improvement efforts in primary care.

IntroductionSelenium (Se) plays a vital role in various physiological processes in plants and is regarded as an essential micronutrient for human health as well.MethodsIn this study, sodium selenite solution at 10, 40, 70, and 100 mg·L-1 concentrations was foliar sprayed, and the strawberry plant growth, antioxidant system, and fruit quality with an emphasis on sugar and acid content were assessed.ResultsThe results showed that 10 mg·L-1 of sodium selenite treatment promoted plant growth, while all the treated concentrations could enhance photosynthesis, the antioxidant system in leaves, the content of Se, and ascorbic acid in fruits. More importantly, 40 mg·L-1 sodium selenite treatment significantly increased fruit weight, total soluble solid, total phenolic content, and anthocyanins, as well as improved the shape index. Furthermore, it decreased the total flavonoid and proanthocyanidin content. Particularly, sodium selenite treatment at 40 and 70 mg·L-1 largely increased the ratio of soluble sugars to titratable acid. The changes of predominant sugars and organic acids during fruit development were further investigated. The sucrose, fructose, and glucose content was upregulated by sodium selenite treatment through upregulating the activities of sucrose phosphate synthase (SPS) and acid invertase, as well as the FaSPS expression. In addition, sodium selenite treatment inhibited the activity of citrate synthase and phosphoenolpyruvate carboxylase, rather than modulating their transcript levels to reduce the citric acid content.ConclusionsThis work presented a potentially efficient approach to enhance plant growth and fruit quality and supplement Se in strawberry, providing insights into the mechanism of regulating sugar and acid metabolism by Se.

Abstract The Tibetan Plateau (TP) is suffering from a substantial decline in terrestrial water storage (TWS) in exorheic basins, threatening water resources that are critical for ∼2 billion people downstream. TWS changes are commonly estimated using gravity satellites through observations of the total terrestrial mass storage (TMS) change, with an implicit assumption of a negligible contribution from sediment transport. Through long‐term (2002–2017) sediment flux observations in seven headwater basins on the TP, we reveal that the gravity satellite‐derived TMS has decreased at a rate of 3.85 ± 0.23 Gt yr−1 in the seven basins, of which 0.35 ± 0.04 Gt yr−1 is contributed by sediment transport. Neglecting this contribution leads to an overestimation of the TWS loss by 10.1 ± 1.3%, equivalent to the annual water demand of an additional 0.62 million people in the surrounding nations. Regionally, the overestimation is surprisingly high in the Indus River and Yarkant River basins, reaching up to 50.8%–77.6%.

Background Chemotactic cytokines play a crucial role in the development of acute myeloid leukemia (AML). Thus, investigating the mechanisms of chemotactic cytokine-related genes (CCRGs) in AML is of paramount importance. Methods Using the TCGA-AML, GSE114868, and GSE12417 datasets, differential expression analysis identified differentially expressed CCRGs (DE-CCRGs). These genes were screened by overlapping differentially expressed genes (DEGs) between AML and control groups with CCRGs. Subsequently, functional enrichment analysis and the construction of a protein-protein interaction (PPI) network were conducted to explore the functions of the DE-CCRGs. Univariate Cox regression, least absolute shrinkage and selection operator (LASSO), and multivariate Cox regression analyses identified relevant prognostic genes and developed a prognostic model. Survival analysis of the prognostic gene was performed, followed by functional similarity analysis, immune analysis, enrichment analysis, and drug prediction analysis. Results Differential expression analysis revealed 6,743 DEGs, of which 29 DE-CCRGs were selected for this study. Functional enrichment analysis indicated that DE-CCRGs were primarily involved in chemotactic cytokine-related functions and pathways. Six prognostic genes (CXCR3, CXCR2, CXCR6, CCL20, CCL4, and CCR2) were identified and incorporated into the risk model. The model’s performance was validated using the GSE12417 dataset. Survival analysis showed significant differences in AML overall survival (OS) between prognostic gene high and low expression groups, indicating that prognostic gene might be significantly associated with patient survival. Additionally, nine different immune cells were identified between the two risk groups. Correlation analysis revealed that CCR2 had the most significant positive correlation with monocytes and the most significant negative correlation with resting mast cells. The tumor immune dysfunction and exclusion score was lower in the high-risk group. Conclusion CXCR3, CXCR2, CXCR6, CCL20, CCL4, and CCR2 were identified as prognostic genes correlated to AML and the tumor immune microenvironment. These findings offerred novel insights into the prevention and treatment of AML.

Kiwifruit bacterial canker, caused by Pseudomonas syringae pv. actinidiae (PSA), poses a grave threat to the global kiwifruit industry. In this study, we examined the role of microRNAs (miRNAs) in kiwifruit’s response to PSA. Kiwifruit seedlings subjected to PSA treatment showed significant changes in both miRNA and gene expression compared to the control group. We identified 364 differentially expressed miRNAs (DEMs) and 7170 differentially expressed genes (DEGs). Further analysis revealed 180 miRNAs negatively regulating 641 mRNAs. Notably, two miRNAs from the miRNA482 family, miRNA-215-3p and miRNA-29-3p, were found to increase kiwifruit’s sensitivity to PSA when overexpressed. These miRNAs were linked to the regulation of NBS-LRR target genes, shedding light on their role in kiwifruit’s defence against PSA. This study offers insights into the miRNA482-NBS-LRR network as a crucial component in enhancing kiwifruit bioresistance to PSA infestation and provides promising candidate genes for further research.

The development of innovative therapeutic strategies for head and neck squamous cell carcinoma (HNSCC) is a critical medical requirement. Antibody-drug conjugates (ADC) targeting tumor-specific surface antigens have demonstrated clinical effectiveness in treating hematologic and solid malignancies. Our investigation revealed high expression levels of SLC3A2 in HNSCC tissue and cell lines. This study aimed to develop a novel anti-SLC3A2 ADC and assess its antitumor effects on HNSCC both in vitro and in vivo. This study developed a potent anti-SLC3A2 ADC (19G4-MMAE) and systematically investigated its drug delivery potential and antitumor efficacy in preclinical models. This study revealed that 19G4-MMAE exhibited specific binding to SLC3A2 and effectively targeted lysosomes. Moreover, 19G4-MMAE induced a significant accumulation of reactive oxygen species (ROS) and apoptosis in SLC3A2-positive HNSCC cells. The compound demonstrated potent antitumor effects derived from MMAE against SLC3A2-expressing HNSCC in preclinical models, displaying a favorable safety profile. These findings suggest that targeting SLC3A2 with an anti-SLC3A2 ADC could be a promising therapeutic approach for treating HNSCC patients.

Introduction Acute-on-chronic liver failure (ACLF) is a prevalent and life-threatening liver disease with high short-term mortality. Although recent clinical trials on the use of mesenchymal stem cells (MSCs) for ACLF treatment have shown promising results, multicentre randomised controlled phase II clinical trials remain uncommon. The primary aim of this trial is to assess the safety and efficacy of different MSCs treatment courses for ACLF.Methods and analysis This is a multicentre, double-blind, two-stage, randomised and placebo-controlled clinical trial. In the first stage, 150 patients with ACLF will be enrolled and randomly assigned to either a control group (50 cases) or an MSCs treatment group (100 cases). They will receive either a placebo or umbilical cord-derived MSCs (UC-MSCs) treatment three times (at weeks 0, 1 and 2). In the second stage, 28 days after the first UC-MSCs infusion, surviving patients in the MSCs treatment group will be further randomly divided into MSCs-short and MSCs-prolonged groups at a 1:1 ratio. They will receive two additional rounds of placebo or UC-MSCs treatment at weeks 4 and 5. The primary endpoints are the transplant-free survival rate and the incidence of treatment-related adverse events. Secondary endpoints include international normalised ratio, total bilirubin, serum albumin, blood urea nitrogen, model for end-stage liver disease score and Child-Turcotte-Pugh score.Ethics and dissemination Ethical approval of this study has been obtained from the Fifth Medical Center of the Chinese PLA General Hospital (KY-2023-3-19-1). All results of the study will be submitted to international journals and international conferences for publication on completion of the study.Trial registration number NCT05985863.

The energy demand has increased significantly in recent years and it is urgent to develop a renewable energy system that is highly efficient and non-noble metal-based. Hydrogen energy is an environmentally friendly energy source with abundant resources, which can be used to solve the problem of high energy demand without greenhouse gas emissions. However, the development of catalysts for hydrogen production technology by electrolysis of water is slow, mainly due to the complexity of the electrolysis hydrogen generation process, low hydrogen production efficiency, weak electrode material activity and high cost. Among the non-noble metal-based catalysts, carbon-based materials have high conductivity, tunable chemical bonding, and easily modified morphology, making them beneficial to achieving efficient hydrogen production, though pure carbon composites suffer from few surface-active sites and unmoderated hydrogen bonding energy, which need to be further optimized. The principle of electrocatalytic hydrogen production from the perspectives of reaction thermodynamics and kinetics is analyzed and discussed in this paper. Thermodynamics of electrocatalytic hydrogen production is reflected by the Gibbs free energy of hydrogen adsorption (ΔGH*) and electrode potential (E). Reaction kinetics of the electrocatalytic hydrogen production process are reflected by overpotential, Tafel slope and exchange current density. Structural optimization methods of carbon-based composite materials and hydrogen production performance after structural optimization are also summarized. Structural optimization methods of carbon-based composite materials mainly include introducing active sites, improving conductivity, increasing specific surface area and introducing self-supporting materials. Finally, prospects are proposed for the development direction and existing problems of electrocatalytic hydrogen production performance of carbon-based composites.

China has made remarkable progress in controlling particulate matter, while O3 pollution over China has become increasingly severe in recent years according to ground observations. Continuous monitoring of dynamic changes in O3 concentrations on regional and national scales can provide valuable insights for pollution control policies. Therefore, an improved similarity distance-based space-time random forest (SDSTRF) model was developed to estimate the near-surface O3 concentration using the surface measurements, satellite O3 precursors, meteorological variables, and other auxiliary information. The O3 concentration data over China were generated based on the developed model with a spatial resolution of 10 km and a temporal resolution of 1 day from 2016 to 2022. The validation results against the ground measurements indicate that the developed SDSTRF model effectively captures O3 variations, achieving a coefficient of determination of 0.83 and a root mean square error of 20.37 μg/m3. The spatiotemporal variations of O3 concentrations were investigated using the generated dataset. A significant increasing trend of 1.243 μg/m3/yr in O3 concentrations was observed in eastern China during the COVID-19 pandemic, which was attributed to changes in NOx concentrations. In this study, the possible reasons for the increase in O3 concentrations are also discussed. Overall, the improved SDSTRF model and the comprehensive analysis of the spatiotemporal variations of near-surface O3 will significantly contribute to achieving clean air in China.

Xanthoceras sorbifolium Bunge oil exhibits favorable characteristics such as excellent low temperature fluidity, high flash point, and degradability. However, it also suffers from drawbacks including low kinematic viscosity at low temperatures and significant loss of kinematic viscosity at high temperatures. In view of these characteristics of X. sorbifolium oil, it was modified in this work with a combination of ethyl cellulose (EC), montmorillonite (MMT), and cetyltrimethylammonium chloride (CTAB). The effects of temperature, time, and additive content on the rheological properties of lubricating oil were studied. The kinematic viscosity of the prepared lubricating oil reached up to 433 mm2/s at 40 °C and 51.1 mm2/s at 100 °C. The coagulation point could be reduced to -25 °C, the friction coefficient was 0.026, and the average wear spot diameter was 0.81 mm, The anti-friction performance was enhanced, and the anti-wear performance decreased somewhat. The prepared lubricating oil had the characteristics of high kinematic viscosity at high temperature and excellent rheological properties at low temperature. It also met the energy and environmental application requirements of a green, environmentally friendly, and biodegradable sustainable development strategy. This study greatly broadens the application range of lubricating oil.

Background: Src homology 2 domain-containing phosphatase 2 (SHP2) is hyper-activated in some solid tumors. Previous findings suggest that the expression of SHP2 in colorectal cancer (CRC) may be associated with prognosis. However, validation with large sample data is lacking. Materials and Methods: Tissue microarrays containing 860 CRCs and 197 mucosal tissues adjacent to the tumors were constructed. Immunohistochemistry was used to evaluate the expression of SHP2. Differences between SHP2 expression and clinicopathological parameters were evaluated. Kaplan–Meier survival curves and log-rank tests were used to analyze the relationships between SHP2 expression and the overall survival of patients. A Cox proportional hazard regression model was used for univariate and multivariate analyses of prognostic factors. Results: SHP2 expression in CRCs tissues was significantly higher than those in adjacent mucosal tissues (P < 0.001). SHP2 expression was related to tumor differentiation, depth of invasion, distant metastasis, vascular tumor thrombus, lymph node metastasis, and TNM classification (P < 0.05). The prognosis of the high-expression group of SHP2 was significantly better than that of the low-expression group (P = 0.008). Univariate analysis showed that the expression of SHP2 was a prognostic factor for CRC (P = 0.008). Multivariate analysis demonstrated that SHP2 remained an independent prognostic factor for CRC (P = 0.033). Conclusion: The expression of SHP2 was significantly higher in CRC tissues than in adjacent normal tissues. High expression of SHP2 was associated with a promising outcome, suggesting that SHP2 may be a favorable prognostic indicator of CRC.

Citrus is one of the world's most economically important fruit crops cultivated by grafting. To support the growth of scion cultivars, rootstock is the primary source of resistance to various abiotic stresses. Herein, seedlings of two genotypes of Citrus junos Sieb. ex Tanaka (the novel rootstock 'Shuzhen No.1' and commonly used rootstock 'Ziyang Xiangcheng'), as well as three commonly used rootstocks including citrange (Citrus sinensis Osbeck. × Poncirus trifoliata Raf.), trifoliate orange (P. trifoliata), and red tangerine (Citrus tangerine Hort. Ex Tanaka), were used as testing materials. The seed characteristics were evaluated, and the rootstock seedlings were subjected to flooding, drought, alkaline, and freezing treatments. Over time, the contents of chlorophyll, soluble sugar, proline, malondialdehyde, and the activity of superoxide dismutase, peroxidase, and catalase in the leaves under different treatments were examined. Furthermore, five citrus varieties were grafted as scions onto one-year-old seedlings from the four rootstocks. Graft success, shoot growth, and leaf greenness were measured and compared. The physiological and biochemical changes in 'Shuzhen No.1' were found to be similar to those in 'Ziyang Xiangcheng'. 'Shuzhen No.1' exhibited greater tolerance to flooding, alkaline, and freezing stress compared to the other four widely used citrus rootstocks, as indicated by physiological and biochemical indexes and principal component analysis. Moreover, the five citrus varieties grafted onto 'Shuzhen No.1' demonstrated vigorous growth and tree vigor. These findings provide valuable insights for the application of 'Shuzhen No.1' and future research on citrus rootstock.

The study examines the corrosion behavior of the (NdSmEuGd)(1-x)/2Dy2xZr2O7 series in 1300 °C molten salt CMAS, demonstrating that the dense layer formed by ZrO2 and apatite could effectively inhibit the penetration of CMAS. Smaller rare-earth ion radii tend to form larger-sized ZrO2, while larger rare-earth ion radii favor apatite formation. Therefore, by carefully balancing the rare earth ion radii in the composition design of high-entropy samples, the levels of ZrO2 and apatite formation can be adjusted to prevent CMAS infiltration. This approach helps identify high-entropy zirconate samples with superior corrosion resistance to CMAS.

The immune microenvironment plays a vital role in bone defect repair. To create an immune microenvironment that promotes osteogenesis, researchers are exploring ways to enhance the differentiation of M2-type macrophages. Functional peptides have been discovered to effectively improve this process, but they are limited by low efficiency and rapid degradation in vivo. To overcome these issues, peptide with both M2 regulatory and self-assembly modules was designed as a building block to construct an ultrasound-responsive nanofiber hydrogel. These nanofibers can be released from hydrogel in a time-dependent manner upon ultrasound stimulation, activating mitochondrial glycolytic metabolism and the tricarboxylic acid cycle, inhibiting reactive oxygen species production and enhancing M2 macrophage polarization. The hydrogel exhibits advanced therapeutic potential for bone regeneration by triggering M2 macrophages to secrete BMP-2 and IGF-I, accelerating the differentiation of bone marrow mesenchymal stem cells (BMSCs) into osteoblasts. Thus, modularly designed biomimetic ultrashort peptide nanofiber hydrogels provide a novel strategy to rebuild osteogenic immune microenvironments for bone repair.

Background: The SYNTAX score II (SS II) has earned widespread recognition for use on individuals with coronary artery disease (CAD) due to its reliable predictions of 4-year all-cause mortality (ACM). This research focuses on substantiating the prognostic significance of using the SS II for patients experiencing concurrent chronic renal insufficiency (CRI) and CAD who have undergone percutaneous coronary intervention (PCI). Methods: This study retrospectively examined 2468 patients with concurrent CAD and CRI who underwent PCI. Based on their SS II, these participants were sorted into low-, medium-, and high-risk groups and monitored over a median of three years. The evaluation of the predictive precision of different SYNTAX scores for clinical outcomes in patients with CRI after PCI involved using time-dependent receiver operating characteristic (ROC) curves. These included the standard SS (SS), SS II, clinical SS (CSS), and residual SS (rSS). The primary outcomes were ACM and cardiac mortality (CM), while the secondary outcomes covered major adverse cardiovascular and cerebrovascular events (MACCEs), stroke, unplanned revascularization, and myocardial infarction (MI). Results: Higher 5-year cumulative incidences of MACCEs, MI, CM, and ACM were observed significantly in patients in the high SS II category relative to those in the low and medium SS II categories. Multivariable Cox regression analysis confirmed that the SS II independently predicts ACM, CM, MI, and MACCEs as a prognostic marker. Additionally, the analysis of the time-dependent ROC curve demonstrated that the areas under the curve (AUC) for predicting CM and ACM were 0.772 and 0.767, respectively, which are superior to those of other SYNTAX scores (p < 0.05). Conclusions: As an independent predictor, the SS II is notable for its ability to forecast long-term adverse outcomes, including MACCEs, CM, ACM, and MI. For patients with coexisting CAD and CRI undergoing PCI, it provides significantly improved prognostic accuracy for 5-year ACM and CM compared to other SYNTAX scores.

The role of biological soil crusts (biocrusts) in ecological restoration is dominated by microbiota. This has been extensively studied in arid ecosystems characterized by nutrient deficiencies and poor soil substrates, including metal(loid)-rich mining regions. However, in the prior study areas, water constraints obscured the toxicity and restriction of succession by heavy metals, which were essential for soil remediation in mining regions using biocrusts. Accordingly, in the current study, we characterized biocrusts from a typical polymetallic tailing wetland and performed pot incubation to evaluate microbial community succession and microbial co-occurrence patterns. Results showed that adequate water and heavy-metal stress produced thicker and more hydrophobic biocrusts than did arid conditions, promoting stronger microbial activity in tailing wetlands. Moreover, the bioinformatic analyses during pot incubation suggest three potential survival strategies. (1) Cyanobacteria variation improved the succession level of cyanobacterial crusts. Even under heavy-metal stress, succession from light to dark cyanobacterial crusts still occurred, with Coleofasciculaceae decreasing 63% and Scytonema increasing 59% compared with the control groups. (2) Keystone taxa variation maintained high inorganic nitrogen turnover. Organisms potentially capable of inorganic nitrogen turnover, including Chloroflexi A4b and SBR1031, maintained a high relative abundance accounting for 42% (15/36) of the keystone taxa. (3) Variations in the interactions among microbiota created closer-knit microbial networks. Compared with the control groups, fewer nodes (15.2%) maintained higher average clustering coefficient values (1.8%) through 14% more positive interactions in the groups exposed to heavy-metal stress at the end of the incubation experiment. This study provides insights into the distinctive survival strategies of biocrusts in metal-rich extreme environments, offering theoretical support for biocrust-mediated ecological restoration in humid metal-contaminated regions.

The capability of 5, 10, 15 mM γ-aminobutyric acid (GABA) to improve the postharvest quality and antioxidant system of strawberry was evaluated in this study. The application of GABA had no effect on fruit skin color and firmness. The weight loss in fruits treated with 10 mM GABA was significantly lower than the control. GABA treatments resulted in higher levels of total soluble sugar, titratable acid, SOD and CAT activities with 10 mM being the most significant effect. Specifically, 10 mM GABA significantly induced the accumulation of fructose, oxalic acid, and succinic acid. Besides, GABA application increased the content of total anthocyanins and total flavonoids, and DPPH radical scavenging activity in fruits. The GABA-treated fruits especially at 5 mM and 10 mM displayed less ROS and MDA. These data suggested that application of 10 mM GABA might be a promising strategy to improve the postharvest marketability of strawberry.

Asparagus, characterized by its high metabolic rate, is susceptible to quality degradation. Proanthocyanidins have antioxidant, antibacterial, antiviral, and other biological functions and can inhibit the production of reactive oxygen species in plants. To enhance the shelf life of asparagus, we investigated the impact of various concentrations of proanthocyanidins on its cold storage and preservation. The findings revealed that proanthocyanidins effectively mitigated water loss, delayed chlorophyll degradation, and prevented firmness decline. Furthermore, they enhanced the activity of antioxidant enzymes (superoxide dismutase, catalase, peroxidase, and polyphenol oxidase), bolstered DPPH free radical scavenging ability, and increased the levels of total phenol, total flavone, rutin, oligomeric procyanidins, proline, and soluble protein. Moreover, proanthocyanidins promoted the accumulation of vitamin C, amino acids, total saponins, and lignin in the later storage stage, contributing to increased mechanical tissue thickness. These results suggest that proanthocyanidins play a crucial role in retarding the deterioration of asparagus quality during storage by affecting the antioxidant capacity and phytochemical (polyphenol,amino acid, total saponin, and lignin) synthesis in asparagus.

Abstract Increasing evidence has confirmed the vital role of Notch signaling in the tumorigenesis of clear cell renal cell carcinoma (ccRCC). The underlying function of long non-coding RNA (lncRNA) related to Notch signaling in ccRCC remains unclear. In present study, the prognostic value and therapeutic strategy of Notch signaling-related lncRNA are comprehensively explored in ccRCC. In total, we acquired 1422 NSRlncRNAs, of which 41 lncRNAs were identified the key NSRlncRNAs associated with the occurrence of ccRCC. The prognostic signature containing five NSRlncRNAs (AC092611.2, NNT-AS1, AGAP2-AS1, AC147651.3, and AC007406.3) was established and validated, and the ccRCC patients were clustered into the high- and low-risk groups. The overall survival of patients in the low-risk group were much more favorable than those in the high-risk group. Multivariate Cox regression analysis indicated that the risk score was an independent prognostic biomarker. Based on the risk score and clinical variables, a nomogram for predicting prognosis of ccRCC patients was constructed, and the calibration curves and DCA curves showed the superior predictive ability of nomogram. The risk score was correlated with immune cell infiltration, targeted therapy or chemotherapy sensitivity, and multiple oncogenic pathways. Additionally, consensus clustering analysis stratified the ccRCC patients into four clusters with obvious different outcomes, immune microenvironments, and expression of immune checkpoints. The constructed NSRlncRNA-based signature might serve as a potential biomarker for predicting prognosis and response to immunotherapy or targeted therapy in patients with ccRCC.

In this study, the influence of nanocrystalline structure on the plasma nitriding (PN) behavior of 17-4PH steel and corresponding corrosion mechanisms under different treatments were systematically investigated. The results showed that nanocrystalline structures introduced by ultrasonic surface rolling process (USRP) would affect the nitrides morphology, composition and distribution via accelerating element diffusion. The fabricated nanocrystalline structures could enhance the corrosion resistance of original and plasma-nitrided 17-4PH steel, which was attributed to its various microstructures. Homogeneous microstructures suppressed or alleviated the formation of micro-galvanic couple in USRP-treated 17-4PH steel and USRP-15 + PN specimen.

The microenvironment of the wound bed is essential in the regulation of wound repair. In this regard, strategies that provide a repairing favorable microenvironment may effectively improve healing outcomes. Herein, we attempted to use electrical stimulation (ES) to boost the paracrine function of adipose-derived stem cells from rats (rASCs). By examining the concentrations of two important growth factors, VEGF and PDGF-AA, in the cell culture supernatant, we found that ES, especially 5 μA ES, stimulated rASCs to produce more paracrine factors (5 μA-PFs). Further studies showed that ES may modulate the paracrine properties of rASCs by upregulating the levels of TRPV2 and TRPV3, thereby inducing intracellular Ca2+ influx. To deliver the PFs to the wound to effectively improve the wound microenvironment, we prepared a heparinized PGA host-guest hydrogel (PGA-Hp hydrogel). Moreover, PGA-Hp hydrogel loaded with 5 μA-PFs effectively accelerated the repair process of the full-thickness wound model in rats. Our findings revealed the effects of ES on the paracrine properties of rASCs and highlighted the potential application of heparinized PGA host-guest hydrogels loaded with PFs derived from electrically stimulated rASCs in wound repair.

An accurate gas utilization model is essential for precisely detecting plant photosynthetic capacity. Existing equipment for measuring the plant photosynthetic rate typically considers the key parameters of mesophyll cell conductance and a photosynthetic model based on the carbon reaction process under direct light conditions. However, the light environment signals received by the plant canopy not only vary significantly in incidence angles, but the effective light intensity also differs greatly from the measured values under vertical incidence conditions. To reduce the deviation between existing photosynthetic models and the actual photosynthetic efficiency of leaves, this study employs the gas diffusion method from engineering, using the finite element approach. Based on elastic mechanics and seepage mechanics, the internal stress field control equation of tomato leaves and the two-phase flow equation under a CO2 porous medium were derived. A mathematical model of porous gas–liquid two-phase fluid-solid coupling was established, solved, and analyzed. Preliminary verification was conducted through tests. The results show that in the initial stage of CO2 entering the leaf, the gas flow velocity is higher because of the larger pressure gradient between the pore and the leaf. In this stage, the gas diffusion rate is higher. As the intake time increases, the pressure gradient gradually decreases, and the inlet velocity slows down. Consequently, the diffusion rate gradually reduces. Because of the coupling of light quantity and light direction, the gas diffusion rate significantly increases compared with the uncoupled model. Additionally, a diffusion model that does not consider fluid–solid coupling will overestimate the gas flow rate as the depth of gas entry increases. Therefore, the internal gas diffusion model must account for the effect of coupling on the diffusion rate.

Background: This study investigated factors influencing discrepancies between fractional flow reserve (FFR) and resting full-cycle ratio (RFR) in the functional assessment of coronary artery stenosis in patients with non-ST-segment elevation acute coronary syndrome (NSTE-ACS). Methods: We included 320 diseased vessels from 253 consecutive patients with NSTE-ACS. Vessels were categorized into four groups based on FFR ≤0.80 and RFR ≤0.89 thresholds: group 1 concordant negative (RFR–/FFR–), group 2 positive RFR and negative FFR (RFR+/FFR–), group 3 negative RFR and positive FFR (RFR–/FFR+), and group 4 concordant positive (RFR+/FFR+). Univariate and multivariate logistic regression analyses were conducted to identify predictors of diagnostic discrepancy between FFR and RFR. Results: Of the 320 diseased vessels, 182 (56.9%) were in group 1 (RFR–/FFR–), 33 (10.3%) in group 2 (RFR+/FFR–), 31 (9.7%) in group 3 (RFR–/FFR+), and 74 (23.1%) in group 4 (RFR+/FFR+). The concordance between FFR and RFR was 80.0%. Notably, left anterior descending artery (LAD) lesions exhibited significantly lower consistency compared to non-LAD lesions (p = 0.001), with distinct differences in FFR and RFR values between these groups (p < 0.001). The presence of a LAD lesion emerged as an independent predictor of diagnostic inconsistency between positive RFR and negative FFR measurements (p = 0.001). Conclusions: LAD involvement independently predicts diagnostic discrepancies between FFR and RFR in evaluating functional coronary artery stenosis in NSTE-ACS patients.

A pumped storage unit is a crucial guarantee in the pursuit of increased clean energy, especially in the progressively severe circumstances of low energy utilization and poor coordination of the integration of volatile renewable energy. However, due to their bidirectional operation design, pumped turbines possess an S-characteristic attribution, wherein the unsteady phenomena of unit vibration, pressure pulsation, and cavitation erosion happen during the start-up process and greatly impact the stable connection to the power grid. Therefore, a systematic study concentrating on an optimal unified paradigm of a start-up strategy for a pumped storage plant is conducted. Model construction, effective analysis, controller design, and collaborative optimization are sequentially expounded. Firstly, a refined start-up nonlinear model of a pumped storage plant with complex boundary conditions is constructed, wherein the delay time of frequency measurement, saturation, and dead zone features are comprehensively taken account. Furthermore, a variable universe fuzzy PID controller and its operation laws are proposed and specifically designed for the speed governing system of the pumped storage plant; the control quality and anti-disturbance performance are verified by a no-load frequency disturbance experiment. On this basis, taking speed overshoot for stationarity and speed rising time for rapidity, a novel open–close loop collaborative fuzzy control strategy is proposed with rotational speed feedback and a variable universe fuzzy PID control. The experiment results show that the proposed unified paradigm has better control performance in various performance indexes, and more balanced control quality and dynamic performance under various complex start-up conditions, which has great application value for ensuring the unit’s timely response to the power grid regulation task and improving the operating stability of the power system.

Fruit softening is a prominent attribute governing both longevity on shelves and commercial worth. Polygalacturonase (PG) plays a major role in strawberry fruit softening. However, the PG gene family in strawberry has not been comprehensively analyzed. In this study, 75 FaPG genes were identified in the octoploid strawberry genome, which were classified into three groups according to phylogenetic analysis. Subcellular localization prediction indicated that FaPGs are mostly localized to the plasma membrane, cytoplasm, and chloroplasts. Moreover, the expression of FaPGs during strawberry development and ripening of ‘Benihoppe’ and its softer mutant was estimated. The results showed that among all 75 FaPGs, most genes exhibited low expression across developmental stages, while two group c members (FxaC_21g15770 and FxaC_20g05360) and one group b member, FxaC_19g05040, displayed relatively higher and gradual increases in their expression trends during strawberry ripening and softening. FxaC_21g15770 was selected for subsequent silencing to validate its role in strawberry softening due to the fact that it exhibited the highest and most changed expression level across different developmental stages in ‘Benihoppe’ and its mutant. Silencing FxaC_21g15770 could significantly improve strawberry fruit firmness without affecting fruit color, soluble solids, cellulose, and hemicellulose. Conversely, silencing FxaC_21g15770 could significantly suppress the expression of other genes related to pectin degradation such as FaPG-like, FaPL, FaPME, FaCX, FaCel, FaGlu, FaXET, and FaEG. These findings provide basic information on the FaPG gene family for further functional research and indicate that FxaC_21g15770 plays a vital role in strawberry fruit softening.

To alleviate the problems of environmental pollution and energy crisis, aggressive development of clean and alternative energy technologies, in particular, water splitting, metal–air batteries, and fuel cells involving two key half reactions comprising hydrogen evolution reaction (HER) and oxygen reduction (ORR), is crucial. In this work, an innovative hybrid comprising heterogeneous Cu/Co bimetallic nanoparticles homogeneously dispersed on a nitrogen-doped carbon layer (Cu/Co/NC) was constructed as a bifunctional electrocatalyst toward HER and ORR via a hydrothermal reaction along with post-solid-phase sintering technique. Thanks to the interfacial coupling and electronic synergism between the Cu and Co bimetallic nanoparticles, the Cu/Co/NC catalyst showed improved catalytic ORR activity with a half-wave potential of 0.865 V and an excellent stability of more than 30 h, even compared to 20 wt% Pt/C. The Cu/Co/NC catalyst also exhibited excellent HER catalytic performance with an overpotential of below 149 mV at 10 mA/cm2 and long-term operation for over 30 h.
