Abstract

Streptococcus agalactiae is not only part of the human intestinal and urogenital microbiota but is also a leading cause of septicemia and meningitis in neonates. Its ability to cause disease depends upon the acquisition of nutrients from its environment, including the transition metal ion zinc. The primary zinc acquisition system of the pathogen is the Adc/Lmb ABC permease, which is essential for viability in zinc-restricted environments. Here, we show that in addition to the AdcCB transporter and the three zinc-binding proteins, Lmb, AdcA, and AdcAII, S. agalactiae zinc homeostasis also involves two streptococcal histidine triad (Sht) proteins. Sht and ShtII are required for zinc uptake via the Lmb and AdcAII proteins with apparent overlapping functionality and specificity. Both Sht-family proteins possess five-histidine triad motifs with similar hierarchies of importance for Zn homeostasis. Independent of its contribution to zinc homeostasis, Sht has previously been reported to bind factor H leading to predictions of a contribution to complement evasion. Here, we investigated ShtII to ascertain whether it had similar properties. Analysis of recombinant Sht and ShtII reveals that both proteins have similar affinities for factor H binding. However, neither protein aided in resistance to complement in human blood. These findings challenge prior inferences regarding the in vivo role of the Sht proteins in resisting complement-mediated clearance. IMPORTANCE This study examined the role of the two streptococcal histidine triad (Sht) proteins of Streptococcus agalactiae in zinc homeostasis and complement resistance. We showed that Sht and ShtII facilitate zinc homeostasis in conjunction with the metal-binding proteins Lmb and AdcAII. Here, we show that the Sht-family proteins are functionally redundant with overlapping roles in zinc uptake. Abstract

International audience; Background: Meningococcal meningitis requires rapid diagnosis and immediate management which is enhanced by the use of PCR for the ascertainment of these infections. However, its use is still restricted to reference laboratories. Methods: We conducted an inter-laboratory study to assess the implementation and the performance of PCR in ten French hospital settings in 2010. Results: Our data are in favour of this implementation. Although good performance was obtained in identifying Neisseria meningitidis positive samples, the main issue was reported in identifying other species (Streptococcus pneumoniae and Haemophilus influenzae) which are also involved in bacterial meningitis cases. Conclusions: Several recommendations are required and, mainly, PCR should target the major etiological agents (N. meningitidis, S. pneumonia, and H. influenzae) of acute bacterial meningitis. Moreover, PCR should predict the most frequent serogroups of Neisseria meningitidis according to local epidemiology. (Clin. Lab. 2012;58:343-346)

Abstract

With the aim of improving the knowledge of Listeria monocytogenes types distributed in poultry, isolates collected from a hundred eviscerated and refrigerated chicken carcasses from four different slaughterhouse plants in northwest Spain were characterised by serotyping and phage typing. Forty-two phage patterns were observed. Eight phage patterns (43, 431387, 43/387/4477/1652 and 43/19/387/575 for strains of serogroup 1/2 and 107, 108/107/47, 2389 and 2389/3274 for strains of serogroup 4) were the most common. Thus, thirty strains (56.60% of phage typeable strains) showed the above-mentioned or indistinguishable (≤1 major phage reaction differences for serogroup ½ strains or ≤2 major differences for serogroup 4) patterns. A wide distribution of some L. monocytogenes phage types was observed because different chicken carcasses (including carcasses from different slaughterhouses) were contaminated with the same or indistinguishable L. monocytogenes phage types. Besides, more than one type (serogroup or ph age type) of L. monocytogenes was detected in 43.75% of positive samples. Abstract

ABSTRACTFollowing a change in surgical practice, we noted that the rate at which Staphylococcus lugdunensiswas isolated from samples from the plastic surgery unit of our hospital increased considerably. We investigated the sources of these S. lugdunensisstrains, and we found that in the case of drain colonization or surgical site infection, the strain was more likely to have come from the patient's skin bacteria when the pubic site had been shaved preoperatively. To test the hypothesis of pubic site colonization, we evaluated the prevalence of S. lugdunensiscarriage among the cutaneous flora of the inguinal area. We found that 22% of 140 incoming patients carried S. lugdunensisin this area and that carriage at both inguinal folds was frequent (68% of carriers). A study of the genetic structure of the total population, including the clinical (n= 18) and the commensal (n= 53) strains, revealed that the diversity of the species was low and that the population was composed of two major groups that diverged at a distance of 35%. No particular characteristics made it possible to distinguish between clinical and commensal strains. Only isolates producing ß-lactamase were homogeneous; six of the eight ß-lactamase-positive strains displayed the same pulsed-field gel electrophoresis pattern.

48. Genetic relationship between Escherichia colistrains isolated from the intestinal flora and those responsible for infectious diseases among patients hospitalized in intensive care units

Abstract

The exact origin of strains of Escherichia coliresponsible for infectious diseases in intensive care units (ICUs) remains partly unknown. Our aim was to determine the nature of the link between strains from the intestinal flora of hospital staff, strains from the intestinal flora of patients hospitalized in ICUs and strains isolated from ICU patients with invasive diseases. For this purpose, 77 strains of E. coliwere genetically characterized by exploring their entire genomes by random amplified polymorphism of DNA (RAPD), and by determining their phylogenetic position in ECOR (E. colireference) groups, the virulence factors harboured (pap, sfa, afa, hly, aerand cnf) and their ability to mutate. The strains isolated from the intestinal flora of hospital staff were found to constitute a genetically heterogeneous population compared with the strains isolated from ICU carriers, which were highly clustered. The latter strains harboured numerous virulence factors, and 80% belonged to the group ECOR B2. The strains isolated from infected patients harboured fewer virulence factors than those from the ICU carriers, and only half belonged to ECOR B2. Moreover, these strains were more genetically related to strains from hospital staff than to strains from ICU carriers. Thus, the exogenous origin of the E. colistrains is probably almost as important as translocation from intestinal flora in ICUs. Moreover, a strong mutator phenotype had a minor, or no, role in the rapid adaptation to modifications in the ecological environment.

50. Gut metabolomic and microbiota analyses in ALS mice reveal specific metabolites despite the absence of significant gut dysbiosis.

Abstract

Objective: Over the past years, interest in the role of gut microbiota in neurodegenerative diseases has emerged. Despite numerous publications over the past decade, both in human and pre-clinical studies, there is no clear consensus on the microbiota's role or involvement in ALS. Few studies on mouse models of ALS highlighted a correlation between specific bacteria species and the prognostic or severity of the disease. Still these results lack reproducibility and remain controverted. In this article we present a study of fecal microbiota in the SOD1 G93A mouse model associated with a metabolomic analysis of cecum content, compared to controls., Methods: Intestinal metabolomic profile and fecal microbiota were assessed in two cohorts of SOD G93A mice compared to wildtype controls at the terminal stage of the ALS disease., Results: Results showed a significant difference in metabolomic profile in SOD1 G93A mice compared to controls but without a marked change in composition and diversity of fecal microbiota. Nevertheless, we observed an increase of Lachnospiraceae family, which are butyrate-producer bacteria, in SOD1 G93A mice. Moreover, some metabolites with significantly different intestinal concentrations are partially produced and linked with intestinal bacteria, such as riboflavin, hippurate, and N-acetylputrescine, leaving us convinced of the interest in looking further into the role of the microbiota in ALS., Conclusions: Despite an alteration of the intestinal metabolome in SOD1 G93A mice, microbiota data did not show significant changes, underlying the need for further research.
