IntroductionRecent research has explored the effectiveness of interactive virtual experiences in managing pain and anxiety in children during routine medical procedures, compared to conventional care methods. However, the influence of the specific technology used as an interface, 3-dimensions (D) immersive virtual reality (VR) vs. 2D touch screens, during pediatric venipuncture, remains unexamined. This study aimed to determine if immersive VR is more effective than a tablet in reducing pain and anxiety during short procedures.MethodsAn interactive game was designed by clinicians and psychologists, expert in pain theory, hypnosis, and procedural pain and anxiety relief, and was tailored for both VR and tablet use. Fifty patients were randomly assigned to either the Tablet or VR group. The primary outcome measures were pain and anxiety levels during the procedure. Secondary outcome measures included the need for physical restraint, duration of the procedure, enjoyment levels, and satisfaction ratings from both parents and nurses.ResultsParticipants, in both groups, had low levels of pain and anxiety. Physical restraint was infrequently used, procedures were brief, and high satisfaction levels were reported by patients, parents, and nurses.DiscussionThis study suggests that the type of technology used as a support for the game has a minimal effect on the child's experience, with both groups reporting low pain and anxiety levels, minimal physical restraint, and high enjoyment. Despite immersive VR's technological advancements, this study underscores the value of traditional tablets with well-designed interactive games in enhancing children's wellbeing during medical procedures.Clinical Trial Registration[ClinicalTrials.gov], identifier [NCT05065307].

Abstract Current differentiation protocols for generating mesencephalic dopaminergic (mesDA) neurons from human pluripotent stem cells result in grafts containing only a small proportion of mesDA neurons when transplanted in vivo. In this study, we develop lineage-restricted undifferentiated stem cells (LR-USCs) from pluripotent stem cells, which enhances their potential for differentiating into caudal midbrain floor plate progenitors and mesDA neurons. Using a ventral midbrain protocol, 69% of LR-USCs become bona fide caudal midbrain floor plate progenitors, compared to only 25% of human embryonic stem cells (hESCs). Importantly, LR-USCs generate significantly more mesDA neurons under midbrain and hindbrain conditions in vitro and in vivo. We demonstrate that midbrain-patterned LR-USC progenitors transplanted into 6-hydroxydopamine-lesioned rats restore function in a clinically relevant non-pharmacological behavioral test, whereas midbrain-patterned hESC-derived progenitors do not. This strategy demonstrates how lineage restriction can prevent the development of undesirable lineages and enhance the conditions necessary for mesDA neuron generation.

Mass reduction of mechanical systems is a recurrent objective in engineering, which is often reached by removing material from its mechanical parts. However, this material removal leads to a decrease of mechanical performances for the parts, which must be minimized and controlled to avoid a potential system failure. To find a middle-ground between material removing and mechanical performances), material must be kept only in areas where it is necessary, for example using stress-driven material removal methods. These methods use the stress field to define the local material removal based on two local parameters: the local volume fraction vf and the structural anisotropy orientation β. These methods may be based on different types of cellular structure patterns: lattice-based or bio-inspired. The long-term objective of this study is to improve the performance of stress-driven methods by using the most efficient pattern. For this purpose, this study investigates the influence of vf and β on the mechanical stiffness of three planar cellular structures called Periodic Stress-Driven Material Removal (PSDMR) structures. The first, taken from the literature, is bio-inspired from bone and based on a square pattern. The second, developed in this study, is also bio-inspired from bone but based on a rectangular pattern. The third is a strut-based lattice pattern well documented in the literature for its isotropic behavior. These three patterns are compared in this study in terms of relative longitudinal stiffness, obtained through linear elastic compressive tests by finite element analysis. It is highlighted that each PSDMR pattern has a specific domain in which it performs better than the two others. In future works, these domains could be used in stress-driven material removal methods to select the most adequate pattern or a mix of them to improve the performances of parts.

Background: Children and adolescents are highly vulnerable to the impact of sustained stressors during developmentally sensitive times. We investigated how demographic characteristics intersect with socioeconomic dimensions to shape the social patterning of quality of life and mental health in children and adolescents, two years into the COVID-19 pandemic. Methods: We used data from the prospective SEROCoV-KIDS cohort study of children and adolescents living in Geneva (Switzerland, 2022). We conducted an intersectional Multilevel Analysis of Individual Heterogeneity and Discriminatory Accuracy by nesting participants within 48 social strata defined by intersecting sex, age, immigrant background, parental education and financial hardship in Bayesian multilevel logistic models for poor health-related quality of life (HRQoL, measured with PedsQL) and mental health difficulties (measured with the Strengths and Difficulties Questionnaire). Results: Among participants aged 2–17 years, 240/2096 (11.5%, 95%CI 10.1–12.9) had poor HRQoL and 105/2135 (4.9%, 95%CI 4.0–5.9) had mental health difficulties. The predicted proportion of poor HRQoL ranged from 3.4% for 6–11 years old Swiss girls with highly educated parents and no financial hardship to 34.6% for 12–17 years old non-Swiss girls with highly educated parents and financial hardship. Intersectional strata involving adolescents and financial hardship showed substantially worse HRQoL than their counterparts. Between-stratum variations in the predicted frequency of mental health difficulties were limited (range 4.4%–6.5%). Conclusions: We found considerable differences in adverse outcomes across social strata. Our results suggest that, post-pandemic, interventions to address social inequities in HRQoL should focus on specific intersectional strata involving adolescents and families experiencing financial hardship, while those aiming to improve mental health should target all children and adolescents.

Behavioral flexibility and timely reactions to salient stimuli are essential for survival. The subcortical thalamic-basolateral amygdala (BLA) pathway serves as a shortcut for salient stimuli ensuring rapid processing. Here, we show that BLA neuronal and thalamic axonal activity in mice mirror the defensive behavior evoked by an innate visual threat as well as an auditory learned threat. Importantly, perturbing this pathway compromises defensive responses to both forms of threats, in that animals fail to switch from exploratory to defensive behavior. Despite the shared pathway between the two forms of threat processing, we observed noticeable differences. Blocking β-adrenergic receptors impairs the defensive response to the innate but not the learned threats. This reduced defensive response, surprisingly, is reflected in the suppression of the activity exclusively in the BLA as the thalamic input response remains intact. Our side-by-side examination highlights the similarities and differences between innate and learned threat-processing, thus providing new fundamental insights.

ABSTRACTNucleus, chromatin, and chromosome organization studies heavily rely on fluorescence microscopy imaging to elucidate the distribution and abundance of structural and regulatory components. Three-dimensional (3D) image stacks are a source of quantitative data on signal intensity level and distribution and on the type and shape of distribution patterns in space. Their analysis can lead to novel insights that are otherwise missed in qualitative-only analyses. Quantitative image analysis requires specific software and workflows for image rendering, processing, segmentation, setting measurement points and reference frames and exporting target data before further numerical processing and plotting. These tasks often call for the development of customized computational scripts and require an expertise that is not broadly available to the community of experimental biologists. Yet, the increasing accessibility of high- and super-resolution imaging methods fuels the demand for user-friendly image analysis workflows. Here, we provide a compendium of strategies developed by participants of a training school from the COST action INDEPTH to analyze the spatial distribution of nuclear and chromosomal signals from 3D image stacks, acquired by diffraction-limited confocal microscopy and super-resolution microscopy methods (SIM and STED). While the examples make use of one specific commercial software package, the workflows can easily be adapted to concurrent commercial and open-source software. Abstract Mind wandering (MW) and mindfulness have both been reported to be vital moderators of psychological wellbeing. Here, we aim to examine how closely associated these phenomena are and evaluate the psychometrics of measures often used to quantify them. We investigated two samples, one consisting of German-speaking unpaid participants (GUP, n $$=$$ = 313) and one of English-speaking paid participants (EPP, n $$=$$ = 228) recruited through MTurk.com. In an online experiment, we collected data using the Mindful Attention Awareness Scale (MAAS) and the sustained attention to response task (SART) during which self-reports of MW and meta-awareness of MW were recorded using experience sampling (ES) probes. Internal consistency of the MAAS was high (Cronbachs $$\alpha$$ α of 0.96 in EPP and 0.88 in GUP). Split-half reliability for SART measures and self-reported MW was overall good with the exception of SART measures focusing on Nogo trials, and those restricted to SART trials preceding ES in a 10 s time window. We found a moderate negative association between trait mindfulness and MW as measured with ES probes in GUP, but not in EPP. Our results suggest that MW and mindfulness are on opposite sides of a spectrum of how attention is focused on the present moment and the task at hand.

In 2023, the European Union will vote on the reauthorization of glyphosate use, renewed in 2017 despite concern on impacts on the environment and public health. A ban is supported by several Member States but rejected by most farmers. What are citizens' preferences to phase out glyphosate? To assess whether taxation could be an alternative to a ban, we conducted a discrete choice experiment in five European countries. Our results reveal that the general public is strongly willing to pay for a reduction in glyphosate use. However, while 75.5% of respondents stated to support a ban in the pre-experimental survey, experimental results reveal that in 73.35% of cases, earmarked taxation schemes are preferred when they lead to a strong reduction in glyphosate use for an increase in food price lower than that induced by a ban. When glyphosate reduction is balanced against its costs, a tax may be preferred.

In Simon-type interference tasks, participants are asked to perform a 2-choice reaction on a stimulus dimension while ignoring the stimulus position. Commonly, robust congruency effects are found; that is, reactions are faster when the relevant stimulus attribute and the assigned response match the location of the stimulus. Simon congruency effects are regularly attributed to a fast, nonverbal processing route. In 3 experiments, we tested the importance of verbal representations in the Simon effect by manipulating the format of representations (verbal vs. nonverbal) with stimulus material (i.e., words vs. gratings) and stimulus arrangement (i.e., horizontally vs. vertically). Results of the first experiment point to a modulation of the Simon effects by both factors when they were manipulated between subjects, up to an inversion of the Simon effect for words presented in vertical arrangement. We replicated the inverse congruency effect for verbal material in vertical arrangement when a within-participant design was used (Experiment 2) and when the impact of reading processes was ruled out (Experiment 3). One cause for this inversion might be the construction of language-based representations that counteract automatic processing given the stimulus arrangement. To investigate this, we assessed individual differences in the use of inner speech for self-instruction. Using hierarchical linear modeling analysis, we found that self-rated evaluative and motivational inner speech processes accounted for a significant portion of the Simon effect. This supports claims that individual differences predict performance even in simple cognitive tasks such as the Simon task and highlights the flexibility of basic cognitive processes.

This paper aims to understand why Ernst Cassirer never published his short but far-reaching reflections on Konrad Fiedler’s theory of art. While both thinkers do share a vision of art as a way to shape (and not simply to imitate) reality, I suggest that they have very different conceptions of artistic objectivity: Cassirer emphasises the symbolic (and therefore cultural) nature of the work of art; Fiedler focuses on the creative process itself, conceived of as an endless elucidation of visibility. In so doing, I demonstrate that Fiedler’s doctrine is not fully compatible with the philosophy of symbolic forms – hence perhaps Cassirer’s reluctance to address the issue publicly.

Mid-SUN proteins are a neglected family of conserved type III membrane proteins of ancient origin with representatives in plants, animals, and fungi. Previous higher plant studies have associated them with functions at the nuclear envelope and the endoplasmic reticulum (ER). In this study, high-resolution confocal light microscopy is used to explore the localisation of SUN3 and SUN4 in the perinuclear region, to explore topology, and to study the role of mid-SUNs on endoplasmic reticulum morphology. The role of SUN3 in the ER is reinforced by the identification of a protein interaction between SUN3 and the ER membrane-bound transcription factor maMYB. The results highlight the importance of mid-SUNs as functional components of the ER and outer nuclear membrane.

Lineage-restricted transcription factor PAX8 is oncogenic in ovarian cancer cells. Here the authors show that PAX8 interacts and recruits a splice variant of the MECOM locus PRDM3 to control the gene expression module involved in adhesion and extracellular matrix, and consequently promotes ovarian tumorigenesis.

As a self‐emissive technology, microLED displays retain all benefits of OLEDs and should be able to improve on many aspects, but will it be enough to guaranty success? In February 2024, leading proponent Apple canceled its microLED smartwatch project. More generally, as microLED keeps getting delayed and OLED keeps improving, the value proposition of microLED is shrinking.
