Abstract

Research was conducted to investigate the function of the beta-ketoacyl and acyl carrier protein components of bacterial type II polyketide synthases using polymerase chain reaction. DNA fragments were ligated to produce plasmids pWEC752 and pWEC769 while expression experiments were performed with either Streptomyces lividans 1326 or S. glaucescens WMH1077. Results showed that each end of the ketosynthase subunits established complexes with the ActI-Orf1 KSalpha.

Abstract

Analysis of the effects of active site mutations on the production of tetracenomycin F2 and C helps investigate the significance of potential active site motifs for acyltransferase and beta-ketoacyl:acyl carrier protein synthase regions in the TemK protein in Streptomyces glaucescens. Plasmids having C173A, C173S and H350L-S351A mutations do not produce any known intermediate, while 351A mutation produces high amounts of tetracenomycin F2. The production of TemK is not affected by any of these mutations.

Abstract

Anthracyclines, such as daunorubicin (DNR) and doxorubicin (DOX), are important antitumor antibiotics produced by microorganisms. They are made through a route involving the formation of a decaketide from propionyl-Coenzyme A and malonyl-Coenzyme A. Aklanonic acid (AA) biosynthesis of ruled by the DNR-DOX polyketide synthase (dps) genes encoding type II polyketide synthase (PKS). The behavour of heterologous PKSs is discussed.

Abstract

Ustilago trichophoraRK089 has been found recently as a good natural malic acid producer from glycerol. This strain has previously undergone adaptive laboratory evolution for enhanced substrate uptake rate resulting in the strain U. trichophoraTZ1. Medium optimization and investigation of process parameters enabled titers and rates that are able to compete with those of organisms overexpressing major parts of the underlying metabolic pathways. Metabolic engineering can likely further increase the efficiency of malate production by this organism, provided that basic genetic tools and methods can be established for this rarely used and relatively obscure species.

Abstract

The use of biodiesel derived crude glycerol for the production of value added chemicals has been discussed frequently. Current glycerol-based microbial production process often suffers from low rates, titers, and yields, hindering their industrial application. Here we report on the identification and optimization of Ustilago trichophora as promising novel production organism for malic acid from glycerol.
