12. Anticipative Tracking with the Short-Term Synaptic Plasticity of Spintronic Devices

Abstract

Real-time tracking of high-speed objects in cognitive tasks is challenging in the present artificial intelligence techniques because the data processing and computation are time-consuming resulting in impeditive time delays. A brain-inspired continuous attractor neural network (CANN) can be used to track quickly moving targets, where the time delays are intrinsically compensated if the dynamical synapses in the network have the short-term plasticity. Here, we show that synapses with short-term depression can be realized by a magnetic tunnel junction, which perfectly reproduces the dynamics of the synaptic weight in a widely applied mathematical model. Then, these dynamical synapses are incorporated into one-dimensional and two-dimensional CANNs, which are demonstrated to have the ability to predict a moving object via micromagnetic simulations. This portable spintronics-based hardware for neuromorphic computing needs no training and is therefore very promising for the tracking technology for moving targets., Comment: 9 pages, 6 figures

Abstract

Artificial intelligence based on artificial neural networks, which are originally inspired by the biological architectures of human brain, has mostly been realized using software but executed on conventional von Neumann computers, where the so-called von Neumann bottleneck essentially limits the executive efficiency due to the separate computing and storage units. Therefore, a suitable hardware platform that can exploit all the advantages of brain-inspired computing is highly desirable. Based upon micromagnetic simulation of the magnetization dynamics, we demonstrate theoretically and numerically that recurrent neural networks consisting of as few as 40 magnetic tunnel junctions can generate and recognize periodic time series after they are trained with an efficient machine-learning algorithm. With ultrahigh operating speed, nonvolatile memory and high endurance and reproducibility, spintronic devices are promising hardware candidates for neuromorphic computing., Comment: 5 pages, 4 figures

Abstract

Spatiotemporal information processing is fundamental to brain functions. The present study investigates a canonic neural network model for spatiotemporal pattern recognition. Specifically, the model consists of two modules, a reservoir subnetwork and a decision-making subnetwork. The former projects complex spatiotemporal patterns into spatially separated neural representations, and the latter reads out these neural representations via integrating information over time; the two modules are combined together via supervised-learning using known examples. We elucidate the working mechanism of the model and demonstrate its feasibility for discriminating complex spatiotemporal patterns. Our model reproduces the phenomenon of recognizing looming patterns in the neural system, and can learn to discriminate gait with very few training examples. We hope this study gives us insight into understanding how spatiotemporal information is processed in the brain and helps us to develop brain-inspired application algorithms., Comment: 9 pages, 6 figures

24. Mechanistic study of NUPR1 in bladder cancer development through transcriptional regulation of CCR2.

Abstract

Nuclear protein‐1 (NUPR1) (also known as p8) is one of the genes associated with transcription factors that participate in various aspects of cancer initiation and development. However, the molecular mechanisms of NUPR1 in bladder cancer (BLCA) remain unclear. We conducted an analysis of the correlation between NUPR1 expression and related genes using the Gene Expression Omnibus (GEO) online database. We employed lentivirus‐mediated small interfering RNA (siRNA) to knockdown the expression of NUPR1 in two human BLCA cell lines. In vitro experiments were conducted to validate the impact of NUPR1 interference on BLCA and the influence of NUPR1 on the transcription of chemokine receptor‐2 (CCR2). Furthermore, transcription factors for CCR2 were predicted using the PROMO database. Co‐immunoprecipitation (Co‐IP) and immunofluorescence double staining were used to detect the binding between NUPR1 and CCAAT/enhancer binding protein γ (CEBPG). In vivo and in vitro experiments were conducted to validate that NUPR1 regulates CCR2 transcription through CEBPG. In vitro experiments indicate that the suppression of NUPR1 inhibited BLCA growth. Analysis of the GEO database revealed a positive correlation between the expression of NUPR1 and CCR2. Luciferase experiments confirmed that NUPR1 influences the transcription of CCR2. Online data indicates that CEBPG is a transcription factor for CCR2. Co‐IP and immunofluorescence double staining confirmed binding between NUPR1 and CEBPG. Luciferase assays and chromatin immunoprecipitation (ChIP) demonstrate that CEBPG regulates the transcription of CCR2. Additionally, rescue experiments at the cellular level and animal experiments validated the aforementioned mechanism. NUPR1 promotes a promotional role in BLCA, and interference with NUPR1 can inhibit the proliferation and invasive abilities of BLCA. There was a correlation between the expressions of NUPR1 and CCR2, and NUPR1 binds with CEBPG in the cell nucleus. Abstract

Aim: To retrieve, analyse and summarize the relevant evidence on the prevention and management of bladder dysfunction in patients with cervical ancer after radical hysterectomy. Design: Overview of systematic reviews. Methods: 11 databases were searched for relevant studies from top to bottom according to the '6S' model of evidence‐based resources. Two independent reviewers selected the articles, extracted the data and appraised the quality of the included reviews based on different types of evaluation tools. Results: A total of 13 studies were identified, including four clinical consultants, four guidelines, four systematic reviews and one randomized controlled trial. 29 best evidence were summarized from five aspects, including definition, risk factors, assessment, prevention and management. Methylation-mediated gene silencing is closely related to the occurrence and development of human tumors. The euchromatic histone lysine methyltransferase 2 (EHMT2, also known as G9a) is highly expressed in many tumors and is generally considered to be an oncogene, which is associated with the poor outcome of many tumors. Combined immunotherapy and immune checkpoint blockade therapy also have good efficacy and certain safety. However, there are still many difficulties in the drugs targeting G9a, and the combined effect and safety of G9a with many drugs is still under study. This article aims to summarize the role and mechanism of G9a and its inhibitors in tumors in the past two years, and to understand the application prospect of G9a from the perspective of diagnosis and treatment. Abstract

Nowadays massive amount of data are available for analysis in natural and social systems. Inferring system structures from the data, i.e., the inverse problem, has become one of the central issues in many disciplines and interdisciplinary studies. In this Letter, we study the inverse problem of stochastic dynamic complex networks. We derive analytically a simple and universal inference formula called double correlation matrix (DCM) method. Numerical simulations confirm that the DCM method can accurately depict both network structures and noise correlations by using available kinetic data only. Abstract

Treatment of castration‐resistant prostate cancer (CRPC) is a long‐standing clinical challenge. Traditionally, CRPC drugs work by either reducing dihydrotestosterone biosynthesis or blocking androgen receptor (AR) signaling. Here it is demonstrated that AR inhibitor treatment gives rise to a drug‐tolerant persister (DTP) state. The thioredoxin/peroxiredoxin pathway is up‐regulated in DTP cells. Peroxiredoxin 5 (PRDX5) promotes AR inhibitor resistance and CRPC development. Inhibition of PRDX5 suppresses DTP cell proliferation in culture, dampens CRPC development in animal models, and stabilizes PSA progression and metastatic lesions in patients. Therefore, the study provides a novel mechanism and potential target for the management of castration‐resistant prostate cancer. [ABSTRACT FROM AUTHOR]
