Among biotechnologies of reproduction in the equine species, artificial insemination remains the most used technology especially for cooled transported sperm. Although the use of INRA96 extender has demonstrated its efficiency for long-term sperm storage at 4°C or 15°C, some stallions ("bad coolers") are excluded from such technology. Some years ago, we demonstrated that liposomes produced from egg yolk (EY) phospholipids could be an alternative to egg yolk plasma in stallion freezing extenders. To develop a new extender for sperm chilling, we evaluated the protective effect of liposomes produced from EY phospholipids on stallion sperm storage at 4°C. The sperm of stallions from two studs was diluted in INRA96 extender (as control) or an experimental extender (EE) composed of INRA96 supplemented with liposomes of EY phospholipids. After 24H (D1), 72H (D3), and 6 days (D6) or 7 days (D7), motility parameters were evaluated using Computer Assisted Semen Analyzer. Our results demonstrated that total and progressive motility decreased significantly after dilution and storage in INRA96 between D1 and D3 (P.05) while no significant decrease was observed between D1 and D3 with EE. Regarding VAP parameter, no significant difference was observed between extenders except at D7 in stud 2. Moreover, total and progressive motility were maintained at a significantly higher level (D3, D6, D7) when sperm was stored in EE compared to INRA96. These promising results demonstrate that the supplementation of INRA96 extender with egg-yolk phospholipids liposomes allows a higher protection to stallion sperm cells.

Most wild and domestic donkey breeds are currently endangered or threatened. Their preservation includes the creation of a Genome Resource Bank. Embryos cryopreservation allows the preservation of genetics from both male and female and is the fastest method to restore a breed. Because embryo production in vivo is limited in equids, our objective was to establish conditions for in vitro production of embryos in donkey using ovum pick up (OPU), IVM, IVF, and in vitro culture of zygotes. Donkey cumulus-oocyte complexes (COCs) were collected by transvaginal ultrasound-guided aspirations OPU in adult cyclic jennies and in vitro matured in tissue culture medium 199 supplemented with fetal calf serum and epidermal growth factor for 24, 30, 34, or 38 hours. They were preincubated with oviductal fluid for 30 minutes, coincubated with frozen-thawed donkey semen treated with procaine for 18 hours, and cultured for 30 hours in Dulbecco's Modified Eagle Medium-F12 supplemented with NaHCO3, fetal calf serum, and gentamycin. From the five OPU sessions, we collected 92 COCs in 193 follicles (48%) with an average of 4.2 COCs per jenny. All COCs were expanded after more than 24-hour IVM. At collection, jennies oocytes contained a germinal vesicle. Metaphase 1 oocytes were observed after 30-hour IVM and 44% were in metaphase 2 after 34-hour IVM. In our conditions, IVM of donkey oocytes was slower than IVM of equine oocytes and optimal duration for donkey oocytes IVM may be 34 hours. Only 15% of jennies oocytes contained two pronuclei after coincubation with donkey spermatozoa and none of them developed further after 48 hours post-IVF. Moreover, some parthenogenetic activation occurred. Thus, the treatment of donkey sperm with procaine may not be efficient for IVF. In conclusion, we established for the first time conditions for OPU in jennies with high recovery rates. We reported that IVM of jennies oocytes can produce 44% of metaphase 2 oocytes after 34 hours in culture and we described for the first time the chronology of IVM of donkey oocytes. Further studies are in progress to establish efficient conditions for IVF and development of donkey zygotes.

Forty-four percent were in metaphase 2 after 34 hours. In our conditions, IVM of donkey oocytes was slower than that of equine oocytes and the optimal duration for donkey oocytes IVM may be 34 hours. Oocytes we co-incubated with frozen-thawed donkey semen treated with procaine for 18 hours and cultured for 30 hours in a Dulbecco Modified Eagle Medium-F12-based medium. Only 15% of jennies oocytes contained 2 pronuclei after co-incubation, and none of them developed further after 48 hours after IVF. Treatment of donkey sperm with procaine may not be efficient for IVF. Some parthenogenetic activation occurred. In conclusion, we confirm that our conditions for OPU in jennies yielded high recovery rates that improved with operator experience. Maturation rates of 44% can be achieved using the IVM medium routinely used for equine oocytes in our lab. Further studies are in progress to establish efficient conditions for IVF and development of donkey zygotes.

The mechanism of fertilization remains largely enigmatic in mammals. Most studies exploring the molecular mechanism underlying fertilization have been restricted to a single species, generally the mouse, without a comparative approach. However, the identification of divergences between species could allow us to highlight key components in the mechanism of fertilization. In the pig, in vitro fertilization (IVF) and polyspermy rates are high, and spermatozoa penetrate easily through the zona pellucida (ZP). In contrast, IVF rates are low in the horse, and polyspermy is scarce. Our objective was to develop a comparative strategy between these two divergent models. First, we compared the role of equine and porcine gametes in the following five functions using intraspecific and interspecific IVF: ZP binding, acrosome reaction, penetration through the ZP, gamete fusion, and pronucleus formation. Under in vitro conditions, we showed that the ZP is a determining element in sperm-ZP attachment and penetration, whereas the capacity of the spermatozoa is of less importance. In contrast, the capacity of the spermatozoa is a key component of the acrosome reaction step. Second, we compared the composition and structure of the equine and porcine ZP. We observed differences in the number and localization of the ZP glycoproteins and in the mesh-like structure of the ZP between equine and porcine species. These differences might correlate with the differences in spermatozoal attachment and penetration rates. In conclusion, our comparative approach allows us to identify determining elements in the mechanism of fertilization.

In the procedure used in this paper, semen was first diluted in INRA82+2% egg yolk (E1) at 37 degrees C. Before or after cooling to 4 degrees C, semen was centrifuged and diluted in E1+2.5% glycerol (E2). Cooled semen was frozen in 0.5-ml straws. Straws were thawed at 37 degrees C for 30s. For fertility trials, frozen ejaculates were used only if total post-thaw motility was above 35%. Most mares were inseminated two times before ovulation with 400 x 10(6) total spermatozoa every 24h. This paper presents post-thaw motility (CASA) and fertility results obtained when some steps of the procedure were evaluated. Use of the first three jets of ejaculate before the centrifugation did not improve post-thaw motility compared to use of the whole semen (25% versus 25%, 2 stallions x 12 ejaculates, P0.80). When the first dilution was performed in E2 at 22 degrees C instead of in E1 at 37 degrees C, motility was slightly improved (38% versus 36%, n283 ejaculates per group, P0.04) but fertility was similar (51% versus 58%, n196 cycles per group, P0.10). Coating the spermatozoa with 0.5, 1, 2, 4 and 8mM of Concanavalin A resulted in unchanged post-thaw motility (6 stallions x 3 ejaculates, P0.05). The extender E2 was modified or supplemented with different substances. Increasing egg yolk concentration from 2 to 4% (v/v) did not increase post-thaw motility (42% versus 34%, 6 stallions x 2 ejaculates, P0.05). Different glycerol concentrations (range: 1.7-3.7%) had no significant effect on post-thaw motility even though 2.4-2.8% resulted in a nonsignificant higher motility (7 stallions x 2 ejaculates, P0.05). Glutamine at 50mM in E2 improved post-thaw motility compared with no glutamine (49% versus 46%, n584 ejaculates per group, P0.0001) but not fertility (53% versus 54%, n451 cycles per group, P0.80). Thawing at 75 degrees C for 10s slightly increased motility after 120 min at 37 degrees C (6 stallions x 1 ejaculate, P0.05) but no effect on per-cycle fertility was noted (32% (19 cycles) versus 41% (17 cycles), P0.50). When post-thaw dilution was performed using a fixed molarity multi-step system (25 mOsm per step) from various osmolarities (900-690 mOsm) to 365 mOsm, motility was unaffected compared with dilution in one step (36% versus 38%, 6 stallions x 1 ejaculate, P0.20).

International audience; The ovarian physiology of the mare is an interesting model. Actually, we could highlight that in contrast with other domestic mammals, no preovulatory LH surge occurs in this species, but LH rises gradually during oestrus with a maximum 1–2 days after ovulation. Moreover, in vitro maturation rate of oocytes remains low in the mare, and in vitro fertilisation methods give only limited success. Thus, follicular development and maturation could involve, at least in part, some species-specific mechanisms and studies on follicular fluid composition may contribute to understanding these events, as well as to alleviate some of the problems encountered. The studies reported here are on the evolution of intrafollicular content of steroids, protein patterns and insulin-like growth factor binding proteins (IGFBPs) profiles during follicular growth, maturation and atresia in the mare, in relation to follicular and oocyte quality. We demonstrated that (1) follicular growth of the dominant follicle is associated with an increase in intrafollicular oestradiol-17β and progesterone levels and a decrease in IGFBP-2 and IGFBP-5, (2) follicular maturation is associated with a decrease in intrafollicular estradiol-17β level, a further increase in progesterone level, a slight increase in IGFBP-2 and finally, the induction of the follicular expression of an Mr 200 000 protein, whose identity is still unknown, and (3) follicular preovulatory regression and atresia are characterised by alterations in steroidogenesis, as well as an increase in intrafollicular levels of IGFBP-2, IGFBP-4 and IGFBP-5. Interestingly, we obtained data which suggest that the expression of the Mr 200 000 protein relates to oocyte resumption of meiosis and cumulus expansion. Finally, relationships between follicular content and follicular quality needs further work in the mare, in order to predict fertility and to increase the embryo production in vivo and in vitro in this species.; Chez la jument, le taux de LH ne présente pas de pic au stade préovulatoire, mais plutôt une augmentation progressive présentant un maximum le lendemain de l’ovulation. Dans ce travail, l’évolution du contenu intrafolliculaire en stéroı̈des, protéines et IGFBP a été étudiée au cours de la croissance, la maturation et l’atrésie folliculaire chez la jument. Elle a été mise en relation avec la qualité folliculaire et ovocytaire. Nous avons montré que: (1) la croissance du follicule dominant se caractérise par l’augmentation des taux intrafolliculaires d’estradiol-17β et de progestérone, et la chute des taux d’IGFBP-2 et d’IGFBP-5, (2) la maturation du follicule dominant s’accompagne de la baisse du taux d’estradiol-17β, l’augmentation du taux de progestérone et d’IGFBP-2, ainsi que par l’expression d’une protéine de Mr 200 000 (p200) dont l’identité est inconnue, et enfin, (3) la régression préovulatoire et l’atrésie folliculaire se caractérisent par l’altération de la stéroidogenèse et l’augmentation des taux d’IGFBP-2, IGFBP-4 et IGFBP-5. Nos résultats montrent que l’expression de la p200 est liée à la maturation ovocytaire et à l’expansion du cumulus.

The supplementation of the freezing diluent with 3 amino acids (glutamine, proline and histidine) and 1 amino acid-related compound (betaine) in preserving stallion spermatozoa diluted in INRA82 extender containing 2.5% (v/v) glycerol and 2% (v/v) egg yolk (control extender) during freezing and thawing was studied at 0, 40, 80, 120 and 160 mM in 20 split ejaculates (10 stallions x 2 ejaculates; Experiment 1). Glutamine and proline were studied at 0, 10, 20, 30, 40, 50, 60, 70 and 80 mM in 20 split ejaculates (10 stallions x 2 ejaculates; Experiment 2). In each experiment, spermatozoa were evaluated after thawing by computer automated sperm analyzer. The percentage of motile spermatozoa (faster than 30 microns/sec) was assessed. In addition, the velocity of the average path (VAP), the straight line velocity (VSL), the curvilinear velocity (VCL) and the amplitude of the lateral head displacement (ALH) were also measured. In Experiment 1, only glutamine (40 mM) significantly improved sperm motility (56.0% +/- 3.0 vs 49.7% +/- 1.6; P < 0.05) compared with the control extender, while velocities were unaffected at concentrations of 40 to 120 mM. However, at 160 mM, a significant decrease in motility and velocity was observed for all amino acids. In Experiment 2, motility in glutamine (range 41.1% +/- 3.8%; 42.4% +/- 3.6) and proline (43.0% +/- 3.7; 45.6% +/- 3.8) extenders compared with the control (34.7% +/- 1.6) was improved significantly (P < 0.05). Sperm velocity was improved at concentrations higher than 40 mM glutamine and 50 mM proline.

The high-molecular-weight proteins of equine follicular fluid were examined to determine whether some polypeptides are unique to certain physiological conditions. Fluids from ovarian follicles of various diameters and physiological stages during the follicular phase were recovered by ultrasound-guided follicular aspiration. Granulosa cells and cumulus-oocyte complexes (COC) were recovered by scraping the intrafollicular wall during puncture. Follicular fluids and corresponding serum, as well as granulosa cell lysates, were analyzed by one-dimensional SDS-PAGE and silver staining. COC morphology was assessed microscopically. A 200-kDa protein band was demonstrated in fluids from preovulatory follicles, in natural conditions or after induction of ovulation. This protein band was absent in fluids from follicles at earlier stages, subordinate follicles, and serum. The presence of this protein at the preovulatory (PO) stage was ascertained through recovery of the fluid from follicles twice during their growth. Its appearance was time dependent after induction of ovulation but was not induced by an intrafollicular injection of a physiological dose of progesterone. We also demonstrated the presence of this 200-kDa protein in granulosa cells lysates recovered from preovulatory follicles. The expression of this protein in the follicular fluid was related to the cumulus aspect and chromatin configuration of the enclosed COC. No relation was found between its presence in the follicular fluid at the PO stage and subsequent ovulation of the punctured follicle or embryo production. The identification of this molecule is approached and discussed. These results show a novel PO stage-related protein in equine follicular fluid, which may be involved in the differentiation and maturation mechanisms occurring in the follicle during the preovulatory period.

Thus, after crossing with the AMH-Cre line, we generated and analyzed a new line that expressed a dominant-negative TRalpha1 isoform (TRalphaAMI) in SCs only. So-called TRalphaAMI-SC (TRalphaAMI/+ Cre+) mice exhibited similar phenotypic features to the knockout line: heavier testicular weight and higher sperm reserve, in comparison with their adequate controls (TRalphaAMI/+ Cre−). Egg yolk is normally used as a protective agent to freeze semen of equine and other species. However, addition of egg yolk in extenders is not without disadvantages and the demand to find cryoprotective alternatives is strong. The objective of this study was to test the cryoprotective capacities of liposomes composed of egg yolk phospholipids. Two experiments were conducted: 1) the first to determine the optimal composition and concentration of liposomes to preserve post-thaw motility and membrane integrity of spermatozoa; 2) the second to assess in vivo the cryoprotective capacities of these liposomes. In Experiment 2, post-thaw motility and membrane integrity of spermatozoa were also analyzed. Experiment 1 demonstrated that liposomes composed of phospholipids E80 (commercial lecithins from egg yolk composed mainly of phosphatidylcholine and phosphatidylethanolamine) and of Hank's salts-glucose-lactose solution (E80-liposomes) were the most efficient in preserving post-thaw motility. The optimal concentration was 4 % (v/v). In Experiment 2, fertility rate after artificial insemination of semen frozen with E80-liposomes was 55 % (22/40) compared with 68 % (27/40) with the control extender containing egg yolk (EY) (p = 0.23). Post-thaw motility parameters were higher with EY than with E80-liposomes (p < 0.0001). For post-thaw membrane integrity no difference was observed between the two extenders (p = 0.08). Liposomes composed of egg yolk phospholipids appeared to be a promising alternative to replace egg yolk in semen freezing extenders in equine species.

This study tested whether variable temperatures (from -0.5 to 15 degrees C) and air exposure could be used under laboratory and under field conditions to store stallion sperm diluted in extender INRA96 without loss of fertility. Experiment 1 (laboratory conditions) measured the effects of two 72 h storage conditions (5 degrees C with air vs. 15 degrees C without air). Experiment 2 (fixed field conditions) measured the effects of 22 h of storage without air in disposable containers maintained at four ambient temperatures (7 degrees C, 17 degrees C, 27 degrees C, 39 degrees C with semen at -0.5 degrees C to 3 degrees C, 4 degrees C to 7 degrees C, 8 degrees C to 10 degrees C, 12 degrees C to 15 degrees C, respectively). Per cycle pregnancy rate (PC) was measured after one artificial insemination (AI) in uterine body of 200 X 10(6) total spermatozoa, 7 h (Experiment 1) or 17 h (Experiment 2) before ovulation. In Experiment 1, PC was similar for both conditions (60% (n = 40 cycles) vs. 63% (n = 40), respectively, 5 stallions X 8 cycles). Only velocity VCL and ALH were slightly higher at 15 degrees C. In Experiment 2, PC was reduced when ambient temperature was low (semen at -0.5 degrees C to 3 degrees C; PC = 25%) rather than intermediate (semen at 4 degrees C to 7 degrees C; PC = 53%) or high (semen at 8 degrees C to 10 degrees C; PC = 50%) (4 stallions X 8 cycles) (P = 0.002). Sperm stored at -0.5 degrees C to 3 degrees C had lower acrosome integrity/responsiveness, similar membrane integrity (HOS test) and motilities, and higher VCL and ALH, than semen stored between 4 and 15 degrees C. These results demonstrate a wide tolerance of equine sperm to variable positive temperatures and air exposure for 22 h storage and more. However, temperatures close to 0 degrees C are detrimental for fertility.

Chantier qualité GA; The effects of in vitro storage on the sperm's ability to undergo the acrosome reaction (AR) have never been studied in avian species despite its major importance for reproduction management. The ability of chicken sperm to undergo the AR was measured after liquid storage at 4 °C and after cryopreservation, and its relationship with other semen quality parameters, including viability, mass motility and objective motility parameters measured by computer semen analyser (CASA) was analysed in two different flocks. The percentage of intact acrosome-reacting spermatozoa (IAR) was dramatically decreased by 48 h liquid storage (loss of 2/3 among the spermatozoa initially able to undergo the AR) whereas motility, viability and morphological integrity were reduced by 10–15%. By contrast, cryopreservation did not affect the induction of AR in flock 1 (29% IAR) whereas it was strongly affected in flock 2 (7% IAR). Motility parameters, viability and morphology were considerably altered by freezing in every case (more that 50% loss). Positive correlations were found between the percentage of intact acrosome-reacting spermatozoa and viability, mass motility and many objective motility parameters. Our results showed that the sperm's ability to undergo the AR was much more affected than other sperm functions after storage at 4 °C, while cryopreservation only had an effect in semen with the lowest initial quality. These results raise questions regarding the specific features of chicken sperm biology that must be taken into account in the treatment of semen.

International audience; A suitable method for the cryopreservation of donkey semen would be very valuable for the ex situ management of genetic diversity in this species. This report uses a variety of observation and trials to evaluate the effect of cryoprotectants in per-cycle pregnancy rates (PC) in equids females (jennies (donkey) and mares (horse)). This was explored by (1) comparing the results of insemination of jennies and mares with cooled or frozen donkey semen, (2) examining the possible toxic effect of the cryoprotectant (CPA) glycerol in these two species and (3) studying alternative solutions. Donkey and horse semen was either used immediately, or cooled according to some steps of the pre-freezing procedure or frozen and thawed. The pre-freezing procedure included semen dilution, centrifugation, resuspension in milk or in INRA82 + 2% egg yolk + various % CPA (expressed as final concentrations in extended semen (v/v)) and then cooling to 4 degrees C. PC was similar in mares and jennies inseminated with donkey semen cooled to 4 degrees C in milk. However, the PC was significantly higher in mares than in jennies when donkey semen was frozen with 2.2% glycerol (36%, n = 50 cycles vs. 11%, n = 38 cycles; P < 0.01). Increasing the concentrations of glycerol (0, 2.2, 3.5, 4.8%) before cooling stallion semen resulted in a progressive decrease in mare PC (87, 53, 53, 13% (n = 15 cycles for each concentration); P < 0.0001). The addition of 2.2% glycerol before cooling donkey semen decreased the PC measured in jennies to 0. The replacement of glycerol by 2% dimethylformamide increased the fertility obtained in jennies with cooled donkey semen (PC: 67%, n= 12 cycles) but did not increase the fertility obtained with frozen-thawed donkey semen (PC: 11%, n = 28 cycles with dimethylformamide vs. 0%, n = 16 cycles with glycerol). In conclusion, this study clearly shows that the ability of jennies to conceive after AI with donkey frozen semen is lower than that of mares. Glycerol affects the fertility of donkey and stallion spermatozoa as early as during the pre-freezing procedure. In consequence, the glycerol level must be low in frozen equine semen to provide good fertility. The toxic dose of glycerol for donkey spermatozoa seems to be almost half that for stallion spermatozoa. Whether this greater sensitivity of donkey spermatozoa to glycerol is responsible for the low success of semen cryopreservation in jennies is not so obvious because replacement of glycerol by dimethylformamide was not much more effective in terms of fertility. Background In human and rodents, sperm-zona pellucida binding is mediated by a sperm surface Galactosyltransferase that recognizes N-Acetylglucosamine residues on a glycoprotein ZPC. In large domestic mammals, the role of these molecules remains unclear: in bovine, they are involved in sperm-zona pellucida binding, whereas in porcine, they are not necessary. Our aim was to clarify the role of Galactosyltransferase and N-Acetylglucosamine residues in sperm-zona pellucida binding in ungulates. For this purpose, we analyzed the mechanism of sperm-zona pellucida interaction in a third ungulate: the horse, since the Galactosyltransferase and N-Acetylglucosamine residues have been localized on equine gametes. Methods We masked the Galactosyltransferase and N-Acetylglucosamine residues before the co-incubation of gametes. Galactosyltransferase was masked either with an anti-Galactosyltransferase antibody or with the enzyme substrate, UDP Galactose. N-Acetylglucosamine residues were masked either with a purified Galactosyltransferase or with an anti-ZPC antibody. Results and discussion The number of spermatozoa bound to the zona pellucida did not decrease after the masking of Galactosyltransferase or N-Acetylglucosamine. So, these two molecules may not be necessary in the mechanism of in vitro sperm-zona pellucida interaction in the horse. Conclusion The involvement of Galactosyltransferase and N-Acetylglucosamine residues in sperm-zona pellucida binding may have been lost during evolution in some ungulates, such as porcine and equine species.

International audience; The chemical composition of freezing extenders plays a major role in sperm cell survival during cryopreservation. In this study we compared two extenders for freezing stallion semen: INRA82 extender (as a control) versus INRA96 (R) extender, both supplemented with egg yolk and glycerol. INRA82 contains milk, whereas INRA96 (R) is a chemically- defined extender developed for fresh semen storage at 4 degrees C or 15 degrees C. Semen from 3 stallions (7 ejaculates per stallion) was frozen in both extenders. In vitro analyses of post-thaw motility of sperm cells (computer-assisted analysis) and of membrane integrity (flow cytometry analysis) were performed. Then a fertility trial was conducted. Inseminations were conducted in a total of 84 mare cycles. INRA96 (R) extender supplemented with egg yolk and glycerol significantly improved per-cycle pregnancy rates compared with INRA82 (71% versus 40%, p < 0.01). In agreement with these fertility results, membrane integrity was better preserved in INRA96 (R) than in INRA82. In contrast, motility parameters were significantly higher in INRA82 than in INRA96 (R). Further research is needed to understand how INRA96 (R) components protect sperm cells during the cryopreservation process and highly increase their fertility potential compared with INRA82 components.

In vitro storage of turkey spermatozoa is performed without consideration of the potential role of seminal plasma on sperm functions. We report the effects of seminal plasma on membrane permeability, lipid metabolism, energy status, motility and fertility of turkey spermatozoa stored at 4 or 20 degrees C. Phospholipid content (1077 nmol/10(9) spz versus 1219 nmol/10(9) spz at 48 h) and membrane permeability of spermatozoa were significantly damaged by the presence of seminal plasma after 48 h of storage at 4 degrees C, whereas damage to ATP content and fertility occurred earlier damaged by this presence (fertility after 24h storage 51% with seminal plasma versus 71% without). At 20 degrees C, seminal plasma decreased the phospholipid content of spermatozoa in the first hour of storage (1326 nmol/10(9) spz versus 1636 nmol/10(9) spz). Twenty-four hours later, this effect was masked by intense lipid peroxidation. These results show that seminal plasma is deleterious to storage of turkey spermatozoa at 4 degrees C and is involved in phospholipid metabolism of spermatozoa. Lipid peroxidation could be responsible for the acceleration of the degradation of sperm phospholipids during storage at 20 degrees C. However, lipid peroxidation seems not to be active at 4 degrees C. In this case, we suggest that phospholipase activation may contribute to sperm degradation, especially in the presence of seminal plasma.

Turkey semen quality is damaged by long term in vitro storage. The objective of the present study was to determine whether changes in energy substrates and antioxidants of semen extender could limit loss of quality and lipid content of turkey spermatozoa during storage. Spermatozoa were incubated in extenders based on Beltsville Poultry Semen Extender (BPSE) to which different energy substrates (acetate, pyruvate and hydroxybutyric acid) or antioxidant (Vitamin E) had been added. Semen was stored at 4 degrees C for 48 h and changes in quality, phospholipid and malondialdehyde (MDA) content of semen were evaluated. Among the different substrates studied, only acetate was able to limit the loss of motility and ATP content after 48 h in vitro storage. Losses of spermatozoal phospholipids were similar when gametes were incubated in an extender without any substrate or in normal BPSE (784-675nmol/10(9) spz versus 837-703 nmol/10(9) spz). However, motility and ATP content were significantly more affected after 48 h of storage in samples incubated without substrates than in BPSE (motility, 2.2 versus 0; ATP, 10 nmol/10(9) spz versus 3 nmol/10(9) spz). The addition of Vitamin E to the extender did not modify either the MDA or phospholipid content of fresh or stored spermatozoa, but increased the motility of stored semen. In conclusion, acetate is an essential substrate for in vitro storage. Spermatozoal phospholipids decreased during storage, but this did not seem to originate from metabolism of endogenous fatty acids. The positive effects of Vitamin E on semen storage did not originate from preservation of lipid oxidation.

In the horse industry, milk or milk-based extenders are used routinely for dilution and storage of semen cooled to 4-8 degrees C. Although artificial insemination (AI) with chilled and transported semen has been in use for several years, pregnancy rates are still low and variable related to variable semen quality of stallions. Over the years, a variety of extenders have been proposed for cooling, storage and transport of stallion semen. Fractionation of milk by microfiltration, ultrafiltration, diafiltration and freeze-drying techniques has allowed preparation of purified milk fractions in order to test them on stallion sperm survival. Finally, a high protective fraction, native phosphocaseinate (NPPC), was identified. A new extender, INRA96, based on modified Hanks' salts, supplemented with NPPC was then developed for use with cooled/stored semen. Four experiments were conducted to compare INRA96 and milk-based extenders under various conditions of storage. The diluted semen was maintained under aerobic conditions when stored at 15 degrees C, and anaerobic conditions when stored at 4 degrees C. In experiment 1, split ejaculates from 13 stallions were diluted either in INRA96 extender then stored at 15 degrees C or diluted in Kenney or INRA82 extenders and then stored at 4 degrees C for 24h, until insemination. In experiment 2, semen from two stallions was extended in INRA96 then inseminated immediately or stored at 15 degrees C for 3 days until insemination. In experiment 3, semen from three stallions was diluted in INRA96 then stored at 15 or 4 degrees C for 24h until insemination, finally, in experiment 4, split ejaculates from four stallions were diluted in INRA96 or E-Z Mixin extenders then stored at 4 degrees C for 24h until insemination. Milk-based semen diluents are known to be practical and effective in protecting equine spermatozoa during storage before artificial insemination. Milk is a biological fluid with a complex composition and contains components which are beneficial or harmful to spermatozoa. The aim of this study was to test the fertility of stallion semen after long-term storage using different milk diluents (INRA 82 or Kenney's diluent) vs one diluent chemically defined (INRA 96), which is composed of efficient milk components and optimized for sperm survival and storage temperature. The milk fraction used was that which best maintained spermatozoal survival based on motility measured in previous studies. Four breeding trials were conducted to determine the influence of combination of new diluent and storage conditions on fertility of the stallion. We compared the standard protocol of storing semen in a skim milk diluent (INRA 82 or Kenney's diluent) at 4 degrees C under anaerobic conditions with the experimental protocol which consisted of storing in a chemically defined, milk-free diluent (INRA 96), at 15 degrees C, under aerobic conditions. After 4 breeding trials, in which the semen was stored for 24 h under the 2 protocols, we obtained 57% (n = 178) and 40% (n = 173) of fertility per cycle using the experimental and the standard protocol respectively (p < 0.001). Another breeding trial was conducted to determine the influence of storage time on the fertility of spermatozoa. We have compared the fertility of semen inseminated immediately (68% of fertility per cycle, n = 50) vs the fertility of semen stored under the experimental protocol for 72 h before insemination (48% of fertility per cycle, n = 52). The experimental protocol improved sperm fertility compared to the standard protocol and seems to be a particular alternative for stallions with cold shock sensitive spermatozoa. Storing semen for 72 h under the experimental protocol seems to be useful in the field.

National audience; The objective of this study was to perform intracytoplasmic sperm injection (ICSI) on in vitro matured equine oocytes and to improve in vitro embryonic development on Vero cells after activation of the microinjected oocytes with calcium ionophore. After maturation (23 or 40 h, 38.5 degrees C, 5 % CO2), the cumulus-oocyte complexes were denuded, centrifuged and all oocytes exhibiting the first polar body were microinjected. ICSI was performed using fresh semen from three fertile stallions. Microinjected oocytes were activated with calcium ionophore A23187 (10 min, 10 mu M) and cultured individually for 7 days on Vero cells in microdrops. In seven trials, 353 cumulus-oocyte complexes were matured and 103 oocytes were microinjected. Eight oocytes were sham microinjected. After ICSI, 85 oocytes (82.5 %) survived the sperm injection procedure. Among the 76 successfully microinjected oocytes, 52 (68 %) were fertilized (two pronuclei, syngamy stage and cleaved oval. Sham microinjected oocytes were not activated. After in vitro culture, 35 ova (46 %) were cleaved 2 days after ICSI and early embryonic development was obtained (three embryos of 23 cells, 50 cells and more than 80 cells) 5 to 7 days after ICSI

New staining methods and automated instruments are now available to evaluate the sperm cell in vitro. Individual compartments of the sperm cell, such as the nucleus and the plasma and acrosomal membranes, may be investigated, as well as the cell function as shown by mitochondria activity and capacitation. Various probes are used and they can be analyzed by direct light or fluorescent microscopy or by flow cytometry. The automated instruments allow objective and accurate analysis and quantification as well as the ability to evaluate large population of cells in a shorter time, thus providing accurate evaluation of sperm quality. However, before these test can be recommended for routine clinical and investigational use, in the stallion, they need to be confirmed on a larger number of stallions and their correlation with traditional semen parameters and with stallion fertility has to be demonstrated.

The prediction of a stallion's fertility is one of the main unresolved problems in horse breeding. Despite many studies of the relationship between seminal characteristics and fertility, we are not able to evaluate with certainty the breeding capacity of a stallion. Stallion selection and fertility prediction are routinely based on the analysis of seminal and behavioural parameters. Basal blood hormone concentrations and their modification after different stimulation are also used for diagnosis. Although the methods developed in some laboratories may explain established subfertility or infertility, mainly detected by abnormal seminal characteristics and/or pregnancy rate, they cannot yet be used for general infertility detection. Such tests are based on spermatozoal morphology, e.g. chromatin and membrane integrity analyses, and spermatozoal functionality, e.g. motility evaluation, zona pellucida-sperm binding assay, and on seminal plasma composition. At this stage we have no idea of the frequency of the abnormalities demonstrated by these methods nor any idea of their correlation with subfertility or infertility.

La plupart des espèces d’ânes sauvages et domestiques est en danger d’extinction. Leur préservation nécessite la cryoconservation du sperme, des ovocytes et des embryons. L’objectif de cette étude était de mettre au point une technique de collecte in vivo et de maturation in vitro des ovocytes d’ânesses. Les ovocytes ont été collectés par ponction folliculaire transvaginale sous guide échographique, placés dans un milieu de maturation in vitro pendant 24, 30, 34 ou 38 heures puis analysés. Quatre-vingt-douze ovocytes ont été collectés dans 193 follicules (48%) avec une moyenne de 4,2 ovocytes par ânesse. Après 34 heures de maturation in vitro, 45% des ovocytes ont été matures. Pour la première fois une technique de collecte in vivo et de maturation in vitro d’ovocytes d’ânesses a été développée. Cette technique permet d’envisager la cryoconservation et la fécondation in vitro de ces ovocytes.

International audience; To fertilize the oocyte, the sperm undergoes physiological and biochemical modifications known as “sperm capacitation”. The aim of this study was to analyze by flow cytometry and immunofluorescence analysis the changes induced by several molecules on stallion sperm viability, plasma membrane fluidity, cholesterol content, calcium influx and tyrosine phosphorylation. Moreover, the modifications induced on glycocalyx sperm sugars were studied by means of lectin-based cell microarray analysis. Frozen sperm samples were incubated with calcium ionophore, MβCD, α-L-fucosidase, neurotensin, HSPA8, cAMP and 4-AP. Flow cytometry analysis showed that membrane fluidity was affected by MβCD, cAMP, neurotensin, HSPA8 and 4-AP; cholesterol depletion was induced by MβCD; an increase of intracellular calcium was produced by calcium ionophore and cAMP; and sperm viability was affected by MβCD. Tyrosine phosphorylation increased when sperm was incubated with MβCD and cAMP. Sperm microarray analysis revealed that i) MβCD and cAMP treatment increased α2,3-linked sialic acid and T antigen, whereas decreased α1,3-linked fucose signals; ii) cAMP alone increased α1,6- and α1,2-linked fucoses; iii) MβCD alone reduced sialylated T antigen, lactosamine, αGal, GalNAc, terminating glycans as well as α 1,6fucose in high-mannose glycans. The combination of MβCD + cAMP treatment preserved the α2,3-linked sialic acid increasing, determined the GalNAc, GlcNAc, α 1,2-linked fucose enhancement, whereas reduced T antigen and high-mannose glycans. Our results determine the specific effect of each molecule in order to establish a combination of several agents to achieve an efficient in vitro capacitation technique for stallion sperm.

For the success of artificial insemination of frozen semen, the composition of the freezing extender is a key factor. The objectives of the thesis were 1) to develop an extender to freeze stallion semen, ready to use and of optimized composition; 2) to acquire knowledge in gametes’ cryobiology. We showed that the cryoprotective ability of an extender based on native phosphocaseinate (INRA96®) supplemented with 2% egg yolk (EY) and 2.5% glycerol was significantly higher than the usual extender based on milk and EY (fertility/cycle of 71% versus 40%, p0.05, n=70cycles). These whole results permitted the development of an extender of standardized composition (patent FR2925917), sterile and marketable in a ready to use form (INRA-Freeze®, IMV-Technologies). In order to optimize the extender and to precisely identify the molecules implicated in the cryoprotection, the EY ,cryoprotectives ability of phospholipids liposomes mainly composed of EY phosphatidylcholine and phosphatidylethanolamine. The promising results we got (fertility/cycle of 55% with liposomes versus 68% with EY, p>0.05, n=80cycles) remain to be improved, in particular by increasing the insaturation degree of the phospholipids’ fatty acids of liposomes., Pour la réussite de l’insémination artificielle de semence congelée, la composition du milieu de congélation est un facteur clef. Les objectifs de la thèse ont été 1) de mettre au point un milieu pour congeler la semence équine, qui soit prêt à l’emploi et de composition optimisée ; 2) d’acquérir des connaissances en cryobiologie des gamètes. Nous avons démontré que le pouvoir cryoprotecteur d’un milieu à base de phosphocaséinate natif (l’INRA96®) supplémenté de 2% de jaune d’oeuf (JO) et de 2.5% de glycérol, était significativement supérieur à celui du milieu habituel, à base de lait et JO (71% de fertilité/cycle versus 40%, p0.05, n=70cycles). L’ensemble de ces résultats a permis la mise au point d’un milieu de formulation standardisée (brevet FR2925917), stérile et commercialisable prêt à l’emploi (INRAFreeze ®, IMV-Technologies). Dans le but d’optimiser le milieu et d’identifier précisément les molécules impliquées dans la cryoprotection, la démarche de fractionnement du JO a été poursuivie. Suite à des analyses in vitro, nous avons testé in vivo le pouvoir cryoprotecteur de liposomes de phospholipides composés essentiellement de phosphatidylcholine et de phosphatidyléthanolamine de JO. Les résultats encourageants obtenus (55% de fertilité/cycle avec les liposomes versus 68% avec le JO, p>0.05, n=80cycles) restent à améliorer, notamment en augmentant le degré d’insaturation des acides gras des phospholipides des liposomes.
