Rising oceanic temperatures create more frequent coral bleaching events worldwide and as such there exists a need for rapid, non-destructive survey techniques to gather greater and higher definition information than that offered by traditional spectral based monitoring systems. Here, we examine thermally induced laboratory bleaching of Montipora capricornis and Montipora confusa samples, utilising hyperspectral data to gain an understanding of coral bleaching from a spectral standpoint. The data revealed several characteristic spectral peaks that can be used to make health determinations. The fluorescence peaks are attributed to fluorescent proteins (FPs) and Chlorophyll-a fluorescence. The reflectance peaks can be attributed to Chlorophyll absorption and accessory pigments such as Peridinin and Diadinoxanthin. Each characteristic spectral peak or ‘marker’ allows for observation of each aspect of coral health and hence, simultaneous monitoring of these markers using hyperspectral imaging techniques provides an opportunity to better understand the processes occurring during bleaching and the rates at which they occur relative to one another.

4057 Background: While several clinical scoring systems exist to aid prognostication and patient (pt) selection for clinical trials in oncology, none are standardly used. We compared the ability of four prognostic scores to predict overall survival (OS) in pts with advanced gastric and esophageal (GE) cancer. Methods: Pts with advanced (unresectable or metastatic) GE cancer receiving first-line palliative-intent systemic therapy at the Princess Margaret Cancer Centre from 2007 to 2020 were included. High prognostic risk pts were identified using four scoring systems: Royal Marsden Hospital (RMH), MD Anderson Cancer Centre (MDACC), Gustave Roussy Immune Score (GRIm-S) and MD Anderson Immune Checkpoint Inhibitor (MDA-ICI) score. OS was estimated using the Kaplan-Meier method and compared between risk groups (high vs. not-high) for each scoring system using the log-rank test. Cox proportional hazards models were used to analyze the association between each prognostic score and OS, adjusting for baseline clinical factors. Harrell’s c-index was used to evaluate predictive discrimination of the models. Time-dependent AUCs were used to measure predictive ability for early death (within 90 days). Results: In total, 451 pts with advanced GE cancer were included. The median age was 59 years, 68% were male, 51% had ECOG status 0-1, 63% presented with de novo metastatic disease. The proportion of pts categorized as high risk was: RMH 25% (N=113), MDACC 13% (N=95), GRIm-S 24% (N=109), MDA-ICI 26% (N=117). The last two years have been a crash course in educating the world about viruses, virology and infectious diseases. Unsurprisingly, viruses have emerged as the harbingers of doom. However, with their newly acquired knowledge, the new armchair virologists fail to grasp the importance of viruses in the living world. They may cause us harm, but without their relentless activity, we’d all be dead anyway. In addition to owing them our lives, they also have the potential to improve our lives with novel biotechnological applications.

Visualization of viruses and their hosts has been paramount to their study and understanding. The direct observation of the morphological dynamics of infection is a highly desired capability and the focus of instrument development across a variety of microscopy technologies. This study demonstrates progress that has been made in exploiting the capabilities offered by HS-AFM to characterise the interactions between coccolithoviruses and their globally important coccolithophore hosts. We observe whole Emiliania huxleyi Virus capsids, transient binding to Emiliania huxleyi derived supported lipid bilayers, and host-virus binding in real-time in an environmentally relevant, aqueous environment.

Changing patterns in diversity are a feature of many habitats, with seasonality a major driver of ecosystem structure and function. In coastal marine plankton-based ecosystems, seasonality has been established through long-term time-series of bacterioplankton and protists. Alongside these groups, fungi also inhabit coastal marine ecosystems. If and how marine fungi show long-term intra- and inter-annual diversity patterns is unknown, preventing a comprehensive understanding of marine fungal ecology. Here, we use a 17-year environmental DNA time-series from the English Channel to determine long-term marine fungal diversity patterns. We show that fungal community structure progresses at seasonal and monthly scales and is only weakly related to environmental parameters. Communities restructured every 52-weeks suggesting long-term stability in diversity patterns. Some major marine fungal genera have clear inter-annual recurrence patterns, re-appearing in the annual cycle at the same period. Low relative abundance taxa that are likely non-marine show seasonal input to the coastal marine ecosystem suggesting land–sea exchange regularly takes place. Our results demonstrate long-term intra- and inter-annual marine fungal diversity patterns. We anticipate this study could form the basis for better understanding the ecology of marine fungi and how they fit in the structure and function of the wider coastal marine ecosystem.

AimsThe majority of pancreatic ductal adenocarcinomas (PDACs) harbour oncogenic mutations inKRASwith variants inTP53,CDKN2AandSMAD4also prevalent. The presence of oncogenic fusions includingNTRKfusions are rare but important to identify. Here we ascertain the prevalence ofNTRKfusions and document their genomic characteristics in a large series of PDAC.MethodsWhole genome sequencing and RNAseq were performed on a series of patients with resected or locally advanced/metastatic PDAC collected between 2008 and 2020 at a single institution. A subset of specimens underwent immunohistochemistry (IHC) analysis. Clinical and molecular characterisation and IHC sensitivity and specificity were evaluated.Results400 patients were included (resected n=167; locally advanced/metastatic n=233). Three patients were identified as harbouring anNTRKfusion, twoEML4-NTRK3(KRAS-WT) and a single novelKANK1-NTRK3fusion. The latter occurring in the presence of a subclonalKRASmutation. Typical PDAC drivers were present including mutations inTP53andCDKN2A. Substitution base signatures and tumour mutational burden were similar to typical PDAC. The prevalence ofNTRKfusions was 0.8% (3/400), while inKRASwild-type tumours, it was 6.25% (2/32). DNA prediction alone documented six false-positive cases. RNA analysis correctly identified the in-frame fusion transcripts. IHC analysis was negative in theKANK1-NTRK3fusion but positive in aEML4-NTRK3case, highlighting lower sensitivity of IHC.ConclusionNTRKfusions are rare; however, with emerging therapeutic options targeting these fusions, detection is vital. Reflex testing forKRASmutations and subsequent RNA-based screening could help identify these cases in PDAC.

Barrows are a prominent feature of Britain's Bronze Age landscape. While they originated as burial monuments, they also appear to have acquired other roles in prehistory. British prehistorians, however, have been hampered in their interpretations of these monuments, as they are wary of speculating about how Bronze Age people might have conceptualised their dead. Here, the authors suggest that a recurring pattern of inversion is significant. They use Conceptual Metaphor Theory to argue that Bronze Age people in Britain saw their dead inhabiting an inverted underworld directly beneath the surface of the earth. This interpretation would explain not only burial practices, but also some of the barrows’ other apparent functions, such as guarding boundaries and controlling routeways.

In the summer months, the waters of the Sargasso Sea are nutrient-limited and strongly stratified, serving as a model system for the predicted warmer and nutrient-limited oceans of the Anthropocene. The dominant microorganisms of surface waters are key drivers of the global carbon cycle. However, the viruses of the Sargasso Sea that shape these host communities and influence host biogeochemical function are not well understood. Here, we apply a hybrid sequencing approach that combines short- and long reads to survey Sargasso Sea phage communities via metagenomics at the viral maximum (80m) and mesopelagic (200m) depths. Taxonomically, we identified 2,301 Sargasso Sea phage populations (~species-level taxonomy) across 186 genera. Over half of the phage populations lacked representation in other global ocean viral metagenomes, whilst 177 phage genera lacked representation in phage isolate databases. Viral fraction and cell-associated viral communities captured in short-read data were distinct and decoupled at both depths, possibly indicating low active lytic viral replication in the Sargasso Sea, with viral turnover occurring across periods longer than the sampling period of three days. Inclusion of long read data was critical for (1) the identification of 79 ecologically important and common viral genomes; (2) capturing the extent of viral genome microdiversity; and (3) enabling the recovery of hypervariable regions in viral genomes predicted to encode proteins involved in host recognition, DNA synthesis and DNA packaging. Host prediction was only possible for ~4% of viral populations.
